Abstract:In this study, we propose a simple, cost-effective, and sensitive high-performance liquid chromatography method with fluorescence detection (HPLC-FLD) for the simultaneous determination of the three bisphenols (BPs): bisphenol A bis (2,3-dihydroxypropyl) ether (BADGE 2H2O), bisphenol F (BPF), and bisphenol E (BPE) in human urine samples. The dispersive solid phase extraction (d-SPE) coupled with solid phase extraction (SPE) procedure performed well for the analytes with recoveries in the range of 74.3–86.5% and relative standard deviations (RSD%) less than 10%. The limits of quantification (LOQs) for all investigated analytes were in the range of 11.42–22.35 ng mL−1. The method was validated at three concentration levels (1 × LOQ, 1.5 × LOQ, and 3 LOQ). During the bisphenols HPLC-FLD analysis, from 6 min a reinforcement (10 or 12) was used, therefore analytes might be identified in the small volume human urine samples. The results demonstrated clearly that the approach developed provides reliable, simple, and rapid quantification and identification of three bisphenols in a urine matrix and could be used for monitoring these analytes.

Abstract:Background: Identification and quantitative determination of analytes released from the packaging material is undoubtedly a difficult and tricky task, requiring the chemical analyst to develop an individual approach to obtain reliable analytical information. Unfortunately, it is still challenging for scientists to determine bisphenols at trace or even ultra-trace levels in samples characterized by a very complex, and often variable, matrix composition. Objective: Optimization and application of QuEChERS/d-SPE coupled with HPLC-DAD (and LC-QqQ-MS) method for the simultaneous determination of bisphenols (A, S, F, B, BADGE and derivatives) in milk samples from a can and breast milk samples have been performed. Methods: Concerning the analysis of unconjugated analytes, after the thawing and shaking the sample (5 mL breast milk or 10 mL milk samples from a can), it was transferred into a 50 mL polypropylene centrifuge tube. For the analysis of the total amount of analytes, prior to the extraction with acetonitrile, a deconjugation step was implemented in a tube by adding to sample, the an Isotopically Labelled Internal Standard (IS) solution (50 ng/mL) and 1 mL of the enzymatic solution with the β-Glucuronidase (3500 U/mL). The mix was homogenized and incubated for 16–18 h at 37 °C. Next, 10 mL of acetonitrile, and a QuEChERS salt packet (4 g anhydrous MgSO4, 1 g NaCl) were added. After shaking and centrifugation, the total acetonitrile layer was isolated in a polypropylene tube evaporate to dryness, and reconstitute in 1.2 mL acetonitrile. During d-SPE step the extract was transferred into a 15 mL polypropylene tube with Z-Sep and primary secondary amine (PSA). Next, shake the tube, store in fridge, and centrifuge for 15 min. The acetonitrile supernatant was obtained with a pipette and evaporated to dryness. Mixture MeOH: water (20:80, v/v) were added to the dry residue and the extract was reconstitute in 200 μL and analyzed by HPLC-DAD and HPLC–QqQ-MS equipment. Conclusion: Six different salts during d-SPE step were evaluated such as: zirconium dioxide-based sorbent (Z-Sep, Z-Sep Plus), primary secondary amine (PSA), octadecyl (C18), EMR-Lipid, Chitin and also their mixtures. Negligible matrix interference was observed for most of the analytes due to application of Z-Sep and PSA in dispersive-solid phase extraction clean-up step. Extraction of target analytes was performed using QuEChERS/d-SPE cleanup, and presents good performance for selected analytes with recoveries in the range of 15–103% and relative standard deviations (RSD) less than 10% in breast milk samples.

A Validated Method for Rapid Determination of Dibenzo-P-dioxins/furans (pcdd/fs), Polybrominated Diphenyl Ethers (pbdes) and Polychlorinated Biphenyls (pcbs) in Human Milk: Focus on Utility of Tandem Solid Phase Extraction (SPE) Cleanup

Abstract:An improved method based on tandem solid phase extraction (SPE) cleanup and gas chromatography-high resolution mass spectrometry (GC-HRMS) has been validated for a rapid determination of dibenzo-p-dioxins/furans (PCDD/Fs), dioxin-like polychlorinated biphenyls (PCBs), marker polychlorinated biphenyls (M-PCBs), and polybrominated diphenyl ethers (PBDEs) using a large volume (50 mL) of human milk. This method was well validated for the measurement of these analytes in human milk from the general population with low limits of detection (LODs, 0.004–0.12 ng/g lipid), satisfactory accuracy (75–120 % of recoveries), and precision [less than 10 % of relative standard deviations (RSDs)]. To comprehensively evaluate the performance of this method, a good, presently validated and routinely used method based on an automated sample clean-up system (ASCS, based on the commercial acid multilayer silica, basic alumina, and carbon columns) was used in parallel for comparison. Compared with the ASCS method, this method presented comparable specificity. Additionally, this method, in contrast to ASCS method, highly reduced consumption of solvents (40 mL versus 500 mL), which results in much lower background in the procedural blank, reduced time, and enhanced sample pretreatment throughput. This method was also applied in a pilot study to measure a batch of human milk samples with satisfactory results.
Determination of 16 Bisphenols in Drinking Water by Online Solid-Phase Extraction-Ultra High-Performance Liquid Chromatography Tandem Mass Spectrometry

Abstract:BackgroundBisphenol compounds are non-persistent environmental endocrine disruptors and frequently detected in drinking water systems, indicating potential human health risks through drinking water. ObjectiveTo establish and optimize a simultaneous determination method for 16 BPs in drinking water by online solid-phase extraction-ultra high-performance liquid chromatography-triple quadrupole mass spectrometry, in order to efficiently monitoring BPs in drinking water. MethodsCandidate online solid-phase extraction conditions, chromatographic columns, mobile phase systems, mass spectrometry parameters, and other conditions were compared by chromatographic peaks of BPs, and processing conditions such as water sample preservation and pretreatment were optimized. The pH level of drinking water samples was adjusted and solid particles were removed. After extraction and purification by an online solid-phase extraction system, samples were detected by ultra-high performance liquid chromatography tandem mass spectrometry and quantified by isotope internal standard method. The proposed method was verified by pure water and terminal tap water, and evaluated by spiked recovery rate and relative standard deviation. Eighty-eight tap water samples from different regions of local pipeline network were collected for method application. ResultsFor the 16 BPs, the calibration curves showed good linearity between 1.0 and 75 ng·L−1 and the correlation coefficients were greater than 0.995. The detection limit of the method was less than 0.30 ng·L−1, and the quantification limit of the method was less than 1.0 ng·L−1. When the spiked concentrations for the 16 BPs were 5.0, 15, and 40 ng·L−1, the average spiked recovery rates of the test substances were between (100 ± 10)%, and the relative standard deviations were all below 10%. In the method application to the local terminal water samples, the positive rates of bisphenol S (BPS), bisphenol A (BPA), and bisphenol AF (BPAF) were as high as 93.2%, 77.3%, and 29.5%, respectively. The concentrations of BPS were from not detected (N.D.) to 37.8 ng·L−1, and the concentrations of BPA were from N.D. to 52.0 ng·L−1. ConclusionThe method using an online solid-phase extraction system is established, featuring simple pre-treatment, small sample volume, high degree of automation, low detection limit, and good accuracy and precision. This method can be applied to the quantitative monitoring of 16 BPs at ng·L−1 level in drinking water.
Application of Solid-Phase Extraction and High-Performance Liquid Chromatography with Fluorescence Detection to Analyze Eleven Bisphenols in Amniotic Fluid Samples Collected during Amniocentesis

Abstract:Amniocentesis involves taking a sample of the amniotic fluid in order to perform a karyotype test and diagnose any genetic defects that may affect the fetus. Amniotic fluid has been collected from patients with an indication for amniocentesis in the 15–26th week of pregnancy. A simple and sensitive high-performance liquid chromatography with fluorescence detection (HPLC-FLD) method for identification and quantification of eleven selected bisphenols in amniotic fluid samples is proposed. The proposed method involved protein precipitation using acetonitrile, and next the extraction and concentration of analytes by solid-phase extraction (SPE). The solid-phase extraction (SPE) procedure with application of Oasis HLB SPE columns performed well for the majority of the analytes, with recoveries in the range of 67–121% and relative standard deviations (RSD%) less than 16%. The limits of detection (LODs) and quantification (LOQs) of all the investigated analytes were in the range of 0.8–2.5 ng mL−1 and 2.4–7.5 ng mL−1 (curves constructed in methanol) and 1.1–5.2 ng mL−1 and 3.2–15.6 ng mL−1 (curves constructed in the amniotic fluid), respectively. The method was validated at the following two concentration levels: 10 ng mL−1 (2 × LOQ) and 20 ng mL−1 (4 LOQ). The results confirm the validity of the SPE procedure and HPLC-FLD method for identification and quantification of bisphenols in amniotic fluid samples collected during an amniocentesis. The result obtained show that HPLC-FLD is a useful method for determination of bisphenol residues at nanogram per milliliter concentrations in amniotic fluid samples. Residues of five analytes (BADGE·2H2O, BPAF, BADGE, BADGE·H2O·HCl and BADGE·2HCl) were detected in amniotic fluid samples. Additionally, the harmfulness of bisphenols as potential pathogens that may cause karyotype disorders and contribute to preterm birth was estimated.

Selection of scFv phages on intact cells under low pH conditions leads to a significant loss of insert-free phages.

Abstract:Display of functional antibody fragments on the surface of filamentous bacteriophages allows fast selection of specific phage antibodies against a variety of target antigens. However, enrichment of single chain variable fragment (scFv)-displaying phages is often hampered by the abundance of bacteriophages lacking antibody fragments. Moderate adhesive binding activities and production advantages of these "empty" phages results in their subsequent enrichment during selection on target cells. To date, very limited effort has been made to develop strategies removing nonspecific binding phages during the selection processes. To efficiently reduce insert-free phages when panning on intact cells, we increased the washing stringency by lowering the pH of the buffer with citric acid. Under standard washing procedures (pH 7.4), only approximately 73% of recovered phages were insert-free after three rounds of selection. Using stringent washing procedures (pH 5.0), approximately 12% of recovered phages contained no scFv. Using this protocol, we have cloned an antibody fragment from a mouse/human hybridoma cell line directed against the disialoganglioside GD2. This study confirms that selection of phage antibodies on cells is efficiently enhanced by assays augmenting the stringency to remove nonspecific binding phages.
Dispersive micro solid phase extraction (DMSPE) using polymer anion exchange (PAX) as the sorbent followed by UPLC-MS/MS for the rapid determination of four bisphenols in commercial edible oils

Abstract:The present work presents a novel and rapid analytical method for the simultaneous analysis of bisphenol A (BPA), bisphenol B (BPB), bisphenol F (BPF) and bisphenol S (BPS) in edible oil based on dispersive micro solid phase extraction (DMSPE) for the first time followed by isotope dilution-ultra high performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). The edible oil sample was dispersed by n-hexane and extracted with ammoniated methanol-water solution. Then the target analytes were dispersedly absorbed using the polymer anion exchange (PAX) as the sorbent and eluted by acidic methanol. After that, four bisphenols were separated on a C18 column by gradient elution with methanol and 0.05% ammonium hydroxide in water as mobile phase, detected by MS/MS under multiple reactions monitoring (MRM) mode and quantified by internal standard method. The PAX amounts, adsorption time, concentrations of formic acid in the elution solvent and volume of elution solvent for the DMSPE technique were optimized. The limit of detection and quantitation (LOD and LOQ), matrix effect, recovery and precision of the developed method were investigated. Results indicated that BPS and the rest three bisphenols displayed excellent linearity in the concentration ranges of 0.1-50μg/L and 0.5-250μg/L, respectively, with correlation coefficients (R2) all larger than 0.998. Achieved MLODs (S/N=3) varied between 0.1-0.4μg/kg for all bisphenols. The mean recoveries at three spiked levels in edible oil were in the range of 87.3-108%. Intra-day precision (n=6) and inter-day precision (n=5) were <9% and <11%, respectively. This method is of rapid-and-simple pretreatment, accurate and sensitive, and suitable for the simultaneous determination of bisphenols in edible oil.

Abstract:This paper developed a novel and environmental microextraction for bisphenol A from milk samples using liquid-liquid microextraction with low-temperature partitioning, which is a sample clean-up step for endocrine disruptors, being a suitable alternative. Furthermore, the microextraction and determination steps were optimized to determine and quantify the bisphenol A from milk. Thus, the determination of it was made using HPLC with fluorescence detection, obtaining the analytical with a linear range from 5.0 to 150 μg L −1 bisphenol (Area = 11,894 C (μg L −1 ) + 22,595) with good linearity (r = 0.9983), quantification (5.0 μg L −1 ) limit, and variation coefficient (4.4%), which were according to the established by regulatory agencies. The bisphenol A microextraction employed only 500 μL of the bovine milk sample and 1000 μL of acetonitrile, obtaining excellent recovery values for (89 − 105%) in UHT and pasteurized milk. This proposed methodology had several advantages in determining the bisphenol A, such as quick analysis, miniaturization of the process, decreasing the solvent and sample volume, and following the Green Analytical Chemistry.

Abstract:Using triacontyl bonded silica (C30) as on-line solid-phase extraction (SPE) material and a specially designed on-line analytical system which allowed large sample volume injection, a high speed and robust on-line SPE-HPLC–MS method was established for the analysis of five estrogens and bisphenol A (BPA) in milk samples. The milk sample is pretreated with acetonitrile for protein precipitation and then treated with primary secondary amine for the removal of polar impurities in the matrix. Then the pretreated sample can be automatically loaded by a LC pump. For effective extraction, an offshoot with NH4Ac solution of high-flow rate was employed to dilute the loaded sample by a mixing tee before sample was loaded onto the C30 extraction column. In this way, large volume injection (1mL in this experiment) could be achieved. Some important parameters such as sample loading flow rate, sample dilution ratio and injection volume were optimized. Under the optimized conditions, the recoveries for all analytes range from 71.4 to 97.1% and reproducibility represented as RSDs are less than 15.0% (n=5) with milk samples spiked at 0.6 and 15ng/mL of each analyte. To the authors’ knowledge, it constitutes the first work describing a C30 on-line SPE-LC–MS analytical method for the screen and monitoring of these estrogens and BPA in milk.
Sensitive determination of polybrominated diphenyl ethers (PBDEs) in water and milk by effervescence-enhanced switchable hydrophilic-hydrophobic solvent-based salting-out microextraction and high-performance liquid chromatography with photodiode array detection (HPLC-PDA)

Abstract:Here is reported effervescent reaction-enhanced switchable hydrophilic-hydrophobic solvent-based salting-out microextraction (ERSSM) with high-performance liquid chromatography with photodiode array detection (HPLC-PDA) for the preconcentration, extraction, and trace-level determination of polybrominated diphenyl ethers (PBDEs) in milk and water. The important variables were rigorously optimized to be 150 μL of hexanoic acid as the extraction solvent, 400 μL of Na 2 CO 3 as the alkaline source, 350 μL of H 2 SO 4 solution as the acid source, and NaCl as the salting-out agent. The optimized conditions provided a linear range from 0.2 to 50 μg L −1 with limits of detection and quantitation from 0.026 to 0.085 μg L −1 and 0.087 to 0.28 μg L −1 for six PBDEs. The intra-day and inter-day precision values were from 2.48% to 6.75%, and the fortified recoveries for PBDEs were between 78.06% and 103.07% in water and milk samples. The developed procedure avoids the use of a traditional organic disperser and mechanical mixing allowing the determination of PBDEs outdoors by synchronous extraction method and high efficiency for the analytes. These advantages suggest a high potential for the routine monitoring of PBDEs in liquid food and environmental samples.

Abstract:ABSTRACT Polybrominated diphenyl ethers (PBDEs), which are used as flame retardants, are widely used additives so many different kind of materials that are consumed by public. Current researches reveal a significant increase in the levels of PBDEs in human biological fluids all around the world. According to International Agency for Research on Cancer (IARC), PBDEs are declarated as carcinogenic agents and additionally a considerable risks have been shown in animal studies such as liver damage, alteration of thyroid hormone levels, neurotoxicity and hazardous effects on development of fetus. Due to the simplicity of human exposure routes such as ingestion, inhalation or dermal contact, it gains high importance to quantify PBDEs in biological fluids. The exposure to PBDEs is critic aspecially during pregnancy, fetus development and infancy. Excretion of PBDEs in human milk is the focus area for researchers in recent years. In this study, we reviewed current methodologies for the determination of PBDEs in human milk including sample preparation step, analytical procedure, validation parameters and observed levels of PBDEs in human milk. Because of the high physico-chemical stability of PBDEs, they have long persistence in human body. Besides, they reside in the lipid fractions of tissues because of their hydrophobic characteristics. Human breast milk has high lipid level, so that it is a suitable matrix for the accumulation and determination of PBDEs.

A Simple and Effective Liquid-Liquid-Liquid Microextraction Method with Ultraviolet Spectrophotometric Detection for the Determination of Bisphenol A in Aqueous Matrices and Plastic Leachates

Abstract:This study shows the development and application of an analytical method for the determination of bisphenol A (BPA) in aqueous samples such as tap, river and mineral water and plastic leachates, based on an extensive literature search to understand the gaps for the determination of BPA in aqueous samples. We found that most of the methods in the literature employ some chromatographic strategy, and, to a much lesser extent, non-chromatographic instrumentation. In this scenario, we show an ultraviolet spectrophotometric-based method that can be used for routine analysis. Sample preparation was conducted by means of solvent extraction followed by back-extraction into an alkaline aqueous solution (liquid-liquid-liquid microextraction) and the detection was performed by UV spectrophotometry at 294 nm. Optimization of variables affecting both extraction and back-extraction was conducted, and the optimal extraction conditions were obtained: 85 mL aqueous sample buffered at pH 10, ionic strength adjusted with NaCl to a 2 mol L-1 final concentration, extraction with 6 mL of ethyl ether for 10 min and back-extraction into 0.5 mL of 2 mol L-1 aqueous NaOH. The following quality parameters were obtained: determination coefficient (R2) > 0.999, intra and inter-day repeatability better than 7.8% and 300 enhancement factor. The method was applied in different aqueous samples with excellent recovery and precision results, and no BPA was detected in natural water, even with the excellent limit of detection (3.5 μg L-1) and limit of quantification obtained. Limitations of this method involve analysis of samples with humic acid concentrations higher than 2 mg L-1 or high concentration of phenols and/or phthalate esters.

Abstract:We developed an analytical method based on ultra-high performance liquid chromatography with UV detection, using a stir bar coated with amino/hydroxyl bifunctional microporous organic network (B-MON), for the analysis of parabens in breast milk samples. B-MON demonstrated superior performance with maximal methylparaben adsorption of 112.15 mg/g. Kinetic fitting revealed that outer diffusion was the key limiting step, and the adsorption was chemisorption. The thermodynamic analysis demonstrated that increased methylparaben adsorption was found at higher temperatures in spontaneous processes. The developed approach showed excellent linearity (R 2 ≥ 0.9964) and a low detection limit (0.01 μg/L). Recoveries ranged from 85.8 to 105.5 % and the relative standard deviation was lower than 9.2 %. Based on the daily exposure assessment, these pollutants do not pose unacceptable health hazards to babies. However, the high detection frequencies (41.9%–93.5 %) suggest that breast milk still should be monitored.

Abstract:A HPLC‐UV method was developed for rapidly and simultaneously analyzing novel and typical bisphenols in building materials, including bisphenol S, diphenolic acid, bisphenol F, bisphenol E, bisphenol A, bisphenol B, bisphenol AF, bisphenol AP, bisphenol C, bisphenol FL, bisphenol Z, bisphenol BP, bisphenol M, and bisphenol P. By using a Kromasil 100–5 C18 column, these bisphenols were completely separated in 40 min via gradually increasing the concentration of methanol in the mobile phase from 45 to 80% during the elution process. In particular, this method achieved the synchronous analysis of bisphenol S, diphenolic acid, bisphenol FL, bisphenol BP, and bisphenol M through HPLC, which were difficult to separate, and had to be identified and detected through MS. The limits of detection of the method ranged from 0.002 to 0.040 mg L−1 for these 14 bisphenols, with the precision less than 4.9% (n = 7, c = 0.05 mg L−1). Automated On-Line Liquid Chromatography–photodiode Array–mass Spectrometry Method with Dilution Line for the Determination of Bisphenol A and 4-Octylphenol in Serum

Abstract:A novel on-line liquid chromatography–photodiode array detection–mass spectrometry (LC–DAD–MS) system was established with restricted-access media (RAM) pre-column and dilution line combined with a column-switching valve. The serum samples were injected directly onto pre-column under diluted condition by dilution line. After elution of proteins in the serum, the analytes were backflushed onto an ODS analytical column using a six-port column-switching device. The influence of the composition of the mobile phase, for instance, organic modifer, ionic strength, pH, dilution times and the rotation time of the switching valve have been investigated using bisphenol A (BPA) and 4-octyphenol (4-OP) as analytes. The evaluations for peak responses and sensitivity were conducted by MS, and proteins were removed by RAM-column with DAD monitoring at 280nm. The peak shape was improved by adding a dilution line, especially in the case of large volume injection (LVI), which increased the sensitivity of the analysis. The selective and sensitive quantification of BPA and 4-OP in serum sample could be finished within 25min. The method had linearity in the range 0.1–500ng/mL with a limit of quantification for BPA and 4-OP of 0.1 and 0.5ng/mL, respectively. The recoveries were in the range of 80–101% with less than 9.0% RSDs. This on-line LC–MS method demonstrates potential application to evaluating the exposure and risk of BPA and 4-OP in human.
Simultaneous Quantification of Α-Lactalbumin and Β-Casein in Human Milk Using Ultra-Performance Liquid Chromatography with Tandem Mass Spectrometry Based on Their Signature Peptides and Winged Isotope Internal Standards

Abstract:In recent years, there is an increasing need to measure the concentration of individual proteins in human milk, instead of total human milk proteins. Due to lack of human milk protein standards, there are only few quantification methods established. The objective of the present work was to develop a simple and rapid quantification method for simultaneous determination of α-lactalbumin and β-casein in human milk using signature peptides according to a modified quantitative proteomics strategy. The internal standards containing the signature peptide sequences were synthesized with isotope-labeled amino acids. The purity of synthesized peptides as standards was determined by amino acid analysis method and area normalization method. The contents of α-lactalbumin and β-casein in human milk were measured according to the equimolar relationship between the two proteins and their corresponding signature peptides. The method validation results showed a satisfied linearity (R2>0.99) and recoveries (97.2–102.5% for α-lactalbumin and 99.5–100.3% for β-casein). The limit of quantification for α-lactalbumin and β-casein was 8.0mg/100g and 1.2mg/100g, respectively. CVs for α-lactalbumin and β-casein in human milk were 5.2% and 3.0%. The contents of α-lactalbumin and β-casein in 147 human milk samples were successfully determined by the established method and their contents were 205.5–578.2mg/100g and 116.4–467.4mg/100g at different lactation stages. The developed method allows simultaneously determination of α-lactalbumin and β-casein in human milk. The quantitative strategy based on signature peptide should be applicable to other endogenous proteins in breast milk and other body fluids.

Abstract:Commonly, analytical methods measuring brominated flame retardants (BFRs) of different chemical polarities in human serum are labor consuming and tedious. Our study used acidified diatomaceous earth as solid-phase extraction (SPE) adsorbent and defatting material to simultaneously determine the most abundant BFRs and their metabolites with different polarities in human serum samples. The analytes include three types of commercial BFRs, tetrabromobisphenol A (TBBPA), hexabromocyclododecane (HBCD) isomers, and polybrominated biphenyl ethers (PBDEs), and dominant hydroxylated BDE (OH-PBDE) and methoxylated BDE (MeO-PBDE) metabolites of PBDEs. The sample eluents were sequentially analyzed for PBDEs and MeO-BDEs on online gel permeation chromatography/gas chromatography-electron capture-negative ionization mass spectrometry (online GPC GC-ECNI-MS) and for TBBPA, HBCD, and OH-BDEs on liquid chromatography-tandem mass spectrometry (LC-MS/MS). Method recoveries were 67–134% with a relative standard deviation (RSD) of less than 20%. Method detection limits (MDLs) were 0.30–4.20 pg/mL fresh weight (f.w.) for all analytes, except for BDE-209 of 16 pg/mL f.w. The methodology was also applied in a pilot study, which analyzed ten real samples from healthy donors in China, and the majority of target analytes were detected with a detection rate of more than 80%. To our knowledge, it is the first time for us in effectively determining BFRs of most types in one aliquot of human serum samples. This new analytical method is more specific, sensitive, accurate, and time saving for routine biomonitoring of these BFRs and for integrated assessment of health risk of BFR exposure.
A miniaturized stir bar sorptive dispersive microextraction method for the determination of bisphenols in follicular fluid using a magnetic covalent organic framework

Abstract:Background Bisphenols, particularly bisphenol A (BPA), are the primary monomers used as additives in the manufacturing of many consumer products. The exposure to these compounds is related to endocrine-disrupting and reproductive effects, among others. For this reason, the development of analytical methods for their determination in biological matrixes is needed to monitor the population exposure to these compounds. Their quantification at ovarian level ( i.e ., follicular fluid) is interesting for the assessment of the bisphenol content to draw conclusions about infertility problems. However, the background does not meet all requirements by focusing mainly on BPA. Results In this work, a miniaturized stir bar sorptive dispersive microextraction (mSBSDME) approach has been developed for the determination of BPA and eight analogues in follicular fluid. In the proposed method, the sample is previously cleaned-up using a zirconia-based solid-phase extraction cartridge, removing proteins and phospholipids, and then subjected to the mSBSDME for the preconcentration of the analytes. For this purpose, a magnetic covalent organic framework was used as sorbent. A Plackett-Burman design was applied to select the significant variables affecting the mSBSDME. Afterwards, the only significant variable ( i.e., sorbent amount) was optimized. Under the optimized conditions, the proposed method was properly validated, and satisfactory analytical parameters in terms of linearity (up to 50 ng mL −1 ), enrichment factors (8.5–14.3), limits of detection in the low ng mL −1 range, and precision (relative standard deviations below 11.5 %) were obtained. Finally, the method was successfully applied to five samples, detecting BPA and other two analogues. Significance This method expands the potential applicability of the mSBSDME to other low-availability complex matrixes, which would otherwise be difficult to analyze. Moreover, it offers a valuable tool for monitoring the female population's exposure to bisphenols with the final aim of evaluating if infertility problems of women might be associated to the exposure to these highly endocrine disrupting compounds.

Abstract:A solid-phase extraction (SPE) coupled with dispersive liquid–liquid microextraction based on the solidification of floating organic drop (DLLME-SFO) method has been developed for extraction and determination of organic bisphenol A (BPA) in different real samples. Bisphenol A was determined by high-performance liquid chromatography-fluorescence detector (HPLC-FL). Variables affecting the performance of both steps were thoroughly investigated. The analytical characteristics of the method were determined. The calibration graph is linear in the range of 0.005–10 ng g−1 with detection limit of 0.002 ng g−1. The relative standard deviation (RSD) for of BPA in different samples are in the range of 6.8–9.6 % (n = 5). The obtained results show that SPE-DLLME-SFO combined with HPLC-FL is a fast and simple method for the determination of BPA in food and environmental samples.

Abstract:Abstract The study focused on the application of GC in the quantitative analysis of bisphenols and their analogues (12 analytes), and the improvement of solid-phase extraction for the whole water analysis of complex water samples. The role of silylation in the qualitative and quantitative analysis of bisphenols was investigated. Partial degradation occurred for selected targets during hot injection with the presence of a silylation agent. A PSA (primary and secondary amines) sorbent placed on the top of the solid-phase extraction (SPE) column sorbent was found to be a matrix component trap, mostly for humic acids. The whole water analysis was performed by washing the filters with methanol and recycling the extract to the sample. The validation of SPE-GC/MS(SIM) gave limits of detection of 1–50 ng/L for ten target bisphenols with a method recovery of between 87 and 133%. The application of the method was tested by the analysis of wastewater sampled from three wastewater treatment plants located in Poland, and municipal surface waters. The only analytes found were BPA and BPS, within the range of 16–1465 ng/L and < MDL-1249 ng/L in wastewater, and 170–3113 ng/L and < MDL-1584 ng/L in surface water, respectively.
