Abstract:A biotechnological technique that involves creating novel pesticides is the nanotechnology of natural compounds, like essential oils. With a lower threat to the environment and public health, these rational approaches provide a more sustainable choice. The present work sought to create novel nanoemulsions of ginger essential oil (G-EO) (Zingiber officinale) and basil essential oil (B-EO) (Ocimum basilicum) to control Odontotermes formosanus (O. formosanus). Stabilization of the ginger nano-emulsion (G-NE) and basil nano-emulsion (B-NE) was assessed by dynamic light scattering (DLS), zeta potential (ZP), and PH measurement, and the characterization was verified by transmission electron microscopy, UV-Vis, energy dispersive spectroscopy, and scanning electron microscopy. Furthermore, a chemical investigation utilizing gas chromatography and mass spectrometers revealed the main components of B-NE, B-EO, G-NE, and G-EO. In addition to assessing the essential oil's termiticidal abilities against O. formosanus in bulk and nanoemulsion formulations. When G-NE and G-EO were both at 17.5 mg/mL, the mortality rates were 100% and 42.87%, respectively. B-EO appeared to have a 45.33% death rate, while B-NE displayed a 100% mortality rate at a dose of 0.273 mg/mL. According to the termiticide appraisal, nanoemulsions have a greater effect on suppressing O. formosanus than essential oils. Additional research was conducted on the impact of G-NE and B-NE on the level of total protein, detoxification enzymes (carboxylesterase, acid, alkaline phosphatase, and acetylcholinesterase), and antioxidant enzyme (peroxidase). It was found that there was a significant change in biochemical activities when compared to the control. Additionally, changes in morphology and histology were seen after exposure to G-NE and B-NE. Moreover, when contrasted to the untreated group, the current study's analysis of the fungal community after exposure to B-NE revealed a high degree of diversity. This work stressed the potential of G-NE and B-NE as natural plant-derived termiticides and suggested a novel technique for sustainability and environmental friendliness in controlling O. formosanus.

Abstract:Aims: Nanoemulsions based on plant essential oils have shown promise as alternatives against fungal pathogens by increasing the solubility and bioavailability of the active compounds of essential oils, which can improve their efficacy and safety. In the present study, we aimed to prepare and characterize nanoemulsions of Lippia origanoides essential oil, and analyze their antifungal activity against C. albicans in planktonic and biofilm form. Additionally, we sought to verify their cytotoxicity. Methods and results: Alginate nanoemulsions were prepared with different concentrations of essential oil, sunflower oil, and surfactant to investigate ideal formulations regarding stability and antifungal efficiency. The results showed the nanoemulsions remained stable for longer than 60 days, with acidic pH, particle sizes ranging from 180.17 ± 6.86 nm to 497.85 ± 253.50 nm, zeta potential from -60.47 ± 2.25 to -43.63 ± 12, and polydispersity index from 0.004 to 0.622. The photomicrographs revealed that the addition of sunflower oil influenced the formation of the particles, forming nanoemulsions. The antifungal results of the essential oil and nanoemulsions showed that the MIC ranged from 0.078 to 0.312 mg ml-1. The nanoemulsions were more effective than the free essential oil in eradicating the biofilm, eliminating up to 89.7% of its mass. With regard to cytotoxicity, differences were found between the tests with VERO cells and red blood cells, and the nanoemulsions were less toxic to red blood cells than the free essential oil. Conclusions: These results show that nanoemulsions have antifungal potential against strains of C. albicans in planktonic and biofilm forms.

Abstract:Cymbopogon martinii essential oil (Palmarosa essential oil, PRO) has a wide range of applications in food, medicine, and cosmetics. Because of its hydrophobicity, instability, and volatility, the best option to safeguard this oil is to encapsulate it using nanoemulsion technology. In this work, we synthesize O/W (oil/water) nanoemulsions from C. martini essential oil and analyze their biological activity. The study uses co-surfactants like ethanol and surfactants like tween 80 and sodium dodecyl sulfate to create O/W nanoemulsions. The nanoemulsions were investigated using UV, fluorescence, and FT-IR fingerprinting techniques in addition to parameters such as pH and conductivity. Additionally, the study explored the biological effects of the nanoemulsions, including their antibacterial, anti-inflammatory, anticancer, and anti-diabetic capabilities. The pharmacokinetics of drug release was also examined. By using GC-FID analysis, it has been determined that geraniol is the primary component of PRO. The concentrations of total phenolic, total flavonoid, and total condensed tannin were found to be 2.91 mg/g GAE, 9.29 mg/g RE, and 3.78 mg/g VAE, respectively. Palmarosa essential oil nanoemulsions (PNE) were discovered to have 93.24% encapsulation efficiency. The nanoemulsions remained stable and spherical even after 50 days of storage. PNE was synthesized with a mean diameter of 460.55 ± 98.65 nm, a polydispersity index (PDI) value of 0.45 ± 0.04, and a zeta potential measurement of 0.1711 ± 0.05 mV. According to a drug kinetic analysis, PNE displayed a first-order model with an R 2 value of 0.930. PNE revealed a dose-dependent 92% inhibition of α-amylase. Additionally, PNE demonstrated potent anti-bacterial activity against the investigated pathogens that varied from 38% to 94%. In the same way, PNE demonstrated potent anti-fungal efficacy against the studied fungal infections, with a range of 50–95%. Anticancer research showed that the PNE-containing cells had a percent viability of 21.29% after 24 hours and 14.92% after 48 hours, respectively. The article explores the possible applications of nanoemulsions made from palmarosa essential oil in the cosmetic, food, and medicinal sectors.

Abstract:Lately, the bacterial multidrug resistance has been a reason to public health concerning around world. The development of new pharmacology therapies against infections caused by multidrug-resistant bacteria is urgent. In this work, we developed 10 NLC formulations composed of essential oils (EO), vegetable butter and surfactant. The formulations were evaluated for long-term and thermal cycling stability studies in terms of (particle size, polydispersion index and Zeta potential). In vitro antimicrobial assays were performed using disk diffusion test and by the determination of the minimum inhibitory concentration (MIC) performed with fresh and a year-old NLC. The most promising system and its excipients were structurally characterized through experimental methodologies (FTIR-ATR, DSC and FE-SEM). Finally, this same formulation was studied through nanotoxicity assays on the chicken embryo model, analyzing different parameters, as viability and weight changes of embryos and annexes. All the developed formulations presented long-term physicochemical and thermal stability. The formulation based on cinnamon EO presented in vitro activity against strains of Acinetobacter baumannii , Klebsiella pneumoniae and Pseudomonas aeruginosa isolated from humans and in vivo biocompatibility. Considering these promising results, such system is able to be further tested on in vivo efficacy assays.

[Neurophysiological characteristics of infants and young children with auditory neuropathy].

Abstract:OBJECTIVE To analyze the neurophysiological characteristics of infants and young children with auditory neuropathy (AN) and explore their clinical significance. METHODS Audiological measurements (acoustic immittance, EOAEs, ABR, CM, MLR and ERP) and peripheral neurological tests were conducted and evaluated in 13 infants and young children with AN. 6 AN patients received CT scan and/or MRI examination. RESULTS All patients had type "A" tympanogram and normal CM. Normal EOAEs were elicited in 12 patients. 8 cases had normal MLR recording and 6 cases had normal ERP (P(300) and MMN). Peripheral neurological tests and CT and/or MRI showed normal results. CONCLUSION The diagnosis of AN in infants and young children should focus on analyzing their neurophysiological characteristics. Combined use of EOAEs, ABR and CM was recommended for hearing screening on newborns with high risk factors.

Abstract:Brazil is renowned for its extensive plant biodiversity, with emphasis on Cymbopogon, C. citratus and C. nardus, with broad antimicrobial potential. Candidemias caused by Candida albicans are highly prevalent in immunosuppressed individuals and are associated with infections by biofilms on medical devices. The aim of this study was to evaluate the antimicrobial potential of essential oils C. citratus and C. nardus against C. albicans in planktonic and biofilm forms. Essential oils were obtained by hydrodistillation and chemical composition evaluated by GC-FID and GC-MS. The minimum inhibitory concentration was determined by the broth microdilution method and the synergy effect of essential oils and amphotericin B were evaluated by the checkerboard test. Biofilm activity was determined by the XTT assay. Cytotoxicity assays performed with VERO cells and molecular docking were performed to predict the effect of oil interaction on the SAP-5 enzyme site. The results showed activity of essential oils against planktonic cells and biofilm of C. albicans. Furthermore, the oils had a synergistic effect, and low cytotoxicity. Molecular docking showed interaction between Cadinene, Caryophyllen oxide, Germacrene D with SAP-5. The results indicate that Cymbopogon spp. studied are anti-Candida, with potential for further application in therapy against infections caused by C. albicans.

Abstract:Nanoemulsions are dispersions of oil-in-water (O/W) and water-in-oil (W/O) immiscible liquids. Thus, our main goal was to formulate a nanoemulsion with low surfactant concentrations and outstanding stability using Copaiba balsam oil (Copaifera sp.). The high-energy cavitation homogenization with low Tween 80 levels was employed. Then, electrophoretic and physical mobility properties were assessed, in addition to a one- and two-year physicochemical characterization studies assessment. Copaiba balsam oil and nanoemulsions obtained caryophyllene as a major constituent. The nanoemulsions stored at 4 ± 2 °C exhibited better physical stability. Two years after formulation, the nanoemulsion showed a reduction in the particle size. The size underwent changes in gastric, intestinal, and blood pH, and the PdI was not changed. In FTIR, characteristic bands of sesquiterpenes and overlapping bands were detected. When subjected to freezing and heating cycles, nanoemulsions did not show macroscopic changes in higher concentrations. Nanoemulsions subjected to centrifuge force by 1000 rpm do not show macroscopic instability and phase inversion or destabilization characteristics when diluted. Therefore, the nanoemulsion showed stability for long-term storage. The nematode Caenorhabditis elegans was used to assess the potential toxicity of nanoemulsions. The nanoemulsion did not cause toxicity in the animal model, except in the highest concentration tested, which decreased the defecation cycle interval and body length. The toxicity and stability outcomes reinforce the nanoemulsions' potential for future studies to explore pharmacological mechanisms in superior experimental designs.

Abstract:This research aims to optimize the parameters for the production of a stable nanoemulsion containing a mixture of essential oils (EOs) with a surfactant blend (Tween and Span 80) and assess its antimicrobial potency against spoilage bacteria and foodborne pathogens. The impact of a surfactant blend's hydrophilic-lipophilic balance (HLB) as well as surfactant:oil ratio was investigated based on the mean particle size (z-average) and polydispersity index (PDI). Results showed HLB value of 12 and a ratio of surfactant to oil at 0.75:1 exhibited an emulsion with the smallest z-average and PDI. Following this, a response surface methodology (RSM) employing central composite design (CCD) was utilized to formulate an optimal nanoemulsion using the microfluidization (MF) technique. The RSM revealed that the microfluidizer pressure of 15,000 psi and 5 cycles produces a z-average of 38.11 nm, PDI of 0.27, ζ-potential of 37.8 mV, and EE of 83.8%. The comparison between the experimental and anticipated results of the optimized nanoemulsion revealed no significant differences (p > 0.05). The optimal nanoemulsion displayed excellent stability during 30 days of storage at both 4 and 30 °C compared to 5 days for coarse emulsion. The nanoemulsion exhibited potent antimicrobial activity, showing minimum inhibitory concentration (MIC) values in the range of 0.019 to 0.156 μL/mL, surpassing the antimicrobial efficacy of the coarse emulsion, which had MIC values between 0.039 to 0.312 μL/mL. Thus, this study emphasized the efficacy of RSM design in developing an optimal nanoemulsion with enhanced antimicrobial properties using EOs, making it a promising natural food preservative.

Abstract:Application of synthetic fungicides in agricultural commodities has been restricted due to development of fungicide resistance fungi and deleterious impact on environment and health of farm animals and humans. Hence, there is an urge for development of mycobiocides, and the present study was undertaken to determine the antifungal activity of Cymbopogon martinii essential oil (CMEO) on post-harvest pathogen Fusarium graminearum. The CMEO was extracted by hydrodistillation and GC-MS chemical profile revealed the presence of 46 compounds and abundant was geraniol (19.06%). The minimum inhibitory concentration and minimum fungicidal concentration of CMEO were determined as 421.7 ± 27.14 and 618.3 ± 79.35 ppm, respectively. The scanning electron microscopic observation of CMEO exposed macroconidia was exhibited a detrimental morphology with vesicles, craters, protuberance, and rough surfaces related to control fungi. The CMEO induced the death of fungi through elevating intracellular reactive oxygen species and lipid peroxidation, and depleting ergosterol content. Regrettably, essential oils are highly volatile and become unstable and lose their biological features on exposure to light, heat, pH, moisture, and oxygen. To overcome these issues, chitosan encapsulated CMEO nanoparticles (Ce-CMEO-NPs) were prepared. The synthesized Ce-CMEO-NPs have spherical morphology with Zeta potential of 39.3-37.2 mV and their corresponding size was found in range of 455-480 nm. The Fourier transform infrared analysis confirmed that bio-active constituents of CMEO were well stabilized due to chitosan conjugation and successfully formed Ce-CMEO-NPs. The in vitro release assay observed that the release of CMEO is stabilized due to the complex formation with chitosan and thereby, increases the lifetime antifungal activity of CMEO by gradual release of antifungal constituents of Ce-CMEO-NPs. In conclusion, antifungal and antimycotoxin activities of CMEO and Ce-CMEO-NPs against F. graminearum were assessed in maize grains under laboratory conditions over a storage period of 28 days. Interestingly, Ce-CMEO-NPs were presented efficient and enhanced antifungal and antimycotoxin activities related to CMEO, and it could be due to perseverance of antifungal activity by controlled release of antifungal constituents from Ce-CMEO-NPs. The study concluded that Ce-CMEO-NPs could be highly appropriate as mycobiocides in safeguarding the agricultural commodities during storage period in agricultural and food industries.

Abstract:In the global context of excessive and indiscriminate application of synthetic agrochemicals, this work combined natural compounds and nanotechnology to develop eco-friendly and sustainable agricultural systems with antifungal action. Cannabis (C) is composed of a large number of phytocannabinoids and terpenes and exhibit important biological properties. Due to the chemical instability of C to avoid their decomposition from environmental exposure, the use of efficient carriers become a promising alternative. Solid Lipid Nanoparticles (SLN) containing C extract were synthesized by homogenization with the ultrasonication method into a lipidic matrix coated with chitosan. The SLN formulations were characterized by dynamic light scattering and field emission scanning electron microscopy , showing a spherical morphology with particles in the range of 150 – 240 nm and low polydispersity indexes. Entrapment efficiency of C was higher than 20% and effectively in vitro released in 24 h. Particularly, the SLN coated with chitosan and loaded with C exhibited an enhanced fungicidal activity against the phytopathogen fungus Fusarium solani sp. Eumartii in a lower concentration than the neat C extract. The results showed a synergistic effect between the positive charges of chitosan and the effect of phytocannabinoids and terpenes of C. These nano-formulations are a promising solution to transform the current agriculture system into a more efficient, eco-friendly and sustainable one.

Abstract:Terbinafine hydrochloride is a synthetic allylamine whose mechanism of action consists of inhibiting the enzyme squalene epoxidase that participates in the first stage of ergosterol synthesis, interfering with fungal membrane function. Ozonated oils are used for topical application of ozone, producing reactive oxygen species that cause cellular damage in microorganisms, therefore being an alternative treatment for acute and chronic skin infections. This study aimed to develop and characterize Eudragit ® RS100 nanocapsules, obtained by interfacial deposition of preformed polymer method, containing 0.5% terbinafine hydrochloride and 5% ozonated sunflower seed oil as a potential treatment against dermatophytes. The polymeric nanocapsules were characterized regarding particle size, zeta potential, pH, drug content, encapsulation efficiency, and stability. The in vitro drug release, in vitro skin permeation, and in vitro antifungal activity were also evaluated. The particle size was around 150 nm with a narrow size distribution, the zeta potential was around + 6 mV, and the pH was 2.2. The drug content was close to 95% with an encapsulation efficiency of 53%. The nanocapsules were capable to control the drug release and the skin permeation. The in vitro susceptibility test showed greater antifungal activity for the developed nanocapsules, against all dermatophyte strains tested, compared to the drug solution. Therefore, the polymeric nanocapsules suspension containing terbinafine hydrochloride and ozonated oil can be considered a potential high-efficacy candidate for the treatment of dermatophytosis, with a possible reduction in the drug dose and frequency of applications. Abstract:The antifungal and antimycotoxigenic activities of the essential oils (EO) from Cuminum cyminum and Laurus nobilis, and their respective principal compounds, cuminaldehyde and 1,8-cineole, were evaluated against fungi of the genus Aspergillus: A. carbonarius, A. niger, A. ochraceus, and A. westerdijkiae. The antifungal activity was determined by the contact method and the mycelial growth of the fungi was evaluated. Scanning electron microscopic (SEM) images were obtained to suggest modes of action of the compounds analysed. The antimycotoxigenic activity was determined by high-performance liquid chromatograph. Aspergillus carbonarius was completely inhibited by cumin EO (500 µl l-1), by laurel EO and by cuminaldehyde (5000 µl l-1). The cumin EO (500 µl l-1) completely inhibited the growth of A. niger. All the samples inhibited the mycelial growth of A. ochraceus, especially cumin EO and cuminaldehyde (250 µl l-1). Aspergillus westerdijkiae was completely inhibited by cumin EO and cuminaldehyde (1000 µl l-1), by laurel EO and 1,8-cineole (10 000 µl l-1). A decrease in the production of ochratoxin A (OTA) was observed post-treatment, except in A. ochraceus, only inhibited by laurel EO. SEM images showed morphological changes in fungal structures and spore inhibition post-treatment. The results confirmed the antifungal and antimycotoxigenic effect of EO and their principal constituents on fungi evaluated.

Abstract:Nanoemulsion is one the most attractive drug delivery for researchers to treat fungal diseases and getting interested in increasing the solubility of low soluble drugs. The objective of this research was to develop a topical nanoemulsion formulation of Posaconazole drug with the intention to enhance its aqueous solubility and local action. By employing High Pressure Homogenization (HPH) method, oil in water nanoemulsion was formulated with 0.2% cinnamon oil as lipid phase, 0.4% tween-80 and poloxamer-188 as surfactants, 0.2% transcutol as a co-surfactant. The physical characteristics of formulations were found to be stable after thermodynamic stability testing. In-vitro diffusion study for optimized nanoemulsion was performed using a dialysis bag membrane and cumulative % drug release was determined. Viscosity and percent drug content was observed to be 0.0593 cps and 90.21 ± 0.23% respectively. The optimized nanoemulsion formulation (F8) was evaluated to be transparent and thermodynamically stable, with -9.46 zeta potential, 78.79 nm particle size, 0.315 polydispersity index. The optimized Nanoemulsion was stable for 3 month in three different temperature conditions. The result from the release study was indicative of improved solubility of Posaconazole, which may serve to boost up the action of the drug for the treatment of fungal diseases. Abstract:In recent years, the study of essential oils as antifungal alternatives and their encapsulation to increase their properties for greater effects has been tested. In this work, nanoparticles of chitosan– Schinus molle L. essential oil (CS-PEO-Np) with a size of 260 ± 31.1 nm were obtained by ionic gelation and evaluated in some growth phases of Aspergillus flavus , a toxigenic fungus. At a concentration of 250 μg/mL of CS-PEO-Np, the A. flavus mycelial growth was inhibited at 97.1% with respect to control, at 96 h of incubation; the germination and viability of spores were inhibited at 74.8 and 40%, respectively, after exposure to 500 μg/mL of these nanomaterials, at 12 h of incubation. The fluorescence images of stained spores with DAPI showed the affectations caused by nanoparticles in the cell membrane, vacuoles and vacuolar content, cell wall, and nucleic acids. For both nanoparticles, CS-Np and CS-PEO-Np, no mutagenic effect was observed in Salmonella Typhimurium; also, the phytotoxic assay showed low-to-moderate toxicity toward seeds, which was dependent on the nanoparticle's concentration. The acute toxicity of CS-PEO-Np to A. salina nauplii was considered low in comparison to CS-Np (control), which indicates that the incorporation of Schinus molle essential oil into nanoparticles of chitosan is a strategy to reduce the toxicity commonly associated with nanostructured materials. The nanoparticulated systems of CS-PEO-Np represent an effective and non-toxic alternative for the control of toxigenic fungi such as A. flavus by delaying the initial growth stage.

Abstract:The Nano-formulation of citronella essential oil ( Cymbopogon nardus (L.) and their mixtures of three adjuvants (Provecta®, Top film®, and PEG600-dioleate) were studied to enhance physico-chemical properties of the Nano-formulation and improve the insecticidal effect against Spodoptera littoralis (Boisd). Characterisation of physicochemical properties of Nano-formulation was studied by calculating droplet size, polydispersity index (PDI), and Zeta-potential parameters. The results showed that the Solid Lipid Nanoparticles (SLNs) mixtures of three adjuvant mixtures had more toxic activity and stability than the Nano-formulation alone. Before storage the acidity, alkalinity (pH), and viscosity exhibited an acidic pH value in the range (4.11–5.34), whereas after three months of storage was recorded high pH, a shift in the pH on storage can indicate the instability of active substances or product under semi field-laboratory conditions. Nano-formulation of the citronella oil mixed with the three adjuvants increased the mortality percentage of S. littoralis larvae.

Abstract:In this study, we developed oregano essential oil-mesoporous nano-silica (OEO-MSNPs), a new and safe eco-friendly antifungal system. MSNPs showed ordered mesoporous structures by TEM, which can provide space for loading OEO. The FTIR characterization of MSNPs and OEO-MSNPs showed that OEO was successfully embedded into the MSNPs. The loading rate of OEO-MSNPs to OEO reached about 60%, and OEO-MSNPs had better antifungal activity against the mold ( Curvularia lunata MF380802.1) found in postharvest decayed Agaricus bisporus . The antifungal effects of OEO-MSNPs against Curvularia lunata MF380802.1 ( C. lunata ) were investigated. Then, our results showed that the antifungal effect of OEO-MSNPs had a minimum inhibitory concentration (MIC) and a minimum fungicidal concentration (MFC) at 0.20 mg/mL and 0.40 mg/mL, respectively. Furthermore, the integrity of cell membranes and nuclear membranes of C. lunata was destroyed during the interaction with OEO-MSNPs, resulting in the exudation of nucleic acids, proteins, and other substances in vitro. This work provides a theoretical basis for the development of effective treatments to address the infection of C. lunata during the storage and preservation of Agaricus bisporus . This newly developed method for encapsulating EOs in MSNPs has potential application in the design of effective antimicrobial materials.

Abstract:Sustainable natural product-based microstructured systems and biopolymers are strong candidates for use in crop protection. Lignin and gelatin microcapsules loaded with orange essential oil were developed with spray-drying in order to enhance its potential. We evaluated the microparticle controlled release mechanisms, biological effects, structural and thermal properties. A quantitative method using headspace-gas chromatography was developed and evaluated for the controlled release of the essential oils. Controlled release studies showed a linear relationship between the biopolymer and essential oil concentrations when retaining volatile compounds. Thermal analyses demonstrated increases in essential oil stability when microencapsulated, especially for lignin as a biopolymer. These results showed that biopolymer type was the main factor influencing quality analysis parameters. Finally, microcapsules loaded with orange essential oil were applied in a bioassay, and showed gains in toxicity against Spodoptera frugiperda compared to non-encapsulated oil.

Abstract:Essential oils (EOs) have antimicrobial properties, but their low solubility in water and strong flavor pose challenges for direct incorporation into food, as they can negatively impact organoleptic properties. To overcome these issues, strategies such as oil-in-water (O/W) nanoemulsions have been developed to improve EO dispersion and protection while enhancing antimicrobial efficacy. The objective of this study was to create sodium alginate-pink pepper essential oil (PPEO) nanoemulsions using microfluidization. Various formulations were assessed for physicochemical, physical, and antimicrobial properties to evaluate their potential in food applications. The microfluidized emulsions and nanoemulsions had droplet sizes ranging from 160 to 443 nm, polydispersity index (PdI) ranging from 0.273 to 0.638, and zeta potential (ζ) ranging from -45.2 to 66.3 mV. The nanoemulsions exhibited Newtonian behavior and remarkable stability after 20 days of storage. Antimicrobial testing revealed effectiveness against Staphylococcus aureus and Listeria monocytogenes, with minimum inhibitory concentrations (MIC) of 200 µg/mL for both microorganisms and minimum bactericidal concentrations (MBC) of 800 µg/mL and 400 µg/mL, respectively, proving that encapsulation of PPEO in nanoemulsions significantly increased its antibacterial activity. These results present the possibility of using PPEO nanoemulsions as a more effective natural alternative to synthetic preservatives in food systems.
