Abstract:A method was developed for the extraction and detection of the polyphosphates in freezing seafood.Chloroform is used to deposit the protein in freezing seafood.The polyphosphates((PO3)nn-、 P2O74-、 P3O105-) was determined by ion chromatography separated by gradient elution of KOH on an Ion Pac AS11 column and detected by suppressed conductivity detector.The method excluded the interference of protein effectively,and the recovery is satisfactory.The method is sensitive,selective and effective,and can be easily performed.

Abstract:To determine whether knowledge was improved as a result of obtaining informed consent from parents for newborn screening of their infants for phenylketonuria (PKU) and other hereditary metabolic disorders, new mothers in seven Maryland hospitals were interviewed either before receiving a standard disclosure (n = 210) or after giving consent (n = 418). The mean knowledge score of the women interviewed after giving consent was significantly higher (P less than .001). Receiving the disclosure was a more powerful predictor of knowledge score, accounting for 40% of the variance, than demographic factors, which accounted for 9%. Women whose consent was obtained just prior to discharge tended to have lower knowledge scores than women whose consent was obtained earlier (P = .03). Women with higher knowledge scores were somewhat less likely to favor consent than women with lower scores. Although consent may not be appropriate for some low-risk procedures, informing parents can be easily and inexpensively accomplished.

Abstract:Due to the rapid decomposition of polyphosphates in aquatic products, it is hard to detect the amount of polyphosphates in aquatic products. In this method, boiling water was used to destroy the substances that induce the decomposition of polyphosphates. In order to remove the impurity, trichloromethane was used as the precipitator. The KOH solution was used as eluent, Ionpac AG11-HC guard column, Ionpac AS11-HC separator column and suppressed conductivity detector were used for the analysis of pyrophosphate, tripolyphosphate and trimetaphosphate. This method is simple and the detection limit is 10.0 mg/kg. The recoveries are 70%-110% for the spiked sample at the concentration level between 10-1000 mg/kg with the RSD less than 12%.
Simultaneous Determination of Six ATP-related Compounds in Aquatic Product Using IP-RPLC

Abstract:In this study,the IP-RPLC method for the determination of six ATP-related compounds(ATP,ADP,AMP,IMP,HxR,Hx)in aquatic product was established.The optimal conditions such as pH,flow rate and concentration of mobile phase were confirmed.The accuracy,precision and limit of quantitation were surveyed and evaluated.The method is based on an extraction of nucleotides from aquatic product with a solution of 10% perchloric acid followed by neutralization by the addition of KOH just prior to HPLC analysis.Chromatographic separations were performed using a Thermo ODS-2 Hypersil(250 mm×4.6 mm,5 μm)column,the column temperature was 25 ℃.The mobile phase consisted of 90% 0.05 mol/L sodium phosphate buffer,10% methanol and 9.5 mmol/L tetrabutylammonium hydroxide,and was adjusted to pH 6.5 with phosphoric acid,the mobile phase was delivered at a flow rate of 0.9 mL/min with equivalent elution,and the detection wave length was 254 nm.The six ATP-related compounds were separated in 13 min.The standard curves of six ATP-related compounds were linear within a range of 2-400 μg/mL,and the correlation coefficient deviations range from 0.999 4 to 1.000 0.The limits of detection(S/N=3)were between 0.1 μg/mL and 0.4 μg/mL.The average recovery of spiked sample deviations range from 82.2% to 110.0%,and the relative standard deviations(RSD)were between 4.2% and 9.5%.In conclusion,the method proved to be simple,rapid,reliableand with high accuracy and precision.The method has practical value for detecting six ATP-related compounds in aquatic product.
Determination of phthalic acid esters and di(2-ethylhexyl) adipate in fish and squid using the ammonium formate version of the QuEChERS method combined with gas chromatography mass spectrometry

Abstract:In the present study, the ammonium formate version of the QuEChERS method, considered highly advantageous in relation to instrument maintenance and other issues, was applied for the first time to extract a group of twelve phthalic acid esters (PAEs, i.e. dipropyl phthalate, DPP; diisobutyl phthalate, DIBP; dibutyl phthalate, DBP; diisopentyl phthalate, DIPP; di-n-pentyl phthalate, DNPP; dihexyl phthalate, DHP; butyl benzyl phthalate, BBP; dicyclohexyl phthalate, DCHP; di(2-ethylhexyl) phthalate, DEHP; di-n-octyl phthalate, DNOP; diisononyl phthalate, DINP; and diisodecyl phthalate, DIDP) and one adipate (di(2-ethylhexyl) adipate, DEHA) from two species of fish (Scomber colias and Katsuwonus pelamis) and one of squid (Loligo gahi). The method was validated in terms of linearity, trueness and matrix effects. Determination coefficients (R2) for matrix-matched calibration curves were higher than 0.99 in all cases, being the lowest calibration levels in the range 0.5-10 ng/g. Mean recovery values were between 70 and 117% with relative standard deviation values ≤20%. Matrix effects were soft (between -20 and +20%) for most analytes and matrices, except in squid samples, which was mostly medium with a moderate ion suppression. The analysis of 10 samples of each type showed the presence of DIBP, DBP and DEHP at concentrations up to 44.2 ± 2.1 ng/g of wet weight in some of the samples and species, still not representing concerning values when considering the daily intake of such species of seafood in the human diet (tolerable daily intake -TDI- values were not exceeded). Results demonstrated that the ammonium formate version of the QuEChERS method can be applied with success for the extraction and determination of the selected PAEs and DEHA in fish and squid samples.

Abstract:Control flow graphs are a well-known representation of the sequential control flow structure of programs with a multitude of applications. Not only single functions but also sets of functions or complete programs can be modeled by control flow graphs. In this case the size of the graphs can grow considerably and thus makes it difficult for software engineers to analyze the control flow. Graph reductions are helpful in this situation. In this paper we define reductions to subsets of nodes. Since executions of programs are represented by paths through the control flow graphs, paths should be preserved. Furthermore, the composition of reductions makes a stepwise analysis approach possible.

Abstract:RATIONALE Mass spectrometric (MS) analysis of low molecular weight polar metabolites can be challenging because of poor chromatographic resolution of isomers and insufficient ionization efficiency. These metabolites include intermediates in key metabolic pathways, such as glycolysis, the pentose phosphate pathway, and the Krebs cycle. Therefore, sensitive, specific, and comprehensive quantitative analysis of these metabolites in biological fluids or cell culture models can provide insight into multiple disease states where perturbed metabolism plays a role. METHODS An ion-pairing reversed-phase ultra-high-performance liquid chromatography (IP-RP-UHPLC)/MS approach to separate and analyze biochemically relevant phosphate- and carboxylic acid-containing metabolites was developed. Diisopropylethylamine (DIPEA) was used as an IP reagent in combination with reversed-phase liquid chromatography (RP-LC) and a triple quadrupole mass spectrometer using selected reaction monitoring (SRM) and negative electrospray ionization (NESI). An additional reagent, hexafluoroisopropanol (HFIP), which has been previously used to improve sensitivity of nucleotide analysis by UHPLC/MS, was used to enhance sensitivity. RESULTS HFIP versus acetic acid, when added with the IP base, increased the sensitivity of IP-RP-UHPLC/NESI-MS up to 10-fold for certain analytes including fructose-1,6-bisphosphate, phosphoenolpyruvate, and 6-phosphogluconate. It also improved the retention of the metabolites on a C18 reversed-phase column, and allowed the chromatographic separation of important isomeric metabolites. This methodology was amenable to quantification of key metabolites in cell culture experiments. The applicability of the method was demonstrated by monitoring the metabolic adaptations resulting from rapamycin treatment of DB-1 human melanoma cells. CONCLUSIONS A rapid, sensitive, and specific IP-RP-UHPLC/NESI-MS method was used to quantify metabolites from several biochemical pathways. IP with DIPEA and HFIP increased the sensitivity and improved chromatographic separation when used with reversed-phase UHPLC.

Abstract:Salmon is a popular food but it is easily susceptible to spoilage by contamination with microorganisms. In this study, a method using hydrophilic interaction chromatography (HILIC)-based solid-phase extraction (SPE) and matrix-assisted laser desorption and ionization time-of-flight/time-of-flight mass spectrometry was developed and applied to reveal the effect of Pseudomonas fluorescens on salmon fillet during the shelf-life period by measuring the changes in the levels of phosphatidylcholine and phosphatidylethanolamine. Fresh samples were inoculated with P. fluorescens (10(6) cfu g(-1)) for 30 s, and lipids were extracted at 0, 24, 48, and 72 h. A homemade SPE cartridge packed with HILIC sorbent (silica derivatized with 1,2-dihydroxypropane) was used for matrix cleanup prior to analysis by mass spectrometry. In total, 30 phospholipids and 16 lysophospholipids were detected and elucidated. The results revealed that the content of phospholipids decreased significantly, whereas that of lysophospholipids increased initially, followed by a gradual reduction as the cold storage time increased. The contamination by P. fluorescens negatively affected the quality of fresh salmon without obvious physical changes, but it posed a potential threat to human health. This study suggests that the well-established method could be used for detecting phospholipids in salmon fillet and perhaps other foods as well.

Abstract:Adenosine triphosphate as a universal energy currency which can directly provide energy required for a multitude of biochemical reactions and biophysical actions through adenosine triphosphatase catalysed hydrolysis. Adenosine triphosphatase activity is thus one important feature for the characterization of protein function and cell activity. Herein, we optimized ion‐pair reversed‐phase HPLC technique for highly efficient separation of adenosine triphosphate, adenosine diphosphate and adenosine monophosphate, and the method demonstrated good linearity. Moreover, by coupling a protein‐removable ultrafiltration, we developed a sensitive and robust approach for quantification of adenosine triphosphatase hydrolytic activity. By this assay, we demonstrated that RecA filaments‐catalysed adenosine triphosphate hydrolysis approached a second‐order reaction, and its rate constant was estimated as 0.057 mM−1min−1. In addition, we explored the effects of DNA length on this reaction and revealed that the increase of the length of single‐stranded DNA can promote the adenosine triphosphatase hydrolytic activity of RecA filaments. All these results confirm the feasibility of this new method in quantification of adenosine triphosphatase hydrolytic activity assays. Compared with previous complicated enzyme‐coupled or homogeneous colorimetric measurements, the developed approach with high resolution separation allows a simple reaction system for adenosine triphosphatase assay and a sensitive detection free of interference from background noise.This article is protected by copyright. Abstract:The combination of ion chromatography (I C) and inductively coupled plasma emission spectroscopy (ICP-ES) was used for the sensitive and specific detection of hypophosphite (PO2), phosphite (PO3) methylphosphonic acid (MPA), and phosphate (PO4). Application of this technique to a wide range of environmental samples proved that reduced phosphorus was present in some situations including process water from thermal phosphorus plants, drinking water contacting cast iron, and phosphorus corrosion inhibitor used in water treatment and in sewage wastewater. Preliminary testing did not detect high concentrations of reduced phosphorus and phosphine in situations where it was previously reported to be very important, including anaerobic digesters in wastewater treatment plants. The new IC-ICP-ES technique is a promising tool for use in corrosion and soil research where phosphites are likely to be present.

Abstract:This article describes an unusual case of a young adult with traumatic obturator fracture-dislocation of the hip, involving a large femoral head fragment and severe delamination of articular cartilage. The dislocation was irreducible by closed reduction because of interposing soft tissues, including the rectus femoris and iliopsoas muscles, and torn joint capsules, and therefore, open reduction was performed using an anterolateral approach in the lateral decubitus position. The large femoral head fragment was released from the ligamentum teres and fixed to the dislocated femoral head with headless screws. The severely delaminated femoral head cartilage was repaired with suture anchors and absorbable sutures. The patient was kept nonweight bearing for 6 weeks postoperatively, and was then allowed to resume full weight bearing gradually. He returned to normal activities of daily living at 14 weeks. At 9 months postoperatively, arthroscopic examination showed complete healing of the fracture and cartilage lesions, and at 12-month follow-up, there was no clinical or radiographic evidence of arthritis or osteonecrosis. The patient had no pain or limp, and achieved an excellent result according to Epstein's clinical evaluation criteria. To our knowledge, no previous report exists on the arthroscopic follow-up of a repaired femoral head cartilage in patients with obturator fracture-dislocation of the hip along with a large femoral head fragment and severe delamination of articular cartilage.

Abstract:The determination of total deoxyribonucleic acid (DNA) concentration is of great importance in many biological and bio-medical analyses. The quantification of DNA is traditionally performed by UV spectroscopy; however the results can be affected greatly by the sample matrix. The proposed method quantifies phosphorus in digested calf thymus DNA and human DNA by high performance liquid chromatography (HPLC) combined with inductively coupled plasma mass spectrometry (ICP-MS). The method presented showed excellent baseline separation between all four DNA mono-nucleotides and 5′UMP. The ability of LC-ICP-MS to provide an internal check that only DNA derived phosphorus was counted in the assay was demonstrated by establishing a mass balance between the total phosphorous signal from undigested DNA and that from the speciated DNA. Column recoveries ranging from 95% to 99% for phosphorus resulted in a mass balance of 95% ± 0.5% for standard nucleotides, determined by LC-ICP-MS, compared to total DNA determined by flow injection coupled to ICP-MS (FI-ICP-MS). The method for quantification was validated by analysis of NIST SRM 2,372; a total speciated DNA recovery of 52.1 ng/μL, compared with an expected value of 53.6 ng/μL, was determined by external calibration. From repeat measurements, a mass balance of 97% ± 0.5% for NIST DNA was achieved. The method limits of detection for individual nucleotides were determined between 0.8 and 1.7 μg L−1 (31P) for individual nucleotides by LC-ICP-MS, and 360 ng L−1 for 5′AMP by direct nebulisation.

A Rapid UHPLC-HILIC Method for Algal Guanosine 5'-Diphosphate 3'-Diphosphate (ppgpp) and the Potential Separation Mechanism.

Abstract:A fast and facile hydrophilic interaction liquid chromatography (HILIC) method was developed and applied to quantify physiologically important ppGpp and its analogues in a tough sample, the astaxanthin-accumulating alga Hameatococcus pluvialis. The method is able to analyze simultaneously seven nucleotides, including ppGpp at the order of pmol g-1 cells within 12 min. Mechanism on the elution order was investigated. It was found that 1) phosphate salt competed for the amide groups on the HILIC column with the phosphate groups of the nucleotides; 2) intramolecular hydrogen bonds might contribute to the elution order by offsetting and reducing the number of free hydrogen acceptor/donor of the nucleotide molecules interacting with the amide groups. This is the first HILIC method for ppGpp, which is feasible and applicable to a wide range of samples, especially tough samples, e.g., algae and plants.

Abstract:objective:A reversed-phase high performance liquid chromatography method for the quantitative determination of free purine and total purine content after hydrolyzing in fishery products was studied,and the contents of free purine and total purines in hairtail surimi were determined.Method:Waters Atlantis dC18 column was used as the stationary phase and 0.02 mol/L KH2PO4(pH 3.62)as mobile phase.Flow rate:1.0 mL/min;Column temperature:30 ℃;Injection volume:10 μL;DAD detector:254 nm;the contents of free purine were determined by hydrolyzing with water at 90 ℃ for 12～14 min;the contents of total purine were determined by hydrolyzing with mixture of trifluoroacetic acid:formid acid:water(5:5:1,V/V/V) at 90℃ for 12 mins;Results:The method had a good resolution in 20 minutes,the peak area of purines and concentration achieved good linear relation ranged from 1 to 40 mg/L,r2>0.9998,the RSD was <2%.Conclusion:The method was simple,rapid and accurate,it can be applied to determining free purine and total purine in fishery products.
Pretreatment Method for the Analysis of Phosphate Oxygen Isotope (δ18op) of Different Phosphorus Fractions in Freshwater Sediments.

Abstract:Accurate measurement of the oxygen isotopic composition of dissolved phosphate (δ18OP) of different phosphorus (P) fractions in lacustrine sediments is very difficult because of the influence of large amounts of impurities. In this study, we developed a five-step method for obtaining high purity Ag3PO4 sample for the analysis of δ18OP of different P fractions in freshwater sediments. Sedimentary P was divided into NaHCO3-P, NaOH-P and HCl-P by chemical sequential extraction. Pretreatment procedures for different sedimentary P fractions were improved in the following respects: 1) abandonment of the magnesium-induced coprecipitation method to avoid the introduction of impurity ions, such as Mg2+ and Cl-, and reduce the loss of P; 2) use of a small amount of non-phosphate activated carbon powder to efficiently remove organic matter in extracts of NaHCO3-P and NaOH-P, and reduce the loss of P; 3) adjustment of the HCl-P extract pH to 4 in order to form Fe(OH)3-PO43- coprecipitate, thereby removing most of metals and Cl-. This method reduces the pretreatment steps, simplifies the operation and increases the recovery of phosphate (90.98%-96.69%). The high purity Ag3PO4 sample can be obtained and the repeatability and accuracy of measured δ18OP is better than 0.3‰, demonstrating high reliability and accuracy. This new method was used to analyze the δ18OP of different P fractions in sediments of a eutrophic lake in southwestern China. The preliminary results indicated that the δ18OP in the sediments can be used to identify different P sources, and provide new insights into sedimentary P cycling. The method established in this study provides a powerful tool for investigating the sources and biogeochemical cycle of P in freshwater sediments.

Abstract:31P nuclear magnetic resonance (NMR) spectra can be biased due to the hydrolysis of labile P species during sample treatment and NMR analysis. This paper offers an approach to circumvent this problem by performing sample preparation and analysis in 18O-enriched medium. Heavy 18O isotope atoms were introduced into the resulting artificial hydrolysis products. The NMR signal of 18O-labeled P was shifted upfield relative to the unlabeled P nuclei in natural metabolites. This isotope shift enabled an immediate differentiation of artificial hydrolysis products from natural metabolites. Moreover, the hydrolysis products could be accurately quantified. Our data suggest that the extent to which artificial hydrolysis alters NMR spectra varies among different types of environmental samples. For instance, 72–84% of the detected monoesters in the organic soils of this study were actually artificially hydrolyzed diesters. By contrast, artificial hydrolysis products in the mineral soils used for this study accounted for less than 6% of the total monoesters. Ephedra and its application to sport performance: another concern for the athletic trainer?

Abstract:OBJECTIVE The ma huang herb, otherwise known as ephedra, has gained widespread popularity as an ergogenic supplement. With the sympathomimetic alkaloid ephedrine as its primary active ingredient, ma huang is marketed to reduce fatigue; increase strength, power, and speed; decrease reaction time; and improve body composition. Although numerous side effects have been associated with the use of ma huang, its popularity in athletes continues to grow. This review provides rationale for the ergogenic claims regarding ma huang and compares and contrasts those claims with data from scientifically controlled investigations. DATA SOURCES MEDLINE and SPORT Discus were searched from 1970 to 2000 using the key words ma huang, ephedra, and ephedrine in combination with humans, exercise, performance, and side effects. DATA SYNTHESIS Ephedrine has been used alone or in combination with other drugs as an effective weight-loss agent. The weight loss has been attributed to thermogenic and lipolytic effects which, in combination with the central nervous system stimulating effects, have also resulted in its use as an ergogenic aid. Most of the scientific data, however, do not support manufacturers' ergogenic claims, and numerous side effects have been associated with ephedrine use. Thus, the safety and efficacy of ma huang as an ergogenic supplement must be questioned. CONCLUSIONS/RECOMMENDATIONS It appears that the risks associated with the use of ma huang far outweigh any possible ergogenic benefits. Thus, it is extremely important that athletic trainers educate athletes on these issues so they can continue to perform at an optimum level in a safe and healthy manner.
Analysis of Trace Phosphonates in Authentic Water Samples by Pre-Methylation and LC-Orbitrap MS/MS.

Abstract:Phosphonate is an important phosphorous species in the effluent of wastewater treatment plant (WWTP), contributing to eutrophication and interfering with phosphate removal in WWTP. It is particularly difficult to determine phosphonates in samples of complex solution chemistry, resulting in very limited information on their presence in environmental matrices. Herein, we proposed a sensitive method based on liquid chromatography tandem mass spectrometry (LC-MS/MS) to determine six quantitatively most important phosphonates even at the ng/L level, i.e., 2-phosphonobutane-1,2,4-tricarboxylic acid, 1-hydroxyethane 1,1-diphosphonic acid, nitrilotris(methylene phosphonic acid), ethylenediamine tetra(methylene phosphonic acid), hexamethylenediamine tetra(methylene phosphonic acid) and diethylenetriamine penta(methylene phosphonic acid). Trimethylsilyldiazomethane (TMSCHN2) derivatization of the target phosphonates is pre-requisite since it could greatly increase the sensitivity up to 2-3 orders of magnitude over direct analysis of the virgin ones. The sample pretreatment methods (including ion exchange and solid phase extraction(SPE)), the derivatization procedures, and the LC-MS/MS conditions were systematically optimized. The limits of quantitation for the six phosphonates in the background of tap water ranged from 1.4 mu g/L to 57 mu g/L for direct analysis, and from 5.0 ng/L to 200 ng/L for SPE enabled pre-concentration analysis, respectively. The reliability of the proposed method was successfully validated by analysis of authentic water samples collected from one river and three WWTPs (0.088-7200 mu g/L phosphonates) with satisfactory recoveries (72-126%).
