Abstract:Selenium (Se) is considered essential for human nutrition as it is involved in the metabolic pathway of selenoproteins and relevant biological functions. Microgreens, defined as tender immature greens, constitute an emerging functional food characterized by overall higher levels of phytonutrients than their mature counterparts. The nutraceutical value of microgreens can be further improved through Se biofortification, delivering Se-enriched foods and potentially an enhanced content of bioactive compounds. The current study defined the effect of sodium selenate applications at three concentrations (0, 8, and 16 μM Se) on the bioactive compounds and mineral content of coriander, green basil, purple basil, and tatsoi microgreens grown in soilless cultivation. Analytical emphasis was dedicated to the identification and quantification of polyphenols by UHPLC-Q-Orbitrap-HRMS, major carotenoids by HPLC-DAD, and macro micro-minerals by ICP-OES. Twenty-seven phenolic compounds were quantified, of which the most abundant were: Chlorogenic acid and rutin in coriander, caffeic acid hexoside and kaempferol-3-O(caffeoyl) sophoroside-7-O-glucoside in tatsoi, and cichoric acid and rosmarinic acid in both green and purple basil. In coriander and tatsoi microgreens, the application of 16 μM Se increased the total phenols content by 21% and 95%, respectively; moreover, it improved the yield by 44% and 18%, respectively. At the same Se dose, the bioactive value of coriander and tatsoi was enhanced by a significant increase in rutin (33%) and kaempferol-3-O(feruloyl)sophoroside-7-O-glucoside (157%), respectively, compared to the control. In green and purple basil microgreens, the 8 μM Se application enhanced the lutein concentration by 7% and 19%, respectively. The same application rate also increased the overall macroelements content by 35% and total polyphenols concentration by 32% but only in the green cultivar. The latter actually had a tripled chicoric acid content compared to the untreated control. All microgreen genotypes exhibited an increase in the Se content in response to the biofortification treatments, thereby satisfying the recommended daily allowance for Se (RDA-Se) from 20% to 133%. The optimal Se dose that guarantees the effectiveness of Se biofortification and improves the content of bioactive compounds was 16 μM in coriander and tatsoi, and 8 μM in green and purple basil.

Abstract:Kale (Brassica oleracea L. var. sabellica L.), kohlrabi (Brassica oleracea L. var. gongylodes L.) and wheat (Triticum aestivum L. cv. Bancal) microgreens were cultivated in presence of selenium 20 μmol L-1 as sodium selenite and sodium selenate mixture. The influence of this biofortification process was evaluated in terms of biomass production, total Se, macro- and micronutrients concentration, polyphenols, antioxidant activity, chlorophylls and carotenoids levels and total soluble proteins content. The results obtained have shown a significant concentration of total Se in the biofortified microgreens of kale (133 μg Se·g-1 DW) and kohlrabi (127 μg Se·g-1 DW) higher than that obtained for wheat (28 μg Se·g-1 DW). The Se uptake in all the species did not produce oxidative damage to the plants reflected in the bioactive compounds, antioxidant capacity or pigments concentration. These Se-enriched microgreens may contribute to the recommended intake of this nutrient in human diet as to overcome Se-deficiency.

Abstract:Selenium (Se) biofortification of plants has been recognized as a good strategy to improve the nutritive value of vegetables and increase Se daily intake in humans. Identifying the most appropriate method to enrich plants is a key issue in the biofortification process. We tested a biofortification technique that produces Se enriched seedlings for transplant, yet barely modifies conventional cultivation techniques. Lettuce (Lactuca sativa L.) and sweet basil (Ocimum basilicum L.) were exposed to selenium by adding 0, 1 and 3 mg L−1 (lettuce) and 0, 2 and 3 mg L−1 (basil) of Se, as sodium selenate, to the growing substrate immediately after sowing. When seedlings reached an appropriate size, they were transplanted into the open field, and plants were grown until maturity. Lettuce and basil seedlings accumulated selenium without any reduction in leaf biomass at maturity. The highest dose of Se induced a higher antioxidant capacity and flavonoid content in both species at both sampling times. At maturity, biofortified plants still showed a higher leaf Se content compared to the control, and would be able to provide from 10% to 17% (lettuce) and from 9% to 12% (basil) of the adequate intake (AI) of Se.

Abstract:Selenium is a micronutrient element that is beneficial for the growth and development of plants. It has antioxidant, anticancer, and antiviral properties that are essential for human and animal health. Low‐consumption mineral elements such as selenium can be included in the diet from various sources. To investigate the growth and phytochemical attributes of a broccoli cultivar "Heracklion", an experiment with five levels of selenium concentration (0, 5, 10, 15, 20 mg/L sodium selenate) was carried out in a randomized complete block design with 3 replications in the field condition. With increasing the concentration of sodium selenate in the foliar application, the accumulation of sodium selenate in broccoli increased and the highest amount (1.47 mg/kg dry weight) was measured at 20 mg/L of sodium selenate. The highest amount of photosynthetic pigments in leaves was recorded at 15 mg/L of sodium selenate. In the case of glucosinolates, with increasing selenium concentration up to 20 mg/L concentration, glucoraphanin, 4‐methoxy glucobrassicin, and aliphatic glucosinolates increased in leaves. It could be demonstrated that foliar application of selenium at 10 mg/L led to an improvement of secondary metabolites, especially glucoraphanin, both in leaves and florets, and could also have a positive effect on human nutrition.

Abstract:Plant biofortification with selenium in interaction with the application of an arbuscular mycorrhizal fungi (AMF)-based formulate,with the goal of enhancing Se bioavailability, is beneficial for the development of the environmentally friendly production of functional food with a high content of this microelement. Research was carried out in order to assess the effects of an AMF-based formulate and a non-inoculated control in factorial combination with two selenium treatments with an organic (selenocystine) or inorganic form (sodium selenate) and a non-treated control on the yield, quality, antioxidant properties, and elemental composition of shallot (Allium cepa L. Aggregatum group). Selenocystine showed the best effect on the growth and yield of mycorrhized plants, whereas sodium selenate was the most effective on the non-inoculated plants. The soluble solids, total sugars, monosaccharides, titratable acidity, and proteins attained higher values upon AMF inoculation. Sodium selenate resulted in higher soluble solids, total sugars and monosaccharide content, and titratable acidity than the non-treated control, and it also resulted in higher monosaccharides when compared to selenocystine; the latter showed higher protein content than the control. Calcium, Na, S, and Cl bulb concentrations were higher in the plants inoculated with the beneficial microorganisms. Calcium and sodium concentrations were higher in the bulbs of plants treated with both the selenium forms than in the control. Selenocystine-treated plants showed enhanced accumulation of sulfur and chlorine compared to the untreated plants. The AMF inoculation increased the bulb selenium content by 530%, and the Se biofortification with selenocystine and sodium selenate increased this value by 36% and 21%, respectively, compared to control plants. The AMF-based formulate led to increases in ascorbic acid and antioxidant activity when compared to the non-inoculated control. The bulb ascorbic acid was increased by fortification with both selenium forms when compared to the non-treated control. The results of our investigation showed that both AMF and selenium application represent environmentally friendly strategies to enhance the overall yield and quality performances of shallot bulbs, as well as their selenium content.

Abstract:Selenium (Se) is an essential trace element for human nutrition and a key component of selenoproteins having fundamental biological and nutraceutical functions. We currently examined lettuce biofortification with Se in an open-gas-exchange growth chamber using closed soilless cultivation for delivering Se-rich food. Morphometric traits, minerals, phenolic acids, and carotenoids of two differently pigmented Salanova cultivars were evaluated in response to six Se concentrations (0–40 μM) delivered as sodium selenate in the nutrient solution. All treatments reduced green lettuce fresh yield slightly (9%), while a decrease in red lettuce was observed only at 32 and 40 μM Se (11 and 21% respectively). Leaf Se content increased in both cultivars, with the red accumulating 57% more Se than the green. At 16 μM Se all detected phenolic acids increased, moreover a substantial increase in anthocyanins (184%) was recorded in red Salanova. Selenium applications slightly reduced the carotenoids content of green Salanova, whereas in red Salanova treated with 32 μM Se violaxanthin + neoxanthin, lutein and β-cryptoxanthin spiked by 38.6, 27.4, and 23.1%, respectively. Lettuce constitutes an ideal target crop for selenium biofortification and closed soilless cultivation comprises an effective tool for producing Se-enriched foods of high nutraceutical value.

Abstract:The biofortification of leafy vegetables with selenium (Se) is a good way to increase human dietary Se intake. In addition, selenium delays plant senescence by enhancing the antioxidant capacity of plant tissues, decreasing postharvest losses. We investigated the effects of selenium addition on the production and quality of sweet basil (Ocimum basilicum) leaves of two harvesting phases, hereafter referred to as cuts, during the crop cycle. Plants were hydroponically grown and treated with 0 (control), 4, 8 and 12 mg Se L−1 as selenate. To evaluate the growth, nutritional value and quality of the basil leaves, selected qualitative parameters were determined at harvest and after five days of storage. Application of Se at varying rates (4, 8 and 12 mg L−1) was associated with an increased leaf selenium concentration in the first, but not the second cut. The application of Se significantly affected the antioxidant capacity as well as the total phenol and rosmarinic acid contents at harvest. The reduction in ethylene production observed in the plants at 4 mg Se L−1 after five days of storage suggests that this Se treatment could be used to prolong and enhance the shelf-life of basil. The daily consumption of 10 g of Se-enriched basil leaves, which, as an example, are contained in a single portion of Italian pesto sauce, would also satisfy the recommended selenium supplementation in humans.

Abstract:One of the important sources of human selenium intake is vegetables, so increasing of selenium in edible parts of plants, biofortification, provides an effective approach to reduce selenium deficiency. Origanum vulgare L. is an important medicinal plant which is also used as a valuable spice. The present study was done to evaluate the ability of selenium accumulation of micropropagated O. vulgare under sodium selenate and/or salicylic acid treatment. Micropropagation of O. vulgare was done from shoot-tips as explants on modified Murashige & Skoog (MS) medium. After 1 month, rooted plantlets were transferred to modified Murashige & Skoog solidified medium containing sodium selenate (0, 25 and 50 μM) and/or salicylic acid (0 and 100 mgL −1 ). After 4 wk, growth, selenium accumulation, biochemical parameters and oxidative stress indicators were determined. The results showed that the plantlets were able to accumulate 16.1 μg g −1 DW and 28.27 μg g −1 DW selenium in shoots at 25 and 50 μM selenate sodium concentrations respectively. Both concentrations of selenium increased shoot dry weight, length of shoot and root, soluble sugars and total protein content, peroxidase and superoxide dismutase activity. On the contrary, malondialdehyde content decreased under selenium treatment. It was found that by increasing selenium concentration the content of total protein, H 2 O 2 and antioxidant enzymes activity increased significantly. Application of salicylic acid in combination with selenium improved growth but did not affect on selenium accumulation. The obtained results can be considered as the first report of biofortification of O. vulgare under tissue culture technique.

Abstract:As an effective neonicotinoid insecticide, imidacloprid (IMI) has been widely used in crop production, but its residue affects normal plant growth. Selenium (Se) is a non-essential mineral nutrient in higher plants, that acts as the active centre of glutathione peroxidase (GSH-Px), which removes harmful peroxides. In this study, we investigated the mechanism by which selenium improves the growth status of IMI-treated garlic plants through analyses of apparent morphology and antioxidant enzyme activity as well as the dynamic changes in nutrients and metabolites in the plants. The results showed that 80 μg/kg Na2SeO3 had a strong effect on alleviating the damage in garlic plants exposed to IMI (1.2 mg/kg) by increasing the absorption of mineral elements to enhance the synthesis of chlorophyll and antioxidant enzymes. A nontarget metabolomics analysis based on gas chromatography-mass spectrometry (GC-MS) indicated that the addition of Na2SeO3 to IMI-treated garlic could reconstruct the plant metabolic distribution by enhancing the nitrogen and indole metabolism, maintaining lower concentrations of secondary metabolites and maintaining the balance of the plant energy metabolism. Our study provides novel insights into the molecular mechanisms by which garlic plants responds to IMI exposure and suggests the use of selenium with IMI-contaminated plants as a solution for the advancement of sustainable agricultural pesticide use.

Abstract:Chinese cabbage ( Brassica rapa ssp. pekinensis ) is a globally cultivated and consumed leafy vegetable due to its abundant plant secondary metabolites and antioxidant compounds, including flavonoids, ascorbic acids, glucosinolates, and vitamins, which have been reported to confer health-promoting effects. Glucosinolates components in leaves of Chinese cabbage plantlets under different concentrations of sodium selenite (0, 30, and 50 μmol/L) were analyzed. Seven glucosinolates were identified and quantified using UHPLC-QTOF-MS. Finally, treatments with 30 and 50 μmol/L Na 2 SeO 3 solution significantly increased the levels of total selenium content as well as total phenols, flavonoids, anthocyanins, and DPPH free radical scavenging ability in Chinese cabbage seedlings. Our results revealed that 30 μmol/L Na 2 SeO 3 effectively enhanced aliphatic glucosinolate levels and total glucosinolate content while causing a significant reduction in indole glucosinolates. Furthermore, downregulation was observed for BrCYP79F1 , BrBCAT4 , and BrMAM1 genes associated with aliphatic glucosinolate synthesis. Conversely, BrMYB28 and BrCYP83A1 genes exhibited significant upregulation. Thus, the positive influence of Na 2 SeO 3 on glucosinolate biosynthesis in Chinese cabbage can be attributed to the upregulation of key genes related to this process.

Abstract:Macroporous cellulose and glass membranes were prepared from filter paper and glass fiber filter, respectively. To enhance their stability, the cellulose membranes were crosslinked with epichlorohydrin, and the glass membranes were crosslinked with glutaraldehyde or organic bifunctional silanes. Several pathways for the modification, activation, and ligand immobilization were used and compared. For cellulose membranes, the diazotization method provided the best results, whereas the glutaraldehyde method provided the best performance for glass membranes, regarding both their stability and ligand immobilization capacity. The characterization of the membranes was made by using a triazine dye, bovine serum albumin, and trypsin as test ligands. The membrane morphologies and the uniformities of ligand distribution across the membrane cartridges were investigated. Numerous affinity ligands were immobilized onto the membranes, and the prepared affinity membranes have been used to separate or purify concanavalin A, peroxidase, protease inhibitors, globulin, fibronectin, and other biomolecules.
Effect of Selenium Enrichment and Type of Application on Yield, Functional Quality and Mineral Composition of Curly Endive Grown in a Hydroponic System

Abstract:Selenium (Se) is an essential element for humans’ health and the recommended amount (RDA) of Se intake per adult is 55–70 μg day−1. The main source for Se intake is represented by diet, but its concentration in vegetables is generally limited with respect to human needs. The aim of this research was to assess the effect of Se application rate and type (fertigation or foliar spray) on yield, functional properties and mineral composition of curly endive grown in a hydroponic system. Five levels of Se (0.0, 1.0, 2.0, 4.0 and 8.0 μmol L−1) were supplied via fertigation or by foliar spray in the form of selenate (Na2SeO4). The results show that a Se implementation of 4 μmol Se L−1 for plants enriched via fertigation and 8 μmol Se L−1 for plants biofortified via foliar spray successfully enhanced production performance parameters such as head weight (42.6% and 27.8%, respectively), SSC (16.7% and 14.3%, respectively), ascorbic acid (33.2% and 33.7%, respectively), total phenolic (58.9% and 54.5%, respectively) and Se concentration (22.2% and 20.3%, respectively). Furthermore, leaf Se concentration in plants enriched via fertigation ranged 0.71–17.61 mg kg−1 of dry weight (DW), whereas, in plant biofortified via foliar spray leaf Se concentration ranged 0.72–12.67 mg kg−1 DW. Plants grown with the highest dosage of Se distributed via fertigation or foliar spray showed a reduction in total-N leaf concentration by 39.4% and 28.6%, respectively, compared with the non-enriched plants. Our results indicate a consumption of 47.4 g day−1 of Se-enriched curly endive grown in soilless culture and treated with 8 μmol L−1 of selenate applied via foliar spray could be sufficient to cover the human physiological needs of this element.

Abstract:Biofortification of selenium (Se) has emerged as an important strategy to enhance Se content in alfalfa, and sulfur (S) can be found to reduce the toxicity of Se to several crops. However, it is not clear if this interaction between Se(IV) and S enhances Se accumulation in alfalfa. In this study, alfalfa was grown on experimental pots and treated with foliar nutrient solutions containing 0–40 mg L −1 sodium selenite (Se(IV)), both with and without supplementary S. Results showed that foliar application of 10 mg L −1 Se(IV) with a S/Se(IV) ratio of 46.83 significantly increased shoot length (28.6%), shoot biomass (39.2%), content of total Se (23.4 times), and total organic Se in alfalfa leaves at the early flowering stage. Levels of selenocysteine (SeCys) and methylselenocysteine (MeSeCys) accounted for 75.2–81.8% of total Se, whereas selenomethionine (SeMet) comprised just 13.7–15.2% of total Se, confirming that SeCys and MeSeCys were the predominant form of Se in Se(IV)-treated leaves. Se-enriched leaves contained higher concentrations of minerals, including iron (33.4%), magnesium (36.3%), manganese (58.6%), zinc (53.2%), copper (34.8%), and total sulfur (22.6%). Concentrations of crude protein were increased by an average of 45.2%. Higher concentrations of S offered a protective effect against Se toxicity and indeed reduced total Se content by 10.9% (20 mg L −1 Se(IV)) and 24.6% (40 mg L −1 Se(IV)), respectively. Based on the above results, the adoption of this agronomic practice could be a promising strategy for Se biofortification and improved nutritional quality in alfalfa grown on Se-deficient soils.

Abstract:Lettuce is a common vegetable in hydroponic production. In this paper, a selenium (Se)-biofortification method was provided. The Se content, speciation, and the effects of different concentrations of selenate and selenite on lettuce growth and amino acids were investigated. The results showed that lettuce had strong ability to accumulate exogenous selenium, and inorganic Se could be effectively converted into organic Se. The proportion of organic Se in the shoots under treatment with 4 μmol L−1 selenite was 100%. Selenomethionine was the main organic Se, accounting for 51% (selenate) and 90% (selenite) of the total Se. Adding Se improves photosynthesis of lettuce and promotes growth. The growth with 2 μmol L−1 selenate and 4 μmol L−1 selenite was better than CK, and the shoot fresh weight was increased by 143.22% and 166.98%, respectively. Furthermore, the optimum Se application is 2 μmol L−1, and some areas can apply 4 μmol L−1 selenite. But Se-excessive areas are not recommended to grow selenium-rich foods. Therefore, lettuce has strong biofortification potential.

Abstract:Production of functional food with high levels of selenium (Se) and iodine (I) obtained via plant biofortification shows significant difficulties due to the complex interaction between the two elements. Taking into account the known beneficial effect of silicon (Si) on plant growth and development, single and joint foliar biofortification of chervil plants with potassium iodide (150 mg L−1) and sodium selenate (10 mg L−1) was carried out in a pot experiment with and without Si nanoparticles foliar supplementation. Compared to control plants, nano-Si (14 mg L−1) increased shoot biomass in all treatments: by 4.8 times with Si; by 2.8 times with I + Si; by 5.6 times with Se + Si; by 4.0 times with I + Se + Si. The correspondent increases in root biomass were 4.5, 8.7, 13.3 and 10.0 times, respectively. The growth stimulation effect of Se, I and I + Se treatments resulted in a 2.7, 3.5 and 3.6 times increase for chervil shoots and 1.6, 3.1 and 8.6 times for roots, respectively. Nano-Si improved I biofortification levels by twice, while I and Se enhanced the plant content of each other. All treatments decreased nitrate levels, compared to control, and increased the photopigment accumulation. Improvement of total antioxidant activity and phenolic content was recorded only under the joint application of Se + I + Si. Foliar nano-Si treatment affected other element content in plants: decreased Na+ accumulation in single and joint supplementation with Se and I, restored Fe, Mn and Cr amount compared to the decreased levels recorded in separately Se and I fortified plants and promoted Al accumulation both with or without Se and I biofortification. The results of this research suggest high prospects of foliar nano-Si supply for enhancing both growth and joint I/Se biofortification of chervil.

Abstract:Abstract Background Selenium (Se) is an essential element for mammals and its deficiency in the diet is a global problem. Plants accumulate Se and thus represent a major source of Se to consumers. Agronomic biofortification intends to enrich crops with Se in order to secure its adequate supply by people. Scope The goal of this review is to report the present knowledge of the distribution and processes of Se in soil and at the plant-soil interface, and of Se behaviour inside the plant in terms of biofortification. It aims to unravel the Se metabolic pathways that affect the nutritional value of edible plant products, various Se biofortification strategies in challenging environments, as well as the impact of Se-enriched food on human health. Conclusions Agronomic biofortification and breeding are prevalent strategies for battling Se deficiency. Future research addresses nanosized Se biofortification, crop enrichment with multiple micronutrients, microbial-integrated agronomic biofortification, and optimization of Se biofortification in adverse conditions. Biofortified food of superior nutritional quality may be created, enriched with healthy Se-compounds, as well as several other valuable phytochemicals. Whether such a food source might be used as nutritional intervention for recently emerged coronavirus infections is a relevant question that deserves investigation.

Abstract:In order to improve the added value and functionality of radish, an agricultural strategy of bio-nano-selenium was established in this study. Using bio-nano-selenium as an exogenous source of selenium fortification, foliar sprays were applied with different concentration gradients and fertilization periods to study its effects on radish yield, quality traits and selenium content in various parts. The results showed that exogenous selenium supplementation increased the root to leaf ratio of radish (125.66%), promoted the underground transfer of nutrients in radish, and increased the yield by 41.56% on average. The soluble solid content (5.85%), content of soluble sugar (25.13%), reducing sugar content (10%) and dry matter content (3.6%) of radish were increased, but the vitamin C content (15.49%) and water soluble protein content (25%) were decreased, and the crude fiber content was not affected. The bio-nano-selenium had a significant effect on the selenium content of radish parts, in the order of radish leaves > radish peel > radish roots > radish core. In this study, the growth, yield, quality and selenium content of various parts of radish was comprehensively studied by using self-developed bio-nano-selenium for the first time. The results showed that exogenous supplementation of bio-nano-selenium could significantly increase the selenium content of radish and overall promote the growth of radish and improve yield and quality traits.

Abstract:Garlic is known for its capacity to accumulate sulfur (S) and selenium (Se) in its bulbs. Therefore, it is a brilliant plant crop for S and Se biofortification and enrichment of consumers' diet with these elements. However, environmental and consumer concerns may limit mineral biofortification of plant crops by chemical fertilizers. This study investigated the effects of using a natural source of S, gypsum (0, 20, and 40 ppm S in the soil) and foliar application of Se (0, 15, and 30 ppm Se) on mineral biofortification of two of Iranian garlic ecotypes (Gorgan and Ramhormoz). After 40 days, the plants were harvested and their mineral composition and bioactive compounds were evaluated. In both ecotypes, the use of S with low Se concentration increased antioxidative capacity of the bulbs. Simultaneous application of S and Se resulted in the highest concentration of total phenolics in both ecotypes. Effect of application of selenium and sulfur on flavonoids' concentration in the bulbs of the ecotypes was different. Gorgan ecotype had the highest flavonoids' concentration under low sulfur concentration, while Ramhormoz ecotype obtained the highest flavonoids following application of high concentrations of S and Se. In both ecotypes, the highest content of allicin was found in high S but with low Se levels. Moreover, in parallel with increasing of S and Se levels, concentration of these elements increased in both ecotypes. The highest concentrations of selenium and sulfur in the bulbs (969.9 μg kg−1 DM and 1.97 mg g−1 DM, respectively) were observed in high S combined with high Se in Gorgan ecotype. Therefore, this study demonstrated the possibility of improving the nutritional value of garlic by simultaneous application of gypsum and Se foliar spray.

Abstract:The ability of some crops to accumulate selenium (Se) is crucial for human nutrition and health. Selenium has been identified as a cofactor of the enzyme glutathione peroxidase, which is a catalyzer in the reduction of peroxides that can damage cells and tissues, and can act as an antioxidant. Plants are the first link in the food chain, which ends with humans. Increasing the Se quantity in plant products, including leafy and fruity vegetables, and fruit crops, without exceeding the toxic threshold, is thus a good way to increase animal and human Se intake, with positive effects on long-term health. In many Se-enriched plants, most Se is in its major organic form. Given that this form is more available to humans and more efficient in increasing the selenium content than inorganic forms, the consumption of Se-enriched plants appears to be beneficial. An antioxidant effect of Se has been detected in Se-enriched vegetables and fruit crops due to an improved antioxidative status and to a reduced biosynthesis of ethylene, which is the hormone with a primary role in plant senescence and fruit ripening. This thus highlights the possible positive effect of Se in preserving a longer shelf-life and longer-lasting quality.
