Abstract:Due to the dissemination of airborne conidia and spores, molds can contaminate various surfaces. In the food industry sector, their presence and development can have health and economic implications. In order to control these undesirable microorganisms, various approaches can be used but the main one relies on the use of disinfectants. The objective of this review is to report the existing studies on the effect of various disinfectant molecules (i.e., sodium hypochlorite, chlorine dioxide, ethanol and other alcohols, hydrogen peroxide, peracetic acid, and quaternary ammonium compounds) on the inactivation of fungal spores. These studies were sorted depending on the targeted fungal species. Noteworthy, in the food industry, four log and three log reductions are required to claim a fungicidal activity for suspension (European Standard 1650, 2019) and surface (European Standard 13697/IN1, 2019) treatments, respectively. Most of the presented studies concerned Penicillium and Aspergillus species (44 and 31% of the literature, respectively). In general, for a given disinfection procedure, ascospores were more resistant than conidia, and Aspergillus conidia were more resistant than Penicillium ones. However, the variability of encountered molds (e.g. species, strains, physiological state) and disinfection procedures (e.g. molecules, concentrations, contact time) affected the efficacy of disinfectants.

Abstract:In vitro and in vivo evaluations of the effects of three commercial disinfectants on isolates of Trichoderma harzianum Rifai and Cladobotryum dendroides (Bull) W Gams & Hoozemans were combined in the development of methodologies for realistic assessment of disinfectant performance. 'Environ' and 'Purogene' incorporated into agar media at 50 mg AI litre-1 were effective in totally preventing the mycelial growth of T harzianum and C dendroides isolates, whereas with 'Sudol' a concentration of 500 mg AI litre-1 was required. Evaluation of a model in vivo system was combined with observations on the fungicidal effects of disinfectants. Spore suspensions of T harzianum (Th1), T harzianum (Th2) and C dendroides prepared from culture washings with sterilised distilled water were used as contaminating inoculum. Environ, Sudol and Purogene in aqueous solutions at 1000 mg AI litre-1 were sprayed onto these building structure surfaces before or after artificial contamination with spore suspensions. Re-isolation from surfaces was carried out using agar swabs, which were prepared in visking tubing, applied to treated surfaces and incubated at 25 degrees C for 24, 48, 72 or 96 h. Environ and Purogene were more effective than Sudol in limiting the recovery of Trichoderma spp and C dendroides. All three disinfectants applied 12 h after artificial contamination onto wood, concrete or glazed surfaces were able to reduce the recovery of these isolates more effectively than when they were applied 12 h before artificial contamination. Greater persistence of contamination was noted on the rougher surfaces (wood and concrete) than on the smoother glazed surface. It was concluded in this in vivo evaluation that, in many cases, the protection achieved by the physical nature of the smooth glazed tile surfaces was equivalent to that available from the application of disinfectants, thus highlighting the attention needed to the nature of building surfaces in the structures used in the mushroom industry. The artificial contamination and re-isolation techniques allowed significant comparison between disinfectant materials, timings and surface types. In the in vivo experiments using a single standardised test concentration, the relative fungicidal effects of the disinfectants, which varied in the complexity of their active ingredients, were better discriminated than the effects observed in vitro, although the general trend was similar.

Abstract:Objectives: The aim of this study was to determine the susceptibility of 77 mould strains: Aspergillus fumigatus (20), Aspergillus flavus (8), Aspergillus niger (4), Aspergillus ochraceus (2), Penicillium citrinum (15), Penicillium crysogenum (14), Penicillium aurantiogriseum (1), Penicillium roquefortii (4), Penicillium paneum (2), Rhizopus spp. (3), Tricoderma spp. (1) and Mucor spp. (3) to biocides. Methods: MIC determination was determined based on CLSI methodology. Results: For hospital acquired strains, MIC50 was 0.5mg/L, MIC90 was 1mg/L for chlorhexidine (CHX); MIC50 was 0.5mg/L, and MIC90 was 1mg/L for benzalkonium chloride (BZC); MIC50 was 1mg/L, and MIC90 was 2mg/L for triclosan (TRC); MIC50 was 1024mg/L, and MIC90 was 2048mg/L for sodium hypochloride (SHC). For feed and food isolates MIC50 was 2mg/L, MIC90 was 8mg/L for CHX, MIC50 was 2mg/L, and MIC90 was 4mg/L for BZC, MIC50 was 2mg/L, and MIC90 was 4mg/L for TRC, MIC50 was 256mg/L, and MIC90 was 512mg/L for SHC. Conclusion: We can conclude that food isolates presented slightly higher MIC50 and MIC90 values for CHX, BNZ and TRC, but not for SHC.
Comparison of the Antifungal Activity of Chlorine Dioxide, Peracetic Acid and Some Chemical Fungicides in Post-harvest Management of Penicillium digitatum and Botrytis cinerea Infecting Sweet Orange and Strawberry Fruits

Abstract:Penicillium digitatum and Botrytis cinerea are some of the most important post-harvest pathogenic fungi infecting vegetables and fruits. This study is aimed to assess the antifungal potency of chlorine dioxide (ClO 2 ) and peracetic acid (PAA) by examining the growth of P. digitatum and B. cinerea on inoculated potato-dextrose-agar medium (PDA), sweet orange and strawberry fruits under in vitro and storehouse conditions. Furthermore, the antifungal efficacy of ClO 2 and PPA was compared with four commercial fungicides namely Thiabendazole, Imazalil, Cyprodinil and Thiophanate-methyl (1000 mg/L). The results indicated that the antifungal activity of ClO 2 and PAA under both in vivo and in vitro conditions were similar to the chemical fungicides. At lower concentration, ClO 2 was more active than PAA. The radial growth of pathogenic fungi in PDA medium was completely inhibited by 100 ppm ClO 2 and 750–900 ppm PAA. Under the storehouse condition, application of 400 ppm ClO 2 and 1050 ppm PAA completely inhibited the test fungi on orange and strawberry fruits. No significant difference was observed in the total sugar content of the fruits following application of the test compounds at these concentrations. This study concludes that ClO 2 and PAA can be used as viable substitutes for chemical fungicides for the management of P. digitatum and B. cinerea in sweet orange and strawberry fruits.
Factors That Interfere in the Action of Sanitizers against Ochratoxigenic Fungi Deteriorating Dry-Cured Meat Products

Abstract:This study verified the factors affecting the antifungal efficacy of sanitizers against ochratoxin A-producing fungi. The fungi Penicillium nordicum, Penicillium verrucosum, and Aspergillus westerdijkiae were exposed to three sanitizers at three concentrations: peracetic acid (0.3, 0.6, 1%), benzalkonium chloride (0.3, 1.2, 2%), and sodium hypochlorite (0.5, 0.75, 1%) at three exposure times (10, 15, and 20 min), three temperatures (10, 25, and 40 °C), and with the presence of organic matter simulating clean (0.3%) and dirty (3%) environments. All the tested conditions influenced the antifungal action of the tested sanitizers. Peracetic acid and benzalkonium chloride were the most effective sanitizers, and sodium hypochlorite was ineffective according to the parameters evaluated. The amount of organic matter reduced the antifungal ability of all sanitizers. The longer exposure time was more effective for inactivating fungi. The temperature acted differently for benzalkonium chloride, which tended to be favored at low temperatures, than for sodium hypochlorite and peracetic acid, which were more effective at higher temperatures. The knowledge of the parameters that influence the action of sanitizers on spoilage fungi is vital in decision-making related to sanitizing processes in the food industry.

Abstract:Introduction. Bacteria in biofilms (BFs) have increased resistance to antibacterial agents, including disinfectants; however, the efficacy level varies depending on the chosen treatment. Therefore, evaluation of efficacy of main disinfectants against BF-residing microorganisms is of scientific and practical interest. The purpose of the study was to explore the effect of disinfectants from various chemical groups on gram-positive and gram-negative bacteria residing in BFs. Materials and methods. The effect of the following disinfectants has been evaluated: alkyldimethylbenzylammonium chloride (ADBAC), tertiary amine (TA), polyhexamethylene guanidine chloride (PHMG), hydrogen peroxide (HP), chloramine (CA), dichloroisocyanuric acid sodium salt (Na DCC), sodium hypochlorite (HC), ethyl alcohol (EA), glutaraldehyde (GA)) against Pseudomonas aeruginosa ATCC 15442 and Staphylococcus aureus ATCC 6538-P BFs. BFs were grown in 96-well plates at 37oC for 24 hours and then exposed to biocide solutions. The efficacy of disinfectants was evaluated by the number of remaining viable cells and BF relative density. Results. The analyzed bacterial strains formed moderate BFs; the average number of viable cells in BFs was 6.51 ± 0.19 lg. The viable bacterial cell counts in BFs reduced by more than 4 lg when exposed to HP solutions at a concentration of 6%, Na DCC solution — 0.1% (by active chlorine), HC — 1% (by active chlorine), CA – 1% (by product), PHMG — 0.05%, TA — 1.0 %. The BF density decreased by more than 70%. ADBAC solutions at concentrations of 0.1–1.0%, TA — 0.05%, HP — 3%, Na DCC solution — 0.05% (by active chlorine) caused a 2-lg reduction in viable cell counts in BFs. The efficacy of chlorine-active compounds and HP increased when 0.5% sulfonol was added. GA (0.25–1.00%) and EA (40–70%) solutions were ineffective against BF microorganisms. Conclusion. A promising potential in combating microbial biofilms is demonstrated by disinfectants from the group of oxidizing agents (chlorine-active and oxygen-containing), TA and PHMG; using ADBAC as an individual compound is ineffective; aldehydes and alcohols are unable to destroy BFs and eliminate microorganisms in them.

Abstract:Using single‐cell technologies, we found that the protein coat of spores was degraded, internal molecules were released, and germination and growth of spores were greatly affected after sodium hypochlorite treatment. Pollution from toxic spores has caused us a lot of problems because spores are extremely resistant and can survive most disinfectants. Therefore, the detection of spore response to disinfectant is of great significance for the development of effective decontamination strategies. In this work, we investigated the effect of 0.5% sodium hypochlorite on the molecular and morphological properties of single spores of Bacillus subtilis using single‐cell techniques. Laser tweezers Raman spectroscopy showed that sodium hypochlorite resulted in Ca2+‐dipicolinic acid release and nucleic acid denaturation. Atomic force microscopy showed that the surface of treated spores changed from rough to smooth, protein shells were degraded at 10 min, and the permeability barrier was destroyed at 15 min. The spore volume decreased gradually over time. Live‐cell imaging showed that the germination and growth rates decreased with increasing treatment time. These results provide new insight into the response of spores to sodium hypochlorite.

Abstract:Heat-resistant fungal conidia are a common source of contamination and can cause significant difficulties in producing spawns. Through the use of PCR method, Aspergillus tubingensis and Aspergillus flavus as common microbial contaminants found in wheat grain spawn were identified that had been sterilized at 120 ºc for 2 h. Since these conidia are highly resistant to standard sterilization techniques, alternative methods were used to treat them with NaOCl and cold plasma and evaluate their effectiveness in reducing contamination. Optical emission spectroscopy (OES) analysis of the plasma showed dominant emissions from the N2 second positive system and N2+ first negative system, while reactive oxygen species (ROS) spectral lines were undetected due to collision-induced quenching effects. Field Emission Scanning Electron Microscopy (FESEM) and Energy Dispersive X-ray Spectroscopy (EDXS) analyses revealed notable alterations in the elemental makeup of conidia surfaces, as evidenced by a marked rise in levels of Na, O, Cl (in the case of NaOCl treatment) and N (in the case of plasma treatment). The conidia size was reduced at lower levels of NaOCl, but with increased concentrations and plasma treatment, the conidia underwent rupture and, in some cases, pulverization. The research suggests that utilizing a combined approach can be highly effective in eliminating heat-resistant fungal conidia and drastically cutting down the sterilization time for producing wheat spawn to only 30 s.

Abstract:Business process redesign (BPR) is a management technique to radically transform organizations for dramatic improvement. Information technology (IT) plays a critical role in BPR. The present study examines the links between IT infrastructure and BPR. The moderating effects of industry type, and information intensity of the firm are also analyzed. Data for the study were gathered through a survey of Fortune 500 US firms at divisional levels. Out of 1,200 questionnaires mailed to Fortune500 firm‐divisions, 124 responses were received. Out of 124 firm‐divisions 73 firm‐divisions were found to be adopting BPR techniques; 39 firm‐divisions were found to be adopting incremental improvement approaches. The rest of the responses were incomplete and could not be used. For data analysis, therefore, only 73 firm‐divisions were considered. The results of the study support the hypotheses that network infrastructure affects the dimensions of BPR (process improvement thrust, and customer focus), but data integration was not found to be significantly affecting the BPR dimensions. The moderating effect of industry type was found to be significantly affecting the relationship between network infrastructure and BPR dimensions. Other relationships were not found to be significant.
Inactivation of fungal spores in water with peracetic acid: Efficiency and mechanism

Abstract:In recent years, peracetic acid (PAA) has been widely used in sewage disinfection due to its strongly sterilizing capacity and less toxic by-products production. However, there was no studies on the inactivation kinetics and mechanism of fungal spores in water by PAA as well as their regrowth. The inactivation efficiency and mechanism of Aspergillus niger and Aspergillus flavus by PAA was illustrated in this study. The results showed that the inactivation of fungal spores fitted to the delayed Chick–Watson model well. The inactivation rate constants were 0.038 and 0.033 L mg−1 min−1, respectively. The effects of pH value and humic acid concentration on the inactivation efficiency were negative and the effect of temperature was positive, while the effect of HCO3 − and Cl− on the disinfection of fungal spores could be negligible. The results of flow cytometric measurements, morphological observation and the leakage of intracellular substances indicated that the inactivation of fungi by PAA was attributed to the penetration of cell membranes rather than the damage of cell membranes. PAA was found to be effective for the inactivation of fungal spores, but was less effective than several traditional disinfectants (Cl2, ClO2) under the same condition. Furthermore, the regrowth of fungal spores after disinfection with PAA was also evaluated. The regrowth of fungal spores that after 1–3 log inactivated in the R2A medium within 5 days was observed, but the regrowth of completely inactivated fungal spores in R2A and in trace element solution was negligible. This study provides the valuable information for the application of PAA in the control of fungal pollution.

Abstract:Fungi represent a considerable challenge given their ubiquitous inhabitation and growth in drinking water distribution systems (DWDSs), yet the germination behavior of fungal spores in DWDSs is largely unknown. This study investigated germination of three chlorine-resistant fungal spores (Penicillium chrysogenum, Cladosporium halotolerans, and Penicillium spinulosum) in synthetic tap water (STW) under varying conditions of disinfectants (free chlorine, FC; monochloramine, NH2Cl), humic acid (HA), and pH. The germination ratios of the tested fungal spores in STW(+HA) were <10% without FC/NH2Cl, while surprisingly increased to a maximum of 67.5% and 44.14% with 2 mg/L FC and 0.4 mg/L NH2Cl, respectively, for C. halotolerans spores. With increasing initial concentrations of FC (0–4 mg/L) and NH2Cl (0–2 mg/L), germination ratios of C. halotolerans spores in STW+HA showed a first increasing and then decreasing trend, suggesting that the stimulation effect was outweighed by inactivation at high CT values. Nontargeted metabolomics analysis identified the decrease of several potential self-inhibitors for spore germination by chlor(am)ination. Patterns of significantly changed metabolites and pathways involving lipid, sugar, and amino acid metabolism were significantly different for C. halotolerans spores treated by FC versus NH2Cl. The results provide new insights into survival strategies and germination mechanisms of fungal spores in chlor(am)ine-disinfected DWDSs.

Efficacy of Hypochlorite in Disinfesting Nonfungal Plant Pathogens in Agricultural and Horticultural Plant Production: A Meta-Analysis

Abstract:Bleach products containing hypochlorite are commonly used as disinfectants to eliminate nonfungal plant pathogens from production surfaces, tools, plant surfaces, irrigation water, and produce dump tanks. Although bleach products are useful, their effectiveness has been reported to vary under specific settings. A meta-analysis was conducted of 86 studies to assess the overall efficacy of hypochlorite against plant pathogenic bacteria, oomycetes, and viruses and to identify factors that explain differences in product efficacy. Hypochlorite resulted in a significant (P < 0.0001) reduction in disease intensity or propagule viability, with a mean Hedges' g standardized difference ([Formula: see text]) of 3.01, indicating that overall, hypochlorite treatments are highly effective. However, heterogeneity in g was significant (P < 0.0001) between studies, wherein 69.8% of the variance observed in g was attributed to true effects. Furthermore, an estimate of between-study variability was moderate (τ2 = 1.46). Random effects (REs) metaregression showed limited effects of moderator variables dosage, contact time, targeted material of treatment, and organism type on product efficacy when all organism types were considered together. Because subgroup [Formula: see text] was significantly higher (P = 0.0070) for oomycetes ([Formula: see text] = 3.30) than for bacteria ([Formula: see text] = 2.19), subsequent metaregressions were performed by organism type. For oomycetes, five RE metaregression models, each containing two moderators and their interaction, resulted in significant (P = 0.05) effects, where models with dosage and time, dosage and genus, time and genus, dosage and target, and time and target accounted for ≤50, 71, 57, 48, and 47%, respectively, of the variance in true effect sizes (R2) associated with [Formula: see text]. For viruses, only the RE metaregression model containing time and target and their interaction resulted in significant (P = 0.0435) effects accounting for 38% of the variance in true effect sizes associated with [Formula: see text]. None of the RE metaregression models for bacteria were significant, although they still accounted for ≤28% of the variance in true effect sizes associated with [Formula: see text]. These results show that although the current recommended rates for dosage and contact time for commercial bleach products are generally expected to result in effective disinfestation, the efficacy against nonfungal plant pathogens is expected to be influenced by the organism type and target being treated with hypochlorite.

Abstract:Bacterial spores in materials and equipment pose significant biosecurity risks, making effective disinfection crucial. This study evaluated Ortho-phthalaldehyde (OPA) and a quaternary ammonia-glutaraldehyde solution (AG) for inactivating spores of Bacillus thuringiensis (BT), B. cereus (BC), and two strains of B. velezensis (BV1 and BV2). Spores of BV1 and BT were treated with 22.5 mg/m 3 OPA by dry fumigation or 1 mg/mL AG by spray for 20 min, according to the manufacturer's recommendation. As no sporicidal effect was observed, OPA was tested at 112.5 mg/m 3 for 40 min, showing effectiveness for BT but not for BV1. Minimum bactericidal concentration (MBC) tests revealed higher MBC values for glutaraldehyde, prompting an overnight test with 112.5 mg/m 3 OPA by dry fumigation and 50 mg/mL AG by spray, using formaldehyde as a control. AG reduced all Bacillus strains, but with limited sporicidal effect. OPA was sporicidal for BT and BV1 but not for BC and BV2, indicating a strain-dependent effect. Formaldehyde performed better overall but did not completely inactivate BV2 spores. Our findings suggest that OPA and AG have potential as formaldehyde replacements in wet disinfection procedures.

[Compatibility of Two Fungal Biocontrol Agents Conidia with Commercial Chemical Acaricides].

Abstract:In this paper, the biological compatibility of fungal biocontrol agents Beauveria bassiana and Paecilomyces fumosoroseus conidia with 10 commercial chemical acaricides were assayed, based on the conidial germination rates in nutritional liquid and on SDAY plate. The results showed that nutritional liquid was more available than SDAY plate in the assay. So far as the 24 h conidial viability concerned, there were significant differences among the test acaricides with the concentrations of recommended for field spray and 5 and 10 fold dilutions, as well as between the two fungal agents. Since acaricides azocyclotin, liuyangmycin, dicofol and avermectin had strong inhibitory effects on the 24 h germination rate of both fungal agents conidia, their combined application with fungal agents was unsuitable for mite control. However, the combined application of pyridaben, propargite, chlorpyrifos, hexythiazox or amitraz with either B. bassiana or P. fumosoroseus was practical, because of their short-term compatibility. When the mixtures of oil-based B. bassiana formulation with the three concentrations of pyridaben, propargite and chlorpyrifos were stored at 4 degrees C or at ambient temperature for 12 months, none of the three chemicals was considered to be good enough for a combined formulation due to the great variability in long-term compatibility. Nevertheless, chlorpyrifos exhibited an encouraging long-term compatibility with B. bassiana, because its low concentration in the fungal formulation did not affect the conidial viability during a 6.5-month period of storage at ambient temperature. When stored at 4 degrees C in dark, the B. bassiana formulation containing low or medium concentration of chlorpyrifos retained the conidial viability of > 90% for up to 12 months.

Abstract:Summary The role of the healthcare environment in the transmission of clinical pathogens is well established. We investigated the chemical stability and sporicidal efficacy of oxidising disinfectant products in the presence of simulated clean and medical dirty conditions. Performance of chlorine-releasing agents (sodium dichloroisocyanurate, chlorine dioxide and hypochlorous acid) was concentration-dependent, with 1000 ppm chlorine showing reduced stability and efficacy in dirty conditions. In contrast peracetic acid product demonstrated stability and consistently achieved efficacy in dirty conditions. These results have implications for clinical practice, as ineffective environmental decontamination may increase the risk of transmission of pathogens that can cause healthcare-associated infections.

Abstract:Abstract: Bacillus cereus spores have the ability to adhere to solid surfaces, including stainless steel, a material widely used in food industries. Adhesion of spores allows recontamination during food processing, and cleaning and disinfection are largely used by industries to control them. Hence, this study aims to assess the detachment capacity (or removing activity) of detergents based on sodium hydroxide, nitric acid, phosphoric acid, and chlorine against two adhered B. cereus spores (one hydrophobic and other hydrophilic) to stainless steel surfaces. Microorganism adhesion on the surfaces reached 5.5 log CFU/cm2 for the two strains studied. Two protocols composed of combinations of chemical compounds, concentration, temperature, and contact time were tested. The inactivation kinetics shapes were convex and modeled by the Weibull model. The effects of temperature and biocide concentration were quantified using a Bigelow-like model. The temperature applied during the cleaning-in-place treatment is an important factor acting on the speed of inactivation or detachment of B. cereus spores. However, this efficiency depends on the hydrophobic characteristics of B. cereus spores. The concentration of detergent and acid also affects the inactivation rate, whereas the characteristic of hydrophobicity does not intervene for the chlorine alkaline treatments.

Abstract:Disinfectants are crucial for root canal therapy (RCT), as metabolism of canal-inhabiting microbes can cause refractory infections. To develop effective yet patient- and environment-friendly disinfectant formulations, we quantitatively assessed the metabolism-inhibiting effects of intracanal disinfectants via D 2 O-probed Single-Cell Raman Spectra (SCRS), using Candida albicans ( C. albicans ) as a pathogen model. For chlorhexidine gluconate (CHX), sodium hypochlorite (NaClO), and hydrogen peroxide (H 2 O 2 ), at their MIC of 4, 168, and 60 μg/ml, respectively, despite the complete growth halt, metabolic activity of individual fungal cells was reduced on average by 0.4%, 93.9%, and 94.1% at 8 h, revealing a “nongrowing but metabolically active” (NGMA) state that may underlie potential refractory infections, particularly for CHX. In contrast, at their Metabolic Activity-based Minimum Inhibitory Concentrations (MIC-MA) of 8, 336, and 120 μg/ml, respectively, metabolic activity of all cells was completely halted throughout 8 h exposure. Moreover, combined use of NaClO+H 2 O 2 (combination at 0.5× MIC-MA each) outperforms solo uses of CHX, NaClO, H 2 O 2 , or other binary combinations. Furthermore, dynamics of SCRS revealed distinct fungicidal mechanisms of CHX, NaClO, H 2 O 2 , and their pairwise combinations. MIC-MA is advantageous in critically assessing antifungal efficacy, and NaClO+H 2 O 2 can potentially serve as a more efficient disinfectant formula for fungal pathogens.

Abstract:Biofilms are composed of complex multi-species in nature, potentially threatening drinking water safety. In this work, the formation of single- and multi-species fungal biofilms formed by Aspergillus niger ( A. niger ) and Aspergillus flavus ( A. flavus ), and the inactivation of mature biofilms using chlor(am)ine were firstly investigated. Results revealed that the antagonistic interaction occurred between A. niger and A. flavus . Chloramination at 20 mg/L for 30 min achieved 74.74 % and 76.04 % inactivation of A. flavus and multi-species biofilm, which were 1.69- and 1.84-fold higher than that of chlorine at the same condition. However, no significant difference was observed in the inactivation of A. niger biofilm between chlorine and monochloramine disinfection due to the lower amount of extracellular polymeric substance produced by it ( p > 0.05). The inactivation of biofilm by monochloramine fitted the Weibull model well. According to the Weibull model, the monochloramine resistance of biofilm were as follows: A. flavus > multi-species > A. niger biofilm. Besides, an increase in reactive oxygen levels, damage of cell membrane, and leakage of intracellular substances in biofilms were observed after chlor(am)ination. More intracellular polysaccharides and proteins were leaked in chloramination inactivation ( p < 0.05). This study provides important implications for controlling fungal biofilm.

Abstract:Aggregation is the primary step prior to fungal biofilm development. Understanding the attributes of aggregation is of great significance to better control the emergence of waterborne fungi. In this study, the aggregation of Aspergills spores (A. flavus and A. fumigatus) under various salt, culture medium, and humic acid (HA) conditions was investigated for the first time, and the inactivation via low-pressure ultraviolet (LPUV) upon aggregated Aspergillus spores was also presented. The aggregation efficiency and size of aggregates increased over time and at low salt (NaCl and CaCl<sub>2</sub>) concentration (10 mM) while decreasing with the continuous increase of salt concentration (100 and 200 mM). Increasing the concentration of culture medium and HA promoted the aggregation of fungal spores. Spores became hydrated, swelled, and secreted more viscous substances during the growth period, which accelerated the aggregation process. Results also suggested that fungal spores aggregated more easily in actual water, posing a high risk of biohazard in real-life scenarios. Inactivation efficiency by LPUV decreased with higher aggregation degrees due to the protection from the damaged spores on the outer layer and the shielding of pigments in the cell wall. Compared to chlorine-based disinfection, the aggregation resulted in the extension of shoulder length yet neglectable change of inactivation rate constant under LPUV treatment. Further investigation of cell membrane integrity and intracellular reactive oxygen species was conducted to elucidate the difference in mechanisms between various techniques. This study provides insight into the understanding and controlling of the aggregation of fungal spores.
