Abstract:Agriotes ustulatus is an economically important click beetle in Europe. A female-produced pheromone, (E,E)-farnesyl acetate, has been identified and is used for monitoring and detecting males. More recently, a floral lure targeting females with modest, but significant, activity has been described. Based on preliminary data, we hypothesized, that similar to the effects on the congeneric A. brevis, addition of the pheromone to the floral lure should improve female A. ustulatus catches. Also, as click beetles have been reported to respond to white light, we studied possible interactions between visual and chemical cues. In field trials, the addition of the synthetic pheromone to the floral lure resulted in a dramatic increase in the number of females trapped, whereas male catches remained unaffected and equal to those in traps baited with pheromone only. A white visual cue did not influence trap catches. Maximum catches of both sexes of A. ustulatus can be achieved using the pheromone and the floral lure inside the same trap. Furthermore, the compounds can be formulated in a single polyethylene bag dispenser, making handling of the trap easier. Due to a much larger proportion of females in the catch, this improved trap may be a promising tool for semiochemical-based, environmentally sound agricultural practice against this important pest.
Behavioural and Electrophysiological Response of Anastrepha fraterculus (Diptera: Tephritidae) to a γ-Lactone Synthetic Semiochemical

Abstract:Attractants are a powerful tool for pest management. The lack of specific attractants for the South American fruit fly, Anastrepha fraterculus, a complex of cryptic species of great economic importance in South America, makes it difficult to monitor the pest in the field. The γ-lactone male sex and aggregation pheromones of several Anastrepha species, naturally released in a 7:3 epianastrephin to anastrephin ratio, and a structurally related naturally occurring γ-lactone ((±)-trans-tetrahydroactinidiolide) with gem-dimethyl groups (dimethyl) at C(4), were evaluated as potential attractants of this species. Different age and mating conditions of A. fraterculus males and females were evaluated during electroantennography (EAG) and field cage experiments in which polymeric lures were deployed to contain 100 mg of attractant. Epianastrephin and dimethyl were EAG+ for all fly conditions, with epianastrephin eliciting the highest response for both sexes and immature flies showing greater responsiveness than mature flies. In the field cage experiments, immature flies were only attracted to leks; virgin females were attracted to leks, dimethyl, and both epianastrephin-anastrephin formulations (95 and 70 wt.% epianastrephin); mature-mated males were attracted to leks, dimethyl and 70 wt.% epianastrephin; and mature-mated females were only attracted to leks. Our bioassays showed a promising performance of the analog dimethyl since it elicited the same response as epianastrephin, requires fewer steps to synthesize, and contains one less chiral center than the natural pheromones. The attraction to leks was recorded for all mating conditions and ages of flies and suggests that air-borne volatiles of calling males contain cues that could act as sensory traps. The addition of any of these compounds in the synthetic attractants may result in a greater attraction and thus deserves further evaluation. Dose-response experiments will provide additional information to move a step forward and validate the results obtained in open-field conditions.

Abstract:Abstract The white-spotted flower chafer (WSFC), Protaetia brevit arsis Lewis, is native to East Asia. Although their larvae are considered a potential resource insect for degrading plant residues, producing protein fodder, and processing to traditional medicine, adult WSFCs inflict damage to dozens of fruit and economic crops. The control of the WSFC still relies heavily on pesticides and the inefficient manual extraction of adults. Here, we report the identification and evaluation of the aggregation pheromone of WSFCs. From the headspace volatiles emitted from WSFC adults, anisole, 4-methylanisole, 2-heptanone and 2-nonanone were identified as WSFC-specific components. However, only anisole and 4-methylanisole elicited positive dose–response relationship in electroantennography tests, and only 4-methylanisole significantly attracted WSFCs of both sexes in olfactometer bioassays and field experiments. These results concluded that 4-methylanisole is the aggregation pheromone of WSFCs. Furthermore, we developed polyethylene vials as long-term dispensers of 4-methylanisole to attract and kill WSFCs. The polyethylene vial lures could effectively attracted WSFCs for more than four weeks. Pheromone-based lures can be developed as an environmentally friendly protocol for monitoring and controlling WSFC adults.

Abstract:Dung beetles are economically important beneficial insects that process dung. To locate this source, they use volatile organic compounds (VOCs). The objectives of the study were to evaluate the attractiveness of ten electrophysiologically-active dung volatiles (phenol, skatole, indole, p -cresol, butanone, butyric acid, eucalyptol, dimethyl sulphide, dimethyl disulphide, and toluene) to dung beetles in the field and to investigate how the composition of volatile blends influences efficacy as lures for use in traps. Six combinations of the compounds were compared with field collected cattle dung bait and a negative control, across three seasons. Both dung and synthetic baits captured all exotic dung beetle species present in the study area. A six-compound mix (M1), comprising major dung volatiles, served as an attractive chemical mixture. The addition of dimethyl sulphide, dimethyl disulphide (M2) and toluene (M4) enhanced attractancy of M1 for dung beetles, while eucalyptol (M3) decreased the attractancy. The degree of attraction by various dung beetle species to synthetic baits varied, but baits proved to be effective, especially for summer trapping. The trap design used in this study presented a convenient and practical way to sample dung beetle and other associated scarabs from open pastures. The attraction of introduced dung beetle species to synthetic baits is documented here for the first time in Australia. In addition, necrophagous Omorgus sp. is reported here for the first time to be attracted to synthetic baits. They showed a significant attraction to the mixture containing dimethyl sulphide and dimethyl disulphide (M2). The current study represents a promising first step towards formulating a synthetic chemical lure for dung beetles, offering a consistent, standardised, and bio-secure trapping method compared to use of naturally occurring dung baits, especially as a multi-species lure.

Optimization of 13-tetradecenyl acetate sex pheromone for trapping Melanotus communis (Coleoptera: Elateridae)

Abstract:Corn wireworm, Melanotus communis Gyllenhal (Coleoptera: Elateridae), is an economically important larval pest of root and tuber crops in the United States. Previous work to estimate field-level abundance of M. communis has focused on grain-based larval baits placed in soil. However, this sampling method is labor intensive and may not estimate population size accurately. Recent discovery of the M. communis sex pheromone, 13-tetradecenyl acetate, provides a new method to monitor this pest during the adult stage. Early studies with this pheromone showed that different trapping methods might enhance catch and improve trap servicing. We hypothesized that placing lures on elevated traps would increase M. communis capture relative to the in-ground pitfall trapping that is currently used. We had 2 objectives for this study: (a) to compare pheromone captures among in-ground pitfall traps, on-ground pitfalls, elevated pitfalls (1 m), or elevated sticky cards (1 m) and (b) test lure longevity by aging the lures outdoors at 8-, 6-, 4-, 2-, and 0-wk intervals prior to trap deployment in the field. Experiments were conducted in North Carolina, Virginia, South Carolina, and Florida during the 2021 and 2022 field seasons. Results highlight large variation in M. communis abundance across the 4 states. We showed that 1 m elevated pheromone traps caught the most beetles. The age of the lure prior to deployment had a significant effect on trap catch. The lures that were aged for fewer weeks attracted significantly more beetles, with 0- and 2-wk-old lures capturing the greatest numbers.

Abstract:Abstract Sustainable, low impact control methods, including mating disruption and microbial insecticides against L. botrana have been available for decades. Yet, successful implementation has been restricted to only a few grapevine districts in the world. A limiting factor is the lack of a female attractant to either monitor or control the damaging sex. Volatile attractants for both female and male insects can be used to assess when L. botrana populations exceed economic thresholds, and to decrease the use of synthetic pesticides within both conventional and pheromone programs. Rather than using host-plant volatiles, which are readily masked by background volatiles released by the main crop, we tested the attractiveness of volatiles that signify microbial breakdown and more likely stand out against the background odour. A two-component blend of 2-phenylethanol (2-PET) and acetic acid (AA) caught significant numbers of both sexes. Catches increased with AA and, to a minimal extent, 2-PET loads. However, a higher load of 2-PET also increased bycatches, especially of Lepidoptera and Neuroptera. Major (ethanol, ethyl acetate, 3-methyl-1-butanol) or minor (esters, aldehydes, alcohols and a ketone) fermentation volatiles, did surprisingly not improve the attraction of L. botrana compared to the binary blend of 2-PET and AA alone, but strongly increased bycatches. The most attractive lure may thus not be the best choice in terms of specificity. We suggest that future research papers always disclose all bycatches to permit evaluation of lures in terms of sustainability.

Abstract:The citrus long-horned beetle, Anoplophora chinensis (Coleoptera: Cerambycidae), is a highly polyphagous species native to eastern and southeastern Asia. Detection of these beetles is dependent on visual surveys, which are inefficient and labor-intensive. The identification and development of pheromone-based lures would help improve survey efforts for A. chinensis should it become established within the US. We identified three antennally active male-produced chemical components (nonanal, 4-(n-heptyloxy)butan-1-ol, and 4-(n-heptyloxy)butanal), which were then tested for behavioral activity at our USDA Quarantine laboratory. When tested together in Y-tube behavioral assays, a three-component blend of the identified compounds was shown to be attractive to adult female A. chinensis. Pheromone production and behavior of A. chinensis and other closely related Anoplophora cerambycid species are discussed in relation to mate finding and potential monitoring.

Abstract:AbstractLasioderma serricorne, also known as cigarette beetle, can exploit a wide variety of stored materials as foods, but it is particularly common on tobacco and herbs. This beetle is a dominant pest species of stored Chinese medicinal materials (CMMs) causing high economic damages, making effective control strategies urgently needed. Behavioural manipulation is an important component of Integrated Pest Management. To the best of our knowledge, plant-borne volatile organic compounds (VOCs) have never been explored to develop lures for managing L. serricorne. In this study, the behavioural responses of L. serricorne to VOCs from four selected CMMs (Euphorbia kansui, Aconitum carmichaelii, Eucommia ulmoides and Pinellia ternata) were studied and their components analysed. Then, the olfactory responses of L. serricorne to the most abundant VOC identified in the preferred CMM, i.e., paeonal, was tested. L. serricorne showed significant differences in its preferences for the VOCs from the four CMMs, i.e, E. kansui > A. carmichaelii > E. ulmoides > P. ternata. From the VOCs of E. kansui, A. carmichaelii, E. ulmoides, and P. ternata, 77, 74, 56, and 81 molecules, were identified, respectively. Paeonal (23.5%), junipene (17.2%), hexanal (17.1%), and benzeneacetonitrile (14.0%) were the most abundant, respectively. Since paeonal dominated the VOC spectrum of the most preferred CMM, this compound was selected for further studies. L. serricorne showed significant positive responses to paeonal tested at various doses, with the most attractive ones being 100 μg and 500 μg. Our findings shed light on the olfactory cues routing the food searching behaviour in the cigarette beetle, providing important information on how L. serricorne targets particular CMMs. The high attractiveness of paeonal at low doses tested here may be exploited further to develop novel monitoring and control tools (e.g., lure-and-kill strategies) against this important stored product pest.

Integrating Early Palliative Care for Patients With HIV

Abstract:Increasingly clinicians are using palliative care to address the symptomatic and psychosocial effects of disease often missed by routine clinical care, termed “early” palliative care. Within an inner-city medical center, we began a program to integrate early palliative care into HIV inpatient care. Patient symptom burden and desired services were assessed and compared to provider perceptions of patient’s needs. From 2010-2012, 10 patients, with a median CD4+ T-cell count of 32.5 cells/μL, and 34 providers completed the survey. Providers ranked their patients’ fatigue, sadness, anxiety, sexual dysfunction, and body image significantly higher than patients it for themselves. Patients ranked medical care, pharmacy, social work, physical therapy, and housing as significantly more important to them than providers estimated them to be. These differences may reflect the fact that physicians often overlook patients’ unmet basic needs. Early palliative care may narrow this gap between providers’ and patients’ perceptions of needs through good communication and targeting barriers, such as housing instability, which are vital to overcome for consistent long-term follow up.

Abstract:The pine sawyer beetle (Monochamus galloprovincialis) is a European vector of the invasive pinewood nematode (Bursaphelenchus xylophilus), one of the most dangerous threats to pine forests worldwide. One recommended method for detecting the nematode is catching and inspecting the M. galloprovincialis beetles. Effective commercial lures are available for that insect but attract non‐target species, particularly beneficial Thanasimus species. Our study aimed to develop an equally effective M. galloprovincialis lure less attractive to beneficial predatory insects. We used electroantennography to test several compounds occurring in the pheromones, hindgut extracts or headspace emissions of non‐Ips bark beetles. The compounds that caused the strongest antennal responses were used along with α‐pinene (host‐tree volatile) and monochamol (2‐undecyloxy‐1‐ethanol, a male‐produced aggregation pheromone of M. galloprovincialis) to prepare the lures for olfactometric and field testing. In the field experiments, the lure containing (+)‐α‐pinene, monochamol, (1R)‐(−)‐myrtenal and (S)‐(−)‐trans‐verbenol was equally effective as the commercial lure Galloprotect Pack. It attracted significantly less Thanasimus formicarius and T. femoralis beetles. Thus, the new lure can serve as a basis for developing an operational lure for monitoring M. galloprovincialis with reduced bycatch of predatory beetles.

Abstract:The grey corn weevil, Tanymecus (Episomecus) dilaticollis, is an economically important polyphagous pest in Eastern and Central Europe. The present research is the first published electrophysiological study of olfactory sensitivity of adult T. dilaticollis to plant volatiles. The electrophysiological responses of male and female antennae were recorded to 20 synthetic volatile organic compounds (VOCs), previously identified to be released from the preferred host plants, maize and sunflower. Antennae of both sexes responded to a wide range of volatiles from different chemical classes: fatty acid derivatives, aromatic compounds and terpenoids. Two green leaf volatiles, (E)-2-hexen-1-ol and (Z)-3-hexen-1-ol, and one terpenoid, (±)-linalool, elicited the strongest electrophysiological responses by male and female antennae. Relatively high electrophysiological activity by T. dilaticollis antennae was also recorded to benzaldehyde, eugenol and caryophyllene. In the field, (E)-2-hexen-1-ol and (Z)-3-hexen-1-ol, when presented individually, caught significantly more male and female T. dilaticollis than unbaited controls. These compounds could be developed into an attractive trap for the detection and monitoring of T. dilaticollis.

Identification of sex attractants for 6 North American click beetle species in 4 tribes of the Elateridae

Abstract:We describe experiments that evaluated potential sex pheromone components for 6 North American click beetle species. In field trials in Illinois, South Carolina, North Carolina, and Virginia, male beetles of 6 species were strongly attracted to geranyl butyrate (Agriotes insanus Candèze), 5-methylhexyl (Z)-4-decenoate (Elater abruptus Say), 11-dodecenyl butyrate (Melanotus ignobilis Melsheimer), and limoniic acid (Gambrinus griseus [Palisot de Beauvois], G. rudis [Brown], and G. plebejus [Say]). Collection of E. abruptus in South Carolina represents a new state record. For each species, capture rates varied geographically and temporally, possibly due to differences in local population densities and regional phenology, or the efficiency of different trap designs or trap placement. Structural similarities were observed between the tested attractants and previously identified pheromones of closely related species. For example, males of A. insanus were attracted to geranyl butyrate, analogous to the terpenoid ester pheromones of a number of European congeners. The attractant for E. abruptus, 5-methylhexyl (Z)-4-decenoate, is an analog of the pheromone of its European congener E. ferrugineus L., and the attractant for M. ignobilis, 11-dodecenyl butyrate, is an analog of attractants of European and Asian congeners. Attraction of the 3 Gambrinus species to limoniic acid parallels recent reports of attraction of the congeners G. seminudus (Van Dyke) and G. ursinus (Van Dyke) to this compound, which was originally identified from closely related species of Limonius. Full identifications of additional sex pheromones for elaterid species should provide a more complete picture of the diversities/similarities of the semiochemicals mediating reproductive behaviors of this biologically diverse and taxonomically complex group.
Bioactive Male-Produced Volatiles from Anastrepha obliqua and their Role in Attraction of Conspecific Females

Abstract:The study of insect semiochemicals, especially pheromones, is of fundamental importance for the development of strategies for controlling agricultural pests. In this study, volatile compounds involved in the communication between males and females of the fruit fly, Anastrepha obliqua (Diptera: Tephritidae), for mating purposes were characterized to develop attractant formulations for females of this species. Extracts containing volatile compounds released by males of A. obliqua were obtained by the dynamic headspace technique and analyzed by gas chromatography coupled with an electroantennographic detector (GC-EAD) and gas chromatography coupled with mass spectrometry (GC-MS). Twenty-one volatile compounds were identified in the aeration extracts of males. Five of them caused EAD responses from the antennae of females: 1-heptanol, linalool, (Z)-3-nonen-1-ol, (E,Z)-3,6-nonadien-1-ol, and (Z,E)-α-farnesene. Six synthetic mixtures of these compounds, including the five-component blend and all possible four-component blends, were formulated in a biopolymer and used in behavioral bioassays conducted in the laboratory arena with conspecific virgin females. One blend of 1-heptanol, linalool, (Z)-3-nonen-1-ol, and (Z,E)-α-farnesene attracted more females than the collection of volatiles from virgin males used as control. The other mixtures were as attractive to A. obliqua females as the control treatment. This study indicates potential for use of these compounds in monitoring and control strategies for this pest.

Effects of volatile compounds emitted by Protea species (Proteaceae) on antennal electrophysiological responses and attraction of cetoniine beetles

Abstract:Evolutionary shifts in pollination systems within a plant genus are commonly associated with changes in floral scent, reflecting selection mediated through the sensory systems of various pollinators. The most common cetoniine beetle pollinator of grassland Protea species in South Africa, Atrichelaphinis tigrina, previously has been shown to have a strong preference for the fruity floral scent of these plants over the weak scent of their bird-pollinated congeners. However, it is not known which of the many compounds found in the scent of beetle pollinated Protea species play a role for pollinator attraction. Electroantennograms (EAG) from A. tigrina beetles were recorded in response to 15 compounds emitted by Protea flower heads. EAG responses to all 15 compounds were significantly greater than those to the paraffin solvent in which they were diluted. The greatest responses were observed for aromatics (anisole, methyl benzoate, methyl salicylate, benzaldehyde) followed by the monoterpene β-linalool, which can comprise up to 66 % of fruity Protea scents. Five compounds that elicited EAG responses (benzaldehyde, β-linalool, (E/Z)-linalool oxide (furanoid), methyl benzoate, and methyl salicylate) were tested in commercially available yellow bucket traps in the field to test their attractiveness to beetles. Traps baited with methyl benzoate, β-linalool, (E/Z)-linalool oxide (furanoid), and methyl salicylate caught significantly more insects than did those containing paraffin only. Methyl benzoate also was more specifically attractive to A. tigrina than was (E/Z)-linalool oxide (furanoid) and paraffin baited controls. A second field experiment using a combination of linalool vs. paraffin baited yellow or green traps showed that trap color had a significant effect on the number of trapped beetles. Yellow traps yielded a ten-fold higher number of insect catches than did green traps. However, the combination of yellow color and the scent compound linalool yielded the highest number of catches. This study has shown that the cetoniine beetle A. tigrina can detect a variety of floral compounds and is attracted to compounds comprising a large proportion of the blend that makes up fruity Protea scents, adding support for the hypothesis that change in scent chemistry during the shift from bird to cetoniine beetle pollination in this genus were mediated by beetle sensory preferences.

Sex Pheromone of the Alfalfa Plant Bug, Adelphocoris lineolatus: Pheromone Composition and Antagonistic Effect of 1-Hexanol (Hemiptera: Miridae)

Abstract:Abstract The sex pheromone composition of alfalfa plant bugs, Adelphocoris lineolatus (Goeze), from Central Europe was investigated to test the hypothesis that insect species across a wide geographical area can vary in pheromone composition. Potential interactions between the pheromone and a known attractant, ( E )-cinnamaldehyde, were also assessed. Coupled gas chromatography-electroantennography (GC-EAG) using male antennae and volatile extracts collected from females, previously shown to attract males in field experiments, revealed the presence of three physiologically active compounds. These were identified by coupled GC/mass spectrometry (GC/MS) and peak enhancement as hexyl butyrate, ( E )-2-hexenyl butyrate and ( E )-4-oxo-2-hexenal. A ternary blend of these compounds in a 5.4:9.0:1.0 ratio attracted male A. lineolatus in field trials in Hungary. Omission of either ( E )-2-hexenyl-butyrate or ( E )-4-oxo-2-hexenal from the ternary blend or substitution of ( E )-4-oxo-2-hexenal by ( E )-2-hexenal resulted in loss of activity. These results indicate that this Central European population is similar in pheromone composition to that previously reported for an East Asian population. Interestingly, another EAG-active compound, 1-hexanol, was also present in female extract. When 1-hexanol was tested in combination with the ternary pheromone blend, male catches were reduced. This compound showed a dose-response effect with small doses showing a strong behavioral effect, suggesting that 1-hexanol may act as a sex pheromone antagonist in A. lineolatus . Furthermore, when ( E )-cinnamaldehyde was field tested in combination with the sex pheromone, there was no increase in male catch, but the combination attracted both males and females. Prospects for practical application are discussed.

Abstract:The spotted asparagus beetle, Crioceris duodecimpunctata (L.) is an invasive host-specific pest of asparagus cultivations. To contribute to the understanding of the role of plant volatiles in host-finding by this species, behavioural and electrophysiological tests were carried out. Y-tube olfactometer bioassays, testing intact or mechanically-damaged cladophylls vs. clean air, revealed sexually-dimorphic responses with males being the only sex attracted to both plant materials. Electroantennographic (EAG) assays showed that antennae of both sexes can perceive a wide range of asparagus volatiles. Male and female EAG profiles were almost similar and (Z)-3-hexen-1-ol was by far the most EAG-active compound. (E)-2-hexenal, (±)-linalool, and 3-heptanone elicited the strongest EAG amplitude within the corresponding chemical groups. Eight of the most EAG-active compounds elicited dose-dependent responses indicating the sensitivity of male and female olfactory systems to changes in stimulus concentration. In a Y-tube olfactometer bioassay, (Z)-3-hexen-1-ol at the doses of 1, 10, and 50 μg did not elicit female attraction whereas a significant attraction at the 10 μg dose and a repellent effect at the 50 μg dose was induced in males. Sexual dimorphism of male behavioural response to host plant volatiles is discussed. This study provides a basis for future investigations that could contribute to the development of semiochemical-based monitoring and management strategies for this pest.

Abstract:Chemosensory proteins (CSPs) are involved in the earliest steps of the olfactory process by binding and transporting odorants and play a crucial role in the insect's search for food and egg-laying sites. In the present study, the tissue expression profiles showed that both CchiCSP3 and CchiCSP5 of Callosobruchus chinensis were highly expressed in the adult antennae. Subsequently, the recombinant CchiCSP3 and CchiCSP5 proteins were analysed using fluorescence competitive binding assays, and both showed binding affinities for the three mung bean volatiles. Molecular docking and site-directed mutagenesis revealed four key amino acid residues in CchiCSP3 (L47, W80, Y81, and L84) and CchiCSP5 (Y28, K46, L49, and I72). Electroantennogram (EAG) and dual-choice biobehavioral assays showed that the antennae of adult C. chinensis were electrophysiologically active in response to stimulation with all three behaviorally active compounds and that octyl 4-methoxycinnamate and β-ionone had a significant luring effect on adult C. chinensis , whereas vanillin had a significant avoidance effect. Our study screened three effective behaviorally active compounds based on the involvement of two CchiCSPs in the recognition of mung bean volatiles, providing an opportunity to develop an alternative control strategy using behavioral disruptors to limit the impact of pests.

Abstract:Abstract Plant volatiles, constitutive or induced, are perceived by insect pests to locate the host plant and also by natural enemies to locate the host insect. These plant volatiles can be utilized to develop attractive or repellant lures for pest management. Studies were carried out to identify the plant volatiles which are induced in pigeonpea, Cajanus cajan (L.) Huth due to the herbivory of blister beetle, Mylabris pustulata . The volatiles from healthy uninfested pigeonpea plants and blister beetle-infested plants were collected using the dynamic headspace collection method with Porapaq Q (80-100 mesh) as adsorbent. Using gas chromatography-mass spectrometry, 28 compounds were identified from uninfested pigeonpea plants whereas 16 compounds were identified from infested plants. A qualitative analysis showed that α -pinene and 3-hexen-2-one were exclusively detected in infested plants and (Z)-3-hexen-1-yl acetate and acetophenone were released in higher quantities from infested plants than from uninfested one. Electrophysiological evaluation of these volatiles along with other plant volatiles showed that blister beetle antennae eliciting higher responses to eucalyptol at a 1- μ g dose, to nerol at a 10- μ g dose and to benzyl acetate at 100- and 1000- μ g doses. Beetle traps with lures of eucalyptol, benzaldehyde, benzyl acetate, and nerol attracted a very small number of blister beetle adults, suggesting the need for further efforts to standardize lure load and trap design.

Behavioral responses of Araecerus fasciculatus (Coleoptera: Anthribidae) to volatiles of selected stored Chinese medicinal plant products

Abstract:The olfactory responses of Araecerus fasciculatus Degeer to the volatiles of 5 traditional Chinese medicine plant products (CMPPs; i.e., Codonopsis pilosula Franch., Ophiopogon japonicus Linn. f., Astragalus membranaceus Fisch., Dendrobium nobile Lindl., and Angelica dahurica Fisch.) were studied using Y-tube, 4-arm, and 6-arm olfactometers. The volatile components of these CMPPs were analyzed using gas chromatography-mass spectrometry (GC-MS). A. fasciculatus significantly preferred the CMPPs volatiles in the order of O. japonicus > C. pilosula > A. membranaceus ≥ D. nobile = A. dahurica. In the volatile profiles of O. japonicus, C. pilosula, A. membranaceus, D. nobile, and A. dahurica, GC-MS identified 27, 61, 63, 52, and 71 components, respectively, and β-elemene (30.58%), hexanal (12.69%), selina-3,7(11)-diene (12.31%), d-limonene (18.59%), and α-curcumene (8.88%) were the most abundant volatile components. A. fasciculatus were attracted to β-elemene, α-selinene, and β-selinene (main components of the volatile profiles of the most preferred CMPPs of O. japonicus) at different concentrations, with 1, 50, and 25 µg/µl being the most attractive, respectively. The olfactory preferences of A. fasciculatus were β-elemene > α-selinene = β-selinene based on the 4-arm olfactometer bioassays for comparisons at their most attractive concentrations. Therefore, differences in the volatile profiles among CMPPs significantly influenced the olfactory responses of A. fasciculatus, offering insight into the mechanisms of host preferences in stored-product pests based on chemical ecology. β-elemene showed the greatest potential to be developed as an attractant for the monitoring and control of this pest.
