Abstract:The “off” response of the human electroretinogram has been shown to resemble an inverted “on” response. The off response can only be obtained from a strongly light-adapted eye. The curve of response amplitude against length of stimulus flash passes through a maximum for flash durations of 12–20 msec, depending on the intensity of adaptation and stimulation. The curve of response amplitude against interval between two very short light flashes passes through a minimum for flash intervals of 15–25 msec under similar conditions of adaptation and stimulation. Algebraic addition of on and off responses or of two on responses has been shown to provide an adequate explanation of the two curves.

Abstract:Ethnopharmacological relevance Gastrodia elata Blume is a traditional Chinese medicine with the effects of improving the deficiency of the body and maintaining health, and polysaccharide (GEP) is one of the effective ingredients to play these activities of G. elata . Traditionally, G. elata is orally administered, so the activities of GEP are associated with digestive and intestinal metabolism. However, the digestive behavior of GEP and its effects on the human gut microbiota are unclear and need to be fully studied. Aim of the study This study aimed to investigate the changes in structural characteristics of GEP during digestion and the related impacts of its digestive product on gut microbiota in human fecal fermentation, and to explain the beneficial mechanism of GEP on human health from the perspective of digestive characteristics and "gut" axis. Materials and methods The changes of reducing sugars, free monosaccharides and physicochemical properties of GEP during digestion were investigated by GPC, HPLC, FT-IR, CD, NMR, SEM, and TGA. Moreover, polysaccharide consumption, pH value changes, SCFAs production, and changes in gut microbiota during fermentation were also discussed. Results During digestion of GEP, glucose was partially released causing a decrease in molecular weight, and a change in monosaccharide composition. In addition, the characteristics of GEP before and after digestion, including configuration, morphology, and stability, were different. The digestive product of GEP was polysaccharide (GEP-I), which actively participated in the fecal fermentation process. As the fermentation time increased, the utilization of GEP-I by the microbiota gradually increased. The abundance of probiotics such as Bifidobacterium , Collinsella , Prevotella , and Faecalibacterium was significantly increased, and the abundance of pathogenic Shigella , Dorea , Desulfovibrio , and Blautia was significantly inhibited, thereby suggesting that GEP has the potential to maintain human health through the "gut" axis. In addition, the beneficial health effects of GEP-I have also been observed in the influence of microbial metabolites. During the fermentation of GEP-I, the pH value gradually decreased, and the contents of beneficial metabolites such as acetic acid, propionic acid, and caproic acid significantly increased. Conclusion The structure of GEP changed significantly during digestion, and its digestive product had the potential to maintain human health by regulating gut microbiota, which may be one of the active mechanisms of GEP.

Intraarticular hip treatment with triamcinolonehexacetonide in juvenile chronic arthritis.

Abstract:To evaluate the effect and tolerance of intraarticular Triamcinolonehexacetonide (TCH) in the course of chronic coxitis in juvenile chronic arthritis (JCA) in an open uncontrolled study. Since 1990 we treated patients < 16 years of age suffering from chronic coxitis with 1 mg/TCH/KG body weight. The patients were checked again 4-8 weeks after the treatment. Clinical and ultrasound courses were recorded with the help of ultrasound and joint scores. The evaluation took place 6, 12, 24 and 36 months after the treatment. At that time we give a report on the 12 months follow-up of 37 hip joints and the 24 months follow-up of 20 hip joints. The immediate effect of TCH influencing mobility, pain sensitivity and joint effusions of the patients is impressing. The long term effect of TCH has to be evaluated by regular check ups for at least 2 years. An individual comparison with the not treated contralateral joint would be desired if ethically justifiable. Most patients suffering from polyarticular diseases with a long course of coxitis needed more than one injection of TCH (mean reinjection time 5.8 months). Any avascular necrosis of the femoral heads or other complications were not observed.

Abstract:To discover anti-pancreatic cancer cells growth inhibitor, a new water-soluble glucan, WTMA, was isolated from the rhizome of Gastrodia elate Bl. The mean molecular weight of the polysaccharide was estimated to be 7.0 x 10(5) Da by high performance gel permeation chromatography (HPGPC). The structure of WTMA was characterized by GC, GC-MS, NMR and MALDI-TOF. WTMA was deduced as an alpha-(1 --> 4)-glucan with alpha-(1 --> 4) linked branches attached to O-6 at branch points. Bioactivity tests in vitro indicated that WTMA and its partial acid hydrolysis-derived mixture product, WTMA-AD-O, could significantly inhibit the proliferation of pancreatic cancer cells. However, WTMA-DE, the degraded polysaccharide without branch chain by isoamylase, did not inhibit the proliferation of pancreatic cancer cells. Thus side chain on WTMA was deduced to be required for the anti-cancer cell activity. Further study demonstrated that WTMA and all of its hydrolysis products had no inhibition effect on liver LO2 cells. (C) 2011 Elsevier Ltd. All rights reserved.

Abstract:The critical current of Nb/sub 3/Sn strands used in Fermilab's high field magnets was measured at low and high fields under various experimental conditions using the voltage-current (V-I) and voltage-field (V-H) methods. The strands were produced using Modified Jelly Roll, Restacked Rod Process and Powder-in-Tube technologies. V-I characteristics at fields above 10-12 T with a smooth transition from the superconducting to normal phase allowed determining strand critical current. V-I and V-H measurements at low fields showed premature quenches due to magnetic instability in strands with high critical current density and large effective filament size. The results of the strand critical current measurements at low and high fields are herein presented.
Structural characterization of the glucan from Gastrodia elata Blume and its ameliorative effect on DSS-induced colitis in mice

Abstract:The polysaccharide glucan was extracted from Gastrodia elata Blume, and its structural characterizations and beneficial effects against acute dextran sulfate sodium (DSS)-induced ulcerative colitis were investigated. The results showed that a polysaccharide GP with a molecular weight of 811.0 kDa was isolated from G. elata Blume. It had a backbone of α-D-1,4-linked glucan with branches of α-d-glucose linked to the C-6 position. GP exhibited protective effects against DSS-induced ulcerative colitis, and reflected in ameliorating weight loss and pathological damages in mice, increasing colon length, inhibiting the expression of inflammatory cytokines tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β), decreasing the levels of inflammatory related proteins NLRP3 and ASC, and elevating the anti-inflammatory cytokine interleukin-10 (IL-10) level in mouse colon tissues. GP supplementation also reinforced the intestinal barrier by promoting the expression of ZO-1, Occludin, and MUC2 of colon tissues, and positively regulated intestinal microbiota. Thus, GP treatment possessed a significant improvement in ulcerative colitis in mice, and it was expected to be developed as a functional food.

Abstract:Peach gum polysaccharides have multiple biological functions, such as anti-oxidation, blood lipids reduction, and immune protection. However, their impact on the gut microbiota was still unclear. Here, high throughput sequencing was used to study the regulatory effects of peach gum polysaccharide on gut microbiota using an in vitro fermentation model of human gut microbiota. The results showed that gut microbiota species increased significantly and α-diversity index (Sobs, Ace, Chao1, Shannon) decreased significantly after polysaccharide fermentation compared with control group (distilled water). At phylum level, the relative abundance of Firmicutes and Actinomyces increased (61.76% vs. 56.64%, 28.24% vs. 23.11%). In contrast, relative abundance of Bacteroides decreased significantly (8.10% vs. 19.21%). At genus level, the relative abundance of Bifidobacteria, Colinella and Parazobacteria increased significantly (21.43% vs. 19.08%, 6.32% vs. 3.73%, 6.37% vs. 0.00%), while the relative abundance of Bacteroides and Prevotella decreased significantly (3.83% vs. 5.52%, 3.49% vs. 12.15%). Precisely, peach gum polysaccharide can effectively regulate the structure of gut microbiota and has potential probiotic effects including anti-obesity, anti-inflammatory, maintenance of gut epithelial barrier and so on.

Abstract:UNLABELLED Only a few parameters such as tumor grade and stage are of value in prognosticating disease course in endometrial carcinoma. Biochemical steroid hormone receptor assays could also be useful but are difficult to perform and interpret. Immunocytochemical assay (ICA) might be the method of choice for detecting endometrial receptors. METHODS Frozen tissue from 78 cases of endometrial adenocarcinoma was examined for the presence of estrogen (ER) and progesterone receptors (PgR) with specific monoclonal anti-receptor antibodies and the peroxidase-antiperoxidase method. In over 60 cases, frozen tissue was also assayed for ER and PgR by biochemical means. RESULTS Fifty-five (71%) of the endometrial carcinomas were ERICA-positive and 55 (71%) PgRICA-positive. Although both ERICA and PgRICA correlated significantly with biochemical ER and PgR only ERICA was predictive of survival. A woman with a negative ERICA was 4 times more likely to die of her disease than if she were ERICA-positive (P = 0.009; mean follow-up, 37.5 months). Three cases ERICA-positive and PgRICA-negative survived while 3 others ERICA-negative and PgRICA-positive died. CONCLUSION ERICA, a technique easy to perform and interpret at the community hospital level, appears to provide prognostic information independent of tumor stage and grade. Such information might be of value in planning postoperative therapies for women with endometrial cancer.

Abstract:A polysaccharide was extracted from the fresh ginger by water extraction and alcohol precipitation. A homogeneous polysaccharide was obtained by Sevage deproteinization, dialysis, DEAE-52 ion exchange chromatography and Sephadex G-100 gel column chromatography. The molecular weight of the ginger polysaccharide was determined by high performance gel permeation chromatography. The monosaccharide composition, scanning electron microscope and other physicochemical parameters of the ginger polysaccharide were further determined. The spectral properties of the ginger polysaccharide were investigated by ultraviolet and infrared spectroscopy. On this basis, methylation analysis, 1D and 2D NMR analysis were used to investigate its possible chemical structure. The results showed that the ginger polysaccharide group was divided into neutral polysaccharide components with high homogeneity, M w = 35.52 kDa, M n = 216.56 kDa and the sugar content was 93.5%. The ginger polysaccharide is mainly composed of Fuc, Ara, Rha, Gal, Glc, Xyl, Man, Gal-UA and Glc-UA, the amount of substance ratio was 1.01:1.96:9.54:14.82:64.20:3.84:2.07:1.41:1.16. To study the structure–activity relationship of ginger polysaccharide, acetylated ginger polysaccharide and phosphate ginger polysaccharide were prepared by chemical methods in this study. The results of antioxidant activity in vitro showed that the ginger polysaccharide and its derivatives had good biological activity in freeing ABTS + radical scavenging, freeing hydroxyl radical scavenging, freeing DPPH radical scavenging and reducing ability. Interestingly, the results showed that the ginger polysaccharide had a certain inhibitory effect on the growth of E. coli in vitro. In general, this study provides a certain experimental basis and reference for the further research on the activity of ginger polysaccharide and its derivatives.

Abstract:This study investigates the optimal extraction conditions, monosaccharide composition, and antioxidant activity analysis of polysaccharides from the aerial parts of three different varietal varieties of Gastrodia (i.e., G. elata Bl. F. elata, G. elata Bl. F. Viridls MaKino , and G. elata Bl. F. Glauca S Chow ). The influence of extraction temperature (30–70 °C), extraction time (15–55 min), and liquid-to-solid ratio (25–65 mL/g) on the yield of polysaccharides was analyzed through single-factor experiments. The response surface methodology was used to optimize the extraction process, and a mathematical model was established to obtain the optimal extraction conditions. The response surface experiment was presented as follows: The optimal extraction conditions for polysaccharides were a liquid-to-solid ratio of 59 mL/g, 56 °C, and 36 min. The polysaccharide yield in G. elata. Bl. F. Glauca S Chow aerial parts under these conditions was 10.90%, which was close to the theoretical value calculated by the model (10.64%). Under the optimal conditions, the yields of polysaccharides followed: G. elata Bl. F. elata (11.32%) > G. elata Bl. F. Glauca S Chow (10.90%) > G. elata Bl. F. Viridls MalKino (10.50%). The polysaccharides were mainly composed of 10 monosaccharides, including glucose, rhamnose, mannose, and xylose. However, the content of monosaccharides in polysaccharides of different varieties of Gastrodia elata Bl aerial parts varied greatly, with glucose and rhamnose being the highest, both exceeding 20%. The polysaccharides in different varieties of Gastrodia elata Bl aerial parts had certain in vitro antioxidant activity. The total reducing power and scavenging rates of 2,2-diphenylpicrylhydrazyl (DPPH) and 2,2′-azinobis-(3-ethylbenzthiazolin-6-sulfonic acid) (ABTS) free radicals increased with the increase of polysaccharide concentration. When the polysaccharide concentration was 1.0 mg/mL, the scavenging rates of DPPH and ABTS free radicals were both over 80%. This study provides a theoretical basis for the further development and utilization of Gastrodia elata Bl aerial part resources.

Abstract:The composition, physicochemical properties, in vitro biological activity, and hypoglycemic activity exhibited by polysaccharides from four varieties of G. elata were investigated in this study; the four extracted GaE polysaccharides were termed as GaE-B (G. elata Bl. f. glauca S. chow polysaccharides), GaE-R (G. elata Bl. f. elata polysaccharides), GaE-Hyb (hybridization of G. elata Bl. f. glauca S. chow and G. elata Bl. f. elata polysaccharides), and GaE-G (G. elata Bl. f. viridis Makino polysaccharides). As revealed by the results, the GaE polysaccharides were found with the same monosaccharide composition, primarily including glucose, whereas the content of each variety was significantly different. In addition, different degrees of differences were found in the in vitro antioxidant and hypoglycemic activity, molecular weight, yield, and chemical composition exhibited by the abovementioned varieties. However, GaE-B and GaE-Hyb were found with similar physical properties, chemical composition, and antioxidant and hypoglycemic activity. GaE-R had the lowest yield, total sugar content, and molecular weight, whereas it involved higher xylose, binding protein, and polyphenols as well as higher antioxidant and hypoglycemic activity. In contrast, GaE-G was found with the highest yield, total sugar content, and molecular weight, whereas it contained the lowest xylose, binding protein, and polyphenols, as well as the weakest antioxidant and hypoglycemic activity. In brief, the polysaccharide of G. elata, a plant resource for homology of medicine and food, could more significantly enhance the biological activity of G. elata as it was released in the process of decocting and stewing. To be specific, the assessment of polysaccharide activity alone suggested that GaE-R was the best.

Abstract:Gastrodia elata Blume (G. elata), listed as one of the 34 precious Chinese medicines, servers a dual purpose as both a medicinal herb and a food source. Polysaccharide is the main active ingredient in G. elata, which has pharmacological activities such as immune regulation, anti-oxidation, anti-cancer, anti-aging, neuroprotection and antibacterial activity and so on. The biological activities of G. elata polysaccharide (GPs) is closely related to its chemical structures. However, no a review has synthetically summarized the chemical structures and pharmacological activities of GPs. This study delves into the chemical structures, pharmacological action of GPs, offering insights for the future development an application of these compounds.

Abstract:Natural polysaccharides interact with gut microbes to enhance human well-being. Grifola frondosa is a polysaccharides-rich edible and medicinal mushroom. The prebiotic potential of G. frondosa polysaccharides has been explored in recent years, however, the relationship between their various structural features and prebiotic activities is poorly understood. In this study, three homogenous polysaccharides GFP10, GFP21 and GFP22 having different molecular weights ( M w ), monosaccharide compositions and glycosidic linkages were purified from G. frondosa , and their effects on intestinal microbial composition were compared. GFP10 was a fucomannogalactan with an M w of 23.0 kDa, and it selectively inhibited Enterobacter , while GFP21 was a fucomannogalactoglucan with an M w of 18.6 kDa, and it stimulated Catenibacterium . GFP22 was a 4.9 kDa mannoglucan that selectively inhibited Klebsiella and boosted Bifidobacterium , Catenibacterium and Phascolarctobacterium , and prominently promoted the production of short-chain fatty acids (SCFAs). The selective modulation of gut microbiota by polysaccharides was structure-dependent. A relatively lower M w and a high proportion of glycosidic linkages like T-Glc p , 1,3-Glc p , 1,3,6-Glc p and 1,4-Glc p might be more easily utilized to produce SCFAs and beneficial for the proliferation of Catenibacterium and Phascolarctobacterium . This research provided a valuable resource for further exploring the structure-activity relationship and prebiotic activity of G. frondosa polysaccharides.

Abstract:A novel polysaccharide, designated GCPB-1b, was obtained from the alkaline extract of the submerged fermentation culturing mycelium powder of Boshuzhi, Ganoderma capense. This polysaccharide was purified by ion-exchange and gel-permeation column chromatography and is a glucan (MW 2847Da). GCPB-1b has a specific optical rotation of [α]D(25)=+154° (c 1.0, H2O). Based on monosaccharide analysis, partial acid hydrolysis, periodic acid oxidation-Smith degradation, methylation analysis, Fourier transform-infrared spectroscopy (FT-IR), gas chromatography-mass spectrometry (GC-MS) and nuclear magnetic resonance (NMR) spectroscopy, the backbone of GCPB-1b was composed of (1→4)-linked-α-d-glucose residues and (1→4,6)-linked-α-d-glucose residues. Furthermore, GCPB-1b had 1-diphenyl-2-picryl-hydrazyl (DPPH) radical-scavenging ability (EC50 3.23μM) according to antioxidant activity tests which was greater than other antioxidants. These data suggest that GCPB-1b holds promise as an anti-aging functional food.

Abstract:The properties of gene and allelic genealogies at a gametophytic self-incompatibility locus in plants have been investigated analytically and checked against extensive numerical simulations. It is found that, as with overdominant loci, there are two genealogical processes with markedly different time scales. First, functionally distinct allelic lines diverge on an extremely long time scale which is inversely related to the mutation rate to new alleles. These alleles show a genealogical structure which is similar, after an appropriate rescaling of time, to that described by the coalescent process for genes at a neutral locus. Second, gene copies sampled within the same functional allelic line show genealogical relationships similar to neutral gene genealogies but on a much shorter time scale, which is on the same order of magnitude as the harmonic mean of the number of gene copies within an allelic line. These results are discussed in relation to data showing trans-specific polymorphisms for alleles at the gametophytic self-incompatibility locus in the Solanaceae. It is shown that population sizes on the order of 4 x 10(5) and a mutation rate per locus per generation as high as 10(-6) could account for estimated allelic divergence times in this family.
Digestive Characteristics of Hericium erinaceus Polysaccharides and Their Positive Effects on Fecal Microbiota of Male and Female Volunteers During in vitro Fermentation

Abstract:Hericium erinaceus polysaccharides (HEPs) have attracted widespread attention in regulating gut microbiota (GM). To investigate digestibility and fermentation of HEPs and their effects on GM composition, three polysaccharide fractions, namely, HEP-30, HEP-50, and HEP-70, were fractionally precipitated with 30%, 50%, and 70% ethanol concentrations (v/v) from hot water-soluble extracts of Hericium erinaceus , respectively. Three kinds of prepared HEPs were structurally characterized and simulated gastrointestinal digestion, and their effects on human fecal microbiota fermentations of male and female and short-chain fatty acid (SCFA) production in vitro were clarified. Under digestive conditions simulating saliva, stomach, and small intestine, HEPs were not significantly influenced and safely reached the distal intestine. After 24 h of in vitro fermentation, the content of SCFAs was significantly enhanced ( p < 0.05), and the retention rates of total and reducing sugars and pH value were significantly decreased ( p < 0.05). Thus, HEPs could be utilized by GM, especially HEP-50, and enhanced the relative abundance of SCFA-producing bacteria, e.g., Bifidobacterium , Faecalibacterium , Blautia , Butyricicoccus , and Lactobacillus . Furthermore, HEPs reduced the relative abundances of opportunistic pathogenic bacteria, e.g., Escherichia-Shigella , Klebsiella , and Enterobacter . This study suggests that gradual ethanol precipitation is available for the preparation of polysaccharides from Hericium erinaceus , and the extracted polysaccharide could be developed as functional foods with great development value.

Abstract:Abstract Background: The primary genetic marker associated with coeliac disease (CD) is the HLA-DQ2 molecule, encoded by the DQA1*0501 and DQB1*0201 genes. Aim: To investigate the frequency of HLA-DQA1*0501 and DQB1*0201 alleles in Jordanian patients with CD and their first-degree relatives. Methods: An allele-specific DNA-based PCR-sequence-specific primer was used to investigate DQA1*0501 and DQB1*0201 alleles in 44 CD patients, 47 first-degree relatives and 53 healthy controls. Results: The mean age of the patients at the time of diagnosis was 13.5 years (range 1–39) and at the time of the study was 19.3 years (range 5–42). The DQA1/B1 (0501; 0201) haplotype was present in 80% of patients and 66% of first-degree relatives compared with 32% of controls (p<0.0001 and <0.01, respectively). The remaining 20% of CD patients who were negative for the DQ2 molecule carried the DQB1*0201 allele only. A statistically significant difference was detected in the DQ2 hetero-dimer frequency between CD patients, their first-degree relatives and controls with a higher frequency in the patients group (p<0.001). Conclusion: The significant differences in the frequency of DQA1*0501, DQB1*0201 alleles in CD patients and their first-degree relatives compared with the control group demonstrated the important role of these alleles in the development of CD in the Jordanian population.
