Abstract:Background: 211At-radiopharmaceuticals are currently the subject of growing studies for targeted alpha therapy of cancers, which leads to the widening of the scope of the targeting vectors, from small molecules to peptides and proteins. This has prompted, during the past decade, to a renewed interest in developing novel 211At-labelling approaches and novel prosthetic groups to address the diverse scenarios and to reach improved efficiency and robustness of procedures as well as an appropriate in vivo stability of the label. Main body: Translated from the well-known (radio)iodine chemistry, the long preferred electrophilic astatodemetallation using trialkylaryltin precursors is now complemented by new approaches using electrophilic or nucleophilic At. Alternatives to the astatoaryl moiety have been proposed to improve labelling stability, and the range of prosthetic groups available to label proteins has expanded. Conclusion: In this report, we cover the evolution of radiolabelling chemistry, from the initial strategies developed in the late 1970's to the most recent findings.

Abstract:Astatine-211 (211At) has emerged as a promising radionuclide for targeted alpha therapy of cancer by virtue of its favorable nuclear properties. However, the limited in vivo stability of 211At-labeled radiopharmaceuticals remains a major challenge. This review provides a comprehensive overview of the current strategies for 211At radiolabeling, including nucleophilic and electrophilic substitution reactions, as well as the recent advances in the development of novel bifunctional coupling agents and labeling approaches to enhance the stability of 211At-labeled compounds. The preclinical and clinical applications of 211At-labeled radiopharmaceuticals, including small molecules, peptides, and antibodies, are also discussed. Looking forward, the identification of new molecular targets, the optimization of 211At production and quality control methods, and the continued evaluation of 211At-labeled radiopharmaceuticals in preclinical and clinical settings will be the key to realizing the full potential of 211At-based targeted alpha therapy. With the growing interest and investment in this field, 211At-labeled radiopharmaceuticals are poised to play an increasingly important role in future cancer treatment.

Stability and in vivo behavior of Rh[16aneS4-diol]211 at complex: a potential precursor for astatine radiopharmaceuticals

Abstract:Introduction: The heavy halogen (211)At is of great interest for targeted radiotherapy because it decays by the emission of short-range, high-energy α-particles. However, many astatine compounds that have been synthesized are unstable in vivo, providing motivation for seeking other (211)At labeling strategies. One relatively unexplored approach is to utilize prosthetic groups based on astatinated rhodium (III) complex stabilized with a tetrathioether macrocyclic ligand - Rh[16aneS(4)-diol](211)At. The purpose of the current study was to evaluate the in vitro and in vivo stability of this complex in comparison to its iodine analog - Rh[16aneS(4)-diol](131)I. Methods: Rh[16aneS(4)-diol](211)At and Rh[16aneS(4)-diol](131)I complexes were synthesized and purified by HPLC. The stability of both complexes was evaluated in vitro by incubation in phosphate-buffered saline (PBS) and human serum at different temperatures. The in vivo behavior of the two radiohalogenated complexes was assessed by a paired-label biodistribution study in normal Balb/c mice. Results: Both complexes were synthesized in high yield and purity. Almost no degradation was observed for Rh[16aneS(4)-diol](131)I in PBS over a 72 h incubation. The astatinated analog exhibited good stability in PBS over 14 h. A slow decline in the percentage of intact complex was observed for both tracers in human serum. In the biodistribution study, retention of (211)At in most tissues was higher than that of (131)I at all time points, especially in spleen and lungs. Renal clearance of Rh[16aneS(4)-diol](211)At and Rh[16aneS(4)-diol](131)I predominated, with 84.1 ± 2.3% and 94.6 ± 0.9% of injected dose excreted via the urine at 4 h. Conclusions: The Rh[16aneS(4)-diol](211)At complex might be useful for constructing prosthetic groups for the astatination of biomolecules and further studies are planned to evaluate this possibility.

Abstract:Astatine-211 is a nuclide with a short half-life of 7.2 h, that show promise for targeted alpha therapy of disseminated cancer. Despite nuclide production being straight-forward using a medium energy cyclotron and an uncomplicated target, not many cyclotrons are currently producing the nuclide. In this work we propose a stream-lined method to produce astatine labelled antibodies that enable production of clinical doses at other sites, remote from the nuclide producing cyclotron. Preconjugating the antibody prior to labelling, quick and efficient astatine recovery from the irradiated target in combination with optimized nuclide production logistics and an efficient synthesis for labelling are all key components to produce a clinical amount, > 300 MBq, of astatinated Trastuzumab.

Abstract:Introduction: Astatine-211 has attained significant interest in the recent times as a promising radioisotope for targeted alpha therapy (TAT) of cancer. In this study, we report the production of 211At via 209Bi (α, 2n) 211At reaction in a cyclotron and development of a facile radiochemical separation procedure to isolate 211At for formulation of nanoradiopharmaceuticals. Methods: Natural bismuth oxide target in pelletized form wrapped in Al foil was irradiated with 30 MeV α-beam in an AVF cyclotron. The irradiated target was dissolved in 2 M HNO3 followed by selective precipitation of Bi as Bi(OH)3 under alkaline condition. The radiochemically separated 211At was used for labeling cyclic RGD peptide conjugated gold nanoparticles (Au-RGD NPs) by surface adsorption. The radiochemical stability of 211At-Au-RGD NPs was evaluated in phosphate buffered saline (PBS) and human serum media. Results: The batch yield of 211At at the end of irradiation was ∼6 MBq.μA-1.h-1. After radiochemical separation, ∼80 % of 211At could be retrieved with >99.9 % radionuclidic purity. Au-RGD NPs (particle size 8.4±0.8 nm) could be labeled with 211At with >99 % radiolabeling yield. The radiolabeled nanoparticles retained their integrity in PBS and human serum media over a period of 21 h. Conclusions: The present strategy simplifies 211At production in terms of purification and would increase affordable access to this radioisotope for TAT of cancer.

Abstract:We have studied the isoheight lines on the WO3 surface as a physical candidate for conformally invariant curves. We have shown that these lines are conformally invariant with the same statistics of domain walls in the critical Ising model. They belong to the family of conformal invariant curves called Schramm-Loewner evolution (or SLE(kappa)), with diffusivity of kappa approximately 3. This can be regarded as the first experimental observation of SLE curves. We have also argued that Ballistic Deposition (BD) can serve as a growth model giving rise to contours with similar statistics at large scales.

Abstract:Antimony‐119 (119Sb) holds promise for radiopharmaceutical therapy (RPT), emitting short‐range Auger and conversion electrons that can deliver cytotoxic radiation on a cellular level. While it has high promise theoretically, experimental validation is necessary for 119Sb in vivo applications. Current 119Sb production and separation methods face robustness and compatibility challenges in radiopharmaceutical synthesis. Limited progress in chelator development hampers targeted experiments with 119Sb. This review compiles literature on the toxicological, biodistribution and redox properties of Sb, along with existing Sb complexes, evaluating their suitability for radiopharmaceuticals. Sb(III) is suggested as the preferred oxidation state for radiopharmaceutical elaboration due to its stability in vivo and lack of skeletal uptake. While Sb complexes with both hard and soft donor atoms can be achieved, Sb thiol complexes offer enhanced stability and compatibility with the desired Sb(III) oxidation state. For 119Sb to find application in RPT, scientists need to make discoveries and advancements in the areas of isotope production, and radiometal chelation. This review aims to guide future research towards harnessing the therapeutic potential of 119Sb in RPT.

Abstract:Developing fast and efficient separation avenues can promote either fundamental study or practical application of astatine-211( 211 At). This work presents a new solid-phase extraction (SPE) separation strategy based on functional resin for recovering 211 At from the irradiated bismuth (Bi) target mildly and rapidly, without switching HNO 3 medium into HCl system. Four functional resins containing acyl-resin, thioether, sulfonate- and sulfonate-groups had been prepared via Friedel-Crafts acylation and alkylation reactions successfully, and subsequently characterized by FTIR, EA, SEM etc. Acyl-resin from these functional materials have shown good retention towards 211 At but low binding affinity to all possible metal impurities (Zn 2+ , Cu 2+ , Ga 3+ and Bi 3+ ) in simulated separation experiments. Static batch sorption experiments indicate that the pseudo-second-order kinetic model is more suitable for describing the binding behavior of 211 At on Acyl-resin. This reveals that chemisorption dominates the reaction between the target radionuclide and functional material. During several practical separations in medically-used scale, over 98.5 % of 211 At can be well retained on SPE column with metal impurities removed in the process of loading target solution and removing non-target radionuclides. More importantly, 60.03 %∼79.05 % of 211 At on the Acyl-resin SPE column is recoverable by NH 2 OH-NH 3 ·H 2 O-CH 3 OH elution process, and over 60 % of the radioactivity distributes in volatile organic solvent (CH 3 OH). The whole SPE separation process can be completed within 20 min, giving 211 At product with high radionuclidic purity (≥ 99 %), low metal impurities (Bi 3+ ≤ 17.37 μg and Cu 2+ ≤ 2.91 μg) and high radiolabeling availability. This work implies that acyl-groups can act as potential binding motifs not only for developing new 211 At separation methods but also designing bifunctional conjugate agents for astatinated compounds based on coordination bonds.

Abstract:Organometallics can be used as precursors for electrophilic 211At‐statinations. In this report, we compared the potential of aryl trimethylsilanes, ‐germanes, or –stannanes to be used as precursors to 211At‐label highly reactive tetrazines. Tetrazines can be used for pretargeted radioligand therapies or be applied as synthons to radiolabel rapidly and orthogonally a broad set of precursors such as peptides, mAbs or nanomedicines. All precursors could successfully be synthesized and radiolabeled. The reactivity of organogermanium reagents ranged between those of respective organotin and organosilicon precursors. Moreover, organogermanium reagents proved promising for accessing more complex and polar tetrazine scaffolds. In contrast to organotin derivatives, the use of protecting groups could be avoided for organogermanium and ‐silicon precursors. The developed 211At‐labeled tetrazines could be labeled in radiochemical conversions of 60‐90%. Organogermanium and ‐silicon precursors were clearly advantaged as additional deprotection steps could avoided. Reported labeling procedures allow astatinations of highly reactive tetrazines to be used for pretargeted approaches or to applied as highly reactive synthons to label the next‐generation of 211At‐labeled radiopharmaceuticals.

Abstract:Radioisotopes of arsenic are of considerable interest to the field of nuclear medicine with unique nuclear and chemical properties making them well-suited for use in novel theranostic radiopharmaceuticals. However, progress must still be made in the production of isotopically pure radioarsenic and in its stable conjugation to biological targeting vectors. This work presents the production and irradiation of isotopically enriched (72)Ge(m) discs in an irrigation-cooled target system allowing for the production of isotopically pure (72)As with capability on the order of 10 GBq. A radiochemical separation procedure isolated the reactive trivalent radioarsenic in a small volume buffered aqueous solution, while reclaiming (72)Ge target material. The direct thiol-labeling of a monoclonal antibody resulted in a conjugate exhibiting exceptionally poor in vivo stability in a mouse model. This prompted further investigations to alternative radioarsenic labeling strategies, including the labeling of the dithiol-containing chelator dihydrolipoic acid, and thiol-modified mesoporous silica nanoparticles (MSN-SH). Radioarsenic-labeled MSN-SH showed exceptional in vivo stability toward dearsenylation.

Abstract:The fascination with superheavy elements (SHE) spans the nuclear physics, astrophysics, and theoretical chemistry communities. Extreme relativistic effects govern these elements' chemistry and challenge the traditional notion of the periodic law. The experimental quest for SHE critically depends on theoretical predictions of these elements' properties, especially chemical homology, which allows for successful prototypical experiments with more readily available lighter homologues of SHE. This work is a comprehensive quantum-chemical investigation into astatine (At) as a non-intuitive homologue of element 113, nihonium (Nh). Combining relativistic coupled-cluster and density functional theory approaches, we model the behaviour of At and AtOH in thermochromatographic experiments on a pristine gold surface. Insights into the electronic structure of AtOH and NhOH and accurate estimates of At–gold and AtOH–gold adsorption energies rationalize recent experimental findings and justify the use of At as a chemical homologue of Nh for the successful design of future experiments on Nh detection and chemical characterization.

Abstract:PurposeAAZTA (1,4-bis (carboxymethyl)-6-[bis (carboxymethyl)]amino-6-methylperhydro-1,4-diazepine) based chelators were initially developed in the context of magnetic resonance imaging. First radiochemical studies showed the capability of AAZTA to form stable complexes with radiolanthanides and moderately stable complexes with 68Ga. For a systematic comparison of the labelling capabilities with current diagnostic and therapeutic trivalent radiometals, AAZTA5 (1,4-bis (carboxymethyl)-6-[bis (carboxymethyl)]amino-6-[pentanoic-acid]perhydro-1,4-diazepine) was synthesized representing a bifunctional version with a pentanoic acid at the carbon-6 atom. To evaluate the effect of adding a targeting vector (TV) to the bifunctional chelator on the complex formation, AAZTA5-TOC was synthesized, radiolabeled and tested in comparison to the uncoupled AAZTA5.MethodsAAZTA5 was synthesized in a 5-step synthesis. It was coupled to the cyclic peptide TOC (Phe1-Tyr3 octreotide) via amide bound formation. AAZTA and AAZTA5-TOC complex formations with 68Ga, 44Sc and 177Lu were investigated at different pH, temperature and precursor amounts. Stability studies against human serum, PBS buffer, EDTA and DTPA were performed.ResultsAAZTA5 and AAZTA5-TOC achieved quantitative labelling (> 95%) at room temperature in less than 5 min with all three nuclides at pH ranges from 4 to 5.5 with low precursor amounts of 1 to 10 nmol. [44Sc]Sc-AAZTA5 complexes as well as [44Sc]Sc-AAZTA5-TOC were completely stable. The 177Lu complexes of AAZTA5 and AAZTA5-TOC showed high stability comparable to the 44Sc complexes. In contrast, the [68Ga]Ga-AAZTA5 complex stability was rather low, but interestingly, [68Ga]Ga-AAZTA5-TOC was completely stable.ConclusionAAZTA5 appears to be a promising bifunctional chelator for 68Ga, 44Sc and 177Lu with outstanding labelling capabilities at room temperature. Complex stabilities are high in the case of 44Sc and 177Lu. While [68Ga]Ga-AAZTA complexes alone lacking stability, [68Ga]Ga-AAZTA5-TOC demonstrated high stability. The latter indicates an interesting feature of [68Ga]Ga-AAZTA5–labelled radiopharmaceuticals.
211 At‐Astatination of Arenes Using Trimethylgermanyl Precursors

Abstract:A set of 211At‐astatoarenes were synthesized from corresponding trimethylgermyl arenes with radiochemical conversions (RCCs) of up to 90%. Both electron rich and electron poor substrates were successfully radiolabeled at room temperature (RT) using relatively low precursor amounts (0.15 μmol/ 0.02 mL solvent (7.5 mM)). Ready access to ortho‐, para‐ and meta‐ astatinated arenes was achievable. Optimized reaction conditions were successfully applied to label a poly (ADP‐ribose) polymerase (PARP) inhibitor with a RCC of approx. 50%. We believe that trimethylgermanyl derivatives are a viable addition to the astatination precursor toolbox and facilitate astatination of arenes. The developed labeling method should easily be applicable for productions under good manufacturing practice (GMP).

Abstract:Abstract Astatine-211 is an attractive radionuclide for use in targeted alpha therapy of blood-borne diseases and micrometastatic diseases. Efficient isolation methods that can be adapted to robust automated 211 At isolation systems are of high interest for improving the availability of 211 At. Based on the early studies of Bochvarova and co-workers involving isolation of 211 At from irradiated thorium targets, we developed a method for 211 At isolation from bismuth targets using tellurium-packed columns. Dissolution of irradiated bismuth targets is accomplished using HNO 3 ; however, 211 At is not captured on the Te column material in this matrix. Our method involves slow addition of aqueous NH 2 OH·HCl to the Bi target dissolved in HNO 3 to convert to a HCl matrix. The amount of NH 2 OH·HCl was optimized because (1) the quantity of NH 2 OH·HCl used appears to affect the radiolabeling yield of phenethyl- closo -decaborate(2-) (B10)-conjugated antibodies and (2) reducing the volume of NH 2 OH·HCl solution can effectively shorten the overall isolation time. A proof-of-concept semi-automated process has been demonstrated using targets containing ~0.96 GBq (~26 mCi) of 211 At. High isolation yields (88–95%) were obtained. Radiochemical purity of the isolated 211 At was assessed by radio-HPLC. Concentrations of Bi and Te contaminants in the 211 At and the astatinated antibodies were evaluated using ICP-MS.

Abstract:Radiotheranostics utilizes a set of radioligands incorporating diagnostic or therapeutic radionuclides to achieve both diagnosis and therapy. Imaging probes using diagnostic radionuclides have been used for systemic cancer imaging. Integration of therapeutic radionuclides into the imaging probes serves as potent agents for radionuclide therapy. Among them, targeted alpha therapy (TAT) is a promising next-generation cancer therapy. The α-particles emitted by the radioligands used in TAT result in a high linear energy transfer over a short range, inducing substantial damage to nearby cells surrounding the binding site. Therefore, the key to successful cancer treatment with minimal side effects by TAT depends on the selective delivery of radioligands to their targets. Recently, TAT agents targeting biomolecules highly expressed in various cancer cells, such as sodium/iodide symporter, norepinephrine transporter, somatostatin receptor, α v β 3 integrin, prostate-specific membrane antigen, fibroblast-activation protein, and human epidermal growth factor receptor 2 have been developed and have made remarkable progress toward clinical application. In this review, we focus on two radionuclides, 225 Ac and 211 At, which are expected to have a wide range of applications in TAT. We also introduce recent fundamental and clinical studies of radiopharmaceuticals labeled with these radionuclides.

Abstract:Targeted alpha therapy of disseminated cancer is an emerging technique where astatine-211 is one of the most promising candidate nuclides. Astatine-211 can be produced in medium energy cyclotrons by alpha particle bombardment of natural bismuth. The produced astatine is then commonly recovered from the irradiated solid target material through dry distillation. The dry distillation process often includes elution and solvation of condensed astatine with chloroform, forming Chloroform Eluate. In this work the handling and safe use of the high activity concentration Chloroform Eluate has been investigated. Correctly performed, evaporation of Chloroform Eluate results in a dry residue with complete recovery of the astatine. The dry residue can then serve as a versatile starting material, using appropriate oxidizing or reducing conditions, for subsequent downstream chemistry. However, it has been found that when evaporating the Chloroform Eluate, astatine can be volatilized if continuing the process beyond the point of dryness. This behavior is more pronounced when the Chloroform Eluate has received a higher absorbed dose. Upon water phase contact of the Chloroform Eluate, a major part of the astatine activity becomes water soluble, leaving the organic phase. A behavior which is also dependent on dose to the solvent.

Abstract:Astatine-211 is an alpha emitter that has been identified as a good candidate for targeted alpha therapy. There is an increasing demand on this radionuclide. Very intense beam from linac being put in operation nowadays could be used to meet this demand. This document presents the design exploration of concepts of liquid bismuth targets dedicated to astatine-211 production. Three concepts are presented and analyzed: a capsule, a fluid loop and a windowless fluid loop. Structural and thermal sizing were performed using mechanical Finite Element models (ANSYS Workbench) and Computational Fluid Dynamic models (FLUENT). Production rates were assessed accordingly. Feasibility and expected performances are discussed in conclusion.
