Abstract:The production of doubled haploid (DH) plants from microspores is an important technique used in plant breeding programs and basic research. Although doubled haploidy efficiencies in wheat and barley are sufficient for breeding purposes, oat (Avena sativa L.) is considered recalcitrant. The objective of this project was to develop a protocol for the production of microspore-derived embryos of oat and further develop these embryos into fertile DH plants. A number of experiments were conducted evaluating the factors influencing microspore embryogenesis, i.e. donor plant conditions, pretreatments, media composition, and culture conditions. The initial studies yielded little response, and it was not until high microspore densities (106 microspores/mL and greater) were used that embryogenesis was achieved. Depending on the treatment, yields of over 5,000 embryos/106 microspores were obtained for breeding line 2000QiON43. The doubled haploidy protocol includes: a 0.3 M mannitol pretreatment of the tillers for 7 days, culture in W14 basal medium with a pH of 6.5–7.5, a microspore density of 106 microspores/mL, and continuous incubation at 28 °C incubation. The resulting embryos observed after 28 days were plated onto solidified W14 medium with 0.8 or 1.0 g/L activated charcoal. A colchicine treatment of 0.2 % colchicine for 4 h resulted in conversion of 80 % of the plants from haploid to DH. This protocol was successful for the production of oat microspore-derived embryos and DH green plants with minimal albinism. DH seed was produced and planted for evaluation in a field nursery.

Abstract:Wide reciprocal hybridisations between Brassica rapa (genome: AA) and Brassica nigra (genome: BB) were performed in the present study. The results indicate that use of B. rapa as female parent and B. nigra as male parent was appropriate for wide hybrid production, and allotetraploids and alloaneuploids could be induced by treating allodiploids with 0.2% colchicine for 16h and 24h. When allodiploids, T-allodiploids (allodiploids after treating with colchicine), allotetraploids and alloaneuploids were transferred to the field, the allotetraploids exhibited more vigorous growth compared to the allodiploids and T-allodiploids. Compared with their parents, the allotetraploids displayed 21.07% polymorphic fragments measured by the sequence related amplified polymorphism (SRAP) analysis, which was the double compared to the allodiploids. Moreover, the novel fragments in allotetraploids were more than twice the number of that in allodiploids. The novel fragments matched homologous functional genes when blasted against genome databases. When the allotetraploids (F1) were self-crossed, the next generation (F2) exhibited the same phenotype and genetic pattern. These results indicated that the degree of genetic changes were positively related with the polyploidy level during the process of heterogeneous chromosomes combinations and chromosome doubling, and both wide hybridisation and genome doubling could affect the genomic architecture in newly formed allopolyploids. Moreover, novel fragments may be correlated with novel phenotypes.

Abstract:The aim of any breeding process is to fully express the targeted, superior/desirable parent characteristic in the progeny. Hybrids are often used in this dynamic, and complex process for which homozygous parents-which may require up to eight generations of back crossing and selection-are required. Doubled haploid (DH) technologies can facilitate the production of true breeding lines faster and in a more efficient manner than the traditional back crossing and selection strategies. Sunflower is the third most important oilseed crop in the world and has no available double haploid induction procedure/technique that can be efficiently used in breeding programs. A reproducible and efficient doubled haploid induction method would be a valuable tool in accelerating the breeding of new elite sunflower varieties. Although several attempts have been made, the establishment of a sunflower doubled haploid induction protocol has remained a challenge owing recalcitrance to in vitro culture regeneration. Approaches for haploid development in other crops are often cultivar specific, difficult to reproduce, and rely on available tissue culture protocols-which on their own are also cultivar and/or species specific. As an out-crossing crop, the lack of a double haploid system limits sunflower breeding and associated improvement processes, thereby delaying new hybrid and trait developments. Significant molecular advances targeting genes, such as the centromeric histone 3 (CenH3) and Matrilineal (MTL) gene with CRISPR/Cas9, and the successful use of viral vectors for the delivery of CRISPR/Cas9 components into plant cells eliminating the in vitro culture bottleneck, have the potential to improve double haploid technology in sunflower. In this review, the different strategies, their challenges, and opportunities for achieving doubled haploids in sunflower are explored.

Abstract:The relation between the activity of the Na+-K+-ATPase and the metabolic source of ATP was investigated in suspensions of MDCK cells. The pump activity of Na+-K+-ATPase was estimated from the initial rate of ouabain-sensitive K+ uptake into K+-depleted cells. Uptake was initiated by the reintroduction of K+ to the medium in which the cells were suspended. The metabolic source of ATP was varied by changing the substrates supplied to the suspension. Cells respiring on glutamine produced ATP from oxidative metabolism alone, whereas cells incubated with glucose and glutamine produced ATP via glycolysis and oxidative phosphorylation. Over a wide range of extracellular K+ concentrations, the initial rate of K+ uptake was faster in cells incubated with glucose and glutamine when compared with cells incubated with glutamine alone. Kinetic analysis together with ouabain-binding data demonstrated that this increase in K+ uptake was due to an increase in maximal velocity (Vmax) at a constant number of Na+-K+-ATPase transport sites. In addition, steady-state studies revealed that the addition of glucose to K+-depleted cells respiring on glutamine alone resulted in a net ouabain-sensitive influx of K+. These data demonstrate that in MDCK cells the maximal capacity for transport via the Na+-K+-ATPase is greater when ATP is produced from both glycolysis and oxidative phosphorylation than when ATP is produced from oxidative phosphorylation alone.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:The doubled haploid technique aims to generate pure inbred lines for basic research and as commercial cultivars. The doubled haploid technique first generates haploid plants and is followed by chromosome doubling, which can be separated in time or overlapped, depending the procedure for each species. For a long time, much effort has been focused on haploid production via androgenesis, gynogenesis, or parthenogenesis. The obtention of haploid plants has frequently required more optimization and has lagged behind research and improvements in chromosome doubling methods. Nevertheless, chromosome doubling has recently been of renewed interest to increase the rates and efficiency of doubled haploid plant production through trialing and optimizing of different procedures. New antimitotic compounds and application methods are being studied to ensure the success of chromosome doubling once haploid material has been regenerated. Moreover, a haploid inducer-mediated CRISPR/Cas9 genome-editing system is a breakthrough method in the production of haploid plant material and could be of great importance for species where traditional haploid regeneration methods have not been successful, or for recalcitrant species. In all cases, the new deployment of this system will demand a suitable chromosome doubling protocol. In this review, we explore the existing doubled haploid and chromosome doubling methods to identify opportunities to enhance the breeding process in major crops.

Abstract:As in any other economically important crop, the possibility of producing fully homozygous, doubled haploid lines in cucumber allows for faster and cheaper breeding. At present, the fastest way to doubled haploidy is the production of cucumber haploid plants and duplication of their chromosomes to make them doubled haploid. In this chapter, we describe a complete protocol to successfully produce cucumber doubled haploid plants, including the evaluation of their ploidy level by flow cytometry. Briefly, this protocol involves a first step of anther culture to induce microspores to divide and proliferate forming calli. The calli produced are isolated from anthers and transferred first to a liquid medium and then to a solid medium to induce organogenesis. Organogenic shoots will eventually give rise to entire DH plants.

Abstract:Techniques for microspore culture of Brassica napus have been improved rapidly, and embryogenesis has been achieved in a wide range of genotypes. Plants regenerated from microspore-derived embryoids can be haploid, diploid or polyploid. From rapeseed microspore culture it is reported that 70%-90% of regenerated plants are haploid. The usual methods of chromosome doubling involve soaking roots (most common) or whole plants in a colchicine solution, or culturing plantlets in colchicine-containing medium in the greenhouse. Other alternatives are injecting colchicine into the secondary buds or applying colchicine-soaked cotton plugs to axillary buds.

Abstract:Double haploid technology (DH) is an essential tool in plant breeding, enabling the rapid production of homozygous lines. However, Cannabis sativa L. has been categorized as recalcitrant to DH induction. In this study, we evaluated the potential to generate DH C. sativa plants via anther culture and indirect de-novo organogenesis. We examined two cultivars, a THCA-dominant cultivar and a CBDA-dominant line. Callus induction success varied, with 29.48% for the THCA cultivar and 71.08% for the CBDA genotype with a regeneration success of 14.45% in 17 weeks for the latter. Mixoploidy in the callus indicated spontaneous genome doubling, while genetic testing confirmed DH nature of the regenerants. This is the first report documenting the successful induction of DH C. sativa plants through de-novo indirect organogenesis. These findings have profound implications for the C. sativa breeding sector by potentially improving efficiency of genome editing and hybrid development in this economically significant species.

Abstract:Since haploid embryoids were first obtained from microspore culture of Brassica napus there has been a rapid progress in the application of this technique in genetic studies and breeding utilization of rapeseed.The relative simplicity of the culture protocol and high-frequency recovery of doubled haploid plants makes microspore culture a potentially powerful technique for varietal development.Nevertheless, there are still some obstacles in the efficient use of this technique such as recalcitrant genotypes, secondary embryogenesis of embryoids, and the lack of efficient chromosome doubling.The present experiment is attempted to improve the production efficiency of doubled haploid rapeseed plants.

Abstract:The microspore culture technique has its wide applications in plant genetic research and breeding programmes in oilseed Brassicas due to its relative simplicity, efficiency in haploid and doubled haploid production, mutation and germplasm generation, and gene transformation. Various factors could influence microspore embryogenesis and haploid production including donor plant genotype, donor plant physiology, microspore developmental stage, culture conditions, culture environment and pretreatments. Stress is also an essential component during embryogenesis induction in microspore culture. Efficient plant regeneration from microspores mostly occurs through direct embryogenesis ensuring minimal occurrence of cytogenetic abnormalities. Appropriate stress conditions such as chilling, partial desiccation, cotyledon excision, and successive subculture of microspore-derived embryos could promote plant development in oilseed rape. Medium renovation, phytohormones and plant growth regulators, and chromosome doubling agents such as colchicine treatment also affect plant regeneration in Brassica species. Compared to colchicine treatments of microspore-derived embryos and plants, immediate colchicine treatment of isolated microspores results in high embryogenesis and diploidisation and low chimeric percentages. The ploidy level of microspore-derived plants of Brassica species could be estimated by different methods at various stages. Mutation breeding techniques are widely used in plant breeding for producing useful mutants and variants. Microspore culture also provides an ideal method for mutation because the mutated traits can be fixed in homozygous condition by chromosome doubling, which can enforce to obtain target mutation traits efficiently. Ultraviolet irradiation, mutagenic agents ethyl methane sulphonate and sodium azide could be applied to isolated microspores and the derived embryos of rapeseed. Utilization of microspore-derived embryos for production of desired traits such as the altered fatty acids, disease resistance and glucosinolate compositions through mutagenesis and selection is advancing and also discussed.

Age and Race Impact the Association between BMI and CVD Mortality in Women

Abstract:Objectives. In previous studies, we have shown that obesity is associated with increased cardiovascular disease (CVD) mortality in white women but not in black women. Earlier research suggests that body mass index (BMI) has a greater effect on CVD mortality in younger white females than older white females, whereas this relationship in black women is not as clear. This study examines the effect of age on the association of BMI to CVD in black and white women. Methods. The Black Pooling Project includes data on 2,843 black women with 50,464 person-years of follow-up, and 12,739 white women with 214,606 person-years of follow-up. A Cox proportional hazards model was used to examine the association between BMI and CVD mortality for specific age/race groups. The younger group was <60 years of age and the older group was >60 years of age. Results. In younger white women, the relative risk (95% confidence interval [CI]) for CVD mortality was significant in obese women (BMI >30 kg/m2) vs. women of normal weight (BMI 18.5–24.9 kg/m2) (1.59 [CI 1.20, 2.09]). Similarly, in older white women, the relative risk for CVD mortality in obese women vs. women of normal weight was significant (1.21 [CI 1.04, 1.41]). There were no such associations for black women. Overweight (BMI 25–29.9 kg/m2) was not associated with increased risk in black or white women. Conclusion. These findings indicate that obesity is associated with a significantly greater risk of CVD mortality among white women, with the strongest association among white women <60 years of age.

Abstract:Abstract Exploitation of biotechnological tools in conventional breeding strategies is the need of the hour for overcoming limitations in rice production and productivity. In addition, improvement in quantity and quality along with resistance to climatic and disease stress in rice require immediate attention. Anther culture has proven its efficiency by instantaneously fixing homozygosity through diploidization of regenerated haploid plants. Therefore, androgenesis provides an efficient platform for developing inbred lines in a short period of time. Although anther culture shows its efficiency in speeding up breeding in several crop species, including rice, associated limitations still prevent the exploitation of its optimum potential. Although anther culture is well exploited in japonica rice breeding, its application in indica rice is limited because of inherent recalcitrant genetic backgrounds. The success of anther culture is determined by several factors that limit the efficiency of androgenesis. Identified constraints are early anther necrosis, poor-callus response, and proliferation, and low green-plant regeneration, along with the most frustrating albinism associated with indica rice, which has been considerably clarified. This chapter details the method of androgenesis and scope for improving the applicability of anther culture producing doubled haploids of rice in order to use it as a complementary tool for precision breeding.

Doubled haploids of novel trigenomic Brassica derived from various interspecific crosses

Abstract:To develop doubled haploid (DH) mapping populations of hexaploid Brassica , 10 F 1 hybrids derived from crosses between allohexaploid Brassica parents were evaluated in this study. The allohexaploid Brassica parents were selfed progenies of unique interspecific crosses between Brassica rapa (genome AA) × B. carinata (BBCC), B. nigra (BB) × B. napus (AACC), and a complex cross between B. juncea (AABB), B. napus and B. carinata , with relatively stable chromosome number (2n = 54). Hexaploid status and chromosome behavior during meiosis I in four promising F 1 hybrids were assessed using microscopy and flow cytometry, and progeny were obtained following microspore culture. Hybrids H11-2 and H16-1 demonstrated higher amenability for embryo generation, plantlet regeneration, and frequency of production of DH microspore-derived progeny of hexaploid DNA content (6 x ) compared to hybrids H08-1 and H24-1. A total of 370 6 x DH progeny were selected out of 693 plantlets from H11-2, 241/436 from H16-1, 23/54 from H08-1, and 21/56 from H24-1. DH progenies of hybrids H11-2 and H16-1 were then designated as promising mapping populations of a new hexaploid Brassica species.
A High-Density Genetic Map of an Allohexaploid Brassica Doubled Haploid Population Reveals Quantitative Trait Loci for Pollen Viability and Fertility.

Abstract:A doubled haploid (DH) mapping population was obtained from microspore culture of an allohexaploid F-1 from the cross between two recently-synthesized allohexaploid Brassica lines. We used single nucleotide polymorphism (SNP) genetic variation based on restriction-site associated DNA (RAD) sequencing to construct a high density genetic linkage map of the population. RAD libraries were constructed from the genomic DNA of both parents and 146 DH progenies. A total of 2.87G reads with an average sequencing depth of 2.59 x were obtained in the parents and of 1.41 x in the progeny. A total of 290,422 SNPs were identified from clustering of RAD reads, from which we developed 7,950 high quality SNP markers that segregated normally (1:1) in the population. The linkage map contained all 27 chromosomes from the parental A, B and C genomes with a total genetic distance of 5725.19 cM and an average of 0.75 cM between adjacent markers. Genetic distance on non-integrated linkage groups was 1534.23 cM, or 21% of total genetic distance. Out of 146 DH progenies, 91 had a complete set of 27 chromosomes as expected of a hexaploid species, and 21 out of 27 chromosomes showed high collinearity between the physical and linkage maps. The loss of chromosome(s) or chromosome segment(s) in the DH population was associated with a reduction in pollen viability. Twenty-five additive QTL were associated with pollen viability and fertility-related traits (seed number, seed yield, pod length, plant height, 1000-seed weight). In addition, 44 intra-genomic and 18 inter-genomic epistatic QTL pairs were detected for 4 phenotypic traits. This provides confidence that the DH population may be selected for improved pollen viability and fertility in a future allohexaploid Brassica species.

Abstract:Abstract Bounteous modern and innovative biotechnological tools have resulted in progressive development in the barley breeding program. Doubled haploids developed (homozygous lines) in a single generation is significant. Since the first discovery of haploid plants in 1920 and, in particular, after discovering in vitro androgenesis in 1964 by Guha and Maheshwari, the doubled haploidy techniques have been progressively developed and constantly improved. It has shortened the cultivar development time and has been extensively used in: genetic studies, gene mapping, marker/trait association, and QTL studies. In barley, the haploid occurrence developed gradually from being a sporadic and random process (spontaneous) to haploid development by in vivo method of modified pollination or by in vitro culture of immature male or female gametophytes. Although significant improvement in DH induction protocols has been made, challenges still exist for improvement in areas such as: low efficiency, albinism, genotypic specificity etc. Here, the paper focuses on: haploidization via different in vitro, in vivo techniques, the recent advances technologies like centromere‐mediated haploidization, hap induction gene, and Doubled haploid CRISPR. The au-courant work of different researchers in barley using these technologies is reviewed. Studies on different factors affecting haploid induction and work on genome doubling of barley haploids to produce DH lines via spontaneous and induced technologies has also been highlighted.

Abstract:Bulb onion (<i>Allium cepa</i> L.) is an allogamous diploid (2n = 16) important for its culinary uses, nutritional value, and medicinal benefits. Despite its economic importance, onion yields and bulb quality are declining, emphasizing the need for new and improved strategies for maintaining and enhancing overall crop quality. Development of inbred lines in onion through traditional breeding is often difficult due to its biennial life cycle, inbreeding depression, and comparatively high heterozygosities. Moreover, genetic research in onion has been hampered by large nuclear genome size. In this regard, gynogenic doubled haploids promise several advantages over inbred lines in support of onion breeding programs and genetic studies. These include complete homozygosity in doubled haploid lines, reduced DNA methylation, elimination of deleterious alleles, and amenability to genetic analysis. This review focuses on the application of in vitro gynogenesis for producing doubled haploids in onion. Factors influencing haploid induction, methods for inducing chromosome doubling and ploidy assessment, evaluation of haploid progenies and doubled haploid lines, and features of doubled haploids potentially useful in crop improvement and genetic studies, are discussed. We identify four major limitations to the success and efficiency of in vitro gynogenesis in onion and discuss strategies for mitigating the negative impacts they pose. This review may be useful to research programs producing doubled haploids in onion or other <i>Allium</i> species using in vitro gynogenesis.
A new method for producing allohexaploid Brassica through unreduced gametes

Abstract:We trialled a two-step method of producing allohexaploid Brassica with three genomes (A, B, and C) derived from pair-wise crossing among three allotetraploid Brassica species. In the first step, the three allotetraploid Brassica species ( Brassica juncea , A j A j B j B j ; Brassica napus , A n A n C n C n ; and Brassica carinata , B c B c C c C c ) were intercrossed in pairs to produce unbalanced trigenomic hybrids: A j A n B j C n , B j B c A j C c and C n C c A n B c . In the second step, these hybrids were crossed with the complementary allotetraploid parent, that is, A j A n B j C n × B c B c C c C c ( B. carinata ), B j B c A j C c × A n A n C n C n ( B. napus ) and C n C c A n B c × A j A j B j B j ( B. juncea ). We hypothesised that the unbalanced trigenomic hybrids would produce high levels of unreduced gametes with the same genome composition as the hybrid. These unreduced gametes would unite with reduced gametes from the complementary allotetraploids to form allohexaploid Brassica progeny (A j A n B c B j C c C n ). From 112 s generation interspecific progeny, two progeny were shown by locus-specific simple sequence repeat markers to be near-allohexaploids derived from an unreduced gamete from C n C c A n B c and a reduced gamete from B. juncea (A j B j ). One of these plants was highly self-fertile, had 50 chromosomes, and inherited patterns of marker alleles (A j A n B c B j C c C n ) that were predicted from first division restitution at meiosis in the C n C c A n B c parent. The second near-allohexaploid had 60 chromosomes, was sterile, and inherited patterns of marker alleles that indicated second division restitution in the C n C c A n B c parent. Trigenomic hybrid Brassica produced by these methods will be valuable bridges to move alleles between allotetraploid species, and may contribute useful meiotic stability alleles to a future allohexaploid species.
