Abstract:Objective: The aim of the present study was to investigate the oral manifestations and salivary composition in type 2 diabetics with periodontitis and to evaluate their association with CA6 gene polymorphism rs2274327.Methods: Oral examination was performed by a single periodontist for 300 type 2 diabetics. Whole unstimulated saliva and blood were collected. The salivary pH, buffer capacity and flow rate were later measured. Immunoglobulin A and electrolytes were assessed using an autoanalyzer. CA6 gene polymorphism rs2274327 was screened by PCR-RFLP assay. The statistical analysis was performed using the SPSS 20.0 version.Results: The salivary pH, buffer capacity and flow rate were significantly lower in the patients carrying TT genotype compared to CC and CT genotype carriers (p < .05). Furthermore, the DMFT index, OHI-s, PI, PPD and CAL were significantly higher in the subjects with TT genotype (p < .05). Carrying at least one T allele seemed to increase the risk of dental caries (OR = 2.59, p < .001), xerostomia (OR = 2.11, p=.003) and taste impairment (OR = 1.97, p < .05).Conclusion: CA6 gene polymorphism rs2274327 seemed to increase the risk of developing, dental caries, periodontitis, xerostomia and taste impairment in type 2 diabetics.

Abstract:Dental caries is a multifactorial disease that can be conceptualized as an interaction between genetic and environmental risk factors. The aim of this study is to examine the effects of AMELX, CA6, DEFB1, and TAS2R38 gene polymorphism and gene-environment interactions on caries etiology and susceptibility in adults. Genomic DNA was extracted from the buccal mucosa, and adults aged 20 to 60 y were placed into 1 of 2 groups: low caries risk (DMFT ≤ 5; n = 77) and high caries risk (DMFT ≥ 14; n = 77). The frequency of AMELX (+522), CA6 (T55M), DEFB1 (G-20A), and TAS2R38 (A49P) single-nucleotide polymorphisms was genotyped with the polymerase chain reaction–restriction fragment length polymorphism method. Environmental risk factors examined in the study included plaque amount, toothbrushing frequency, dietary intake between meals, saliva secretion rate, saliva buffer capacity, mutans streptococci counts, and lactobacilli counts. There was no difference between the caries risk groups in relation to AMELX (+522) polymorphism (χ 2 test, P > 0.05). The distribution of CA6 genotype and allele frequencies in the low caries risk group did not differ from the high caries risk group (χ 2 test, P > 0.05). Polymorphism of DEFB1 (G-20A) was positively associated, and TAS2R38 (A49P) negatively associated, with caries risk (χ 2 test, P = 0.000). There were significant differences between caries susceptibility and each environmental risk factor, except for the saliva secretion rate (Mann-Whitney U test, P = 0.000). Based on stepwise multiple linear regression analyses, dental plaque amount, lactobacilli count, age, and saliva buffer capacity, as well as DEFB1 (G-20A), TAS2R38 (A49P), and CA6 (T55M) gene polymorphism, explained a total of 87.8% of the variations in DMFT scores. It can be concluded that variation in CA6 (T55M), DEFB1 (G-20A), and TAS2R38 (A49P) may be associated with caries experience in Turkish adults with a high level of dental plaque, lactobacilli count, and age and when saliva buffer capacity is low.

Abstract:UNLABELLEDRecent studies have shown that vitamin D receptor (VDR) gene polymorphism had regulatory effects on bone mineral density (BMD) and bone turnover. The VDR gene has also been indicated as a candidate gene for the susceptibility of osteoporosis. However, it is unclear whether VDR genotypes could be associated with alveolar bone loss of patients with periodontitis, or whether vitamin D receptor gene could be a candidate gene for susceptibility to periodontitis. The purpose of this study was to answer these two questions.METHODSTwenty-four cases of adult periodontitis (AP), 37 cases of early onset periodontitis (EOP) and 39 healthy controls were recruited for the study. Individual samples of venous blood and DNA were obtained from each subject. Genotypes of the TaqI VDR gene were determined by PCR and TaqI restriction endonuclease digestion.RESULTSOne out of 24 AP patients, nine out of 37 EOP patients and two out of 39 healthy controls were detected with Tt genotype, while the rest had the TT genotype. The detected frequency of Tt genotype was significantly higher in EOP patients (24.3%) than in AP patients (4.2%) and healthy controls (5.1%). The frequency of t allele was also significantly higher in EOP patients. There was no statistical difference in the distribution of TaqI VDR genotypes between AP patients and healthy controls. The study suggests that Tt genotype might be a risk indicator for the susceptibility to EOP. Carriage of the allele (t) of the TaqI VDR gene may increase the risk of developing EOP.

Abstract:OBJECTIVE:To investigate the relationship between vitamin D receptor (VDR) gene polymorphisms and chronic periodontitis(CP).METHODS:Buccal swabs from 105 patients with mild/mode-rate CP and 85 severe CP were collected, DNA was extracted from these buccal swabs using the TIANamp Swab DNA Kit [TIANGEN Biotech (Beijing) CO.Ltd]. The VDR rs1544410 and rs731236 were genotyped by the Sequenom MassARRAY system (Shanghai Benegene Biotechnology Co. Ltd), which was based on MALDI-TOF (matrix-assisted laser desorption ionization time-of-flight) technology. The distribution of the genotypes and allele frequencies were analyzed.RESULTS:The frequencies of the rs1544410 A allele and AA+AG genotype were significantly higher in severe CP than in mild/moderate CP of all the patients and the female patients respectively (all the patients: P=0.006, 0.007; the female patients: P=0.001, 0.001). The frequencies of the rs731236 C allele and CC+CT genotype were significantly higher in severe CP than in mild/moderate CP of all the patients and the female patients respectively (all the patients: P=0.003, 0.004; the female patients: P<0.001, <0.001).CONCLUSION:Gene polymorphisms of VDR rs731236 and rs1544410 may be associated with severe CP in Chinese Han population.

Abstract:OBJECTIVE:To investigate the relationship between vitamin D receptor (VDR) gene polymorphisms and periodontitis. METHODS:Ninety patients with aggressive periodontitis(AgP), 34 patients with chronic periodontitis and 91 healthy controls were recruited in this study. VDR gene was analyzed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) with Taq I restriction endonuclease. RESULTS:The detection frequency of Tt genotype was higher in the aggressive periodontitis patients than in the healthy controls (16.7% vs 7.7%, P>0.05). The difference between the female aggressive periodontitis patients and healthy controls (20.8% vs 4.2%, P<0.05) was statistically significant, but no difference was found between the male patients and controls (10.8% vs 11.6%, P>0.05). There was a strong association between aggressive periodontitis and Tt genotype in females (AgP patients vs healthy controls, OR=6.02). The detection frequency of Taq I ER-alpha genotypes was not statistically different between the chronic periodontitis patients and healthy controls. CONCLUSION:In female Han Chinese population, the Tt VDR genotype may be associated with aggressive periodontitis.
Relationship in development of malocclusions to polymorphisms of selected vitamin D receptors

Abstract:Background: The development of malocclusion is related to various factor, many of which are still not fully explained. The steroid hormone, 1,25-dihydroxyvitamin D3, has pleiotropic effects. It plays a key role in skeletal metabolism and the control of cell repair by attaching to the nuclear vitamin D steroid receptor (VDR). This vitamin affects bone turnover through the processes of bone tissue formation and resorption via its action on cells of the osteoblastic and osteoclastic lineage, exerts a modulating effect on the immune system, and is involved in the regulation of cell proliferation and differentiation. The role of vitamin D3 (VD3) and its receptor polymorphisms is a rarely studied topic in dentistry. Due to the proven influence on bone turnover processes and immune responses, the main research topic is its relation to periodontal diseases, but so far, its role in the formation and development of malocclusions has not been assessed. Objectives: This study aimed to assess the association of selected VDR polymorphisms: Cdx2 (rs11658820), TaqI (rs7975232), BsmI (rs1544410), ApaI (rs7975232), and FokI (rs2228570) with the development of malocclusions. Material and methods: A prospective observational study was performed. The examination consisted of a medical interview, intraand extraoral orthodontic diagnosis, alginate impression, cone beam computed tomography (CBCT), and venous blood sample to obtain genomic DNA and assess VDR polymorphisms. Results: The rs11658820 polymorphism causes an almost 4-fold increase in the probability of the presence of a malocclusion. GT and TT genotypes of rs7975232 are also associated with a similar risk - almost 6 and almost 5 times higher, respectively. In turn, the effect of the rs2228570-AG and GG genotype polymorphisms on the occurrence of transversal anomalies was demonstrated (odds ratio (OR) = 8.46 and OR = 6.92, respectively). Conclusions: The association of individual polymorphisms with specific malocclusions should be carefully assessed, especially since some trends have been indicated.

Abstract:Objective:To explore the relationship between vitamin D receptor TaqI gene polymorphism and chronic periodontitis.Method:Search was conducted on the following electronic databases:Chinese Bio-medicine Database,China Academic Journals Full-text Database,Medline to identify all the relevant case-control trials.Meta analysis was performed with RevMan4.2 to investigate heterogeneity,to combine the studies and to evaluate the publication bias.Result:Totally 5 studies were recruited.The cases in CP group were 388,in the control group were 355.The pooled OR and 95%CI of genotype TT,Tt,tt were 1.29(0.89;1.87),0.84(0.58;1.21)and 0.75(0.36;1.59)(p>0.05).The pooled OR and 95%CI of alleles T and t were 1.24(0.92;1.68)and 0.80(0.59;1.09)(p>0.05).Conclusion:No evidence showed that vitamin D receptor TaqI gene polymorphism was associated with chronic periodontitis.
Vitamin D Metabolism-related Gene Polymorphisms in Crohn's Disease

Abstract:Objectives: To observe single nucleotide polymorphisms (SNPs) of vitamin D metabolism-related factors, vitamin D receptor (VDR) and vitamin D binding protein (DBP), in Crohn's disease (CD). Methods: 121 patients with CD and 381 controls were enrolled. SNP (rs731236) in VDR gene and SNPs (rs4588, rs7401 and rs2282679) in DBP gene were typed in patients with CD and controls by gene sequencing. Results: In our case-control cohort, no significant difference was observed on CD risk for any of the four SNPs (rs731236, rs4588, rs7401 and rs2282679) in vitamin D metabolism-related genes (P>0.05). Furthermore, there was no association between CD susceptibility and the haplotypes of DBP gene (P> 0.05). Conclusions: Our study suggests that the four SNPs (rs731236, rs4588, rs7401 and rs2282679) in vitamin D metabolism-related genes may have no correlation with susceptibility of CD in Chinese Han population. SNP (rs731236) in VDR gene may play a role in the aetiology of CD among affected males. However, our findings need to be confirmed in multi-centre studies.

Abstract:Background: Vitamin D is a potent immune modulator and is associated with autoimmune diseases, predominantly Type 1 diabetes (T1D). Vitamin D status and its receptor gene polymorphisms in T1D are not yet investigated in the South Indian population.Objective: The present study focuses on exploring the significance of vitamin D levels and Vitamin D receptor (VDR) gene polymorphisms with the risk of T1D in the South Indian population.Methods: A total of 120 T1D patients along with 214 unaffected first-degree relatives (FDRs) were included in this study. Genotyping of VDR polymorphisms at four different loci (FokI- F/f, BsmI- B/b, TaqI- T/t, and ApaI- A/a) was assessed through the ARMS-PCR method. Serum vitamin D levels were measured in 98 T1D patients and 75 age and sex-matched siblings by the ELISA method.Results: Vitamin D deficiency (VDD) was observed in a higher proportion of T1D patients than in controls (52% vs. 32%; p<0.03). The frequency of the FokI-FF genotype was significantly higher (OR=1.66; p<0.03) in T1D patients conferring a susceptible association with the disease. Nevertheless, the increased frequency of heterozygous Ff genotype (OR=0.57; p<0.02) among controls may confer a protective association with T1D. Further, the TDT analysis revealed over-transmission of ApaI-A (T: U=15/5; p<0.006) and BsmI-B alleles (T: U=17/5; p<0.01) and under-transmission of BsmI-b/ApaI-a/TaqI-T haplotype (T: U=5.4/14.4; p=0.04) from parents to T1D patients.Conclusion: The present study concludes that VDD is a major contributing risk factor to T1D development in the South Indian population. Furthermore, the FokI-FF genotype, BsmI-B, and ApaI-A alleles are positively associated with T1D risk. Whereas, the FokI-Ff genotype and BsmI-b/ApaI-a/TaqI-T haplotype are negatively associated with the disease.

A Novel Bayes' Theorem-Based Saliency Detection Model

Abstract:SUMMARY We propose a novel saliency detection model based on Bayes’ theorem. The model integrates the two parts of Bayes’ equation to measure saliency, each part of which was considered separately in the previous models. The proposed model measures saliency by computing local kernel density estimation of features in the center-surround region and global kernel density estimation of features at each pixel across the whole image. Under the proposed model, a saliency detection method is presented that extracts DCT (Discrete Cosine Transform) magnitude of lo-cal region around each pixel as the feature. Experiments show that the proposed model not only performs competitively on psychological patterns and better than the current state-of-the-art models on human visual ﬁxation data, but also is robust against signal uncertainty.

Abstract:Background: Oral cancer is known as one of the most common cancers, with a poor prognosis, related to delayed clinical diagnosis, either due to the lack of particular biomarkers related to the disease or costly therapeutic alternatives.Aims and Objectives: In this study association of single nucleotide polymorphism (Taq1, T>C) in Vitamin D receptor gene with oral cancer and pre oral cancer was studied.Materials and Methods: Total 230 patients of precancerous oral lesions (Leukoplakia 70, Oral Sub mucous fibrosis 90, Lichen Planus 70), 72 oral cancer patients and 300 healthy control subjects were genotyped by PCR-RFLP methods. Chi-square test was used for calculation of genotype and allele frequencies.Results: Mutant genotype CC as well as C allele were found to significantly decrease the risk of oral disease (P value=0.04, OR=0.60 and P value=0.02, OR=0.75 respectively). In particular, compared to non smokers, smokers with TC & CC genotypes were at decrease risk of oral diseases (P value=0.0001, OR=0.04). The mutant allele genotype CC as well as the mutant allele C showed protective association with leukoplakia (P value=0.01, OR=0.39 & P value=0.009, OR=0.59 respectively). However, individual with CC genotype had developed high cell differentiated grade at diagnosis (OR= 3.78, P value= 0.008).Conclusions: This study concludes that VDR (Taq1) polymorphism is associated with oral cancer and pre oral cancer susceptibility in North Indian population.

Abstract:Aims Type 2 diabetes mellitus (T2DM) is a complex and multifaceted disease that contributes significantly to Bangladesh's disease burden. Both polygene abnormalities and environmental factors contribute to this genetic condition. Vitamin D receptor (VDR) has immunomodulatory functions that may contribute to the development of type 2 diabetes. This investigation examined the association between the vitamin D receptor gene (rs2228570) FokI polymorphism and genetic susceptibility to T2DM in the Bangladeshi population. Methods A total of 203 subjects (108 clinically identified T2DM patients and 95 healthy controls) participated in this research study with the ethical committee's approval. Genomic DNA was isolated from venous blood collected from the volunteers with prior consent. Extracted DNA was then genotyped for single nucleotide polymorphism (SNPs) for VDR (rs2228570) FokI gene by PCR-RFLP analysis, where the genotypes were assessed by agarose gel electrophoresis. Results Genotype distribution for VDR (rs2228570) FokI polymorphism exhibited a significant difference between T2DM patients and the control group, whereas allele frequencies for both genes did not differ evidently between the patient and control group. Conclusions Our finding demonstrates a possible link between the risk of T2DM and the FokI polymorphism of the VDR (rs2228570) gene.

Abstract:Objective: The aim of this study was to assess the role of ENAM rs3796703, LTF rs1126478, and TNF-alpha rs1800629 in high caries susceptibility.Design: The present case-control study included 1005 unrelated children under 4 years old: 505 with severe caries (dmft index >= 4) and 500 who were caries-free (dmft index = 0 and without white-spot lesions). Questionnaires were obtained from parents and gardians about the children's diet and oral behavioural habits. All the children received dental examinations and oral swabbing for human genomic DNA collection. ENAM rs3796703, LTF rs1126478, and TNF-alpha rs1800629 were genotyped by Sanger sequencing.Results: The frequency of the ENAM rs3796703 T allele (6.7% in the caries group and 4.2% in the caries-free group), CT genotype (12.7% in the caries group and 8.4% in the caries-free group), TNF-alpha rs1800629 A allele (4.8% in the caries group and 6.8% in the caries-free group), and AG genotype (8.7% in the caries group and 13.2% in the caries-free group) were significantly different between the caries and caries-free groups (p < 0.05). No significant difference was found in the LTFrs1126478 allele frequency and genotype distribution between the two groups. The ENAM rs3796703 CT genotype increased caries susceptibility by 60.9% compared to the CC genotype (beta = 0.746, OR = 1.609), and the TNF-alpha rs1800629 AG genotype reduced caries susceptibility by 47.4% compared to the GG genotype (beta = -0.642, OR = 0.526). In terms of habits covariates, prolongation of night feeding time by 1 month increased caries susceptibility by 3.3% (beta = 0.033, OR = 1.033); additionally, sweets and acidic drinks consumption 1-2 times per day increased caries susceptibility by 218.2% (beta =1.158, OR = 3.182), and consumption 3 or more times pe/r day increased susceptibility by 883.5% (13 = 2.286, OR = 9.835) compared to non-consumption. Topical fluoride application decreased caries susceptibility by 43.0% (beta = -0.562, OR = 0.570).Conclusions: The ENAM and TNF-alpha genes are likely associated with caries experience in Chinese children. The ENAM rs3796703 CT genotype might be involved in caries susceptibility, while TNF-alpha rs1800629 AG genotype might be involved in caries protection. (C) 2017 Published by Elsevier Ltd.

Abstract:Solid-state 13C nuclear magnetic resonance (NMR) spectroscopy was applied to intact and isolated loblolly pine wood samples to identify potential structural changes induced by tree age, milling, lignin extraction, or naturally occurring mutations. Special attention was paid to ketone and aldehyde as well as nonpolar alkyl groups, which could be observed at low concentrations (<2 in 1000 C) using improved spinning-sideband suppression with gated decoupling. Carbonyl structures were present in intact wood, and there are more keto groups than aldehydes. Their concentrations increased from juvenile to mature wood and with milling time, whereas extraction did not alter the C=O fraction. Significant amounts of aldehyde and dihydroconiferyl alcohol residues were present in coniferyl aldehyde dehydrogenase-deficient wood, confirming solution-state NMR spectra of the corresponding lignin. These results demonstrate the utility of solid-state NMR as an assay for changes in the lignin structure of genetically modified plants.
Vitamin D Receptor Gene Polymorphism and Vitamin D Status in Population of Patients with Cardiovascular Disease—A Preliminary Study

Abstract:The association between vitamin D receptor (VDR) polymorphism and the risk of cardiovascular diseases (CVD) remains unclear. This study aimed to assess a relationship between the VDR genotypes, plasma concentrations of vitamin D metabolites, and the occurrence of cardiovascular and metabolic disorders. Fifty-eight patients treated for various cardiological afflictions were included. Identification of VDR polymorphisms: ApaI, TaqI, BsmI, and FokI were carried out using the PCR-RFLP method. Plasma concentrations of 25-hydroxyvitamin-D2, 25-hydroxyvitamin-D3, and 3-epi-25-hydroxyvitamin D3 were assessed by the UPLC-MS/MS method. Lower incidence of BsmI AA genotype in the studied patients was observed compared with healthy controls, but the difference was insignificant. Among patients with the TT genotype, frequency of hypertension was higher than among carriers of other ApaI genotypes (p < 0.01). In addition, carriers of the TT ApaI, TC TaqI, and GA BsmI genotypes had an increased risk of obesity, while the presence of the FokI TT genotype was associated with a higher incidence of heart failure and hypertension. In conclusion, the BsmI AA genotype can be protective against CVD, but this observation needs study on a larger group of patients. Particular VDR genotypes were associated with 25-hydroxyvitamin-D levels, and the mechanism of this association should be further investigated.

Association of Genetic Variants in Enamel-Formation Genes with Dental Caries: A Meta- and Gene-Cluster Analysis

Abstract:Previous studies have reported the association between multiple genetic variants in the enamelformation genes and the risk of dental caries with inconsistent results. We performed a systematic literature search of the PubMed, Cochrane Library, HuGE and Google Scholar databases for studies published before March 21, 2020 and conducted meta-, gene-based and gene-cluster analysis on the association between genetic variants in the enamel-formation genes and the risk of dental caries. We identified 21 relevant publications including a total of 24 studies for analysis. The genetic variant rs17878486 in AMELX was significantly associated with dental caries risk (OR = 1.40, 95% CI: 1.02-1.93, P = 0.037). We found no significant association between the risk of dental caries with rs12640848 in ENAM (OR = 1.15, 95% CI: 0.88-1.52, P = 0.310), rs1784418 in MMP20 (OR = 1.07, 95% CI: 0.76-1.49, P = 0.702) and rs3796704 in ENAM (OR = 1.06, 95% CI: 0.96-1.17, P = 0.228). Gene-based analysis indicated that multiple genetic variants in AMELX showed joint association with the risk of dental caries (6 variants; P < 10(-5)), so did genetic variants in MMP13 (3 variants; P = 0.004), MMP2 (3 variants; P < 10(-5)), MMP20 (2 variants; P < 10(-5)) and MMP3 (2 variants; P < 10(-5)). The gene-cluster analysis indicated a significant association between the genetic variants in this enamel-formation gene cluster and the risk of dental caries (P < 10-5). The present meta-analysis revealed that genetic variant rs17878486 in AMELX was associated with dental caries, and multiple genetic variants in the enamel-formation genes jointly contributed to the risk of dental caries, supporting the role of genetic variants in the enamelformation genes in the etiology of dental caries. TLR2 and TLR4 Polymorphisms Are Not Associated with Dental Caries in Polish Children

Abstract:The aim of the present study was to analyze the association of the TLR2 (Toll-like receptor 2 gene) 2258G>A (rs5743708), TLR4 (Toll-like receptor 4 gene) 896A>G (rs4986790), and TLR4 1196C>T (rs4986791) polymorphisms with dental caries in Polish children. The participants, 261 15-year-old children, were divided into two groups: 82 cases (i.e., children with DMFT (Decayed, Missing, and Filled Teeth) index >5, having either moderate or high caries experience, assigned as the "higher" caries experience group) and 179 controls (i.e., children with DMFT ≤ 5, having either low or very low caries experience, assigned as the "lower" caries experience group). Genomic DNA was isolated from buccal swabs, and genotyping was determined by means of real-time PCR (polymerase chain reaction). There were no significant differences in the genotype or allele distributions in all tested SNPs (single nucleotide polymorphisms) between children with "higher" caries experience and those with "lower" caries experience. TLR4 haplotype frequencies did not differ significantly between cases and controls. In an additional analysis with another case definition applied (subjects with DMFT ≥ 1 were assigned as "cases", whereas children with DMFT = 0 were assigned as "controls"), no significant differences in the TLR2 and TLR4 genotype, allele frequencies, and TLR4 haplotype frequencies were found between the case and the control groups. The results of the present study broaden our knowledge on the potential genetic factors that might affect caries risk and suggest that TLR2 rs5743708 and TLR4 rs4986790 and rs4986791 SNPs are not associated with dental caries susceptibility in Polish children.
