Abstract:Melatonin, a natural agent, has multiple functions in animals as well as in plants. However, its possible roles in plants under abiotic stress are not clear. Nowadays, soil salinity is a major threat to global agriculture because a high soil salt content causes multiple stresses (hyperosmotic, ionic, and oxidative). Therefore, the aim of the present study was to explore: (1) the involvement of melatonin in biosynthesis of photosynthetic pigments and in regulation of photosynthetic enzymes, such as carbonic anhydrase (CA) and ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco); (2) the role of melatonin in osmoregulation by proline and carbohydrate metabolism; and (3) the function of melatonin in the antioxidant defense system under salinity. Outcomes of the study reveal that under non-saline conditions, application of melatonin (20 and 50 μM) improved plant growth, viz. shoot length, root length, shoot fresh weight (FW), root FW, shoot dry weight (DW), root DW and leaf area and physio-biochemical parameters [chlorophyll (Chl) a and b, proline (Pro) and total soluble carbohydrates (TSC) content, and increased the activity of CA and Rubisco]. However, tomato seedlings treated with NaCl exhibited enhanced Chl degradation, electrolyte leakage (EL), malondialdehyde (MDA) and reactive oxygen species (ROS; superoxide and hydrogen peroxide). ROS were detected in leaf and root. Interestingly, application of melatonin improved plant growth and reduced EL, MDA and ROS levels through upregulation of photosynthesis enzymes (CA, Rubisco), antioxidant enzymes (superoxide dismutase, catalase, glutathione reductase and ascorbate reductase) and levels of non-enzymatic antioxidants [ascorbate (ASC) and reduced glutathione (GSH)], as well as by affecting the ASC—GSH cycle. Additionally, exogenous melatonin also improved osmoregulation by increasing the content of TSC, Pro and Δ1-pyrroline-5-carboxylate synthetase activity. These results suggest that melatonin has beneficial effects on tomato seedlings growth under both stress and non-stress conditions. Melatonin’s role in tolerance to salt stress may be associated with the regulation of enzymes involved in photosynthesis, the antioxidant system, metabolism of proline and carbohydrate, and the ASC—GSH cycle. Also, melatonin could be responsible for maintaining the high ratios of GSH/GSSG and ASC/DHA.

Exogenous Potassium (K+) Positively Regulates Na+/H+ Antiport System, Carbohydrate Metabolism, and Ascorbate–Glutathione Cycle in H2S-Dependent Manner in NaCl-Stressed Tomato Seedling Roots

Abstract:Potassium (K+) is one of the vital macronutrients required by plants for proper growth and blossoming harvest. In addition, K+ also plays a decisive role in promoting tolerance to various stresses. Under stressful conditions, plants deploy their defense system through various signaling molecules, including hydrogen sulfide (H2S). The present investigation was carried out to unravel the role of K+ and H2S in plants under NaCl stress. The results of the study show that NaCl stress caused a reduction in K+ and an increase in Na+ content in the tomato seedling roots which coincided with a lower H+-ATPase activity and K+/Na+ ratio. However, application of 5 mM K+, in association with endogenous H2S, positively regulated the Na+/H+ antiport system that accelerated K+ influx and Na+ efflux, resulting in the maintenance of a higher K+/Na+ ratio. The role of K+ and H2S in the regulation of the Na+/H+ antiport system was validated by applying sodium orthovanadate (plasma membrane H+-ATPase inhibitor), tetraethylammonium chloride (K+ channel blocker), amiloride (Na+/H+ antiporter inhibitor), and hypotaurine (HT, H2S scavenger). Application of 5 mM K+ positively regulated the ascorbate–glutathione cycle and activity of antioxidant enzymes that resulted in a reduction in reactive oxygen species generation and associated damage. Under NaCl stress, K+ also activated carbohydrate metabolism and proline accumulation that caused improvement in osmotic tolerance and enhanced the hydration level of the stressed seedlings. However, inclusion of the H2S scavenger HT reversed the effect of K+, suggesting H2S-dependent functioning of K+ under NaCl stress. Therefore, the present findings report that K+, in association with H2S, alleviates NaCl-induced impairments by regulating the Na+/H+ antiport system, carbohydrate metabolism, and antioxidative defense system.

Abstract:Soil salinization is a serious threat to agricultural production because it limits the growth and development of crops. Melatonin is a pleiotropic molecule widely used to alleviate abiotic stressors, including salt stress. The objective of the present study was to investigate the function of melatonin in metabolic processes related to the mitigation of salt-induced growth inhibition in tomato seedlings. The hydroponically grown tomato seedlings were foliar sprayed with 100 μM melatonin and salt (NaCl) treatment was applied in tomato roots for 7 days. The experiment was composed of four treatments: (1) control; (2) melatonin (MT, 100 μM); (3) salt (125 mM NaCl); and melatonin + NaCl (MT + salt, 100 μM + 125 mM NaCl). The results revealed that melatonin treatment significantly mitigated salt-induced adverse effects as evidenced by higher biomass production, greater accumulation of K + , and lower content of Na + in salt-stressed tomato leaves and roots, resulting in lower Na + /K + ratio as compared with only salt-stressed seedlings. Melatonin application enhanced photosynthetic efficiency through the elevation of gas exchange and chlorophyll fluorescence parameters values, along with the increment of carbon assimilation enzymes activity. Moreover, sucrose content and sucrose phosphate synthase (SPS) activity were increased in melatonin-received tomato seedlings over to salt-stressed plants. Melatonin also regulates gene expression related to sugar transporters and carbohydrate metabolism in plants exposed to salinity stress. Additionally, plants treated with melatonin showed higher nitrate reductase and nitrate reductase activity by 78% and 62%, respectively, compared with plants under salt stress. Similarly, the nitrogen assimilation capacity (higher nitrate, NO 3 − level) was increased in melatonin treatment plants. Taken together, the present research findings indicate that melatonin attenuates salinity-caused growth inhibition through the coordination action of carbohydrate and nitrogen metabolism and the balancing of Na + /K + homeostasis in tomato seedlings.

Abstract:Melatonin (Mel) is a multifunctional signaling molecule, quantified in a wide range of higher plants. It acts as a pleiotropic molecule and is involved in numerous signaling and physiological processes, particularly under harsh environmental conditions in plants. In the present study, the defensive role of Mel was explored in salt (NaCl)-stressed tomato (Solanum lycopersicum) plants. Mel (50 or 100 µM) was supplied to the foliage of tomato plants at 30 days after transplantation (DAT), grown without stress, and in the presence of NaCl (100 or 150 mM) stress. Salt stress negatively affected photosynthetic efficiency, cell viability, plant growth, fruit quality, and impaired root morphology, and stomatal physiology, but induced the accumulation of reactive oxygen species ((H2O2 and O2·−), malondialdehyde (MDA) content, and electrolyte leakage. Interestingly, foliar supplementation of Mel to stressed and non-stressed plants lowered MDA levels, reduced oxidative stress and promoted photosynthesis, improved cell viability, stomatal behavior, and plant growth, and boosted the activity of catalase (CAT), peroxidase (POX), and superoxide dismutase (SOD), and elevated proline content which bestowed endurance and reduced the salt-induced oxidative damage.

Abstract:Troponins are recommended as the biomarkers of choice for the detection of myocardial infarction (MI) by the Universal Definition.1 This is well accepted for the diagnosis of type 1 or spontaneous MI, in which any elevation in the setting of ischemia is required but not for the diagnosis of type 4a or MI associated with percutaneous coronary intervention (PCI), in which an isolated 3× the upper reference limit elevation is required. Creatine kinase MB (CKMB) is widely favored because a number of studies support a relationship between CKMB elevation after PCI and long-term mortality with the cut-point appearing to be about 8 to 10× elevation of CKMB.2–5 Article see p 150 Troponins are much more sensitive than CKMB in detecting small areas of myocyte necrosis, but small amounts of myocyte necrosis may not be related to increased mortality6,7 In a meta-analysis of 4 studies with 2359 patients undergoing PCI, increases in troponin of >3 times upper reference limit were found in 14.5% and were associated at 18-month follow-up with increased events (death, MI, repeat target vessel PCI, and coronary artery bypass grafting) (odds ratio [OR], 2.25; 95% confidence interval [CI], 1.26–4.00).8 However, none of the studies used a 99th percentile cutoff with a sensitive troponin assay to define a normal baseline. The situation is complex because PCI may reduce the risk of death in high-risk patients, and the risk of a small amount of myocardial injury related to PCI may be balanced by the success of the PCI. Abstract:Soil salinity poses a significant threat to agricultural productivity, impacting the growth and yield of wheat (Triticum aestivum L.) plants. This study investigates the potential of melatonin (MT; 100 µM) and hydrogen sulfide (H2S; 200 µM sodium hydrosulfide, NaHS) to confer the tolerance of wheat plants to 100 mM NaCl. Salinity stress induced the outburst of reactive oxygen species (ROS) resulting in damage to the chloroplast structure, growth, photosynthesis, and yield. Application of either MT or NaHS augmented the activity of antioxidant enzymes, superoxide dismutase, ascorbate peroxidase, glutathione reductase, and reduced glutathione (GSH) levels, upregulated the expression of Na+ transport genes (SOS1, SOS2, SOS3, NHX1), resulting in mitigation of salinity stress. Thus, improved stomatal behavior, gas-exchange parameters, and maintenance of chloroplast structure resulted in enhanced activity of the Calvin cycle enzymes and overall enhancement of growth, photosynthetic, and yield performance of plants under salinity stress. The use of DL-propargylglycine (PAG, an inhibitor of hydrogen sulfide biosynthesis) and p-chlorophenyl alanine (p-CPA, an inhibitor of melatonin biosynthesis) to plants under salt stress showed the comparative necessity of MT and H2S in mitigation of salinity stress. In the presence of PAG, more pronounced detrimental effects were observed than in the presence of p-CPA, emphasizing that MT was involved in mitigating salinity through various potential pathways, one of which was through H2S.

Abstract:Melatonin (MT) has been reported to regulate certain plant physiological processes and promote tolerance to different environmental stresses such as salinity. Green bean (Phaseolus vulgaris L. cv Royal Nel) seedlings were exposed to 200 mM NaCl stress with or without pre-treatment with 150 μM MT. Salt stress led to a lower chlorophyll content, a reduced photosynthetic activity, increased reactive oxygen species (ROS) contents, and decreased photosystem II (PSII) activity. The application of exogenous MT to green bean seedlings under salt stress improved photosynthetic activiy and alleviated the oxidative damages by enhancing the activity of antioxidant enzymes. The expression of catalase (CAT1), glutathione reductase (GR), superoxide dismutase (CuZnSOD1), ascorbate peroxidase (APX), Peroxiredoxin Q (PrxQ), and 2-cysteine peroxiredoxin (2-Cys-Prx) encoding genes was significantly increased under salt stress in green bean seedling compared with the untreated control. However, plants treated with exogenous MT and NaCl had 28.8, 21.1, 26.1, 20, 26.2, and 22.4% higher CuZnSOD, CAT1, APX, GR, PrxQ and 2-Cys-Prx transcript levels, respectively, compared to NaCl stress alone. Our study revealed the protective mechanisms mediated by exogenous MT application in NaCl stress alleviation and our findings could be used in the management of green bean cultivation in salinity-prone soils.

Abstract:The beneficial roles of melatonin (Mel) and gibberellic acid (GA3) in the biosynthesis of photosynthetic pigments, osmoregulation, and methylglyoxal (MG) detoxification and antioxidant system were studied in tomato (Solanum lycopersicum L. cv. Five Star) seedlings under NaCl stress. The exogenous application of Mel (100 µM) and GA3 (1.4 µM) together more efficiently affected growth performance of seedlings under salt stress. The decreased chlorophyll (Chl) degradation and Chl-degrading enzyme (chlorophyllase) activity in seedlings receiving Mel plus GA3 resulted in increased Chl content by upregulating Chl synthesizing enzyme (δ-aminolevulinic acid dehydratase) under salinity. Exogenous Mel plus GA3 suppressed the overproduction of reactive oxygen species (ROS; superoxide and hydrogen peroxide) and activity of glycolate oxidase. Application of Mel with GA3 reduced MG content by enhancing the activity of enzymes (glyoxalase I and glyoxalase II) involved in the MG detoxification system. Both Mel and GA3 together protected seedlings from ROS induced damage by regulating Δ1-pyrroline-5-carboxylate synthetase activity, and content of proline (Pro) and glycine betaine (GB). Seedlings receiving Mel + GA3 exhibited a substantial upregulated activity of catalase, ascorbate peroxidase, glutathione reductase, dehydroascorbate reductase, monodehydroascorbate reductase, glutathione peroxidase, polyphenol oxidase and lipoxygenase, and redox homeostasis that reduced oxidative damage induced by salinity. These outcomes advocate that Mel and GA3 played beneficial roles in Chl, Pro and GB biosynthesis, and improved redox homeostasis, and MG detoxification and antioxidant system under salt stress.

Abstract:Being sessile, plants' exposure to various environmental stresses during their life cycle is inevitable which can affect their yield and productivity. This study investigates the protective effect of exogenous application of melatonin on 30-day-old tomato plants under NaCl and cadmium stress treatments. Plant growth, photosynthetic pigment, and antioxidant enzymes of plants exposed to NaCl, cadmium (Cd), and NaCl + Cd stress and melatonin treatment on them were analysed. The plants under NaCl- and Cd-induced stress produced an increased amount of ROS and lipid peroxidation, which were significantly reduced upon melatonin application. The maximum quantum energy (F V /F M ) and performance index (PI ABS ) significantly improved in melatonin-treated plants under stress conditions. Redox homeostasis was maintained with a significant increase in superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase. This study suggests that exogenous melatonin improves plants' ability to overcome the combination stress caused by NaCl and Cd by increasing overall photosynthetic capacity and modulating redox balance.

They regulate stomatal movement and enhance drought resistance. However, their physiological relationship is not well understood. Here, their crosstalk involved in osmotic stress resistance in Arabidopsis thaliana was studied. Exogenous H2S and MEL closed stomata under normal or osmotic stress conditions and increased the relative water contents of plants under osmotic stress conditions. At the same time, exogenous H2S and MEL responded to osmotic stress by increasing the content of proline and soluble sugar, and reducing malondialdehyde (MDA) content and relative conductivity. Using mutants in the MEL-associated production of serotonin N-acetyltransferase (snat), caffeic acid O-methyltransferase (comt1) and N-acetylserotonin methyltransferase (asmt), we determined that H2S was partially dependent on MEL to close stomata. Additionally, the overexpression of ASMT promoted stomatal closure. Exogenous H2S increased the transcription levels of SNAT, ASMT and COMT1. Furthermore, exogenous H2S treatments increased the endogenous MEL content significantly. At the post-translational level, H2S sulfhydrated the SNAT and ASMT, but not COMT1, enzymes associated with MEL production. Thus, H2S appeared to promote stomatal closure in response to osmotic stress by increasing the transcription levels of MEL synthesis-related genes and the sulfhydryl modification of the encoded enzymes. These results increased our understanding of H2S and MEL functions and interactions under osmotic stress conditions.

Abstract:Melatonin is a ubiquitous signaling plant hormone that plays a crucial role in regulating the growth and development of plants under stress conditions. Since a few species have been investigated to unveil the effect of exogenous melatonin on salt stress, in the current research study, we investigated the effects of melatonin by measuring the photosynthetic characteristics and antioxidant defense system of maize seedling under salt stress (200 mM NaCl), along with different concentrations of melatonin (MT1—30, MT2–60, and MT3–90 µM) treatments. Salt stress reduced the plant growth characteristics and photosynthetic efficiency by increasing the ROS accumulation and reducing the antioxidant enzyme activities of maize seedling. However, pretreatment with melatonin on roots alleviated NaCl-induced decrease in photosynthetic rate and oxidative stress in a dose-dependent manner. Our results revealed that exogenous application of melatonin at an optimum concentration (60 µM) under salt stress conditions significantly increased the growth of plants, chlorophyll content, photosynthetic efficiency, antioxidant enzymes activities, i.e., superoxide dismutase (SOD), peroxide (POD), catalase (CAT) and ascorbate peroxidase (APX), and reduced the reactive oxygen species (ROS). This study unraveled the crucial role of melatonin in salt stress mitigation and thus can be implicated in the management of salinity in maize seedling.

Abstract:Salinity causes yield and quality losses in agricultural production and therefore great economic losses around the world. Hydrogen sulfide (H2S) is known to play a crucial role to ease physiological and metabolic processes in plants, and also increases the tolerance of the plant against many abiotic stress conditions. In this study, we investigated the effects of H2S treatments (0, 25, 50, 75 and 100 μM NaHS were applied as H2S donor) to the tomato seedlings to alleviate the harmful effects of salt stress (0, 75 and 150 mM NaCl). There was a significant decrease in plant growth and development in parallel with the increased salt level. Visible changes in plant development were observed after the dose of 75 mM NaCl in the tomato seedling. The effects of different doses of exogenous H2S treatment were found to be significant. H2S treatment increased the stress tolerance in tomato seedlings by arranging the mineral element and hormone content. Furthermore, H2S relieved the effect of stress in plants by increasing photosynthetic activity (photosynthesis rate (Pn), transpiration rate (Tr), stomatal conductivity (gs) and intercellular CO2 concentration (Ci)) of the plant. In addition, the effect of H2S on salt stress tolerance in tomato seedlings may be due to its positive effect on mineral element contents. As a result, based on the beneficial effects of H2S in tomato seedlings under salt stress, this treatment can be considered as an alternative resilience method for cultivation in saline soils or irrigation with low quality waters.

Abstract:The interactions between nitric oxide (NO) and melatonin in alleviating sodium chloride (NaCl) toxicity in plants are poorly comprehended. Here, the associations between the exogenous application of melatonin and endogenous NO levels in inducing tomato seedlings' defense response during NaCl toxicity were investigated. The results indicated that the application of melatonin (150 μM) increased height (23.7%) and biomass (32.2%), improved chlorophyll (a (137%) and b (92.8%)), and proline metabolisms, and reduced the contents of superoxide anion radicals (49.6%), hydrogen peroxide (31.4%), malondialdehyde (38%), and electrolyte leakage (32.6%) in 40-day-old tomato seedlings grown under NaCl (150 mM) treatment. Melatonin increased the antioxidant defense system in NaCl-stressed seedlings by increasing the activity of the antioxidant enzymes. Melatonin also improved N metabolism and endogenous NO content in NaCl-stressed seedlings by upregulating the activity of enzymes implicated in N assimilation. Furthermore, melatonin improved ionic balance and reduced Na content in NaCl-exposed seedlings by upregulating the expression of genes involved in K/Na ratio homeostasis ( NHX1-4 ) and increasing the accumulation of mineral nutrients (P, N, Ca, and Mg). However, the addition of cPTIO (100 μM; an NO scavenger) reversed the beneficial impacts of melatonin, indicating the effective function of NO in melatonin-induced defense mechanisms in NaCl-stressed tomato seedlings. Therefore, our results revealed that melatonin improves the tolerance of tomato plants during NaCl toxicity by mediating internal NO.

Mitogen-Activated Protein Kinase Is Involved in Salt Stress Response in Tomato (Solanum lycopersicum) Seedlings

Abstract:Salt stress impairs plant growth and development, thereby causing low yield and inferior quality of crops. In this study, tomato (Solanum lycopersicum L. 'Micro-Tom') seedlings treated with different concentrations of sodium chloride (NaCl) were investigated in terms of decreased plant height, stem diameter, dry weight, fresh weight, leaves relative water content and root activity. To reveal the response mechanism of tomato seedlings to salt stress, the transcriptome of tomato leaves was conducted. A total of 6589 differentially expressed genes (DEGs) were identified and classified into different metabolic pathways, especially photosynthesis, carbon metabolism, biosynthesis of amino acids and mitogen-activated protein kinase (MAPK) signaling pathway. Of these, approximately 42 DEGs were enriched in the MAPK signaling pathway, most of which mainly included plant hormone, hydrogen peroxide (H2O2), wounding and pathogen infection signaling pathways. To further explore the roles of MAPK under salt stress, MAPK phosphorylation inhibitor SB203580 (SB) was applied. We found that SB further decreased endogenous jasmonic acid, abscisic acid and ethylene levels under salt stress condition. Additionally, in comparison with NaCl treatment alone, SB + NaCl treatment reduced the content of O2- and H2O2 and the activities of antioxidant enzyme and downregulated the expression levels of genes related to pathogen infection. Together, the results revealed that MAPK might be involved in the salinity response of tomato seedlings by regulating hormone balance, ROS metabolism, antioxidant capacity and plant immunity.
Effects of foliar spraying with melatonin and chitosan Nano-encapsulated melatonin on tomato (Lycopersicon esculentum L. cv. Falcato) plants under salinity stress

Abstract:Melatonin has been found to be crucial in the growth and development of plants under stress conditions. In this study, the effects of melatonin and nano melatonin regarding the growth and development of tomato plants, along with their photosynthetic pigment, phenol, and antioxidant activity, were investigated under saline conditions. The study was conducted using a completely randomized design with three replications, and the applied treatments were salt stress and foliar spraying of melatonin at a concentration of 0 (control), melatonin (Mel), and nano capsule-melatonin (Nano-Mel) at 500 µM. Salinity treatments included application of sodium chloride with two concentration of 0 mM NaCl (S1) and 50 mM NaCl (S2). Under saline conditions, Mel and Nano-Mel increased both shoot and root fresh and dry weights, improved relative water content (RWC), and enhanced antioxidant activity and phenolic content. Salinity elevated leaf ABA content, unaffected by Mel or Nano-Mel. Chlorophyll fluorescence and SPAD values demonstrated resilience to salinity with Mel and Nano-Mel applications. Nano-Mel notably mitigated Na + accumulation in leaves under salinity, helping maintain K + homeostasis. Proline levels rise due to salinity but decreased with Mel and Nano-Mel treatments. Electrolyte leakage (EL) increased under salinity but is significantly reduced by Mel, indicating enhanced membrane stability. The findings reveal that salinity stress significantly reduced plasma membrane intrinsic protein (PIP) expression in roots and leaves, whereas Mel and Nano-Mel treatments enhance PIP expression, particularly in roots. The study concludes that Mel and Nano-Mel effectively alleviate salinity-induced stress, promoting growth and maintaining physiological homeostasis in tomato plants.
Beneficial Roles of Melatonin on Redox Regulation of Photosynthetic Electron Transport and Synthesis of D1 Protein in Tomato Seedlings under Salt Stress

Abstract:Melatonin is important in the protection of plants suffering various forms of abiotic stress. The molecular mechanisms underlying the melatonin-mediated protection of their photosynthetic machinery are not completely resolved. This study investigates the effects of exogenous melatonin applications on salt-induced damage to the light reaction components of the photosynthetic machinery of tomato seedlings. The results showed that melatonin pretreatments can help maintain growth and net photosynthetic rate (PN) under salt stress conditions. Pretreatment with melatonin increased the effective quantum yield of photosystem II (ΦPSII), the photochemical quenching coefficient (qP) and the proportion of PSII centers that are "open" (qL) under saline conditions. In this way, damage to the photosynthetic electron transport chain (PET) in photosystem II (PSII) was mitigated. In addition, melatonin pretreatment facilitated the repair of PSII by maintaining the availability of D1 protein that was otherwise reduced by salinity. The ROS levels and the gene expressions of the chloroplast TRXs and PRXs were also investigated. Salt stress resulted in increased levels of reactive oxygen species (ROS), which were mitigated by melatonin. In tomato leaves under salt stress, the expressions of PRXs and TRXf declined but the expressions of TRXm1/4 and TRXm2 increased. Melatonin pretreatment promoted the expression of TRXf and the abundances of TRXf and TRXm gene products but had no effects on the expressions of PRXs. In summary, melatonin improves the photosynthetic activities of tomato seedlings under salt stress. The mechanism could be that: (1) Melatonin controls ROS levels and prevents damaging elevations of ROS caused by salt stress. (2) Melatonin facilitates the recovery of PET and D1 protein synthesis, thus enhancing the tolerance of photosynthetic activities to salinity. (3) Melatonin induces the expression of TRXf and regulates the abundance of TRXf and TRXm gene products, which may facilitate repair of the light reaction parts of the photosynthetic machinery.

Abstract:Melatonin (N-acetyl-5-methoxytryptamine) is an important molecule in plants that regulates growth and development and responds to abiotic stresses. In this study, we studied the role of exogenous melatonin in alleviating salt stress in cucumber. The results showed that exogenous melatonin pretreatment could improve cell viability, protect photosynthesis, increase antioxidant enzyme activity, inhibit active oxygen explosion, reduce malondialdehyde (MDA) content and relative conductivity in cucumber Seedlings under salt stress compared with salt stress alone. Gene expression analysis showed that melatonin significantly increased the expression of antioxidant enzyme gene, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase genes, mitogen-activated protein kinase (MAPK) genes (MAPK3, MAPK4, MAPK6) and salt overly sensitive (SOS) genes (SOS1, SOS2, SOS3) under salt stress. diphenyleneiodonium (DPI) or dimethylthiourea (DMTU) significantly reduced melatonin-induced defense responses, including decreased antioxidant enzyme activity and photosynthesis protection, as well as the expression of stress-related genes. Similar to DPI and DMTU, U0126 (MEK inhibitor) also reduces melatonin-induced defense responses. In conclusion, our study shows that exogenous melatonin treatment can effectively alleviate the damage of salt stress on cucumber seedlings, and the alleviation effect is related to hydrogen peroxide (H2O2). And there may be interactions between melatonin-mediated salt tolerance and the MAP kinase signaling pathway.

Abstract:Soil salinity is a severe threat to agricultural production. Most saline soils turn alkaline, increasing the soil pH and, in turn, hampering the growth and development of crops. In this study, the effects of a foliar spray of melatonin (MT; 100 μmol·L−1) on the pH of the root environment, growth of tomato seedlings, endogenous MT levels, rapid chlorophyll fluorescence induction kinetics, and key enzymes of the Calvin cycle under alkaline (60 mmol·L−1 NaHCO3) stress were studied in Riegel 87-5 tomatoes. The results revealed that the growth and photosynthesis of tomato seedlings were inhibited by increased pH in the root environment under alkali stress; however, the application of exogenous MT reduced the pH of the root environment, alleviated the inhibition of growth of tomato seedlings under alkali stress, increased the content of photosynthetic pigments, alleviated the damage of the donor and acceptor sides of the photosynthetic electron transport chain, increased the activity and efficiency of photosynthetic electron transport, and optimized the share of the light energy allocated to PSII reaction centers. Increased expression levels of Calvin-cycle enzymes, including fructose-1,6-bisphosphate aldolase (FBA), fructose-1,6-bisphosphate esterase (FBP), and phosphoglycerate kinase (PGK), led to enhanced photosynthetic performance in tomato seedlings. Exogenous MT boosted endogenous MT levels and stimulated the production and secretion of organic acids in the root system. This regulation of organic acid content reduced the environmental pH in the inter-root zone, alleviating the damage caused by alkali stress. This study indicated that the exogenous administration of MT may mediate an increase in endogenous MT levels, regulate the efficiency of photosynthesis and root pH levels, and play a crucial role in mitigating injury caused by alkali stress in tomato seedlings.
