Abstract:Here, we explore the possible ancient fungal species in the gut of Ötzi, the Iceman, a naturally mummified human found in the Tyrolean Alps (border between Italy and Austria). While ancient DNA (aDNA) has been extensively used to study human, animal, and plant evolution, this research focuses on ancient microbial diversity, specifically fungi. Fungal DNA is often underestimated in metagenomic samples, however here we hypothesise the possibility of retrieving ancient fungal sequences from Ötzi’s gut. A robust bioinformatic pipeline has been developed to detect and authenticate fungal aDNA from stomach, small intestine, and large intestine samples. We revealed the presence of genus, with and as possible species, that were particularly abundant in the stomach and small intestine. We suggest that Ötzi may have consumed these fungi accidentally, likely in association with other elements of his diet, and they thrived in his gut after his death due to their adaptability to harsh and cold environments. This research provides insight into the coexistence of ancient humans with specific fungal species and proposes and validates a conservative bioinformatic approach for detecting fungal aDNA in historical metagenomic samples.

AMS 14C Dating of Equipment from the Iceman and of Spruce Logs from the Prehistoric Salt Mines of Hallstatt

Abstract:This paper summarizes radiocarbon measurements of mainly botanical samples from the Iceman (“Ötzi”) and from his discovery site, an Alpine glacier at the Austrian-Italian border. The results were obtained by accelerator mass spectrometry (AMS) at 3 different laboratories (Vienna, Austria; Uppsala, Sweden; Gif-sur-Yvette, France) between 1992 and 1997. All the dates, except 2, are consistent with the time period 3360–3100 BC, as previously determined from bone and tissue samples from the Iceman himself. The 2 exceptional dates from wooden artifacts suggest that the site of the Iceman was used as a mountain pass for millennia prior to and after the lifetime of “Ötzi”. For a 2nd sample complex, we studied logs from the beginning of salt mining in the world's oldest salt mines at Hallstatt in Upper Austria. 14C AMS measurements were performed in Vienna on spruce samples found in the prehistoric mines and from a log-house on the surface. Data evaluation included “wiggle matching” of different sets of tree rings. The results suggest that salt mining in the Hallstatt region took place in the 14th–13th century BC, well before the so-called Hallstatt period. We discuss in some detail the chemical pretreatment of the samples and the data evaluation. We also present a comprehensive survey of 14C dates available in the literature concerning both botanical remains from the vicinity of the Iceman and from the earliest salt mining in Hallstatt.

Abstract:The Epigravettian human remains from Riparo Tagliente in northern Italy represent some of the earliest evidence of human occupation in the southern Alpine slopes after the Last Glacial Maximum. Genomic analyses of the 17,000-year-old Tagliente 2 mandible revealed the oldest presence of a genetic profile with affinities to the Near East in the Italian peninsula, which later became the most widespread hunter-gatherer ancestry across Europe. However, a comparable biomolecular characterization of the Tagliente 1 burial remains unavailable, preventing us from defining its biological relationships with Tagliente 2. Here, we apply paleogenomic, isotopic, and radiocarbon dating analyses on a femur fragment of Tagliente 1 and compare the reconstructed data with previously reported results from Tagliente 2. Despite their different isotopic signatures and non-overlapping radiocarbon dates, we reveal that the two human remains belong to the same male individual. We determine that the distinct isotopic values can be explained by different dietary practices during lifetime, whereas the non-overlapping radiocarbon dates can be caused by minimal radiocarbon contamination, possibly deriving from chemical treatments for conservation purposes. These findings highlight the importance of interdisciplinary biomolecular studies in offering new perspectives on the Palaeolithic fossil record and addressing long-standing bioarchaeological questions.

Abstract:Several reports indicate that protein kinase C (PKC) plays a role in insulin-induced glucose transport in certain cells. The precise effects of insulin on specific PKC isoforms are as yet unknown. Utilizing primary cultures of rat skeletal muscle, we investigated the possibility that insulin may influence the activation state of PKC isoenzymes by inducing their translocation and tyrosine phosphorylation. This, in turn, may mediate insulin effects on glucose transport. We identified and determined the glucose transporters and PKC isoforms affected by insulin and 12-O-tetradecanoylphorbol-13-acetate (TPA). Insulin and TPA each caused an increase in glucose uptake. Insulin translocated GLUT3 and GLUT4 without affecting GLUT1. In contrast, TPA translocated GLUT1 and GLUT3 without affecting GLUT4. Insulin translocated and tyrosine phosphorylated and activated PKC-beta2 and -zeta; these effects were blocked by phosphatidylinositol 3-kinase (PI3K) inhibitors. TPA translocated and activated PKC-alpha, -beta2, and -delta; these effects were not noticeably affected by PI3K inhibitors. Furthermore, wortmannin significantly inhibited both insulin and TPA effects on GLUT translocation and glucose uptake. Finally, insulin-induced glucose transport was blocked by the specific PKC-beta2 inhibitor LY379196. These results indicate that specific PKC isoenzymes, when tyrosine-phosphorylated, are implicated in insulin-induced glucose transport in primary cultures of skeletal muscle.
The “angioletti” of Palermo: the health and development of mummified non-adults in late modern Palermo, Sicily (1787–1880 CE)

Abstract:The Capuchin Catacombs in Palermo, Sicily, have been home to non-adult mummified remains since the seventeenth century CE. Despite the increasing numbers of scientific studies conducted at this site, very little research has focused specifically on the youngest members of late modern (1787–1880 CE) society. This research aims to redress the balance by examining 43 individuals to gain insight into the demographic profile of mummified non-adults, to characterize their health status and possible cause of death, and to better understand the funerary treatment offered to the youngest members of society. A portable X-ray unit was used to capture anteroposterior and lateral images of each mummy; this facilitated age estimation, the identification of pathological and/traumatic lesions, and evidence of conservation and the mummification process more generally. This study revealed that regardless of age and health status at the time of death, the mortuary rite performed was primarily influenced by the wealth and social standing of the deceased’s kin. No demographic trends were observed in the data and the lack of evidence of metabolic, neoplastic, and traumatic bone lesions suggest these non-adults died from short-term, acute illnesses. Even when individuals did display evidence of chronic health conditions that would have impacted their day-to-day lives (e.g., B035), they were not excluded from this mortuary tradition on the basis of their long-term health and care requirements in life. Artifacts were found with all individuals examined and were associated with the mummification process, conservation of mummies, and/or their display. This research has ultimately demonstrated that non-invasive imaging can be used to gain a more comprehensive understanding of the lives and deaths of non-adults inhabiting late modern Palermo.

A probable case of Hand-Schueller-Christian's disease in an Egyptian mummy revealed by CT and MR investigation of a dry mummy

Abstract:The challenging mission of paleopathologists is to be capable to diagnose a disease just on the basis of limited information gained by means of one or more paleodiagnostic techniques. In this study a radiologic, anthropologic and paleopathologic analysis of an ancient Egyptian mummy through X-rays, CT and MR was conducted. An Ancient Egyptian mummy ("Mistress of the house", Archeological Museum, Zagreb, Croatia) underwent digital radiography, computed tomography and magnetic resonance imaging employing 3-dimensional ultra-short-echo time (UTE) sequence, that allows to image ancient dry tissue. Morphological observations on the skull and pelvis, the stages of epiphyseal union and dental wear indicated that the remains are those of a young adult male. Multiple osseous lytic lesions were observed throughout the spine as well as on the frontal, parietal, and occipital bone, orbital wall and the sella turcica of the sphenoid. Considering the sex and age of the individual and the features of the lesions, the authors propose the diagnosis of Hand-Schueller-Christian's disease. This is the first study to have effectively used MR images in the differential diagnosis of a disease. It also confirmed the CT value in revealing central nervous system involvement just by detecting skeletal lesions. Although the mummy was previously dated to 3rd century B.C. based on the properties of the sarcophagi, the sex of the mummy suggests that it was most probably transferred into these sarcophagi in later times. The mummification techniques used and radiometric data (C14) dated it to 900-790. B.C.

Abstract:Recent excavations at Ranis (Germany) identified an early dispersal of Homo sapiens into the higher latitudes of Europe by 45,000 years ago. Here we integrate results from zooarchaeology, palaeoproteomics, sediment DNA and stable isotopes to characterize the ecology, subsistence and diet of these early H. sapiens . We assessed all bone remains ( n = 1,754) from the 2016–2022 excavations through morphology ( n = 1,218) or palaeoproteomics (zooarchaeology by mass spectrometry ( n = 536) and species by proteome investigation ( n = 212)). Dominant taxa include reindeer, cave bear, woolly rhinoceros and horse, indicating cold climatic conditions. Numerous carnivore modifications, alongside sparse cut-marked and burnt bones, illustrate a predominant use of the site by hibernating cave bears and denning hyaenas, coupled with a fluctuating human presence. Faunal diversity and high carnivore input were further supported by ancient mammalian DNA recovered from 26 sediment samples. Bulk collagen carbon and nitrogen stable isotope data from 52 animal and 10 human remains confirm a cold steppe/tundra setting and indicate a homogenous human diet based on large terrestrial mammals. This lower-density archaeological signature matches other Lincombian–Ranisian–Jerzmanowician sites and is best explained by expedient visits of short duration by small, mobile groups of pioneer H. sapiens .

Abstract:The reconstruction of past life based on archaeozoology is a challenging domain that offers a range of valuable details concerning former human and animal populations. Additionally, the ancient era is a source of information for human and veterinary medicine, as well as for other biological sciences. This report highlights a pathological lesion identified during the investigation of a horse skeleton from a pit dated La Tène in Alba County (Romania). The left scapula with lesions was collected from the skeleton of a buried 7-8-year-old male horse. The aforementioned bone underwent gross, radiological, and computerized tomography evaluation. Macroscopically, a collar of supraarticular cancellous hyperostosis was detected, most likely as a consequence of an invasive chronic phlegmonous periarthritis and/or bursitis of the infraspinate muscle following probably a penetrating cutaneous wound in the shoulder region. A suppurative periarthritis\bursitis of the infraspinate muscle situated nearby caused, apparently, supraglenoidian periosteitis responsible for osteophytes and exostoses formation in the neck region of the scapula.
"This I Suffered in the Short Space of my Life". The Epitaph for Lucius Minicius Anthimianus (CIG 3272; Peek GVn 1166)

Abstract:Herewith we present an interdisciplinary study of the metrical funerary inscription from the third century CE (CIG 3272; Peek GV 1166). This emotional Greek epitaph reports the short life (from birth to death) of the 4 year old Lucius Minicius Anthimianus. This is the first detailed study since the dissertation by Klitsch (1976). The inscription presents an ideal case for a truly interdisciplinary study of the patient-history, in that its interpretation involves the study of Greek literature and linguistics, epigraphy, social and religious history, and ancient medicine. It also offers ample opportunity to show the contradictions inherent in proposing retrospective diagnosis, without neglecting the relevant information modern medicine has to offer for the interpretation of this case history. We argue that Lucius' father was most probably a physician, that the text of the inscription stems from expert knowledge of ancient medicine and that the traditional retrospective diagnosis of this case, tuberculosis, is an untenable hypothesis.

Biocultural evidence of precise manual activities in an Early Holocene individual of the high-altitude Peruvian Andes

Abstract:Objectives: Cuncaicha, a rockshelter site in the southern Peruvian Andes, has yielded archaeological evidence for human occupation at high elevation (4,480 masl) during the Terminal Pleistocene (12,500-11,200 cal BP), Early Holocene (9,500-9,000 cal BP), and later periods. One of the excavated human burials (Feature 15-06), corresponding to a middle-aged female dated to ~8,500 cal BP, exhibits skeletal osteoarthritic lesions previously proposed to reflect habitual loading and specialized crafting labor. Three small tools found in association with this burial are hypothesized to be associated with precise manual dexterity. Materials and methods: Here, we tested this functional hypothesis through the application of a novel multivariate methodology for the three-dimensional analysis of muscle attachment surfaces (entheses). This original approach has been recently validated on both lifelong-documented anthropological samples as well as experimental studies in nonhuman laboratory samples. Additionally, we analyzed the three-dimensional entheseal shape and resulting moment arms for muscle opponens pollicis. Results: Results show that Cuncaicha individual 15-06 shows a distinctive entheseal pattern associated with habitual precision grasping via thumb-index finger coordination, which is shared exclusively with documented long-term precision workers from recent historical collections. The separate geometric morphometric analysis revealed that the individual's opponens pollicis enthesis presents a highly projecting morphology, which was found to strongly correlate with long joint moment arms (a fundamental component of force-producing capacity), closely resembling the form of Paleolithic hunter-gatherers from diverse geo-chronological contexts of Eurasia and North Africa. Discussion: Overall, our findings provide the first biocultural evidence to confirm that the lifestyle of some of the earliest Andean inhabitants relied on habitual and forceful precision grasping tasks.

Abstract:Oxygen isotope ratios of organisms are closely linked to the hydrological environment in which they grow up. This is especially the case for mammals. Mammal-bone phosphate is formed at a constant body temperature and, thus, relatively insensitive temperature-dependent isotope fractionation. The applicability of oxygen isotopes from mammal-bone phosphate for environmental reconstructions is tested here using bones of deer, domestic pig, and domestic cattle from 16 archaeological sites situated along a North-South transect crossing the Alps. Bones of 118 specimens in total were analyzed, which covered an age span from the Late Neolithic to the Roman period. The main control on oxygen-isotope ratios was site altitude. Significant negative correlations between phosphate-isotope values and altitude were registered especially for cattle and pig. Possibly nonlocal pig bones from one site were deviating from this altitudinal pattern and were excluded from further correlations. Modern equations for translating oxygen-isotope ratios of the three species to those of source water were applied. The reconstructed source water isotopic composition showed a similar altitudinal relation as modern Alpine precipitation. As a consequence, our study confirms the utility of oxygen-isotope ratios of mammal-bone phosphate for palaeoaltitudinal reconstructions.

Abstract:Worldwide, the study and examination of human remains and the circumstances of their acquisition for anatomical collection have received great interest. As part of provenance research projects, a large number of collections are being investigated to determine whether the human remains have been acquired in a correct or unlawful way because the people could have been killed in order to be used as "anthropological objects" for research purposes and to become so-called "specimens". These topics have also been addressed by the Institute of Anatomy at the University Medical Center Rostock. The role of radiology in this interdisciplinary project will be presented using selected examples. The anatomical collection at the University of Rostock includes 40 human skulls, 14 plaster casts, 6 Egyptian mummy heads, and 1 full-body mummy. In addition to the examination by a historian, an anthropologist, and forensic pathologists, additional computed tomography was carried out on nine skulls and the full-body mummy. Micro-computed tomography was also carried out on seven skulls in order to enable a look behind the mummification material and tissue remains. (Micro-)computed tomography was able to close diagnostic gaps and the results presented some rather unexpected findings. Due to interdisciplinary collaboration, individual fates could be determined, which provided information about the individual's life and death circumstances. None of the examined individuals showed evidence of colonial-era injustice or the use of violence that would have led to their inclusion in the collection. (Micro-)computed tomography was a valuable addition to this provenance research project.

Polar Thigh and Other Medical Observations During the Longest Solo Unsupported One-Way Polar Ski Expedition in Antarctica

Abstract:Antarctic expeditions are exceptional challenges for the human body, and medical issues such as nutritional deficiencies, polar anemia, cold injuries, and even death have been reported. We prospectively investigated medical issues encountered by a 33-year-old female adventurer completing the longest solo unsupported one-way polar ski expedition in Antarctica, covering 1484.53 km in 70 days and 16 h from Hercules Inlet to the Ross Ice Shelf, in temperatures estimated from -12 to -50°C and wind speeds of up to 60 mi/h. The adventurer developed a debilitating overuse musculoskeletal neck injury due to poor environmental conditions (ie, poor visibility, wind, and soft snow) while pulling a heavy sled and continuously holding the neck in a flexed position for checking directions on a mounted compass. The adventurer further developed a cold injury on the left calf (polar thigh) that gradually worsened and became ulcerated and more painful throughout the expedition. Potential risk factors included cold ambient temperatures, exposure to windy conditions, and clothing. After the expedition, this injury was treated with a skin graft. Important changes in body mass were observed (ie, reduction of body mass by 26.7% after the expedition) as well as general fatigue, muscle soreness, and sleep deterioration. This case study contributes to the body of knowledge of medical issues during Antarctic expeditions and provides the first scientific account of an adventurer with polar thigh. Further research into this condition is recommended.

Abstract:<p>Skeletal remains were retrieved from a cave in an arid desert environment. The submitted material consisted of dried grass, soil, and malodorous skeletal remains wrapped in leather. After examination, it was revealed that the smell had not been due to recent decomposition but to adipocere. The cause of death was listed as "undetermined" and a suggested date of death as "unknown." Although adipocere usually requires a damp environment or submersion in water to develop, this case provides further evidence for the rare possibility of adipocere formation in an otherwise very dry environment. Wrapping of the body in leather material had produced an impermeable barrier which had sequestrated and preserved bodily fluids allowing anaerobic decay with fat hydrolysis and adipocere formation. Microenvironments may have potentially significant and quite idiosyncratic effects on decompositional processes which may initially confuse investigations.</p>

Abstract:Objective: To gain insights on possible impairment of a Middle Bronze Age individual with bilateral humerus varus buried with a sword in Northeastern Italy. Materials: A skeleton of a 40-50-year-old male from Olmo di Nogara (Italy) compared to other males from the same necropolis and to Neolithic and Iron Age samples from Italy. Methods: Macroscopic/X-rays analysis for pathological diagnosis and cross-sectional geometric analysis. Results: Both humeri of the individual appear short with destruction of the humeral heads, showing severe osteoarthrosis and flattening of the scapular glenoid cavities. The individual showed appreciable humeral bilateral asymmetry; there is no evidence for sustained immobilization. Conclusions: The pathological modifications suggest a diagnosis of bilateral humeral varism probably following an injury at birth. The individual's life was likely not significantly affected, as evidence suggests that he remained active and possibly used weapons. Significance: Biomechanical analyses provided a useful tool to reconstruct the life of the subject within the community, showing that this individual's apparent upper limb abnormalities did not exempt him from a role as a warrior and highlighting the importance of the warrior identity in this Bronze Age society. Limitations: The complex interaction between epiphyseal damage and shortening of the humerus makes it difficult to assess activity patterns. Only severe impairment leading to long-term immobilization can be excluded for this individual. Suggestions for further research: Cross-sectional geometry may be used in other cases of humerus varus or bone dysplasia to investigate functional impairment.

The application of a functionalized film method allowed us to perform a non-invasive sampling. Paleo-proteomics confirmed the presence of lung tissue-specific proteins (organ specific) as well as the presence of proteins linked to severe inflammation. Paleoproteomics and paleometabolomics further allowed the identification of the main components of Nebiri's embalming recipe: animal fats and glue, balms, essential oils, aromatic plants, heated Pistacia, and coniferous resins. Both the use of Pistacia and coniferous resins in an early 18th Dynasty individual confirm Nebiri's high social status. The technique applied offers a targeted approach to the chemical characterization of human tissues, embalming compounds, and organic materials layering in pottery. The ability of the functionalized film method to harvest all types of compounds, from macromolecules (i.e., proteins) to small molecules (i.e., organic acids) opens a new path in the study of ancient material culture; furthermore, it allows to perform untargeted analysis, which is necessary when no a priori information is available.

The habitat utilization and environmental resilience of Homo heidelbergensis in Europe

Abstract:The European populations of Homo heidelbergensis may have contributed to the genetic heritage of modern Eurasians. A better understanding of the possible effects of palaeoenvironmental alterations on the evolution of ancient humans can help to understand the origin of developed traits. For this purpose, the spatiotemporal alterations of physical factors were modelled in Europe for the period of 670–190 ka, covering the existence of Homo heidelbergensis in Europe. The factors included the following: paleoclimatic conditions, climatic suitability values of ancient humans, two prey species, and the European beech. Furthermore, the distribution and features of wood used for toolmaking were also investigated. Finally, changes in the relative mortality risk, the percentage of the body covered by clothing, and daily energy expenditure values in the coldest quarter of the year were modelled. The results suggest that H. heidelbergensis inhabited dominantly temperate regions in Europe where prey such as red deer were present. In the northern regions, European beech trees were abundant. When making wood tools, they preferred relatively light but not the strongest woods, which were readily available in the vicinity of the sites. Although hard and heavy woods were also occasionally used, at a European level, significant changes were observed in the relative mortality risk, the percentage of the body covered by clothing, and daily energy expenditure values during the period of 670–190 ka. However, substantial differences between archaeological sites in these values, indicating somewhat ecological variations, were not found during the studied period.

Abstract:The use of diagenetic alterations in bone microstructure ('histotaphonomy') as indicators of funerary treatment in the past and for post-mortem interval calculations in forensic cases has received increasing attention in the last decade. Studies have used histological changes to conclude in-situ decomposition, mummification, infanticide and post-mortem interval. There has been very little attempt to experimentally validate the links between decomposition, depositional conditions, time-since-death and microscopic changes in human bone so that meaningful interpretations of archaeological and forensic observations can be made. Here, we address this problem experimentally using the largest sample of human remains from anatomical donors and the longest-term deposition framework to date. This study tests one key assumption of histotaphonomy; that putrefaction during the early stages of decay is reflected in bone microanatomy and composition. Seventeen human donors and six pigs were deposited on the surface in a known Australian environment and left to decompose between 463 and 1238 days. All remains underwent all stages of decomposition reaching skeletonisation. Rib and femur samples were analysed using conventional histological methods and scanning electron microscopy, by applying the Oxford Histological Index, and examining collagen birefringence, microcracking and re- and de mineralisation. Biomolecular changes of the femoral samples were analysed using Fourier-transform infrared (FTIR) spectroscopy. The results indicate that bioerosion in human bone does not occur due to putrefaction. There were no correlations between bone histology and the following variables: human vs pigs, season, primary vs secondary deposition, position, fresh vs frozen and time-since-deposition. Furthermore, no trends were observed between biomolecular changes and time-since-deposition. The study also shows that pigs cannot be used as substitutes for human remains for bone biodegradation research. This is the first, controlled, larger scale study of human remains providing a lack of support for a long-assumed relationship between putrefaction and bone histology bioerosion. Using bone degradation as an argument to prove putrefaction, in-situ decomposition and early taphonomic processes cannot be supported based on the experimental human data presented.
