Abstract:THE HUMAN immunodeficiency virus (HIV) epidemic has moved into the heterosexual population in the United States. This probably started in the 1970s but was making significant inroads by 1982 as determined from a backcalculation model. 1 Given the methodologic difficulties in estimating HIV incidence, trend data may give the most reliable picture of the emerging epidemic. As of March 1992, a total of 12 881 patients with the acquired immunodeficiency syndrome (AIDS) had been reported who acquired their infection through heterosexual intercourse, 5100 men and 7781 women. 2 Women have outnumbered men in the heterosexual contact transmission category in the United States since the epidemic was recognized in 1981. 3 Four-year trends in reported AIDS cases show that women accounted for an increasing proportion of total cases between 1988 (10.4%) and 1991 (12.8%) and heterosexual contact accounted for a greater proportion of female cases each year from 1988 through 1991

Abstract:Treatment with transition metal complexes is an efficient method to fight with cancer. Therefore, a new transition metal complex formulated as [Pd(1, 3-pn)(acac)]Cl (pn and acac stand for propylendiamine and acetylacetonate, respectively) was synthesized and analyzed using 1H NMR, Fourier transform infrared, electronic absorption spectroscopy techniques as well as elemental analysis and conductivity measurement. The geometry optimization, frontier molecular orbital (FMO) analysis, molecular electrostatic potential (MEP), natural bond orbital (NBO) analysis and nonlinear optical (NLO) property were accomplished by density functional theory (DFT) at B3LYP level with 6-311G(d,p)/aug-cc-pVTZ-PP basis set. Preliminary determination of antitumor activity and lipophilicity of this metal complex was performed experimentally and the promising results were obtained. This encouraged us to study the interaction and binding mode/modes of this complex with DNA as the primary receptor for the chemotropic drugs and BSA as the transporter protein in the circulatory system. For this reason, the binding of newly made complex was assessed in-vitro under physiological state using experimental and in-silico molecular modeling studies. So, the CT-DNA binding study of this complex was explored using spectrofluorometric as well as spectrophotometric techniques, viscosity and gel electrophoresis experiments. Furthermore, fluorescence, UV–Vis, Fo¨$$\ddot{\mathrm{o}}$$rster resonance energy transfer and circular dichroism studies were carried out for BSA binding. The experimental and computational interaction studies showed that [Pd(1, 3-pn)(acac)]Cl complex binds to the minor groove of CT-DNA and interacts with BSA by van der Waals forces and hydrogen bond.Graphical abstract

PITIA: an FPGA for throughput-intensive applications

Abstract:In this paper, we present a novel, high throughput field-programmable gate array (FPGA) architecture, PITIA, which combines the high-performance of application specific integrated circuits (ASICs) and the flexibility afforded by the reconfigurability of FPGAs. The new architecture, which targets datapath circuits, uses the concepts of wave steering and pipelined interconnects. We discuss the FPGA architecture and show results for performance, power consumption, clock network performance, and routability. Results for some commonly used datapath designs are encouraging with throughputs in the neighborhood of 625MHz in 0.25-/spl mu/m 2.5-V CMOS technology. Results for random benchmark circuits are also shown. We characterize designs according to their Rent's exponents and argue that designs with predominantly local interconnects are the best fit in PITIA. We also show that as technology scales down toward deep submicron, PITIA shows an increasing throughput performance.
Synthesis, crystal structure, cytotoxicity, in-detail experimental and computational CT-DNA interaction studies of 2-picolinate Pd(II) and Pt(II) complexes

Abstract:A new Pd(II) complex of formula [Pd(en)(2-pyc)]+ (where, en is ethylenediamine and 2-pyc is 2-pyridinecarboxylate anion) and its reported Pt(II) analogue, i.e. [Pt(en)(2-pyc)]+ have been made by an improved synthetic procedure, yielding above 80%. They have been characterized by FT-IR, UV-Vis, 1H NMR, 13C NMR, conductivity and elemental analysis. Single crystal structural determination of [Pt(en)(2-pyc)]+ displayed that the Pt(II) cation in this complex coordinated by 2-pyc and en each as five member chelate resulting in slightly distorted square-planar array. The time-dependent spectroscopic analysis of these compounds in aqueous medium demonstrated their structural stabilities. The cytotoxic activities of Pd(II) and Pt(II) complexes, free 2-pyc and carboplatin (as standard drug) were assayed in-vitro against the HCT-116 and MCF-7 as cancerous and MCF 10 A and CCD-841 as normal cell lines. They showed the IC50 order of: carboplatin > 2-pyc > Pt(II) > Pd(II) and lower activities against non-cancerous cells. CT-DNA binding of the Pd(II), Pt(II) and 2-pyc free ligand were explored individually. In this relation, UV-Vis and fluorescence titrations disclosed quenching of CT-DNA absorption and emissions by the compounds via dynamic mechanism and formation of H-bonds and van der Waals forces between them. The interaction was further validated and verified by viscosity measurements and gel electrophoresis. Partition coefficient determination showed that all three compounds have more lipophilicity than cisplatin. Furthermore, docking analysis and molecular dynamics simulation were done to evaluate the nature of interaction between aforementioned compounds and CT-DNA. The finding results demonstrated that these agents interact with CT-DNA via groove binding and were in agreement with experimental results.Communicated by Ramaswamy H. Sarma.

Abstract:Scientists should not forget that the rate of death as a result of cancer is far more than that of other diseases like influenza or coronavirus (COVID-19), so the research in this field is of cardinal significance. Therefore, a new and hydrophilic palladium(II) complex of the general formula [Pd(bpy)(proli-dtc)]NO3, in which bpy and proli-dtc are 2,2'-bipyridine and pyrroline dithiocarbamate ligands, respectively, was synthesized and characterized utilizing spectral and analytical procedures. Density functional theory (DFT) calculation was also performed with B3LYP method in the gas phase. The DFT and spectral analysis specified that the Pd(II) atom is found in a square-planar geometry. HOMO/LUMO analysis, quantum chemical parameters and MEP surface of the complex were investigated to acquire an intuition about the nature of the compound. Partition coefficient and water solubility determination showed that both lipophilicity and hydrophilicity of the compound are more than cisplatin. The 50% inhibition concentration (IC50) value was evaluated against K562 cancer cells, the obtained result has revealed a promising cytotoxic effect. DNA and BSA binding of the complex were explored through multi-spectroscopic (UV–Vis, fluorescence, FRET, and CD) and non-spectroscopic (gel electrophoresis, viscosity and docking simulation) techniques. The obtained findings demonstrated that the complex strongly interacts with CT-DNA by hydrophobic interactions and possesses medium interaction with BSA via hydrogen bond and van der Waals forces, thus BSA could efficiently carry out complex transportation. Furthermore, the results of docking simulation agree well with the experimental findings. In conclusion, the new Pd(II) complex has cytotoxic activity and could interact with DNA and BSA effectively.Graphical abstract

Abstract:In the present research, the feasibility of a Cobalt(III) complex containing β–amino alcohol ligands for affinity with the target calf thymus DNA is demonstrated. In the title complex, [Co(C 11 H 15 N 2 O 2 ) 2 ]Cl, the Co(III) atom is six-coordinated with four N atoms and two O atoms from (2-[(E)-({2-[(2-Hydroxyethyl) amino]ethyl}imino)methyl]phenol) ligand (L). To investigate the molecular interaction between the synthesized complex and DNA, some multi-spectroscopic approaches associated with molecular docking were employed in the physiological buffer (pH 7.4). The results indicated that the Co(III) complex proved to be a minor groove binder with a preference for the A-T region, which was substantiated by displacement studies with Hoechst33258 and Methylene blue (MB) as minor groove binder and intercalator. In addition, the results of the molecular docking study revealed that the Co(III) complex approached the gap between the DNA minor grooves near the spot where the Hoechst was. Furthermore, the results of the cytotoxicity and apoptosis tests for the MCF-7 cell line were also indicative of the positive effects of the complex on controlling the growth and viability of breast cancer. Abstract:The present article investigates whether the cis and trans isomers of the same compound affect their antitumor and DNA-binding properties. Thus, we have synthesized cis -[Pd(L-Ala) 2 ] ( I ) and trans -[Pd(L-Ala) 2 ] ( II ) (Ala = L-alanine) complexes by modifications to improve the reported procedure. Cytotoxicity tests of these two isomers along with carboplatin (as a known drug) against MCF-7 and HCT-116 (as cancerous) and MCF10A and CCD-841 (as normal cell lines) were performed under the same experimental conditions. The results follow the sequence I > II > carboplatin for both cell lines. DNA-binding studies were carried out in a medium similar to that of physiological conditions (pH = 7 and 0.02 M NaCl). Results of UV–vis absorption provides apparent binding constant, K app , and midpoint transition of native CT-DNA to refolded structure, L 1/2 , suggesting interaction at quite low concentrations of these metal complexes. Fluorescence emission intensity of the CT-DNA-EB system demonstrates static quenching predominates in the binding process. Moreover, negative values of Δ H 0 and Δ S 0 suggested that the hydrogen bonds and van der Waals forces are the interaction forces between the complexes with the CT-DNA groove. Other studies include gel electrophoresis, viscosity experiments, partition coefficient, computational energy minimization, molecular docking and molecular dynamics simulations. All these studies indicate that the cis isomer has better interaction towards CT-DNA and are in accord with fluorescence, UV–vis and cytotoxicity studies. The present work gives valuable indications for further design of cis and trans metal-based isomers as potential anticancer agents.

Abstract:Since the design of metal complexes with better biological activities is important, herein a new palladium(II) complex bearing en and acac (en and acac stand for ethylenediamine and acetylacetonato, respectively) as its ligands, [Pd(en)(acac)]NO 3 complex, was synthesized and fully characterized. Quantum chemical computations of the palladium(II) complex were done via DFT/B3LYP method. Cytotoxicity activity of the new compound on leukemia cell line (K562) was assessed via MTT method. The findings indicated that the metal complex has remarkable cytotoxic effect than cisplatin. OSIRIS DataWarrior software was employed to calculate in-silico physicochemical and toxicity parameters of the synthesized complex which rendered significant results. To comprehend the interaction type of new metal compound with macromolecules, the in depth investigation of interaction of mentioned complex with CT-DNA and BSA was accomplished by fluorescence, UV-Visible absorption spectroscopy, viscosity measurement, gel electrophoresis, FRET analysis and circular dichroism (CD) spectroscopy. On the other hand, computational molecular docking was carried out and the obtained data demonstrated that H-bond and van der Waals forces are the dominant forces for the binding of the compound to the mentioned biomolecules. Molecular dynamics simulation was also done and confirmed the stability of best docked pose of palladium(II) complex inside DNA or BSA over the time and in presence of water solvent. Also, Our own N-layered Integrated molecular Orbital and molecular Mechanics (ONIOM) methodology based on the hybridization of quantum mechanics and molecular mechanics (QM/MM) methodology was accomplished to inquire about binding of Pd(II) complex with DNA or BSA. The in silico studies of the designed complex and its ligands were accomplished by OSIRIS DataWarrior software Interaction with CT-DNA and BSA was assessed by various spectroscopic methods. Molecular docking simulation supported the interaction with both macromolecules. Based on ONIOM analysis, the structures of the complex and biomolecules are altered after binding.
Two Novel Cadmium(ii) Complexes with Demethylcantharate and Polypyridyl: Crystal Structure, Interactions with DNA and Bovine Serum Albumin

Abstract:Two novel mixed-ligand complexes [Cd2(DCA)2(bipy)2(H2O)2] (1) and [Cd2(DCA)2(phen)2]·4H2O (2) (DCA=demethylcantharate, 7-oxabicyclo[2,2,1]heptane-2,3-dicarboxylate, C8H8O5; bipy=2,2′-bipyridine, C10H8N2; phen=phenanthroline, C12H8N2) were synthesized and characterized by elemental analysis, infrared spectra, thermogravimetric analysis and X-ray diffraction. The complexes crystallized in the triclinic crystal system with P1¯ space group. The complex was binuclear molecule. In complex 1, Cd(II) ion was seven-coordinated by two nitrogen atoms from bipy, four oxygen atoms of carboxyl groups from DCA and one water molecule. In complex 2, each Cd(II) ion was six-coordinated by two nitrogen atoms from phen, one ether oxygen atom and three oxygen atoms of carboxyl groups from DCA. DNA binding properties of the complexes were investigated by electronic absorption spectra, fluorescence spectra and viscosity measurements. Results indicated that the complexes bound to DNA via partial intercalation mode. The values of binding constants Kb/(M−1) were 1.03×104 (1) and 5.26×104 (2) at 298K. Meanwhile, the complexes could quench the fluorescence of bovine serum albumin (BSA) through static quenching with the binding constants KA/(M−1) of 2.87×105 (1) and 3.87×105 (2) and the binding site of 0.98 (1) and 0.97 (2). In addition, complex 2 had stronger inhibition ratios than Na2(DCA) against human hepatoma cells (SMMC7721) lines and human gastric cancer cells (MGC80-3) lines in vitro. The IC50 values of complex 2 against the two tested cells were 25.54±6.66 and 19.14±1.05μM, respectively.

Abstract:Cyclic voltammetry (CV) and UV–Visible spectroscopy (UV–Vis) techniques were used to calculate binding parameters of 4-methyl-5-((phenylimino)methyl)-3H-1,2-dithiole-3-thione (MPDT) and 5-(4-fluorophenyl)-3H-1,2-dithiole-3-thione (FPDT) with DNA. The results obtained from both techniques were confirmed by computational molecular docking using AutoDock molecular docking software. The anodic peak potential shift in CV indicated an intercalative mode of binding. The binding constants (M−1) of the adducts MPDT-DNA and FPDT-DNA obtained from voltammetric measurements were found to be 8.0 × 104 and 2.4 × 104, respectively, with binding free energy being −27.99 and −25.01 KJ mol−1, respectively. These results are in good agreement with those obtained from UV–Visible spectroscopic studies. The diffusion coefficients of MPDT and FPDT (2.06 × 10−10 and 2.42 × 10−9, respectively) were found to be higher than those of DNA-bound compounds (1.27 × 10−10 and 1.65 × 10−9 cm2/s, respectively). The binding free energy of MPDT and FPDT to DNA was also calculated by molecular docking study. The docking study gave excellent approximation with experimental results, shedding light on the sites of binding.

Abstract:In this study we have investigated, by combining microbial and microscopical techniques, the adhesion ability of bacteria present in Tuber borchii ectomycorrhizosphere. Our data demonstrate that a common pool of bacteria - Pseudomonas, Bacillus, Micrococcus and Moraxella - occurs in all ectomycorrhizal homogenates and that most of these bacteria are able to attach in vitro to plant cells.

Abstract:A new series of Pd (II) complexes of methyl substituted benzylamine ligands (BLs) has been synthesized and characterized via spectroscopic techniques such as UV/Vis. FTIR, LCMS, 1 H, and 13 C NMR. The UV/Vis study in DMSO, DMSO + water, and DMSO + PBS buffer (pH = 7.2) confirmed their molecular sustainability in liquids. Their in vitro anticancer activity against breast cancer cell lines such as MCF-7 and MDA-MB-231 makes them interesting for in vivo analysis. Their stronger DNA binding activity (DBA) compared with free ligand suggested them as a good DNA binder. DBA was further confirmed by physicochemical studies such as surface tension and viscosity of complex + DNA which inferred the disruption of DNA and intercalation of complexes, respectively. Their % binding activity, % disruption of DNA base pairs (DNABP), and % intercalating strength are reported in this paper for the first time for better understanding of DNA binding mechanism. Along with this, their scavenging activity (SA) determined through DPPH free radical and the results indicate good antioxidant behaviour of complexes.

Abstract:The interaction of poly(diallyldimethyl ammonium chloride) (PDDA) with DNA has been studied by spectroscopic analysis of different kinds of nucleic acid probes, microscopic IR- and CD-spectroscopy. The two spectroscopic changes of ethidium bromide (EB) on its binding to DNA, namely red-shift of its maximum absorption wavelength (νmax) and enhancement of fluorescence intensity are utilized to study the interaction of PDDA with DNA. Reversion of νmax and decrease in fluorescence of DNA–EB system on addition of PDDA show the dissociation of DNA/EB complex and the formation of the DNA/PDDA complex. At the same time, the binding constant of EB and the number of binding sites per nucleotide decrease with increase in the concentrations of PDDA, indicating non-competitive inhibition of EB binding to DNA in the presence of PDDA. From the Scatchard analysis, we obtain the association constant of PDDA to DNA is 4.7×104M−1. Contrary to EB, the binding of PDDA with DNA can greatly enhance the fluorescence intensity of groove-binding probes, Hoechst 33258 and DAPI, which can also be used to investigate the formation of the DNA/PDDA complex. The fluorescence analysis shows that the interaction of DNA with PDDA may have effect on the conformation of secondary structure of DNA. IR-spectra show that PDDA interacts with DNA through both the phosphate groups and the bases of DNA and the formation of DNA/PDDA complex causes the change of the conformation of the DNA secondary structure, which is further proved by CD-spectra.
A mechanism – based perspective on the interplay of new drug candidate with biomolecules

Abstract:This research deals with the synthesis and analysis of a novel Pd(II) compound, [Pd(dach)(SA)], featuring 1,2-diaminocyclohexane (dach) and salicylic acid (SA). The compound's structure was refined and its electronic properties including HOMO-LUMO energies, molecular electrostatic potential and natural bond analysis were calculated using B3LYP functional of DFT. Biologically, the compound demonstrated significant antitumor efficacy against the K562 leukemia cell line (IC 50 = 37 μM). In-silico methods predicted the favorable characteristics and low risks of new compound. Interactions with DNA and serum albumin were explored using both experimental and computational techniques, revealing that the compound binds through hydrogen bonds and van der Waals forces having K b , DNA = 1.7 × 10 5 , ΔG° DNA = - 7.32 kcal/mol, K b , BSA = 1.25 × 10 6 and ΔG° BSA = - 8.36 kcal/mol, as indicated by fluorescence spectroscopy. UV–Vis spectroscopy showed a reduction in the absorption spectra of both DNA and BSA upon interaction. DNA binding affinity was confirmed via gel electrophoresis and viscosity measurements. Molecular docking suggested the groove binding with DNA, and site I of BSA, primarily through hydrogen binding. The stability and conformational changes of these interactions over the time were affirmed by molecular dynamics simulations and QM/MM calculations. These comprehensive studies underscore the potential of [Pd(dach)(SA)] complex as a candidate for cancer therapy, highlighting its strong binding interactions with crucial biomolecules.

Abstract:The binding properties of cationic porphyrin–phenylpiperazine hybrids to calf thymus (CT) DNA were investigated by using absorption, fluorescence and circular dichroism (CD) spectra, and the apparent affinity binding constants (Kapp) of the porphyrins for CT DNA were determined by using a competition method with ethidium bromide (EB). Intercalation of porphyrin into CT DNA occurred when two phenylpiperazines were introduced at cis position onto the periphery of cationic porphyrin. The photocleavages of pBR322 plasmid DNA by the porphyrins were consistent with the values of Kapp. With [porphyrin]/[DNA base pairs] ratio increased, the binding mode tended to be outside binding, and the cleavage abilities of the porphyrins varied. In the presence of sodium azide, a quencher of 1O2, the cleavage of DNA by the porphyrin of intercalation was less inhibited.

Abstract:The properties to be an active drug candidate of the complex Pt(TEEDA)Cl2, C1; Pd(TEEDA)Cl2, C2 and their hydrolysed product [Pt(TEEDA)(OH2)2]2+, C1' and [Pd(TEEDA)(OH2)2]2+, C2' were predicted by Lipinski's rule of 5 and PASS (prediction of activity spectra for substances) web tool. Their structural profile, HOMO-LUMO energy and electronic potential surface ware analysed by DFT calculation. Their TD-DFT spectra were compared with experimental UV-Vis spectra. The hydrolysis mechanisms of C1 & C2 to the diaqua form C1' and C2' were extensively investigated by DFT method in different levels of theory and using CPCM/water model and compared with recognised Pt based anticancer drugs. All the stationary states, including the transition state for the reactions were identified by the DFT calculation. The IRC calculation confirmed that the transition states are well connected and corelate with reactants and products. Interaction of the complexes with DNA & HSA was also investigated by molecular docking study.
Synthesis of a Family of Pd(II) Complexes Using Pyridyl-Ketone Ligands: Crystal Structure, Thermal, Physicochemical, XRD/HSA, Docking, and Heck Reaction Application

Abstract:Four Pd(II) complexes, (dpk)PdCl2 (complex-1), and (dpk)Pd(OAc)2 (complex-2) have been prepared using di(2-pyridyl) ketone as the chelate ligand (dpk). The (dpk·EtOH)PdCl2 (complex-3) and (dpk·EtOH)Pd(OAc)2 (complex-4) were synthesized by selectively introducing complex-1 and complex-2 to an EtOH in situ nucleophilic addition reaction on the O=C of the dpk ligand, respectively. All complexes were characterized using CHN-EA, UV-vis, FT-IR, FAB-MS, EDX, TGA, and NMR physicochemical tools. The XRD-crystallography technique was employed to ascertain the structure of complex-3. The analysis revealed a monoclinic/P21/c crystal system characterized by a square planar structure oriented in the cis direction around the Pd center. Several C-H···Cl and O-H···O H-bonds constructing 2D-S12 and S7 synthons were confirmed via XRD/HSA interactions. The influence of EtOH addition to the O=C group of dpk in (dpk)PdCl2 was documented by using UV-vis/FT-IR spectra and TGA analysis. As catalysts, all complexes have demonstrated a notable catalytic function in the Heck reaction, resulting in a high yield under gentle conditions using iodobenzene and methyl acrylate as model reactions. Moreover, the complex-1 and complex-3 docking activity was evaluated against 1BNA-DNA.
Synthesis, characterization, interaction with DNA, and cytotoxic effect in vitro of new mono- and dinuclear Pd(II) and Pt(II) complexes with benzo[d]thiazol-2-amine as the primary ligand

Abstract:A series of novel Pd(II) and Pt(II) complexes, [PdL(2)Cl(2)]·DMF (1), [Pd(2)(L-H)(2)(bpy)Cl(2)]·(H(2)O)(2)·DMF (2), [Pd(2)(L-H)(2)(phen)Cl(2)]·2H(2)O (3), [PtL(2)Cl(2)]·H(2)O (4), [Pt(2)(L-H)(2)(bpy)Cl(2)]·2H(2)O (5), and [Pt(2)(L-H)(2)(phen)Cl(2)]·H(2)O (6), where bpy = 2,2'-bipyridine, phen = 1,10-phenanthroline, and L = 1,3-benzothiazol-2-amine, have been synthesized and characterized. The competitive binding of the complexes to DNA has been investigated by fluorescence spectroscopy. The values of the apparent DNA binding constant, calculated from fluorescence spectral studies, were 3.8 × 10(6) (K(app)(4)), 2.9 × 10(6) (K(app)(1)), 2.4 × 10(6) (K(app)(6)), 2.0 × 10(6) (K(app)(5)), 1.2 × 10(6) (K(app)(3)), and 6.9 × 10(5) (K(app)(2)). The binding parameters for the fluorescence Scatchard plot were also determined. On the basis of the data obtained, it indicates that the six complexes bind to DNA with different binding affinities in the relative order 4 > 1 > 6 > 5 > 3 > 2. Viscosity studies carried out on the interaction of complexes with Fish Sperm DNA (FS-DNA) suggested that all complexes bind by intercalation. Gel electrophoresis assay demonstrates that all the complexes can cleave the pBR 322 plasmid DNA and bind to DNA in a similar mode. The cytotoxic activity of the complexes has been also tested against four different cancer cell lines. The results show that all complexes have activity against KB, AGZY-83a, Hep-G2, and HeLa cells. In general, the Pt(II) complexes were found to be more effective than the isostructural Pd(II) complexes. The mononuclear complexes exhibited excellent activity in comparison with the dinuclear complexes in these four cell lines. Moreover, on the KB cell line (the human oral epithelial carcinoma), the observed result seems quite encouraging for the six complexes with IC(50) values ranging from 1.5 to 8.6 μM. Furthermore, apoptosis assay with hematoxylin-eosin staining shows treatment with the six complexes results in morphological changes of KB cells. The results induce apoptosis in KB cells.
Experimental and Computational Studies on the Interaction of DNA with Hesperetin Schiff Base CuII Complexes

Abstract:The interactions with calf thymus DNA (CT-DNA) of three Schiff bases formed by the condensation of hesperetin with benzohydrazide (HHSB or L1H3), isoniazid (HIN or L2H3), or thiosemicarbazide (HTSC or L3H3) and their CuII complexes (CuHHSB, CuHIN, and CuHTSC with the general formula [CuLnH2(AcO)]) were evaluated in aqueous solution both experimentally and theoretically. UV–Vis studies indicate that the ligands and complexes exhibit hypochromism, which suggests helical ordering in the DNA helix. The intrinsic binding constants (Kb) of the Cu compounds with CT-DNA, in the range (2.3–9.2) × 106, from CuHTSC to CuHHSB, were higher than other copper-based potential drugs, suggesting that π–π stacking interaction due to the presence of the aromatic rings favors the binding. Thiazole orange (TO) assays confirmed that ligands and Cu complexes displace TO from the DNA binding site, quenching the fluorescence emission. DFT calculations allow for an assessment of the equilibrium between [Cu(LnH2)(AcO)] and [Cu(LnH2)(H2O)]+, the tautomer that binds CuII, amido (am) and not imido (im), and the coordination mode of HTSC (O−, N, S), instead of (O−, N, NH2). The docking studies indicate that the intercalative is preferred over the minor groove binding to CT-DNA with the order [Cu(L1H2am)(AcO)] > [Cu(L2H2am)(AcO)] ≈ TO ≈ L1H3 > [Cu(L3H2am)(AcO)], in line with the experimental Kb constants, obtained from the UV–Vis spectroscopy. Moreover, dockings predict that the binding strength of [Cu(L1H2am)(AcO)] is larger than [Cu(L1H2am)(H2O)]+. Overall, the results suggest that when different enantiomers, tautomers, and donor sets are possible for a metal complex, a computational approach should be recommended to predict the type and strength of binding to DNA and, in general, to macromolecules.

Abstract:Me2dpq, a bioactive polypyridyl quinoxaline‐based ligand, and its unique binuclear Co(II)‐Me2dpq complex were synthesized and characterized by single crystal X‐ray diffraction and other spectroscopic approaches. The ligand and Co(II)‐complex were tested for DNA binding, antioxidant scavenging, and cytotoxicity. DNA binding mechanism was investigated using molecular docking. A quinoxaline‐based ligand, 6,7‐dimethyl‐2,3‐di(2‐pyridyl)quinoxaline (Me2dpq), and its unique binuclear Co(II)‐Me2dpq complex were prepared and analyzed using single crystal X‐ray diffraction and several spectroscopy techniques. Hirshfeld surface analyses were also used to clarify the Me2dpq ligand and the Co(II)‐Me2dpq crystal packing. The Co(II)‐Me2dpq complex crystallized in triclinic P‐1 space group and exhibited a distorted trigonal bipyramidal geometry around the cobalt(II) ion. Each Co(II) ion coordinated to a Me2dpq ligand through two N donor centers from pyridyl and quinoxaline rings along with a terminal Cl and two bridged Cl atoms. The binding properties of the ligand and its complex with CT‐DNA were studied by several techniques, namely UV–Vis spectroscopy, fluorescence quenching, viscosity measurements, thermal denaturation, and gel electrophoresis. The CT‐DNA binding interaction tests demonstrated that both the Co(II)‐Me2dpq complex and its parent ligand bind to DNA in an intercalative way. The antioxidant studies of the novel Co(II)‐Me2dpq complex illustrated significant activity against DPPH• and OH•· radicals. The in vitro cytotoxic effect of Me2dpq ligand and its Co(II)‐complex on MCF‐7 and Hep‐G2 tumor cell lines was evaluated by using the MTT assay. The studies showed that the Co(II)‐Me2dpq complex had superior activity toward the tested cell lines. Molecular docking studies of synthesized compounds were employed to figure out the way they would associate with a biologically macromolecular target B‐DNA (PDB: 1BNA). Thus, this study is anticipated to provide novel opportunities for the successful utilization of the synthesized compounds in many medical domains.
