Abstract:Anaerobic digestion (AD) of effluents in sugarcane biorefineries may improve the energy balance in the bioethanol production process and the environmental suitability of wastes disposal. This study analyses the inclusion of AD of vinasse (stillage) into different configurations of sugarcane biorefineries, using technical, economic and environmental metrics. The three base-case scenarios are a first generation (1G) ethanol plant, an integrated first and second generation (1G2G) ethanol plant with alcoholic fermentation of the hemicellulose fraction (C5), and an 1G2G ethanol plant with biodigestion of the C5 fraction. These three base scenarios are compared with the similar ones including the biodigestion of vinasse. The implementation of vinasse biodigestion had positive impact on the productivity and sustainability of sugarcane biorefineries. The use of biogas from vinasse for a 1G ethanol plant increased in 9.20% the surplus of electric energy yielded to the grid. This scenario had a positive net present value, NPV: + 11.5 × 10<sup>6</sup> USD, assuming minimum acceptable rate of return, MARR = 11%. For the 1G2G + C5 fermentation scenario the NPV was + 4.63 × 10<sup>6</sup> USD. Assuming NPV = 0, the IRR for 1G becomes 19.7% and for 1G2G + C5, 13.6%, an IRR higher than the MARR. The inclusion of AD of vinasse when the C5 fraction is biodigested was not an economically feasible option. In all scenarios, vinasse biodigestion reduced environmental impacts, and the 1G2G processes exhibit better results than the consolidated 1G for almost all environmental impact categories. The AD of vinasse can be considered an environmental-friendly and economical-feasible technology to improve the process in sugarcane biorefineries.

Abstract:Vinasse, a residue from bioethanol production containing high organic matter concentration, was used as substrate in submerged fermentation of Pseudomonas aeruginosa PA1 for biosurfactant production. About 2.7 g/L of rhamnolipids was obtained, with surface tension of 29.2 mN/m and critical micelle concentration of 80.3 mg/L. After separation of rhamnolipid and biomass, residual fermentation media were submitted to anaerobic biodegradation in mesophilic conditions. The residual medium derived from fermentation with vinasse diluted to 1 : 1, without addition of nitrogen, C : N 21, and for 168 h, led to 63.2% chemical oxygen demand (COD) removal and 97.6 mL CH4/g CODremoved. Compared to results obtained with fresh vinasse (73.7% COD removal and 112.4 mL CH4/g CODremoved), it could be concluded that both processes can be integrated in order to add value to the residue and obtain energy, reducing production costs and at the same time environmental impacts related to vinasse disposal.

Abstract:Improving anaerobic digestion of sugarcane vinasse - a high-strength wastewater from ethanol distillation - is a subject of great interest, in view of the reduction of the pollutants and recovery of methane and valuable metabolites as byproducts. Through metatranscriptomic analysis, this study evaluated the active microbiome and metabolic pathways in a continuous acidogenic reactor: Stage 1S (control): 100% sucrose-based substrate (SBS); Stage 2SV (acclimation): 50% SBS and 50% vinasse; Stage 3V: 100% vinasse. Metatranscriptome obtained from each Stage was subjected to taxonomic and functional annotations. Under SBS feeding, pH dropped to pH 2.7 and biohydrogen production was observed. As vinasse was added, pH increased to 4.1-4.5, resulting in community structure and metabolite changes. In Stage 3V, biohydrogen production ceased, and propionate and acetate prevailed among the volatile fatty acids. Release of homoacetogenesis enzymes by Clostridium ljungdahlii and of uptake hydrogenase (EC 1.12.99.6) by Pectinatus frisingensis were linked to hydrogen consumption in Stages 2SV and 3V. Metabolic pathways of vinasse compounds, such as carbohydrates, malate, oxalate, glycerol, sulfate and phenol, were investigated in detail. In pyruvate metabolism, gene transcripts of oadA (oxaloacetate decarboxylase) and mdh (malate dehydrogenase), were upregulated in Stage 3V, being mostly attributed to P. frisingensis. Acetate formation from vinasse degradation was mainly attributed to Megasphaera and Clostridium, and propionate formation to P. frisingensis. Glycerol removal from vinasse exceeded 99%, and gene transcripts encoding for glpF (glycerol uptake facilitator protein), glpK (glycerol kinase) and glpABC (glycerol-3-phosphate dehydrogenase) were expressed mostly by Pectinatus and Prevotella. mRNA profiling showed that active bacteria and gene expression greatly changed when vinasse replaced sucrose, and Pectinatus was the main active bacterium degrading the searched compounds from vinasse. The identification of the main metabolic routes and the associated microorganisms achieved in this work contributes with valuable information to support further optimization of fermentation towards the desired metabolites.

Abstract:Anaerobic digestion (AD) of residues from integrated first- and second-generation ethanol (1G2G) biorefineries is a sustainable method for energy recovery through biogas production. This study evaluated the co-digestion of 1G vinasse, 2G vinasse and pentose liquor (from the pretreatment of sugarcane bagasse for 2G ethanol production) compared to individual digestions using biochemical methane potential (BMP) assays. The results showed some “key” micronutrients from the substrates that affected methane (CH4) production, while their balance provided by co-digestion achieved high digestibility (95%). High iron (Fe) and nickel (Ni) concentrations, in addition to furfural (0.33 g L−1) in pentose liquor seemed to decrease its CH4 production potential. Despite these adverse effects observed in mono-digestion, co-digestion was beneficial for this substrate, increasing digestibility (52%) and BMP (118%). The highest BMP was observed in vinasse 2G (631 ± 6 NmL CH4 gTVS−1), with no significant difference compared to the adjusted modified Gompertz model (624 ± 10 NmL CH4 gTVS−1). The co-digestion system also presented the highest specific CH4 production rate (20 ± 1 NmL CH4 gTVS−1day−1) and shortened the lag phase by 19% compared to the AD of isolated 1G vinasse with the second lowest BMP value (494 ± 11 NmL CH4 gTVS−1).
Sugarcane wastes as microbial feedstocks: A review of the biorefinery framework from resource recovery to production of value-added products

Abstract:Sugarcane industry is a major agricultural sector capable of producing sugars with byproducts including straw, bagasse, and molasses. Sugarcane byproducts are no longer wastes since they can be converted into carbon-rich resources for biorefinery if pretreatment of these is well established. Considerable efforts have been devoted to effective pretreatment techniques for each sugarcane byproduct to supply feedstocks in microbial fermentation to produce value-added fuels, chemicals, and polymers. These value-added chains, which start with low-value industrial wastes and end with high-value products, can make sugarcane-based biorefinery a more viable option for the modern chemical industry. In this review, recent advances in sugarcane valorization techniques are presented, ranging from sugarcane processing, pretreatment, and microbial production of value-added products. Three lucrative products, ethanol, 2,3-butanediol, and polyhydroxyalkanoates, whose production from sugarcane wastes has been widely researched, are being explored. Future studies and development in sugarcane waste biorefinery are discussed to overcome the challenges remaining.
Thermophilic Anaerobic Digestion of Vinasse in Pilot Plant UASB Reactor

Abstract:Vinasse from sugar-cane alcohol production is generated in great amounts in Brazil and is disposed on soil “in natura”. Only a few distilleries use mesophilic anaerobic digestion, so far, for gas recovery and vinasse treatment or conditioning, due to an unfavorable economical balance. Thermophilic anaerobic digestion was studied in a large-scale pilot-plant (75 m3 UASB reactor), since vinasses are generated at high temperatures, and no experiments on this subject are reported in the literature, except for a few laboratory assays using pure substrates and other wastewaters. This work demonstrates the feasibility of sugar-cane vinasse thermophilic anaerobic digestion in UASB reactors. The reactor was operated for 280 days, as far as possible like a full-scale plant; at applied organic loading rates of 25-30 kg COD/m3.day, 72% COD removal and 10 Nm3gas/m3.day could be achieved. Granulation of the sludge was essential in order to maintain good stability of the process; almost no soda was necessary for pH control. For full-scale implementation, re-start of the thermophilic process, after several months without feeding, has to be studied yet.

Abstract:The energy crisis resulted in increasing awareness that alternative sources of energy should be considered. During this time, Brazil implemented ethanol production from sugarcane as biofuel. However, during this process, large amounts of residues are generated, such as vinasse. This residue can be treated anaerobically to generate methane as a source of bioenergy with the use of sequencing batch reactors operated with immobilized biomass (AnSBBR). In this work, tests were conducted in an AnSBBR laboratory-scale reactor, and the main results regarding the kinetic model fitting and performance of substrate consumption (83 %), methane content in the biogas (77 %), applied organic load (5.54 g COD L−1 day−1), methane productivity (973 N-mL CH4 L−1 day−1), and yield (9.47 mol CH4 kg COD−1) show that AnSBBR is a promising technological alternative. After tests conducted in a laboratory-scale reactor, an industrial reactor was scaled and was also operated in a sequencing batch with immobilized biomass (AnSBBR) for the anaerobic treatment of vinasse with the goal of generating methane and environmental suitability to further disposal in soil. The calculations were performed based on data from a sugar and alcohol plant located in São Paulo, Brazil. This study proposes to the operation of the industrial scale reactor was the association of four AnSBBR (each one with a volume of 15849 m3) operating in parallel (with a feeding and discharge time of 4 h and a reaction time of 8 h), with the goal of adapting the treatment system from a discontinuous operation to a continuous operation. In this industrial scenario, the methane production was estimated at 1.65 × 106 mol CH4 day−1, and the energy was approximately 17 MW, increasing the possible energy recovery contained in sugarcane from 93 to 96 %.

Abstract:The use of agro-industrial and urban organic waste to produce energy seems to be one of the most attractive options for the future of energy supply. Anaerobic digestion is a promising process for treating organic waste and producing biogas. Biogas is a mixture of gases, primarily consisting of methane, carbon dioxide, and hydrogen sulfide, that can be used for electrical and thermal energy generation or as fuel. However, it is still necessary to improve the technology associated with its production, adapting it to an efficient, safe, and competitive scale. Various methodologies are used to support the decision on process parameters and operating conditions to find the most effective and sustainable ones. In this study, an exergy analysis was performed to determine the exergetic efficiency and exergy destruction in the biogas process produced from the anaerobic digestion of vinasse. The analysis was performed through mass, energy, and exergy balances using literature data and computer simulation and considering the use of a continuous stirred tank reactor. The results show that the exergy destroyed in the CSTR biodigester was 1338.9 kW, accounting for 57.2% of the overall exergy destruction in the process. The heat exchanger showed the highest exergy efficiency (99.95%), with only 5.77 kW of exergy being destroyed. The authors suggest for future works that it would be interesting to perform a life cycle assessment (LCA) and exergoeconomic analysis to evaluate the environmental and economic impacts associated with biogas production from the anaerobic digestion of sugarcane vinasse.

Abstract:This paper reviews the recent achievements in the enhanced production of volatile fatty acids (VFAs) from waste activated sludge (WAS). The enhanced strategies are divided into two approaches. The first strategy focuses on the regulation of carbon-to-nitrogen (C/N) ratio by co-digestion of WAS with carbon-rich substrates, including municipal solid wastes (MSW), marine algae, agricultural residues, and animal manures. The other strategy is to enhance the solubilization and hydrolysis of WAS or inhibit the methanogenesis by applying various pretreatments, such as mechanical, chemical, enzymatic, and thermal pretreatment. Finally, the applications of WAS-derived VFAs are discussed. The future researches in enhancing VFAs production and wide application of the VFAs from both technical and economic perspectives are proposed.

Abstract:The demand for new products derived from agro-industrial residues has increased recently. Furthermore, vinasse, a wastewater from ethanol production, needs treatment to be reused in the sugarcane industry, reducing industrial water consumption. This study performed vinasse filtration with charcoal from industrial sugarcane residues and used the filtered molasses dilution in ethanol fermentation. There were five treatments in randomized blocks with three repetitions. The treatments were deionized water and natural vinasse as positive and negative controls, respectively, and filtered vinasse from charcoal made from bamboo, sugarcane bagasse, and straw. Hence, fermentation for ethanol production was performed. The filtered vinasse with all types of charcoals showed lower soluble solids, total residual reducing sugar, higher ethanol concentration, and fermentative efficiency compared to in-nature vinasse in the wine. Filtered vinasse from bagasse and straw charcoals shows 81.14 and 77.98% efficiency in ethanol production, respectively, close to deionized water (81.49%). For an industrial proposal, vinasse filtration may occur in a continuum process with recovery of charcoal by regeneration. Charcoal renewal should increase porosity for subsequent vinasse filtration. The process is feasible to apply using a product of sugarcane residue to treat wastewater and reduce industrial water consumption and vinasse disposal.

Abstract:The process of producing sustainable aviation fuel (SAF) from organic waste involves the use of volatile fatty acids (VFAs) as intermediates that are obtained via arrested anaerobic digestion (AAD) and VFA recovery. This recovery process often requires several steps, including dewatering, filtration, extraction, and purification. The recovery of VFAs is crucial for their upgrading and can pose a challenge in the production of SAF from organic waste due to high costs and compatibility issues. This review discusses various dewatering methods, including centrifuges, belt filter presses, and screw presses, and explores conditioning technologies that can improve dewatering performance. It also introduces filtration technologies, with a focus on dynamic filtration, which shows promise in addressing the issue of membrane fouling. Additionally, the review describes extraction technologies such as electrodialysis, adsorption, and liquid–liquid extraction (LLE). By providing insights into these different techniques, the review aims to contribute to the development of an integrated VFA recovery process with low carbon footprint.

Vinasse from sugarcane bagasse (hemicellulose) acid hydrolysate and molasses supplemented: biodegradability and toxicity

Abstract:Vinasse sugarcane is a valuable byproduct of the ethanol production process, presenting a perspective of volume increase with the development of second generation ethanol (2GE). However, this byproduct needs new methods of treatment and management for sustainability. Besides that, 2GE vinasse can be associated with some compounds (such as furan derivatives, phenolic compounds and organic acids), depending on the process used to solubilize hemicellulose, which could compromise vinasse destination or utilization. For this reason, detoxification methods of the hemicellulosic hydrolysates, from which vinasse is obtained in subsequent steps, are crucial. This study aimed to investigate whether the biological detoxification of vinasse from 2GE presents a difference concerning the microbial activity of biodegradation and toxicity when compared to vinasse without the detoxification process. Two vinasses (1, before; and 2, after detoxification) from fermented sugarcane bagasse (hemicellulose fraction) acid hydrolysate (supplemented with its molasses), under different concentrations: 2.5; 5 e 10% were evaluated. Their physicochemical characterization, biodegradation microbial activity (through Bartha and Pramer respirometric method, with total count of heterotrophic bacteria and fungi), and toxicity evaluation (through bioassays with Lactuca sativa at concentraction: 2.5; 5 e 10% and Daphnia similis to 1.5; 2.5; 3.5; 4.5; 5 and 10%) were performed. The results indicated high mineral and organic matter, which under a specific circumstance (2.5% of soil conditioning), enabled high efficiency in biodegradation (>80%). The bioassays with L. sativa signaled negative effect for radicular growth when the vinasses were applied at a concentration of 5 and 10% (sublethal effect and delayed root growth). Acute effects were observed in D. similis, with 50% of immobilization, at concentrations of 4.13% and 4.74% for vinasses 1 and 2, respectively. These results indicate that the biodegradation of vinasse from sugarcane bagasse acid hydrolysate occurs at relatively low levels (up to 5%) and suggests that higher concentrations (≥10%) may impair the growth of soil-associated microorganisms.
Two problems in one shot: Vinasse and glycerol co-digestion in a thermophilic high-rate reactor to improve process stability even at high sulfate concentrations

Abstract:Anaerobic co-digestion (AcoD) of sugarcane vinasse and glycerol can be profitable because of the destination of two biofuel wastes produced in large quantities in Brazil (ethanol and biodiesel, respectively) and the complementary properties of these substrates. Thus, the objective of this study was to assess the effect of increasing the organic loading rate (OLR) from 2 to 20 kg COD m-3 d-1 on the AcoD of vinasse and glycerol (50 %:50 % on a COD basis) in a thermophilic (55 °C) anaerobic fluidized bed reactor (AFBR). The highest methane production rate was observed at 20 kg COD m-3 d-1 (8.83 L CH4 d-1 L-1), while the methane yield remained stable at around 265 NmL CH4 g-1 CODrem in all conditions, even when influent vinasse reached 1811 mg SO42- L-1 (10 kg COD m-3 d-1). Sulfate was not detected in the effluent. Bacterial genera related to sulfate removal, such as Desulfovibrio and Desulfomicrobium, were observed by means of shotgun metagenomic sequencing at 10 kg COD m-3 d-1, as well as the acetoclastic archaea Methanosaeta and prevalence of genes encoding enzymes related to acetoclastic methanogenesis. It was concluded that process efficiency and methane production occurred even in higher sulfate concentrations due to glycerol addition.
Harnessing the Energy Potential and Value-Added Products from the Treatment of Sugarcane Vinasse: Maximizing Methane Production Through Co-Digestion with Sugarcane Molasses and Enhanced Organic Loading

Abstract:This study assessed the impact of organic loading rate (OLR) on methane (CH 4 ) production in the anaerobic co-digestion (AcoD) of sugarcane vinasse and molasses (SVM) (1:1 ratio) within a thermophilic fluidized bed reactor (AFBR). The OLR ranged from 5 to 27.5 kg COD.m −3 .d −1 , with a fixed hydraulic retention time (HRT) of 24 h. Organic matter removal varied from 56 to 84%, peaking at an OLR of 5 kg COD.m −3 .d −1 . Maximum CH 4 yield (MY) (272.6 mL CH 4 .g −1 COD rem ) occurred at an OLR of 7.5 kg COD.m −3 .d −1 , while the highest CH 4 production rate (MPR) (4.0 L CH 4 .L −1 .d −1 ) and energy potential (E.P.) (250.5 kJ.d −1 ) were observed at an OLR of 20 kg COD.m −3 .d −1 . The AFBR exhibited stability across all OLR. At 22.5 kg COD.m −3 .d −1 , a decrease in MY indicated methanogenesis imbalance and inhibitory organic compound accumulation. OLR influenced microbial populations, with Firmicutes and Thermotogota constituting 43.9% at 7.5 kg COD.m −3 .d −1 , and Firmicutes dominating (52.7%) at 27.5 kg COD.m −3 .d −1 . Methanosarcina (38.9%) and hydrogenotrophic Methanothermobacter (37.6%) were the prevalent archaea at 7.5 kg COD.m −3 .d −1 and 27.5 kg COD.m −3 .d −1 , respectively. Therefore, this study demonstrates that the organic loading rate significantly influences the efficiency of methane production and the stability of microbial communities during the anaerobic co-digestion of sugarcane vinasse and molasses, indicating that optimized conditions can maximize energy yield and maintain methanogenic balance.

Abstract:This review addresses the critical issue of a rapidly increasing worldwide waste stream and the need for sustainable management. The paper proposes an integrated transformation toward a next-generation methanization process, which leads not only to treating waste but also to converting it into higher value compounds and greener energy. Although the current and commonly used anaerobic digestion process is useful for biogas production, it presents limitations of resource exploitation and some negative environmental impacts. Focusing on the acidogenic stage in waste stream processing, the paper discusses the recent strategies to enhance the recovery of volatile fatty acids (VFAs). These acids serve as precursors for synthesizing a variety of biochemicals and biofuels, offering higher value products than solely energy recovery and soil fertilizers. Additionally, the importance of recycling the fermentation residues back into the biorefinery process is highlighted. This recycling not only generates additional VFAs but also contributes to generating clean energy, thereby enhancing the overall sustainability and efficiency of the waste management system. Moreover, the review discusses the necessity to integrate life cycle assessment (LCA) and techno-economic analysis (TEA) to evaluate the environmental impacts, sustainability, and processing costs of the proposed biorefinery.

Abstract:The increasing ethanol production brings many advantages to Brazil. However, it generates high quantities of vinasse from 10 to 15 l per liter of alcohol produced. This residue is applied as a fertilizer in sugarcane crops but, it presents a potential risk to soil quality when high doses of vinasse are applied in the same area over years. So, one solution would be to transport it to distant locations, but this would mean an increase in costs. Thus, concentrated vinasse, wherein the volume is reduced by evaporation process, becomes a viable alternative and is being increasingly used by sugarcane mills in Brazil. The aim of this study was to evaluate the net nitrogen mineralization and potential nitrogen mineralization of soil that received three doses of concentrated and not concentrated vinasse. The not concentrated vinasse is a good alternative to be applied, but higher doses promote N losses by denitrification due to the high water content. The highest values of potential nitrogen mineralization and, lowest values of constant of mineralization in the treatments with concentrated vinasse indicate that this residue releases N at a slower speed than in treatments with not concentrated vinasse, suggesting that there will be N availability in the subsequent crops.

Abstract:pH is notably known as the main variable defining distinct metabolic pathways during sugarcane vinasse dark fermentation. However, different alkalinizing (e.g. sodium bicarbonate; NaHCO3) and/or neutralizing (e.g. sodium hydroxide; NaOH) approaches were never directly compared to understand the associated impacts on metabolite profiles. Three anaerobic structured-bed reactors (AnSTBR) were operated in parallel and subjected to equivalent operational parameters, except for the pH control: an acidogenic-sulfidogenic (R1; NaOH + NaHCO3) designed to remove sulfur compounds (sulfate and sulfide), a hydrogenogenic (R2; NaOH) aimed to optimize biohydrogen (bioH2) production, and a strictly fermentative system without pH adjustment (R3) to mainly evaluate lactic acid (HLa) production and other soluble metabolites. NaHCO3 dosing triggered advantages not only for sulfate reduction (up to 56%), but also to enhance the stripping of sulfide to the gas phase (75-96% of the theoretical sulfide produced) by the high and constant biogas flow resulting from the CO2 released during NaHCO3 dissociation. Meanwhile, molasses-based vinasse presented higher potential for bioH2 (up to 4545 mL-H2 L-1 d-1) and HLa (up to 4800 mg L-1) production by butyric-type and capnophilic lactic fermentation pathways. Finally, heterolactic fermentation was the main metabolic route established when no pH control was provided (R3), as indicated by the high production of both HLa (up to 4315 mg L-1) and ethanol (1987 mg L-1). Hence, one single substrate (from which one single source of inoculum was originated) offers a wide range of metabolic possibilities to be exploited, providing substantial versatility to the application of anaerobic digestion in sugarcane biorefineries.
Techno-economic and Life Cycle Assessments for Bioenergy Recovery from Acid-Hydrolyzed Residues of Sugarcane Bagasse in the Biobased Xylose Production Platform

Abstract:The acid-hydrolyzed residues of lignocellulosic biomass are the primary organic solid wastes during xylose production, while their management and/or valorization have often been overlooked. Here, based on the xylose production platform, four scenarios were established for techno-economic and life cycle assessments (TEA-LCA) of sugarcane bagasse-derived hydrolysis residue obtained from H2SO4 pretreatment, including landfill (baseline), incineration with cogeneration system, bioethanol production, and biomethane production. According to the TEA results, the landfill scenario exhibited the lowest economic benefits, with an internal rate of return (IRR) of 18.8% and a net present value (NPV) of 23,270,090 $. In contrast, the incineration scenario showed the best economic results, with IRR and NPV improving by 4.7% and 68.8%, respectively. The bioethanol and biomethane scenarios exhibited an IRR of 0.2% and 2.2% higher than the IRR of the landfill, respectively. Furthermore, the economics of all four scenarios were sensitive to plant capacity, hemicellulose to xylose conversion rate, and xylose price. The LCA showed that landfill was the worst option, followed by bioethanol and incineration, while the biomethane scenario reduced 39.3% of the adverse impacts using the ReCiPe single score, as it avoided a higher amount of production of synthetic fertilizer and natural gas. The environmental performance was sensitive to the conversion rate of hemicellulose to xylose as per the sensitivity analysis. This study indicated that the valorization of acid-hydrolyzed residues into bioenergy was attractive for the xylose plant, and give the decision-makers a clear perspective for developing a sustainable disposal policy for acid-hydrolyzed residues.
