Abstract:Hydrogen sulfide (H2S) and nitric oxide (NO) are important signaling molecules that regulate several physiological functions. Understanding the chemistry behind their interplay is important for explaining these functions. The reaction of H2S with S-nitrosothiols to form the smallest S-nitrosothiol, thionitrous acid (HSNO), is one example of physiologically relevant cross-talk between H2S and nitrogen species. Perthionitrite (SSNO(-)) has recently been considered as an important biological source of NO that is far more stable and longer living than HSNO. In order to experimentally address this issue here, we prepared SSNO(-) by two different approaches, which lead to two distinct species: SSNO(-) and dithionitric acid [HON(S)S/HSN(O)S]. (H)S2NO species and their reactivity were studied by (15)N NMR, IR, electron paramagnetic resonance and high-resolution electrospray ionization time-of-flight mass spectrometry, as well as by X-ray structure analysis and cyclic voltammetry. The obtained results pointed toward the inherent instability of SSNO(-) in water solutions. SSNO(-) decomposed readily in the presence of light, water, or acid, with concomitant formation of elemental sulfur and HNO. Furthermore, SSNO(-) reacted with H2S to generate HSNO. Computational studies on (H)SSNO provided additional explanations for its instability. Thus, on the basis of our data, it seems to be less probable that SSNO(-) can serve as a signaling molecule and biological source of NO. SSNO(-) salts could, however, be used as fast generators of HNO in water solutions.

Abstract:Generation of S -nitrosothiol (RSNO) and nitric oxide (NO) mediated by zinc(II) coordination motifs are of prime importance for understanding the role of zinc(II)-based cofactors in various redox-signalling pathways. This study uniquely employs a set of mononuclear [ L 2 Zn II ] 2+ cores (where L = Me 4 PzPz/Me 2 PzPy/Me 2 PzQu ) for introducing subtle alterations of the primary coordination sphere and investigates the role of ligand tuning on the transformation of NO 2 – in the presence of thiols. Single crystal X-ray diffraction (SCXRD) analyses on [ L 2 Zn II –X](X) (where X = perchlorate/triflate) illustrate consistent changes in the bond distances, thereby showing variations of the metal–ligand interactions depending on the nature of the heterocyclic donor arms (pyrazole/pyridine/quinoline). Moreover, such tuning of the ligands affects the Lewis-acidity of the [ L 2 Zn II ] 2+ cores as evaluated by 31 P NMR and SCXRD studies on the 1:1 acid-base adducts [ L 2 Zn II (OPEt 3 )] 2+ . Crystallographic and 15 N NMR spectroscopic analyses on the nitrite complexes [ L 2 Zn II (κ 2 -nitrite)](ClO 4 ) reveal that the chemical environment of the nitrite anions in these complexes are nearly identical, despite the dissimilarity in the Lewis-acidity of [ L 2 Zn II ] 2+ cores. Interestingly, RSNO and NO generation from the reactions of [ L 2 Zn II (κ 2 -nitrite)](ClO 4 ) with 4- tert -butylbenzylthiol ( t BuBnSH) exhibits that [( Me 2 PzQu ) 2 Zn] 2+ core is the most efficient in promoting nitrite-thiol interactionsdue to the ease of available hemilabile coordination site at the Lewis acidic [Zn II ]. Detailed UV-vis studies in tandem with computational investigation, for the first time, provide an unambiguous demonstration of nitrous acid (HNO 2 ) intermediate generated through an intramolecular proton-transfer from thiol to nitrite at zinc(II).

Abstract:The redox chemistry of H2S with NO and other oxidants containing the NO group is discussed on a mechanistic basis because of the expanding interest in their biological relevance, with an eye open to the chemical differences of H2S and thiols RSH. We focus on the properties of two "crosstalk" intermediates, SNO- (thionitrite) and SSNO- (perthionitrite, nitrosodisulfide) based in the largely controversial status on their identity and chemistry in aqueous/nonaqueous media, en route to the final products N2O, NO2-, NH2OH/NH3, and S8. Thionitrous acid, generated either in the direct reaction of NO + H2S or through the transnitrosation of RSNO's (nitrosothiols) with H2S at pH 7.4, is best described as a mixture of rapidly interconverting isomers, {(H)SNO}. It is reactive in different competitive modes, with a half-life of a few seconds at pH 7.4 for homolytic cleavage of the N-S bond, and could be deprotonated at pH values of up to ca. 10, giving SNO-, a less reactive species than {(H)SNO}. The latter mixture can also react with HS-, giving HNO and HS2- (hydrogen disulfide), a S0(sulfane)-transfer reagent toward {(H)SNO}, leading to SSNO-, a moderately stable species that slowly decomposes in aqueous sulfide-containing solutions in the minute-hour time scale, depending on [O2]. The previous characterization of HSNO/SNO- and SSNO- is critically discussed based on the available chemical and spectroscopic evidence (mass spectrometry, UV-vis, 15N NMR, Fourier transform infrared), together with computational studies including quantum mechanics/molecular mechanics molecular dynamics simulations that provide a structural and UV-vis description of the solvatochromic properties of cis-SSNO- acting as an electron donor in water, alcohols, and aprotic acceptor solvents. In this way, SSNO- is confirmed as the elusive "yellow intermediate" (I412) emerging in the aqueous crosstalk reactions, in contrast with its assignment to polysulfides, HSn-. The analysis extends to the coordination abilities of {(H)SNO}, SNO-, and SSNO- into heme and nonheme iron centers, providing a basis for best unraveling their putative specific signaling roles.

Spontaneous and Selective Formation of HSNO, a Crucial Intermediate Linking H2S and Nitroso Chemistries

Abstract:Thionitrous acid (HSNO), a potential key intermediate in biological signaling pathways, has been proposed to link NO and H2S biochemistries, but its existence and stability in vivo remain controversial. We establish that HSNO is spontaneously formed in high concentration when NO and H2S gases are mixed at room temperature in the presence of metallic surfaces. Our measurements reveal that HSNO is formed by the reaction H2S + N2O3 → HSNO + HNO2, where N2O3 is a product of NO disproportionation. These studies also suggest that further reaction of HSNO with H2S may form HNO and HSSH. The length of the S-N bond has been derived to high precision and is found to be unusually long: 1.84 Å, the longest S-N bond reported to date for an R-SNO compound. The present structural and, particularly, reactivity investigations of this elusive molecule provide a firm foundation to better understand its potential physiological chemistry and propensity to undergo S-N bond cleavage in vivo.

Abstract:Nitrite-to-NO transformation is of prime importance due to its relevance in mammalian physiology. Although such a one-electron reductive transformation at various redox-active metal sites (e.g., Cu and Fe) has been illustrated previously, the reaction at the [ZnII] site in the presence of a sacrificial reductant like thiol has been reported to be sluggish and poorly understood. Reactivity of [(Bn 3 Tren)ZnII–ONO](ClO4) (1), a nitrite-bound model of the tripodal active site of carbonic anhydrase (CA), toward various organic probes, such as 4-tert-butylbenzylthiol ( t BuBnSH), 2,4-di-tert-butylphenol (2,4-DTBP), and 1-fluoro-2,4-dinitrobenzene (F-DNB), reveals that the ONO-moiety in the [ZnII]–nitrite coordination motif of complex 1 acts as a mild electrophile. t BuBnSH reacts mildly with nitrite at a [ZnII] site to provide S-nitrosothiol t BuBnSNO prior to the release of NO in 10% yield, whereas the phenolic substrate 2,4-DTBP does not yield the analogous O-nitrite compound (ArONO). The presence of sulfane sulfur (S0) species such as elemental sulfur (S8) and organic polysulfides ( t BuBnS n Bn t Bu) during the reaction of t BuBnSH and [ZnII]–nitrite (1) assists the nitrite-to-NO conversion to provide NO yields of 65% (for S8) and 76% (for t BuBnS n Bn t Bu). High-resolution mass spectrometry (HRMS) analyses on the reaction of [ZnII]–nitrite (1), t BuBnSH, and S8 depict the formation of zinc(II)-persulfide species [(Bn 3 Tren)ZnII–S n –Bn t Bu]+ (where n = 2, 3, 4, 5, and 6). Trapping of the persulfide species ( t BuBnSS–) with 1-fluoro-2,4-dinitrobenzene (F-DNB) confirms its intermediacy. The significantly higher nucleophilicity of persulfide species (relative to thiol/thiolate) is proposed to facilitate the reaction with the mildly electrophilic [ZnII]–nitrite (1) complex. Complementary analyses, including multinuclear NMR, electrospray ionization-MS, UV–vis, and trapping of reactive S-species, provide mechanistic insights into the sulfane sulfur-assisted reactions between thiol and nitrite at the tripodal [ZnII]-site. These findings suggest the critical influential roles of various reactive sulfur species, such as sulfane sulfur and persulfides, in the nitrite-to-NO conversion.

Abstract:Third-party payers increasingly are offering insured populations a choice of health insurance packages. This paper addresses the issue of adverse selection: the tendency of high-risk and thus high-cost individuals to disproportionately choose comprehensive plans and low-risk individuals to disproportionately choose plans with limited benefits. The adverse selection issue is first analyzed as it applies generally to multiple-choice plans, and then a methodology is developed for avoiding adverse selection in the case of the Medicare voucher plan. Although developed specifically for the Medicare voucher plan, the proposed pricing system could be applied to any insured population that is offered a multiple-choice insurance plan.
Nitrosopersulfide (SSNO(-)) accounts for sustained NO bioactivity of S-nitrosothiols following reaction with sulfide

Abstract:Sulfide salts are known to promote the release of nitric oxide (NO) from S-nitrosothiols and potentiate their vasorelaxant activity, but much of the cross-talk between hydrogen sulfide and NO is believed to occur via functional interactions of cell regulatory elements such as phosphodiesterases. Using RFL-6 cells as an NO reporter system we sought to investigate whether sulfide can also modulate nitrosothiol-mediated soluble guanylyl cyclase (sGC) activation following direct chemical interaction. We find a U-shaped dose response relationship where low sulfide concentrations attenuate sGC stimulation by S-nitrosopenicillamine (SNAP) and cyclic GMP levels are restored at equimolar ratios. Similar results are observed when intracellular sulfide levels are raised by pre-incubation with the sulfide donor, GYY4137. The outcome of direct sulfide/nitrosothiol interactions also critically depends on molar reactant ratios and is accompanied by oxygen consumption. With sulfide in excess, a 'yellow compound' accumulates that is indistinguishable from the product of solid-phase transnitrosation of either hydrosulfide or hydrodisulfide and assigned to be nitrosopersulfide (perthionitrite, SSNO(-); λ max 412 nm in aqueous buffers, pH 7.4; 448 nm in DMF). Time-resolved chemiluminescence and UV-visible spectroscopy analyses suggest that its generation is preceded by formation of the short-lived NO-donor, thionitrite (SNO(-)). In contrast to the latter, SSNO(-) is rather stable at physiological pH and generates both NO and polysulfides on decomposition, resulting in sustained potentiation of SNAP-induced sGC stimulation. Thus, sulfide reacts with nitrosothiols to form multiple bioactive products; SSNO(-) rather than SNO(-) may account for some of the longer-lived effects of nitrosothiols and contribute to sulfide and NO signaling.

Abstract:A 21-year-old man presented in a state of delirium with compulsive, pathologic laughter and no focal neurologic findings. Brain imaging revealed a lenticulostriate artery infarct of the caudate nucleus. The absence of neurological signs made the diagnosis difficult, but the presence of delirium was a clue to the existence of structural brain disease.

Abstract:Long considered vital to antioxidant defenses, thiol chemistry has more recently been recognized to be of fundamental importance to cell signaling. S-nitrosothiols-such as S-nitrosoglutathione (GSNO)-and hydrogen sulfide (H2S) are physiologic signaling thiols that are regulated enzymatically. Current evidence suggests that they modify target protein function primarily through post-translational modifications. GSNO is made by NOS and other metalloproteins; H2S by metabolism of cysteine, homocysteine and cystathionine precursors. GSNO generally acts independently of NO generation and has a variety of gene regulatory, immune modulator, vascular, respiratory and neuronal effects. Some of this physiology is shared with H2S, though the mechanisms differ. Recent evidence also suggests that molecules resulting from reactions between GSNO and H2S, such as thionitrous acid (HSNO), could also have a role in physiology. Taken together, these data suggest important new potential targets for thiol-based drug development.

Abstract:Recently, it is needed to have high performance and use capacity efficiently at the same time on the heterogeneous disk environment. But it is difficult to do so because the performance and capacity of each disk are different. We think to extend AOD method for this aim. This method uses the difference of access load between primary data and backup data and balance access load and data amount simultaneously on the homogeneous environment. In this paper, we consider to maximize usable capacity by the position of backup and the disk sequencing. Furthermore, we arrange the primary data in order to balance the performance based on its Bandwidth to Space Ratio.

Abstract:Interactions between small inorganic molecules are fundamental to the understanding of basic reaction mechanisms and some of the initial processes of chemical evolution that preceded organic molecules and led to the origin of life. The kinetics of these processes are suitable for the fast generation of a variety of new chemical entities and the propagation of a cascade of chemical reactions, a property that is ideal for signaling purposes even in biological systems. NO and H2S are such molecules that are nowadays recognized as biological gasotransmitters involved in the regulation of physiological functions through protein modifications such as S-nitrosothiol, disulfide, and persulfide formations. In this Viewpoint, we review the current understanding of interactions of NO (and organic and metal nitrosyl species) with H2S, in both chemical and biochemical contexts. Through the formation of HNO, (H)SNO (and its isomers), (H)SSNO, and polysulfides, these two gasotransmitters initiate reaction networks with significant roles in cell signaling. The chemical reactivities and biological effects of these nitrogen and sulfur species are still unresolved, and, thus, a cross-talk between all of them represents a challenging interdisciplinary field that awaits exciting new findings. We tackle some of the intriguing and open questions and provide perspectives for future research directions.
S-Transnitrosation reactions of hydrogen sulfide (H2S/HS-/S2-) with S-nitrosated cysteinyl thiols in phosphate buffer of pH 7.4: Results and review of the literature

Abstract:Cysteine (CysSH) and its derivatives including N-acetylcysteine (NAC) and glutathione (GSH), and cysteine residues in proteins and enzymes are nitrosated with nitric oxide (NO) reaction products such as N2O3 to form S-nitrosated cysteine thiols (RCysSNO). RCysSNO undergo with cysteine thiols (RCysSH) S-transnitrosation reactions, thereby transferring reversibly their nitrosyl (+NO) group to RCysSH to form RCysSNO. •NO release from RCysSNO and S-transnitrosation are considered the most important features and signalling pathways of RCysSNO. Hydrogen sulfide (H2S: pKa1, 7; HS-: pKa2, 12.9) is an endogenous product of cysteine metabolism. We hypothesized that RCysSNO would also undergo S-transnitrosation reaction with H2S/HS-/S2- to form thionitrite (ONS-), the smallest S-nitrosated thiol. This article describes spectrophotometric and mass spectrometric investigations of S-transnitrosation reactions in phosphate buffered saline (PBS) of pH 7.4 between H2S/HS-/S2- (supplied as Na2S) and S-nitrosoglutathione (GSNO), S-nitroso-l-cysteine (CysSNO), S-nitroso-N-acetyl-l-cysteine (SNAC), and the synthetic S-nitroso-N-acetyl-l-cysteine ethyl ester (SNACET). For comparison, we also investigated the reactions of H2S/HS-/S2- with NO+BF4- and NO2+BF4-, direct ON+ and O2N+ donors, respectively, and assumed formation of ONS- and thionitrate (O2NS-), respectively. Addition of Na2S (at 1 mM) to buffered RCysSNO solutions resulted in decreases of the absorbance at 340 nm and concomitant increases in the absorbance at 410 nm depending upon the nature and concentration of RCysSNO (range, 25-1000 μM). The reactivity order of RCysSNO against H2S/HS-/S2- was: CysSNO > SNACET > GSNO > SNAC. Our spectrophotometric and GC-MS analyses indicate that H2S/HS-/S2- and RCysSNO undergo multiple reactions. Major final reaction products were found to be nitrite and nitrate. ONS- and O2NS- were not detected by GC-MS, suggesting rapid and complete S/O-exchange from water at pH 7.4. GC-MS analyses of ethyl acetate extracts of reaction mixtures suggested formation of tetrasulfur (S4), the precursor of elemental sulfur (S8). The broad absorbance around 410 nm and the turbidity occurred in RCysSNO/Na2S reaction mixtures support formation of polysulfides (polysulfanes) and colloidal sulfur. The reaction of NO+BF4- and NO2+BF4- with H2S/HS-/S2- differed from the S-transnitrosation reactions of RCysSNO, with NO+BF4- being more reactive than NO2+BF4-. In this article, we also briefly review and discuss recent published work dealing with the reaction of H2S/HS-/S2- with low- and high-molecular-mass S-nitrosated thiols. This research area is highly challenging and controversial with respect to the primarily formed reaction products. The synthesis of structurally well-characterized, pure stable-isotope labelled species of putative reaction products, including ONS-, O2NS- and ONSS-, and their use in combination with mass spectrometry coupled to chromatography, e.g. GC-MS and LC-MS/MS, are indispensable in exploring the complex interaction of the two gasotransmitters, H2S and •NO.

Abstract:Since the heydays of Reactive Sulfur Species (RSS) research during the first decade of the Millennium, numerous sulfur species involved in cellular regulation and signalling have been discovered. Yet despite the general predominance of organic species in organisms, recent years have also seen the emergence of inorganic reactive sulfur species, ranging from inorganic polysulfides (HSx-/Sx2-) to thionitrous acid (HSNO) and nitrosopersulfide (SSNO-). These inorganic species engage in a complex interplay of reactions in vitro and possibly also in vivo. Employing a combination of spectrophotometry and sulfide assays, we have investigated the role of polysulfanes from garlic during the release of nitric oxide (•NO) from S-nitrosoglutathione (GSNO) in the absence and presence of thiol reducing agents. Our studies reveal a distinct enhancement of GSNO decomposition by compounds such as diallyltrisulfane, which is most pronounced in the presence of cysteine and glutathione and presumably proceeds via the initial release of an inorganic mono- or polysulfides, i.e., hydrogen sulfide (H₂S) or HSx-, from the organic polysulfane. Albeit being of a preliminary nature, our spectrophotometric data also reveals a complicated underlying mechanism which appears to involve transient species such as SSNO-. Eventually, more in depth studies are required to further explore the underlying chemistry and wider biological and nutritional implications of this interplay between edible garlic compounds, reductive activation, inorganic polysulfides and their interplay with •NO storage and release.

Abstract:A smart molecule, QT490, containing thiosemicarbazide moiety acts as a highly selective turn-on in vitro NO sensor through the unprecedented NO-induced transformation of thiosemicarbazide moiety to 1,3,4-oxadiazole heterocycle with the concomitant release of HSNO, thereby eliminating any interference from various endogenous biomolecules including dehydroascorbic acid, ascorbic acid, etc. The kinetic studies of the reactions between QT490 and NO provide a mechanistic insight into formation of HSNO/RSNO from the reaction between H2S/RSH and NO in the biological system. This novel probe is non-cytotoxic, cell permeable, water-soluble, and appropriate for intracellular cytoplasmic NO sensing with the possibilities of in vivo applications.

Abstract:Hard end point studies represent the best available evidence for demonstrating the cardiovascular and renal protection that is achievable with a given treatment in hypertensive patients, yet properly designed end point studies require large cohorts of patients and long follow-up, are expensive, and do not provide any insight on the mechanisms that lead to the clinical manifestations. Studies that are based on the incidence of preclinical alterations, i.e., the surrogate end points, may circumvent these limitations provided that their relationship with the major cardiovascular events is scientifically proved. In this respect, among the many surrogate end points that are under investigation, left ventricular hypertrophy, microalbuminuria, and treatment-induced diabetes seem most promising for replacing the hard end points in view of their undisputed mechanistic relationship with the clinical events and of the mounting evidence indicating that from their changes it is possible to predict the clinical outcome of patients. In addition, the limited resources that are required to carry out this kind of investigations make them preferable to hard end point studies for anticipating the cardiovascular and renal benefit associated with the use of antihypertensive agents.

Abstract:The conversion of NO into its congeners, nitrosonium (NO+) and nitroxyl (HNO/NO-) species, has important consequences in NO metabolism. Dinitrosyl iron complex (DNIC) combined with thiol ligands was shown to catalyze the conversion of NO into NO+, resulting in the synthesis of S-nitrosothiols (RSNO) both in vitro and in vivo. The formation mechanism of DNIC was proposed to involve the intermediate release of nitroxyl. Since the detection of hydroxylamine (as the product of a rapid reaction of HNO/NO- with thiols) is taken as the evidence for nitroxyl generation, we examined the formation of hydroxylamine, RSNO, and nitrite (the product of a rapid reaction of NO+ with water) in neutral solutions containing iron ions and thiols exposed to NO under anaerobic conditions. Hydroxylamine was detected in NO treated solutions of iron ions in the presence of cysteine, but not glutathione (GSH). The addition of urate, a major "free" iron-binding agent in humans, to solutions of GSH and iron ions, and the subsequent treatment of these solutions with NO increased the synthesis of GSNO and resulted in the formation of hydroxylamine. This caused a loss of urate and yielded a novel nitrosative/nitration product. GSH attenuated the urate decomposition to such a degree that it could be reflected as the function of GSH:urate. Results described here contribute to the understanding of the role of iron ions in catalyzing the conversion of NO into HNO/NO- and point to the role of uric acid not previously described.
