Abstract:The endogenous components of okara can affect the extraction and modification of insoluble dietary fiber (IDF). This study was intended to investigate the effect of extraction and purification of IDFs from okara affected by the endogenous components and composite enzyme Viscozyme® L. After that, the physicochemical, functional, and structural characteristics of IDFs were analyzed. The results showed that with the purification of degreasing, deproteinizing and destarching, Viscozyme® L can accurately act on the cell wall of okara, leading to the increase in specific surface area of IDF and the formation of honeycomb-like structure. In accordance with the X-ray diffraction and Fourier-transform infrared spectrum investigations, relative crystallinity decreased and certain cellulose components were reallocated and changed into soluble dietary fiber. Moreover, the modified IDF showed improved water-holding capacity (10.73-11.59 g/g), oil-holding capacity (5.37-6.60 g/g), and swelling capacity (8.99-12.37 mL/g), leading to improved adsorption capacities of glucose (maximum: 39.60 mg/g), cholesterol (14.54-33.56 mg/g at pH 7; 10.94-19.37 mg/g at pH 2), and cholate and better cation-exchange capacity. The outcomes demonstrated the potential for the acquired IDFs, particularly the high-purity modified-IDFs T3 and T4 , to be exploited as functional fiber components in the food sector. PRACTICAL APPLICATION: This study advances the understanding of the internal mechanism of modified insoluble dietary fiber (IDF) with different purities in okara. The obtained high-purity modified IDF can be employed as a functional fiber raw material in the manufacture of functional foods, and can also be utilized to assist in the treatment of some illnesses due to its well-established in vivo function.

Abstract:High purity insoluble dietary fiber (HPIDF) was extracted from Okara by compound enzyme method, and solid emulsifiers with different particle sizes were prepared by wet grinding. Its composition, structure and physicochemical properties were studied, and the influence mechanism of solid emulsifiers with different particle sizes on emulsifying properties and interface stability of Pickering emulsion was systematically studied. The results showed that the particle size of HPIDF decreased significantly, the ζ-potential, contact Angle and swelling capacity of HPIDF ncrease significantly (p < 0.05). HPIDF forms an adsorption layer at the oil-water interface, and some of them are connected to form a bridge network structure, which plays a role of steric hindrance. And the emulsion has excellent stability under different environmental factors. HPIDF are suitable raw materials as natural food-grade solid emulsifiers. It is cost-effective and eco-friendly to realize the high-value utilization of Okara resources, reduce resource waste, and extend the industrial chain.

Abstract:Okara, as a by-product of soybean processing, is rich in insoluble dietary fiber (IDF), which is a carbohydrate polymer with various insoluble polysaccharides. Nowadays, the extraction of IDF with excellent functional properties has become a research hotspot. In this work, we further proposed an alternating alkali/ultrasound method for the efficient extraction of IDF. The sequential treatments of alkali (A-ISF), alkali-ultrasonic (AU-ISF), ultrasonic-alkali (UA-ISF), ultrasonic-alkali-ultrasonic (UAU-ISF) and alkali-ultrasonic-alkali (AUA-ISF) were applied to extract insoluble soybean fiber (ISF). FTIR and XRD results proved the typical structure of ISFs, and TGA results demonstrated the improved thermal stability of UAU-ISF and AUA-ISF. Chemical composition measurement showed that UAU-ISF and AUA-ISF exhibited higher cellulose content (>83 %). SEM results revealed that ultrasonic treatment led to a decomposition of okara matrix and significant porous structure in ISFs with an amplified collapse effect, resulting in an increase of the pore size of ISFs, and strengthening the properties of UAU-ISF and AUA-ISF in higher water (>15 g/g)/oil (>12 g/g) holding capacities, cholesterol binding capacity (>36 mg/g), and cation exchange capacity (>0.3 mmol/g), thus providing new insights for the preparation of ISF with high functional properties that are beneficial for human intestinal health.

Abstract:The insoluble dietary fiber from ginseng residue (ginseng-IDF) was extracted using the AOAC (Association of Official Analytical Chemists) method with content of 68.61%. Ginseng-IDF had a polysaccharide content of 18.87%, uronic acid content of 7.85%, protein content of 6.52%, and had ideal water-holding capacity (17.66 g/g), swelling ability (15.05 mL/g), and oil-holding capacity (1.78 g/g). Scanning electron microscope, Fourier transform infrared spectroscopy, and X-ray diffraction analyses suggested that ginseng-IDF had the typical structures of hydrolysis fiber, polysaccharide functional groups, and crystal structure of cellulose. Different fiber components give ginseng-IDF a specified range of pyrolysis temperature, and it is suitable for application in food processing lower than 300 °C. In addition, ginseng-IDF exhibited notable glucose and sodium cholate adsorption, significantly improved nitrite adsorption at pH 2.0 and cholesterol adsorption at pH 7.0. The above results show that ginseng-IDF could be used as an ideal functional ingredient in food processing.

Abstract:Soluble dietary fiber (SDF), an important prebiotic, has attracted growing attention, due to its great health effects and wide application. This study focused on the preparation of SDF from fermented okara. The yield of SDF obtained through Penicillium expansum fermentation (FSDF) reached 45.63 % (w/w) under the optimal conditions (pH 6.7, inoculum size 9.5 %, and time 29 h) by response surface methodology, which were 1.92 and 4.43 times higher than those of phosphate-citric acid treatment and untreated okara. Infrared spectra and X-ray diffraction indicated that three SDFs had similar spectral distribution and crystalline region. Moreover, FSDF displayed looser and more porous microstructures. Meanwhile, the composition ratio of monosaccharides has changed. FSDF exhibited higher water solubility (97.46 %), glucose adsorption capacity (203.73 mg/g), sodium cholate adsorption capacity (13.07 mg/g), cholesterol adsorption capacity (6.69– 7.62 mg/g) and radical (ABTS + , hydroxyl and DPPH) scavenging capacity. Additionally, three SDFs didn't degrade by upper gastrointestinal tract and could improve the proportion of beneficial intestinal flora in vitro, such as Lactobacillus and Bifidobacterium. Overall, the FSDF prepared in this study was a functional ingredient with great potential in foods.

Abstract:The microstructure, physicochemical properties and adsorption capacities of deoiled red raspberry pomace (DRP) and its total dietary fiber (DRPF) were investigated in the study, aiming to evaluate their potential to use as functional food ingredients. The chemical composition showed that both DRP and DRPF were mainly made of lignin. Compared to DRP, DRPF exhibited significantly decreased total contents of sugar, fat, and crude protein, from 13.37%, 5.07% and 2.58% to 1.54%, 1.17% and 1.37%, respectively. The thermal gravity analysis revealed that DRP and DRPF were stable under 162 degrees C and 197 degrees C, respectively. DRPF showed higher water-holding capacity (8.97 g/g vs. 2.62 g/g in DRP), swelling ability (10.18 mL/g vs. 6.93 g/g in DRP) and water solubility (0.25% vs. 0.07% in DRP). DRP had significantly higher in vitro cholesterol, and glucose adsorption capacities. However, better adsorption capacity of toxic ions was shown in DRPF. Based on chemical composition and characteristics, DRP and DRPF are both ideal supplements for dietary fiber and might be suitable to use as anti-obesity ingredients. Moreover, DRPF may be a better choice for toxic ions removing food.

Abstract:In order to improve the function of Shatian pomelo sponge layer insoluble dietary fiber (IDF), it was modified by high hydrostatic pressure (HHP, 400 MPa, 15 min), cellulose hydrolysis (CH, 0.4% cellulase, 70 min) and alkaline hydrolysis (AH, 1.4% sodium hydroxide solution, 20 min), respectively. Compared with the unmodified IDF, the molecular weight and particle size of the three modified IDFs were both decreased, while their void structure was increased. Specifically, the IDF treated with AH resulted in the smallest particle size (60.63 μm) and a honeycomb-like void structure, which explained why AH-treated IDF showed the strongest glucose and cholesterol adsorption capacities in vitro. The contents of rhamnose, xylose, galactose and galacturonic acid in IDF were significantly increased after HHP treatment (H-IDF). Moreover, H-IDF and CH-treated IDF (C-IDF) exhibited similar capacities for oil-holding and cation-exchange. H-IDF presented the strongest swelling capacity and excellent capacity for cholesterol adsorption. Thus, HHP modified Shatian pomelo peel sponge layer IDF can not only improve the utilization of by-products, but also has potential application value as a novel ingredient in hypolipidemic and hypoglycemic foods products.

Abstract:Dietary fiber, which is utilized to make functional meals, is an important component for promoting human health and managing calorie consumption. In this study, three different particle sizes of OIDF (Okara insoluble dietary fiber) were characterized. Their lipid-lowering effects and the impacts on gut microbiota were determined by OIDF intervention in high-fat diet rats. Scanning electron microscopy (SEM) results showed that the three particle sizes of OIDF have different morphologies. Fourier transform infrared spectroscopy (FT-IR) results showed that the three sources of IDF samples have similar active groups, but the thermogravimetric analysis/differential scanning calorimetry (TGA/DSC) and X-ray diffraction (XRD) showed that three different particle sizes of OIDF have different retention and crystallinity. Among the three OIDFs, OIDF-10 exhibited the stronger WSC, OHC, CAC, and SCAC. The results after the feeding showed that the OIDF of three particle sizes could improve the elevation of blood lipids and the disturbance of gut microbiota caused by the high-fat diet. Therefore, this study demonstrated the functional significance of the three particle sizes of OIDF and provided a reference for its application in functional food processing, aiming at maintaining healthy blood lipid and intestinal flora levels.

Abstract:This study aimed to prepare soybean dregs dietary fibre (DF) using physically assisted chemical (KHMSO) modification and study its structure, function and vitro simulation experiments. The soluble dietary fibre (SDF) content in KHMSO increased and insoluble dietary fibre (IDF) content decreased. The modified DF surface becomes irregular and rough, and the results of XPS fitting indicated that the DF structure had different peak-splitting groups. The KHMSO-treated group had the lowest digestion rate in gastric fluid and the highest digestibility in intestine fluid. The OD600 of fecal cultures was increased to 0.915, and the increased abundance of microbiota was associated with the metabolism of SCFAs, such as Lachnospiraceae, as well as the higher n-butyric acid in the KHMSO-treated group compared to the other groups and lower than the inulin, suggesting KHMSO might enhance the production of functional foods aimed at promoting intestinal health.

Abstract:To improve its functional properties, insoluble fiber of bamboo shoot shell (BIDF) was modified by enzymatic hydrolysis and dynamic high pressure micro-fluidization (DHPM). The results showed that, after enzymatic hydrolysis and DHPM treatment, the significantly decreased particle sizes and the marked microstructural changes of BIDF powders were noticed, especially for a honey-comb appearance and large cavities were clearly visible on the surface of DHPM-modified fiber. Crystallinity and thermal stability of modified fibers increased, due to the fact that part of lignin and hemicellulose were removed during the treatments, which was further confirmed by the FT-IR spectra. Compared with unmodified and enzymatic hydrolyzed fibers, DHPM-modified fiber had not only higher water holding capacity, but also more promising binding capacities for oil, nitrite ion, glucose and cholesterol, which might dependent on its decreased particle size and porous structure. The present study suggested that DHPM modification could effectively improve functional properties of BIDF, which promotes its use in food applications.

Abstract:Utilization of by-products remains a critical technical challenge in the food industry. This work aimed to improve the gelation performance of insoluble soybean fiber (ISF) from okara based on structural modifications via pH-shifting during steam-cooking treatment. Okara was intrinsically compact and tightly aggregated. Its amorphous region was degraded by steam-cooking at pH 3 and inter-molecular H-bonds were reduced, resulting in increased content of cellulose from 37.86 wt% to 43.32 wt% and crystallinity degree from 7.79% to 40.10%. The compact structure of okara was dissociated into flexible fibers by steam-cooking at pH 11, allowing for higher porosity with a specific surface area reaching 1.4428 m 2 /g. However, there were no significant changes in microstructure except for a decreased particle size as pH approached neutral. The outstanding structure and adsorption capacity of ISF supported the higher gelatin performance. The hydrogels with a self-supporting and dense network structure were observed after continuous stirring from ISF steam-cooked at pH 3, 4, and 11, which was attributed to H-bonds between ISF , while okara and other ISF suspensions exhibited good fluidity. These findings provide a new method for ISF treated by steam-cooking to enable hydrogel formation for potential applications in reconstituted foods and low-calorie gel systems.

Abstract:The objective of this study was to investigate the effect of particle size and microstructure on the physical and functional properties of soybean insoluble dietary fiber (IDF) in aqueous solution. Different IDF suspensions were obtained by ball milling (BM) and high-pressure homogenization (HPH) treatments. The results showed that the BM treatment significantly reduced the particle size distribution of the IDF, and the additional HPH treatment (BM-HPH) caused the IDF to become more porous, resulting in the formation of a honeycomb structure. Samples with a honeycomb network structure had higher hydration capacity and solubility. The water holding capacity, swelling capacity and solubility of I8 reached 18g/g, 72ml/g and 35%, respectively. I8 had the highest emulsifying stability and suspension stability. These results of the present study suggested that the reduction in particle size of IDF cannot fully account for its functional properties, of which the honeycomb structure played a more significant role. From the results of this study, it was clear that the functional properties of IDF depend on its small particle size and honeycomb structure. The present study indicated that a combination of BM and HPH treatments provides a new perspective formodifying and utilizing dietary fiber.

Effects of physical method and enzymatic hydrolysis on the properties of soybean fiber-rich stabilizer for oil in water emulsions

Abstract:Background: Okara is a by-product from the soybean industry and an abundant resource of insoluble soybean fiber (ISF). ISF with various properties could be obtained by different extraction methods. It is an attractive option to utilize okara by taking advantage of ISF as an emulsifier or stabilizer. Results: Compared with the untreated ISF (ISFUT ), superfine grinding reduced the particle size and viscosity of ISF (ISFSG ). Steam explosion increased the water solubility from 17.5% to 51.7% but decreased the water holding capacity and swelling capacity of ISF (ISFSE ) from 15.0 and 14.0 g/g to 4.2 and 3.3 g/g, respectively. Emulsions prepared by ISFUT and ISFSG before or after enzymatic hydrolysis presented large oil droplets and were unstable. Although emulsions prepared by ISFSE after enzymatic hydrolysis (ISFSE-E ) showed flocculation, the volume-weighted average diameter (19.7 μm) were the smallest while the viscosity and viscoelastic modulus were the highest, and exhibited excellent physical stability during storage. Conclusion: ISF obtained by physical and hydrolysis treatment displayed diverging physicochemical properties while ISF prepared by steam explosion-enzymatic hydrolysis presented the best potential to stabilize emulsions. The present study could provide novel information about the utilization of okara by the application of ISF as an emulsifier or stabilizer. © 2023 Society of Chemical Industry.
Removal of bound polyphenols and its effect on structure, physicochemical and functional properties of insoluble dietary fiber from adzuki bean seed coat

Abstract:Adzuki bean seed coat is an underutilized by-product of the adzuki bean paste process, rich in insoluble dietary fibers and bound polyphenols, which have diverse bioactivities. This work aimed to investigate the content of bound polyphenols in adzuki bean seed coat insoluble dietary fiber (AIDF) and their effects on the structure, physicochemical properties, and functional properties of insoluble dietary fiber. Alkaline treatment was used to remove the bound polyphenols from AIDF, resulting in dephenolized dietary fiber (AIDF-DF). The FT-IR, XRD and SEM results showed that there was no considerable structural difference between AIDF and AIDF-DF. The CLSM demonstrated that bound polyphenols could be removed from AIDF by alkaline treatment. Compared with AIDF, the water retention capacity, oil retention capacity, and swelling capacity of AIDF-DF were decreased by 6.87%, 34.42%, and 15.71%, respectively. AIDF also showed strong hypolipidemic capacities in vitro. What's more, DPPH decreased by 88.98%–92.22%, ABTS+ decreased by 74.60%–75.61%, and FRAP value decreased by 76.29%–89.44% in AIDF-DF when compared with AIDF. Therefore, the results will provide a better understanding of the interaction between cereal-based DF and its bound polyphenols.
Structure, physicochemical properties and effects on nutrients digestion of modified soluble dietary fiber extracted from sweet potato residue

Abstract:Alkaline hydrogen peroxide (AHP) modification increased the yield and the content of soluble dietary fiber extracted from sweet potato residue. As compared to the original sweet potato dietary fiber (O-SPDF), AHP modified SPDF (A-SPDF) had a smaller molecular weight of 69073.59 Da and a lower zeta-potential of -27 mV. Monosaccharide composition analysis revealed that A-SPDF had a higher proportion of pectin polysaccharides with homogalacturonan (HG) and rhamnogalacturonan-Ⅰ (RG-Ⅰ) backbones. Fourier transformed infrared (FT-IR), scanning electron microscopy (SEM), and X-ray diffraction (XRD) analyses were employed to determine the structural differences between A-SPDF and O-SPDF. Characterization of their rheological properties showed that A-SPDF exhibited higher viscosity than O-SPDF at concentrations of 2%, 4% and 6%, respectively. Additionally, A-SPDF had a more gel-like behavior than O-SPDF in the presence of Ca2+, supporting the better functional properties of A-SPDF as determined by water holding capacities (WHC), oil holding capacities (OHC), and water swelling capacities (WSC). Furthermore, A-SPDF performed significantly better in inhibiting starch hydrolysis and reducing the glucose diffusion rate using an Infogest simulated digestion model. However, SPDFs had no impact on the digestion rate of protein. Our results suggested that A-SPDF has the potential to serve as a novel food additive and functional hydrocolloid to attenuate nutrients digestion related disorders, which forms the scientific basis for the better utilization of sweet potato residue and further develop sweet potato dietary fiber (SPDF) as a functional food and/or additive in the food industry.

Abstract:This study was designed to explore the dietary fiber potential of Lycium Barbarum residue, aiming to ascertain its viability as a sustainable dietary fiber source. The study examined the extraction of water-insoluble dietary fiber (LBIDF) from Lycium barbarum residue (LBR) using the enzymatic method. The comparison involved analyzing the monosaccharide composition, microstructure, physicochemical properties, and adsorption properties of LBR and LBIDF. The results indicated that LBIDF possesses the ideal physicochemical properties compared to LBR. Through instrumental analysis, it has been demonstrated that LBIDF possesses a specific structure and functional group of fibers, making it suitable for processing below 300 °C. Furthermore, LBIDF exhibited an optimal functional capacity for cholesterol (41.749 mg/g vs. 31.464 mg/g in LBR), bile acid (30.623 mg/g vs. 19.875 mg/g in LBR), nitrite ion (1159.394 μmol/g vs. 783.261 μmol/g in LBR), glucose (3.552 mmol/g vs. 2.664 mmol/g in LBR), and glucose dialysis retardation index (6.43% vs. 13.52% in LBR). LBIDF can inhibit the digestion of fat and starch in vitro. These findings implied that dietary fiber samples from LBR may offer potential as functional food components.

Abstract:Bagasse is one of major by-product of sugar mills, but its utilization is limited by the high concentration of lignin. In this study, the optimal alkaline hydrogen peroxide (AHP) treatment conditions were determined by the response surface optimization method. The results showed that the lignin removal rate was 62.23% and the solid recovery rate was 53.76% when bagasse was prepared under optimal conditions (1.2% H2O2, 0.9% NaOH, and 46 °C for 12.3 h), while higher purity of bagasse insoluble dietary fiber (BIDF) was obtained. To further investigate the modification effect, AHP assisted with high-temperature-pressure cooking (A-H) and enzymatic hydrolysis (A-E) were used to modify bagasse, respectively. The results showed that the water holding capacity (WHC), oil holding capacity (OHC), bile salt adsorption capacity (BSAC), and nitrite ion adsorption capacity (NIAC) were significantly improved after A-H treatment. With the A-E treatment, cation exchange capacity (CEC) and BSAC were significantly increased, while WHC, OHC, and glucose adsorption capacity (GAC) were decreased. Especially, the highest WHC, OHC, BSAC and NIAC were gained by A-H treatment compared to the A-E treatment. These changes in the physicochemical and functional properties of bagasse fiber were in agreement with the microscopic surface wrinkles and pore structure, crystallinity and functional groups. In summary, the A-H modification can effectively improve the functional properties of bagasse fiber, which potentially can be applied further in the food industry.

Abstract:In this study, okara dietary fiber samples were prepared through a combination of solid-state fermentation using Aspergillus oryzae and ultrasonication. Subsequent infrared and UV–visible spectral analyses revealed that such treatment resulted in an increase in the oligosaccharide content of these samples through the enhanced cleavage of hydrogen bonds. Scanning electron microscopy and X-ray evaluation further confirmed the changes in the structural makeup of these okara SDF (soluble dietary fiber) samples relative to control samples. Following combination treatment, the soluble dietary fiber levels in these samples were three times higher than in control samples at 18.06 g/100 g. This treatment also led to a significant increase in the water holding, oil holding, and swelling capacities of the prepared fiber samples, whereas the capacity for water retention was slightly reduced. Ultrasonication alone failed to impact SDF oil holding capacity, nor did it impact cholesterol absorption at a pH of 2 used to simulate the stomach microenvironment. Together, these data thus offer a valuable reference for further work aimed at extracting soluble dietary fiber from okara.
Effect of fermentation and dynamic high pressure microfluidization on dietary fibre of soybean residue

Abstract:Soybean residue is the main by-product of making soybean milk and tofu, and there is considerable interest in its recovery, recycling and upgrading. This work was to study the effect of fermentation with lactic acid bacteria and dynamic high pressure microfluidization (DHPM) on fibre fractions, fibre composition, surface topography and X-ray diffraction of dietary fibre in soybean residue. The results show that both fermentation and DHPM increased soluble dietary fibre (6.4-9.7 g/100 g and 6.4-14.0 g/100 g, respectively) and decreased the insoluble:soluble ratio (11.6-7.8 and 11.6-4.5, respectively). The minimum insoluble:soluble ratio (2.5) was obtained in samples with combined fermented and DHPM-treated at 200 MPa. The loss of hemicellulose was observed after fermentation and DHPM, while the cellulose content did not show significant differences. Microstructural and crystal structure analysis indicated that fermentation resulted in the modification of the fibrous structure with reduced crystallinity and DHPM damaged the structure to form rugged surface.
