Abstract:The impact of DMPP (3,4-dimethylpyrazole phosphate), applied at two doses (low: recommended for agronomic use; high: > 100 × the recommended), on the function, diversity, and dynamics of target microorganisms (ammonia-oxidizing microorganisms, AOM), functionally associated microorganisms (nitrite-oxidizing bacteria (NOB) and denitrifiers), and total prokaryotic and fungal microbial communities was assessed in two loamy soils, mainly differing in pH (acidic vs. alkaline), in a 35-day microcosm study. This was achieved via monitoring inorganic N-pools, potential nitrification (PN) rates, amoA gene and transcripts abundance, the abundance of other phylogenetic marker genes (nxrB, narG, nirS, nirK, nosZ, 16S rRNA, 18S rRNA), and amplicon sequencing of amoA, 16S rRNA, and ITS. Overall, DMPP was more persistent in the acidic soil. Its low dose successfully inhibited nitrification in the alkaline but not in the acidic soil, where effective inhibition was observed only at the high dose. This was mainly attributed to the consistently higher activity of DMPP towards ammonia-oxidizing bacteria (AOB) prevailing in the alkaline soil, unlike ammonia-oxidizing archaea (AOA) whose abundance and transcriptional activity was reduced only by the high dose. DMPP, at the high dose, reduced the abundance of Nitrobacter but not Nitrospira NOB, while its low dose increased the abundance of denitrifying bacteria, prokaryotic, and fungal populations in the alkaline soil. Amplicon sequencing revealed that DMPP imposed significant changes in the composition of the prokaryotic, fungal, and AOB communities in both soils, unlike AOA which were less responsive. These were associated with dose-dependent changes in the abundance of bacteria and fungi known to control key soil functions implying possible effects for the soil ecosystem homeostasis. Our study paves the way for a more comprehensive analysis of the effects of NIs on the soil microbial community, beyond the current focus on target AOM.

Abstract:PurposeThe application of organic and inorganic fertilizers to soil can result in increased gaseous emissions, such as NH3, N2O, CO2, and CH4, as well as nitrate leaching, contributing to climate warming and ground and surface water pollution, particularly in regions with hot climates, where high temperatures and high soil nitrification rates often occur. The use of nitrification inhibitors (NIs) has been shown to effectively decrease nitrogen (N) losses from the soil-plant system.Materials and methodsNon-disruptive laboratory incubation experiments were conducted to assess the extent to which temperature (20 and 30 °C) and nutrient source (mineral and organic fertilizers) influence the rate of carbon (C)- and N-related microbial processes in soil in response to the NI 3,4-dimethylpyrazole phosphate (DMPP). Furthermore, short-term changes in the ability of microbes to degrade C substrates were evaluated in disruptive soil microcosms using microbial community-level physiological profiling and the abundance of the bacterial 16S rRNA gene as a measure of total bacterial population size.Results and discussionDMPP reduced net nitrification after 2 and 4 weeks of incubation at 30 and 20 °C by an average of 78.3 and 84.5 %, respectively, and with similar dynamics for mineral or organic fertilization. The addition of labile organic matter with cattle effluent led to a rapid increase in C mineralization that was significantly reduced by DMPP at both temperatures, whereas no changes could be detected after the addition of mineral fertilizer. The culturable heterotrophic microorganisms showed metabolic diversification in the oxidation of C sources, with organic fertilizer playing a major role in the substrate utilization patterns during the first week of incubation and the DMPP effects prevailing from day 14 until day 28. Furthermore, the copy number of the bacterial 16S rRNA gene was reduced by the application of DMPP and organic fertilizer after 28 days.ConclusionsOur results show the marked efficiency of DMPP as an NI at elevated temperatures of incubation and when associated with both mineral and organic fertilization, providing support for its use as a tool to mitigate N losses in Mediterranean ecosystems. However, we also observed impaired C respiration rates and bacterial abundances, as well as shifts in community-level physiological profiles in soil, possibly indicating a short-term effect of DMPP and organic fertilizers on non-target C-related processes and microorganisms.

Abstract:Nitrous oxide (N2O) emissions have been increasing as a result of intensive nitrogen (N) fertilisation. Soil nitrification and denitrification are the main sources of N2O, and the use of ammonium-based fertilisers combined with nitrification inhibitors (NIs) could be useful in mitigating N2O emissions from agricultural systems. In this work we looked at the N2O mitigation capacity of two dimethylpyrazol-based NIs, 3,4-dimethylpyrazole phosphate (DMPP) and 2-(N-3,4-dimethyl-1H-pyrazol-1-yl) succinic acid isomeric mixture (DMPSA), on soil nitrifying and denitrifying microbial populations under two contrasting soil water contents (40% and 80% soil water filled pore space; WFPS). Our results show that DMPP and DMPSA are equally efficient at reducing N2O emissions under 40% WFPS conditions by inhibiting bacterial ammonia oxidation. In contrast, at 80% WFPS DMPSA was less efficient than DMPP at reducing N2O emissions. Interestingly, at 80% WFPS, where lowered oxygen availability limits nitrification, both DMPP and DMPSA not only inhibited nitrification but also stimulated N2O reduction to molecular nitrogen (N2) via nitrous oxide reductase activity (Nos activity). Therefore, in this work we observed that DMP-based NIs stimulated the reduction of N2O to N2 by nitrous oxide reductase during the denitrification process.

Abstract:The application of nitrification inhibitors (NIs) based on ammonium (NH4+) is considered to be an efficient way to reduce nitrogen (N) loss by delaying the nitrification process through influencing ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB). However, the inhibition mechanisms of NIs on AOA and AOB in different soils remain unclear. Hence, we explored the two commonly used NIs (3, 4-dimethylepyrazole phosphate (DMPP) and dicyandiamide (DCD) and their combination (DMPP + DCD) on the soil nitrification and abundance of ammonia oxidizers (AOA and AOB) in three black soils (HLJ, NA, and DA) with different physicochemical properties using a 120-day incubation experiment. The results demonstrated that NIs significantly increased NH4+-N concentrations and decreased NO3−-N concentrations in all three tested soils. There was no significant difference in inhibiting nitrification in HLJ among all NI treatments, while DCD was more efficient in NA, DMPP + DCD had better efficiency in DA. The potential nitrification rate (PNR) was greatly decreased by NIs addition, and PNR was significantly positively correlated with AOB (p < 0.05). AOA was dominant in the acid soil. All NI treatments significantly inhibited soil nitrification through inhibiting the growth of AOB in the two soils with higher pH. The abundance of AOA and AOB was significantly correlated with different soil types (positively correlated with soil pH, and negatively correlated with organic matter). Moreover, soil pH and soil organic matter were considered to be the most important factors influencing the inhibition efficiency of NIs and the abundance of AOA and AOB. The application of the NIs combination (DMPP + DCD) was considered to be the most cost-effective way to inhibit soil nitrification in soil with higher pH and lower SOM, which provides a theoretical basis for a field experiment.

Abstract:This study explored the effects of combined urea and 3,4-dimethylpyrazole phosphate (DMPP) on several components critical to the soil system: net nitrification rates; communities of targeted ammonia oxidizers [ammonia-oxidizing archaea (AOA) and bacteria (AOB) and complete ammonia-oxidizing bacteria (comammox)]; non-targeted nitrite-oxidizing bacteria (NOB) and bacteria. We conducted the study in two contrasting soils (acidic and neutral) over the course of 28 days. Our results indicated that DMPP had higher inhibitory efficacy in the acidic soil (30.7%) compared to the neutral soil (12.1%). The abundance of AOB and Nitrospira -like NOB were positively associated with nitrate content in acidic soil. In neutral soil, these communities were joined by the abundance of AOA and Nitrobacter -like NOB in being positively associated with nitrate content. By blocking the growth of AOB in acidic soil—and the growth of both AOB and comammox in neutral soil—DMPP supported higher rates of AOA growth. Amplicon sequencing of the 16S rRNA gene revealed that urea and urea + DMPP treatments significantly increased the diversity indices of bacteria, including Chao 1, ACE, Shannon, and Simpson in the acidic soil but did not do so in the neutral soil. However, both urea and urea + DMPP treatments obviously altered the community structure of bacteria in both soils relative to the control treatment. This experiment comprehensively analyzed the effects of urea and nitrification inhibitor on functional guilds involved in the nitrification process and non-targeted bacteria, not just focus on targeted ammonia oxidizers.

Abstract:Application of nitrification inhibitors (NIs) in agricultural systems is an important strategy to enhance fertilizer nitrogen use efficiency and mitigate soil nitrous oxide (N2O) emissions. Here, we conducted a global meta-analysis of 88 published studies to assess the response of N2O-related functional gene and transcript abundances, and community structure to NIs application. Application of NIs significantly reduced the abundance of ammonia-oxidizing bacteria ammonia monooxygenase (AOB amoA) genes, AOB amoA transcript and nitrite reductase (nirS and nirK) genes. The effectiveness of NIs on reducing the AOB amoA abundance was influenced by N form, soil texture, soil pH and the experimental type (field vs. laboratory). Specifically, NIs were more effective when a mixed inorganic and organic N source was applied to a medium-textured soils. The NIs effectiveness increased with increasing soil pH. The response of AOB amoA abundance to NIs application was not affected by NI type, N rate, soil moisture, soil temperature and soil organic carbon (SOC). The inhibitory effect of NIs on nirS abundance increased with increasing soil temperature. NIs decreased soil nitrifying enzyme activity (NEA) and denitrifying enzyme activity (DEA) by 34.5 % and 27.0 %, respectively, leading to an overall 63.6 % reduction of N2O emissions. Soil NEA correlated positively with the abundance and community structure of AOB amoA but not with AOA amoA. Decrease in DEA with NIs application coincided with the decreasing nirS and nirK abundances. This global-scale assessment demonstrates that the effectiveness of NIs in reducing N2O emissions was attributed to the inhibiting effects on AOB amoA, nirS and nirK genes. Our findings highlight that NIs' inhibition effects on bacterial ammonia-oxidizing community and the encode enzymes in transformation of nitrite to nitric oxide are the main mechanisms for mitigation of N fertilizer-induced N2O emissions.

Abstract:The nitrification inhibitor 3,4-dimethylpyrazole phosphate (DMPP) is a powerful tool that can be used to promote nitrogen (N) use efficiency and reduce N losses from agricultural systems by slowing nitrification. Mounting evidence has confirmed the functional importance of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) in nitrification and N2O production; however, their responses to DMPP amendment and the microbial mechanisms underlying the variable efficiencies of DMPP across different soils remain largely unknown. Here we compared the impacts of DMPP on nitrification and the dynamics of ammonia oxidizers between an acidic pasture soil and an alkaline vegetable soil using a (15)N tracing and (13)CO2-DNA-stable-isotope probing (SIP) technique. The results showed that DMPP significantly inhibited nitrification and N2O production in the vegetable soil only, and the transient inhibition was coupled with a significant decrease in AOB abundance. No significant effects on the community structure of ammonia oxidizers or the abundances of total bacteria and denitrifiers were observed in either soil. The (15)N tracing experiment revealed that autotrophic nitrification was the predominant form of nitrification in both soils. The (13)CO2-DNA-SIP results indicated the involvement of AOB in active nitrification in both soils, but DMPP inhibited the assimilation of (13)CO2 into AOB only in the vegetable soil. Our findings provide evidence that DMPP could effectively inhibit nitrification through impeding the abundance and metabolic activity of AOB in the alkaline vegetable soil but not in the acidic pasture soil, possibly due to the low AOB abundance or the adsorption of DMPP by organic matter. Importance: The combination of the (15)N tracing model and (13)CO2-DNA-SIP technique provides important evidence that the nitrification inhibitor DMPP could effectively inhibit nitrification and nitrous oxide emission in an alkaline soil through influencing the abundance and metabolic activity of AOB. In contrast, DMPP amendment has no significant effect on nitrification or nitrifiers in an acidic soil, potentially owing to the low abundance of AOB and the possible adsorption of DMPP by organic matter. Our findings have direct implications for improved agricultural practices through utilizing the nitrification inhibitor DMPP in appropriate situations, and they emphasize the importance of microbial communities to the efficacy of DMPP.

Abstract:Up to 70% of the nitrogen (N) fertilizer applied to agricultural soils is lost through microbially mediated processes, such as nitrification. This can be counteracted by synthetic and biological compounds that inhibit nitrification. However, for many biological nitrification inhibitors (BNIs), the interaction with soil properties, nitrifier specificity, and effective concentrations are unclear. Here, we investigated three synthetic nitrification inhibitors (SNIs) (DCD, DMPP, and nitrapyrin) and three BNIs (methyl 3(4-hydroxyphenyl) propionate (MHPP), methyl 3(4-hydroxyphenyl) acrylate (MHPA), and limonene) in two agricultural soils differing in pH and nitrifier communities. The efficacies of SNIs and BNIs were resilient to short-term pH changes in the neutral pH soil, whereas the efficacy of some BNIs increased by neutralizing the alkaline soil. Among the BNIs, MHPA showed the highest inhibition and was, together with MHPP, identified as a putative AOB/comammox-selective inhibitor. Additionally, MHPA and limonene effectively inhibited nitrification at concentrations comparable to those used for DCD. Moreover, we identified the effective concentrations at which 50 and 80% of inhibition is observed (EC 50 and EC 80 ) for the BNIs, and similar EC 80 values were observed in both soils. Overall, our results show that these BNIs could potentially serve as effective alternatives to SNIs currently used.

Abstract:The application of the nitrification inhibitor, 3,4-dimethylpyrazole-phosphate (DMPP), is considered as an effective strategy to mitigate agricultural nitrogen loss. However, the inhibitory effect of DMPP on nitrification is variable and the importance of the soil microbial community composition to the variability is poorly understood. In this study, nine soils were collected across three land uses to investigate the impact of DMPP on nitrification and associated dynamics of ammonia oxidizers in a 28-day microcosm incubation. The results showed that the efficacy of DMPP at inhibiting net nitrification rates varied highly from no effect to 63.6 % during the first week of incubation. The abundance of ammonia-oxidizing bacteria (AOB), rather than ammonia-oxidizing archaea (AOA), was significantly correlated with nitrate concentrations across three land uses and significantly inhibited by DMPP addition. DMPP had higher efficacy in neutral and alkaline wheat and vegetable soils, compared with pasture soils. Canonical correspondence analysis suggested that soil pH was the most influential factor explaining the community composition of AOB and AOA in the collected soils. However, neither ammonium nitrate nor DMPP addition had a significant effect on the community composition of AOB or AOA during the incubation indicated by non-metric multidimensional scaling ordination. Taken together, our findings indicated that DMPP slowed nitrification by inhibiting the growth of AOB, and DMPP application affected the abundance of AOB more than the ammonia oxidizer community composition.

Abstract:Dicyandiamide (DCD) and nitrapyrin (NP) are nitrification inhibitors (NIs) used in agriculture for over 40 years. Recently, ethoxyquin (EQ) was proposed as a novel potential NI, acting through its derivative quinone imine (QI). Still, the specific activity of these NIs on the different groups of ammonia-oxidizing microorganisms (AOM), and mostly their effects on other soil microbiota remain unknown. We determined the impact of QI, and comparatively of DCD and NP, applied at two doses (regular versus high), on the function, diversity, and dynamics of target (AOM), functionally associated (nitrite-oxidizing bacteria-NOB), and off-target prokaryotic and fungal communities in two soils mainly differing in pH (5.4 versus 7.9). QI was equally effective to DCD but more effective than NP in inhibiting nitrification in the acidic soil, while in the alkaline soil QI was less efficient than DCD and NP. This was attributed to the higher activity of QI toward AOA prevailing in the acidic soil. All NIs induced significant effects on the composition of the AOB community in both soils, unlike AOA, which were less responsive. Beyond on-target effects, we noted an inhibitory effect of all NIs on the abundance of NOB in the alkaline soil, with Nitrobacter being more sensitive than Nitrospira. QI, unlike the other NIs, induced significant changes in the composition of the bacterial and fungal communities in both soils. Our findings have serious implications for the efficiency and future use of NIs on agriculture and provide unprecedented evidence for the potential off-target effects of NIs on soil microbiota. IMPORTANCE NIs could improve N use efficiency and decelerate N cycling. Still, we know little about their activity on the distinct AOM groups and about their effects on off-target soil microorganisms. Here, we studied the behavior of a new potent NI, QI, compared to established NIs. We show that (i) the variable efficacy of NIs across soils with different pH reflects differences in the inherent specific activity of the NIs to AOA and AOB; (ii) beyond AOM, NIs exhibit negative effects on other nitrifiers, like NOB; (iii) QI was the sole NI that significantly affected prokaryotic and fungal diversity. Our findings (i) highlight the need for novel NI strategies that consider the variable sensitivity of AOM groups to the different NIs (ii) identify QI as a potent AOA inhibitor, and (iii) stress the need for monitoring NIs' impact on off-target soil microorganisms to ensure sustainable N fertilizers use and soil ecosystem functioning.
The Efficacy of 3,4-Dimethylpyrazole Phosphate on N2O Emissions is Linked to Niche Differentiation of Ammonia Oxidizing Archaea and Bacteria Across Four Arable Soils

Abstract:The nitrification inhibitor, 3,4-dimethylpyrazole phosphate (DMPP), can be used to reduce N2O emissions from agricultural ecosystems. However, the effectiveness of DMPP varies among soils, and this is due to both abiotic (e.g. soil properties) and biotic factors (e.g. ammonia oxidizers and denitrifier communities). Understanding the nature of these effects is necessary to improve the efficacy of DMPP, therefore encouraging wider adoption and environmental benefits. In particular, soil microbial properties associated with variation in efficacy remain largely unknown. In this study four contrasting arable soils (a grey desert soil, an alluvial paddy soil, a loess formed paddy soil, and a red soil), were characterized based on DMPP inhibition of N2O emissions and associated microbial functional guilds. DMPP significantly inhibited nitrification and N2O emissions, with an average inhibitory rate ranging from 41.7% in a red soil to 90.0% in a grey desert soil. Ammonia oxidizing bacteria (AOB) and archaea (AOA) exhibited contrasting response patterns to DMPP addition. Notably, suppression of N2O emissions by DMPP only occurred alongside fluctuations in AOB abundance. However, when AOB were inhibited, AOA abundance increased. Soil-dependent response patterns to DMPP were observed for ammonia oxidizers and denitrifiers in terms of community structure. Partial least squares path modeling (PLS-PM) found that abiotic factors, particularly pH, and biological factors such as ammonia oxidizer communities, were closely linked to N2O emissions. Our findings provide evidence that: (i) DMPP effectively inhibits nitrification through inhibiting the abundance of AOB across soil types; (ii) releasing AOA from the competition with AOB allows AOA to efficiently grow and multiply, even under high ammonium conditions; and (iii) abiotic factors play a more important role than biotic factors in soil N2O emissions.

Abstract:An aerobic 15N microcosmic experiment was conducted to compare the inhibitory effects of the biological nitrification inhibitor (BNI), methyl 3-(4-hydroxyphenyl) propionate (MHPP) at rates of 500 and 1000 mg kg−1 with the synthetic nitrification inhibitor 3,4-dimethylpyrazole phosphate (DMPP) at 1% of applied NH4+, on the gross nitrification rate (n_gross) and on the abundance and community composition of ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) of two contrasting soils (pH: 5.10 vs. 8.15, clay content 17.8 vs. 30.8). DMPP inhibited 56.6% of n_gross in the acidic soil and 50.3% in the calcareous soil, whereas MHPP inhibited 18.3–55.5% of n_gross in the acidic soil and 14.1–20.2% in the calcareous soil. MHPP used at the high rate showed the same inhibition on n_gross as DMPP in the acidic soil but not in the calcareous soil. DMPP and MHPP likely regulated n_gross by causing niche differentiation between AOA and AOB. Moreover, the community composition of AOB was more sensitive to nitrification inhibitor application than that of AOA, particularly in the acidic soil. However, the response of AOB community composition was less sensitive to the application of MHPP than to that of DMPP. MHPP mainly targeted Nitrosospira clusters 3a.2, 3b.2, and 9 of the AOB in the acidic soil.

Abstract:This study evaluated the effects of nitrification inhibitor 3,4-dimethylpyrazole phosphate (DMPP) and fungicide iprodione on net nitrification rates and abundances of functional genes related to nitrification and denitrification in an agricultural soil. Single DMPP application or repeated iprodione applications decreased net nitrification rates in the test soil. However, when the DMPP and iprodione were applied together, they could generate antagonistic effects on the inhibitions of net nitrification rates. Repeated iprodione applications reduced ammonia oxidizing archaea and bacteria (AOA and AOB) amoA gene abundances, while DMPP application decreased AOB amoA gene abundances only. The abundances of narG and nirK genes were negatively affected by repeated iprodione applications. Our results demonstrated that combined applications of DMPP and iprodione could generate antagonistic effects on the inhibitions of net nitrification rates and discrepant impacts on the abundances of functional genes related to soil denitrification.

Abstract:Nitrification inhibitors (NIs) 3,4-dimethylpyrazole phosphate (DMPP) and dicyandiamide (DCD) have been used extensively to improve nitrogen fertilizer utilization in farmland. However, their comparative effects on ammonia-oxidizing bacteria (AOB) and ammonia-oxidizing archaea (AOA) in agricultural soils are still unclear. Here, we compared the impacts of these two inhibitors on soil nitrification, AOA and AOB abundance as well as their community structure in a vegetable soil by using real-time PCR and terminal restriction fragment length polymorphism (T-RFLP). Our results showed that urea application significantly increased the net nitrification rates, but were significantly inhibited by both NIs, and the inhibitory effect of DMPP was significantly greater than that of DCD. AOB growth was more greatly inhibited by DMPP than by DCD, and the net nitrification rate was significantly related to AOB abundance, but not to AOA abundance. Application of urea and NIs to soil did not change the diversity of the AOA community, with the T-RFs remaining in proportions that were similar to control soils, while the community structure of AOB exhibited obvious shifts within all different treatments compared to the control. Phylogenetic analysis showed that all AOA sequences fell within group 1.1a and group 1.1b, and the AOB community consisted of Nitrosospira cluster 3, cluster 0, and unidentified species. These results suggest that DMPP exhibited a stronger inhibitory effect on nitrification than DCD by inhibiting AOB rather than AOA.

Abstract:Nonphotochemical quenching (NPQ) is a fundamental mechanism in photosynthesis which protects plants against excess excitation energy and is of crucial importance for their survival and fitness. Recently, carotenoid radical cation (Car*+) formation has been discovered to be a key step for the feedback deexcitation quenching mechanism (qE), a component of NPQ, of which the molecular mechanism and location is still unknown. We have generated and characterized carotenoid radical cations by means of resonant two color, two photon ionization (R2C2PI) spectroscopy. The Car*+ bands have maxima located at 830 nm (violaxanthin), 880 nm (lutein), 900 nm (zeaxanthin), and 920 nm (beta-carotene). The positions of these maxima depend strongly on solution conditions, the number of conjugated C=C bonds, and molecular structure. Furthermore, R2C2PI measurements on the light-harvesting complex of photosystem II (LHC II) samples with or without zeaxanthin (Zea) reveal the violaxanthin (Vio) radical cation (Vio*+) band at 909 nm and the Zea*+ band at 983 nm. The replacement of Vio by Zea in the light-harvesting complex II (LHC II) has no influence on the Chl excitation lifetime, and by exciting the Chls lowest excited state, no additional rise and decay corresponding to the Car*+ signal observed previously during qE was detected in the spectral range investigated (800-1050 nm). On the basis of our findings, the mechanism of qE involving the simple replacement of Vio with Zea in LHC II needs to be reconsidered.

Abstract:Nitrification inhibitors (NIs) are considered as an effective strategy for reducing nitrification rate and related environmental nitrogen (N) loss. However, whether plant-derived biological NIs had an advantage over chemical NIs in simultaneously inhibiting nitrification rate and N 2 O production remains unclear. Here, we conducted an aerobic 15 N microcosmic incubation experiment to compare the effects of a biological NI (methyl 3-(4-hydroxyphenyl) propionate, MHPP) with three chemical NIs, 2-chloro-6-(trichloromethyl) pyridine (nitrapyrin), dicyandiamide (DCD), and 3,4-dimethylpyrazole phosphate (DMPP) on (i) gross N mineralization and nitrification rate and (ii) the relative importance of nitrification and denitrification in N 2 O emission in a calcareous soil. The results showed that DMPP significantly inhibited m _gross rate ( P < 0.05), whereas DCD, nitrapyrin, and MHPP only numerically inhibited it. Gross N nitrification ( n _gross ) rates were inhibited by 9.48% in the DCD treatment to 51.5% in the nitrapyrin treatment. Chemical NIs primarily affected the amoA gene abundance of ammonia-oxidizing bacteria (AOB), whereas biological NIs affected the amoA gene abundance of ammonia-oxidizing archaea (AOA) and AOB. AOB's community composition was more susceptible to NIs than AOA, and NIs mainly targeted Nitrosospira clusters of AOB. Chemical NIs of DCD, DMPP, and nitrapyrin proportionally reduced N 2 O production from nitrification and denitrification. However, the biological NI MHPP stimulated short-term N 2 O emission and increased the proportion of N 2 O from denitrification. Our findings showed that the influence of NIs on gross N mineralization rate ( m _gross ) was dependent on the NI type. MHPP exhibited a moderate n _gross inhibitory capacity compared with the three chemical NIs. The mechanisms of chemical and biological NIs inhibiting n _gross can be partly attributed to changes in the abundance and community of ammonia oxidizers. A more comprehensive evaluation is needed to determine whether biological NIs have advantages over chemical NIs in inhibiting greenhouse gas emissions.

Abstract:Biological nitrogen fixation and nitrification inhibitor applications contribute to improving soil nitrogen (N) availability, however, free-living N fixation affected by nitrification inhibitors has not been effectively evaluated in soils under different weed management methods. In this study, the effects of the nitrification inhibitors dicyandiamide (DCD) and 3, 4-dimethylpyrazole phosphate (DMPP) on the nitrogenase, nifH gene，and diazotrophic communities in soils under different weed management methods (AMB, weeds growth without mowing or glyphosate spraying; GS, glyphosate spraying; MSG, mowing and removing weeds and glyphosate spraying; and WM, mowing aboveground weeds) were investigated. Compared to the control counterparts, the DCD application decreased soil nitrogenase activity and nifH gene abundance by 4.5 % and 37.9 %, respectively, under the GS management method, and the DMPP application reduced soil nitrogenase activity by 20.4 % and reduced the nifH gene abundance by 83.4 % under the MSG management method. The application of nitrification inhibitors significantly elevated soil NH4+-N contents but decreased NO3--N contents, which had adverse impacts on soil nifH gene abundance and nitrogenase activity. The nifH gene abundances were also negatively impacted by dissolved organic N and Geobacter but were positively affected by available phosphorus and diazotrophic community structures. Nitrification inhibitors significantly inhibited Methylocella but stimulated Rhizobiales and affected soil diazotrophic communities. The nitrification inhibitors DCD and DMPP significantly altered soil diazotrophic community structures, but weed management outweighed nitrification inhibitors in reshaping soil diazotrophic community structures. The non-targeted effects of the nitrification inhibitors DMPP and DCD on soil free-living N fixation were substantially influenced by the weed management methods.
Contribution of airway closure to chronic postbronchiolitis airway dysfunction in rats.

Abstract:Genetically susceptible Brown Norway rats develop a chronic asthmalike syndrome after recovering from viral bronchiolitis at an early age. We hypothesized that airway closure is an important mechanism of airflow obstruction in postbronchiolitis rats. Rats were studied 8-12 wk after inoculation with Sendai virus or sterile vehicle at 3-4 wk of age. Under light pentobarbital anesthesia, rats were instrumented with an orotracheal catheter and an esophageal pressure monitor and placed in a total body plethysmograph. Lung volumes and forced-expiratory maneuvers were measured using the Boyle's law method and software-controlled valving of positive and negative pressures to elicit lung inflations and rapid deflations; pulmonary resistance was measured during spontaneous tidal breathing; and quasi-static pressure-volume curves were obtained with passive inflations and deflations in fully anesthetized, paralyzed rats. Compared with controls, the postbronchiolitis rats had elevated pulmonary resistance and reduced forced-expiratory volume in 0.2 s. Most of the reduced forced-expiratory volume in 0.2 s was associated with reduced forced vital capacity, indicating premature airway closure as a prominent mechanism. The reduced airflow in postbronchiolitis rats was highly dependent on lung volume, being nearly normal at 70% lung capacity, but sevenfold less than normal at 30% lung capacity. Increased respiratory system hysteresis between functional reserve capacity and total lung capacity was evidence for increased airway closure at normal end-expiratory lung volumes in postbronchiolitis rats. We conclude that airway instability and closure is a prominent mechanism of the chronic airway dysfunction in rats that have recovered from viral bronchiolitis at an early age.

Abstract:ABSTRACT The use of biological nitrification inhibitors (BNIs) holds a great potential to effectively reduce nitrogen losses from agroecosystems and conforms with the current move toward ecological-intensified agriculture. Knowledge of the activity of BNIs to soil nitrifiers is limited and is generally based on a single Nitrosomonas europaea bioassay. We determined the in vitro activity of multiple plant-derived compounds as BNIs such as (i) root-derived compounds [sakuranetin, methyl 3-(4-hydroxyphenyl)-propionate (MHPP), and zeanone]; (ii) other phytochemicals (caffeic, quinic, chlorogenic, and shikimic acids); and (iii) analogs of statins (simvastatin), triazoles (1-butyl-4-propyl-triazole, 1,4-dibutyltriazole), and zeanone (2-methoxy-1,4-naphthoquinone) on distinct soil-derived ammonia-oxidizing bacteria (AOB) ( Nitrosospira multiformis and Nitrosomonas europaea ), ammonia-oxidizing archaea (AOA) ( Candidatus Nitrosotalea sinensis and Candidatus Nitrosocosmicus franklandianus), and a nitrite-oxidizing bacterium (NOB) ( Nitrobacter sp.). Our results indicate that AOA were more sensitive than AOB to BNIs. Sensitivity within the AOA group was BNI dependent, unlike AOB, for which N. multiformis was consistently more sensitive than N. europaea . Several compounds were inhibitory to Nitrobacter sp. with MHPP and caffeic acid being more potent against NOB compared to the ammonia-oxidizing strains, an observation with potential implications for soil quality and productivity. Overall, zeanone was the most potent ΒNI against ammonia oxidizers, while caffeic acid was the most potent BNI against Nitrobacter sp. We provide pioneering evidence for the activity range of multiple BNIs on soil nitrifiers, stress the need for revisiting the biological screening systems currently used for BNI determination, and advocate for a more thorough monitoring of the impact of BNI candidates on a range of target and non-target microorganisms. IMPORTANCE Synthetic nitrification inhibitors are routinely used with nitrogen fertilizers to reduce nitrogen losses from agroecosystems, despite having drawbacks like poor efficiency, cost, and entry into the food chain. Plant-derived BNIs constitute a more environmentally conducive alternative. Knowledge on the activity of BNIs to soil nitrifiers is largely based on bioassays with a single Nitrosomonas europaea strain which does not constitute a dominant member of the community of ammonia-oxidizing microorganisms (AOM) in soil. We determined the activity of several plant-derived molecules reported as having activity, including the recently discovered maize-isolated BNI, zeanone, and its natural analog, 2-methoxy-1,4-naphthoquinone, on a range of ecologically relevant AOM and one nitrite-oxidizing bacterial culture, expanding our knowledge on the intrinsic inhibition potential of BNIs toward AOM and highlighting the necessity for a deeper understanding of the effect of BNIs on the overall soil microbiome integrity before their further use in agricultural settings.

Effects of the nitrification inhibitor nitrapyrin on urea-based fertilizers in a Mediterranean calcareous soil: N dynamics and microbial functional genes.

Abstract:Nitrogen (N) fertilization is an essential agronomic practice, which increases crop yields and improves soil fertility. Globally, more than 110 x 109 kg of chemical N fertilizers are applied each year with urea-N being one of the most affordable options. Upon urea hydrolysis, any portion not assimilated by crops is either volatilized as NH3 or microbially nitrified (i.e., NH4+ oxidized) to leachable NO3- and NO2-. Nitrification inhibitors (NI) are increasingly co-applied as a sustainable agricultural practice and block the process of nitrification, resulting in a temporal increase of NH4+ in the soils. Several studies have documented the effectiveness of NIs in retaining soil NH4+ and increasing crop yields, but less is known about the effects of NIs on the fate of urea&#8211;N and the overall impact of NIs on the soil microbial community.In a 60 day soil mesocosm experiment, we investigated the effects of Nitrapyrin (NI; 2-chloro-6-(trichloromethyl)pyridine) co-applied with a selection of urea-based fertilizers: urea (U); U with urease inhibitors (U+UI); methylene-urea (MU); and zeolite-coated urea (ZU), on a typical Mediterranean soil under ambient summer conditions. We showed that NI applied with urea fertilizers resulted in a slower decay of extractable NH4+ with a concurrent increase in NH3 volatilization. Integrated measures of soil NH4+ were 1.5 to 3.3-fold greater when NI was applied. At the same time, there was a 10 to 60% reduction in integrated measures of NO3- and NO2- when NI was applied with the tested fertilizer types, except MU fertilizer where the integrated measures of NO3- and NO2- doubled. Upon urea hydrolysis, the released NH4+ was transformed to NO3- and NO2-, which subsequently decreased in concentration following a typical nitrification - denitrification pathway in the absence of plants. Soil N2O emissions from urea fertilizers were reduced by 40% with UI, 50% with NI, and 66% with NI + UI.Interestingly, 15 days after the application of NI, there was a decrease in bacterial abundance (eub genes; qPCR) in all fertilized treatments. NI dramatically reduced the abundance of ammonia-oxidizing microbes (amoA genes) and there were fewer bacteria associated with denitrification genes (nirK, nirS, nosZ) when NI was applied.&#160;At the end of the experiment, there was no significant difference in total N among all fertilized soils. Total N was in excess when compared to the control, and it was a considerable N pool potentially immobilized in microbial biomass in the absence of crops.In conclusion, the use of NI doubled NH4+ retention in the soil and decreased soil N2O emission by 50%, through negatively affecting ammonia oxidizing and denitrifying microbes and subsequently reducing soil available NO3- and NO2-. The application of NIs should be carefully planned and synchronized (timing) with crop growth to reduce subsequent N transformations and N loss to the environment.Keywords: urea, zeolite, methylene-urea, nitrification inhibitor, nitrapyrin, calcareous soil, soil nitrogen
