Abstract:Background: Benzo[a]pyrene (BaP) is an airborne pollutant resulting from incomplete combustions and was classified by IARC as possible carcinogenic compound to human beings. Human beings can be exposed to airborne BaP via inhalation and dermal pathways in both indoor and outdoor environment. Consequently, proper assessment of exposure to airborne BaP is very important. Aims: The aim of this study is to assess exposure of human beings to both gas- and particle-phases of BaP via inhalation and dermal pathways for both indoor and outdoor environment of a typical residential condition in Beijing. Methods: Equilibrium indoor concentrations within a typical Beijing residential home of both phases BaP were calculated. Exposure to both phases BaP via inhalation pathway can be estimated with the inhalation rate and the exposure duration. Exposure to gas-phase BaP via dermal pathway can be estimated with the skin area contact with the gas, the exposure duration and the overall skin permeability coefficient of gases. Exposure parameters based on the survey in North China were utilized in this study. Results: The total exposure is highest in winter, which is 3.08×10-3(?g/kg/d), and is lowest in summer, which is 3.27×10-4(?g/kg/d). The indoor exposure to both phases of BaP is at least 10 times higher than that of outdoor environment. The contribution of exposure to gas-phase BaP by inhalation varies from 5.94% to 6.54% and the contribution to particle-phase BaP by inhalation is between 90.59% and 92.91%. The occupancy of dermal exposure to gas-phase BaP is from 0.66% to 3.18%. Conclusions: 1) The exposure to BaP via inhalation and dermal pathways decreased with winter, spring, autumn and summer. 2) The indoor exposure to both gas and particle-phases of BaP much higher than that of outdoor environment. 3) The exposure to particle-phase BaP by inhalation is the most important exposure pathway, followed by which is the exposure to gas-phase BaP by inhalation. The exposure to gas-phase BaP via dermal pathway is the least important.
Analysis of 200 food items for benzo[a]pyrene and estimation of its intake in an epidemiologic study

Abstract:Animal studies have shown that dietary intake of benzo[a]pyrene (BaP), a polycyclic aromatic hydrocarbon (PAH), causes increased levels of tumors at several sites, particularly in the upper gastrointestinal tract. However, the role of dietary intake of BaP and cancer in humans is not clear. We created a BaP database of selected food products that could be linked to Food Frequency Questionnaires (FFQs) to estimate BaP intake. BaP levels were measured for each food line-item (composite samples) which consisted of a variety of foods in a FFQ. Composite sample parts were derived from the Second National Health and Nutrition Examination Survey (NHANES II) which represents the most common food items consumed by the general population. Meat samples were cooked by different techniques in controlled conditions, and by various restaurants and fast-food chains. Non-meat products were purchased from the major national supermarket chains. The quantities of BaP were measured using a thin-layer chromatography (TLC)/spectrofluorometer technique and were highly correlated with both BaP (r=0.99) [corrected] and sum of carcinogenic PAH (r=0.98) measured by HPLC technique. We linked our database to the results from a FFQ and estimated the daily BaP intake of various food items in 228 subjects in the Washington, DC metropolitan area. The highest levels of BaP (up to about 4 ng BaP/g of cooked meat) were found in grilled/barbecued very well done steaks and hamburgers and in grilled/barbecued well done chicken with skin. BaP concentrations were lower in meats that were grilled/barbecued to medium done and in all broiled or pan-fried meat samples regardless of doneness level. The BaP levels in non-meat items were generally low. However, certain cereals and greens (e.g. kale, collard greens) had levels up to 0.5 ng/g. In our population, the bread/cereal/grain, and grilled/barbecued meat, respectively, contributed 29 and 21 percent to the mean daily intake of BaP. This database may be helpful in initial attempts to assess dietary BaP exposures in studies of cancer etiology.
Measurement of Human CYP1A2 Induction by Inhalation Exposure to Benzo(a)pyrene Based on in Vivo Isotope Breath Method.

Abstract:Cytochrome P450 1A2 (CYP1A2) is an enzyme involved in the metabolic activation of certain carcinogens, and inducible by toxic substrates. To date, few studies have investigated in vivo CYP1A2 induction in humans and its relationship to polycylic aromatic hydrocarbons (PAHs) like benzo(a)pyrene (BaP). Non-smoking healthy male coke-oven workers (n = 30) were recruited as 'exposure' group, and non-smoking healthy office workers in the same city (n = 10) were selected as 'control' group, to test whether high inhalation exposure to PAHs can induce CYP1A2 activity in human livers. Significantly higher inhalation exposure of PAHs were found among the exposure group compared to the control. Inhalation BaP exposure concentration in the exposure group was more than 30 times higher than the control group (p < 0.001). However, the exposure group did not exhale significant higher levels of (13)CO2/(12)CO2 in breath samples (p = 0.81), and no significant relationship was found between the inhaled BaP concentration and the (13)CO2/(12)CO2 ratio (p = 0.91). A significant association was found between the (13)CO2/(12)CO2 exhalation and dietary BaP intake level. Hepatic CYP1A2 activity/induction level was not effected by inhaled BaP but was altered by ingestion of BaP.
Comparison of urinary 3-hydroxybenzo(a)Pyrene (3-OHBaP) and trans-anti-7,8,9,10-tetrahydroxy-7,8,9,10-tetrahydrobenzo(a)Pyrene (TetraolBaP) as biomarkers of exposure to carcinogenic BaP

Abstract:Introduction Biomonitoring of exposure to carcinogenic Benzo(a)Pyrene is generally based on measurement of urinary 3-hydroxybenzo(a)pyrene (3-OHBaP), but its analysis is complex and only reflects the BaP detoxification pathway. TetraolBaP, another BaP metabolite resulting from the metabolic activation pathway, is now available but has not yet been studied in occupational settings or compared with 3-OHBaP. Methods Biomonitoring was carried out on 118 subjects working in the aluminium smelting industry. 3 urine samples were collected from each subject at the beginning and end of the working week. Pyrene metabolite (1-hydroxypyrene) and the two BaP biomarkers (3-OHBaP and TetraolBaP) were analysed using LC-Fluorescence and GC-NCI-MS-MS. Results The workers studied were found to be highly exposed, with 1-OHP and 3-OHBaP frequently exceeding maximum recommended values in occupational settings. Maximum concentrations were measured at end of shift+16h for all biomarkers, highlighting dermal exposure and/or temporary storage. Correlations were strong between 1-OHP and 3-OHBaP (r = 0.68–0.75) as well as between 3-OHBaP and TetraolBaP (r = 0.67–0.78), and moderate between 1-OHP and TetraolBaP (r = 0.59–0.76). While TetraolBaP levels were higher at low PAH exposures, TetraolBaP increased much more slowly at high exposures, indicating progressive saturation of the bioactivation pathway. The [3-OHBaP]/[TetraolBaP] ratio was found to be significantly lower in chronically exposed workers. Urinary TetraolBaP levels corresponding to 1-OHP (2.5 μg/L or 1 μmol/mol creatinine) or 3-OHBaP (0.4 nmol/mol creatinine) guidance values were found to range between 0.84 and 0.95 nmol/mol creatinine. Conclusions TetraolBaP, resulting from carcinogenic BaP's metabolic activation pathway, was shown to be a diagnostically specific and sensitive biomarker for determining subjects' toxic internal exposure to PAHs in different contexts (occupational settings, environment) and assessing health risks.

Head, Shoulders, Knees, and Toes: Placement of Wearable Passive Samplers Alters Exposure Profiles Observed

Abstract:Chemical exposures are a major risk factor for many diseases. Comprehensive characterization of personal exposures is necessary to highlight chemicals of concern and factors that influence these chemical exposure dynamics. For this purpose, wearable passive samplers can be applied to assess longitudinal personal exposures to airborne contaminants. Questions remain regarding the impact of sampler placement at different locations of the body on the exposure profiles observed and how these placements affect the monitoring of seasonal dynamics in exposures. This study assessed personal air contaminant exposure using passive samplers worn in parallel across 32 participant's wrists, chest, and shoes over 24 h. Samplers were analyzed by thermal desorption gas chromatography high-resolution mass spectrometry. Personal exposure profiles were similar for about one-third of the 275 identified chemicals, irrespective of sampler placement. Signals of certain semivolatile organic compounds (SVOCs) were enhanced in shoes and, to a lesser extent, wrist samplers, as compared to those in chest samplers. Signals of volatile organic compounds were less impacted by sampler placement. Results showed that chest samplers predominantly captured more volatile exposures, as compared to those of particle-bound exposures, which may indicate predominant monitoring of chemicals via the inhalation route of exposure for chest samplers. In contrast, shoe samplers were more sensitive to particle-bound SVOCs. Seventy-one chemicals changed across participants between winter and summer in the same manner for two or more different sampler placements on the body, whereas 122 chemicals were observed to have seasonal differences in only one placement. Hence, the placement in certain cases significantly impacts exposure dynamics observed. Passive sampling of polybrominated diphenyl ethers in indoor and outdoor air in Shanghai, China: seasonal variations, sources, and inhalation exposure

Abstract:Ninety-seven seasonal, passive indoor and outdoor air samples were collected in Shanghai to study polybrominated diphenyl ethers (ΣPBDEs, 16 congeners including BDE-209), their concentrations, composition profiles, seasonal variations, influencing factors, emission sources, and human inhalation exposure. In summer, median indoor concentrations of Σ15PBDEs (excluding BDE-209) were 82 pg m−3 in offices and 30 pg m−3 in homes, ∼3 times the winter concentrations. The average summer concentration of 130 pg m−3 BDE-209 in homes was higher than that in offices (which was 90 pg m−3); in winter, home and office concentrations were similar (46 and 47 pg m−3, respectively). For outdoor air, the median concentration of Σ15PBDEs in summer (12 pg m−3) was twice the winter concentration (6 pg m−3), while the summer median concentration of BDE-209 (398 pg m−3) was half the winter concentration (794 pg m−3). Higher concentrations of Σ15PBDEs indoors compared with outdoors showed that the lower brominated BDEs found were mainly from indoor sources. Meanwhile, the much lower indoor concentration of BDE-209 compared with the outdoors showed that BDE-209 came mainly from outdoor sources. The data set also indicated that electric/electronic appliances were the main sources of indoor ΣPBDEs, and old appliances emitted more lower brominated BDEs, while industrial emissions should be the main source of the outdoor BDE-209. Median daily human exposures to Σ15PBDEs and BDE-209 through inhalation were estimated to be 0.23 and 1.73 ng day−1 in winter and 0.65 and 2.28 ng day−1 in summer for adults. The human inhalation exposure to ΣPBDEs (3.44 ng day−1 for adults and 1.33 ng day−1 for toddlers) was comparable to that from eating contaminated fish for both toddlers and adults in Shanghai.

Abstract:To obtain a comprehensive understanding of the main source and route of human exposure to polybrominated diphenyl ethers (PBDEs), the daily intake and uptakes through inhalation, ingestion, and dermal contact for Shanghai residents were estimated on the basis of the PBDE concentrations in the air obtained in the present study and previous data reported in the literature. The PBDE concentrations in the gas and particle phases collected in Shanghai were 0.99-57.5 and 0.1-234 pg/m(3), respectively. The contamination levels of PBDEs in the air in Shanghai were similar to or slightly lower than the data from other regions. The estimated total daily intakes of PBDEs through the three routes were 607 and 1,636 ng/day for children and adults, respectively, while they decreased to 63.0 and 93.1 ng/day when the uptake efficiency (which is the fraction of contaminants that reaches the systemic circulation) of PBDEs was added to calculation. The results showed that dust is the main source of human exposure to PBDEs when PBDE uptake efficiency was not considered. It accounted for 66.2-79.2% of the total PBDE intake. However, food is the main source, which accounted for 66.6-75.1%, when the uptake efficiency was added to calculation. Among the three routes, dermal contact (53.1-76.6%) is the main pathway, whereas ingestion (84.7-92.9%) is the main one when the uptake efficiency was considered. Furthermore, risk assessment showed that the PBDE exposure amount would not cause obvious non-cancer and cancer risks to local residents.
Measurement of urinary Benzo[a]pyrene tetrols and their relationship to other polycyclic aromatic hydrocarbon metabolites and cotinine in humans

Abstract:Biomonitoring of exposure to polycyclic aromatic hydrocarbons (PAHs) typically uses measurement of metabolites of PAHs with four or less aromatic rings, such as 1-hydroxypyrene, even though interest may be in exposure to larger and carcinogenic PAHs, such as benzo[a]pyrene (B[a]P). An improved procedure for measuring two tetrol metabolites of B[a]P has been developed. Using 2 mL urine, the method includes enzymatic deconjugation of the tetrol conjugates, liquid-liquid extraction, activated carbon solid phase extraction (SPE) and Strata-X SPE, and gas chromatography-electron capture negative ionization-tandem mass spectrometric determination. Limits of detection were 0.026 pg/mL (benzo[a]pyrene-r-7,t-8,t-9,c-10-tetrahydrotetrol, BPT I-1) and 0.090 pg/mL (benzo[a]pyrene-r-7,t-8,c-9,c-10-tetrahydrotetrol, BPT II-1). We quantified BPT I-1 and BPT II-1 in urine from a volunteer who consumed one meal containing high levels of PAHs (barbequed chicken). We also measured urinary concentrations of BPT I-1 and BPT II-1 in smokers and nonsmokers, and compared these concentrations with those of monohydroxy PAHs (OH-PAHs) and cotinine. Urinary elimination of BPT I-1 and BPT II-1 as a function of time after dietary exposure was similar to that observed previously for OH-PAHs. While the median BPT I-1 concentration in smokers' urine (0.069 pg/mL) significantly differs from nonsmokers (0.043 pg/mL), BPT I-1 is only weakly correlated with cotinine. The urinary concentration of BPT I-1 shows a weaker relationship to tobacco smoke than metabolites of smaller PAHs, suggesting that other routes of exposure such as for example dietary routes may be of larger quantitative importance.

Abstract:The margins of exposure (MOE) and incremental lifetime cancer risk (ILCR) both indicated that the dietary exposure to BaP of the participants was generally safe, but there were still some people with certain carcinogenic risks. From MD trajectories, BaP binding does not affect the global motion of individual nucleic acid sequences, but local weak non‐covalent interactions changed greatly; it also weakens molecular interactions of nucleic acid with Bacillus stearothermophilus DNA polymerase I large fragment (BF), and significantly changes the cavity structure of recognition interface. As an extremely strong polycyclic aromatic hydrocarbon carcinogen, benzo[α]pyrene (BaP) is often produced during food processing at high temperatures. Recently, food safety, as well as toxicity mechanism and risk assessment of BaP, has received extensive attention. We first constructed the database of BaP pollution concentration in Chinese daily food with over 104 data items; collected dietary intake data using online survey; then assessed dietary exposure risk; and finally revealed the possible toxicity mechanism through four comparative molecular dynamics (MD) simulations. The statistical results showed that the concentration of BaP in olive oil was the highest, followed by that in fried meat products. The margins of exposure and incremental lifetime cancer risk both indicated that the dietary exposure to BaP of the participants was generally safe, but there were still some people with certain carcinogenic risks. Specifically, the health risk of the core district population was higher than that of the noncore district in Bashu area, and the female postgraduate group was higher than the male group with bachelor degree or below. Multi-residue analysis of several high-production-volume chemicals present in the particulate matter from outdoor air. A preliminary human exposure estimation

Abstract:A multi-residue method based on gas chromatography-mass spectrometry combined with pressurised liquid extraction was developed to determine seven organophosphate esters (OPEs), six phthalate esters (PAEs), four benzotriazoles (BTRs), five benzothiazoles (BTHs) and four benzenesulfonamides (BSAs) in particulate matter samples from outdoor air. All of these compounds are among the named high-production volume chemicals (HPVCs) and some of them have shown to be harmful to human, therefore they have been subject for legal regulation in order to control their production and usage. Under optimised conditions, high recovery values (>80%) and low detection limits (pg m-3) were obtained for most of the compounds with accuracy values between 83% and 118%. Some samples from two locations surrounded by different industry activities showed the widespread occurrence of all the PAEs, followed by some OPEs. Diethylhexyl phthalate (DEHP) was the most abundant compound with concentrations ranging from 1.9 to 97.7 ng m-3. With the concentrations found, estimated daily intakes through outdoor inhalation were calculated for each contaminant and for different population groups classified by age (infants, children and adults) in two possible exposure scenarios (low and high). Then, hazard quotients and carcinogenic risks were estimated for several compounds, those that had toxicological parameters available. This preliminary result showed no significant risks via ambient inhalation for the exposed population, however more research is needed to confirm the present results.
A realistic human skin model to study benzo[a]pyrene cutaneous absorption in order to determine the most relevant biomarker for carcinogenic exposure

Abstract:Polycyclic aromatic hydrocarbons (PAH) are ubiquitous pollutants, among which benzo[a]pyrene (B[a]P) is the only compound classified carcinogenic to humans. Besides pulmonary uptake, skin is the major route of PAH absorption during occupational exposure. Health risk due to PAH exposure is commonly assessed among workers using biomonitoring. A realistic human ex vivo skin model was developed to explore B[a]P diffusion and metabolism to determine the most relevant biomarker following dermal exposure. Three realistic doses (0.88, 8.85 and 22.11 nmol/cm2) were topically applied for 8, 24, and 48 h. B[a]P and its metabolites were quantified by liquid chromatography coupled with fluorimetric detection. The impact of time, applied dose, and donor age were estimated using a linear mixed-effects model. B[a]P vastly penetrated the skin within 8 h. The major metabolites were 3-hydroxybenzo[a]pyrene (3-OHB[a]P) and 7,8,9,10-tetrahydroxy-7,8,9,10-tetrahydrobenzo[a]pyrene (B[a]P-tetrol). This latter predominantly derives from the most carcinogenic metabolite of B[a]P, benzo[a]pyrene-7,8-diol-9,10-epoxide (BPDE), as well as benzo[a]pyrene-9,10-diol-7,8-epoxide (reverse-BPDE). Benzo[a]pyrene-trans-7,8-dihydrodiol (B[a]P-7,8-diol) was a minor metabolite, and benzo[a]pyrene-trans-4,5-dihydrodiol (B[a]P-4,5-diol) was never quantified. Unmetabolized B[a]P bioavailability was limited following dermal exposure since less than 3% of the applied dose could be measured in the culture medium. B[a]P was continuously absorbed and metabolized by human skin over 48 h. B[a]P-tetrol production became saturated as the applied dose increased, while no effect was measured on the other metabolic pathways. Age had a slight positive effect on B[a]P absorption and metabolism. This work supports the relevance of B[a]P-tetrol to assess occupational exposure and carcinogenic risk after cutaneous absorption of B[a]P.

Deployment Optimization of a Boom for FalconSAT-3 using Elastic Memory Composite Material

Abstract:Rationale for the inclusion of deployable structures onboard small satellites is ever increasing. For example, replacing traditional mass-expulsion control thrusters with micropropulsion ion thrusters on extendible booms can significantly reduce fuel mass requirements for attitude control systems. However, current flight-heritage booms are rendered inadequate when held to the stringent mass and mechanical requirements necessary to justify such a change. To address the deficiencies of existing boom technologies, a new generation of deployable structures must be developed. Paramount in this endeavor will be the ability to incorporate new materials into the design of next-generation deployable space structures. One promising new material/ technology for longeron members of deployable booms is TEMBOTM Elastic Memory Composite (EMC). EMC retains the structural properties of traditional fiber reinforced composites, i.e. high stiffness to mass ratio, while possessing the ability to behave as a shape memory material. These characteristics enable the primary structural component of a boom to additionally function as the primary deployment mechanism. This paper will focus on the developmental efforts encountered while advancing EMC from a material concept to a viable boom technology. In particular, this paper will introduce a family of deployable EMC booms and then outline the down select process employed during the development of the baseline United States Air Force Academy FalconSat-3 microsat boom.
Long-term spatiotemporal variation of benzo[a]pyrene in Japan: Significant decrease in ambient concentrations, human exposure, and health risk

Abstract:Although Benzo[a]pyrene (BaP) is considered carcinogenic to humans, the health effects of exposure to ambient levels have not been sufficiently investigated. This study estimated the long-term spatiotemporal variation of BaP in Japan over nearly two decades at a fine spatial resolution of 1 km. This study aimed to obtain an accurate spatiotemporal distribution of BaP that can be used in epidemiological studies on the health effects of ambient BaP exposure. The annual BaP concentrations were estimated using an ensemble machine learning approach using various predictors, including the concentrations and emission intensities of the criteria air pollutants, and meteorological, land use, and traffic-related variables. The model performance, evaluated by location-based cross-validation, exhibited satisfactory accuracy (R2 of 0.693). Densely populated areas showed higher BaP levels and greater temporal reduction, whereas BaP levels remained higher in some industrial areas. The population-weighted BaP in 2018 was 0.12 ng m-3, a decrease of approximately 70% from its 2000 value of 0.44 ng m-3, which was also reflected in the estimated excess number of lung cancer incidences. Accordingly, the proportion of BaP exposure below 0.12 ng m-3, which is the BaP concentration associated with an excess lifetime cancer risk of 10-5, reached 67% in 2018. Our estimates can be used in epidemiological studies to assess the health effects of BaP exposure at ambient concentrations.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) can cause adverse effects on human health. The relative contributions of their two major intake routes (diet and inhalation) to population PAH exposure are still unclear. We modeled the contributions of diet and inhalation to the overall PAH exposure of the population of Beijing in China, and assessed their human incremental lifetime cancer risks (ILCR) using a Mont Carlo simulation approach. The results showed that diet accounted for about 85% of low-molecular-weight PAH (L-PAH) exposure, while inhalation accounted for approximately 57% of high-molecular-weight PAH (H-PAH) exposure of the Beijing population. Meat and cereals were the main contributors to dietary PAH exposure. Both gaseous- and particulate-phase PAHs contributed to L-PAH exposure through inhalation, whereas exposure to H-PAHs was mostly from the particulate-phase. To reduce the cancer incidence of the Beijing population, more attention should be given to inhaled particulate-phase PAHs with considerable carcinogenic potential.

Abstract:Daily dietary and inhalation exposures to 16 parent polycyclic aromatic hydrocarbons (PAHs) and urinary excretion of 13 monohydroxy metabolites (OHPAHs) were monitored for 12 non-smoking university students in Beijing, China, during a controlled feeding experiment. The relationship between the urinary excretion of OHPAHs and the uptake of PAHs was investigated. The results suggest severe exposure of the subjects to PAHs via both dietary and inhalation pathways. Large increase of most urinary OHPAHs occurred after the ingestion of lamb kabob. Higher concentrations of OHPAHs were observed for female subjects, with the intakes of parent PAHs lower than those by males, likely due to the gender differences in metabolism. It appears that besides 1-PYR, metabolites of PHE could also be used as biomarkers to indicate the short-term dietary exposure to PAHs and urinary 3-BaA may serve as the biomarker for inhalation intake of high molecular weight PAHs.
The past 40 years' assessment of urban-rural differences in Benzo[a]pyrene contamination and human health risk in coastal China

Abstract:China has implemented many environmental regulations to battle against polycyclic aromatic hydrocarbon (PAH) contamination since the 1990s. It remains unclear how the exposure levels of PAHs changed quantitatively since reform and opening up in 1978 in China, whether the human health risks decreased or not, and how about the discrepancy between urban and rural areas. Here, taking Benzo[a]pyrene (BaP) in the rapidly urbanized Bohai region of China as a case, we used the improved Berkeley-Trent-Urban-Rural model to simulate the multimedia concentrations of BaP from 1980 to 2020 based on BaP emissions at a regional scale. The total emission of BaP in 1990 was the highest, with a value of 240 t, while the urban emission peaked in 2010. The BaP emissions from rural areas were two to seven times higher than urban areas, and the differences became smaller over time. Despite this, the average modeled BaP concentrations in urban air and soil were two to tens fold higher than in rural areas, particularly in highly urbanized or industrialized cities. Mostly, the concentrations of BaP in rural areas peaked in 1990, while those in urban areas peaked in 1990 or 2010. Early urbanized Beijing and Tianjin were the hot-spot cities of BaP contamination before 2000, while after 2010, higher concentrations were found in late industrialized Shandong and Hebei. BaP posed potential cancer risks to local residents, and air inhalation accounted for more than 80 % of the total risk. Under the stronger implementation of environmental regulations since the 1990s, it showed great health benefits, particularly for the urban residents in Beijing and Tianjin. The biggest decline in cancer risk was found in the period 2010-2020, and the average decreasing rates were 61.4 % and 57.4 % for urban and rural areas, respectively.

Abstract:Tetrabromobisphenol-A (TBBPA) is an emerging organic pollutant and a commonly used brominated flame retardant that has received much attention owing to its toxicity. Although TBBPA is ubiquitously detected in atmospheric particulate matter and dust, few studies have investigated the sub-chronic inhalation exposure to TBBPA. To further understand the excretion characteristics and tissue accumulation of TBBPA after inhalation exposure, we used the rat model to conduct a sub-chronic inhalation exposure study. Male rats were administered with different doses of aerosol TBBPA (12.9, 54.6, 121.6, and 455.0 mg/m3). TBBPA was found in the excretion (feces and urine) and all the target tissues (lung, liver, heart, thymus gland, spleen, testicles, muscles, kidneys, brain and serum). Feces were the main route of excretion, which contributed 19.18% to 72.54% (urine <0.10%). TBBPA excretion through feces following inhalation administration was much higher than that following oral and dermal exposure, thereby indicating lower bioavailability of TBBPA under inhalation exposure. Liver and serum showed higher levels of TBBPA compared with those of other tissues, thereby suggesting tissue-specific accumulation of TBBPA in rats. Owing to the relative non-invasiveness of serum sampling and greatest TBBPA concentration among the tissues, serum is a suitable matrix for estimation of TBBPA bioaccumulation after inhalation exposure.

Abstract:Phthalate esters (PAEs) are widely used as plasticizers in consumer products. PAEs are a group of environmental hormone which disrupts human and animals' endocrine systems. Different occupational groups are exposed to various levels of PAEs. In the present study, four typical occupational groups were chosen, including doctors, college teachers, college students, and drivers who worked in public traffic system. In order to understand the exposure levels to PAEs via inhalation, air samples were collected from multiple microenvironments including indoor and outdoor in Hangzhou to measure the gas and particle concentrations of six PAEs, together with time spent in different microenvironments of these four groups. A comprehensive PAEs exposure model was built to estimate the daily PAEs exposure through inhalation, oral and dermal pathways. The Monte Carlo simulation results show that doctors were exposed to the highest level of PAEs, and consequently had the highest health risk among these four occupational groups. In contrast, college students had the lowest health risk. By setting the exposure level of staying in residences as the baseline, doctors and drivers were two occupations exposed to high PAEs health risk. Di-(2-ethylhexyl) phthalate (DEHP) was the largest contributor among the six phthalates, posing moderate health risk (10-5-10-6) to every occupation. For traffic microenvironments alone, the total exposure levels for different transportation modes were in the descending order of busses, cars, cabs, tubes, motor bikes, and walking.

Abstract:Environmental exposures are one of the greatest threats to human health, yet we lack tools to answer simple questions about our exposures: what are our personal exposure profiles and how do they change overtime (external exposome), how toxic are these chemicals, and what are the sources of these exposures? To capture variation in personal exposures to airborne chemicals in the gas and particulate phases and identify exposures which pose the greatest health risk, wearable exposure monitors can be deployed. In this study, we deployed passive air sampler wristbands with 84 healthy participants (aged 60–69 years) as part of the Biomarkers for Air Pollutants Exposure (China BAPE) study. Participants wore the wristband samplers for 3 days each month for five consecutive months. Passive samplers were analyzed using a novel gas chromatography high resolution mass spectrometry data-processing workflow to overcome the bottleneck of processing large datasets and improve confidence in the resulting identified features. The toxicity of chemicals observed frequently in personal exposures were predicted to identify exposures of potential concern via inhalation route or other routes of airborne contaminant exposure. Three exposures were highlighted based on elevated toxicity: dichlorvos from insecticides (mosquito/malaria control), naphthalene partly from mothballs, and 183 polyaromatic hydrocarbons from multiple sources. Other exposures explored in this study are linked to diet and personal care products, cigarette smoke, sunscreen, and antimicrobial soaps. We highlight the potential for this workflow employing wearable passive samplers for prioritizing chemicals of concern at both the community and individual level, and characterizing sources of exposures for follow up interventions.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) are a class of important organic pollutants widely emitted from anthropogenic activities, with a general distribution in the gas and particulate phases. Some PAHs are carcinogenic, teratogenic, and mutagenic. Inhalation exposure to PAHs is correlated with adverse health outcomes in the respiratory and cardiovascular systems. Thus, it is significant to determine the exposure level of the general population. This study summarizes the evaluation methods for PAH exposure, focusing on different exposure parameters. External exposure can be determined via the collection of the environmental pollution concentration through active samplers or passive samplers during environmental monitoring or personal sampling. Time-activity patterns give critical exposure information that captures the exposure period, origin, and behaviors. Modeling is a labor-less approach for human exposure estimation, and microenvironmental exposure requires specific research. It is important to select appropriate methods to quantify the exposure level to provide accurate data to establish the exposure–risk relationship and make scientific suggestions for the protection of public health.
