Abstract:The prevalence of the two most common neurodegenerative diseases, Parkinson’s disease (PD) and Alzheimer’s Disease (AD), are expected to rise alongside the progressive aging of society. Both PD and AD are classified as proteinopathies with misfolded proteins α-synuclein, amyloid-β, and tau. Emerging evidence suggests that these misfolded aggregates are prion-like proteins that induce pathological cell-to-cell spreading, which is a major driver in pathogenesis. Additional factors that can further affect pathology spreading include oxidative stress, mitochondrial damage, inflammation, and cell death. Nanomaterials present advantages over traditional chemical or biological therapeutic approaches at targeting these specific mechanisms. They can have intrinsic properties that lead to a decrease in oxidative stress or an ability to bind and disaggregate fibrils. Additionally, nanomaterials enhance transportation across the blood-brain barrier, are easily functionalized, increase drug half-lives, protect cargo from immune detection, and provide a physical structure that can support cell growth. This review highlights emergent nanomaterials with these advantages that target oxidative stress, the fibrillization process, inflammation, and aid in regenerative medicine for both PD and AD.

Abstract:Alzheimer's disease (AD), one of the most common chronic neurodegenerative diseases , is characterized by memory impairment, synaptic dysfunction, and character mutations. The pathological features of AD are Aβ accumulation, tau protein enrichment, oxidative stress, and immune inflammation. Since the pathogenesis of AD is complicated and ambiguous, it is still challenging to achieve early detection and timely treatment of AD. Due to the unique physical, electrical, magnetic, and optical properties of nanoparticles (NPs), nanotechnology has shown great potential for detecting and treating AD. This review provides an overview of the latest developments in AD detection via nanotechnology based on NPs with electrochemical sensing, optical sensing, and imaging techniques. Meanwhile, we highlight the important advances in nanotechnology-based AD treatment through targeting disease biomarkers, stem-cell therapy and immunotherapy. Furthermore, we summarize the current challenges and present a promising prospect for nanotechnology-based AD diagnosis and intervention.

White blood cell count nadir following remission induction chemotherapy is predictive of outcome in older adults with acute myeloid leukemia

Abstract:Kinetics of white blood cell (WBC) elimination following induction chemotherapy for older adults with acute myeloid leukemia (AML) may serve as a surrogate for its effectiveness and safety by enabling real-time prognostication. We reviewed 122 older adults with AML treated at the Cleveland Clinic. Recursive partitioning analysis was used to identify optimal cut points in nadir WBC count and time to WBC nadir that correlate with survival. Multivariable analysis identified time to WBC nadir less than or equal to 10 days (HR 2.15, 95%CI 1.12 – 4.12, p = 0.02), low WBC nadir (less than 0.04×109/l, HR 2.68, 95%CI 1.15 – 6.23, p = 0.02) and high WBC nadir (greater than 0.12×109/l HR 1.5, 95%CI 0.96 – 2.37, p = 0.08), as predictors of worse outcomes. Time to WBC nadir predicts survival. The absolute WBC nadir value follows a J-curve, with lower value indicating a worse outcome.

Abstract:Alzheimer’s disease (AD) is a devastating disease of the aging population characterized by the progressive and slow brain decay due to the formation of extracellular plaques in the hippocampus. AD cells encompass tangles of twisted strands of aggregated microtubule binding proteins surrounded by plaques. Delivering corresponding drugs in the brain to deal with these clinical pathologies, we face a naturally built strong, protective barrier between circulating blood and brain cells called the blood–brain barrier (BBB). Nanomedicines provide state-of-the-art alternative approaches to overcome the challenges in drug transport across the BBB. The current review presents the advances in the roles of nanomedicines in both the diagnosis and treatment of AD. We intend to provide an overview of how nanotechnology has revolutionized the approaches used to manage AD and highlight the current key bottlenecks and future perspective in this field. Furthermore, the emerging nanomedicines for managing brain diseases like AD could promote the booming growth of research and their clinical availability.

Abstract:Alzheimer’s disease（AD） is a progressive and fatal neurodegenerative condition and the most prevalent cause of dementia. This disease is characterized by progressive cognitive impairment. The prevalence of AD is currently affecting more than 35 million people and is rising worldwide. No efficient therapy is currently available due to low drug potency and a number of various obstacles to delivery. Recent nanotechnological advancements have the potential to offer promising therapeutic options. Progress on nanomaterials as well as their applications in biomedicine is receiving increasing attention, especially the advantages of nanomaterial-based drug delivery systems. The aim of this review is to comprehensively summarize the latest developments in nanomaterial-based strategies for AD treatment, including nanoparticles, liposomes and other options for the delivery of therapeutic compounds and scaffolds for cell delivery strategies. Future research directions are also proposed. We hope this review can provide important information to guide the future development of nanomaterials in AD treatment.

Abstract:Abstract: The potential of nanotechnology in advancing the diagnosis and treatment of neurodegenerative diseases is explored in this comprehensive literature review. Through an examination of various studies that utilize nanotechnology models. The aim of this paper is to evaluate whether these models can be considered a significant breakthrough in the field of neurodegeneration. The findings of these studies suggest that nanotechnology has the capacity to improve existing therapeutic approaches, create novel and safe compounds, and develop more precise imaging techniques and diagnostic methods for neurodegenerative diseases. With the emergence of the nanomedicine era, a new and innovative approach to diagnosing and treating these conditions has been introduced. Notably, the researchers' development of a nanocarrier drug delivery tool demonstrates immense potential compared to conventional therapy, as it maximizes therapeutic efficacy and minimizes undesirable side effects.

Abstract:BACKGROUND Partial cystectomy is a bladder preserving treatment option for patients with urachal adenocarcinoma of the bladder. The procedure involves the full thickness surgical removal of the tumour, the surrounding bladder wall and the urachal remnant. Oncologically, it is imperative that the excision margins are clear of disease, whereas a functionally adequate residual bladder capacity is valued by the patient. It is not always possible to palpate the extent of the tumour intra-abdominally. We describe a simple technique of using methylene blue to outline the border of the planned excision.

Thus, effective therapeutic approaches are indeed necessary to address this growing concern. Current treatment regimens primarily help to relieve the symptoms, nevertheless, with nanotechnology-based approaches the nerve damage can be regressed along with a reduction in inflammation and improved brain delivery. Researchers and healthcare professionals are working diligently to develop innovative treatments and interventions to improve the quality of life for individuals affected by AD. This review paper aims to provide comprehensive insights on disease pathogenesis, FDA-approved treatment, recent advancements in drug therapy, and innovative formulation approaches along with an update on drugs in different clinical phases. Abstract:Alzheimer's disease (AD) is considered to have a multifactorial etiology. The hallmark of AD is progressive neurodegeneration, which is characterized by the deepening loss of memory and a high mortality rate in the elderly. The neurodegeneration in AD is believed to be exacerbated following the intercoupled cascades of extracellular amyloid beta (Aβ) plaques, uncontrolled microglial activation, and neuroinflammation. Current therapies for AD are mostly designed to target the symptoms, with limited ability to address the mechanistic triggers for the disease. Abstract:Background: Vilazodone is the latest US Food and Drug Administration approved antidepressant agent available in the USA. Its putative mechanism of antidepressant action enhances the release of serotonin across the brain’s serotonergic pathways specifically by inhibiting the serotonin transporter, similar to a selective serotonin reuptake inhibitor (SSRI), and simultaneously stimulating serotonin-1a receptors via partial agonism, similar to the anxiolytic buspirone. This combined activity in the single vilazodone agent has been termed by the authors as being a serotonin partial agonist and reuptake inhibitor or (SPARI). Methods: A MEDLINE and Internet search was conducted and the resultant preclinical and clinical evidence was reviewed. The authors attempt to review laboratory data, animal model data and human trial data to develop a translational theory on the mechanism of antidepressant action of this agent and also its adverse effect potential. Results: Randomized, controlled empirical data for vilazodone have gained it approval for treating major depressive disorder. It combines two well known pharmacodynamic mechanisms of serotonergic action into a novel agent. Although no head-to-head studies against other antidepressants have been published, the efficacy data for vilazodone appear comparable to other known antidepressants, with similar gastrointestinal side effects to SSRI or serotonin norepinephrine reuptake inhibitor (SNRI) antidepressants, but possibly with a lower incidence of sexual side effects and weight gain. Discussion: Vilazodone will lend itself to the current armamentarium in the treatment of major depressive disorder and may hold promise for patients who cannot tolerate other antidepressants. Its unique SPARI mechanism of action could also be efficacious for patients who do not respond to SSRI or SNRI antidepressant monotherapies.

Abstract:In recent years, the incidental rate of neurodegenerative disorders has increased proportionately with the aging population. Alzheimer's disease (AD) is one of the most commonly reported neurodegenerative disorders, and it is estimated to increase by roughly 30% among the aged population. In spite of screening numerous drug candidates against various molecular targets of AD, only a few candidates - such as acetylcholinesterase inhibitors are currently utilized as an effective clinical therapy. However, targeted drug delivery of these drugs to the central nervous system (CNS) exhibits several limitations including meager solubility, low bioavailability, and reduced efficiency due to the impediments of the blood-brain barrier (BBB). Current advances in nanotechnology present opportunities to overcome such limitations in delivering active drug candidates. Nanodrug delivery systems are promising in targeting several therapeutic moieties by easing the penetration of drug molecules across the CNS and improving their bioavailability. Recently, a wide range of nano-carriers, such as polymers, emulsions, lipo-carriers, solid lipid carriers, carbon nanotubes, metal based carriers etc., have been adapted to develop successful therapeutics with sustained release and improved efficacy. Here, we discuss few recently updated nano-drug delivery applications that have been adapted in the field of AD therapeutics, and future prospects on potential molecular targets for nano-drug delivery systems.

Abstract:Alzheimer's disease (AD) still prevails and continues to increase indiscriminately throughout the 21st century, and is thus responsible for the depreciating quality of health and associated sectors. AD is a progressive neurodegenerative disorder marked by a significant amassment of beta-amyloid plaques and neurofibrillary tangles near the hippocampus, leading to the consequent loss of cognitive abilities. Conventionally, amyloid and tau hypotheses have been established as the most prominent in providing detailed insight into the disease pathogenesis and revealing the associative biomarkers intricately involved in AD progression. Nanotheranostic deliberates rational thought toward designing efficacious nanosystems and strategic endeavors for AD diagnosis and therapeutic implications. The exceeding advancements in this field enable the scientific community to envisage and conceptualize pharmacokinetic monitoring of the drug, sustained and targeted drug delivery responses, fabrication of anti-amyloid therapeutics, and enhanced accumulation of the targeted drug across the blood-brain barrier (BBB), thus giving an optimistic approach towards personalized and precision medicine. Current methods idealized on the design and bioengineering of an array of nanoparticulate systems offer higher affinity towards neurocapillary endothelial cells and the BBB. They have recently attracted intriguing attention to the early diagnostic and therapeutic measures taken to manage the progression of the disease. In this article, we tend to furnish a comprehensive outlook, the detailed mechanism of conventional AD pathogenesis, and new findings. We also summarize the shortcomings in diagnostic, prognostic, and therapeutic approaches undertaken to alleviate AD, thus providing a unique window towards nanotheranostic advancements without disregarding potential drawbacks, side effects, and safety concerns.

Abstract:Alzheimer's disease (AD) is a chronic neurodegenerative disorder associated with dementia. To overcome the limitations of diagnosis and treatment nanotechnology research is bringing more needed and optimum clinical results. The use of nanocarriers for delivering bio-actives to treat neurodegenerative diseases has shown many benefits over common therapies such as the possibility of loading various payloads, controllable delivery, and amenability to surface modification with target ligands that can promote the crossing of the blood-brain barrier (BBB) for efficient distribution of bioactive into the brain. This review spotlights the applications of nanocarriers such as magnetic nanoparticles, carbon nanotubes, quantum dots, polymeric nanocapsules, multifunctional nanoliposomes, and nanoemulsions in the treatment of neurodegenerative diseases. The production of biosensors for the identification of Alzheimer's disease biomarkers has made considerable progress due to advances in nanoscience. The special properties of nanomaterials (such as graphene, carbon nanotubes, silver, gold, magnetic nanoparticles, polymers, and quantum dots) can improve the optical and electrochemical behavior of transducers. Therefore, electrochemical and optical biosensors with better sensitivity, selectivity, and prolonged stability desired. After all, the methods that focused on the identification of multiple analytes should be strengthened to reduce misdiagnosis.

Abstract:Introduction: Alzheimer's Disease (AD) is a progressive neurodegenerative pathology characterized by the presence of neuritic plaques and neurofibrillary tangles. The most important markers in AD pathology include excessive accumulation of amyloid beta (Aβ42) and phosphorylated tau (P-tau) proteins. One of the possible therapeutic strategies entails the elimination of such deposits by inhibiting Aβ aggregation. For years, one of the major problems in the treatment of AD has been the limited ability to deliver drugs to the brain for reasons related to poor solubility, low bioavailability, and the impact of the blood-brain barrier (BBB).Areas covered: In recent years, the authors have observed an increasing scientific interest in nanotechnological solutions as the factors potentially capable of facilitating the treatment of neurodegenerative diseases. The authors discuss recent reports regarding the use of nanotechnology in the therapy and treatment of AD.Expert opinion: The current advances in nanotechnology promise a chance to overcome the obstacles posed by said limitations. The size and diversity of nanoparticles in terms of both composition and shape create new possibilities for a variety of therapeutic applications, also in the context of the treatment and diagnostics of neurodegenerative diseases, for instance in combination with magnetic resonance imaging.

Abstract:Two-dimensional (2D) nanomaterials have been widely used in pharmaceutical applications due to their advantages of higher drug loading, controllable drug release, high stability, biodegradability, biocompatibility, and generally lower toxicity. This review presents state-of-the-art advancement in the context of utilizing 2D nanomaterials for the diagnosis and treatment of Alzheimer's disease (AD). The core pathological features of AD include amyloid plaques formed by amyloid beta protein (Aβ) aggregation in the cerebral cortex and hippocampus, and the self-assemble of Aβ peptides into β-sheet-rich aggregates. Early and accurate detection of AD biomarkers provide potential means for its diagnosis and treatment. Studies have indicated that 2D nanomaterials can significantly improve the detection accuracy and specificity of AD biomarkers (such as Aβ protein, tau protein, acetylcholinesterase (AChE), Acetylcholine (ACh), MicroRNAs, and S100β protein). In the treatment of AD, inhibiting the aggregation of Aβ and transforming the β-sheet structure of Aβ aggregates into harmless structures has been proved to be the effective strategies. 2D nanomaterials have application potential in the treatment of AD because they can degrade Aβ peptides or inhibit Aβ peptide aggregation through photodynamic or photothermal effects. However, most of the in vivo cytotoxicity mediated by nanomaterials has not yet been resolved.

Conditions, like Alzheimer's, Parkinson's, and Huntington's diseases, among others, share common characteristics, such as neuronal loss, misfolded protein aggregation, and nervous system dysfunction. One of the major obstacles in treating these diseases is the presence of the blood-brain barrier, limiting the delivery of therapeutic agents to the central nervous system. Nanotechnology offers promising solutions to overcome these challenges. In Alzheimer's disease, NPs loaded with various compounds have shown remarkable promise in preventing amyloid-beta (Aβ) aggregation and reducing neurotoxicity. Parkinson's disease benefits from improved dopamine delivery and neuroprotection. Huntington's disease poses its own set of challenges, but nanotechnology continues to offer innovative solutions. The promising developments in nanoparticle-based interventions for neurodegenerative diseases, like amyotrophic lateral sclerosis (ALS) and multiple sclerosis (MS), have offered new avenues for effective treatment. Nanotechnology represents a promising frontier in biomedical research, offering tailored solutions to the complex challenges posed by neurodegenerative diseases. While much progress has been made, ongoing research is essential to optimize nanomaterial designs, improve targeting, and ensure biocompatibility and safety. Nanomaterials possess unique properties that make them excellent candidates for targeted drug delivery and neuroprotection. They can effectively bypass the blood-brain barrier, opening doors to precise drug delivery strategies. This review explores the extensive research on nanoparticles (NPs) and nanocomposites in diagnosing and treating neurodegenerative disorders. Abstract:Alzheimer's disease (AD) remains one of the most challenging neurodegenerative disorders to treat, with oxidative stress playing a significant role in its pathology. Recent advancements in nanoenzymes technology offer a promising approach to mitigate this oxidative damage. Nanoenzymes, with their unique enzyme-mimicking activities, effectively scavenge reactive oxygen species and reduce oxidative stress, thereby providing neuroprotective effects. This review delves into the underlying mechanisms of AD, focusing on oxidative stress and its impact on disease progression. We explore the latest developments in nanoenzymes applications for AD treatment, highlighting their multifunctional capabilities and potential for targeted delivery to amyloid-beta plaques. Despite the exciting prospects, the clinical translation of nanoenzymes faces several challenges, including difficulties in brain targeting, consistent quality production, and ensuring safety and biocompatibility. We discuss these limitations in detail, emphasizing the need for rigorous evaluation and standardized protocols. This paper aims to provide a comprehensive overview of the current state of nanoenzymes research in AD, shedding light on both the opportunities and obstacles in the path towards effective clinical applications.

Abstract:Neuronanomedicine merges nanotechnology and neuroscience in the pursuit of engineering therapeutic interventions for neurological disorders, including Alzheimer's disease (AD) and Parkinson's disease (PD). While no nanoparticle‐based drug delivery systems (NDDSs) are yet approved for use for targeting the central nervous system, this review critically analyses the development of NDDSs for the improvement of currently approved therapeutics for the symptomatic treatment of AD and PD. It showcases how NDDSs can help therapeutic payloads overcome existing limitations, such as insufficient drug accumulation in the brain and limited effectiveness, by enhancing their pharmacokinetics, bioavailability, brain penetration and accumulation, and overall therapeutic efficacy through drug encapsulation, manipulation of nanoparticle properties, and nanoparticle surface functionalisation. However, we also draw attention to widespread issues in the field that impede progress, including the poor selection of in vitro models and the inadequate design of pre‐clinical in vivo studies. We further advocate for greater standardisation of study design and reporting requirements in the future, which would likely enhance outcomes and expedite the translation of neuronanomedicines.

Abstract:Abstract Alzheimer's disease is a debilitating, progressive neurodegenerative disorder characterized by the progressive accumulation of abnormal proteins, including amyloid plaques and intracellular tau tangles, primarily within the brain. Lysosomes, crucial intracellular organelles responsible for protein degradation, play a key role in maintaining cellular homeostasis. Some studies have suggested a link between the dysregulation of the lysosomal system and pathogenesis of neurodegenerative diseases, including Alzheimer's disease. Restoring the normal physiological function of lysosomes hold the potential to reduce the pathological burden and improve the symptoms of Alzheimer's disease. Currently, the efficacy of drugs in treating Alzheimer's disease is limited, with major challenges in drug delivery efficiency and targeting. Recently, nanomaterials have gained widespread use in Alzheimer's disease drug research owing to their favorable physical and chemical properties. This review aims to provide a comprehensive overview of recent advances in using nanomaterials (polymeric nanomaterials, nanoemulsions, and carbon-based nanomaterials) to enhance lysosomal function in treating Alzheimer's disease. This review also explores new concepts and potential therapeutic strategies for Alzheimer's disease through the integration of nanomaterials and modulation of lysosomal function. In conclusion, this review emphasizes the potential of nanomaterials in modulating lysosomal function to improve the pathological features of Alzheimer's disease. The application of nanotechnology to the development of Alzheimer's disease drugs brings new ideas and approaches for future treatment of this disease.
