Abstract:Traditional spectral analysis is prone to integration and signals overlap. It makes the extracted information unavailable or inaccurate, limiting its application in the detection of complex components. Therefore, two-dimensional correlation spectroscopy (2D-COS) that was superior to traditional spectral analysis has been established, by extracting useful information under certain chemical or physical stimulus from a series of spectra to improve the spectral resolution. In this study, with the identification of the different origins and parts of Panax notoginseng as an example. First, as a comparison, the attenuated total reflection infrared spectroscopy (ATR-FTIR) of P. notoginseng samples combined with chemometric methods was applied to identify it. Then, we generated synchronous and asynchronous 2D-COS spectral images from the fingerprint regions in the spectrum, and established a Residual convolutional neural network (ResNet) to identify the different origins and parts. Finally, the externally verified was applied to evaluate the accuracy of the model. By comparison, the 2D-COS was more suitable for the identification of the origins and parts of P. notoginseng than traditional spectroscopy. The identification accuracy of these samples of synchronous 2D-COS spectral images in the training set and test set were both greater than 99%. And the results of the externally verified indicated that synchronous 2D-COS spectral images were significantly outperforming asynchronous 2D-COS spectral images, and the sensitivity, specificity, and accuracy of the samples are both better than 0.99. Furthermore, when the sample number was relatively small or large differences in sample number were large, the synchronous 2D-COS spectral images could also successfully distinguish the different origins and parts well of P. notoginseng. In general, the ResNet model could provide a better discriminant model, and perform high-resolution processing.

Abstract:Puerariae Thomsonii Radix (PTR) is not only widely used in disease prevention and treatment but is also an important raw material as a source of starch and other food. The growth years of PTR are closely related to its quality. The rapid and nondestructive identification of growth year is essential for the quality control of PTR and other traditional Chinese medicines. In this study, we proposed a convolutional neural network (CNN)-based classification framework in conjunction with hyperspectral imaging (HSI) technology for the rapid identification of the growth years of PTRs. Traditional treatment methods (i.e., multiplicative scatter correction, standard normal variate, and Savitzky-Golay smoothing) combined with machine learning algorithms (i.e., random forest, logistic regression, naive Bayes, and eXtreme gradient boost) were used as baseline models. Among them, the F1-score of CNN-based models based on PTRs' outer surfaces was over 90%, outperforming all the other baseline models. These results showed that it was feasible to use a deep learning algorithm in conjunction with HSI technology to identify the growth years of PTR. This method provides a fast, nondestructive, and simple method of identifying the growth years of PTR. It can be easily applied to other scenarios, such as for the identification of the locality or years of growth for other traditional Chinese herbs.

Abstract:<p>Polysaccharide, the main component of <em>Polygonatum kingianum</em>, has been proven to have many pharmacological effects, such as antioxidant, anti-aging and anti-bacteria. The accumulation of polysaccharides was greatly influenced by the growth age of <em>P. kingianum</em>. Therefore, a fast and low-cost chemometric method is needed to identify the growth age. Attenuated total reflection-Fourier transform infrared (ATR-FTIR) and ultraviolet visible (UV-Vis) spectroscopy of <em>P. kingianum</em> with four growth ages (80 samples) were collected and analyzed by chemometric methods. The correlation between 13 agronomic traits and polysaccharide accumulation was discussed. A data fusion strategy was used to identify the growth age of <em>P. kingianum</em> by extracting the latent variables (LVs) and principal components (PCs) of the spectra. The results founded that the polysaccharide content showed a negative correlation the fibrous root number, leaf length, leaf width, stem diameter and fresh weight on the ground part. The ATR-FTIR and UV-Vis spectra in <em>P. kingianum</em> with different growth ages were similar. The regions of 1750–1500 cm<sup>−1</sup>, 1000–750 cm<sup>−1</sup> and 200–250 nm, 330–360 nm are important variables for discriminating four growth ages of <em>P. kingianum</em>. The classification effect of data fusion models was better than that of single spectra models with 100% accuracy as well as perfect sensitivity and specificity. Hierarchical cluster analysis shown that the samples were divided into two categories: one 5-year-old and the other 2, 3 and 4-year-old. In summary, the polysaccharide content in 4-year-old <em>P. kingianum</em> by seed reproduction was the highest. Data fusion of ATR-FTIR and UV-Vis spectra combined with PLS-DA models could be used to accurately distinguish <em>P. kingianum</em> in different growth ages, which was valuable for the identification and evaluation of other medicinal and edible homologous plants. Meanwhile, it also provides a theoretical basis for the harvesting age and cultivation conditions of <em>P. kingianum</em>.</p>

Abstract:To identify wild and cultivated Gastrodia elata quickly and accurately, this study is the first to apply three‐dimensional correlation spectroscopy (3DCOS) images combined with deep learning models to the identification of G. elata. The results concluded that the deep learning model is more effective than the traditional chemometric model and has greater potential for application in the identification of wild and cultivated G. elata. To identify wild and cultivated Gastrodia elata quickly and accurately, this study is the first to apply three‐dimensional correlation spectroscopy (3DCOS) images combined with deep learning models to the identification of G. elata. The spectral data used for model building do not require any preprocessing, and the spectral data are converted into three‐dimensional spectral images for model building. For large sample studies, the time cost is minimized. In addition, a partial least squares discriminant analysis (PLS‐DA) model and a support vector machine (SVM) model are built for comparison with the deep learning model. The overall effect of the deep learning model is significantly better than that of the traditional chemometric models. The results show that the model achieves 100% accuracy in the training set, test set, and external validation set of the model built after 46 iterations without preprocessing the original spectral data. The sensitivity, specificity, and the effectiveness of the model are all 1. The results concluded that the deep learning model is more effective than the traditional chemometric model and has greater potential for application in the identification of wild and cultivated G. elata.

A new effective method for identifying boletes species based on FT-MIR and three dimensional correlation spectroscopy projected image processing

Abstract:This study proposed the necessity of identifying the species for boletes in combination with the medicinal value, nutritional value and the problems existing in the industrial development of boletes. Based on the preprocessing of Fourier transform mid-infrared spectroscopy (FT-MIR) by 1st, 2nd, SNV, 2nd + MSC and 2nd + SG, Multilayer Perceptron (MLP) and CatBoost models were established. To avoid complex preprocessing and feature extraction, we try deep learning modeling methods based on image processing. In this paper, the concept of three-dimensional correlation spectroscopy (3DCOS) projection image was proposed, and 9 datasets of synchronous, asynchronous and integrative images are generated by computer method. In addition, 18 deep learning models were established for 9 image datasets with different sizes. The results showed that the accuracy of the three types of synchronous spectral models reached 100%, while the accuracy of the asynchronous spectral and integrative spectral models of 3DCOS projection images were 96.97% and 97.98% in the case of big datasets, which overcame the defects of poor modeling effect of asynchronous spectral and integrative spectral in previous two-dimensional correlation spectroscopy (2DCOS) studies. In conclusion, the modeling results of 3DCOS projection images are perfect, and we can apply this method to other identification fields in the future.

Abstract:Amomum tsao-ko (A. tsao-ko), is a medicine and food homology plant with high economic value. Different drying temperature affects flavor and quality of A. tsao-ko, which is closely related to its economic value and consumer acceptance. This research investigated the feasibility of combining Fourier-transform near infrared spectroscopy (FT-NIR) with chemometrics and machine learning to the identification of drying temperatures of A. tsao-ko. A total of 626 fresh samples from Honghe Prefecture, Yunnan, were dried to constant weight at 40 ℃ (62 h), 50 ℃ (54 h), 60 ℃ (43 h), 70 ℃ (35 h) and 80 ℃ (25 h) using an electric thermostatic drying oven. Principal component analysis (PCA) was used to observe the clustering of samples treated at different drying temperatures. Partial least squares-discriminant analysis (PLS-DA), support vector machines (SVM), and Residual Neural Network (ResNet) are used to build discriminant models. The accuracy of GS-SVM, GA-SVM, and ResNet models was more than 99% in the test set for identifying the drying temperature of A. tsao-ko. In contrast, the ResNet model based on synchronous two-dimensional correlation spectroscopy (2DCOS) images obtained the most satisfactory discrimination results. The use of appropriate data pre-processing (SD) can optimize the performance of the model to a certain extent and improve the accuracy of discrimination. This study demonstrates the feasibility of FT-NIR in the identification of the drying temperature of A. tsao-ko, providing a rapid and nondestructive method for the assessment and control of A. tsao-ko quality.

Abstract:A novel method was developed for the rapid determination of multi-indicators in corni fructus by means of near infrared (NIR) spectroscopy. Particle swarm optimization (PSO) based least squares support vector machine was investigated to increase the levels of quality control. The calibration models of moisture, extractum, morroniside and loganin were established using the PSO-LS-SVM algorithm. The performance of PSO-LS-SVM models was compared with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN). The calibration and validation results of PSO-LS-SVM were superior to both PLS and BP-ANN. For PSO-LS-SVM models, the correlation coefficients (r) of calibrations were all above 0.942. The optimal prediction results were also achieved by PSO-LS-SVM models with the RMSEP (root mean square error of prediction) and RSEP (relative standard errors of prediction) less than 1.176 and 15.5% respectively. The results suggest that PSO-LS-SVM algorithm has a good model performance and high prediction accuracy. NIR has a potential value for rapid determination of multi-indicators in Corni Fructus.
On-Line Monitoring of Extraction Process of Flos Lonicerae Japonicae Using Near Infrared Spectroscopy Combined with Synergy Interval Pls and Genetic Algorithm

Abstract:There is a growing need for the effective on-line process monitoring during the manufacture of traditional Chinese medicine to ensure quality consistency. In this study, the potential of near infrared (NIR) spectroscopy technique to monitor the extraction process of Flos Lonicerae Japonicae was investigated. A new algorithm of synergy interval PLS with genetic algorithm (Si-GA-PLS) was proposed for modeling. Four different PLS models, namely Full-PLS, Si-PLS, GA-PLS, and Si-GA-PLS, were established, and their performances in predicting two quality parameters (viz. total acid and soluble solid contents) were compared. In conclusion, Si-GA-PLS model got the best results due to the combination of superiority of Si-PLS and GA. For Si-GA-PLS, the determination coefficient (Rp2) and root-mean-square error for the prediction set (RMSEP) were 0.9561 and 147.6544μg/ml for total acid, 0.9062 and 0.1078% for soluble solid contents, correspondingly. The overall results demonstrated that the NIR spectroscopy technique combined with Si-GA-PLS calibration is a reliable and non-destructive alternative method for on-line monitoring of the extraction process of TCM on the production scale.

Abstract:Recently, deep learning has presented as a powerful approach to overcome the deficiencies of the conventional biochemical approaches. In this study, a method for discriminating medicinal plant Tetrastigma hemsleyanum from different origins was proposed using near-infrared spectroscopy (NIRS) and deep learning models. Support vector machine (SVM), self-adaptive evolutionary extreme learning machine (SAE-ELM), and convolutional neural network (CNN) were used to process the near-infrared spectral data (4000-5600 cm-1). The results indicated that the average recognition accuracy of SVM on the test set samples (n = 60) reached 90%. The average recognition accuracy of SAE-ELM was 98.3%, while CNN correctly discriminated 100% of T. hemsleyanum from different origins. Notably, CNN avoids tedious redundant data preprocessing and is also able to save the trained model for the next call to achieve rapid detection. As above, this study provides an effective deep learning-based method for discriminating the geographical origins of T. hemsleyanum as well as providing a convenient and satisfactory approach to ensure the famous-region of other medicinal plants.
The Storage Period Discrimination of Bolete Mushrooms Based on Deep Learning Methods Combined With Two-Dimensional Correlation Spectroscopy and Integrative Two-Dimensional Correlation Spectroscopy

Abstract:Boletes are favored by consumers because of their delicious taste and high nutritional value. However, as the storage period increases, their fruiting bodies will grow microorganisms and produce substances harmful to the human body. Therefore, we need to identify the storage period of boletes to ensure their quality. In this article, two-dimensional correlation spectroscopy (2DCOS) images are directly used for deep learning modeling, and the complex spectral data analysis process is transformed into a simple digital image processing problem. We collected 2,018 samples of boletes. After laboratory cleaning, drying, grinding, and tablet compression, their Fourier transform mid-infrared (FT-MIR) spectroscopy data were obtained. Then, we acquired 18,162 spectral images belonging to nine datasets which are synchronous 2DCOS, asynchronous 2DCOS, and integrative 2DCOS (i2DCOS) spectra of 1,750–400, 1,450–1,000, and 1,150–1,000 cm –1 bands. For these data sets, we established nine deep residual convolutional neural network (ResNet) models to identify the storage period of boletes. The result shows that the accuracy with the train set, test set, and external validation set of the synchronous 2DCOS model on the 1,750–400-cm –1 band is 100%, and the loss value is close to zero, so this model is the best. The synchronous 2DCOS model on the 1,150–1,000-cm –1 band comes next, and these two models have high accuracy and generalization ability which can be used to identify the storage period of boletes. The results have certain practical application value and provide a scientific basis for the quality control and market management of bolete mushrooms. In conclusion, our method is novel and extends the application of deep learning in the food field. At the same time, it can be applied to other fields such as agriculture and herbal medicine.

Abstract:Introduction Medicinal Polygonatum species is a widely used traditional Chinese medicine with high nutritional value, known for its anti‐fatigue properties, enhancement of immunity, delays aging, improves sleep, and other health benefits. However, the efficacy of different species varies, making the quality control of medicinal Polygonatum species increasingly important. Polysaccharides are important in medicinal Polygonatum species because of their potential functional properties, such as antioxidation, hypoglycemia, protection of intestinal health, and minimal toxicological effects on human health, as well as high polysaccharide levels. Objective This study developed a qualitative medicinal Polygonatum species model and a polysaccharides predictive model based on attenuated total reflection Fourier transform infrared spectroscopy (ATR‐FTIR) combined with a multivariate analysis approach. Materials and Methods ATR‐FTIR spectral information of 334 medicinal Polygonatum species samples was collected and the spectral information of different modes was analyzed. The ATR‐FTIR spectral differences of three medicinal Polygonatum species were studied by multivariate analysis combined with four spectral preprocessing and three variable selection methods. For the prediction of polysaccharides in Polygonatum kingianum Collett & Hemsl. (PK), we initially determined the actual content of 110 PK polysaccharide samples using the anthrone‐sulfuric acid method, then established partial least squares regression (PLSR) and kernel PLSR models in conjunction with attenuated total reflectance Fourier transform infrared (ATR‐FTIR) spectroscopy. Results In the visualization analysis, the orthogonal partial least squares‐discriminant analysis (OPLS‐DA) model based on second‐order derivative (SD) preprocessing was most suitable for medicinal Polygonatum species species binary classification, spectral differences between Polygonatum cyrtonema Hua (PC) and other species are evident; in the hard modeling, SD preprocessing improves the accuracy of non‐deep learning models for the classification of three medicinal Polygonatum species. In contrast, residual neural network (ResNet) models were the best choice for species identification without preprocessing and variable selection. In addition, the partial least squares regression (PLSR) model and Kernel‐PLSR model can quickly predict PK polysaccharides content, among them, the Kernel‐PLSR model with SD pretreatment has the best prediction performance, residual prediction deviation (RPD) = 7.2870, Rp = 0.9905. Conclusion In this study, we employed ATR‐FTIR spectroscopy and various treatments to discern different medicinal Polygonatum species. We also evaluated the effects of preprocessing methods and variable selection on the prediction of PK polysaccharides by PLSR and Kernel‐PLSR models. Among them, the ResNet model can achieve 100% correct classification of medicinal Polygonatum species without complex spectral preprocessing. Furthermore, the Kernel‐PLSR model based on SD‐ATR‐FTIR spectra had the best performance in polysaccharides prediction. In summary, by integrating ATR‐FTIR spectroscopy with multivariate analysis, this research accomplished the classification of medicinal Polygonatum species and the prediction of polysaccharides. The methodology offers the benefits of speed, environmental sustainability, and precision, highlighting its significant potential for practical applications. In future research, on the one hand, it can be further investigated using a portable infrared spectrometer, and on the other hand, infrared spectroscopy can also be applied to the prediction of other chemical components of medicinal Polygonatum species.

Abstract:Dendrobium officinale is a medicinal plant with high commercial value. The Dendrobium officinale market in Yunnan is affected by the standardization of medicinal material quality control and the increase in market demand, mainly due to the inappropriate harvest time, which puts it under increasing resource pressure. In this study, considering the high polysaccharide content of Dendrobium leaves and its contribution to today's medical industry, (Fourier Transform Infrared Spectrometer) FTIR combined with chemometrics was used to combine the yields of both stem and leaf parts of Dendrobium officinale to identify the different harvesting periods and to predict the dry matter content for the selection of the optimal harvesting period.

Abstract:The outer membrane (OM) protein components of a Vibrio cholerae O1 and four V. cholerae O139 strains, collected from cholera patients, were analysed by SDS-PAGE. A protein of 69 kDa molecular mass was observed only when the OMPs were prepared from strains grown in synthetic broth. As a result of passage in the rabbit ileal loop (RIL), virulence was enhanced, and a protein component around 18 kDa of the V. cholerae O139 OM became the major protein component. On immunoblot analysis with rabbit antiserum against V. cholerae O139 OM, it was shown that, apart from the major protein component of V. cholerae O1 OM of around 45 kDa and that of V. cholerae O139 OM of around 38 kDa, all other minor protein components were cross-reactive between the two serogroups. In immunoblot assays with convalescent sera obtained from V. cholerae O139-infected patients, it was observed that in addition to the lipopolysaccharide (LPS)-induced antibody, only the 38 kDa major protein component elicited considerable levels of antibody in the patient. Minor OM components of 18 kDa were detected in the immunoblot analysis by LPS-directed antibody, however, as the OM proteins are known to be associated with LPS.
Identification of Dendrobium Using Laser-Induced Breakdown Spectroscopy in Combination with a Multivariate Algorithm Model

Abstract:Dendrobium, a highly effective traditional Chinese medicinal herb, exhibits significant variations in efficacy and price among different varieties. Therefore, achieving an efficient classification of Dendrobium is crucial. However, most of the existing identification methods for Dendrobium make it difficult to simultaneously achieve both non-destructiveness and high efficiency, making it challenging to truly meet the needs of industrial production. In this study, we combined Laser-Induced Breakdown Spectroscopy (LIBS) with multivariate models to classify 10 varieties of Dendrobium. LIBS spectral data for each Dendrobium variety were collected from three circular medicinal blocks. During the data analysis phase, multivariate models to classify different Dendrobium varieties first preprocess the LIBS spectral data using Gaussian filtering and stacked correlation coefficient feature selection. Subsequently, the constructed fusion model is utilized for classification. The results demonstrate that the classification accuracy of 10 Dendrobium varieties reached 100%. Compared to Support Vector Machine (SVM), Random Forest (RF), and K-Nearest Neighbors (KNN), our method improved classification accuracy by 14%, 20%, and 20%, respectively. Additionally, it outperforms three models (SVM, RF, and KNN) with added Principal Component Analysis (PCA) by 10%, 10%, and 17%. This fully validates the excellent performance of our classification method. Finally, visualization analysis of the entire research process based on t-distributed Stochastic Neighbor Embedding (t-SNE) technology further enhances the interpretability of the model. This study, by combining LIBS and machine learning technologies, achieves efficient classification of Dendrobium, providing a feasible solution for the identification of Dendrobium and even traditional Chinese medicinal herbs.

Abstract:Boletes are favored by consumers because of their unique flavor, rich nutrition and delicious taste. However, the different nutritional values of each species lead to obvious price differences, so shoddy products appear on the market, which affects food safety. The aim of this study was to find a rapid and effective method for boletes species identification. In this paper, 1,707 samples of eight boletes species were selected as the research objects. The original Mid-Infrared (MIR) spectroscopy data were adopted for support vector machine (SVM) modeling. The 11,949 spectral images belong to seven data sets such as two-dimensional correlation spectroscopy (2DCOS) and three-dimensional correlation spectroscopy (3DCOS) were used to carry out Alexnet and Residual network (Resnet) modeling, thus we established 15 models for the identification of boletes species. The results show that the SVM method needs to process complex feature data, the time cost is more than 11 times of other models, and the accuracy is not high enough, so it is not recommended to be used in data processing with large sample size. From the perspective of datasets, synchronous 2DCOS and synchronous 3DCOS have the best modeling results, while one-dimensional (1D) MIR Spectrum dataset has the worst modeling results. After comprehensive analysis, the modeling effect of Resnet on the synchronous 2DCOS dataset is the best. Moreover, we use large-screen visualization technology to visually display the sample information of this research and obtain their distribution rules in terms of species and geographical location. This research shows that deep learning combined with 2DCOS and 3DCOS spectral images can effectively and accurately identify boletes species, which provides a reference for the identification of other fields, such as food and Chinese herbal medicine.

Abstract:Introduction: The cultivation and sale of medicinal plants are some of the main ways to meet the increased market demand for plant-based drugs. Panax notoginseng is a widely used Chinese medicinal material. The growth and accumulation of bioactive constituents mainly depend on a satisfactory growing environment. Additionally, the occurrence of market fraud means that care should be taken when purchasing. Methods: In this study, we report the correlation between saponins and climate factors based on high performance liquid chromatography (HPLC), and evaluate the influence of climate factors on the quality of P. notoginseng. In addition, the synchronous two-dimensional correlation spectroscopy (2D-COS) images of near infrared (NIR) data combined with the deep learning model were applied to traceability of geographic origins of P. notoginseng at two different levels (district and town levels). Results: The results indicated that the contents of saponins in P. notoginseng are negatively related to the annual mean temperature and the temperature annual range. A lower annual mean temperature and temperature annual range are favorable for the content accumulation of saponins. Additionally, high annual precipitation and high humidity are conducive to the content accumulation of Notoginsenoside R1 (NG-R1), Ginsenosides Rg1 (G-Rg1), and Ginsenosides Rb1 (G-Rb1), while Ginsenosides Rd (G-Rd), this is not the case. Regarding geographic origins, classifications at two different levels could be successfully distinguished through synchronous 2D-COS images combined with the residual convolutional neural network (ResNet) model. The model accuracy of the training set, test set, and external validation is achieved at 100%, and the cross-entropy loss function curves are lower. This demonstrated the potential feasibility of the proposed method for P. notoginseng geographic origin traceability, even if the distance between sampling points is small. Discussion: The findings of this study could improve the quality of P. notoginseng, provide a reference for cultivating P. notoginseng in the future and alleviate the occurrence of market fraud.
Portable near-infrared spectroscopy combined with Machine learning algorithms for the origin identification and quality evaluation of Acanthopanax senticosus

Abstract:The aim of this study is to develop a comprehensive quality assessment method for Acanthopanax senticosus using near-infrared (NIR) spectroscopy combined with machine learning algorithms. A qualitative discriminant analysis model was established to distinguish the samples of Acanthopanthus from different habitats, and the overall prediction accuracy was more than 85%. Moreover, A partial least squares regression (PLSR) model was built to predict the response values of Acanthopanax senticosus at each retention time, with the Acanthopanax senticosus dataset serving as the Y matrix and the NIR spectral dataset as the X matrix. The transformed Acanthopanax senticosus fingerprint closely matched the actual fingerprint, exhibiting high average similarity between the predicted and measured fingerprints for the test set samples (Pearson correlation coefficient = 0.8553, cosine similarity = 0.8627). The results of the cluster analysis demonstrated a high level of consistency. Converting NIR spectral fingerprints into HPLC fingerprint format enhanced interpretability and more intuitively displayed the components responsible for the diversity among samples from different origins. Meanwhile, partial least squares regression (PLSR) model and support vector machine regression (SVR) model for the prediction of the contents of moisture, total solids, protocatechuic acid, syringin and chlorogenic acid contents were successfully established. In addition, the models obtained by SVR show better quantitative predictive ability in general. The NIR spectra of Acanthopanax senticosus can well predict the values of moisture, total solids, protocatechuic acid, syringin and chlorogenic acid content with RMSEP values of 0.182, 0.797, 0.003, 0.007, 0.026, respectively. Overall, the results indicate that this rapid quality evaluation system can be utilized for the quality control of Acanthopanax senticosus.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Background</h3><p>Paridis Rhizoma (PR) is a famous traditional herbal medicine. Apart from two officially recorded species, viz. <em>Paris polyphylla</em> Smith var. <em>yunnanensis</em> (Franch.) Hand. - Mazz. (PPY) and <em>P. polyphylla</em> Smith var. <em>chinensis</em> (Franch.) Hara (PPC), there are still many other species used as folk medicine. It is necessary to understand the metabolic differences among <em>Par</em>is species.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Purpose</h3><p>To establish a strategy that can discover species-specific steroidal saponin markers to distinguish closely-related <em>Paris</em> herbs for quality and safety control.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>A new strategy of molecular-networking-guided discovery of species-specific markers was proposed. Firstly, the ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS) was applied to obtain the MS and MS/MS data of all samples. Then, molecular networking (MN) was created using MS/MS data to prescreen the steroidal saponins for subsequent analysis. Next, the principal component analysis (PCA) and orthogonal partial least square discriminant analysis (OPLS-DA) models were established to discover potential markers. Finally, the verification, identification and distribution of chemical markers were performed.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>A total of 126 steroidal saponins were screened out from five species using MN. Five species were classified successfully by OPLS-DA model, and 18 species-specific markers were discovered combining the variable importance in the projection (VIP) value, P value (one-way ANOVA) and their relative abundance. These markers could predict the species of <em>Paris</em> herbs correctly.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>These results revealed that this new strategy could be an efficient way for chemical discrimination of medicinal herbs with close genetic relationship.</p>
