Abstract:Phytolacca americana L. is of great interest as a traditional additive in various folk remedies in several countries, including Turkey. We aimed to determine the chemical profile (assisted by high-Performance liquid chromatography-electrospray ionization-tandem mass apectrometry (HPLC-ESI-MS/MS) experiments of three extracts obtained by different polarity solvents viz. ethyl acetate (to extract semipolar compounds), methanol and water (to extract highly polar metabolites) from P. americana leaves. Their anti-diabetic effects were investigated in vitro by assessing their inhibition toα-amylase and α-glucosidase. Assessment of the neuroprotective potential of the three extracts was carried out against acetyl-(AChE) and butyryl-(BChE) cholinesterase enzymes. HPLC-ESI-MS/MS experiments showed a total of 17 chromatographic peaks primarily classified to six flavonoids, two saponins, and six fatty acids. Antioxidant assays revealed remarkable activity for the ethyl acetate and methanol extracts. The BChE inhibition was considerably more significant (4.08 mg galantamine equivalent (GALAE)/g) for the ethyl acetate extract, whereas the methanol extract had good inhibitory efficacy for AChE (2.05 mg GALAE/g). Through network pharmacology, the compounds' mechanism of action of targeted key gene in their associated diseases were identified. The hubb gene signal transducer and activator of transcription 3 (STAT3) and tumour necrosis factor (TNFα) where the P. americana compound's site of action in inflammation bowel disease. The results offer possibilities for the prospective application of P. americana in metabolic regulation, blood glucose control, and as a source of bioactive compounds with cholinesterase enzyme inhibitory characteristics which could be of relevance in the cosmetic or pharmaceutical industry for combating melanogenesis.Communicated by Ramaswamy H. Abstract:Boerhavia diffusa is a great tropical plant and is widely used for various traditional purposes. In the present study, we examined the influence of solvents (dichloromethane, ethyl acetate, methanol and infusion (water)) on chemical composition and biological capabilities of B. diffusa. An UHPLC-HRMS method was used to determine the chemical characterization. The biological ability was examined for antioxidant, enzyme inhibitory and anti-cancer effects. To evaluate antioxidant effects, different chemical methods (ABTS, DPPH, CUPRAC, FRAP, metal chelating and phosphomolybdenum) were applied. With regard to enzyme inhibitory properties, cholinesterases, amylase, glucosidase and tyrosinase were used. The MDA-MB-231 breast cancer cell line was chosen to determine anticancer activity. Based on the UHPLC-HRMS analysis, 37 specialized metabolites were dereplicated and identified in the studied extracts. Results revealed the presence of 15 hydroxybenzoic, hydroxycinnamic, acylquinic acids, and their glycosides, one rotenoid, seven flavonoids, 12 fatty acids and two other glycosides. Among the tested extracts, the methanol extract showed a stronger antioxidant ability compared with other extracts. The methanol extract also showed the best inhibitory effects on tyrosinase and glucosidase. In the anti-cancer evaluation, the methanol extract showed stronger anticancer effects compared with water extract. In summary, our observations can contribute to the establishment of B. diffusa as a potential candidate for functional applications in the preparation.

Abstract:The members of the genus Hypericum have great potential to develop functional uses in nutraceutical and pharmaceutical applications. With this in mind, we aimed to determine the chemical profiling and biological properties of different extracts (ethyl acetate, methanol and water) from two Hypericum species (H. montbretii and H. origanifolium). We combined two approaches (LC-DAD-MS and LC-NMR) to identify and quantify chemical compounds of the extracts. Antioxidant properties (free radical quenching, reducing power and metal chelating) and enzyme inhibitory effects (cholinesterase, tyrosinase, amylase and glucosidase) were determined as biological properties. The tested extracts were rich in caffeic acid derivatives and flavonoids, and among them, 3-caffeoyl quinic acid and myricetin-3-O-rhamnoside were found to be the main compounds. The total phenolic and flavonoid levels were determined to be 50.97–134.99 mg GAE/g and 9.87–82.63 mg RE/g, respectively. With the exception of metal chelating, the methanol and water extracts showed stronger antioxidant properties than the ethyl acetate extracts. However, different results were obtained for each enzyme inhibition assay, and in general, the ethyl acetate extracts present more enzyme-inhibiting properties than the water or methanol extracts. Results from chemical and biological analyses were combined using multivariate analysis, which allowed establishing relationships between composition and observed effects of the Hypericum extracts based on the extraction solvents. To gain more insights between chemical compounds and enzyme-inhibiting effects, we performed molecular docking analysis. We observed favorable interactions between certain compounds and the tested enzymes during our analysis, confirming the data obtained from the multivariate approach. In conclusion, the obtained results may shed light on the road from natural sources to functional applications, and the tested Hypericum species may be considered potential raw materials, with promising chemical constituents and biological activities.

Abstract:To investigate the chemical composition and therapeutic properties of the Tanacetum nitens ( Boiss. & Noë) Grierson aerial parts, organic solvent extracts were prepared by sequential maceration in hexane, dichloromethane, ethyl acetate, and methanol; aqueous extracts were obtained by maceration or infusion. Spectroscopic extract analysis revealed T. nitens as a promising source of phytoconstituents with potential therapeutic applications. Tanacetum nitens ( Boiss. & Noë) Grierson is an aromatic perennial herb used in Turkish traditional medicine to treat headache, fever, and skin diseases. This study aimed to investigate the chemical composition, antioxidant, enzyme inhibition, and cytotoxic properties of T. nitens aerial parts. Organic solvent extracts were prepared by sequential maceration in hexane, dichloromethane, ethyl acetate, and methanol while aqueous extracts were obtained by maceration or infusion. Nuclear magnetic resonance (NMR) and LC‐DAD‐MS analysis allowed the identification and quantification of different phytoconstituents including parthenolide, tanacetol B, tatridin B, quinic acid derivatives, β‐sitosterol, and glycoside derivatives of quercetin and luteolin. The type and amount of these phytochemicals recovered by each solvent were variable and significant enough to impact the biological activities of the plant. Methanolic and aqueous extracts displayed the highest scavenging and ions‐reducing properties while the dichloromethane and ethyl acetate extracts exerted the best total antioxidant activity and metal chelating power. Results of enzyme inhibition activity showed that the hexane, ethyl acetate, and dichloromethane extracts had comparable anti‐acetylcholinesterase activity and the latter extract revealed the highest anti‐butyrylcholinesterase activity. The best α‐amylase and α‐glucosidase inhibition activities were obtained from the hexane extract. The dichloromethane and ethyl acetate extracts exhibited the highest cytotoxic effect against the prostate carcinoma DU‐145 cells. In conclusion, these findings indicated that T. nitens can be a promising source of biomolecules with potential therapeutic applications.

Abstract:Carissa carandas L. (Apocynaceae) is widely distributed in tropical and subtropical regions of Asia including Pakistan, India, Afghanistan, and Sri Lanka. C. carandas is considered as an integral component of traditional medicinal systems to combat several health ailments. The present study aimed to assess this plant's phytochemical contents and biological potential by performing sequential extraction, adopting a bioassay-guided approach. C. carandas powder was extracted with n-hexane to remove fatty substances and then residues were sequentially extracted with dichloromethane, methanol, and 50% methanol. All the sequential crude extracts were evaluated for phytochemical contents (total phenolics, flavonoids, and anthocyanins), in vitro antioxidant activity (FRAP, DPPH), in vitro anti-inflammatory activity (serum and egg albumin denaturation), in vivo anti-inflammatory activity (carrageenan- and formaldehyde-induced paw edema), and in vitro antimicrobial activity. Active crude extract was then partitioned using the liquid-liquid separation method followed by further separation of the active fraction by RP-HPLC. The active fraction was then subjected to LC-ESI-MS/MS analysis for tentative identification of bioactive metabolites responsible for its bioactive properties, followed by HPLC quantification. The analysis revealed methanol extract to have more phytochemical contents, radical scavenging properties, reduced inflammation in both models (in vitro and in vivo), and antimicrobial properties against urinary tract infection-causing agents as compared to dichloromethane and 50% methanol extracts. The ethyl acetate fraction obtained after liquid-liquid partitioning (LLP) of the active methanol extract exhibited more activity as compared to C. carandas methanol extract. RP-HPLC sub-fractionation yielded seven sub-fractions, but a slight decrease in biological potential was recorded. Therefore, LLP fraction B was subjected to further analysis. LC-ESI-MS/MS analysis led to the tentative identification of phenolic acids (chlorogenic acid, quinic acid), flavonoids (quercetin), and anthocyanins (peonidin-3-arabinoside, delphinidin-3-galactoside, delphinidin-3-rutinoside) in the active LLP ethyl acetate fraction. Chlorogenic acid, ellagic acid, and quinic acid were quantified as 17.6 μg/mg, 5.90 μg/mg, and 3.30 μg/mg, respectively, on a dry weight basis by HPLC. C. carandas may be considered a promising therapeutic plant, and the results of the current study provide more evidence to support the assertions made in ancient medical traditions. These findings highlight its promising applications in health, medicine, cosmetics, preservatives, and as a natural coloring agent.

Phytochemical Analysis and Biological Evaluation of Carob Leaf (Ceratonia siliqua L.) Crude Extracts Using NMR and Mass Spectroscopic Techniques

Abstract:Carob leaves have gained attention for their bioactive properties and traditional medicinal uses, including as treatment for diabetes, digestive disorders, and microbial infections. The aim of this study was to explore the phytochemical composition of carob leaf acetone extracts using advanced spectroscopic techniques. The combined use of heteronuclear nuclear magnetic resonance (NMR) experiments with 1D selective nuclear Overhauser effect spectroscopy (NOESY) offers detailed structural insights and enables the direct identification and quantification of key bioactive constituents in carob leaf extract. In particular, the NMR and mass spectrometry techniques revealed the presence of myricitrin as a predominant flavonoid, as well as a variety of glycosylated derivatives of myricetin and quercetin, in acetone extract. Furthermore, siliquapyranone and related gallotannins are essential constituents of the extract. The potent inhibitory effects of the carob leaf extract on Staphylococcus aureus (MIC = 50 μg mL−1) and a-glucosidase enzyme (IC50 = 67.5 ± 2.4 μg mL−1) were also evaluated. Finally, the antibacterial potency of carob leaf constituents were calculated in silico; digalloyl-parasorboside and gallic acid 4-O-glucoside exert a stronger bactericidal activity than the well-known myricitrin and related flavonoids. In summary, our findings provide valuable insights into the bioactive composition and health-promoting properties of carob leaves and highlight their potential for pharmaceutical and nutraceutical applications.

Abstract:Breonadia salicina (Vahl) Hepper & J.R.I. Wood is widely distributed throughout Africa. It is used ethnobotanically to treat various diseases. However, the metabolic profile of the Breonadia species is not well characterized and the metabolites that are responsible for the bioactivity of this plant remain unknown. Therefore, there is a need to determine the phytochemical and bioactivity profile to identify metabolites that contribute to the antidiabetic, anti-inflammatory and antiproliferation activity, including the genotoxicity and cytotoxic effects, of Breonadia salicina. The study is aimed at exploring the metabolomic profile antidiabetic, anti-inflammatory and antiproliferation activity, as well as the genotoxicity and cytotoxicity effects, of constituents of B. salicina. The compounds in the B. salicina extract were analyzed by ultra-performance liquid chromatography with quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS), and the resultant data were further analyzed using a molecular networking approach. The crude stem bark and root extracts showed the highest antidiabetic activity against α-amylase at the lowest test concentration of 62.5 μg/mL, with 74.53 ± 0.74% and 79.1 ± 1.5% inhibition, respectively. However, the crude stem bark and root extracts showed the highest antidiabetic activity against α-glucosidase at the lowest test concentration of 31.3 μg/mL, with 98.20 ± 0.15% and 97.98 ± 0.22% inhibition, respectively. The crude methanol leaf extract showed a decrease in the nitrite concentration at the highest concentration of 200 μg/mL, with cell viability of 90.34 ± 2.21%, thus showing anti-inflammatory activity. No samples showed significant cytotoxic effects at a concentration of 10 μg/mL against HeLa cells. Furthermore, a molecular network of Breonadia species using UPLC-QTOF-MS with negative mode electrospray ionization showed the presence of organic oxygen compounds, lipids, benzenoids, phenylpropanoids and polyketides. These compound classes were differentially distributed in the three different plant parts, indicating the chemical differences between the stem bark, root and leaf extracts of B. salicina. Therefore, the identified compounds may contribute to the antidiabetic and anti-inflammatory activity of Breonadia salicina. The stem bark, root and leaf extracts of B. salicina yielded thirteen compounds identified for the first time in this plant, offering a promising avenue for the discovery of new lead drugs for the treatment of diabetes and inflammation. The use of molecular networking produced a detailed phytochemical overview of this Breonadia species. The results reported in this study show the importance of searching for bioactive compounds from Breonadia salicina and provide new insights into the phytochemical characterization and bioactivity of different plant parts of Breonadia salicina.

Abstract:Caralluma europaea Guss. ( C. europaea ) is a medicinal plant used for cancer treatment. However, these treatments may be associated with complications that need to be investigated. This work aims to evaluate not only the chemical composition but also the hepatoprotective and anticancer properties of C. europaea extracts. The chemical constitution of the hydroethanolic extract was explored using gas chromatography-mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC). The hydroethanolic extract, flavonoids, and polyphenols-rich extract at 100, 15, and 50 mg/kg, respectively, were administered to acetaminophen-treated rats for seven days. We used Western blotting and Real-Time quantitative Polymerase Chain Reaction (RT-qPCR) to determine the protein and the mRNA levels of cancer stemness markers in pancreatic cancer cell lines MIA PaCa-2 and BxPC-3 exposed to increasing doses of C. europaea extracts. In silico analysis was used to evaluate the effects of phenolic compounds revealed in C. europaea on caspase-3 and HSP90, and on liver damage on CYP2E1. The primary phenolics detected by GC-MS and HPLC were ferulic acid and benzofurazan. The positive control group showed an increase in AST, ALT, ALP, triglycerides, and VLDL levels. C. europaea extracts demonstrated hepatoprotective effects by ameliorating acetaminophen-induced alterations of biochemical and hispathological parameters. Immunoblotting and RT-qPCR profiling of cancer stemness markers indicated a reduction in the expression levels of Oct-4 and Nanog proteins, as well as a reduction in the mRNA levels of CD133 by 50–60% and Sox2 by 80–90% in pancreatic cancer cells. Molecular docking showed that naringenin presented the highest docking Gscore on CYP2E1 (−8.199) and HSP90 (−7.742). In conclusion, C. europaea extracts could be considered as a safe and promising therapeutic strategy to sensitize pancreatic cancer cells to chemotherapy.

Abstract:Background: Salvia verbenaca of the Lamiaceae family is a Mediterranean plant widely used in the Moroccan traditional folk medicine. The aim of this work was to explore the phytochemical composition of Salvia verbenaca extracts and its antioxidant activity. Methods: Separation and identification of the major phytochemicals present in the two hexane and ethyl acetate explored extracts have been achieved through ultra-high-performance liquid chromatography separation technique coupled to photodiode array and high-resolution time-of-flight mass spectrometry detectors. Antioxidant activity of the obtained extracts was evaluated through DPPH• (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2'-azinobis(3-ethylbenzothiazoline6-sulfonic acid) diammonium salt) assays and the obtained results were compared to TROLOX (6-hydroxy2,5,7,8-tetramethylchroman-2-carboxylic acid). Results: Using the analysis technique indicated above, eighteen phytochemicals belonging to phenolic acids, phenolic diterpenes and flavonoids have been characterized on the basis of the obtained UV and mass spectroscopic properties and on the subsequent fragmentations. The antioxidant activity of the explored extracts showed potential scavenging activities compared to TROLOX. A detailed discussion of the attained results has been presented considering the activities observed of each extract. Conclusions: The research herein presented an analysis technique allowing to screen Salvia verbenaca phytochemicals. The explored plant could be considered as a source of functional phenolic compounds. These could be useful for further pharmacological studies such as new drugs design after clinic and its safety evaluation. It is thus hoped that the information presented here might prompt further studies that will possibly lead to development of therapeutic agents from this plant.

Abstract:In the present study, the methanolic extract of Calotropis procera root bark was subjected to solvent-solvent partitioning using n-hexane fraction (HF), dichloromethane fraction (DMF), ethyl acetate fraction (EAF), and methanol fraction (MF). The resultant fractions were tested for antioxidant activity using in vitro radical scavenging assay. Preliminary phytochemical investigation revealed the presence of varying proportions of secondary metabolites in solvent fractions such as glycosides, flavonoids, triterpenoids, sterols, and polyphenolic compounds. The total phenolic content of EAF was 25.7 ± 3.12 mg TAE/g followed by 19.05 ± 3.29 mg TAE/g in DMF. The total flavonoid content was 13.69 ± 1.74 mg QUE/g in DMF and 11.4 ± 1.88 mg QUE/g in EAF. The EAF showed significant radical-scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH, IC50 = 369.87 μg/mL), nitric oxide (IC50 = 317.46 μg/mL), hydrogen peroxide (H2O2 , IC50 = 396.85 μg/mL), and hydroxyl radicals (IC50 = 195.39 μg/mL). DMF was most effective in scavenging superoxide radicals (IC50 = 679.60 μg/mL), while greater metal chelating activity was exhibited by MF (IC50 = 614.73 μg/mL). Moreover, the total antioxidant activity for EAF was found to be 94.14 ± 9.114 mg AAE followed by DMF (68.10 ± 8.78 mg AAE). EAF also significantly reduced the formation of thiobarbituric acid reactive substances in a dose-dependent manner in CuSO4. The observed antioxidant effect might be attributed to the presence of secondary metabolites. Subsequent GC-MS analysis of EAF confirmed the presence of lupeol, α-amyrin, β-amyrin, and ursolic acid. The current investigation reveals that the high polyphenolic and antioxidant pentacyclic triterpenes in EAF of C. procera root bark methanol extract correlates with its good antioxidant activity and can provide protection against free radicals-induced damage in a variety of chronic health conditions.

Abstract:In disease treatment, the utilisation of medicinal plants has witnessed a discernible rise, driven by concerns over the adverse effects associated with synthetic drugs available in the market. Analyses of the plant Christia vespertilionis (L.f.) Bakh. F., indigenous to Malaysia, has suggested its antidiabetic property linked to α-glucosidase inhibition, but metabolites responsible for antidiabetic are unexplored. The metabolomics approaches and molecular docking simulations were integrated to identify the putative α-glucosidase inhibitors and their enzyme interaction. In this study, the crude leaves extracted from this plant were extracted using solvents of varying polarity, followed by gas and liquid chromatography coupled with mass spectrometry metabolomics. The highest inhibition activity in a mixture of n-hexane and ethyl acetate (1:1, v/v)) was observed. Six putative metabolites corresponding to antidiabetic activity were identified: palmitic acid (2), linolenic acid (4), 7-tetradecenal (5), aloeemodin-8-monoglucoside (14), bruceine I (15), and sanjidin B (16). The mechanism of action of all the identified compounds is competitive, mainly involving hydrophobic and hydrogen bonding interactions with the protein residues. Compounds 14, 15, and 16 exhibited strong binding capabilities with both enzyme crystal structures compared to the positive control, quercetin. The metabolites extracted from C. vespertilionis leaves have demonstrated promising antidiabetic effects. These antidiabetic compounds can potentially commercialise new drug candidates in managing diabetes conditions.

Abstract:Background: Canavalia rosea is a common tropical seacoast flowering plant from the family of Fabaceae which is reported as bay bean and coastal jack bean; has a wide range of therapeutic and nutraceutical properties. Aim: The present research aims to explore some pharmacological insights of the methanol extract C. rosea of leaves (MECR) and its chloroform fraction (CFCR) and n-hexane fraction (NFCR) through in-vitro and in-silico approaches. Methods: Different fractions of C. rosea were subjected to ferric reduction assay and total phenolic and flavonoid content assay to explore their antioxidant potential. The anti-inflammatory activity was assessed on the hypotonic-induced human erythrocyte-lysis model, while the protein denaturation method was applied for screening anti-arthritis properties of plant extract; and the cytotoxic activity was evaluated by brine shrimp lethality bioassay. In the in-silico study, molecular docking, pass prediction and ADME/T, analysis was used to investigate anti-arthritic, anti-inflammatory, antioxidant and cytotoxic potency of five selected compounds of C. rosea. Finally, the quantum chemical density functional test analysis was applied to investigate the chemical and physical properties of those compounds. Results: All the soluble organic extracts of C. rosea demonstrated moderate toxicity with strong antioxidants potential, in which MECR manifested the peak level of scavenging activity (2.49) on ferric reduction assay. MECR, CFCR & NFCR significantly protected lysis of human erythrocyte membrane induced by hypotonic solution, whereas MECR and CFCR were exhibited partially equal inhibitory activity. The in-vitro anti-arthritis assay, MECR, NFCR and CFCR showed strongly significant (p˂0.001) inhibitory potency at 500 μg/mL & 1000 μg/mL concentrations. The molecular docking simulations revealed strong binding of all compounds to specific receptors, with Rutin showing the highest biological reactivity followed by Daucosterol, β-Sitosterol, Stigmasterol, and Guanidine. All the compounds showed favorable reactivity patterns, with O and H atoms poised for nucleophilic and electrophilic attacks in chemical density functional calculations. Conclusion: The recent investigation suggests that C. rosea could have the potential source of antioxidant, anti-inflammatory, anti-arthritis and cytotoxic activity, prompting further investigation into their mechanisms.

Abstract:Caralluma edulis is a leafless succulent herb native to India and Pakistan, where it is used as emergency food and as potential herbal medicine. It was investigated for its bioactive chemicals and further medicinal properties. The objective of the current study was to accomplish the chemical and biological characterization of C. edulis . Chemical profiling was done through estimation of total bioactive contents and UHPLMS analysis. Biological screening was achieved through six different antioxidant and enzyme inhibition assays. Estimation of its phenolic and flavonoid contents revealed that ethyl acetate fraction (Ce-E) contains 14.15 mg GAE/g extract and 26.13 mg RE/g extract of phenolics and flavonoids, respectively, followed by the methanolic (Ce-M) and the water (Ce-W) soluble fractions. In free radical inhibitory assays, Ce-E fraction also exhibited highest activity (DPPH: 19.72 mg TE/g extract, ABTS: 48.66 mg TE/g extract) followed by Ce-M, whereas, in CUPRAC and FRAP assays, all the extracts, except water fraction, exhibited nearly equal potential in the range of 53.85–57.96 and 25.42–32.12 mg TE/g extract, respectively. In metal chelating antioxidant assay, only Ce-M and Ce-W fractions displayed considerable activities (25.88 and 23.55 mg EDTAE/g extract respectively), whereas, Ce-H exhibited the highest activity in phosphomolybdenum assay (1.03 mmolTE/g extract), against BChE (6.43 mg GALAE/g extract), α-glucosidase and α-amylase (6.98 and 0.47 mmol ACAE/g, respectively) and tyrosinase (64.46 mg KAE/g extract) enzymes. Ce-M and Ce-E also exhibited significant activity against tyrosinase (59.85 and 58.40 mg KAE/g extract, respectively). Crude methanolic extract (Ce-M) was analyzed to unveil its secondary metabolic picture; the UHPLCMS analysis discloses 105 compounds as phenolic acids, flavonoids, steroids and their glycosides, which makes the C. edulis extract a strong candidate for nutraceutical and functional food ingredient. Some of the metabolites were docked against all the tested enzymes to predict mode of actions and to substantiate the pharmaceutical nature of C. edulis .

Abstract:Ultrasound-assisted solvent extraction (UASE) was appslied to extract phytoconstituents from Semecarpus anacardium L. The effective extraction parameters were optimized using the Response Surface Methodology (RSM) with Central Composite Face Centered design (CCFC), and the optimization parameters were validated through Adaptive Neuro-Fuzzy Inference System (ANFIS) and Machine Learning (ML) algorithm models. The four independent parameters, i.e. ethanol concentration ( X 1 : 62–72% v/v), temperature ( X 2 : 38–48°C), ultrasonic-exposure time ( X 3 : 12–24 min), and particle size ( X 4 : 2–5 mm) were optimized. The maximum yield of phytocompounds was achieved at X 1 = 67%, X 2 = 43°C, X 3 = 18 min, and X 4 = 3.5 mm. The optimized yield of total phenolic, flavonoid, and antioxidant activity was considered when selecting process parameters. In this scenario, the optimized yields of total polyphenolic content (y 1 = 619.25 mg gallic acid equivalent (GAE)/g), total flavonoid content (y 2 = 151.24 mg Quercetin equivalents (QE)/g), free radical scavenging abilities (%DPPH*sc (y 3 = 66.41%), and %ABTS*sc (y 4 = 56.94%). The LC-MS peaks identify the presence of semecarpetin, butein, and amentoflavone, whereas GS-MS peaks represent seventeen gaseous components. Furthermore, the bioactive-rich optimized extract was nontoxic and supported the growth of macrophage- and epithelial cells in vitro . Conclusively, optimizing the UASE parameters enhanced the yield of bioactive compounds of S. anacardium L.

Abstract:Dysphania ambrosioides has been reported to have many medicinal properties, due to its possession of a multitude of biologically active molecules contained in its leaves. However, very few studies have been reported to evaluate their pharmacological properties. Consequently, in the present study, many computational tools have been performed to predict drug similarity and ADMET properties. Besides, the inhibitory potential of D.ambrosioides major compounds against Bacterial, Fungal and cardiovascular main receptor targets has been investigated. This study suggests that Carvone oxide, 5‐Isopropenyl‐2‐Methylenecyclohexanol, and Caryophyllene oxide were the most active molecules belonging to D. ambrosioides Leaves, possessing drug‐likeness with satisfactory bioactivity scores, having good pharmacokinetic values. Metabolism and toxicities were further studied using FAME3, GLORY, and pred‐hERG. Slight cardiotoxicity and cytotoxicity were predicted, respectively, for Caryophyllene oxide and Carvone oxide, 5‐Isopropenyl‐2‐Methylenecyclohexanol. Good inhibitory activities of the three compounds against Bacterial, Fungal, and Cardiovascular receptor targets. Hence, this is a comprehensive in silico approach to evaluate D.ambrosioides Leaves main phytocompounds in the background of its potential in future drug development

Abstract:Premna barbata Wall. ex Schauer is used traditionally as folkloric medicines for the treatment of differentpathological conditions. The first reported constituent from the bark of the plant was an iridoid glycoside premnosidic acid having antioxidant activity. Other species of this genus i.e., Premna latifolia, Premna tomentosa has shown to have antioxidant and cytotoxic activities. Despite the ethnomedicinal uses, no scientific evidence in support of antitumor activity on the stem bark of Premna barbata is reported so far. Hence, the current work aims to assess anticancer potentiality of different extracts of P. barbata on various cancer cell lines. Different extracts i.e., Petroleum ether extract (PBPE), Chloroform extract (PBCE), Ethyl acetate extract (PBEE) and Alcoholic extract (PBAE) were prepared and on each extract in-vitro antiproliferative activity was performed by using SRB assay. The most potent extract i.e., PBEE was then evaluated for antioxidant activity. Qualitative phytochemical investigation of different extracts indicates the presence of proteins, carbohydrates, steroids/triterpenoids, alkaloids, glycosides, flavonoids, and phenolic constituents. Ethyl acetate extract of Premna barbata gives potent cytotoxic activity in all cancer cell lines but more selectively to human colon cancer cell line (COLO-205) with GI 50 44.6 μg/ml. The phenolic and flavonoid content in ethyl acetate extract was 3.43±0.09 mg GE/g and 4.28±0.18mg QE/g respectively. Nineteen compounds were observed in positive (+) ESI mode chromatograms when LC-MS analysis was carried out. The LC-MS analysis by positive ionization mode, the predicted compounds such as Geniposidic acid (synonym: Premnosidic acid) and Rutin were detected. The cytotoxicity observed on cancerous cell lines in our study may be due to the presence of observed compounds. So, it can be concluded that Premna barbata stembark has remarkable cytotoxic activity against different tumor cell lines but the effect is more on colon cell lines as compared to others.

Abstract:Rapid screening of botanical extracts for the discovery of bioactive natural products was performed using a fractionation approach in conjunction with flow-injection high-resolution mass spectrometry for obtaining chemical fingerprints of each fraction, enabling the correlation of the relative abundance of molecular features (representing individual phytochemicals) with the read-outs of bioassays. We applied this strategy for discovering and identifying constituents of Centella asiatica (C. asiatica) that protect against Aβ cytotoxicity in vitro. C. asiatica has been associated with improving mental health and cognitive function, with potential use in Alzheimer's disease. Human neuroblastoma MC65 cells were exposed to subfractions of an aqueous extract of C. asiatica to evaluate the protective benefit derived from these subfractions against amyloid β-cytotoxicity. The % viability score of the cells exposed to each subfraction was used in conjunction with the intensity of the molecular features in two computational models, namely Elastic Net and selectivity ratio, to determine the relationship of the peak intensity of molecular features with % viability. Finally, the correlation of mass spectral features with MC65 protection and their abundance in different sub-fractions were visualized using GNPS molecular networking. Both computational methods unequivocally identified dicaffeoylquinic acids as providing strong protection against Aβ-toxicity in MC65 cells, in agreement with the protective effects observed for these compounds in previous preclinical model studies.

Abstract:The over production of free radicals in the human body is known to cause oxidative stress which plays an important role in the pathogenesis of many diseases. Therefore, the search for natural antioxidants would continue to be a major area of research for many decades. This study was to investigate the in vitro antioxidant activities and to determine the phytochemicals present in Cassia sieberiana. The methanol stem bark extract of C. sieberiana was subjected to phytochemical screening and Gas Chromatography-Mass Spectrometry (GC-MS) analysis. Its antioxidant activities were assayed in vitro using nitric oxide scavenging assay, ferric reducing antioxidant potential assay, lipid peroxidation inhibitory assay and 2-diphenyl-1-picrylhydrazyl scavenging assay. The phytochemical screening of the stem bark extract of C. sieberiana showed saponins, alkaloids, flavonoids, steroids, tannins, phlobatannins, terpernoids, cardiac glycosides, phenolics and reducing sugar. The GC-MS Table revealed that the methanol extract has 51 bioactive compounds with 3-0-Methyl-d-glucose having the highest percentage of 12.16 % while 5.alpha-Androstan-3-beta-ol,4,4-dimethyl- having the lowest percentage of 0.09 %. The antioxidant study showed concentration-dependent radical scavenging activity of the methanol stem bark extract of C. sieberiana. This concentration-dependent result was also observed when compared with ascorbic acid used as the standard drug. The presence of different phytochemicals such as alkaloids, saponins, tannins, phenols, flavonoids, among other things, might be responsible for C. sieberiana antioxidant activity. The study concluded that the methanol stem bark extract of C. sieberiana contains 51 bioactive compounds and showed good antioxidant activity which could be therapeutic in several human diseases. Abstract:This study was designed to check the potential of secondary metabolites of the selected plants; Citrullus colocynthis, Solanum nigrum, Solanum surattense, Calotropis procera, Agave americana, and Anagallis arvensis for antioxidant, antibacterial, antifungal, and antidiabetic agents. Plant material was soaked in ethanol/methanol to get the crude extract, which was further partitioned via solvent extraction technique. GCMS and FTIR analytical techniques were applied to check the compounds responsible for causing antioxidant, antimicrobial, and antidiabetic activities. It was concluded that about 80% of studied extracts/fractions were active against α-amylase, ranging from 43 to 96%. The highest activity (96.63%) was exhibited by butanol fractions of A. arvensis while the least response (43.65%) was shown by the aqueous fraction of C. colocynthis and the methanol fraction of fruit of S. surattense. The highest antioxidant activity was shown by the ethyl acetate fraction of Anagallis arvensis (78.1%), while aqueous as well as n-hexane fractions are the least active throughout the assay. Results showed that all tested plants can be an excellent source of natural products with potential antimicrobial, antioxidant, and antidiabetic potential. The biological response of these species is depicted as a good therapeutic agent, and, in the future, it can be encapsulated for drug discovery.
