Abstract:The exponentially increasing population poses a serious threat to global food security. Concurrently, the climate change condition also limits crop productivity by enhancing the effect of biotic and abiotic stressors. The traditional crop improvement programs are not enough to meet the food and nutritional requirements of such a progressive population. Recently, the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein (Cas) based genome editing tool adopted from bacterial adaptive immune system against invading foreign DNA has demonstrated its tremendous potential in the sector of crop improvement. CRISPR/Cas-mediated targeting can activate, repress, or completely abolish the gene function. CRISPR/Cas-mediated interventions can produce biofortified crops by targeting negative regulators or activating positive regulators for nutrients. Thus, it can address nutritional security concerns. The advancement in CRISPR/Cas-mediated genome editing, encompassing base and prime editing has paved the way to modify an organism's genome in a predictable and precise manner. The use of morphogenetic regulators can omit the problem of tissue culture stages, which is one of the major bottlenecks in plant genome editing. CRISPR/Cas-based genome editing has been performed in many crop plants to induce biotic and abiotic stress tolerance, increase quality and nutritional values, enhance productivity, and prevent post-harvest losses. In this review article, we summarize the progress, challenges opportunities and regulatory landscape of genome editing for the improvement of various traits in crop plants.

Abstract:The global climate change and unfavourable abiotic and biotic factors are limiting agricultural productivity and therefore intensifying the challenges for crop scientists to meet the rising demand for global food supply. The introduction of applied genetics to agriculture through plant breeding facilitated the development of hybrid varieties with improved crop productivity. However, the development of new varieties with the existing gene pools poses a challenge for crop breeders. Genetic engineering holds the potential to broaden genetic diversity by the introduction of new genes into crops. But the random insertion of foreign DNA into the plant's nuclear genome often leads to transgene silencing. Recent advances in the field of plant breeding include the development of a new breeding technique called genome editing. Genome editing technologies have emerged as powerful tools to precisely modify the crop genomes at specific sites in the genome, which has been the longstanding goal of plant breeders. The precise modification of the target genome, the absence of foreign DNA in the genome-edited plants, and the faster and cheaper method of genome modification are the remarkable features of the genome-editing technology that have resulted in its widespread application in crop breeding in less than a decade. This review focuses on the advances in crop breeding through precision genome editing. This review includes: an overview of the different breeding approaches for crop improvement; genome editing tools and their mechanism of action and application of the most widely used genome editing technology, CRISPR/Cas9, for crop improvement especially for agronomic traits such as disease resistance, abiotic stress tolerance, herbicide tolerance, yield and quality improvement, reduction of anti-nutrients, and improved shelf life; and an update on the regulatory approval of the genome-edited crops. This review also throws a light on development of high-yielding climate-resilient crops through precision genome editing.

Abstract:Vegetable crops are known as protective foods due to their potential role in a balanced human diet, especially for vegetarians as they are a rich source of vitamins and minerals along with dietary fibers. Many biotic and abiotic stresses threaten the crop growth, yield and quality of these crops. These crops are annual, biennial and perennial in breeding behavior. Traditional breeding strategies pose many challenges in improving economic crop traits. As in most of the cases the large number of backcrosses and stringent selection pressure is required for the introgression of the useful traits into the germplasm, which is time and labour-intensive process. Plant scientists have improved economic traits like yield, quality, biotic stress resistance, abiotic stress tolerance, and improved nutritional quality of crops more precisely and accurately through the use of the revolutionary breeding method known as clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR-associated protein-9 (Cas9). The high mutation efficiency, less off-target consequences and simplicity of this technique has made it possible to attain novel germplasm resources through gene-directed mutation. It facilitates mutagenic response even in complicated genomes which are difficult to breed using traditional approaches. The revelation of functions of important genes with the advancement of whole-genome sequencing has facilitated the CRISPR-Cas9 editing to mutate the desired target genes. This technology speeds up the creation of new germplasm resources having better agro-economical traits. This review entails a detailed description of CRISPR-Cas9 gene editing technology along with its potential applications in olericulture, challenges faced and future prospects.

The modifications to the genetic makeup of crops, genetic engineering has facilitated the development of varieties with improved traits such as pest and disease resistance, herbicide tolerance, enhanced nutritional content, and resilience to environmental stresses. This review article provides a comprehensive analysis of the current status, benefits, challenges, and future prospects of genetic engineering in agriculture, the historical evolution of genetic engineering technologies and their application in agriculture. The significant advancements in techniques such as CRISPRCas9, TALENs, ZFNs, and RNA interference, which have enabled targeted and efficient genetic modifications and explores the wide range of genetically modified (GM) crops that have been developed and commercialized, including Bt cotton, Roundup Ready soybeans, and Golden Rice, emphasizing their impact on crop yields and agricultural sustainability, the benefits of genetic engineering, underscoring its role in increasing crop productivity, reducing the reliance on chemical pesticides and herbicides, and enhancing the nutritional quality of food crops. The potential of genetic engineering to develop crops with novel traits, such as biofortified crops and those with improved tolerance to abiotic stresses, the challenges and concerns associated with genetic engineering, including regulatory and biosafety issues, environmental impacts, ethical considerations, and economic challenges, the review discusses the future prospects of genetic engineering in agriculture, considering the integration of genetic engineering with other emerging technologies like precision agriculture and digital farming. Abstract:The fact that the human population is projected to cross the mark of staggering 8.3 billion by the year 2030 has raised the serious concerns to ensure the food security. Traditional plant breeding has played a pivotal role in the development of numerous high yielding cultivars. However, the over-exploitation of cultivated pool of crops and significantly limited success rate of tapping potential of wild germplasm has narrowed down the chances of crop improvements. Along with these breeding constraints, the global climate perturbation scenario has challenged the ongoing efforts being made in cultivar development. Plant undergoes epigenetic changes, which are heritable in nature, to respond to environmental stimuli. Various epigenetic-level alternations such as methylation patterns and histone modifications are observed when plant undergoes the biotic as well as abiotic stress events. The alterations induce variations in the patterns of gene expression, thereby facilitating plant to adapt the stress conditions. These epigenetic marks in the context of breeding are termed as epi-breeding and the epialleles derived from epimutations, either via spontaneous manner or chemical mutations, which are stably inheriting and fixed can greatly contribute in the expression of the target gene. The advents in the genome editing tools such as ZFNs, TALENs and CRISPR have revolutionized the way of tinkering the genome in precise and efficient manner. Harnessing the potential of these genome editing tools for engineering crop epigenome to turn on the switch for desirable gene or suppress the overexpression of undesirable gene can yield more precise and stable epigenetic modifications which can ultimately lead to the development of climate-smart varieties.

Abstract:Conventional plant breeding has contributed enormously towards feeding the world and has played crucial roles in the development of modern society. The conventional method creates variation by transferring genes between or within the species. In general, these methods are more expensive and takes more time, to overcome these limitations, new technology is required. Genome editing is a powerful tool for biotechnology applications, with the capacity to alter the function of any gene. With the availability of gene information for the majority of the traits, genome editing emerged as a potential to create a new variation with the introduction of any transgene. The important genome editing tools used nowadays are ZFNs, TALEN, Pentatricopeptide repeats protein, adenine base editor, RNA interference, and CRISPR/Cas9. These tools have opened a new era for crop improvement. Due to the complex genetic architecture of most traits, it is challenging to edit genes controlling them. To overcome these challenges, genome editing provides a broader perspective. Among the above-mentioned tools, CRISPR/Cas9 is the most powerful tool for gene editing. These technologies are being used to create abiotic and biotic resistance crop varieties.

Abstract:Genome-editing (GE) is having a tremendous influence around the globe in the life science community. Among its versatile uses, the desired modifications of genes, and more importantly the transgene (DNA)-free approach to develop genetically modified organism (GMO), are of special interest. The recent and rapid developments in genome-editing technology have given rise to hopes to achieve global food security in a sustainable manner. We here discuss recent developments in CRISPR-based genome-editing tools for crop improvement concerning adaptation, opportunities, and challenges. Some of the notable advances highlighted here include the development of transgene (DNA)-free genome plants, the availability of compatible nucleases, and the development of safe and effective CRISPR delivery vehicles for plant genome editing, multi-gene targeting and complex genome editing, base editing and prime editing to achieve more complex genetic engineering. Additionally, new avenues that facilitate fine-tuning plant gene regulation have also been addressed. In spite of the tremendous potential of CRISPR and other gene editing tools, major challenges remain. Some of the challenges are related to the practical advances required for the efficient delivery of CRISPR reagents and for precision genome editing, while others come from government policies and public acceptance. This review will therefore be helpful to gain insights into technological advances, its applications, and future challenges for crop improvement.

Abstract:A world with zero hunger is possible only through a sustainable increase in food production and distribution and the elimination of poverty. Scientific, logistical, and humanitarian approaches must be employed simultaneously to ensure food security, starting with farmers and breeders and extending to policy makers and governments. The current agricultural production system is facing the challenge of sustainably increasing grain quality and yield and enhancing resistance to biotic and abiotic stress under the intensifying pressure of climate change. Under present circumstances, conventional breeding techniques are not sufficient. Innovation in plant breeding is critical in managing agricultural challenges and achieving sustainable crop production. Novel plant breeding techniques, involving a series of developments from genome editing techniques to speed breeding and the integration of omics technology, offer relevant, versatile, cost-effective, and less time-consuming ways of achieving precision in plant breeding. Opportunities to edit agriculturally significant genes now exist as a result of new genome editing techniques. These range from random (physical and chemical mutagens) to non-random meganucleases (MegaN), zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), clustered regularly interspaced short palindromic repeats (CRISPR)/associated protein system 9 (CRISPR/Cas9), the CRISPR system from Prevotella and Francisella1 (Cpf1), base editing (BE), and prime editing (PE). Genome editing techniques that promote crop improvement through hybrid seed production, induced apomixis, and resistance to biotic and abiotic stress are prioritized when selecting for genetic gain in a restricted timeframe. The novel CRISPR-associated protein system 9 variants, namely BE and PE, can generate transgene-free plants with more frequency and are therefore being used for knocking out of genes of interest. We provide a comprehensive review of the evolution of genome editing technologies, especially the application of the third-generation genome editing technologies to achieve various plant breeding objectives within the regulatory regimes adopted by various countries. Future development and the optimization of forward and reverse genetics to achieve food security are evaluated.

Abstract:Genome editing technology revolutionized crop improvement technology through sequence-specific, precise, site-directed, safe genetic manipulation and combat the major 21st century challenge such as achieving world food security meeting rising global food demand and improving food nutrition in the face of rapidly changing climate conditions. Crop improvement using conventional and molecular breeding approaches takes time, causing biosafety concerns and cannot equipoise with raising demand. Genome editing system like zinc finger nuclease (ZFN), transcription activator-like effector nucleases (TALEN), and clustered regularly interspaced short palindromic repeats (CRISPR) made a desirable targeted modification in crops for improving crop yield, nutraceutical quality and also enhance tolerance to environmental stress (biotic or abiotic) through add the desirable trait(s) and remove the undesirable. Genome manipulation tools progression creates new breakthroughs and speeds up crop improvement through site-directed mutagenesis efficiently for crop improvements to meet the ever-increasing global demand for food and produce more resilient crop with great flexibility to combat climate change.

Abstract:Crop improvement is a continuous process in agriculture which ensures ample supply of food, fodder and fiber to burgeoning world population. Despite tremendous success in plant breeding and transgenesis to improve the yield-related traits, there have been several limitations primarily with the specificity in genetic modifications and incompatibility of host species. Because of this, new breeding techniques (NBTs) are gaining worldwide attention for crop improvement programs. Among the NBTs, genome editing (GE) using site-directed nucleases (SDNs) is an important and potential technique that overcomes limitations associated with classical breeding and transgenesis. These SDNs specifically target a compatible region in the gene/genome. The meganucleases (MgN), zinc finger nucleases (ZFN), transcription activator-like effectors nucleases (TALENs), and clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated endonuclease (Cas) are being successfully employed for GE. These can be used for desired or targeted modifications of the native endogenous gene(s) or targeted insertion of cis/trans elements in the genomes of recipient organisms. Applications of these techniques appear to be endless ever since their discovery and several modifications in original technologies have further brought precision and accuracy in these methods. In this review, we present an overview of GE using SDNs with an emphasis on CRISPR/Cas system, their advantages, limitations and also practical considerations while designing experiments have been discussed. The review also emphasizes on the possible applications of CRISPR for improving economic traits in crop plants.

Abstract:Climate change is one of the burdensome factors of agricultural productivity. Adverse biotic and abiotic stress impacts directly on plants resulting in poor productivity and yield loss. Modern protocols in genome editing using CRISPR, TALENs, ZFNs and Meganucleases enable editing at the precise site. The versatility of genome editing tools makes its application useful in fields like crop improvement, gene functional analyses, pathway research, studying animal models, genetic analyses, epigenetic research, drug development and biofuels research etc. The technological advancements in genome editing are adopted in both monocots and dicots for crop improvement. These technologies are precise, independent of breeding limitations like unexplored germplasm resources and reduce breeding cycles drastically from decades to years to meet the global requirements. The application of genome editing in agriculture proves to be a potential source for developing crops with biotic and abiotic stress, improved yield with better nutritional qualities.

Abstract:Sustainable intensification of agriculture in Africa is essential for accomplishing food and nutritional security and addressing the rising concerns of climate change. There is an urgent need to close the yield gap in staple crops and enhance food production to feed the growing population. In order to meet the increasing demand for food, more efficient approaches to produce food are needed. All the tools available in the toolbox, including modern biotechnology and traditional, need to be applied for crop improvement. The full potential of new breeding tools such as genome editing needs to be exploited in addition to conventional technologies. Clustered regularly interspaced short palindromic repeats/CRISPR-associated protein (CRISPR/Cas)-based genome editing has rapidly become the most prevalent genetic engineering approach for developing improved crop varieties because of its simplicity, efficiency, specificity, and easy to use. Genome editing improves crop variety by modifying its endogenous genome free of any foreign gene. Hence, genome-edited crops with no foreign gene integration are not regulated as genetically modified organisms (GMOs) in several countries. Researchers are using CRISPR/Cas-based genome editing for improving African staple crops for biotic and abiotic stress resistance and improved nutritional quality. Many products, such as disease-resistant banana, maize resistant to lethal necrosis, and sorghum resistant to the parasitic plant Striga and enhanced quality, are under development for African farmers. There is a need for creating an enabling environment in Africa with science-based regulatory guidelines for the release and adoption of the products developed using CRISPR/Cas9-mediated genome editing. Some progress has been made in this regard. Nigeria and Kenya have recently published the national biosafety guidelines for the regulation of gene editing. This article summarizes recent advances in developments of tools, potential applications of genome editing for improving staple crops, and regulatory policies in Africa.

Abstract:Conventional breeding approaches have played a significant role in meeting the food demand remarkably well until now. However, the increasing population, yield plateaus in certain crops, and limited recombination necessitate using genomic resources for genomics-assisted crop improvement programs. As a result of advancements in the next-generation sequence technology, GABs have developed dramatically to characterize allelic variants and facilitate their rapid and efficient incorporation in crop improvement programs. Genomics-assisted breeding (GAB) has played an important role in harnessing the potential of modern genomic tools, exploiting allelic variation from genetic resources and developing cultivars over the past decade. The availability of pangenomes for major crops has been a significant development, albeit with varying degrees of completeness. Even though adopting these technologies is essentially determined on economic grounds and cost-effective assays, which create a wealth of information that can be successfully used to exploit the latent potential of crops. GAB has been instrumental in harnessing the potential of modern genomic resources and exploiting allelic variation for genetic enhancement and cultivar development. GAB strategies will be indispensable for designing future crops and are expected to play a crucial role in breeding climate-smart crop cultivars with higher nutritional value.

Abstract:Genome-editing technologies have revolutionized research in plant biology, with major implications for agriculture and worldwide food security, particularly in the face of challenges such as climate change and increasing human populations. Among these technologies, clustered regularly interspaced short palindromic repeats [CRISPR]–CRISPR-associated protein [Cas] systems are now widely used for editing crop plant genomes. In this review, we provide an overview of CRISPR–Cas technology and its most significant applications for improving crop sustainability. We also review current and potential technological advances that will aid in the future breeding of crops to enhance food security worldwide. Finally, we discuss the obstacles and challenges that must be overcome to realize the maximum potential of genome-editing technologies for future crop and food production.

Abstract:Genome editing can transform agriculture and shape the future of food by improving crop yields and animal productivity, which in turn can help to achieve food security for the growing world population. CRISPR–Cas-based technologies are powerful gene editing tools that are applied to various food products. In this Review, we discuss the applications of CRISPR–Cas aimed at increasing the nutritional value of crops through macronutrient engineering and biofortification or the reduction of the amount of antinutrients. We examine the role of CRISPR–Cas in improving the flavour of crops and reducing post-harvest losses to increase consumer acceptance and decrease food waste. We also highlight the gene editing of animal food products and probiotics. We summarize the regulations for approval of gene-edited foods worldwide and the progressively evolving public view. Finally, we explore the strategies that can help to enhance the efficiency of genome editing techniques and the acceptance of genome-edited foods in the global market, and extend the technology to low-resource settings.

Abstract:Genome editing (GE) has opened up new vistas for precise trait manipulation in different crops. The other main attractions of GE has been the production of transgene free trait manipulated plants even though the GE involves a genetic transformation step to introduce the GE machinery into the plant cells. This enables better social acceptability of the genome edited plants and already in many countries including India, the edited plants with SDN1 (site directed nuclease – class I) strategy have been exempted from the purview of regulations by statutory bodies (such as GEAC in india) that monitor and regulate the release of genetically modified organisms. Among the several tools of GE that have been discovered and harnessed, CRISPR/Cas9 has been the most efficient and widely used. CRISPR/Cas9 mediated GE has been demonstrated and adopted in oilseed crops already. However, there are limitations and bottle necks such as amenability of the crop for in vitro genetic manipulation and the availability of functionally validated gene models for trait manipulation, which are limiting application of GE widely in oilseed crops. We discuss here the prospects of GE in oilseed crops.

Abstract:Presently, the field of plant breeding is in the genomics era, where innovative techniques are being integrated to accelerate and enhance the efficiency of breeding. Traditional plant breeding methods rely on maintaining plant germplasm with desirable agronomic traits from distinct plants produced through crosses or mutagenesis. However, advancements in genetic engineering encompass all forms of genetic modification through recombinant DNA technology (RDT) and cell fusion mechanisms. These approaches shed light on areas involving mutant organisms, DNA replication, genetic linkage resolution, genetically modified organisms (GMOs), protein sequencing, functional genomics, and computational genomics alterations in genetic engineering. The integration of structural genomics into breeding and eugenics analysis has resulted in a vast knowledge base on crop genetics, species divergence, and molecular origin of traits, as well as the evolutionary history of crop lineage from ancient ancestral species. The genomic data and advancements have proven essential in identifying rare genes, alleles, or local lesions crucial to significant agronomic traits, thereby expediting breeding cycles. This article aims to explore the potential of emerging genetic engineering technologies, including synthetic biology and genome editing, to further advance crop genetics and plant breeding.

Abstract:Genome-editing tools including meganucleases, zinc finger nucleases, transcription activator-like effector nucleases and clustered regularly interspaced short palindromic repeats (CRISPR) system have been applied to improve the quality of staple, oilseed, and horticultural crops with great accuracy and efficiency compared to conventional breeding. In particular, the CRISPR method has proven to be a feasible, cost-effective and versatile tool allowing precise and efficient editing of plant genomes in recent years, showing great potential in crop improvement. Until now, various genome-edited crops with enhanced commercial value have been developed by not only global companies but also small laboratories in universities, suggesting low entry barriers with respect to manpower and capital. In this study, we review the current applications of genome editing technologies to improve the nutritional and functional quality and preferred traits of various crops. Combining this rapidly advancing genome-editing technology and conventional breeding will greatly extend the potential of genome-edited crops and their commercialization.

Abstract:Cereal crops like rice, maize, barley, and wheat are the major staple food crops, and their nutrition and processing quality requires a dedicated long-term breeding program. However, genome editing (GE) could be a potential approach for the targeted improvement of cultivated crop plants via specifically selected alternation in their genome. Various GE approaches have been previously used, which make use of zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs), and clustered regularly interspaced short identical and inverted repeats CRISPR/Cas9 (CRISPR-associated protein 9). Genome modification methods have been implemented for the desired trait improvement in numerous crops as well in some gymnosperms and angiosperm. CRISPR/Cas9-based methods have provided a great capacity to genetic engineering due to their competency, simplicity, and the ability to adopt in comparison to other GE techniques. These methods have provided capacity to gain an exact and deeper understanding of GE. But in plant research, crop system specifically aims at genetic improvement by targeting desired biosynthesis pathways. This urges to develop new technologies for specific mutagenesis and accurate genome editing of the key genes involved in different biosynthetic pathways, and targeted control of their gene expression at transcriptional or post-transcriptional level.
