Abstract:Small intestinal submucosa (SIS) membranes as a decellularized tissue are known to be a natural nanofibrous biomaterial mainly made of type I collagen fibers and containing some growth factors (fibroblast growth factor 2 and transforming growth factor beta) desired in tissue engineering. Here we show that the SIS membranes can promote the formation of bone mineral hydroxylapatite (HAP) crystals along the collagen fibers constituting the membranes from a HAP-supersaturated solution. The resultant biomineralized HAP-SIS scaffolds were found to promote the attachment, growth and osteogenic differentiation of mesenchymal stem cells (MSCs) in both basal and osteogenic media by the evaluation of osteogenic marker formation. More importantly, the HAP-SIS scaffolds could induce the osteogenic differentiation in the basal media without osteogenic supplements due to the presence of HAP crystals in the scaffolds. Histological characterization of the MSC-seeded scaffolds showed that HAP-SIS scaffolds are biocompatible and promote the formation of new tissue in vitro. The biomineralized SIS membranes mimic some aspects of natural bone in terms of the composition and nanostructures and can find potential use in bone tissue engineering.

Abstract:Knowledge-based systems that have made the computer-aided design (CAD) of analog circuits feasible are discussed. The three systems-Idac, Oasys, and Opasyn-were formally announced in 1987. Although they differ widely in philosophy, all use common building blocks and produce sized-schematic diagrams showing how transistors, capacitors, and so forth are connected, complete with the components' values-from which custom circuits can be synthesized. These building blocks, however, unlike the ones used in semicustom analog IC design, are not fixed designs from a library. Rather, they can be varied infinitely, according to rules given to the tools by human experts, so that they approach the ideal performance far more closely than is possible with a limited choice of fixed blocks. All told, Idac, Oasys, and Opasyn can automatically synthesize analog circuits from 13 classes of analog building blocks and can produce over 100 distinct circuit topologies. The characteristics of the three systems are discussed and compared.<<ETX>>
A 3D-Printed Polycaprolactone/Marine Collagen Scaffold Reinforced with Carbonated Hydroxyapatite from Fish Bones for Bone Regeneration

Abstract:In bone tissue regeneration, extracellular matrix (ECM) and bioceramics are important factors, because of their osteogenic potential and cell-matrix interactions. Surface modifications with hydrophilic material including proteins show significant potential in tissue engineering applications, because scaffolds are generally fabricated using synthetic polymers and bioceramics. In the present study, carbonated hydroxyapatite (CHA) and marine atelocollagen (MC) were extracted from the bones and skins, respectively, of Paralichthys olivaceus. The extracted CHA was characterized using Fourier transform infrared (FTIR) spectroscopy and X-ray diffraction (XRD) analysis, while MC was characterized using FTIR spectroscopy and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The scaffolds consisting of polycaprolactone (PCL), and different compositions of CHA (2.5%, 5%, and 10%) were fabricated using a three-axis plotting system and coated with 2% MC. Then, the MC3T3-E1 cells were seeded on the scaffolds to evaluate the osteogenic differentiation in vitro, and in vivo calvarial implantation of the scaffolds was performed to study bone tissue regeneration. The results of mineralization confirmed that the MC/PCL, 2.5% CHA/MC/PCL, 5% CHA/MC/PCL, and 10% CHA/MC/PCL scaffolds increased osteogenic differentiation by 302%, 858%, 970%, and 1044%, respectively, compared with pure PCL scaffolds. Consequently, these results suggest that CHA and MC obtained from byproducts of P. olivaceus are superior alternatives for land animal-derived substances.
Preparation and characterization of two novel osteoinductive fishbone-derived biphasic calcium phosphate bone graft substitutes

Abstract:Journal of Biomaterials Applications, Ahead of Print. Many studies have reported on the conversion of natural resources into xenografts with hydroxyapatite (HA) as major component, but the extraction of biphasic calcium phosphate (HA/β-TCP) from animal bones and transformation into bone graft substitutes are rarely reported. In this research, two kinds of fish bones were made into granular porous biphasic calcium phosphate bone graft substitutes with particle sizes between 500 to 1000 μm through a series of preparation procedures (Salmo salar calcined at 900°C named Sa900 and Anoplopoma fimbria calcined at 800°C named An800). The chemical composition was characterized by X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR). The morphology and porous structure of the scaffolds were comparatively analyzed by scanning electron microscopy (SEM) and mercury porosimeter. The specific surface area of materials was measured by the nitrogen adsorption technique based on BET theory. Cytotoxicity and ectopic osteogenesis were also carried out to investigate the biocompatibility and osteoinductive potential of these materials. The results showed that both fishbone-derived scaffolds were composed of HA and β-TCP with different proportions, and numerous interconnected pores with different sizes were observed at the surface of materials. An800 had higher total porosity reaching 74.8% with higher interconnectivity and micropores mostly distributed at 0.27 μm and 0.12 μm, while Sa900 had a higher specific surface area and higher intraparticle porosity with nanopores mostly distributed at 0.07 μm. CCK-8 assays and Live/dead staining demonstrated excellent biocompatibility. Material-induced osteoid formation were observed on the interface of both internal pores and periphery of materials after implantation in muscle pouch of Wistar rats for 8 weeks which indicated some extent of osteoinductive potential of materials. The possible mechanism of material-induced osteogenesis and the effects of chemical composition, surface topography, and spatial structure on osteogenesis were also discussed in this paper.

Abstract:The high prevalence of bone defects has become a worldwide problem. Despite the significant amount of research on the subject, the available therapeutic solutions lack efficiency. Autografts, the most commonly used approaches to treat bone defects, have limitations such as donor site morbidity, pain and lack of donor site. Marine resources emerge as an attractive alternative to extract bioactive compounds for further use in bone tissue-engineering approaches. On one hand they can be isolated from by-products, at low cost, creating value from products that are considered waste for the fish transformation industry. One the other hand, religious constraints will be avoided. We isolated two marine origin materials, collagen from shark skin (Prionace glauca) and calcium phosphates from the teeth of two different shark species (Prionace glauca and Isurus oxyrinchus), and further proposed to mix them to produce 3D composite structures for hard tissue applications. Two crosslinking agents, 1-[3-(dimethylamino)propyl]-3-ethylcarbodiimide hydrochloride/N-Hydroxysuccinimide (EDC/NHS) and hexamethylene diisocyanate (HMDI), were tested to enhance the scaffolds’ properties, with EDC/NHS resulting in better properties. The characterization of the structures showed that the developed composites could support attachment and proliferation of osteoblast-like cells. A promising scaffold for the engineering of bone tissue is thus proposed, based on a strategy of marine by-products valorisation.

Abstract:Critical-sized bone defects remain challenging in the clinical setting. Autologous bone grafting remains preferred by clinicians. However, the use of autologous tissue is associated with donor-site morbidity and limited accessibility to the graft tissue. Advances in the development of synthetic bone substitutes focus on improving their osteoinductive properties. Whereas osteoinductivity has been demonstrated with ceramics, it is still a challenge in case of polymeric composites. One of the approaches to improve the regenerative properties of biomaterials, without changing their synthetic character, is the addition of inorganic ions with known osteogenic and angiogenic properties. We have previously reported that the use of a bioactive composite with high ceramic content composed of poly(ethyleneoxide terephthalate)/poly(butylene terephthalate) (1000PEOT70PBT30, PolyActive, PA) and 50% beta-tricalcium phosphate (β-TCP) with the addition of zinc in a form of a coating of the TCP particles can enhance the osteogenic differentiation of human mesenchymal stromal cells (hMSCs) (3). To further support the regenerative properties of these scaffolds, inorganic ions with known angiogenic properties, copper or cobalt, were added to the coating solution. β-TCP particles were immersed in a zinc and copper or zinc and cobalt solution with a concentration of 15 or 45 mM. 3D porous scaffolds composed of 1000PEOT70PBT30 and pure or coated β-TCP were additively manufactured by 3D fibre deposition. The osteogenic and angiogenic properties of the fabricated scaffolds were tested in vitro through culture with hMSCs and human umbilical vein endothelial cells, respectively. The materials were further evaluated through ectopic implantation in an in vivo mini-pig model. The early expression of relevant osteogenic gene markers (collagen-1, osteocalcin) of hMSCs was upregulated in the presence of lower concentration of inorganic ions. Further analysis will focus on the evaluation of ectopic bone formation and vascularisation of these scaffolds after implantation in a mini-pig ectopic intramuscular model.

Abstract:With demand for alternatives to autograft and allograft materials continuing to rise, development of new scaffolds for bone tissue repair and regeneration remains of significant interest. Engineered collagen‐calcium phosphate (CaP) constructs can offer desirable attributes, including absence of foreign body response and possession of inherent osteogenic potential. Despite their promise, current collagen‐CaP constructs are limited to nonload‐bearing applications. In this article, we describe a process for creating decellularized sheets of highly aligned, natively cross‐linked, and mineralized collagen fibrils, which may be useful for developing multilaminate collagen‐CaP constructs with improved mechanical properties. Decellularized bovine tendons were cryosectioned to produce thin sheets of aligned collagen fibrils. Mineralization of the sheets was then performed using an alternate soaking method incorporating a polymer‐induced liquid precursor (PILP) process to promote intrafibrillar mineralization, along with incorporation of physiologically relevant amounts of citrate, Mg, and carbonate. Characteristics of the produced scaffolds were assessed using energy‐dispersive X‐ray spectroscopy (EDX), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Scaffolds were also compared with both native bovine cortical bone and pure hydroxyapatite using X‐ray powder diffraction (XRD), and Fourier transform infrared spectroscopy attenuated total reflection (FTIR‐ATR). Structural and chemical analyses show that the scaffold preparation process that we described is successful in creating mineralized collagen sheets, possessing a mineral phase similar to that found in bone as well as a close association between collagen fibrils and mineral plates.

Abstract:Fucoidan (Fuc), a natural polysaccharide derived from brown seaweed algae, and gelatin (Gel) were conjugated to form a template for preparation of biomimetic scaffolds for potential applications in bone tissue regeneration. To the Fuc–Gel we then incorporated the peptide sequence MTNYDEAAMAIASLN (MTN) derived from the E-F hand domain, known for its calcium binding properties. To mimic the components of the extracellular matrix of bone tissue, the Fuc–Gel–MTN assemblies were incubated in simulated body fluid (SBF) to induce biomineralization, resulting in the formation of β-tricalcium phosphate, and hydroxyapatite (HAp). The formed Fuc–Gel–MTN–beta–TCP/HAP scaffolds were found to display an average Young’s Modulus value of 0.32 GPa (n = 5) with an average surface roughness of 91 nm. Rheological studies show that the biomineralized scaffold exhibited higher storage and loss modulus compared to the composites formed before biomineralization. Thermal phase changes were studied through DSC and TGA analysis. XRD and EDS analyses indicated a biphasic mixture of β-tricalcium phosphate and hydroxyapatite and the composition of the scaffold. The scaffold promoted cell proliferation, differentiation and displayed actin stress fibers indicating the formation of cell-scaffold matrices in the presence of MT3C3-E1 mouse preosteoblasts. Osteogenesis and mineralization were found to increase with Fuc–Gel–MTN–beta–TCP/HAP scaffolds. Thus, we have developed a novel scaffold for possible applications in bone tissue engineering.

Abstract:To obtain the biomimetic scaffolding materials for bone tissue engineering, poly(lactide-co-glycolide) (PLGA) nanofibrous mesh (NFM) was mineralized in a 5x simulated body fluid (SBF) for different time after it was treated by air plasma for 15min and subsequent collagen coating. The apatite particles were nucleated on the surface of individual nanofibers, gradually grew up, and finally covered the whole NFM surface. The mineral aggregates were mainly composed of tiny hydroxyapatite (HA) nanoparticles, whose content reached a constant value of 54 mu g center dot cm-2 after 9 days. The collagen coating and apatite deposition enhanced the NFM strength pronouncedly too. In vitro cell culture demonstrated that the non- or less mineralized NFMs were more beneficial of cell spreading and proliferation than those highly mineralized NFMs, but the latter ones could strongly promote secretion of alkaline phosphatase (ALP) by osteoblasts after cultured for 14 days. Moreover, the highly mineralized NFMs also could significantly up-regulated ALP activity and calcium synthesis of bone marrow mesenchymal stem cells (BMSCs), demonstrating that these NFMs are more favorable of the osteoblast phenotype expression and osteogenic induction. Therefore, the biomimetic apatite deposited PLGA/collagen NFM could be a promising candidate scaffold for bone tissue engineering.

Abstract:It is of great significance to develop osteoinductive artificial scaffold for bone repair and regeneration. We constructed a biomimetic apatite interface on electmspun polycaprolactone fibers by combining layer-by-layer (LbL) nanocoating with mineralization to fabricate an osteoinductive artificial scaffold. After polydopamine modification, cationic type-1 collagen and anionic chondroitin sulfate were sequentially adsorbed on the fiber surface. The fibers coated with the multilayer components served as the precursor matrix to induce apatite deposition. By adjusting the number of the layers and duration of mineralization, the nanoscale morphology of composite fibers was optimized. When ten bilayers of the collagen and chondmitin sulfate were deposited onto the fibers followed by one day-mineralization, the obtained polycaprolactone-apatite composite scaffolds significantly promoted the adhesion, proliferation, and osteogenic differentiation of MC3T3-E1 cells. In a subcutaneous implantation in mice, this composite fiber membrane enhanced in vivo ectopic osteogenesis. Our nanoarchitectural scaffolds were able to mimic the composition and structure of the bone matrix to a certain extent, holding great potential for bone repair and regeneration.

Abstract:Objective To explore the feasibility of the preparation of a new scaffold material by mineralizing with calcium phosphate.Methods The collagen sponges were mineralized with calcium acetate and sodium phosphate reagents by using precipitation method and microwave method respectively.The surface characteristics of surface-modified collagen sponges were analyzed by scanning electron microscopy ( SEM),X-ray diffractometer (XRD) and fourier transform infrared spectrometer (FT-IR).A pilot was made with cranial defect model.Then 6 mm diameter defects were created.The histomorphometric surface-modified collagen sponges were compared with collagen sponges and Bio-Oss after 4,8,12 weeks.Results SEM scanning indicated that collagen sponges were mostly coated with calcium phosphate by the precipitation method and the microwave method.Carbonate apatite existed.Microwave method was better than precipitation method.The histological observations showed calcified newly formed bone surrounding surface-modified collagen sponges.Massive osteocytes existed.Conclusions The collagen sponges mineralized by calcium phosphate has better properties of bone formation.Surface-modified collagen sponge as a scaffold material is a promising biomaterial.

Abstract:Bone loss due to trauma and tumors remains a serious clinical concern. Due to limited availability and disease transmission risk with autografts and allografts, calcium phosphate bone fillers and growth factor-based substitute bone grafts are currently used in the clinic. However, substitute grafts lack bone regeneration potential when used without growth factors. When used along with the added growth factors, they lead to unwanted side effects such as uncontrolled bone growth. Collagen-based hydrogel grafts available on the market fail to provide structural guidance to native cells due to high water-solubility and faster degradation. To overcome these limitations, we employed bioinspired material design and fabricated three different hydrogels with structural features similar to native collagen at multiple length-scales. These hydrogels fabricated using polyionic complexation of oppositely charged natural polysaccharides exhibited multi-scale architecture mimicking nanoscale banding pattern, and microscale fibrous structure of native collagen. All three hydrogels promoted biomimetic apatite-like mineral deposition in vitro elucidating crystalline structure on the surface while amorphous calcium phosphate inside the hydrogels resulting in mineral-hydrogel nanocomposites. When evaluated in a non-load bearing critical size mouse calvaria defect model, chitosan - kappa carrageenan mineral-hydrogel nanocomposites enhanced bone regeneration without added growth factors compared to empty defect as well as widely used marketed collagen scaffolds. Histological assessment of the regenerated bone revealed improved healing and tissue remodeling with mineral-hydrogel nanocomposites. Overall, these collagen-inspired mineral-hydrogel nanocomposites showed multi-scale hierarchical structure and can potentially serve as promising bioactive hydrogel to promote bone regeneration.Statement of SignificanceHydrogels, especially collagen, are widely used in bone tissue engineering. Collagen fibrils play arguably the most important role during natural bone development. Its multi-scale hierarchical structure to form fibers from fibrils and electrostatic charges enable mineral sequestration, nucleation, and growth. However, bulk collagen hydrogels exhibit limited bone regeneration and are mostly used as carriers for highly potent growth factors such as bone morphogenic protein-2, which increase the risk of uncontrolled bone growth. Thus, there is an unmet clinical need for a collagen-inspired biomaterial that can recreate structural hierarchy, mineral sequestration ability, and stimulate recruitment of host progenitor cells to facilitate bone regeneration. Here, we propose collagen-inspired bioactive mineral-hydrogel nanocomposites as a growth factor-free approach to guide and enhance bone regeneration.

Abstract:Composite biomaterial scaffolds consisting of natural polymers and bioceramics may offer an alternative to autologous grafts for applications such as bone repair. Herein, we sought to investigate the possibility of incorporating marine coral microparticles into a collagen-based scaffold, a process which we hypothesised would enhance the mechanical properties of the scaffold as well its capacity to promote osteogenesis of human mesenchymal stromal cells. Cryomilling and sieving were utilised to achieve coral microparticles of mean diameters 14 μm and 64 μm which were separately incorporated into collagen-based slurries and freeze-dried to form porous scaffolds. X-ray diffraction and Fourier transform infrared spectroscopy determined the coral microparticles to be comprised of calcium carbonate whereas collagen/coral composite scaffolds were shown to have a crystalline calcium ethanoate structure. Crosslinked collagen/coral scaffolds demonstrated enhanced compressive properties when compared to collagen only scaffolds and also promoted more robust osteogenic differentiation of mesenchymal stromal cells, as indicated by increased expression of bone morphogenetic protein 2 at the gene level, and enhanced alkaline phosphatase activity and calcium accumulation at the protein level. Only subtle differences were observed when comparing the effect of coral microparticles of different sizes, with improved osteogenesis occurring as a result of calcium ion signalling delivered from collagen/coral composite scaffolds. These scaffolds, fabricated from entirely natural sources, therefore show promise as novel biomaterials for tissue engineering applications such as bone regeneration.

Objective:To describe the epidemiology of AIDS-associated KS (AIDS-KS) in Europe and to determine whether it occurs with increased frequency in southern Europe. Methods:Analysis of the ‘European non-aggregate AIDS data set’, as of September 1995. Countries with a cumulative total of ≥ 50 KS cases as the presenting manifestation of AIDS were included. Homosexual men were excluded from south versus non-south comparisons because of possible confounding effects due to their route of HIV transmission. Results:KS was the presenting manifestation of AIDS for 13.3% (16 367 out of 122 679) of men and 2% (491 out of 24 826) of women. In all countries, the risk for KS was higher in individuals who acquired HIV infection via sexual rather than parenteral transmission. Among AIDS patients, there is little difference by sex in the risk of KS in injecting drug users (IDU) or transfusion recipients. The percentage with KS increased with age among homosexual and bisexual men, from 10% in the age group 15–19 years to 23% in the age group 30–39 years. In all countries, the percentage with KS declined over time. The risk of KS was not significantly higher in southern Europe. The percentage with KS in southern Europe was slightly lower than in northern Europe (P > 0.1) in male IDU (1.8% versus 2.1%), and only slightly higher (P > 0.1) in female IDU (1.5% versus 1.1%), in male transfusion recipients (3.5% versus 3.0%), in female transfusion recipients (2.4% versus 2.3%), and in both heterosexual men (7.5% versus 6.2%) and women (2.0% versus 1.6%) excluding those originating from countries where heterosexual HIV transmission is frequent. Conclusions:The strong geographic predilection described for classical KS in southern Europe was not seen for AIDS-KS. If KS is caused by a viral infection in an immunodeficient host, our findings suggest the geographical variations in classical KS are not due to variation in prevalence of the causative virus but may be due to geographical variations in the prevalence of a form of mild immunodeficiency.
Fish Scale Derived Hydroxyapatite Incorporated 3D Printed PLA Scaffold for Bone Tissue Engineering

Abstract:Bone defect repair, particularly in the alveolar region, remains a significant hurdle in periodontics. In recent years, the spotlight in regenerative medicine has fallen on 3D-printed bone scaffolds, especially those constructed of polylactic acid (PLA) infused with hydroxyapatite. This research introduced a novel approach by developing a 3D-printed PLA scaffold enriched with hydroxyapatite derived from fish skin waste (FSHA). Mechanical compression tests revealed that the 3D-printed PLA-FSHA scaffolds had a compressive strength (13.4±5.53 MPa) in the same ballpark as their reference PLA counterparts (20.3±1.08 MPa). Scanning electron micrographs highlighted an average pore size in the scaffold (572 ± 33 μm) conducive to angiogenesis and facilitating cell migration and proliferation. In vitro, cytotoxicity was ascertained using the MTT assay on L929 fibroblast cells. Further in vitro cytocompatibility assessments through actin-DAPI staining and measurements of bone regeneration markers - alkaline phosphatase, osteocalcin and osteopontin-demonstrated that the PLA-FSHA scaffolds not only were biocompatible but also showcased performance on par with the commercial graft, osseograft. This lays the foundation for future in vivo evaluations of bone regenerative capabilities.

Abstract:Simulating the hierarchical structure of natural bone, mineralized synthetic polymer fibrils such as poly(L-lactic acid) (PLLA) in the form of microspheres show considerable potential as injectable scaffolds for bone defect repair. However, the hydrophobicity of PLLA restricts the deposition of bone-like apatite on the surface of PLLA directly, limiting its application in osteogenesis. In this paper, after sodium trimetaphosphate (STMP) modifi-cation, nanofibrous PLLA microspheres were immersed into 2.5 times simulated body fluid (2.5 xSBF) for 3 days to produce abundant bone-like apatite on the surface of microspheres. The bioactivity in vitro and bone repair ability in vivo of surface mineralized PLLA microspheres were evaluated. The results indicated that surface mineralized nanofibrous PLLA microspheres improved MSCs proliferation and bone-related gene expression, including OPN, RUNX-2 and OCN. After the surface mineralized nanofibrous PLLA microspheres were implanted into rat skulls to repair bone defects (5 mm) for 6 weeks, the micro-CT and histology results showed that the surface mineralized nanofibrous PLLA microspheres improved bone regeneration compared to pure nanofibrous PLLA microspheres. Our study provides a feasible strategy for the rapid preparation of biomimetic mineralized PLLA scaffolds, the novel mineralized microspheres showed considerable bone repair potential.

Abstract:Tissue engineering scaffolds have presented effective value in bone repair. However, the integration of the diverse components, complex structures, and multifunction to impart the scaffolds with improved applicability is still a challenge. Here, we propose a novel fish-derived scaffold combined with photothermal therapy and mesenchymal stem cells (MSCs) to promote bone regeneration. The fish-derived scaffold is composed of the decellularized fish scale and gelatin methacrylate synthesized from fish gelatin (fGelMA), which can promote the proliferation and osteogenesis of MSCs with no obvious immunological rejection. Furthermore, the black phosphorus (BP) nanosheets are incorporated into the fGelMA hydrogel network, which can endow the hydrogel with the capacity of photothermal conversion stimulated by near-infrared (NIR) light. The fish-derived scaffold can promote the osteogenesis process of MSCs with higher expression of osteogenic markers and higher mineralization assisted by the NIR light in vitro. The regeneration of mice calvarial defect has also been accelerated by the scaffold with photothermal therapy and MSCs. These results suggest that the fish-derived scaffold, photothermal therapy, and MSCs-based regenerative therapy is a promising clinical strategy in bone regeneration.

Abstract:We present two patients of univentricular physiology, who underwent diaphragmatic plication following the complication of diaphragmatic paralysis resulting from a bidirectional Glenn procedure. Over several months, complex connections developed between aortopulmonary collateral arteries, resulting in large shunts around the plication sites and an increased central pulmonary artery (PA) pressure to 14–15 mmHg. Most blood flow from these connections was reversed in the lower PAs of the affected side, reaching the contralateral lungs through the central PAs. Selective angiography identified almost all of the feeding arteries and complex connections. Aggressive coil embolization at these sites decreased the PA pressure to approximately 10 mmHg, enabling the Fontan procedure. © 2013 Wiley Periodicals, Inc.
