Abstract:Salicylic acid (SA) is an important regulator of plant resistance to biotrophic and hemi-biotrophic pathogens. The enhanced pseudomonas susceptibility 1 (eps1) mutant in Arabidopsis thaliana is hypersusceptible to both virulent and avirulent strains of the bacterial pathogen Pseudomonas syringae. Through positional cloning, the EPS1 gene was isolated and found to encode a novel member of the BAHD acyltransferase superfamily. Pathogen-induced accumulation of SA and expression of pathogenesis-related (PR) genes were compromised in the eps1 mutant. SA could induce PR1 gene expression and restore disease resistance in the eps1 mutant. These results suggest that EPS1 functions upstream of SA and may be involved directly in synthesis of a precursor or a regulatory molecule for SA biosynthesis. Mutations of EPS1 or other genes important for SA accumulation or signaling conferred enhanced resistance to the necrotrophic fungal pathogens Botrytis cinerea and Alternaria brassicicola in the Nossen-0 background but had little effect in the Columbia-0 background. These results suggest that there is natural variation among Arabidopsis ecotypes with respect to the antagonistic cross-talk between defense signaling pathways against various types of microbial pathogens.

Abstract:Endogenous salicylic acid (SA) levels increase and several families of pathogenesis-related genes (including PR-1 and PR-2) are induced during the resistance response of tobacco to tobacco mosaic virus (TMV) infection. We have found that at a temperature (32 degrees C) that prevents the induction of PR genes and resistance, the increases in SA levels were eliminated. However, when the resistance response was restored by shifting inoculated plants to lower temperatures, SA levels increased dramatically and preceded PR-1 gene expression and necrotic lesion formation associated with resistance. SA was also found in a conjugated form whose levels increased in parallel with the free SA levels. This SA beta-glucoside (SAG) was as active as SA in inducing PR-1 gene expression. PR-1 gene induction by SAG was preceded by a transient release of SA. The existence of a mechanism that releases SA from SAG suggests a possible role for SAG in the maintenance of systemic acquired resistance. Previously, we identified a soluble salicylic acid-binding protein (SABP) in tobacco whose properties suggest that it may play a role in transmitting the SA signal during plant defence responses. This SABP has been purified 250-fold by sequential chromatography on DEAE-Sephacel, Sephacryl S-300, Blue Dextran-Agarose and Superose 6. Several monoclonal antibodies (mAbs) raised against the highly purified SABP immunoprecipitated the SA-binding activity and a 280 kDa protein. This 280 kDa protein also co-purified with the SA-binding activity during the various chromatography steps, suggesting that it was responsible for binding SA. Immunoblot analysis with the SABP-specific mAbs also detected the 280 kDa protein in highly purified preparations of SABP. However, in crude homogenates these mAbs only recognized a 57 kDa protein. These and other results suggest that SABP is a multimeric complex which contains, at least, a 57 kDa protein and whose components are readily crosslinked during purification.

Abstract:Salicylic acid (SA) and N -hydroxypipecolic acid (NHP) are two central plant immune signals involved in both resistance at local sites of pathogen infection (basal resistance) and at distal uninfected sites after primary infection (systemic acquired resistance). Major discoveries and advances have led to deeper understanding of their biosynthesis and signaling during plant defense responses. In addition to their well-defined roles in immunity, recent research is emerging on their direct mechanistic impacts on plant growth and development. In this review, we will first provide an overview of how SA and NHP regulate local and systemic immune responses in plants. We will emphasize how these two signals are mutually potentiated and are convergent on multiple aspects—from biosynthesis to homeostasis, and from signaling to gene expression and phenotypic responses. We will then highlight how SA and NHP are emerging to be crucial regulators of the growth-defense balance, showcasing recent multi-faceted studies on their metabolism, receptor signaling and direct growth/development-related host targets. Overall, this article reflects current advances and provides future outlooks on SA/NHP biology and their functional significance as central signals for plant immunity and growth. Because global climate change will increasingly influence plant health and resilience, it is paramount to fundamentally understand how these two tightly linked plant signals are at the nexus of the growth-defense balance.

Abstract:Systemic acquired resistance (SAR) occurs when primary infected leaves produce several SAR-inducing chemical or mobile signals that are transported to uninfected distal parts via apoplastic or symplastic compartments and activate systemic immunity. The transport route of many chemicals associated with SAR is unknown. Recently, it was demonstrated that pathogen-infected cells preferentially transport salicylic acid (SA) through the apoplasts to uninfected areas. The pH gradient and deprotonation of SA may lead to apoplastic accumulation of SA before it accumulates in the cytosol following pathogen infection. Additionally, SA mobility over a long distance is essential for SAR, and transpiration controls the partitioning of SA into apoplasts and cuticles. On the other hand, glycerol-3-phosphate (G3P) and azelaic acid (AzA) travel via the plasmodesmata (PD) channel in the symplastic route. In this review, we discuss the role of SA as a mobile signal and the regulation of SA transport in SAR.

Abstract:Plants respond to biotic stress by inducing a variety of responses, which not only protect against the immediate diseases but also provide immunity from future infections. One example is systemic acquired resistance (SAR), which provides long-lasting and broad-spectrum protection at the whole plant level. The induction of SAR prepares the plant for a more robust response to subsequent infections from related and unrelated pathogens. SAR involves the rapid generation of signals at the primary site of infection, which are transported to the systemic parts of the plant presumably via the phloem. SAR signal generation and perception requires an intact cuticle, a waxy layer covering all aerial parts of the plant. A chemically diverse set of SAR inducers has already been identified, including hormones (salicylic acid, methyl salicylate), primary/secondary metabolites (nitric oxide, reactive oxygen species, glycerol-3-phosphate, azelaic acid, pipecolic acid, dihyroabetinal), fatty acid/lipid derivatives (18 carbon unsaturated fatty acids, galactolipids), and proteins (DIR1-Defective in Induced Resistance 1, AZI1-Azelaic acid Induced 1). Some of these are demonstrably mobile and the phloem loading routes for three of these SAR inducers is known. Here we discuss the recent findings related to synthesis, transport, and the relationship between these various SAR inducers.

Abstract:Systemic signalling involves a complex network of signal transduction and amplification that leads to the activation of defence genes and establishment of systemic resistance throughout the entire plant. On microbial invasion, pathogen‐associated molecular patterns and effectors are frequently detected and recognised by plant receptors. Localised perception at the infection site rapidly triggers a cascade of early signalling events, including protein phosphorylation and production of reactive oxygen species. Subsequently, secondary signal molecules are synthesised and involved in amplification of defence signalling and the establishment of systemic acquired resistance (SAR). An increasing number of long‐distance signalling intermediates has been identified. Most of these act together and specifically promote systemic defence via the regulation of the local release of a set of systemically mobile signals. In the systemic tissue, the induction of SAR depends on synergistic interactions between systemic and ubiquitous salicylic acid‐associated immune signals. Local resistance to pathogen infection generally results from PAMP (pattern‐associated molecular pattern)‐triggered immunity and microbial effector‐triggered immunity. Microbial infection of local tissues often leads to systemic acquired resistance (SAR) in distal tissues which provides long‐lasting broad‐spectrum resistance to a variety of pathogens. Reactive oxygen species mediate systemic immunity acting as both cell‐to‐cell systemic signals and as local signalling partners upstream of other phloem‐mobile signals. Mobile signals for SAR that are translocated via the vasculature may include methyl salicylate, the C9 lipid peroxidation product azelaic acid, one or more lipid transfer proteins, the diterpene dihydroabietinal and the nonprotein amino acid pipecolic acid. Many of the SAR mobile signals cooperate with each other and with salicylic acid in one or more parallel and synergistic signalling pathways. As a form of priming, SAR is supported by epigenetic regulation of defence gene expression.
A narrowband interference suppression algorithm for time synchronization

Abstract:In this paper, we propose an algorithm to suppress the narrowband interference (NBI) during the time synchronization. By exploring the difference between the time domain autocorrelation matrix of the NBI and that of the desired signal, the NBI is suppressed in the eigen-domain of the NBI. Compared with the traditional frequency domain techniques, the proposed algorithm does not encounter the problem of spectral leakage, thus can yield better performance when the NBI power is high. The probabilities of false alarm and detection are analyzed to illustrate the performance of the proposed algorithm. For the practical implementation, a blind adaptive approach is developed to estimate the inverse of the autocorrelation matrix of the received signal. Simulation results are presented to demonstrate the effectiveness and advantage of the proposed algorithm.
The effect of contact lens cleaners on lens parameters and surface quality of the Novalens.

Abstract:The purpose of this study is to evaluate the effect of one abrasive cleaner, one non-abrasive cleaner and one non-abrasive alcohol-based cleaner on the lens parameter stability of the NOVALENS. Twenty-four lenses, all NOVALENS material of identical design, were cleaned 100 times each. Eight lenses were cleaned with MiraFlow (CIBA Vision), eight lenses with Optic-Free Daily Cleaner and eight lenses with the Gas Permeable Daily Cleaner (PBH). The base curve radius, center thickness and power were verified immediately before, immediately after and (per manufacturer's recommendation) 4 hours after cleaning at baseline, 25, 50, 75 and 100 cleanings. Lens toricity of approximately 0.50D was found with all three cleaners after 100 cleanings. No change in lens power or center thickness was found. It was concluded that extreme care must be taken in handling the NOVALENS due to the potential of induced warpage.

Abstract:Abstract N-hydroxypipecolic acid (NHP) accumulates in the plant foliage in response to a localized microbial attack and induces systemic acquired resistance (SAR) in distant leaf tissue. Previous studies indicated that pathogen inoculation of Arabidopsis (Arabidopsis thaliana) systemically activates SAR-related transcriptional reprogramming and a primed immune status in strict dependence of FLAVIN-DEPENDENT MONOOXYGENASE 1 (FMO1), which mediates the endogenous biosynthesis of NHP. Here, we show that elevations of NHP by exogenous treatment are sufficient to induce a SAR-reminiscent transcriptional response that mobilizes key components of immune surveillance and signal transduction. Exogenous NHP primes Arabidopsis wild-type and NHP-deficient fmo1 plants for a boosted induction of pathogen-triggered defenses, such as the biosynthesis of the stress hormone salicylic acid (SA), accumulation of the phytoalexin camalexin and branched-chain amino acids, as well as expression of defense-related genes. NHP also sensitizes the foliage systemically for enhanced SA-inducible gene expression. NHP-triggered SAR, transcriptional reprogramming, and defense priming are fortified by SA accumulation, and require the function of the transcriptional coregulator NON-EXPRESSOR OF PR GENES1 (NPR1). Our results suggest that NPR1 transduces NHP-activated immune signaling modes with predominantly SA-dependent and minor SA-independent features. They further support the notion that NHP functions as a mobile immune regulator capable of moving independently of active SA signaling between leaves to systemically activate immune responses.

Abstract:As the most widely used herbal medicine in human history and a major defense hormone in plants against a broad spectrum of pathogens and abiotic stresses, salicylic acid (SA) has attracted major research interest. With applications of modern technologies over the past 30 years, studies of the effects of SA on plant growth, development, and defense have revealed many new research frontiers, and continue to deliver surprises. In this review, we provide an update on recent advances in our understanding of SA metabolism, perception, and signal transduction mechanisms in plant immunity. An overarching theme emerges that SA executes its many functions through intricate regulation at multiple steps: SA biosynthesis is regulated both locally and systemically, while its perception occurs through multiple cellular targets, including metabolic enzymes, redox regulators, transcription cofactors, and most recently, an RNA-binding protein. Moreover, SA orchestrates a complex series of posttranslational modifications of downstream signaling components and promotes the formation of biomolecular condensates that function as cellular signaling hubs. SA also impacts wider cellular functions through crosstalk with other plant hormones. Looking into the future, we propose new areas for exploration on SA functions, which will undoubtedly uncover more surprises for many years to come.

Abstract:Salicylic acid (SA) is an important phytohormone, well‐known for its regulatory role in shaping plant immune responses. In recent years, significant progress has been made in unravelling the molecular mechanisms underlying SA biosynthesis, perception, and downstream signalling cascades. Through the concerted efforts employing genetic, biochemical, and omics approaches, our understanding of SA‐mediated defence responses has undergone remarkable expansion. In general, following SA biosynthesis through Avr effectors of the pathogens, newly synthesized SA undergoes various biochemical changes to achieve its active/inactive forms (e.g. methyl salicylate). The activated SA subsequently triggers signalling pathways associated with the perception of pathogen‐derived signals, expression of defence genes, and induction of systemic acquired resistance (SAR) to tailor the intricate regulatory networks that coordinate plant immune responses. Nonetheless, the mechanistic understanding of SA‐mediated plant immune regulation is currently limited because of its crosstalk with other signalling networks, which makes understanding this hormone signalling more challenging. This comprehensive review aims to provide an integrated overview of SA‐mediated plant immunity, deriving current knowledge from diverse research outcomes. Through the integration of case studies, experimental evidence, and emerging trends, this review offers insights into the regulatory mechanisms governing SA‐mediated immunity and signalling. Additionally, this review discusses the potential applications of SA‐mediated defence strategies in crop improvement, disease management, and sustainable agricultural practices.

Abstract:The small phenolic compound salicylic acid (SA) plays an important regulatory role in multiple physiological processes including plant immune response. Significant progress has been made during the past two decades in understanding the SA-mediated defense signaling network. Characterization of a number of genes functioning in SA biosynthesis, conjugation, accumulation, signaling, and crosstalk with other hormones such as jasmonic acid, ethylene, abscisic acid, auxin, gibberellic acid, cytokinin, brassinosteroid, and peptide hormones has sketched the finely tuned immune response network. Full understanding of the mechanism of plant immunity will need to take advantage of fast developing genomics tools and bioinformatics techniques. However, elucidating genetic components involved in these pathways by conventional genetics, biochemistry, and molecular biology approaches will continue to be a major task of the community. High-throughput method for SA quantification holds the potential for isolating additional mutants related to SA-mediated defense signaling.

Abstract:To elude or minimize the effects of disease and herbivory, plants rely on both constitutive and inducible defenses. In response to attack by pathogens or pests, plants activate signaling cascades leading to the accumulation of endogenous hormones that trigger the induction of defenses. Salicylic acid (SA), jasmonic acid (JA), and ethylene (E) are plant-specific hormones involved in communicating the attack by many pathogens and pests in a broad range of plant species. SA, JA and E signaling cascades do not activate defenses independently, but rather establish complex interactions that determine the response mounted in each condition. Deployment of defenses is energetically costly, so a trade-off between the activation of resistance against a particular pest or pathogen and down regulation of other defenses is common. Conversely, activation of broad range resistance in response to an initial attack may serve to deter opportunistic agents. Thus, the interaction among SA, JA and E defense signaling pathways can be antagonistic, cooperative or synergistic, depending on the plant species, the combination of organisms attacking the plants, and the developmental and physiological state of the plant. A characterization of the interactions among defense signaling pathways and the determination of the molecular components mediating cross-talk between the different pathways will be essential for the rational design of transgenic plants with increased resistance to disease and/or herbivores without critically compromising other agronomic traits.

Abstract:In the absence of adaptive immunity displayed by animals, plants respond locally to biotic challenge via inducible basal defense networks activated through recognition and response to conserved pathogen-associated molecular patterns. In addition, immunity can be induced in tissues remote from infection sites by systemic acquired resistance (SAR), initiated after gene-for-gene recognition between plant resistance proteins and microbial effectors. The nature of the mobile signal and remotely activated networks responsible for establishing SAR remain unclear. Salicylic acid (SA) participates in the local and systemic response, but SAR does not require long-distance translocation of SA. Here, we show that, despite the absence of pathogen-associated molecular pattern contact, systemically responding leaves rapidly activate a SAR transcriptional signature with strong similarity to local basal defense. We present several lines of evidence that suggest jasmonates are central to systemic defense, possibly acting as the initiating signal for classic SAR. Jasmonic acid (JA), but not SA, rapidly accumulates in phloem exudates of leaves challenged with an avirulent strain of Pseudomonas syringae . In systemically responding leaves, transcripts associated with jasmonate biosynthesis are up-regulated within 4 h, and JA increases transiently. SAR can be mimicked by foliar JA application and is abrogated in mutants impaired in jasmonate synthesis or response. We conclude that jasmonate signaling appears to mediate long-distance information transmission. Moreover, the systemic transcriptional response shares extraordinary overlap with local herbivory and wounding responses, indicating that jasmonates may be pivotal to an evolutionarily conserved signaling network that decodes multiple abiotic and biotic stress signals.

Abstract:N-hydroxy pipecolic acid (NHP) plays an important role in plant immunity. In contrast to its biosynthesis, our current knowledge with respect to the transcriptional regulation of the NHP pathway is limited. This study commences with the engineering of Arabidopsis plants that constitutively produce high NHP levels and display enhanced immunity. Label-free proteomics reveals a NAC-type transcription factor (NAC90) that is strongly induced in these plants. We find that NAC90 is a target gene of SAR DEFICIENT 1 (SARD1) and induced by pathogen, salicylic acid (SA), and NHP. NAC90 knockout mutants exhibit constitutive immune activation, earlier senescence, higher levels of NHP and SA, as well as increased expression of NHP and SA biosynthetic genes. In contrast, NAC90 overexpression lines are compromised in disease resistance and accumulated reduced levels of NHP and SA. NAC90 could interact with NAC61 and NAC36 which are also induced by pathogen, SA, and NHP. We next discover that this protein triad directly represses expression of the NHP and SA biosynthetic genes AGD2-LIKE DEFENSE RESPONSE PROTEIN 1 (ALD1), FLAVIN MONOOXYGENASE 1 (FMO1), and ISOCHORISMATE SYNTHASE 1 (ICS1). Constitutive immune response in nac90 is abolished once blocking NHP biosynthesis in the fmo1 background, signifying that NAC90 negative regulation of immunity is mediated via NHP biosynthesis. Our findings expand the currently documented NHP regulatory network suggesting a model that together with NHP glycosylation, NAC repressors take part in a 'gas-and-brake' transcriptional mechanism to control NHP production and the plant growth and defense trade-off.

Abstract:Salicylic acid (SA) is an important plant hormone that is best known for mediating host responses upon pathogen infection. Its role in plant defense activation is well established, but its biosynthesis in plants is not fully understood. SA is considered to be derived from two possible pathways; the ICS and PAL pathway, both starting from chorismate. The importance of both pathways for biosynthesis differs between plant species, rendering it hard to make generalizations about SA production that cover the entire plant kingdom. Yet, understanding SA biosynthesis is important to gain insight into how plant pathogen responses function and how pathogens can interfere with them. In this review, we have taken a closer look at how SA is synthesized and the importance of both biosynthesis pathways in different plant species.

Abstract:Plants produce diverse small molecules rapidly in response to localized pathogenic attack. Some of the molecules are able to migrate systemically as mobile signals, leading to the immune priming that protects the distal tissues against future infections by a broad-spectrum of invaders. Such form of defense is unique in plants and is known as systemic acquired resistance (SAR). There are many small molecules identified so far with important roles in the systemic immune signaling, some may have the potential to act as the mobile systemic signal in SAR establishment. Here, we summarize the recent advances in SAR research, with a focus on the role and mechanisms of different small molecules in systemic immune signaling.

Abstract:Plant peptide signaling is an upcoming topic in many areas of plant research. Our recent findings show that the tyrosine sulfated peptide receptors PSKR1 and PSY1R are not only involved in growth and development but also in plant defense. They modulate salicylate- and jasmonate-dependent defense pathways in an antagonistic manner and this phenomenon might be dependent on the age and developmental stage of the plant. Here we discuss how the endogenous peptides might integrate growth, wounding, senescence and the opposing defense pathways against biotrophic and necrotrophic pathogens for increased fitness of the plant.
