Protein Free Spreading Method for Electron Microscopic Studies of DNA Compaction Stimulated by Interactions with Histone H 1 in Surface Films

Abstract:The application of the protein-free spreading method for electron microscopic strudies of DNA-histone H1 interactions stimulating DNA compaction is described. The method is based on the formation of the thin film containing DNA molecules on the surface of the hypophase of histone H1 solution. Histone H1 molecules interact with the spread in the surface film DNA molecules giving rise to mainly intramolecular DNA-histone H1 complexes which are transferred to the electron microscopic grids and studied in EM. This method has many advantages over traditional EM methods of DNA-ligand interactions studies and gives new important information about DNAhistone H1 compaction. The protein free spreading technique was applied to studies of linear, circular DNA and complexes of kinetoplast DNA compaction stimulated by histone H1. The results show that histone H1 stimulates side-by-side association of DNA duplex fiber and the models of DNA folding in the compact structures observed are offered. The protein free spreading technique can be applied to studies of D NA compaction with other types of DNA binding agents.
Sport Specialization and Quality of Life among Middle- and High-School Long-Distance Runners of Different Injury Status: A Retrospective Study

Abstract:Running-related injuries are prevalent in adolescent long-distance runners. The aim of our retrospective study was to compare differences in sport specialization, running habits,quality of life, and sleep habits among middle-and high-school long-distance runners of different injury statuses. Middle- and high-school long-distance runners across the United States were recruited via cross-country coaches and athletic directors between January and May 2020. Participants completed an online survey with questions related to demographics, sport specialization, running habits, quality of life, sleep, and self-reported injury history. Overall, 306 participants completed the survey (male = 107, female = 176, unspecified = 23; age = 15.7 ± 1.1 years). Of the participants, 178 (58.2%) reported no history of injury, 101 (33.0%) reported a previous injury, and 27 (8.8%) reported a current injury. Middle- and high-school runners with a current injury reported significantly lower overall health (p= .01) and average distance per run (p = .05) than uninjured runners. No significant differences were found among injury status and sport specialization level, quality of life, sleep habits, or running volume (p> .05). Runners with a self-reported previous or current injury do not appear to be classified as high-specialized runners more frequently than uninjured runners.

Abstract:This study aimed to characterize running-related injuries (RRIs), explore their relationship with run and resistance training (RT) parameters, and identify perceived prevention measures among adult recreational runners. An anonymous online survey was designed and distributed via social media and email. Data were analyzed with chi-square, t-test, or analysis of variance (ANOVA), with significance accepted at p ≤ 0.05. Data from 616 participants (76.8% female, age: 42.3 ± 10.5 y) were analyzed. Most runners (84.4%) had an injury history, with 44.6% experiencing one in the past year. The most common RRI sites included the foot/ankle (30.9%) and knee (22.2%). RRI prevalence was higher in those running >19 miles weekly (48.4%, p = 0.05), but there were no differences based on RT participation status. Among those using RT, relatively more RRIs were observed in runners who trained the hip musculature (50.3%, p = 0.005) and did not include the upper body (61.6%, p < 0.001). A disproportionately high RRI prevalence was found for several of the other risk-reduction strategies. RRIs remain a substantial problem, particularly around the ankle/foot and knee. Higher run volume and performance motives were positively associated with RRIs. Most runners incorporated RRI risk-reduction techniques, with over half using RT. The current study did not determine whether preventative strategies were implemented before or after injury; therefore, prospective studies controlling for previous injuries are required to evaluate the effectiveness of RT in preventing future RRIs.

Abstract:Background: Training characteristics, such as volume and duration, have been studied in relation to running-related injury (RRI) risk, with mixed findings in adult runners. There is a lack of research assessing how training characteristics relate to RRI in youth runners, despite the high RRI rates observed in this population. Purpose: To prospectively determine associations between (1) total weekly running volume and duration and (2) week-to-week changes in running volume, duration, intensity and training stress with in-season RRI among high school cross-country runners. Methods: Runners completed a preseason demographics and injury history survey and daily surveys regarding training distance, duration, intensity and current RRI. Values were summed weekly and change scores were calculated relative to the prior week. Runners completing ≥75% of daily surveys were analysed; sensitivity analyses for those completing ≥50% and ≥90% were conducted. Generalised estimating equations assessed associations between change in each predictor, including interactions with sex and RRI within the subsequent week, controlling for year in school, prior RRI and repeated observations. Results: 434 runners enrolled in the study; 161 (37%) completed ≥75% of daily surveys. No associations between total volume, total duration or week-to-week change in training characteristics and in-season RRI were observed (p≥0.54). Sensitivity analyses did not detect any significant associations. Conclusion: Total weekly running volume and duration and weekly changes in training were not associated with RRI. RRI are multifactorial and assessing the interaction between training characteristics and other lifestyle factors is likely necessary for determining RRI risk in youth runners.

Abstract:BACKGROUND: The popularity of running continues to increase, which means that the incidence of running-related injuries will probably also continue to increase. Little is known about risk factors for running injuries and whether they are sex-specific.OBJECTIVES: The aim of this study was to review information about risk factors and sex-specific differences for running-induced injuries in adults.SEARCH STRATEGY: The databases PubMed, EMBASE, CINAHL and Psych-INFO were searched for relevant articles.SELECTION CRITERIA: Longitudinal cohort studies with a minimal follow-up of 1 month that investigated the association between risk factors (personal factors, running/training factors and/or health and lifestyle factors) and the occurrence of lower limb injuries in runners were included.DATA COLLECTION AND ANALYSIS: Two reviewers' independently selected relevant articles from those identified by the systematic search and assessed the risk of bias of the included studies. The strength of the evidence was determined using a best-evidence rating system. Sex differences in risk were determined by calculating the sex ratio for risk factors (the risk factor for women divided by the risk factor for men).MAIN RESULTS: Of 400 articles retrieved, 15 longitudinal studies were included, of which 11 were considered high-quality studies and 4 moderate-quality studies. Overall, women were at lower risk than men for sustaining running-related injuries. Strong and moderate evidence was found that a history of previous injury and of having used orthotics/inserts was associated with an increased risk of running injuries. Age, previous sports activity, running on a concrete surface, participating in a marathon, weekly running distance (30-39 miles) and wearing running shoes for 4 to 6 months were associated with a greater risk of injury in women than in men. A history of previous injuries, having a running experience of 0-2 years, restarting running, weekly running distance (20-29 miles) and having a running distance of more than 40 miles per week were associated with a greater risk of running-related injury in men than in women.CONCLUSIONS: Previous injury and use of orthotic/inserts are risk factors for running injuries. There appeared to be differences in the risk profile of men and women, but as few studies presented results for men and women separately, the results should be interpreted with caution. Further research should attempt to minimize methodological bias by paying attention to recall bias for running injuries, follow-up time, and the participation rate of the identified target group.

Abstract:Background Given the high incidence and heavy burden of running related injuries, large-scale, prospective multifactorial investigations examining potential risk factors are warranted. This study aimed to identify factors associated with running related injuries and to evaluate their potential in injury screening. Study design Prospective cohort study. Materials and methods Two hundred and seventy-four recreational runners were recruited. Clinical measures (strength, range of motion, foot position), injury and training history (via questionnaire), impact loading (via accelerometery) and running technique measures were collected at baseline. Runners were tracked for injury for one year via fortnightly check-ins. A binary logistic regression, (injury versus no injury), was performed for each variable univariably, and then adjusting for age, sex and mileage. A multivariable regression was also performed to evaluate the model’s discriminative ability. Results Of the 225 runners included in the final analysis 52% experienced a running related injury. Injury history in the past year, less navicular drop, and measures of running technique (knee, hip, and pelvis kinematics) were associated with increased odds of injury ( p < .05). The multivariable logistic regression model was statistically significant, χ 2 (11) = 56.45, p < .001, correctly classifying 74% of cases with a sensitivity and specificity of 72% and 76%, respectively. The area under the receiver operating characteristic curve was 0.79 (CI 95% = 0.73–0.85), demonstrating acceptable discriminative ability. Conclusions This study found a number of clinical and running technique factors to be associated with prospective running related injuries among recreational runners. With the exception of injury history, the factors identified as being significantly associated with injury may be modifiable and therefore, could form the basis of interventions. Range of motion, spatiotemporal parameters and strength measures were not associated with injury and thus their utilisation in injury prevention practices should be reconsidered.

Abstract:Running is a popular form of exercise that is easily accessible to various populations; endurance running, defined as distances beyond 5 km, continues to grow within the sport. Endurance running-related injuries are common in the lower extremities and are primarily overuse related. A multitude of risk factors for injury exist, including extrinsic factors, such as running distance and frequency, and intrinsic factors, such as biomechanics and nutrition status. Training and rehabilitation techniques vary with a general focus on strengthening and gradual increase in activity, but evidence is mixed, and it is difficult to generalize programs across different running populations. Management of specific running groups, including youth runners, is an area in which additional research is needed. New treatments, such as orthobiologics and wearable technology, have promising potential to optimize performance and recovery and minimize injury. However, they need to be further evaluated with high-quality studies.

Abstract:We report on three epidemiologic studies of orthopedic injuries in exercisers. One group of 438 men and women ran approximately 25 mi per week; 24% reported an injury during the previous year. Higher weekly running mileage and heavier individuals were more likely to report an injury. Injuries were not associated with speed of running, age, gender, type of surface, time of day, or stretching habit. In a second study, rates of injury for the foot, knee, hip, back, shoulder, and elbow were examined in runner (n =2,102) and nonrunner (n =724) patients at a preventive medicine clinic. Only knee injury rates were significantly higher in runners. Third, participants (n=968) in worksite-sponsored exercise programs were evaluated for orthopedic injuries. Rates of exercise injuries were relatively low (net rate = 12%/year in exercisers). Risk of injury in participants starting a walking, running, or jogging program was associated (p <.05) with age, sex, body mass index, flexibility, cardiorespiratory endurance, and number of sit-ups in 1-min.

Abstract:In recent years, the surge in sport and exercise participation, particularly in running, has coincided with the widespread adoption of running-related technology, such as fitness trackers. This study investigates the correlation between the use of running-related technology and running-related injuries among recreational and elite long-distance runners. We conducted a quantitative, cross-sectional online survey of 282 adult runners. Data were analyzed using descriptive statistics and a multivariable logistic regression analysis. Participants, with an average age of 37.4 years, reported varied running experience, with 90.07% utilizing running-related technology during their runs to some degree, primarily smartwatches like Garmin and Apple Watch. Running-related technology users showed a higher likelihood of experiencing running-related injuries compared to non-users (OR = 0.31, p < 0.001). However, those who utilized the metrics obtained from running-related technology to guide their training decisions did not exhibit a higher risk of injury. This nuanced relationship highlights the importance of considering individual training behaviors and the potential psychological impacts of technology on running practices. The study underscores the need for future research integrating biomechanical and psychosocial factors into running-related technology to enhance injury prevention strategies.

Abstract:BackgroundDespite a rapidly growing body of research, a systematic evidence compilation of the risk and protective factors for middle- and long-distance running-related injury (RRI) was lacking.ObjectivesOur objective was to compile the evidence about modifiable and non-modifiable training-related and behavioral risk and protective factors for middle- and long-distance RRI.MethodsWe searched five databases (PubMed, CINAHL, MEDLINE, SPORTDiscus, and PsycINFO) for the dates 1 January 1970 to 31 December 2015, inclusive, for original peer-reviewed articles. The eligible designs were cross-sectional, case–control, longitudinal observational studies, and randomized controlled trials involving runners competing at distances from ≥800 m to ≤42.2 km. Outcomes were any specific and/or general RRI, and exposures included training-related and behavioral factors. We extracted authors and date, study design, injury type(s), descriptors and comparators for each exposure, and results and measures of association from the selected studies. Methodological quality was independently appraised using two separate checklists: a modified checklist for observational study designs and the Physiotherapy Evidence Database (PEDro) scale for randomized controlled trials.ResultsAmong 73 articles eligible for inclusion, 19 (26.0%) and 30 (41.0%) were of high or satisfactory methodological quality, respectively. As a non-modifiable exposure, a history of previous injury was found to be associated with an increased risk of both general and specific RRI. In terms of modifiable exposures, irregular and/or absent menstruation was found to be associated with an increased risk of stress fracture development, whereas the use of oral contraceptives was found to be associated with a decreased risk. High clinical, methodological, and statistical heterogeneity meant it was not feasible to estimate a pooled effect size across similar studies.ConclusionsA history of previous injury was associated with an increased risk of both general and specific RRI. The use of oral contraceptives was found to be associated with a decreased risk of skeletal stress fracture. Conversely, irregular and/or absent menstruation was associated with an increased risk. The varied effect directions and/or a number of statistically insignificant results associated with the majority of factors hindered our ability to draw any definitive conclusions about their relationship to RRI risk.

LOWER QUARTER- AND UPPER QUARTER Y BALANCE TESTS AS PREDICTORS OF RUNNING-RELATED INJURIES IN HIGH SCHOOL CROSS-COUNTRY RUNNERS

Abstract:Background: While cross-country running is a popular interscholastic sport, it also has a high incidence of running-related injuries (RRIs). Recent literature suggests that functional tests may identify athletes at increased risk of injury. The Y-Balance Test (YBT) is an objective measure used to assess functional muscle strength, balance, and expose asymmetries between tested limbs. Purpose/hypothesis: The purpose of this study was to determine if the YBT could predict RRI in high school cross-country runners. It was hypothesized that an asymmetric right (R)/left (L) YBT reach distance for the lower or upper extremities would be associated with an increased risk of RRI. Study design: Prospective observational cohort. Methods: One hundred forty-eight athletes (80 girls, 68 boys) who competed in interscholastic cross-country in Southern California during the 2015 season participated in the study. Prior to the cross-country season, the runners completed Lower-Quarter YBT (LQ-YBT) and Upper-Quarter YBT (UQ-YBT) testing to assess lower and upper extremity asymmetry, respectively. The runners were prospectively monitored for RRI occurrence throughout the season using the Daily Injury Report form. Results: Forty-nine runners (33.1%) incurred a RRI during the 2015 season, with the lower leg (shin/calf) and knee the most common RRI sites. Girls had a higher RRI occurrence (38.8%) than boys (26.5%) (p = 0.12). Boys had greater raw scores for LQ-YBT R and L anterior (ANT), posteromedial (PM), posterolateral (PM) and composite reach distances than girls (p<0.05). With the exception of normalized superolateral reach distance, boys had significantly greater scores for raw and normalized R and L UQ-YBT reach distances and raw composite scores than girls (p<0.05). After adjusting for prior RRI, while boy runners with a LQ-YBT PM reach difference ≥4.0 cm were five times more likely to incur a RRI (Adjusted odds ratio [AOR] = 5.05, 95% CI: 1.3-19.8; p = 0.02), girl runners with a UQ-YBT inferolateral (IL) reach difference ≥ 4.0 cm were 75% less likely to incur a RRI (AOR = 0.25, 95% CI: 0.1-0.7; p = 0.005). By lower extremity body region, boy runners with a UQ-YBT superolateral (SL) reach difference ≥ 4.0 cm were seven times more likely to incur a hip/thigh/knee RRI [AOR] = 7.20, 95% CI: 1.1-45.6; p = 0.002). Conclusion: Greater lower extremity (PM) or upper extremity (SL) reach distance asymmetry, as measured by the LQ-YBT or UQ-YBT, respectively, were associated with RRI in boy high school cross-country runners. Abstract:This study aims to investigate anthropometric and training characteristics associated with injuries in runners using a complex approach. We conducted a cross-sectional study with 97 Brazilian runners of both sexes (age: 39.1 ± 11.5 years; body mass index: 24.2 ± 4.2 kg/m 2 ). A web survey was used for data collection, including age, sex, body weight, body height, training volume/frequency, session duration, running pace and injury information (yes or no; the number of injuries). The partial correlation parameter was used for network estimation, considering both sexes. For women, injury occurrence was positively and strongly associated with training frequency ( r = 0.57) and the number of competitions ( r = 0.64), while for men, a negative and strong association was found with age ( r =-0.69), and positive associations were observed with BMI ( r = 0.89) and training volume ( r = 0.82). Network centrality showed that training session duration and the number of competitions/year presented a higher expected influence value for women (−1.47) and men (−1.31). Using a non-linear approach with Brazilian non-professional runners, training characteristics had a stronger influence on running-related injuries.

Worldwide, reliable comparable data exist and indicate a downward trend in fatal electric accidents. Difficulties were encountered in compiling international statistics because of differences in how accident data were defined and recorded, variations in mandatory practices, lack of suitable data and indices, accident insurance systems, and lack of correlation between technical, financial, and medical aspects.
Impact of COVID-19 Social Distancing Restrictions on Training Habits, Injury, and Care Seeking Behavior in Youth Long-Distance Runners

Abstract:Purpose: The COVID-19 pandemic impacted the sporting and exercise activities of millions of youth. Running is an activity that could be maintained while social distancing restrictions were implemented during the pandemic. If running-related injuries do occur, these restrictions may also influence the access to care or care seeking behavior of this population. Therefore, the purpose of this study was to determine if the social distancing restrictions during the 2020 COVID-19 pandemic influenced training habits, injury, and care seeking behavior in youth long-distance runners. Methods: A customized, open online questionnaire was provided to runners 9-19 years of age who participated in long-distance running activities including team/club cross-country, track and field (distances ≥800 m), road races, or recreational running. Participants responded to questions about demographics, running habits, RRIs, and health care provider visits 6-months before as well as during social distancing restrictions due to COVID-19. Wilcoxon signed rank tests compared differences for ratio data and Chi-square tests were used to compare proportions before and during COVID-19 social distancing restrictions. Statistical significance was set at p ≤ 0.05. Results: A total of 287 youth long-distance runners (male = 124, female = 162, unspecified = 1; age = 15.3 ± 1.7 years; running experience = 5.0 ± 2.3 years) participated. Compared to their pre-COVID-19 responses, youth long-distance runners reported lower distances run per week (p < 0.001), fewer runs per week (p < 0.001), fewer hard runs per week (p < 0.001), fewer number of injuries (p < 0.001), and fewer injuries per 1,000 km (p = 0.002) during the COVID-19 social distancing restrictions. A lower proportion of participants reported in-person health care provider visits (p < 0.001) and a lower proportion of visits were made to an athletic trainer during COVID-19 social distancing restrictions compared to prior to COVID-19 (p < 0.001). Conclusion: The COVID-19 pandemic resulted in significant decreases in both training and injuries which were different compared to previous reports in an adult population. Many of the runners who sustained an injury during COVID-19 social distancing restrictions did not seek care, with the most prominent reduction in visits to an athletic trainer. This could impact future injury and chronic pain.

Main outcome measures: Electronic patient records were analyzed from the AT-PBRN from 59 athletic trainers across 14 states between 2010 and 2022. Summary statistics (frequency, percentages, median, and interquartile ranges [IQR]) were used to describe injury (age at injury, sex, sport, body part, and diagnosis) and treatment characteristics (type of treatment, number of visits, and number of procedures per visit). Results: Football (17.7%, n = 179), basketball (17.6%, n = 178), and soccer (14.9%, n = 151) reported the highest number of injuries. The ankle (17.2%, n = 174), knee (16.5%, n = 167), and head (14.1%, n = 143) were the most common injury locations. Concussion (13.0%, n = 131), ankle strain/sprain (12.4%, n = 125), and thigh/hip/groin sprain/strain (11.1%, n = 112) were the most reported diagnoses. Therapeutic exercise or activities (27.6%, n = 1068), athletic trainer evaluation or re-evaluation (24.7%, n = 957), and hot/cold packs (19.8%, n = 766) were the most common services, with a median of 2 visits (IQR = 1 to 4) and 2 procedures (IQR = 1 to 2) per visit. Conclusions: Football, basketball, and soccer reported the most musculoskeletal injuries for middle school-aged athletes. Concussions were the most frequent diagnosis, followed by ankle sprains/strains. Our findings are similar to prior investigations at the high school and collegiate levels. Treatments at the middle school level were also similar to those that have been previously reported at the high school level with therapeutic exercise/activity, athletic trainer evaluation or re-evaluation, and hot/cold packs being the most common treatments. This information may be useful for informing patient care decisions at the middle-school level.
The Prevention and Nursing Care of Common Injuries in Long-Distance Running of College Students

Abstract:As a favorite sport of teachers and students, long-distance running can enhance physical fitness. However, due to nonstandard movements in sports, teachers and students get injured psysically. Taking the prevention of long-distance running injuries of teachers and students in colleges and universities as the research goal, this article investigates the teachers and students of a physical education college in Shanxi Province by means of questionnaire survey, counts the functional indexes of teachers and students in long-distance running for one year and analyzes the injuries. The results show that the injury rate of teachers and students is 45.5%; Teachers and students with only one injury are the most, and knee injuries are the most common, with a mild injury rate of 60.98% and concentrated in November-December and June-July. The main reasons for the injuries of teachers and students are poor physical fitness, insufficient warm-up, and poor sports equipment. Through full warm-up exercise and adequate rest, common injuries in long-distance running can be effectively prevented.

Abstract:The primary purpose of this study was to examine how the type and magnitude of changes in running behavior, as a consequence of COVID-19 pandemic restrictions, influence running-related injuries. Secondarily, we aimed to examine how lifestyle and psychosocial well-being measures may influence running behavior change. An online survey was advertised to individuals over the age of 18 that currently run or have previously participated in running for exercise. The survey questions examined injury history and new injuries sustained during COVID-19 restrictions, as well as changes related to training behavior changes, training environment changes, social behaviors, and psychosocial well-being. Changes reflected differences in running behaviors prior to COVID-19 restrictions (1 month prior to COVID-19 restrictions being imposed) and during COVID-19 restrictions (May 5 to June 10, 2020). A total of 1,035 runners were included in the analysis. Current injuries sustained during COVID-19 occurred in 9.5% of the runners. Injured runners made a greater number of total changes ( p = 0.031) as well as training-related ( p = 0.042) and environment-related ( p = 0.017) changes compared with uninjured runners. A significant relationship was found between injury and those that reported less time to exercise to changes in work environment ( p = 0017). This study highlights the multi-dimensional nature of running-related injuries and the need to consider the interaction of multiple changes in running behavior, rather than isolating single factors. Greater understanding of the underlying causes of running-related injuries can help reduce the risk of future injury.

Abstract:QUESTIONS: What is the incidence of running-related injuries (RRIs) in recreational runners? Which personal and training characteristics predict RRIs in recreational runners?DESIGN: Prospective cohort study.PARTICIPANTS: A total of 200 recreational runners answered a fortnightly online survey containing questions about their running routine, races, and presence of RRI. These runners were followed-up for a period of 12 weeks.OUTCOME MEASURES: The primary outcome of this study was running-related injury. The incidence of injuries was calculated taking into account the exposure to running and was expressed by RRI/1000 hours. The association between potential predictive factors and RRIs was estimated using generalised estimating equation models.RESULTS: A total of 84 RRIs were registered in 60 (31%) of the 191 recreational runners who completed all follow-up surveys. Of the injured runners 30% (n=18/60) developed two or more RRIs, with 5/18 (28%) being recurrences. The incidence of RRI was 10 RRI/1000 hours of running exposure. The main type of RRI observed was muscle injuries (30%, n=25/84). The knee was the most commonly affected anatomical region (19%, n=16/84). The variables associated with RRI were: previous RRI (OR 1.88, 95% CI 1.01 to 3.51), duration of training although the effect was very small (OR 1.01, 95% CI 1.00 to 1.02), speed training (OR 1.46, 95% CI 1.02 to 2.10), and interval training (OR 0.61, 95% CI 0.43 to 0.88).CONCLUSIONS: Physiotherapists should be aware and advise runners that past RRI and speed training are associated with increased risk of further RRI, while interval training is associated with lower risk, although these associations may not be causative.
