Abstract:Hyperglycemia contributes to the excessive proliferation and migration of vascular smooth muscle cells (VSMC), which are closely associated with atherosclerosis. MicroRNAs (miRNAs/miRs) constitute a novel class of gene regulators, which have important roles in various pathological conditions. The aim of the present study was to identify miRNAs involved in the high glucose (HG)-induced VSMC phenotype switch, and to investigate the underlying mechanism. miRNA sequencing and reverse transcription-quantitative PCR results indicated that inhibition of miR-125a expression increased the migration and proliferation of VSMCs following HG exposure, whereas the overexpression of miR-125a abrogated this effect. Furthermore, dual-luciferase reporter assay results identified that 3-hydroxy-3-methyglutaryl-coA reductase (HMGCR), one of the key enzymes in the mevalonate signaling pathway, is a target of miR-125a. Moreover, HMGCR knockdown, similarly to miR-125a overexpression, suppressed HG-induced VSMC proliferation and migration. These results were consistent with those from the miRNA target prediction programs. Using a rat model of streptozotocin-induced diabetes mellitus, it was demonstrated that miR-125a expression was gradually downregulated, and that the expressions of key enzymes in the mevalonate signaling pathway in the aortic media were dysregulated after several weeks. In addition, it was found that HG-induced excessive activation of the mevalonate signaling pathway in VSMCs was suppressed following transfection with a miR-125a mimic. Therefore, the present results suggest that decreased miR-125a expression contributed to HG-induced VSMC proliferation and migration via the upregulation of HMGCR expression. Thus, miR-125a-mediated regulation of the mevalonate signaling pathway may be associated with atherosclerosis.
Methyltransferase Like 3-Mediated N6-methylatidin Methylation Inhibits Vascular Smooth Muscle Cells Phenotype Switching Via Promoting Phosphatidylinositol 3-Kinase Mrna Decay.

Abstract:N6-methylatidine (m6A) is involved in post-transcriptional metabolism and a variety of pathological processes. However, little is known about the role of m6A in vascular proliferative diseases, particularly in vascular smooth muscle cells (VSMCs) phenotype switching-induced neointimal hyperplasia. In the current study, we discovered that methyltransferase like 3 (METTL3) is a critical candidate for catalyzing a global increase in m6A in response to carotid artery injury and various VSMCs phenotype switching. The inhibited neointimal hyperplasia was obtained after in vivo gene transfer to knock-down Mettl3. In vitro overexpression of Mettl3 resulted in increased VSMC proliferation, migration, and reduced contractile gene expression with a global elevation of m6A modification. In contrast, Mettl3 knockdown reversed this facilitated phenotypic switch in VSMCs, as demonstrated by downregulated m6A, decreased proliferation, migration, and increased expression of contractile genes. Mechanistically, Mettl3 knock-down was found to promote higher phosphatidylinositol 3-kinase (Pi3k) mRNA decay thus inactivating the PI3K/AKT signal to inhibit VSMCs phenotype switching. Overall, our findings highlight the importance of METTL3-mediated m6A in VSMCs phenotype switching and offer a novel perspective on targeting METTL3 as a therapeutic option for VSMCs phenotype switching modulated pathogenesis, including atherosclerosis and restenosis.
MiR-126 Promotes Endothelial Cell Apoptosis by Targeting PI3K/Akt in Rats with Lower Limb Arteriosclerosis Obliterans.

Abstract:OBJECTIVE:To investigate the influence of micro ribonucleic acid (miR)-126 on the rats with lower limb arteriosclerosis obliterans (ASO).MATERIALS AND METHODS:Male Sprague- Dawley rats aged 3 months old were randomly divided into Sham operation group (Control group, n=10) and Model group (n=10), and the model of lower limb ASO was established. After modeling, the expression of miR-126 in arteries was detected using quantitative Real Time-Polymerase Chain Reaction (qRT-PCR), and the change in the downstream signaling pathway was examined via Western blotting. The human umbilical vein endothelial cells (HUVECs) were induced by oxidized low-density lipoprotein (Ox-LDL) to establish the model of endothelial injury, followed by detection of miR-126 expression. Then, the Luciferase assay was performed to verify the downstream target gene of miR-126. After being cultured, HUVECs were set as Control group, Ox-LDL induction group, and Ox-LDL + miR-126 inhibitor group, and the expressions of phosphorylated-protein kinase B (p-Akt) and cleaved cysteine-aspartic protease-3 (Caspase-3) were detected in the above groups.RESULTS:After the establishment of the model, the expression level of miR-126 was raised in vessels, but the phosphatidylinositol 3-hydroxy kinase (PI3K)/Akt signals were weakened (p<0.01). Ox-LDL-induced endothelial cell apoptosis promoted the expression of miR-126, and the difference was statistically significant. The bioinformatics analysis results showed that PI3KR2 was a direct target of miR-126, which was also proven via the Luciferase assay. Moreover, the transfection with miR-126 inhibitor into endothelial cells suppressed Ox-LDL-induced cell apoptosis, thereby persistently activating the PI3K/Akt signaling pathway (p<0.01).CONCLUSIONS:In rats with lower limb arteriosclerosis obliterans (ASO), miR-126 represses the PI3K/Akt signaling pathway to accelerate endothelial cell apoptosis.
RelB Represses Mir-193A-5p Expression to Promote the Phenotypic Transformation of Vascular Smooth Muscle Cells in Aortic Aneurysm

Abstract:Aortic aneurysm (AA) is a potentially fatal disease with the possibility of rupture, causing high mortality rates with no effective drugs for the treatment of AA. The mechanism of AA, as well as its therapeutic potential to inhibit aneurysm expansion, has been minimally explored. Small non-coding RNA (miRNAs and miRs) is emerging as a new fundamental regulator of gene expression. This study aimed to explore the role and mechanism of miR-193a-5p in abdominal aortic aneurysms (AAA). In AAA vascular tissue and Angiotensin II (Ang II)-treated vascular smooth muscle cells (VSMCs), the expression of miR-193a-5 was determined using real-time quantitative PCR (RT-qPCR). Western blotting was used to detect the effects of miR-193a-5p on PCNA, CCND1, CCNE1, and CXCR4. To detect the effect of miR-193a-5p on the proliferation and migration of VSMCs, CCK-8, and EdU immunostaining, flow cytometry, wound healing, and Transwell Chamber analysis were performed. In vitro results suggest that overexpression of miR-193a-5p inhibited the proliferation and migration of VSMCs, and its inhibition aggravated their proliferation and migration. In VSMCs, miR-193a-5p mediated proliferation by regulating CCNE1 and CCND1 genes and migration by regulating CXCR4. Further, in the Ang II-induced abdominal aorta of mice, the expression of miR-193a-5p was reduced and significantly downregulated in the serum of patients with aortic aneurysm (AA). In vitro studies confirmed that Ang II-induced downregulation of miR-193a-5p in VSMCs by upregulation of the expression of the transcriptional repressor RelB in the promoter region. This study may provide new intervention targets for the prevention and treatment of AA.

Abstract:Aberrant smooth muscle cells (SMCs) play important roles in the formation of abdominal aortic aneurysm (AAA). Although the molecular mechanism of AAA formation has been investigated, there is a lack of understanding concerning the role of microRNAs (miRNAs) in AAA, which the current study aimed to address. Firstly, miRNA array analysis was performed in order to compare the miRNA profiles in a mouse model of AAA with those in normal control mice, and differentially expressed miRNAs were identified. miR-129-5p was selected for further analysis, and was used to transfect human SMCs. The results of an MTT assay revealed that miR-129-5p inhibited the proliferation of SMCs, and flow cytometry indicated that apoptosis was induced. Bioinformatic analysis predicted that Wnt5a was the potential target gene of miR-129-5p, and this was verified by luciferase assay. In summary, miR-129-5p inhibits cellular proliferation, induces apoptosis and modulates the Wnt5a signaling pathway in SMCs.

Abstract:Background Abdominal aortic aneurysms (AAA) are characterized by loss of vascular smooth muscle cells (SMCs). Growth factors induce proliferation and cell survival of SMCs. PI 3-kinase (PI3K) is an important downstream mediator in growth factor signaling. Objective This project is based on the hypothesis that PI3K-mediated SMC proliferation can compensate for the loss of SMCs in AAA and thus inhibit AAA progression. Therefore, we determined effects of reduced PI3K activity in SMCs on AAA progression, cell proliferation as well as vascular remodeling. Methods To investigate the role of PI3K, we analyzed mice and aortic SMCs harboring a smooth muscle-specific deficiency of the PI3K catalytic subunit p110α (SM-p110α-/-). AAA development in SM-p110α-/- mice and wild-type (WT) littermates was induced by perfusion of the infrarenal aortic segment with porcine pancreatic elastase (PPE). AAA diameter was determined by echocardiography. Structural changes in the aorta were identified by (immuno)histochemical staining. SMC proliferation was measured in vitro by BrdU incorporation or real-time cell analysis. p110α induced signal transduction pathways were characterized by Western blot and transcriptome analysis. Results PPE treatment led to a massive loss of SMCs in the compromised aortic segment after 3 days in both SM-p110α-/- and WT mice. The aortic diameter was enlarged after 28 days in all PPE-treated mice. However, the increase was significantly higher in SM-p110α-/- mice (70.2±10.8%, n=9) compared to WT animals (42.4±6.0%, n=10) (p<0.05). A similar effect was obtained in aged (untreated) mice: In 24-month-old SM-p110α-/- mice, abdominal aortic lumen diameter was increased by 42% compared to 3-month-old animals, but only by 15% in corresponding WT mice. PPE-treated SM-p110α-/- mice were characterized by reduced vascular remodeling with less PCNA-positive proliferating cells compared to WT animals. In addition, proliferation of aortic p110α-/- SMCs in serum-containing medium was impaired. Mechanistic analyses showed that PDGF- and insulin-induced activation of AKT as well as AKT-dependent phosphorylation and inactivation of the transcription factors FOXO-1, -3 and -4 were significantly reduced in p110α-/- SMCs compared to WT cells. Further analyses revealed that specific inhibition of FOXO1 by AS1842856 rescued PDGF or serum stimulated proliferation of p110α-/- SMCs. In addition, expression analysis of FOXOs in patients revealed significantly increased FOXO1 expression in AAA compared to aortic samples from healthy individuals. Conclusion Deficiency of the catalytic PI3K isoform p110α in SMCs promotes the development and progression of AAA. p110α/AKT-dependent inactivation of FOXO1 induces SMC proliferation, which can ameliorate aortic wall damage. As FOXO1 inhibitors and a recently developed p110α activator are available, the PI3K/AKT/FOXO1 signaling cascade may provide a therapeutic target to influence the stability of the aortic wall in AAA.

The Compatibility of Alisma and Atractylodes Affects the Biological Behaviours of VSMCs by Inhibiting the miR-128-5p/p21 Gene

Abstract:Objective. The compatibility of Alisma and Atractylodes (AA) has been estimated to exhibit antiatherosclerotic effects, but the mechanism remains unclear. This study aimed to identify the role of AA in oxidized low-density lipoprotein (ox-LDL)-induced vascular smooth muscle cell (VSMC) behaviours and to explore the effects of microRNAs (miRNAs). Methods. A scratch wound-healing assay was used to detect the migration of VSMCs, and immunocytochemistry and western blotting for SM22ɑ were used to evaluate phenotypic transformation. Bromodeoxyuridine (BrdU) immunocytochemistry and flow cytometry were applied to detect the proliferation of VSMCs. miRNA microarray profiling was performed using Lianchuan biological small RNA sequencing analysis. VSMCs were transfected with the miR-128-5p mimic and inhibitor, and the migration, phenotypic modulation, and proliferation of VSMCs were investigated. The 3′UTR-binding sequence site of miR-128-5p on the p21 gene was predicted and assessed by luciferase assays. Result. AA and the extracellular regulated protein kinase 1/2 (ERK1/2) blocker U0126 markedly inhibited migration, elevated smooth muscle 22α (SM22α) expression, repressed VSMC proliferation, elevated miR-466f-3p and miR-425-3p expression, and suppressed miR-27a-5p and miR-128-5p expression in ox-LDL-induced VSMCs. miR-128-5p targets the tissue inhibitor of metalloproteinases (TIMPs), silent information regulator 2 (SIRT2), peroxisome proliferator-activated receptor (PPAR), and p21 genes, which are linked to the behaviours of VSMCs. The miR-128-5p mimic promoted the migration and proliferation of VSMCs and suppressed p21, p27, and SM22ɑ expression. The inhibitor increased p21, p27, and SM22ɑ expression and repressed the migration, phenotypic transformation, and proliferation of VSMCs. miR-128-5p directly targeted the 3′UTR-binding sequences of the p21 gene, negatively regulated p21 expression, and supported the proliferation of VSMCs. Conclusion. Our research showed that the migration, phenotypic transformation, and proliferation of ox-LDL-induced VSMCs were repressed by AA through inhibiting miR-128-5p by targeting the p21 gene, which may provide an effective option for the treatment of atherosclerosis.

Inhibition Effect of Microrna-124 on Rat Vascular Smooth Muscle Cell Proliferation and Its Molecular Mechanism

Abstract:Objective To investigate the inhibition effect and molecular mechanism of microRNA-124 (miR-124) on rat vascular smooth muscle cell proliferation. Methods We applied 10 male 11-week-old spontaneously hypertensive rats (SHR) as experimental group and 10 age matched Wistar Kyoto (WKY) rats as control group. Aortic smooth muscle cells were isolated from the medial layer of thoracic aorta of SHR and WKY rats and cultured in Dulbecco's modified eagles medium (DMEM). Quantitative real-time PCR (RT-PCR) was used to detect the expression of miR-124 in SHR and WKY aortic smooth muscle cells. The SHR aortic smooth muscle cells were transfected with either miR-124 mimics or NC mimics control. MTT assay was used to explore the proliferation of SHR aortic smooth muscle cells in vitro. The targeted gene of miR-124 was predicted by bioinformatics, and verified by dual luciferase reporter assay. The over-expression of miR-124 in SHR aortic smooth muscle cells was analyzed by the mRNA RT-PCR and protein expression of Meox2 by Western blot analyses. Finally, SHR aortic smooth muscle cells proliferation was also detected using MTT assay after treated with siMeox2. Results The expression of miR-124 in SHR aortic smooth muscle cells was 0.22 times of WKY aortic smooth muscle cells (P<0.01). Over-expression of miR-124 could significantly inhibit SHR aortic smooth muscle cells proliferation. Bioinformatics analysis and dual luciferase reporter assay demonstrated that Meox2 was a target gene of miR-124. The mRNA level of Meox2 in SHR aortic smooth muscle cells after treated with miR-124 mimics was 0.29 times of transfected NC mimics control (P<0.01). Western blot showed that Meox2 protein was also decreased after treated with miR-124 mimics. Knock-down Meox2 could also arrest the SHR aortic smooth muscle cells proliferation. Conclusions The expression of miR-124 is down-regulated in SHR, which may be mediated by Meox2 regulation of SHR aortic smooth muscle cells proliferation and inhibit the occurrence of hypertensive disease. Rapamycin Attenuates a Murine Model of Thoracic Aortic Aneurysm by Downregulating the Mir-126–3P Mediated Activation of MAPK/ERK Signalling Pathway

Abstract:Thoracic aortic aneurysm (TAA) is fatal diseases, which leads to aortic rupture and sudden death. Blood pressure-lowering drugs are ineffective for most of the patients. Our previous study demonstrated the inhibition of endothelial secreted miR-126-3p by rapamycin ameliorate the aneurysmal phenotype of smooth muscle cells (SMCs) in vitro. Hence, this study aimed to evaluate the modulation and mechanism of miR-126-3p in a murine model of TAA (Fbn1(c139G/+)). Our results showed that noticeable disturbed flow (DF) was observed in the aorta of Fbn1(c139G/+) mice, and the expression of miR-126-3p was significantly increased under the DF in the cell chamber. This finding was also confirmed by tests in the corresponding DF area of the human aortic aneurysm tissue. Constant rapamycin administration significantly ameliorates the incidence and severity of Fbn1(c139G/+) mice characterized by decreased aortic media degradation, macrophage infiltration and MMP2/9 expression in the aortic wall. Mechanistic studies showed that rapamycin attenuates TAA progression by inhibiting miR-126-3p through ERK1/2 inactivation. (C) 2019 Elsevier Inc. All rights reserved.
MicroRNA-370-3p Inhibits Human Vascular Smooth Muscle Cell Proliferation Via Targeting KDR/AKT Signaling Pathway in Cerebral Aneurysm.

Abstract:OBJECTIVE:Cerebral aneurysm is a common vascular disease with high morbidity and mortality. Vascular smooth muscle deletion or dysplasia is an important reason for the development of cerebral aneurysm. MiRNAs participate in a variety of biological functions through inhibiting target gene translation. The aim of the present study was to evaluate the role of miRNAs in the regulation of vascular smooth muscle cell proliferation.MATERIALS AND METHODS:MiRNA and mRNA expressions were tested by Real-time PCR. Cell cycle was detected by flow cytometry. Cell viability was evaluated by MTT assay. HUASMC cell proliferation was determined by BrdU assay. Protein expressions were determined using Western blot. MiRNA target gene was confirmed by luciferase assay.RESULTS:MiR-370-3p expression was increased in cerebral aneurysm tissues. Ectopic expression of miR-370-3p suppressed proliferation of vascular smooth muscle cells and blocked cell cycle. Numerous cell proliferation and apoptosis-related factors were down-regulated by miR-370-3p. Results of target prediction database and dual-luciferase assay revealed that KDR is a direct target of miR-370-3p. Importantly, FOXO1 activity and AKT and FOXO1 phosphorylation were inhibited by miR-370-3p. We suggest that miR-370-3p directly targets KDR, resulting in the activation of AKT signaling pathway.CONCLUSIONS:MiR-370-3p was involved in the development of cerebral aneurysm by targeting KDR and blocking AKT/FOXO1 signaling pathway. The results provide theoretical basis for further investigation of potential clinical prevention and treatment of cerebral aneurysm.
MicroRNA-127-3p regulates inflammatory response in injury-induced vascular remodeling and cellular senescence in vascular smooth muscle cells through mTor

Micro RNAs (miRs) are promising therapeutic targets due to their cell-specific functions. Targeting miR-127-3p emerges as an attractive approach to enhance vascular healing and limit angioplasty-related complications. Here, we focus on elucidating the role of miR-127-3p in vascular smooth muscle cells (VSMC) to characterize their role in cellular function and vascular senescence. Methods Utilizing reverse transcription and qRT-PCR we checked expression levels of miR-127-3p in human and murine aortic VSMCs. After overexpression and downregulation via transfection of pre-miR and antagomiR, we evaluated the influence of miR-127-3p on cellular functions such as proliferation and migration of human (non-) replicative senescent VSMCs. Potential targets were selected via target prediction analysis and verified on mRNA and protein levels. Morphological changes were visualized via high-resolution fluorescence microscopy. Senescence marker expression was checked after transfection by qRT-PCR. Results MiR-127-3p is highly upregulated in femoral artery tissue on day 10 after wire-induced injury in C57Bl6 mice, serving as restenosis model (p<0.001) and aged C57BL/6 mice (3 months vs. 22 months, p<0.0001). Replicative senescent human VSMC exhibit increased miR-127-3p expression (p<0.05). In vitro, miR-127-3p overexpression reduces proliferation in senescent (p<0.001) and non-senescent (p<0.01) cells. It hinders migration in non-senescent cells (p<0.0001) but has a reverse effect in senescent cells (p<0.01). Potential targets affecting cellular functions, inflammatory pathways, and cellular aging were identified as mTOR, AXL, and TEAD3 (p<0.05). Transfection of pre-miR leads to increase in cell size and the acquisition of a flattened morphology of human VSMC. P14arf and laminB1, as senescence-associated genes, showed downregulation after overexpression of miR-127-3p on mRNA level (p<0.05). When VSMCs were stimulated with anti-miR-127-3p, increased mRNA levels of p14arf and laminB1 were detected (p<0.05). Conclusion This study identifies miR-127-3p as a contributor to cellular functions including cellular proliferation and migration and seems to have opposing effects in non- and replicative senescent VSMC. Further research is needed to validate miR-127-3p as a therapeutic target for vascular aging and remodeling in vitro and in vivo.

We aimed to investigate the role of p55 gamma, a regulatory subunit of PI3Ks, in vascular smooth muscle cell (VSMC) proliferation and neointimal formation. Methods and results We identified p55 gamma as an important factor that suppresses VSMC proliferation and injury-evoked neointimal formation. Western blot and mRNA analyses showed that p55 gamma expression declined in balloon-injured rat carotid arteries and in response to PDGF-BB and serum treatment in cultured VSMCs. Overexpression of p55 gamma inhibited, whereas short hairpin RNA knockdown of p55 gamma promoted PDGF-BB- and serum-induced VSMC proliferation. Importantly, in vivo adenoviral gene transfer of p55 gamma into carotid arteries attenuated, while knockdown of p55 gamma enhanced balloon injury-induced neointimal formation. Furthermore, p55 gamma sequentially up-regulated p53 and p21, resulting in cell-cycle arrest in S phase; small-interfering RNA knockdown of either p53 or p21 blocked p55 gamma-induced VSMC growth arrest. Mechanistically, p55 gamma interacted with and stabilized p53 protein by blocking mouse double minute 2 homologue-mediated p53 ubiquitination and degradation, subsequently activating its target gene p21. Concurrently, p55 gamma up-regulated Bcl-xl expression, resulting in non-apoptotic growth arrest effect. Conclusion These findings mark p55 gamma as a novel upstream regulator of the p53-p21 signalling pathway that negatively regulates VSMC proliferation, suggesting that malfunction of p55 gamma may trigger vascular proliferative disorders.

Abstract:Objective: To investigate the protective efficacy of miR-155 on down regulating NoxA1 gene expression, resulting in an inhibitory effect of vascular smooth muscle cell (VSMC) over proliferation and thus alleviating the progression of coronary artery atherosclerosis in mouse disease model. Methods: Carotid artery of C57BL6 mouse was used for VSMC primary culture. The isolated VSMC was transfected with recombinant Pad2YFG adenovirus fluorescent vector with miR-155 fragment (mimic or mutant). Fluorescence microscope was applied to observe the transfection rate of miR-155 into VSMC. Artificial artery-injured mouse model fed with high-fat diet was established as host to accept VSMC transplantation via tail vein injection for 3 consecutive days. Aortic and carotid arteries were extracted at week 6. Distribution of miR-155 was quantified; protein and RNA were extracted to detect NoxA1 and NADPH expression in vehicle control, miR-155 mimic group, and the inhibitor group. Immunohistochemistry was performed in arteries section to compare the thickness of neointima and assess the severity of AS of each group. Results: The miR-155 distribution was observed varied at week 6 in control, miR-155 mimic and inhibitor groups. The NoxA1 protein expression in VSMC was decreased in mimic group at week 6 vs control and inhibitor groups (p<0.05); no significant difference of NADPH expression was observed in all groups. The NoxA1 and NADPH gene expression in VSMC were both found reduced compared with those of control group at week 6 (p<0.05). Immunohistochemistry of artery frozen sections figured out that the thickness of neointima of carotid artery in miR-155 mimic group was significantly lower vs control and inhibitor groups (p<0.01) at week 6. Conclusion: miR-155 transfection into VSMC seems to have reversed regulatory effect on NoxA1 expression resulting in amelioration of atherosclerotic lesion in artificial AS mouse model. A therapeutic strategy based on the functional role of miR-155 in VSMC over proliferation-based atherosclerosis would need to complementarily-synthesize miRNAs, which is a promising therapeutic tool due to the enhanced stability and sustained effects after intravenous injection in the arterial wall.

Abstract:Numerous microRNAs participate in regulating the pathological process of atherosclerosis. We have found miR‐130a is one of the most significantly down‐regulated microRNAs in arteriosclerosis obliterans. Our research explored the function of miR‐130a in regulating proliferation by controlling autophagy in arteriosclerosis obliterans development. A Gene Ontology (GO) enrichment analysis of miR‐130a target genes indicated a correlation between miR‐130a and cell proliferation. Thus, cell cycle, CCK‐8 assays and Western blot analysis were performed, and the results indicated that miR‐130a overexpression in vascular smooth muscle cells (VSMCs) significantly attenuated cell proliferation, which was validated by an in vivo assay in a rat model. Moreover, autophagy is thought to be involved in the regulation of proliferation. As our results indicated, miR‐130a could inhibit autophagy, and ATG2B was predicted to be a target of miR‐130a. The autophagy inhibition effect of miR‐130a overexpression was consistent with the effect of ATG2B knockdown. The results that ATG2B plasmids and miR‐130a mimics were cotransfected in VSMCs further confirmed our conclusion. In addition, by using immunohistochemistry, the positive results of LC3 II/I and ATG2B in the rat model and artery vascular tissues from the patient were in accordance with in vitro data. In conclusion, our data demonstrate that miR‐130a inhibits VSMCs proliferation via ATG2B, which indicates that miR‐130a could be a potential therapeutic target that regulates autophagy in atherosclerosis obliterans.

Abstract:Proliferation of vascular smooth muscle cells (VSMCs) is a primary mechanism underlying cardiovascular proliferative disorders. Phosphoinositide 3-kinase (PI3K)-Akt (or protein kinase B) axis has been assigned at the center of pathways that regulate cell proliferation. Here we demonstrate that enhanced PI3K-Akt signaling by mitogenic stimulation or arterial injury profoundly elevates expression of receptor interacting protein 3 (RIP3) in primary cultured rat VSMCs and in vivo and that the up-regulation of RIP3 leads to VSMC growth arrest and apoptosis via inhibiting the PI3K-Akt signaling pathway, thereby alleviating balloon injury-induced neointimal formation. Specifically, mitogenic stimulation with platelet-derived growth factor-BB or angiotensin II leads to a profound increase in RIP3 expression, which is abolished by inhibition of PI3K or Akt, and increased PI3K-Akt signaling by expression of a constitutively active PI3K mutant also elevates RIP3 expression. Importantly, adenoviral overexpression of RIP3 not only triggers apoptosis but also causes cell cycle arrest at G1/G0 phases that is associated with suppressed Akt activation. In sharp contrast, RIP3 gene silencing enhances serum- and platelet-derived growth factor-induced cell proliferation and Akt activation. In vivo adenoviral gene delivery of rat RIP3 (rRIP3) increased apoptosis and reduced VSMC proliferation, thus, effectively alleviating balloon injury-induced neointimal formation. The growth-suppressive and pro-apoptotic effects are independent of rRIP3 Ser/Thr kinase activity, because overexpression of a kinase-inactive mutant of rRIP3, similar to its wild type, is sufficient to induce growth arrest and apoptosis. These findings reveal a novel growth-suppressive action of RIP3, marking RIP3 as an important factor to prevent excessive mitogenic stimulation- or injury-induced vascular smooth muscle cells hyperplasia.

Abstract:<span> Abdominal aortic aneurysm (AAA) is an aging-related degenerative disease. miR-188-5p was reported to induce cell senescence and play a key role in aging-related disease. Therefore, in this study, we investigated miR-188-5p expression during progression in experimental AAAs. Furthermore, we investigated whether inhibition of miR-188-5p could suppress AAA progression. Experimental AAAs were created in 9–12-week-old male C57BL/6J mice by transient intra-aortic infusion of porcine pancreatic elastase. Expression of miR-188-5p levels were assessed in aneurysmal and control aortae during the progression of aneurysm. For inhibition experiment, miR-188 inhibiting group mice were injected with AAV2-miR188-5p sponge through tail vein and control group mice were injected with AAV2-CMV-GFP. Influences on experimental AAA progression were assessed by measurements of aortic diameter and histopathologic analysis at sacrifice. Meanwhile, immunohistochemistry and fluorescence in situ hybridization were used to determine the inflammatory cells infiltration and colocalization of miR-188-5p in aortic sections. Expression of miR-188-5p is upregulated during progression of AAA. Importantly, miR-188-5p inhibition treatment prevented enlargement of experimental aneurysms. Meanwhile, miR-188-5p inhibition regimens attenuated medial elastin degradation, smooth muscle cell depletion, and mural angiogenesis and the accumulation of macrophages, T cells, and angiogenesis. Furthermore, colocalization of miR188-5p with CD68 and CD3 was observed, which suggest miR-188-5p was expressed mainly in infiltrated macrophages and T cells. Expression of miR-188-5p is increased in experimental AAAs. Treatment with miR-188-5p inhibition limits experimental AAA progression, with histologic evidence of reduced neovessels and attenuated mural leukocyte infiltration. These findings underscore the potential significance of miR-188-5p in aneurysm pathogenesis and as a target for suppression of AAA disease.</span>

Abstract:Aortic dissection (AD) has a rapid onset and a high mortality. This study explored mechanism of miR-146b and PI3k/Akt on aortic dissection. Human aortic vascular smooth muscle cells (T/GHA-VSMC) were assigned into blank group, empty vector group, miR-146b group and miR-146b group with nanoparticles as carrier (vector+miR-146b group), followed by analysis of cell proliferation, apoptosis and migration. Results showed that, expressions of PI3k/Akt, miR-146b and cell proliferation in the empty vector, miR-146b, and vector+miR-146b groups were lower than in the blank group (P <0.05), with lowest expression level in the empty vector group (P <0.05). Moreover, cell apoptosis was increased (P <0.05) and migration ability was decreased (P <0.05) after transfection, and miR-146b was found to target the PI3k/Akt. Nanoparticle-mediated miR-146b can inhibit the proliferation and migration of T/GHA-VSMC in AD patients, which is related to PI3k/Akt signaling. miR-146b may become a potential molecular target for AD, which can alleviate the occurrence and development of AD.

Abstract:Abstract Accumulating evidence shows that the abnormal proliferation and migration of vascular smooth muscle cells (VSMCs) can significantly affect the long‐term prognosis of coronary artery bypass grafting. This study aimed to explore the factors affecting the proliferation, migration, and phenotypic transformation of VSMCs. First, we stimulated VSMCs with different platelet‐derived growth factor‐BB (PDGF‐BB) concentrations, analyzed the expression of phenotype‐associated proteins by Western blotting, and examined cell proliferation by scratch wound healing and the 5‐ethynyl‐2‐deoxyuridine (EdU) assay. VSMC proliferation was induced most by PDGF‐BB treatment at 20 ng/mL. miR‐200a‐3p decreased significantly in A7r5 cells stimulated with PDGF‐BB. The overexpression of miR‐200a‐3p reversed the downregulation of α‐SMA ( p < 0.001) and the upregulation of vimentin ( p < 0.001) caused by PDGF‐BB. CCK8 and EdU analyses showed that miR‐200a‐3p overexpression could inhibit PDGF‐BB‐induced cell proliferation ( p < 0.001). However, flow cytometric analysis showed that it did not significantly increase cell apoptosis. Collectively, the overexpression of miR‐200a‐3p inhibited the proliferation and migration of VSMCs induced by PDGF‐BB, partly by affecting phenotypic transformation‐related proteins, providing a new strategy for relieving the restenosis of vein grafts.
