Abstract:Reactive oxygen species(ROS) mainly produced by oxidative stress,not only causes DNA damage and genetic unstability,but also closely involves in organ lesion and tumor development.Recently,taking advantage of ROS difference between the normal cells and tumor cells,ROS responsive nano-delivery system is designed.Drugs only release within the tu mor micro-environment at high level of ROS,which ensures high selectivity and therapeutic efficacy.The newly progress made in this field will be briefly discussed in this paper.

Abstract:Recently, inorganic nanomaterials have received considerable attention for use in biomedical applications owing to their unique physicochemical properties based on their shapes, sizes, and surface characteristics. Photodynamic therapy (PDT), sonodynamic therapy (SDT), and chemical dynamic therapy (CDT), which are cancer therapeutics mediated by reactive oxygen species (ROS), have the potential to significantly enhance the therapeutic precision and efficacy for cancer. To facilitate cancer therapeutics, numerous inorganic nanomaterials have been developed to generate ROS. This mini review provides an overview of the generation mechanisms of ROS by representative inorganic nanomaterials for cancer therapeutics, including the structures of engineered inorganic nanomaterials, ROS production conditions, ROS types, and the applications of the inorganic nanomaterials in cancer PDT, SDT, and CDT.

Abstract:Given the fact that excessive levels of reactive oxygen species (ROS) induce damage to proteins, lipids, and DNA, various ROS-generating agents and strategies have been explored to induce cell death and tumor destruction by generating ROS above toxic threshold. Unfortunately, hypoxia in tumor microenvironment (TME) not only promotes tumor metastasis but also enhances tumor resistance to the ROS-generated cancer therapies, thus leading to ineffective therapeutic outcomes. A variety of nanotechnology-based approaches that generate or release O 2 continuously to overcome hypoxia in TME have showed promising results to improve the efficacy of ROS-generated cancer therapy. In this minireview, we present an overview of current nanomaterial-based strategies for advanced cancer therapy by modulating the hypoxia in the TME and promoting ROS generation. Particular emphasis is put on the O 2 supply capability and mechanism of these nanoplatforms. Future challenges and opportunities of design consideration are also discussed. We believe that this review may provide some useful inspiration for the design and construction of other advanced nanomaterials with O 2 supply ability for overcoming the tumor hypoxia-associated resistance of ROS-mediated cancer therapy and thus promoting ROS-generated cancer therapeutics.

Abstract:Intracellular reduction–oxidation (redox) homeostasis, mediated mainly by reactive oxygen species (ROS), plays a significant role in physiological function maintenance and regulation of organisms. The intrinsic biochemical properties of ROS remarkably influence the growth, differentiation, metastasis, and death of tumor cells, leading to the exploitation of these active chemical species for cancer therapy. Due to significant advances in nanotechnology, a variety of nanomaterials have been developed to alter the spatiotemporal dynamic behaviors of ROS in physiological environments. In this review, we provide a detailed description and analysis of the latest advances in redox regulation–based nanotherapies, focusing on the basic material chemistry of nanomaterials through which ROS can be generated or scavenged to improve the anticancer therapeutic effects. In addition, key issues and core technologies in the development of ROS–based nanotherapies are discussed, aimed at unlocking the inherent power of nanotechnology to optimize therapeutic outcomes.Developments in this growing field are expected to contribute to the further basic research and clinical applications of redox–based nanotherapies.

Abstract:Tumor resistance to apoptosis and the immunosuppressive tumor microenvironment are two main reasons for the poor outcomes of tumor treatment. Pyroptosis is a newly discovered form of programmed cell death, featuring surprising proinflammatory activity. Recent evidence shows that tumor cell pyroptosis leads to strong inflammatory responses and significant tumor regression, which is different from apoptotic cell death. Strategies based on different types of nanoparticles for pyroptosis induction have become a useful tool for tumor treatment. Reactive oxygen species (ROS) is considered an efficient way to cause pyroptosis, and ROS‐based nanotechnologies have been a powerful strategy for pyroptosis induction and cancer treatments. This review covers recent progress in ROS‐induced pyroptosis and nanotherapy to trigger ROS‐mediated pyroptosis in cancer.

Abstract:Developing efficient cancer treatment modalities is crucial, given the tens of thousands of deaths that cancer causes each year. Although there are various treatment modalities available, such as chemotherapy, photodynamic therapy (PDT), photothermal therapy (PTT), chemodynamic therapy (CDT), sonodynamic therapy (SDT), etc., individual treatment approaches have limitations in achieving efficient tumor ablation due to the complex tumor microenvironment and the shortfalls of single therapeutic modality. The recent advancements in material science have led to the emergence of efficient cascade-based nanosystems that optimize these treatment modalities. Among such modalities, cascade therapy triggered by reactive oxygen species (ROS) has proven particularly effective through the synergism between photodynamic and chemodynamic therapies. This review focuses on the recent advances in nanosystem-based synergistic therapy via a cascade of ROS generated by photosensitizers, sonosensitizers, and Fenton reactions, emphasizing the design and cascade therapy mechanism of these approaches. Furthermore, the article covers the challenges and future outlook of such ROS generation-based cascade strategies for cancer therapy.

Abstract:Elevated oxidative stress is a representative hallmark of tumor cells. The development of redox-responsive nanovectors capable of being selectively activated in cancer cells has emerged as a popular approach to fabricate anticancer agents. However, despite tremendous achievements of redox-responsive nanotherapeutics, many of them suffer from limited therapeutic efficacy due to the unsatisfactory selectivity of pathological tissues over normal ones. On the one hand, although cytosolic glutathione (GSH) is much higher than that in plasma, the cytosolic GSH concentrations of cancer cells and healthy cells is not significantly different. On the other hand, elevated ROS levels are not sufficient to selectively activate the therapeutic agents in cancer cells. To this end, amplification of intracellular ROS levels to enhance the selective responsiveness of redox-responsive nanotherapeutics in cancer cells is of increasing interest. In this Minireview, redox-responsive nanovectors responding to intracellular endogenous redox species is first briefly introduced. Subsequently, the strategies to engineer cancer cells as intelligent nanoreactors aiming at amplification of intracellular oxidative stress by either boosting ROS generation and/or suppressing antioxidation systems are discussed. These ROS-generating platforms can be either directly used for chemodynamic therapy (CDT) or be coupled with conventional redox-responsive nanotherapeutics, representing a novel and promising methodology to improve therapeutic efficacy.

Abstract:Head and neck squamous cell carcinoma (HNSCC) is the most common type of head and neck cancer (HNC). Conventional treatment of HNSCC involves surgical removal, together with or without chemoradiotherapy. Because of the special location of head and neck tumors, trauma of surgical intervention often impairs physiological functions and causes disfigurement, seriously affecting the quality of life of HNSCC patients. High levels of reactive oxygen species (ROS) have been reported to cause cancer cell death by inducing apoptosis, autophagy, necroptosis and ferroptosis. Based on this, some non-invasive nanotherapeutics have aroused considerable attention, which could promote the generation of ROS with or without external stimuli, damaging biological macromolecules to kill cancer cells. Herein, the authors present the mechanisms of ROS-based cancer therapy and summarize different nanotherapeutics against HNSCC, including photodynamic therapy (PDT), sonodynamic therapy (SDT), radiation therapy (RT), and chemodynamic therapy (CDT). Furthermore, ROS-sensitive drug release nanosystems and some synergistic therapeutic modalities for HNSCC are reviewed. Finally, challenges and future perspectives of these treatments are also discussed for further research directions, which can facilitate the development of ROS-based nanotherapeutics for clinical application.

Abstract:Altered redox metabolism is one of the hallmarks of tumor cells, which not only contributes to tumor proliferation, metastasis, and immune evasion, but also has great relevance to therapeutic resistance. Therefore, regulation of redox metabolism of tumor cells has been proposed as an attractive therapeutic strategy to inhibit tumor growth and reverse therapeutic resistance. In this respect, nanomedicines have exhibited significant therapeutic advantages as intensively reported in recent studies. In this review, we would like to summarize the latest advances in nanomaterial-assisted strategies for redox metabolic regulation therapy, with a focus on the regulation of redox metabolism-related metabolite levels, enzyme activity, and signaling pathways. In the end, future expectations and challenges of such emerging strategies have been discussed, hoping to enlighten and promote their further development for meeting the various demands of advanced cancer therapies. It is highly expected that these therapeutic strategies based on redox metabolism regulation will play a more important role in the field of nanomedicine.

Abstract:Reactive oxygen species (ROS)-responsive nanocarriers have aroused widespread interest in recent years. On the one hand, a high ROS level has been detected in many types of tumor cells. On the other hand, ROS generation is also induced during photodynamic, sonodynamic, or chemodynamic therapy. In addition, multiple types of polymers are sensitive to ROS. Therefore, numerous ROS-responsive polymeric nanocarriers with unique ROS-responsive characteristics have been developed. This review discusses ROS-sensitive polymeric nanocarriers to improve drug delivery efficacy. In particular, ROS-responsive nanocarriers for synergistic cancer therapy are highlighted. The development of novel ROS-sensitive nanocarriers holds great potential for combining ROS-mediated therapy, such as photodynamic therapy, and other therapies to achieve synergistic anticancer efficacy.

Abstract:Reactive oxygen species (ROS)-based nanodynamic therapies are attractive treatment cancer strategies with excellent characteristics such as high selectivity, low toxicity and low multidrug resistance. Metal-organic frameworks (MOFs) and MOFs-based nanoplatforms exhibit promising properties such as self-reactive properties, morphology diversity, high loading capacities, easy functionalization and biocompatibility, providing more possibilities for the design of nanodynamic therapies. In this review, we focus on these excellent properties of MOFs and the construction of MOFs-based nanodynamic therapies for cancer treatment, including photodynamic therapy (PDT), chemodynamic therapy (CDT), and sonodynamic therapy (SDT). In addition, the optimization strategies of MOFs-based nanodynamic therapies are summarized in various aspects, meanwhile the research and applications of synergistic cancer therapies are systematically reviewed. Finally, we analyze the prospects and challenges of clinical applications of MOFs-based nanodynamic therapies, providing a valid reference and a solid foundation for further research.

Abstract:Numerous pathological conditions, including cancer, inflammatory diseases, and neurodegenerative diseases, are accompanied by overproduction of reactive oxygen species (ROS). This makes ROS vital flagging molecules in disease pathology. ROS-responsive drug delivery platforms have been developed. Nanotechnology has been broadly applied in the field of biomedicine leading to the progress of ROS-responsive nanoparticles. In this review, we focused on the production and physiological/pathophysiological impact of ROS. Particular emphasis is put on the mechanisms and effects of abnormal ROS levels on oxidative stress diseases, including cancer, inflammatory disease, and neurodegenerative diseases. Finally, we summarized the potential biomedical applications of ROS-responsive nanocarriers in these oxidative stress diseases. We provide insights that will help in the designing of new ROS-responsive nanocarriers for various applications.

Abstract:Accumulating evidence shows that elevated levels of reactive oxygen species (ROS) are associated with cancer initiation, growth, and response to therapies. As concentrations increase, ROS influence cancer development in a paradoxical way, either triggering tumorigenesis and supporting the proliferation of cancer cells at moderate levels of ROS or causing cancer cell death at high levels of ROS. Thus, ROS can be considered an attractive target for therapy of cancer and two apparently contradictory but virtually complementary therapeutic strategies for the regulation of ROS to treat cancer. Despite tremendous resources being invested in prevention and treatment for cancer, cancer remains a leading cause of human deaths and brings a heavy burden to humans worldwide. Chemotherapy remains the key treatment for cancer therapy, but it produces harmful side effects. Meanwhile, the process of de novo development of new anticancer drugs generally needs increasing cost, long development cycle, and high risk of failure. The use of ROS-based repurposed drugs may be one of the promising ways to overcome current cancer treatment challenges. In this review, we briefly introduce the source and regulation of ROS and then focus on the status of repurposed drugs based on ROS regulation for cancer therapy and propose the challenges and direction of ROS-mediated cancer treatment.

Abstract:One fundamental mechanism widely described for nanotoxicity from nanomaterials involves oxidative damage due to generation of free radicals and other reactive oxygen species (ROSs). Indeed, the ability of nanoscale materials to facilitate the transfer of electrons, and thereby promote oxidative damage or in some instances provide antioxidant protection, may be a fundamental property of nanomaterials. Effective methods are needed to assess oxidative damage elicited by nanoscale materials. The production of ROSs induced by nanomaterials is a double-edged sword, bringing not only the benefits of efficient nanomaterials for therapeutic treatment of diseases, but also possible health and environmental risks associated with them. Therefore, it is important to give a brief review on ROSs of nanomaterials and their relation in various biomedical applications.

Abstract:Functional subcellular organelle mitochondria are emerging as a crucial player and driver of cancer. Precision medicine targeting mitochondria can change nutrient availability and redox homeostasis in cancer cells. This review highlights how the modification capable of manipulating nanomaterials for reactive oxygen species (ROS) generation strategies can influence the rate of mitochondrial ROS production and their implementations in cancer therapy. Functional subcellular organelle mitochondria are emerging as a crucial player and driver of cancer. For maintaining the sites of cellular respiration, mitochondria experience production, and accumulation of reactive oxygen species (ROS) underlying oxidative damage in electron transport chain carriers. Precision medicine targeting mitochondria can change nutrient availability and redox homeostasis in cancer cells, which might represent a promising strategy for suppressing tumor growth. Herein, this review highlights how the modification capable of manipulating nanomaterials for ROS generation strategies can influence or compensate the state of mitochondrial redox homeostasis. We propose foresight to guide research and innovation with an overview of seminal work and discuss future challenges and our perspective on the commercialization of novel mitochondria‐targeting agents.

Abstract:Nano-biotechnologies which combine diagnosis with therapy in an integrated nanoplatform provide a promising prospect for cancer theranostics. Currently, the development of personalized medicine requires the exploitation of "smart" theranostic nanoplatforms with specific targeting, precise cargoes release, non-invasive therapeutics for cancers and so on. High levels of hydrogen peroxide (H2O2) and glutathione (GSH) are prominent features of tumor microenvironment (TME), which are distinctly different from healthy tissues. Accordingly, extensive redox-responsive theranostic nanoplatforms have been exploited by modulating intracellular redox homeostasis. In this review, we first summarized the recent advances of overexpressed H2O2- and antioxidant GSH-responsive nanoplatforms for tumor diagnose and treatment. Then, the strategies by synergistically boosting reactive oxygen species (ROS) production and GSH depletion for amplifying oxidative stress are highlighted. At last, the prospects and controversies of stimuli-driven nanotheranostics are also discussed for future development.

Abstract:Reactive Oxygen Species (ROS)‐regulating nanomedicines for disease treatments mainly include ROS‐upregulating nanomedicines and ROS‐downregulating nanomedicines. Here, ROS‐upregulating nanomedicines can exert the toxic effect by employing nanoplatforms to enhance ROS generation in pathological sites for ROS‐induced toxic therapy, and ROS‐downregulating nanomedicines can scavenge excess ROS to maintain normal physiological process and avoid oxidative stress injuries for antioxidant therapy.Reactive oxygen species (ROS) play an essential role in physiological and pathological processes. Studies on the regulation of ROS for disease treatments have caused wide concern, mainly involving the topics in ROS‐regulating therapy such as antioxidant therapy triggered by ROS scavengers and ROS‐induced toxic therapy mediated by ROS‐elevation agents. Benefiting from the remarkable advances of nanotechnology, a large number of nanomaterials with the ROS‐regulating ability are developed to seek new and effective ROS‐related nanotherapeutic modalities or nanomedicines. Although considerable achievements have been made in ROS‐based nanomedicines for disease treatments, some fundamental but key questions such as the rational design principle for ROS‐related nanomaterials are held in low regard. Here, the design principle can serve as the initial framework for scientists and technicians to design and optimize the ROS‐regulating nanomedicines, thereby minimizing the gap of nanomedicines for biomedical application during the design stage. Herein, an overview of the current progress of ROS‐associated nanomedicines in disease treatments is summarized. And then, by particularly addressing these known strategies in ROS‐associated therapy, several fundamental and key principles for the design of ROS‐associated nanomedicines are presented. Finally, future perspectives are also discussed in depth for the development of ROS‐associated nanomedicines.

Abstract:The therapeutic efficiency of reactive oxygen species (ROS)‐based nanotherapeutics is restrained by the rigorous production conditions of relatively sufficient and kinetically matching supply of intracellular substrates. The cumulative disruption of redox homeostasis and consequent pathology (e.g., Parkinson's disease) with low levels of substrates in living organisms may provide a promising model for ROS‐based therapy. Herein, a catechol chemistry‐mediated ternary nanostructure is prepared for long‐lasting generation of oxidative •OH in weakly acidic, low H2O2 homeostasis conditions of tumor. This platform employs mesoporous polydopamine (MPDA) as the porous redox mediator, while PDA‐induced sequential precipitation and biomineralization lead to hydroxy iron oxide (FeOOH) as the "iron reservoir," and calcium phosphate (CaP) as the pH‐sensitive sheddable shell. In weakly acidic conditions, the CaP layer can be degraded to expose the catalytic surface of Fe‐dopamine interplay, where FeOOH dissolution, Fe(III) chelation, Fe(III) reduction, Fe(II) release take place sequentially and continuously for Fe(II) recycling and Fenton catalysis. Both in vitro and in vivo studies verify the significant inhibition of cancer cells and tumor regression, which can also be strengthened by the local photothermal heating. This work establishes the first paradigm of pathologically inspired nanohybrids of ROS generators with long‐lasting efficacy for cancer therapy.

Abstract:Reactive oxygen species (ROS) play a dual role in the initiation, development, suppression, and treatment of cancer. Excess ROS can induce nuclear DNA, leading to cancer initiation. Not only that, but ROS also inhibit T cells and natural killer cells and promote the recruitment and M2 polarization of macrophages; consequently, cancer cells escape immune surveillance and immune defense. Furthermore, ROS promote tumor invasion and metastasis by triggering epithelial-mesenchymal transition in tumor cells. Interestingly, massive accumulation of ROS inhibits tumor growth in two ways: (1) by blocking cancer cell proliferation by suppressing the proliferation signaling pathway, cell cycle, and the biosynthesis of nucleotides and ATP and (2) by inducing cancer cell death via activating endoplasmic reticulum stress-, mitochondrial-, and P53- apoptotic pathways and the ferroptosis pathway. Unfortunately, cancer cells can adapt to ROS via a self-adaption system. This review highlighted the bidirectional regulation of ROS in cancer. The study further discussed the application of massively accumulated ROS in cancer treatment. Of note, the dual role of ROS in cancer and the self-adaptive ability of cancer cells should be taken into consideration for cancer prevention.

Abstract:Triggered by the endogenous chemical energy in the tumor microenvironment (TME), chemodynamic therapy (CDT) as an emerging non‐exogenous stimulant therapeutic modality has received increasing attention in recent years. The chemodynamic agents can convert internal hydrogen peroxide (H2O2) into the lethal reactive oxygen species (ROS) hydroxyl radicals (•OH) for oncotherapy. Compared with other therapeutic modalities, CDT possesses many notable advantages, such as tumor‐specific, highly selective, fewer systemic side effects, and no need for external stimulation. Nevertheless, mild acid pH, low H2O2 content, and overexpressed reducing substance in TME severely suppressed the CDT efficiency. With the rapid development of nanotechnology, some kinds of nanomaterials have been utilized with improved CDT efficiency. In particular, the excellent photo‐, ultrasound‐, magnetic‐, and other stimuli‐response properties of nanomaterials make it possible for combination cancer therapy of CDT with other therapeutic modalities, and it has shown superior anti‐cancer activity than monotherapies. Therefore, it is necessary to summarize the application of nanomaterial‐based chemodynamic cancer therapy. In this review, the various nanomaterials‐based nanoplatforms for CDT and its combinational therapies are summarized and discussed, aiming to provide inspiration for the design of better‐quality agents to promote the CDT development and lay the foundation for its future conversion to clinical applications.
