Abstract:CD36 is a multifunctional cell surface glycoprotein that acts as a surface receptor for thrombospondin (TSP), and thereby may mediate adhesive interactions between cells and substrata, platelets and other cells, and macrophages and apoptotic neutrophils. The identity of the TSP binding site on CD36 is controversial and may involve more than one structural domain. We have constructed a series of recombinant bacterial GST/CD36 fusion proteins that span nearly all of the CD36 molecule and have demonstrated that fusion proteins containing the region extending from amino acid 93 to 120 formed specific, saturable, and reversible complexes with TSP. As with intact CD36, binding was calcium-dependent, was independent of which ligand was immobilized, and was blocked by monoclonal antibodies to both CD36 and TSP. Stoichiometry and affinity of the fusion proteins for TSP were consistent with that of the intact protein. We also demonstrated that these fusion proteins competitively inhibited binding of TSP to purified platelet CD36 and to cell surface CD36 on peripheral blood monocytes and CD36 cDNA-transfected melanoma cells. These data demonstrate that the region between amino acids 93 and 120 has all of the characteristics required of the TSP binding domain.
Effect of Β-Glucosidase As a Feed Supplementary on the Growth Performance, Digestive Enzymes and Physiology of Broilers

Abstract:The effects of beta-glucosidase on the overall growth performance and a set of physiological parameters of broilers were investigated. 240 male, one-day old Avine broiler chickswere randomly allocated to four treatment groups and fed with a corn-soybean meal supplemented with 0% (control), 0.2%, 0.4% and 0.6% beta-glucosidase. The 0.2% beta-glucosidase group, but not the 0.4% and 0.6% beta-glucosidase groups; showed a significantly increased average daily weight gain (p < 0.05) over that of the control. All three beta-glucosidase feed groups showed significantly higher feed conversion ratios than the control group (p < 0.05). Feed supplementation of 0.2% beta-glucosidase significantly raised the contents of serum isoflavone aglycones as shown by decreases of genistin and daizin (p < 0.01) and an increase of daidzein (p < 0.01). The 0.2% beta-glucosidase feeding significantly increased the intestinal amylase activity while it had little effect on lipase and trypsin activities (p > 0.05). 0.2% beta-glucosidase feeding also significant elevated the levels of high-density lipoprotein cholesterol and malate dehydrogenase while lowering the level of low-density lipoprotein cholesterol (LDL-C). Finally, beta-glucosidase improved the anti-oxidative activities of the animals; the 0.2% beta-glucosidase feed group had higher activities of superoxide dismutase (p < 0.05), glutathione peroxidase and glutathione reductase in the liver (p < 0.05). and malondialdehyde level in the serum (p < 0.05).

Abstract:Endo -xylanase and endo -glucanase are supplemented to poultry diets in order to improve nutrient digestion and non-starch polysaccharide (NSP) fermentation. Here, the action of these enzymes on alcohol insoluble solids (AIS) from wheat and maize grains as well as its implications for starch digestion in milled grains were evaluated in vitro , under conditions mimicking the poultry digestive tract. For wheat AIS, GH11 endo -xylanase depolymerized soluble arabinoxylan (AX) during the gizzard phase, and proceeded to release insoluble AX under small intestine conditions. At the end of the in vitro digestion (480 min), the endo -xylanase, combined with a GH7 endo -β-1,4-glucanase, released 30.5 % of total AX and 18.1 % of total glucan in the form of arabinoxylo- and gluco-oligosaccharides, as detected by HPAEC-PAD and MALDI-TOF-MS. For maize AIS, the combined enzyme action released 2.2 % and 7.0 % of total AX and glucan, respectively. Analogous in vitro digestion experiments of whole grains demonstrated that the enzymatic release of oligomers coincided with altered grain microstructure, as examined by SEM. In the present study, cell wall hydrolysis did not affect in vitro starch digestion kinetics for cereal grains. This study contributes to understanding the action of feed enzymes on cereal NSP under conditions mimicking the poultry digestive tract.

Abstract:The non-starch polysaccharides (NSPs) in cell walls can act as a barrier for digestion of intracellular nutrients. This effect is called "cage effect." Part of the success of fibrolytic enzymes in broiler feed is assumed to be attributed to cage effect reduction. Further, changes in viscosity and potential prebiotic action should also be considered. The aim of this study was to gain insight into the relative importance of the cage effect in xylanase efficacy in broilers. Using a 2×2 factorial design, 24 pens with 30 Ross 308 male chicks were fed corn-soy based diets consisting of normal and freeze-thawed (5 d at -18°C) corn, both with and without xylanase. The freeze-thaw method was used to eliminate the cage effect, whereas a corn-based diet was used to exclude viscosity effects. Body weights (BW), feed intake (FI), and feed conversion ratio (FCR) were determined at d 13, 26, and 39. A balance study was executed at the end of the growing phase. These birds were euthanized at d 34 (non-fasted) to determine the viscosity of digesta, blood metabolites, intestinal morphology, and microbiota composition. During the finisher period, there was a significant interaction between enzyme supplementation and freeze-thawing for FCR, in which FCR was improved by freeze-thawed corn and tended to be improved by normal corn+enzyme compared with the control group. The improvement in performance (finisher period) of freeze-thawed corn and xylanase coincided with increased gut absorption of glucose (based on postprandial plasma concentrations) and increased number of Clostridiumcluster IV in the caecum, and agreed with the higher gut villus height. In addition, xylanase inclusion significantly increased the postprandial plasma glycine and triglycerides concentration, and led to elevated bacterial gene copies of butyryl CoA:acetate CoA-transferase, suggesting a prebiotic effect of xylanase addition through more than just the cage effect reduction. The applied model managed to rule out viscosity by using corn, and it was possible to isolate the cage effect by freeze-thawing the dietary corn.

Abstract:While some progress has been made in stemming forest loss in the tropics, deforestation continues to threaten tropical biodiversity. Many tropical protected areas exist as “islands” surrounded by continuing land use change and development. To understand the interconnections between land cover, biodiversity, and population growth, we use a unique dataset from the Tropical Ecology Assessment and Monitoring (TEAM) Network together with Landsat derived land cover products, population data from the Gridded Population of the World (GPW), and roads data from the Global Roads Open Access Dataset (gROADS). We find significant relationships between road network length and forest loss and between road network length and agricultural development, but no clear links between population growth and land use change. This reaffirms the importance of planning future road extensions to limit their effects on tropical protected areas.
Effects of Xylanase on Nutrients Utilization and Digestive Organs Development in Broilers.

Abstract:The trial was conducted to study the effects of different levels of xylanase on the nutrients utilization,digestive-related organs development of broilers fed wheat-based diets.Two hundred and forty 1-day-old Arbor Acres broilers were allocated to 4 dietary treatments(with 5 replicates of 12 chicks each) in a randomized complete block design.A wheat-corn-based basal diet containing 40% wheat was designed with 0,500,1 000 and 5 000 U/kg xylanase supplemented,respectively.The trial lasted for 21 d.The results showed that supplementing xylanase improved the apparent dry matter digestibility(P=0.093) and AME(P=0.110),but the apparent utilization ratio of crude protein,Ca and P and fecal moisture ratio was not significantly improved in broilers at 17～21 d(P0.05).No significant differences were observed in the nutrients utilization among the xylanase groups(P0.05).Supplementation of 1 000 U/kg xylanase decreased significantly the relative length of ileum of broilers at 21 days of age(P0.05).No significant differences were observed in the relative weight of intestine,gaster,pancreas and spleen of broilers at 21 days of age among groups(P0.05).Supplementation of xylanase did not affect significantly tibia weight and length,ash and P content in tibia of the broilers at 21 days of age.These data indicated that xylanase,even over supplementation did not significantly improve nutrients utilization and most of digestive-related organs development of broilers fed diets with 40% wheat,and increasing supplementation of xylanase had no deleterious effects.

Abstract:Xylanase has been demonstrated to improve growth performance of broilers fed wheat- or corn-based diets due to its ability to degrade arabinoxylans (AX). However, content and structure of AX in corn and wheat are different, comparing effects of xylanase on cecal microbiota of broilers fed corn- or wheat-based diets could further elaborate the mechanism of the specificity of xylanase for different cereal grains. Thus, a total of 192 one-day-old broilers were randomly allotted into four dietary treatments, including wheat-soybean basal diet, wheat-soybean basal diet with 4,000U/kg xylanase, corn-soybean basal diet, and corn-soybean basal diet with 4,000U/kg xylanase to evaluate interactive effects of xylanase in corn- or wheat-based diets on broilers cecal microbiota during a 6-week production period. The results indicated that bacterial community clustering was mainly due to cereal grains rather than xylanase supplementation. Compared with broilers fed wheat-based diets, corn-based diets increased alpha-diversity and separated from wheat-based diets ( p <0.05). Xylanase modulated the abundance of specific bacteria without changing overall microbial structure. In broilers fed wheat-based diets, xylanase increased the abundance of Lactobacillus , Bifidobacterium , and some butyrate-producing bacteria, and decreased the abundance of non-starch polysaccharides-degrading (NSP) bacteria, such as Ruminococcaceae and Bacteroidetes ( p <0.05). In broilers fed corn-based diets, xylanase decreased the abundance of harmful bacteria (such as genus Faecalitalea and Escherichia-Shigella ) and promoted the abundance of beneficial bacteria (such as Anaerofustis and Lachnospiraceae_UCG_010 ) in the cecum ( p <0.05). Overall, xylanase supplementation to wheat- or corn-based diets improved broilers performance and cecal microbiota composition. Xylanase supplementation to wheat-based diets increased the abundance of butyrate-producing bacteria and decreased the abundance of NSP-degrading bacteria. Moreover, positive effects of xylanase on cecal microbiota of broilers fed corn-based diets were mostly related to the inhibition of potentially pathogenic bacteria, and xylanase supplementation to corn-based diets slightly affected the abundance of butyrate-producing bacteria and NSP-degrading bacterium, the difference might be related to lower content of AX in corn compared to wheat.

The effect of a heat-stable xylanase on digesta viscosity, apparent metabolizable energy and growth performance of broiler chicks fed a wheat-based diet

Abstract:Feed costs represent a significant portion of the cost of poultry production. This study, in 3 experiments, was conducted to evaluate the effectiveness of a heat-stable xylanase (XYL) as a dietary supplement and its effect on digesta viscosity, nitrogen-corrected apparent metabolizable energy (AMEn), and live performance in broiler chicks. Experiment 1: the objective was to determine the effects of the amount and type of enzyme supplementation on digesta viscosity, AMEn, and bird performance using 7 diets. The dietary treatments were: no supplementation (C), 5 levels of XYL (1 to 16 ppm), or supplementation with a carbohydrase cocktail (CC). Experiment 2: the objective was to determine the interaction of the dietary XYL and the energy content of the feed. There were 2 levels of XYL (0 and 20 ppm) and 3 dietary energy levels (2,770, 2,920, and 3,070 kcal/kg ME). Experiment 3: the objective was to determine the interaction of the dietary XYL and feed form. The treatments were: 5 levels of XYL (0 to 40 ppm) and 2 feed forms (mash and crumble). Broiler chicks were reared in battery cages to 21 d. Statistical analysis of the data was completed using Proc GLM of SAS (9.2) (SAS Institute, Cary, NC). In experiment 1, increasing XYL (0 to 16 ppm) resulted in a decrease in digesta viscosity and an increase in AMEn. The XYL included as low as 1 ppm resulted in a significant increase in AMEn which reached 5% with 16 ppm XYL. In contrast, increase in BWG (4%) above values with the basal diet was greatest with 1 ppm XYL. In experiment 2, the caloric content of the diet influenced the increase in AMEn with inclusion of XYL, 8% and 6% increases with 2,920 kcal/kg and 3,070 kcal/kg diets, respectively. Without addition of XYL, BWG was significantly lower when fed the diet with the highest energy content. In experiment 3, feed form x XYL influenced the effect of XYL on BWG. The BWG was greater when birds were fed the crumble diet with XYL vs when they were fed the mash feed with XYL. The xylanase proved effective for broilers to 21 d when fed the diets used herein with changes in digesta viscosity, increased dietary AMEn, and improved bird performance represented by either BW gain or FCR.
Effect of endo-1,4-beta-xylanase supplementation to low-energy diets on performance, blood constituents, nutrient digestibility, and gene expressions related growth of broiler chickens

Abstract:The presence of soluble and insoluble non-starch polysaccharides (NSP) was reported to reduce nutrient utilisation, and adversely impact the broilers' growth performance; accordingly, NSP-degrading enzymes are essential supplements to cereal-based diets. Therefore, the current trial was conducted to characterise the impacts of supplemental xylanase (Xyl) to diets with low-ME levels on performance, carcass traits, blood parameters, nutrient digestibility and some genes expressions in broiler chickens. A total of 600 1-day-old Ross 308 male broiler chicks were randomly assigned to 6 treatments with 10 replications of 10 birds each per group in a completely randomised design. The 6 treatments were as follow: (1) basal diets with balanced ME content served as control (positive control, PC), (2) low-energy diet (negative control 1 [NC1]; ME content reduced by 70 kcal/kg compared with PC), (3) low-energy diet (negative control 2 [NC2]; ME content reduced by 140 kcal/kg compared with PC), (4) NC1 + 100 g/ton xylanase (NC1 + 100Xyl), (5) NC2 + 100 g/ton xylanase (NC2 + 100Xyl), and (6) NC1 + 50 g/ton xylanase (NC1 + 50Xyl). At the end of the experiment (35 days of age), the reduction of energy in the NC diets yielded lower live body weight (BW) and total body weight gain (BWG) (p ˂ 0.001); however, it significantly increased feed intake (p ˂ 0.05), leading to worst feed conversion ratio (FCR) and European production efficiency factor (EPEF) (p ˂ 0.01) than PC. There was non-significant variation in final BW, BWG, FCR, or EPEF between the PC group and the NC groups supplemented with Xyl. Carcass yield, gizzard, liver and, muscle relative weights were not influenced by dietary treatments; while broilers fed diet with low-energy diets with or without Xyl addition had lower abdominal fat (p ˂ 0.01) than PC. Furthermore, broilers fed on low-ME diets supplemented with Xyl showed a reduction in plasma total cholesterol (p ˂ 0.05) and low density lipoprotein (p ˂ 0.01) levels. Greater antibody titre against Newcastle disease (p ˂ 0.05) was recorded in the NC1 + 100Xyl and NC2 + 100Xyl groups. The addition of Xyl to low-energy diets significantly improved (p ˂ 0.05) fibre digestibility compared to the PC group. Moreover, enzyme supplementation increased muscle total lipids content and decreased muscle thiobarbituric acid retroactive substance content. In addition, enzyme supplementation increased gene expression related to growth and gene expression related to fatty acid synthesis. It was concluded that a low-ME diet might diminish broiler performance, whereas Xyl supplementation to low-ME diets beneficially affected growth performance, abdominal fat percentage, nutrient digestibility and immunity for broilers, and gene expressions related to growth and fatty acid synthesis in broiler chickens fed low-energy diets.
Effect of Β‐glucosidase on the Meat Quality and Digestibility in Broilers

Abstract:SummaryThis study investigated the effects of supplementary β‐glucosidase on the carcass composition, meat quality, weight of digestive organ and apparent digestibility in male broilers. Two hundred and forty male, 1‐day avine broiler chicks were randomly allocated into four treatment groups and fed with corn–soya bean meal supplemented with 0 (control), 0.6, 1.2 and 1.8 U/g of β‐glucosidase respectively. The results showed that there were no significant differences (p > 0.05) among groups in carcass composition (percentages of eviscerated yield, half‐eviscerated yield, muscle yield of breast and leg). However, adding 0.6 U/g β‐glucosidase to the diet not only altered the meat quality by decreasing the drip loss ratio (p < 0.05) and relative lightness (L*) value (p < 0.01), increasing relative redness (a*) value (p < 0.01), but also significantly decreased the pancreas to body weight ratio (p < 0.05), however, with little effect on liver, proventriculus and gizzard to body weight ratio (p > 0.05). The length and width of duodenum villus were not affected by the addition of β‐glucosidase, but the coefficients of total tract apparent digestibility of protein and fat increased by 9.02% (p < 0.05) and 7.40% (p < 0.01) respectively; the parameters of ash were not affected by β‐glucosidase addition (p < 0.05). This study provided valuable information for evaluation of the effect of supplementary β‐glucosidase on the meat quality and digestibility of broilers.
The Benefits of Exogenous Xylanase in Wheat–Soy Based Broiler Chicken Diets, Consisting of Different Soluble Non-Starch Polysaccharides Content

Abstract:Four wheat-based diets with either low soluble non-starch polysaccharide (NSPs, 13 g/kg) content (low viscosity, LV) or high NSPs content (33.5 g/kg; high viscosity, HV), without and with exogenous xylanase (XYL), were fed to male Ross 308 broiler chickens from 7 to 21 days age. The enzyme was supplemented at 100 FXU/kg diet, and its preparation was based on endo-1,4-beta-xylanase produced by Aspergillus oryzae. Each diet was fed to eight pens, with five birds in each pen, following randomisation. Chicks fed XYL had an improved feed efficiency, hepatic coenzyme Q10, cecal butyric acid concentration, nitrogen digestibility (p < 0.05) and increased dietary ME (p < 0.001). Compared to HV, birds fed LV diets had reduced weight of proventriculus, gizzard and the pancreas and higher blood glutathione peroxidase and dietary ME (p < 0.05), but no differences were observed on nutrient digestibility and growth performance variables. This also suggests that birds may tolerate a greater dietary NSPs content; thus, further benefits may be obtained by the application of XYL in low energy wheat-based diets.
Xylanase and xylo- oligosaccharide prebiotic improve the growth performance and concentration of potentially prebiotic oligosaccharides in the ileum of broiler chickens

Abstract:<span>1. The objective of this study was to investigate the effect of supplementing broiler diets with xylanase or xylo- oligosaccharide (XOS) on growth performance, the concentration of non-starch polysaccharide (NSP) hydrolysis products in the ileum and concentration of short chain fatty acids (SCFA) in the caeca of broiler chickens.2. In total, 500 male Ross 308 broilers were used in this 29-day (d) study. The treatments were organised into a 2 × 2 plus 1 factorial arrangement consisting of two additives (xylanase or XOS) at two levels (low or high) plus a control treatment with no additives. This gave five treatments with 100 birds in each treatment group. The diets were slightly deficient in protein by 20 g/kg and energy by 1 MJ/kg.3. On d 14 and 28, two birds per pen were euthanised, the caeca content collected and analysed for short chain fatty acid (SCFA) concentration. On d 29, six birds per pen were euthanised and ileal digesta were collected and analysed for the concentration of NSP fractions.4. On d 14, caecal acetic acid, iso-butyric acid, iso-valeric acid, n-valeric acid and total SCFA concentrations were significantly greater (P ≤ 0.05) when diets were supplemented with XOS compared with xylanase.5. Ileal concentration of arabinose, galactose and glucuronic acid (GlucA2) were significantly greater (P ≤ 0.05) in the insoluble NSP fraction when diets were supplemented with a high level of xylanase, compared with the control treatment. Ileal concentration of fructose was significantly greater (P ≤ 0.05) in the water soluble NSP when a high level of xylanase or low level of XOS were included in the diet compared with the control.6. It was concluded that xylanase and XOS had similar effects on NSP concentration and SCFA in the caeca, although there was little effect on performance. This observation demonstrated further benefits of xylanase supplementation in wheat-based broiler diets beyond digesta viscosity reduction and the release of extra nutrients.</span>

Abstract:Exogenous carbohydrases are commonly added to monogastric animal feed to degrade non-starch polysaccharides (NSP) to improve zootechnical performance and nutrient digestion. In the current study, the effects of dietary supplementation of a new, intrinsically thermostable, monocomponent xylanase on intestinal parameters, zootechnical performance and carcass traits of broilers were evaluated. A total of 720 1-day-old broilers were randomly allotted to four wheat-based diets with different levels of xylanase (T1: control; T2: 30,000 U/g; T3: 45,000 U/g; T4: 90,000 U/g) and fed from 1 to 35 days of age. The data showed that xylanase supplementation reduced the intestinal viscosity significantly, especially in the ileum. Duodenum and cecum pH was not influenced by xylanase supplementation, while T3 treatment showed the lowest pH value in jejunum and ileum. Xylanase supplementation reduced significantly feed conversion ratio (FCR) without affecting feed intake (FI). T2 treatment exhibited a higher body weight gain (BWG) compared the other treatments. Live weight, carcass weight, legs, and liver weight were significantly higher in T3 and T4 treatments compared to T1. No significant differences on foot pad lesions were observed among the four treatments. In conclusion, dietary supplementation of this new xylanase significantly improves intestinal tract viscosity and affects beneficially broilers’ performance and carcass traits.

Abstract:In a 67 year old patient with multiple cardiovascular risk factors a livedo reticularis, ischemic acral lesions as well as deterioration of renal function five weeks after cardiac catheterism and aorto-coronary bypass surgery led to suspicion of cholesterol embolism. Fundoscopy revealed cholesterol crystals in retinal vascular branches thus delivering important diagnostic information. Atheromatous lesions of the entire aorta and the ilio-femoral arteries were possible sources for embolism. The outcome was favorable. The lesions of the toes regressed and renal failure did not progress to dialysis. The diagnostic steps taken, the clinical picture of cholesterol embolism, the use of imaging and therapeutic options are discussed.
Changes in Growth Performance and Ileal Microbiota Composition by Xylanase Supplementation in Broilers Fed Wheat-Based Diets

Abstract:Xylanase exerts key roles in improving growth performance and intestinal health of broilers fed wheat-based diets. However, knowledge is limited regarding effects of xylanase supplementation on ileal microbiota in broilers. A total of 128 one-day-old broilers (initial BW 48.03 ± 0.33 g) were selected to investigate effects of xylanase (AT-xynA) on growth performance, ileal morphology, microbiota composition, immune response, antioxidant capacity, and endocrine peptide levels in broilers. Broilers were randomly allotted into two dietary treatments ( n = 8), namely, a wheat-soybean basal diet and a basal diet with 4,000 U/kg AT-xynA (XY). On days 7, 14, 21, and 42, broilers were weighted and ileal tissues were sampled. Ileal digesta samples were collected for analyzing microbiota composition on days 21 and 42. The results showed that AT-xynA could improve average daily weight gain and average daily feed intake, and there were interactions between diet and age of broilers ( p < 0.05). On days 21 and 42, xylanase supplementation decreased ileal microbiota α-diversity, and the relative abundance of potentially pathogenic microbiota, such as phylum Proteobacteria, family Moraxellaceae and Staphylococcaceae, genus Staphylococcus , Pseudomonas , Streptococcus , and Enterococcus , increased the abundance of Lactobacillus ( p < 0.05). Moreover, the reduction in acetate concentration and abundance of short-chain fatty acid-producing bacteria was also observed in broilers from XY group ( p < 0.05). AT-xynA increased ileal villus height, glucagon-like peptide-1, and insulin-like growth factor-1 concentrations and decreased interleukin-1β, interleukin-6, tumor necrosis factor-α, and malondialdehyde content in broilers, and these positive effects on intestinal health were greater in young broilers. In conclusion, xylanase supplementation to wheat-based diets could improve ileal intestinal morphology and immune function, and alleviate excess fermentation of bacteria, which may be related to changes of intestinal microbiota. In addition, the positive effects of xylanase on intestinal health were more pronounced in young broilers, thus contributing to subsequent improvement in growth performance of broilers.

Abstract:This paper discusses the structural difference and role of xylan, procedures involved in the production of xylooligosaccharides (XOS), and their implementation into animal feeds. Xylan is non-starch polysaccharides that share a β-(1-4)-linked xylopyranose backbone as a common feature. Due to the myriad of residues that can be substituted on the polymers within the xylan family, more anti-nutritional factors are associated with certain types of xylan than others. XOS are sugar oligomers extracted from xylan-containing lignocellulosic materials, such as crop residues, wood, and herbaceous biomass, that possess prebiotic effects. XOS can also be produced in the intestine of monogastric animals to some extent when exogenous enzymes, such as xylanase, are added to the feed. Xylanase supplementation is a common practice within both swine and poultry production to reduce intestinal viscosity and improve digestive utilization of nutrients. The efficacy of xylanase supplementation varies widely due a number of factors, one of which being the presence of xylanase inhibitors present in common feedstuffs. The use of prebiotics in animal feeding is gaining popularity as producers look to accelerate growth rate, enhance intestinal health, and improve other production parameters in an attempt to provide a safe and sustainable food product. Available research on the impact of xylan, XOS, as well as xylanase on the growth and health of swine and poultry, is also summarized. The response to xylanase supplementation in swine and poultry feeds is highly variable and whether the benefits are a result of nutrient release from NSP, reduction in digesta viscosity, production of short chain xylooligosaccharides or a combination of these is still in question. XOS supplementation seems to benefit both swine and poultry at various stages of production, as well as varying levels of XOS purity and degree of polymerization; however, further research is needed to elucidate the ideal dosage, purity, and degree of polymerization needed to confer benefits on intestinal health and performance in each respective species.

Abstract:This study aimed to investigate the effects of different levels of xylanase supplementation in a wheat-based diet on growth performance, short-chain fatty acids, intestinal health, microbial composition, and serum metabolism. A total of 1200 male chicks were randomly assigned to four wheat-based diet treatments: Group C (adding 0 mg/kg of xylanase), Group L (adding 50 mg/kg of xylanase), Group M (adding 100 mg/kg of xylanase), and Group H (adding 150 mg/kg of xylanase). The experiment lasted for 56 days. The results indicated that Group H broilers experienced a decreased feed-to-gain ratio throughout the study period. Additionally, dietary supplementation with xylanase led to an increase in the physical barrier, as indicated by increased VH and VH/CD in the gut (p < 0.05). Furthermore, levels of D-lactic acid and endotoxin were reduced. Xylanase supplementation also increased the abundance of Muc-2, ZO-1, and Occludin (p < 0.05). Moreover, xylanase supplementation enhanced the activity of sucrase and maltase in the duodenum (p < 0.05), which may be attributable to the upregulation of the abundance of SI and MGA (p < 0.05). Furthermore, xylanase addition promoted propionic acid produced by specific bacteria, such as Phascolarctobacterium, and influenced the microbial composition to some extent, promoting intestinal health. Additionally, 150 mg/kg of xylanase supplementation increased the amino acid, peptide, and carbohydrate content and upregulated the metabolism of amino acids related to histidine, cysteine, methionine, and other pathways (p < 0.05). These findings suggest adequate xylanase supplementation can enhance nutritional digestibility and absorption, improve growth performance, stimulate endogenous enzyme activity, optimize intestinal morphology and barrier function, and positively influence acid-producing bacteria and amino acid metabolic pathways.
Impact of fermentable fiber, xylo-oligosaccharides and xylanase on laying hen productive performance and nutrient utilization

Abstract:This study evaluated the impact of feeding xylo-oligosaccharides (XOS), fermentable fiber in the form of wheat bran (WB), and xylanase (XYL) on laying hen productive performance and nutrient digestibility. The hypothesis was that the WB would provide the microbiota in the hindgut with fermentable dietary xylan, and the XOS and XYL would further upregulate xylan fermentation pathways, resulting in improved nutrient utilization. Isa Brown hens (n = 96) were obtained at 39 wk of age. They were fed 12 dietary treatments, 8 hens per treatment, for 56 d. A commercial laying hen ration was fed, and for half of the treatments 10% of this ration was directly replaced with WB. The diets were then supplemented with either 1) no supplements; 2) XOS 50 g/t; 3) XOS 2000 g/t; 4) XYL (16,000 BXU/kg); 5) XYL + XOS 50 g/t, or 6) XYL + XOS 2,000 g/t. Hen performance and egg quality were measured every 14 d. On d56, ileum digesta samples were collected for determination of starch, nonstarch polysaccharide (NSP), XOS, protein, energy, and starch digestibility. Ceca digesta samples were also collected for analysis of XOS, short chain fatty acid (SCFA), xylanase and cellulase activity and microbial counts. Feeding 2,000 g/t XOS increased ileal protein digestibility. Combined 2,000 g/t XOS and XYL increased cecal Bifidobacteria concentration. This combination also increased cecal xylanase activity in birds fed the control diet. Cecal cellulase activity was improved by feeding WB, XYL, and 2,000 g/t XOS. XYL increased cecal lactate production. Feeding 2,000 g/t XOS with WB increased insoluble NSP degradability and shell breaking strength at d56. In summary, supplementing laying hen diets with fermentable fiber, XYL and XOS increases utilization of dietary xylan, improving nutrient utilization, performance, and gastrointestinal health.

Abstract:BACKGROUND Increased arterial stiffness has been shown to be associated with coronary heart disease (CHD). However, it remains unclear as to whether the second derivative of the finger photoplethysmogram (SDPTG), a non-invasive method for the assessment of arterial stiffness, is useful for the estimation of risk of CHD in the general population. METHODS AND RESULTS The SDPTG in 211 subjects (age: 63+/-15 years, range: 21-91 years, 93 males) was recorded without apparent atherosclerotic disorders from a community. The relationship between the SDPTG indices (b/a and d/a) and coronary risk factors (n=211) or the Framingham risk score (n=158, age: 60+/-12 years, range: 30-74 years, 63 males) were analyzed. The SDPTG indices significantly correlated with the Framingham risk score in both genders (b/a; r(male) =0.43, r(female) =0.54 and d/a; r(male) =-0.38, r(female) =-0.58), as well as several coronary risk factors. In the receiver operating characteristics curve analyses, the b/a discriminated high-risk subjects for CHD, who were in the highest quintile of the Framingham risk score in each gender, with a sensitivity and specificity of 0.85 and 0.58 in males and 0.83 and 0.72 in females, respectively. CONCLUSIONS These results suggest that the SDPTG is useful for the estimation of risk of CHD in the general population.
Effect of dietary supplementation of xylanase in a wheat-based diet containing canola meal on growth performance, nutrient digestibility, organ weight, and short-chain fatty acid concentration in digesta when fed to weaned pigs

Abstract:This study was designed to determine the effect of dietary supplementation of xylanase on growth performance, nutrient digestibility, organ weight, digesta pH, and concentration of short-chain fatty acids (SCFA) of weaned pigs fed wheat-canola meal (CM) diets over a 35-d period. A total of 144 piglets (72 barrows and 72 gilts) weaned at 18 ± 2 d of age, with initial body weight (BW) of 6.2 ± 0.7 kg, received one of eight dietary treatments based on randomized complete block design. BW and feed intake were recorded weekly to calculate average daily gain (ADG), average daily feed intake (ADFI), and feed efficiency (G:F). Treatments consisted of a control wheat-soybean meal-based diet and wheat-regular (RCM), dehulled (DCM), or coarse CM (CCM) without and with 16,000 BXU/kg xylanase (Econase XT). All diets contained 500 FTU/kg of phytase (Quantum Blue 5G) and titanium dioxide (0.3%). Apparent total tract digestibility (ATTD) of neutral detergent fiber (NDF), crude protein (CP), phosphorous (P), calcium (Ca), dry matter, and diet were determined. On day 35, one pig per pen was euthanized to evaluate the main factors of protein, xylanase supplementation, and sex on organ weight, ileal and colon digesta pH, and colon digesta concentrations of SCFA. The main factors did not affect growth performance. Xylanase supplementation improved nutrient digestibilities in all diets and increased ileal and colonic digesta pH without affecting the growth performance of weaned pigs fed wheat and CM-based diets. A protein-xylanase effect (P < 0.05) resulted in increasing the ATTD of NDF from 28% to 32% and from 29% to 37% for RCM and DCM, respectively. The ATTD of CP was greater (P < 0.05) with xylanase supplementation (75% vs. 70%). Xylanase supplementation increased ATTD of P and Ca. A three-way interaction (P < 0.05) for protein-xylanase-gender for colon pH, acetic, and propionic acid in the colon digesta of pigs indicated that, in addition to the protein source, piglet sex could have influenced how xylanase works. Xylanase supplementation increased (P < 0.05) the weight of the liver and spleen and tended (P < 0.10) to increase the size of the kidney. In conclusion, dietary supplementation of xylanase increased nutrient digestibility and digesta pH but did not influence the growth performance of weaned pigs fed wheat and CM-based diets over a 35-d period.
