Castleman ’ s Disease of the Retroperitoneum : With Special Reference to IgG 4-Related Disorder

Abstract:Localized Castleman’s disease (CD) has been divided two types, the classical hyaline vascular (HV) type and the rare plasma cell (PC) type. Recently, we have reported two cases of IgG4-related disorder of the retroperitoneum showing PC type of CD. To further clarify the clinicopathological findings of CD of the retroperitoneum, eight such cases have been studied. A single lesion was located in the retroperitoneum (n=3), ureter (n=2) and renal hilum (n=2). One case had bilateral ureter lesions. The HV type of CD accounts for approximately 90% of cases. However, 50% (n=4) of our cases were the PC type of CD. Three of the four lesions of HV type had lymph node lesions, whereas all four PC type of CD were soft tissue masses. These clinicopathologic findings appear quite different from previous descriptions. Immunohistochemical study demonstrated numerous IgG4 plasma cells accounting for more that 50% of IgG4 cells in three cases of the four PC type of CD. Moreover, serum IgG4 concentration was increased in two of the four cases of PC type of CD that were examined. The serum interleukin-6 levels were within the normal range in two cases of PC type that were examined. The present study suggests that a majority of the PC type of CD arising in the retroperitoneum appears to be an IgG4-related disorder. 〔J Clin Exp Hematopathol 50(1) : 39-44, 2010〕
Cold acclimation increases depolarization resistance and tolerance in muscle fibers from a chill-susceptible insect, Locusta migratoria

Abstract:Cold exposure depolarizes cells in insects due to a reduced electrogenic ion transport and a gradual increase in extracellular K+ concentration ([K+]). Cold-induced depolarization is linked to cold injury in chill-susceptible insects, and the locust, Locusta migratoria, has been shown to improve cold tolerance following cold acclimation through depolarization resistance. Here we investigate how cold acclimation influences depolarization resistance and how this resistance relates to improved cold tolerance. To address this question, we investigated if cold acclimation affects the electrogenic transport capacity and/or the relative K+ permeability during cold exposure by measuring membrane potentials of warm- and cold-acclimated locusts in the presence and absence of ouabain (Na+-K+ pump blocker) or 4-aminopyridine (4-AP; voltage-gated K+ channel blocker). In addition, we compared the membrane lipid composition of muscle tissue from warm- and cold-acclimated locust and the abundance of a range transcripts related to ion transport and cell injury accumulation. We found that cold-acclimated locusts are depolarization resistant due to an elevated K+ permeability, facilitated by opening of 4-AP-sensitive K+ channels. In accordance, cold acclimation was associated with an increased abundance of Shaker transcripts (gene encoding 4-AP-sensitive voltage-gated K+ channels). Furthermore, we found that cold acclimation improved muscle cell viability following exposure to cold and hyperkalemia even when muscles were depolarized substantially. Thus cold acclimation confers resistance to depolarization by altering the relative ion permeability, but cold-acclimated locusts are also more tolerant to depolarization.

Here we investigate how prolonged exposure to temperatures just above the supercooling point affects ion balance in freeze-avoiding mountain pine beetle (Dendroctonus ponderosae) larvae in autumn, mid-winter, and spring, and relate it to organismal recovery times and survival. Hemolymph ion balance was gradually disrupted during the first day of exposure, characterized by hyperkalemia and hyponatremia, after which a plateau was reached and maintained for the rest of the seven-day experiment. The degree of ionoregulatory collapse correlated strongly with recovery times, which followed a similar asymptotical progression. Mortality increased slightly during extensive cold exposures, where hemolymph K+ concentration was highest, and a sigmoidal relationship was found between survival and hyperkalemia. Thus, the cold tolerance of the freeze-avoiding larvae of D. ponderosae appears limited by the ability to prevent ionoregulatory collapse in a manner similar to chill-susceptible insects, albeit at much lower temperatures. Based on these results, we propose that a prerequisite for the evolution of insect freeze-avoidance may be a convergent or ancestral ability to maintain ion homeostasis during extreme cold stress.

Abstract:Low temperature causes most insects to enter a state of neuromuscular paralysis, termed chill coma. Susceptibility of insect species to enter chill coma is tightly correlated to the species distribution limits and for this reason it is important to understand the cellular processes that underlie chill coma. It is known that muscle function is markedly depressed at low temperature and this suggests that chill coma is partly caused by impairment in the muscle per se. To find the cellular mechanism(s) underlying muscle dysfunction at low temperature, we examined the effect of low temperature (5°C) on several events in the excitation-contraction-coupling in the migratory locust (Locusta migratoria). Intracellular membrane potential recordings during single nerve stimulations showed that 70% of fibers at 20°C produced an action potential (AP), while only 55% of the fibers were able to fire AP at 5°C. Reduced excitability at low temperature was caused by ∼80% drop in L-type Ca2+ current and a depolarizing shift in its activation of around 20 mV, which means that a larger endplate potential would be needed to activate the muscle AP at low temperature. In accordance we showed that intracellular Ca2+ transients were largely absent at low temperature following nerve stimulation. In contrast, maximum contractile force was unaffected by low temperature in chemically skinned muscle bundles which demonstrates that the function of the contractile filaments are preserved at low temperature. These findings demonstrate that reduced L-type Ca2+ current is likely the most important factor contributing to loss of muscle function at low temperature in locust.

Abstract:The arizonins, a novel complex of antibiotics related to kalafungin, were discovered in the fermentation broth of Actinoplanes sp. AB660D-122. Comparative taxonomic studies indicated that the culture is a new species and therefore has been designated Actinoplanes arizonaensis sp. nov. Two members of the complex, arizonins A1 and B1, exhibit moderate to potent in vitro antimicrobial activity against pathogenic strains of Gram-positive bacteria.

Abstract:Background: Many insect species have evolved the ability to survive extracellular freezing. The search for the underlying principles of their natural freeze tolerance remains hampered by our poor understanding of the mechanistic nature of freezing damage itself. Objectives: Here, in search of potential primary cellular targets of freezing damage, we compared mitochondrial responses (changes in morphology and physical integrity, respiratory chain protein functionality, and mitochondrial inner membrane (IMM) permeability) in freeze-sensitive vs. freeze-tolerant phenotypes of the larvae of the drosophilid fly, Chymomyza costata . Methods: Larvae were exposed to freezing stress at −30°C for 1 h, which is invariably lethal for the freeze-sensitive phenotype but readily survived by the freeze-tolerant phenotype. Immediately after melting, the metabolic activity of muscle cells was assessed by the Alamar Blue assay, the morphology of muscle mitochondria was examined by transmission electron microscopy, and the functionality of the oxidative phosphorylation system was measured by Oxygraph-2K microrespirometry. Results: The muscle mitochondria of freeze-tolerant phenotype larvae remained morphologically and functionally intact after freezing stress. In contrast, most mitochondria of the freeze-sensitive phenotype were swollen, their matrix was diluted and enlarged in volume, and the structure of the IMM cristae was lost. Despite this morphological damage, the electron transfer chain proteins remained partially functional in lethally frozen larvae, still exhibiting strong responses to specific respiratory substrates and transferring electrons to oxygen. However, the coupling of electron transfer to ATP synthesis was severely impaired. Based on these results, we formulated a hypothesis linking the observed mitochondrial swelling to a sudden loss of barrier function of the IMM.

Dietary potassium and cold acclimation additively increase cold tolerance in Drosophila melanogaster

Abstract:In the cold, chill susceptible insects lose the ability to regulate ionic and osmotic gradients. This leads to hemolymph hyperkalemia that drives a debilitating loss of cell membrane polarization, triggering cell death pathways and causing organismal injury. Biotic and abiotic factors can modulate insect cold tolerance by impacting the ability to mitigate or prevent this cascade of events. In the present study, we test the combined and isolated effects of dietary manipulations and thermal acclimation on cold tolerance in fruit flies. Specifically, we acclimated adult Drosophila melanogaster to 15 or 25 degrees C and fed them either a K+-loaded diet or a control diet. We then tested the ability of these flies to recover from and survive a cold exposure, as well as their capacity to protect transmembrane K+ gradients, and intracellular Na+ concentration. As predicted, cold-exposed flies experienced hemolymph hyperkalemia and cold-acclimated flies had improved cold tolerance due to an improved maintenance of the hemolymph K+ concentration at low temperature. Feeding on a high-K+ diet improved cold tolerance additively, but paradoxically reduced the ability to maintain extracellular K+ concentrations. Cold-acclimation and K+-feeding additively increased the intracellular K+ concentration, aiding in maintenance of the transmembrane K+ gradient during cold exposure despite cold-induced hemolymph hyperkalemia. There was no effect of acclimation of diet on intracellular Na+ concentration. These findings suggest intracellular K+ loading and reduced muscle membrane K+ sensitivity as mechanisms through which cold-acclimated and K+-fed flies are able to tolerate hemolymph hyperkalemia.

Glial voltage-gated K+ channels modulate the neural abiotic stress tolerance of Drosophila melanogaster

Abstract:Severe abiotic stress causes insects to lose nervous function and enter a state of paralytic coma. Central to this loss of function is a spreading depolarization (SD), where a characteristic collapse of ion gradients depolarizes neuronal and glial membranes and rapidly shuts down the CNS. Despite representing a critical limit to CNS function, the stress threshold that elicits SD can be altered by the process of acclimation, though the mechanisms underlying this response remain largely unknown. Here, we made electrophysiological measurements of SD and investigated the role of K+ channels in acclimation of the CNS stress response of Drosophila melanogaster. First, we demonstrate that improved cold tolerance in the CNS elicited by cold acclimation was abolished by pharmacological blockade of K+ channels with voltage-gated K+ channels representing most of this effect. Next, we used the UAS/Gal4 model system to screen for candidate genes encoding glial voltage-gated K+ channels and found that knockdown of sei- and Shaw-encoded channels mimicked the effect of K+ blockade in cold-acclimated flies. Furthermore we show that the knockdown of glial sei-encoded channels also impair tolerance to anoxia and heat stress. These findings suggest that voltage-gated K+ channels, especially those encoded by sei, are integral to the CNS stress- and acclimation-response and we posit that this is elicited through mechanisms involving glial spatial buffering and barrier function. Establishing such causal links between tissue-specific expression of candidate genes and SD mechanisms will inevitably aid our understanding of insect ecophysiology and SD-related neuropathologies.

P38 MAPK is a Component of the Signal Transduction Pathway Triggering Cold Stress Response in the MED Cryptic Species of Bemisia Tabaci

Abstract:Cold stress responses help insects to survive under low temperatures that would be lethal otherwise. This phenomenon might contribute to the invasion of some Bemisia tabaci cryptic species from subtropical areas to temperate regions. However, the molecular mechanisms regulating cold stress responses in whitefly are yet unclear. Mitogen-activated protein kinases (MAPKs) which including p38, ERK, and JNK, are well known for their roles in regulating metabolic responses to cold stress in many insects. In this study, we explored the possible roles of the MAPKs in response to low temperature stresses in the Mediterranean cryptic species (the Q-biotype) of the B. tabaci species complex. First, we cloned the p38 and ERK genes from the whitefly cDNA library. Next, we analyzed the activation of MAPKs during cold stress in the Mediterranean cryptic species by immuno-blotting. After cold stress, the level of phospho-p38 increased but no significant change was observed in the phosphorylation of ERK and JNK, thus suggesting that the p38 might be responsible for the defense response to low temperature stress. Furthermore, we demonstrated that: i) 3 min chilling at 0°C was sufficient for the activation of p38 MAPK pathway in this whitefly; and ii) the amount of phosphorylated p38 increased significantly in the first 20 min of chilling, reversed by 60 min, and then returned to the original level by 120 min. Taken together, our results suggest that the p38 pathway is important during response to low temperature stress in the Mediterranean cryptic species of the B. tabaci species complex.

Abstract:Insect cold survival has drawn increased attention in recent years, prompting a new classification of survival phenotypes that reflect the ecological and physiological differences in the manner in which insects survive the winter. In addition, more is known about the nature of cold injury and its effects on an insect system from the whole body to the subcellular level, although much remains to be discovered. Other than the detection of misfolded protein by the heat shock response, signal transduction of low temperatures by insects is largely unexplored. Recent advances in Drosophila and a few vertebrate systems reveal the potential importance of transient receptor potential ion channels in low temperature signal transduction, and membrane fluidity also possibly plays a role in the detection of low temperatures by insects. Physiological changes that occur in insect cells as a result of low temperatures or in anticipation of low temperatures include the induction of a suite of heat shock proteins, metabolic polyol synthesis, antifreeze protein production, and the induction of extracellular ice nucleation. In addition, a variety of new genes are now known to be induced upon cold exposure, but the roles of these molecules in cold survival are still largely unknown.

Abstract:There is a growing appreciation that insect distribution and abundance are associated with the limits of thermal tolerance, but the physiology underlying thermal tolerance remains poorly understood. Many insects, like the migratory locust (Locusta migratoria), suffer a loss of ion and water balance leading to hyperkalaemia (high extracellular [K+]) in the cold that indirectly causes cell death. Cells can die in several ways under stress, and how they die is of critical importance to identifying and understanding the nature of thermal adaptation. Whether apoptotic or necrotic cell death pathways are responsible for low-temperature injury is unclear. Here, we use a caspase-3 specific assay to indirectly quantify apoptotic cell death in three locust tissues (muscle, nerves and midgut) following prolonged chilling and recovery from an injury-inducing cold exposure. Furthermore, we obtain matching measurements of injury, extracellular [K+] and muscle caspase-3 activity in individual locusts to gain further insight into the mechanistic nature of chilling injury. We found a significant increase in muscle caspase-3 activity, but no such increase was observed in either nervous or gut tissue from the same animals, suggesting that chill injury primarily relates to muscle cell death. Levels of chilling injury measured at the whole animal level, however, were strongly correlated with the degree of haemolymph hyperkalaemia, and not apoptosis. These results support the notion that cold-induced ion balance disruption triggers cell death but also that apoptosis is not the main form of cell damage driving low-temperature injury.
Sex-dependent latent chilling injury changes estimates of thermal tolerance in a model insect

Abstract:Thermal tolerance limits are closely associated with insect distribution. Thermal injury sets limits to mobility or survival after removal from the stress, and these limits are frequently integrated into models describing or predicting climate suitability for species of interest. Cold stress severity, sex, and prior thermal acclimation status can all influence lower thermal limits. There is a growing understanding of chilling injuries initially manifest, but we poorly understand repair or further injury that may happen after rewarming. We exposed male and female to an acute or chronic cold stress before assessing their mobility over a 24 h period. Females progressively worsened under both conditions, but male mobility neither worsened nor improved. Female mobility declined slower in flies recovering at cooler temperatures, and cold acclimation significantly mitigated latent injury in females following the same degree of initial injury, regardless of recovery temperature. We conclude that latent chilling injury can be sex-specific, occurs independently from mechanisms driving tissue damage in the cold, is temperature-dependent, and is mitigated by prior thermal acclimation. We argue that latent chilling injury and the factors that influence it should be more carefully considered in estimating tolerance limits.

Insect fat body cell morphology and response to cold stress is modulated by acclimation

Abstract:Mechanistic understanding about the nature of cellular cryoinjury and mechanisms by which some animals survive freezing while others do not, is currently lacking. Here we exploited the broadly-manipulable freeze tolerance of larval malt flies (Chymomyza costata) to uncover cell and tissue morphological changes associated with freeze mortality. Diapause induction, cold acclimation, and dietary proline supplementation generate malt fly variants ranging from weakly to extremely freeze tolerant. Using confocal microscopy and immunostaining of the fat body, Malpighian tubules, and anterior midgut we described tissue and cytoskeletal (F-actin and α-tubulin) morphologies among these variants after exposure to various cold stresses (from chilling at -5°C to extreme freezing at -196°C), and upon recovery from cold exposure. Fat body tissue appeared to be the most susceptible to cryoinjury; freezing caused coalescence of lipid droplets, loss of α-tubulin structure, and apparent aggregation of F-actin. A combination of diapause and cold acclimation substantially lowered the temperature at which these morphological disruptions occurred. Larvae that recovered from a freezing challenge repaired F-actin aggregation but repaired neither lipid droplet coalescence nor α-tubulin structure. Our observations indicate that lipid coalescence and damage to α-tubulin are non-lethal forms of freeze injury, and suggest that repair or removal (rather than protection) of actin proteins is a potential mechanism of acquired freeze tolerance.

Abstract:Using the tropical crustacean Macrobrachium rosenbergii we investigate two popular hypotheses proposed to explain loss of function in ectotherms exposed to critically high and low temperatures. Specifically, we examine whether acute cold stress disrupts hemolymph and muscle ion balance or causes a loss of oxygen availability. We found that acute cold stress causes loss of righting response at 13 °C, but that a cold-induced loss of ion-balance only occurs after onset of mortality. In regards to oxygen availability, we found no decrease in hemolymph oxygen content during cold exposure, and no changes in the concentrations of the anaerobic end products l-lactate and succinate in the tail muscle of the shrimp. Therefore, our results support neither of these two popular hypotheses and it remains unknown what physiological perturbations determine the lower limits of thermal tolerance in Macrobrachium rosenbergii.
Mathematical modeling and analysis of the heat shock protein response during thermal stress in fish and HeLa cells

Abstract:The climate change has the potential to dramatically affect species' thermal physiology and may change the ecology and evolution of species' lineages. In this work, we investigated the transition of dynamics in the heat shock response when the thermal stress approaches the upper thermal limits of species to understand how the climate change may affect the heat shock responses in ectotherms and endotherms. The heat shock protein 70, HSP70, prevents protein denaturation or misfolding under thermal stresses. When thermal stress increases, the number of misfolded proteins increases, which leads to high levels of HSP70 protein. However, when temperatures approach limits of thermal tolerance (i.e., the critical thermal maximum, CTmax, for ectotherms and the superior critical temperature, SCT, for endotherms), levels of HSP70 protein synthesis start to decrease. Thus, we hypothesized that the temperature at the first reduction of HSP abundance indicates the thermal limits of the species. In this work, we provide a mathematical model to investigate the behavior of the heat shock responses related to HSP70 protein. This model captures the dynamics of HSP70 protein and Hsp70 mRNA, in HeLa cells (i.e., representative for endotherms) and multiple species of fishes (i.e., representative for ectotherms) with different acclimation histories. Based on our hypothesis of the relationship between the HSP70 protein level and CTmax/SCT, our model provides three methods to predict the CTmax of fishes with varying acclimation temperature and the SCT of HeLa cells. The CTmax increases as the acclimation temperature increases in fishes, however the CTmax plateaus when the acclimation temperature is higher than 20°C in brook trout, a representative cool water salmonid. Our model also captures the situation that the heat shock reaction in fish is more sensitive to the heat shock temperature than HeLa cells, when the heat shock temperature is lower than the upper thermal tolerance. However, both fish and HeLa cells are sensitive to the heat shock temperature when the temperature reaches the thermal tolerance limits. Additionally, our sensitive analysis result indicates that the status of some components in the heat shock reaction changes when the temperature reaches the thermal tolerance resulting in failure in protein refolding in fish and speeding up the refolding process in HeLa cells. Mathematical analysis is also applied on a simplified mathematical model to investigate the detailed dynamics of the model, such as the steady states of the substrate, Hsp70 mRNA, and HSP70 protein, at different thermal stresses. The comparison between the original model and simplified model shows that the inhibition on HSP70 protein transcription by thermal stresses leads to the reduction in HSP70 protein at extreme temperatures.

Abstract:antibody and rheumatoid factor were not found. Biochemical screening for viral hepatitis did not reveal antibodies against any type of viral hepatitis. Immunofixa-tion was normal and cryoglobulins were not detected. Histological evaluation of liver biopsy demonstrated macronodular cirrhosis, portal tract fibrosis and prolifer-ating biliary tracts with infiltrates of mainly T lymphocytes and few plasma cells. As an associated autoimmune dis-ease, Hashimoto’s thyroiditis with slightly elevated thyroid-stimulating hormone 3.9 mU/l (0.2–3.8) and thyroid autoantibodies 8 U/l (0–5) were found and a therapy with 50 g levothyroxine was initiated.I n our patient the diagnosis of AIH was made according to suggested parameters [4] . AIH is known to respond to steroid therapy with clinical and biochemical im-provement to normal liver parameters. Af-ter initiation of a corticosteroid therapy the patient recovered and all liver labora-tory parameters remained within normal limits. A 6-mg budesonide therapy was suggested but incompletely continued. However, for 2 years the controls showed GOT and GPT elevated up to 96 U/l ( ! 35 )and 100 U/l ( !3 5), respectively, but all oth-er laboratory parameters remained within normal limits. A utoimmune hepatitis (AIH) is a chronic autoimmune disease of the liver with unexplained etiology, potentially progressing to hepatic failure [2] . So far, AIH associated with neuropathy has been described in a few individual patients [3] . Here we describe a patient suffering from AIH who developed a motor-axonal poly-neuropathy. In 2004, a 61-year-old female Cauca-sian patient presented with an 8-kg weight loss during the preceding 3 months, fa-tigue, abdominal distension, yellow scler-ae, jaundice, pale stool, and dark urine. Abstract:The question of optimization of ion channel function to surrounding temperatures in poikilothermic organisms remains largely uninvestigated. Here, we addressed it by studying the temperature-dependence of L-type Ca2+ channels (LTCCs) in Drosophila larval muscles in the context of their modulation by protein kinase A (PKA). LTCC currents were recorded between 4 and 30°C. Different aspects of LTCC function reached maxima between 15 and 25°C: conductance, tail current amplitude, inactivation rate, and the level of basal up-regulation by PKA (26% at 21°C). Anomalous temperature-dependencies of LTCC conductance and kinetics were similar in control and in the presence of the PKA inhibitor H-89. Analysis of deactivation kinetics revealed excessive tail currents at lower temperatures (up to 15°C), indicative of voltage-dependent facilitation of LTCCs. Tail current magnitude gradually decreased with temperature from a maximum at 15°C until a nearly complete disappearance at 30°C. Elimination of excessive tail currents at higher temperatures coincided with unusual slowing of inactivation, suggesting disruption of the facilitation by rising temperature, possibly through depletion of the pool of contributing channels. Overall, these results suggest the presence of a physiological plasticity optimum of LTCC function in the temperature range of normal Drosophila development.

Abstract:Abstract Cold temperatures can be fatal to insects, but many species have evolved the ability to cold acclimate, thereby increasing their cold tolerance. While there is a growing body of knowledge concerning the mechanisms underlying cold tolerance, relatively little is known concerning how insects sense noxious cold (cold nociception), or how cold nociception might function in cold tolerance. It has been previously shown that Drosophila melanogaster larvae perform highly stereotyped, cold-evoked behaviors under the control of noxious cold-sensing neurons (nociceptors) innervating the barrier epidermis. In the present study, we first sought to describe cold-nociceptive behavior among 11 drosophilid species with differing cold tolerances and from differing climates. Behavioral analyses revealed that the predominant cold-evoked response among drosophilid larvae is a head-to-tail contraction (CT) behavior, which is likely inherited from a common ancestor. However, despite lack of phylogenetic signal (suggesting trait lability), the CT behavior was transient and there was no clear evidence that cold sensitivity was related to thermal environment; collectively this suggests that the behavior might not be adaptive. We therefore sought to uncover an alternative way that cold nociception might be protective. Using a combination of cold-shock assays, optogenetics, electrophysiology, and methods to genetically disrupt neural transmission, we demonstrate that cold sensing neurons in Drosophila melanogaster (Class III nociceptors) are sensitized by and critical to cold acclimation. Moreover, we demonstrate that cold acclimation can be optogenetically-evoked, sans cold. Collectively, these findings reveal that cold nociception constitutes a peripheral neural basis for Drosophila larval cold acclimation. Significance Statement Many insects adapt to cold in response to developmental exposure to cool temperatures. While there is a growing body of knowledge concerning the mechanisms underlying cold tolerance, it is unknown how sensory neurons might contribute. Here, we show that noxious cold sensing (cold nociception) is widely present among drosophilid larvae, and that cold-sensing neurons (Class III cold nociceptors) are necessary and sufficient drivers of cold acclimation. This suggests that cold acclimation has, at least in part, a neural basis.
