Abstract:Collagen-chitosan composite film modified with grapheme oxide (GO) and 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC), termed CC-G-E film, was loaded with basic fibroblast growth factor (bFGF) as the development of an efficacious wound healing device. In this study we report a novel drug delivery system that prevents the initial burst release and loss of bioactivity of drugs in vitro and in vivo applications. The results showed that CC-G-E film possessed improved thermal stability and a higher rate of crosslinking with increased mechanical properties when the dosage of GO was between 0.03% and 0.07%. It was shown that the in vitro release of bFGF from CC-G-E film continued for more than 28d. Furthermore, the CC-G-E films demonstrated excellent in vitro biocompatibility following culture with L929 fibroblasts in terms of cell adhesion and proliferation. CC-G-E films were implanted into Sprague-Dawley rats to characterize their ability to repair full-thickness skin wounds. Results showed that the CC-G-E film accelerated the wound healing process compared with the blank control. Based on all the results, it was concluded that CC-G-E film operates as a novel drug delivery system and due to its performance in wound remodeling, has potential to be developed as a wound dressing material.
Carboxymethyl chitosan-methacrylic acid gelatin hydrogel for wound healing and vascular regeneration

Abstract:At present, wound dressings in clinical applications are primarily used for superficial skin wounds.However these dressings have significant limitations, including poor biocompatibility and limited ability to promote wound healing. To address this issue, t this study used aldehyde polyethylene glycol as the cross-linking agent to design a carboxymethyl chitosan-methacrylic acid gelatin hydrogel with enhanced biocompatibility, which can promote wound healing and angiogenesis.The CSDG hydrogel exhibites acid sensitivity, with a swelling ratio of up to 300%. Additionally, it exhibited excellent resistance to external stress, withstanding pressures of up to 160 kPa and self-deformation of 80%. Compared to commercially available chitosan wound gels, the CSDG hydrogel demonstrates excellent biocompatibility, antibacterial properties, and hemostatic ability. Both in vitro and in vivo results showed that the CSDG hydrogel accelerated blood vessel regeneration by upregulating the expression of CD31, IL-6, FGF, and VEGF, thereby promoting rapid healing of wounds. In conclusion, this study successfully prepared the CSDG hydrogel wound dressings, providing a new approach and method for the development of hydrogel dressings based on natural macromolecules.

Abstract:The purpose of this study is to prepare polysaccharide-based hydrogel dressing integrated with microspheres which has excellent self-healing ability and sustained antibacterial function for wound healing application. The hydrogel is derived from carboxymethyl chitosan (CMCS) and carboxymethyl cellulose (CMC) via the Schiff’s base reaction, without the addition of chemical catalyst. Also, the CMCS microspheres containing an antibacterial drug, e.g., tetracycline hydrochloride (TH), were prepared by an emulsion cross-linking. Parameters were optimized during the preparation experiment to improve various properties of the microspheres/hydrogel composite dressing. This gel dressing has good elasticity, when compressive strain was 0.7 mm/mm, the tangent elastic modulus was up to 1.7 MPa. Furthermore, its drug sustained release performance was significantly better than the drug directly embedded in the hydrogel. TH could sustain release stably within 12 h, without obvious burst release phenomenon. Antibacterial experiments showed that 20 mg/mL microspheres made composite dressing a better performance than control microspheres and hydrogel. Potential of the composite scaffolds was further demonstrated by culture of human adipose-derived stem cells (ASCs) in vitro. Cell culture showed that this composite hydrogel could support survival and proliferation of ASCs. These characteristics provide a potential opportunity to use this composite gel dressing in drug delivery and wound healing applications.

Abstract:Chitosan and pectin are employed here in the formation of biodegradable hydrogels. The addition of cellulose nanowhiskers (CNW) into the hydrogels was accomplished through two methods: a) By swelling, after the polymeric network formation, and b) inclusion in polymer solutions before gelation, with varying CNW content. Comprehensive physico‐chemical, thermal, microscopic, gel fraction, and water vapor transmission analyses were conducted, complemented by antibacterial and viability assessments. CNWs were successfully synthesized and incorporated within the hydrogel by both methods. Moreover, the degradation temperature (Tmax) of hydrogels was higher with CNW (0% CNW: 205.5 °C, 10% CNW: 217.0 °C). CNW aggregates were observed in hydrogels with 20 and 30% content but no aggregation was observed with 4 and 10%. The antimicrobial properties were not affected by the addition of CNW, and the cytotoxicity tests revealed better results with 10% CNW compared with 20%. The study signifies the versatility of CNWs as effective reinforcements, influencing structural, thermal, and biological aspects of the hydrogels. The proposed methodology, emphasizing CNW incorporation, lays the groundwork for innovative biomaterials with promising medical implications. This investigation encourages further exploration and validation of these composite hydrogels in vivo for practical medical usage in wound dressings. This article is protected by copyright. Abstract:Gelatin (GA), hyaluronic acid (HA) and cellulose nanocrystals (CNC) are promising materials for skin wound care. In this study the GA-HA-CNC hydrogels were prepared by cross-linking and freeze-drying. The composition and mechanism of GA-HA-CNC hydrogels were confirmed by FTIR. The morphology and pore size were obtained by SEM. We accessed the physical property from rheological results and swelling ratio. NIH-3T3 cells were inoculated into the hydrogels and cultured for different days, then we analyzed the cytotoxicity of the prepared hydrogels by CCK-8 methods and live/dead pictorial diagram using staining kits. FTIR revealed the combination between GA, HA and CNC was attributed to the amide bond and hydrogen bonding. SEM results showed that the drying GA-HA-CNC hydrogels were spongy, with the pore diameter about 80–120 µm. CNC significantly enhanced the property of the hydrogels and play a vital role according to the rheology and swelling results. The cells culture results showed that NIH-3T3 cells can attached to, grow, and proliferate well on the GA-HA-CNC hydrogels. In conclusion, the natural GA-HA-CNC hydrogel has great potential for the skin wound repair.

Abstract:Human umbilical cord-mesenchymal stem cells (hUCMSCs) can secrete a variety of cytokines and growth factors promoting wound repair. Hydrogel is suitable biomaterial to supply niche for cells adhesion and survival. This study constructed a functional injectable thermo-sensitive hydrogel (chitosan/glycerol phosphate sodium/cellulose nanocrystals, CS/GP/CNC) encapsulated hUCMSCs to repair full-thickness cutaneous wound. Addition of CNC to the CS/GP system not only accelerated the gel speed, but also greatly improved the mechanical properties of the gel and decreased degradation rate. The novel hydrogel was injectable and low toxicity. Histological detection showed that hydrogel-hUCMSCs combination significantly accelerated wound closure, microcirculation, tissue remodeling, re-epithelialization and hair follicle regeneration, and inhibited over-inflammation in the central and surrounding wounds. The hydrogel-hUCMSCs combination promoted collagen deposition and keratinocyte mature marker K1 expression, decreased inflammatory factors secretion namely TNF-α and IL-1β. The present data provides a potential strategy for treatment of non-healing chronic cutaneous wounds.

Abstract:Recent years have demonstrated that biopolymers can serve as effective woundhealing agents. We hereby report chitosan-oxidized cellulose (CS.OMCC) based hydrogels loaded with eugenol with distinctive antimicrobial, biocompatible, and angiogenic characteristics. CS.OMCC hydrogels in four different ratios were synthesized through Schiff base (-CN) between chitosan (CS) and oxidized microcrystalline cellulose (OMCC). The swelling and degradation study analyzed the maximum absorption strength and weight loss of 1165% and 56.8 %, respectively. Hydrogels were potent against E. coli and S. aureus . The zones of inhibition were dependent upon the chitosan and eugenol, where 30 % eugenol (Eu) loaded CS.OMCC 4:1 hydrogel revealed greater zones (17 mm). The hydrogels demonstrated favorable biocompatibility, adhesion, and migration potential of mouse fibroblast cells. Moreover, the in-vivo potential by Chorioallantoic membrane (CAM) assay displayed an increase in the number, length, and size of blood vessels, contributing superior angiogenic potential. Increased expression in the case of VEGF further supported the role of hydrogels in promoting angiogenesis. Our study presents eugenol loaded chitosan-oxidized cellulose hydrogels that presented a promising solution to target bacterial infection and promote angiogenesis, thereby promoting the wound healing process.

Abstract:Wound dressing with an improved structural and functional recapitulation of damaged organs, efficient self-healing and antibacterial properties that can well integrate with tissue are urgently needed in wound management. Supramolecular hydrogels confer control over structural properties in a reversible, dynamic and biomimetic fashion. Herein, a kind of injectable, self-healing and antibacterial supramolecular hydrogel with multi-responses were fabricated by mixing phenylazo-terminated Pluronic F127, quaternized chitosan-graft-cyclodextrin and polydopamine coated tunicate cellulose nanocrystals under physiological conditions. By exploiting the photoisomerization of azobenzene under different wavelengths, a supramolecular hydrogel featuring a changing crosslink density of network was obtained. The corporation of polydopamine coated tunicate cellulose nanocrystals strengthens the hydrogel network with Schiff base bonds and hydrogen bonds, which avoids complete gel-sol transition. The inherent antibacterial property, drug release behavior, self-healing ability, hemostatic performance and biocompatibility were investigated to confirm superiority in wound healing. Moreover, the curcumin loaded hydrogel (Cur-hydrogel) showed multi-responsive release profiles (light, pH, and temperature). A full-thickness skin defect model was built to confirm that Cur-hydrogels significantly accelerated wound healing rate with better granulation tissue thickness and collagen disposition. Overall, the novel photo-responsive hydrogel with coherent antibacterial property has great potential in the healthcare of wound healing.
In situ injectable nano-composite hydrogel composed of curcumin, N,O-carboxymethyl chitosan and oxidized alginate for wound healing application

Abstract:In this paper, an in situ injectable nano-composite hydrogel composed of curcumin, N,O-carboxymethyl chitosan and oxidized alginate as a novel wound dressing was successfully developed for the dermal wound repair application. Nano-curcumin with improved stability and similar antioxidant efficiency compared with that of unmodified curcumin was developed by using methoxy poly(ethylene glycol)-b-poly(ε-caprolactone) copolymer (MPEG-PCL) as carrier followed by incorporating into the N,O-carboxymethyl chitosan/oxidized alginate hydrogel (CCS-OA hydrogel). In vitro release study revealed that the encapsulated nano-curcumin was slowly released from CCS-OA hydrogel with the diffusion-controllable manner at initial phase followed by the corrosion manner of hydrogel at terminal phase. In vivo wound healing study was performed by injecting hydrogels on rat dorsal wounds. Histological study revealed that application of nano-curcumin/CCS-OA hydrogel could significantly enhance the re-epithelialization of epidermis and collagen deposition in the wound tissue. DNA, protein and hydroxyproline content in wound tissue from each group were measured on 7th day of post wounding and the results also indicated that combined using nano-curcumin and CCS-OA hydrogel could significantly accelerate the process of wound healing. Therefore, all these results suggested that the developed nano-curcumin/CCS-OA hydrogel as a promising wound dressing might have potential application in the wound healing.

Abstract:Cutaneous T cell lymphoma (CTCL) is a generic classification of clonally‐derived malignancies of phenotypic helper/inducer T cells with a propensity to infiltrate the skin, migrate into the epidermis, localize in T cell zones of lymphoid structures and spare the bone marrow. One clinical presentation has a tendency to evolve into another as subclones of progressively less mature and less epidermotropic neoplastic cells arise and overgrow the relatively more mature other subclones. In this manner, localized, scaling plaques of typical parapsoriasis develop into more infiltrated plaques of classical mycosis fungoides which are themselves predecessors of tumor or erythroderma stage CTCL. With loss of epidermotropism, systemic dissemination occurs: first microscopically with blood involvement and seeding of internal organs and finally with visceral tumor formation. The relationship between CTCL and adult T cell lymphoma/leukemia is unclear because of tremendous overlap in the clinical and immunologic findings and because a fraction of patients with classical CTCL have low titers of anti‐HTLV‐1 antibodies. It is important to distinguish the skin‐limited and systemic phases of CTCL in order to select appropriate treatments. New diagnostic techniques which are quite helpful include immunotyping with monoclonal antibodies, karyotype analysis and T cell receptor studies. Photopheresis, a very promising new therapeutic approach for the management of patients with systemically disseminated disease, and a suggested pathogenetic scheme are discussed.

A Novel Composite Hydrogel Composed of Formic Acid-Decellularized Pepsin-Soluble Extracellular Matrix Hydrogel and Sacchachitin Hydrogel as Wound Dressing to Synergistically Accelerate Diabetic Wound Healing

Abstract:Extracellular matrix (ECM) hydrogel can create a favorable regenerative microenvironment and act as a promising dressing for accelerating the healing of diabetic wound. In this study, a simple and effective decellularization technique was developed and optimized to obtain acellular extracellular matrix (aECM) from porcine skin. It was found that decellularization at 30% formic acid for 72 h effectively decellularized porcine skin while retaining >75% collagen and ~37% GAG in the aECM with no presence of nuclei of cellular remnants. aECM hydrogel was fabricated by digesting aECM with pepsin in various acidic solutions (0.1 N HCl, glycolic acid (GA) and 2-pyrrolidone-5-carboxylic acid (PCA)) and then treated with a pH-controlled neutralization and temperature-controlled gelation procedure. Based on physical characterizations, including SDS-PAGE, rheological analysis and SEM analysis, aECMHCl hydrogels fabricated at 25 mg/mL in 0.1 N HCl were selected. Four polymeric ECM-mimic hydrogels, including sacchachitin (SC), hyaluronic acid (HA) and chitosan (CS) and three composite hydrogels of combining SC either with aECMHCl,25 (aECMHCl/SC), HA (HA/SC) or CS (SC/CS) were prepared and evaluated for WS-1 cell viability and wound-healing effectiveness. Cell viability study confirmed that no hydrogel dressings possessed any toxicity at all concentrations examined and ECMHCl, HA and ECMHCl/SC at higher concentrations (>0.05%) induced statistically significant proliferation. Diabetic wound healing study and histological examinations revealed that ECMHCl/SC hydrogel was observed to synergistically accelerate wound healing and ultimately stimulated the growth of hair follicles and sweat glands in the healing wound indicating the wound had healed as functional tissues. The results support the great potential of this newly produced ECMHCl/SC composite hydrogel for healing and regeneration of diabetic wounds.

Abstract:Delayed or chronic wound healing is one of severe clinical issues. Developing scaffold materials capable of supporting cells and inducing tissue regeneration remains a challenge. Here, a polysaccharide-based hydrogel is constructed for promoting full-thickness skin wound healing in mouse model. The engineering hydrogel consists of a dynamic crosslinking network formed by the Schiff base reaction between aldehyde-containing xyloglucan and methacrylated chitosan. Its reversible gel-sol-gel transition upon shearing force is highly beneficial to completely cover and fill irregular wound shape. The second covalent cross-linking network achieved by photo-initiated polymerization offers a feasible way to tune the mechanical property of hydrogel after injection, with an ideal mechanical adaptivity for clinical application. Remarkably, both in vitro and in vivo evaluations demonstrate that the hydrogel with endogenously bioactive galactoside units can promote cell spheroid formation and accelerate wound healing by expediting re-epithelialization, collagen deposition, angiogenesis as well as the formation of hair follicles.

Abstract:A novel multifunctional poly(γ-glutamic acid)/silk sericin (γ-PGA/SS) hydrogel has been developed and used as wound dressing. The physical and chemical properties of the γ-PGA/SS gels were systemically investigated. Furthermore, these γ-PGA/SS gels have been found to promote the L929 fibroblast cells proliferate, and in the in vivo study, significant stimulatory effects were also observed on granulation and capillary formation on day 9 in H-2-treated wounds, indicating that this new complex hydrogel could maintain a moist healing environment, protect the wound from bacterial infection, absorb excess exudates, and promote cell proliferation to reconstruct damaged tissue. Considering the simple preparation process and excellent biological property, this γ-PGA/SS hydrogel might have a wide range of applications in biomedical and clinical areas.

Abstract:CONTEXT: Growth factors act in an integrated manner to promote the wound-healing process. However, probably due to early inactivation of these molecules in the wound site, their topical administration scarcely leads to a significant improvement in chronic wound repair.OBJECTIVES: With the aim of identifying improved therapeutics, a sodium carboxymethyl chitosan-recombinant human epidermal growth factor conjugate (NaCMCh-rhEGF) was developed. It is believed that conjugation will protect rhEGF against proteolysis and will mediate rhEGF release by α-amylase.MATERIAL AND METHODS: As hydrogels possess most of the desirable characteristics of an ideal dressing, we used our previously described chitosan-based hydrogel as a carrier for NaCMCh-rhEGF nanoparticles to make a novel wound dressing system. To evaluate the biological activity of NaCMCh-rhEGF and free rhEGF, the proliferation of fibroblasts was measured using a colorimetric assay. Additionally the stability of conjugated and free rhEGF against proteases was estimated.RESULT AND DISCUSSION: In vitro results revealed that the conjugated form exhibited more stability against proteolysis and also preserved its biological activity. Furthermore, in vivo studies were performed using an excision wound model on diabetic rats. After 15 d, the wound area in NaCMCh-rhEGF-hydrogel dressing group was significantly smaller than other groups and showed histological parameters equal to positive wound control group.CONCLUSION: A polymer conjugated rhEGF was developed that was more stable against proteases and reserved the biological activity of the drug. This dressing appears to be a competent candidate for chronic wound healing.

Abstract:Wound healing is a complex project, and effectively promoting skin repair is a huge clinical challenge. Hydrogels have great prospect in the field of wound dressings because their physical properties are very similar to those of living tissue and have excellent properties such as high water content, oxygen permeability and softness. However, the single performance of traditional hydrogels limits their application as wound dressings. Therefore, natural polymers such as chitosan, alginate and hyaluronic acid, which are non-toxic and biocompatible, are individually or combined with other polymer materials, and loaded with typical drugs, bioactive molecules or nanomaterials. Then, the development of novel multifunctional hydrogel dressings with good antibacterial, self-healing, injectable and multi-stimulation responsiveness by using advanced technologies such as 3D printing, electrospinning and stem cell therapy has become a hot topic of current research. This paper focuses on the functional properties of novel multifunctional hydrogel dressings such as chitosan, alginate and hyaluronic acid, which lays the foundation for the research of novel hydrogel dressings with better performance.

Abstract:<p>Effective wound dressings are of great significance in preventing infections and promoting wound healing. However, most existing hydrogel dressings have an inadequacy in either mechanical performance, biological activities, or versatilities. Here we presented a double-network cross-linked polysaccharide-based hydrogel composed of collagen peptide-functionalized carboxymethyl chitosan (CS) and oxidized methacrylate sodium alginate (SA). The hydrogel possessed interconnected porous morphologies, suitable swelling ratios, excellent mechanical properties, and favorable biocompatibility. Meanwhile, the <em>in vivo</em> studies using a mouse full-thickness skin defect model showed that the double-network CS/SA hydrogel significantly accelerated wound healing by regulating the inflammatory process, promoting collagen deposition, and improving vascularization. Therefore, the functionalized double-network hydrogel should be a potential candidate as wound dressings</p>

Abstract:Hydrogel has emerged as one of the most promising dressings for wound healing owing to the creation of moist environment. However, it suffers from poor water retention capability and weak mechanical property. Therefore, development of dressings with durable moisture retention, robust mechanical property, and self-healing performance is urgently needed in practical applications. Herein, we developed a bilayered composite dressing consisting of breathable and hydrophobic polyurethane/polydimethylsiloxane (PU/PDMS) nanofibrous membrane and double-crosslinked chitosan-based hydrogel with dynamic Schiff base bonds and non-dynamic photo-crosslinking bonds. Combined with the nanofibrous membrane as protection and mechanical support, the bilayered hydrogel composites presented enhanced water retention capability to provide sustained moist environment, excellent stretchable and compressive performance to bear deformation, fast self-healing and good cyclic resilience to endure frequent movements. The in vivo study indicated their effective therapeutic effect in a rat model with full-thickness skin wounds. This study demonstrated that the hydrogel/nanofibrous membrane composite represented a promising candidate as dressing material for wound healing.

Abstract:The wound dressings' lack of antioxidant and antibacterial properties, and delayed wound healing limit their use in wound treatment and management. Recent advances in dressing materials are aimed at improving the limitations discussed above. Therefore, the aim of this study includes the preparation and characterization of oxidized hydroxyethyl cellulose (OHEC) and ferulic acid-grafted chitosan (CS-FA) hydrogel loaded with green synthesized selenium nanoparticles (Se NPs) (OHEC-CS-FA-Se NPs named as nanohydrogel) for diabetic wound healing. The structure and properties of the hydrogel was characterized by FTIR, FE-SEM, HR-TEM, EDAX, UV–Vis spectrophotometry, XRD, DLS, zeta potential and rheological studies. The findings of these experiments demonstrate that nanohydrogel possesses a variety of outstanding qualities, including an optimal gel time, good swelling characteristics, a fair water retention rate, a good degradation rate, and strong mechanical stability. Nanohydrogel has been shown to have a synergistic impact by significantly increasing antioxidant activity by scavenging ABTS and DPPH radicals. The nanohydrogel's strong biocompatibility was confirmed by cytocompatibility testing using L929 mouse fibroblast cells. In addition, the wound healing potential of nanohydrogel was tested on L929 cells by an in vitro scratch assay and the nanohydrogel showed a wound closure rate of 100 % after 12 h. In addition to this study, nanohydrogel has demonstrated significant antimicrobial properties against human and wound infection causing pathogens such as Bacillus subtilis , methicillin-resistant Staphylococcus aureus (MRSA), Escherichia coli , and Pseudomonas aeruginosa . In the animal model, almost complete diabetic wound healing was achieved on day 14 after application of the nanohydrogel. The results obtained indicate that the multifunctional bioactive nature of OHEC-CS-FA-Se NPs showed exceptional antioxidant and antibacterial potential for the treatment of infected and chronic wounds.
