Abstract:The objective of this study was to evaluate the effect of low dietary protein sources on the gut microbiota in finishing pigs. Thirty-six Duroc x Landrace x Large White finishing barrows were randomly allocated to four equal groups. Soybean meal (SBM) as a protein source is a control group and had a general protein level of 15% (SBM15). The pigs fed soybean meal (SBM11), soybean-cottonseed corn germ meal (SCCM11), and cottonseed-corn germ meal (CCM11) were offered low protein (11%) diets. The results showed that the ileum and feces microbiota of the SCCM11 group were significantly increased compared to the SBM15 group (P<0.05). At the phylum level, the relative abundance of Proteobacteria phyla was significantly increased in the ileum microbiota of the SCCM11 group compared to the SBM15 group (P<0.05). At the genus level, the abundance of Lactobacillus and Bifidobacterium showed a significant increase in the ileum microbiota of the SCCM11 group (P<0.05). The abundance of Ruminococcaceae, Phascolarctobacterium was significantly increased in the feces microbiota of the SCCM11 group than the other three groups (P<0.05). These results indicated that the SCCM11 group with added crystalline amino acid may modulate intestinal microbiota.

Abstract:This study was conducted to evaluate the effects of phytosterols (PS) and phytosterol esters (PSE) on C57BL/6 mice. Three groups of 34 six-week-old C57BL/6 mice of specific pathogen free (SPF) grade, with an average initial body weight (IBW) of 17.7g, were fed for 24 days either natural-ingredient diets without supplements or diets supplemented with 89 mg/kg PS or diets supplemented with 400 mg/kg PSE. Growth performance, blood biochemistry, liver and colon morphology as well as intestinal flora status were evaluated. Both PS and PSE exhibited growth promotion and feed digestibility in mice. In blood biochemistry, the addition of both PS and PSE to the diet resulted in a significant decrease in Total Cholesterol (TC) and Triglyceride (TG) levels and an increase in Superoxide Dismutase (SOD) activity. No significant changes in liver and intestinal morphology were observed. Both increased the level of Akkermansia in the intestinal tract of mice. There was no significant difference between the effects of PS and PSE. It was concluded that dietary PS and PSE supplementation could improve growth performance, immune performance and gut microbiome structure in mice, providing insights into its application as a potential feed additive in animals production.

Abstract:Lactobacillus overgrowth has been reported in patients with short bowel syndrome (SBS) (1–4). We previously reported D-lactic acidosis that Lactobacillus overgrowth caused in a male SBS patient (5). He had severe anemia related to vitamin B12 deficiency 9 years later. It has been generally accepted that B12 deficiency in SBS patients results from the inadequate absorption of B12 and/or the bacteria–host competition for uptake of B12 (6). In the present study, we suspected the recurrence of Lactobacillus overgrowth as a possible cause, because some lactobacilli require B12 for growth. Therefore, we analyzed the fecal flora and the B12 auxotrophy of Lactobacillus isolates.
Degree of Dietary Refinement Alters Apparent Gut Permeability

Abstract:Previous studies in our lab showed less refined diets reduced gut permeability (GI‐p) as assessed by 4hr plasma FITC‐dextran (FITC‐d) concentration in conjunction with reduced adiposity. To determine if degree of dietary refinement altered gut health and lipoprotein biology, we conducted a 14 week feeding trial in which weanling C57Bl/6 mice (n=15/diet) were fed one of four diets differing in dietary protein source (soy or milk protein) and degree of refinement. All diets provided 20%EN PRO, 30% EN fat, and 50% EN CHO. Purified diets contained either isolated soy protein (ISP) or dried whole milk powder (DWMP) as the protein source, while non‐purified diets contained either soybean meal (SPC), or dried whole milk powder (DMC) as protein sources. Purified diets contained 14% neutral detergent fiber (NDF) as cellulose, while non‐purified soy and milk protein diets provided 11.4% and 8.4% NDF, respectively, from wheat middlings, corn, corn gluten meal and soybean meal (SPC diet). All diets provided an average 3.3 %N as analyzed by Rapid N‐cube (Elementar, Germany). Mice fed purified diets were heavier than those fed non‐purified diets, with ISP‐fed mice being heaviest (p<0.033). Mice consumed similar amounts of energy in the different diets. Fecal %N was significantly increased in the non‐purified SPC‐ and DMC‐fed mice (2.99%, 2.98% respectively), being almost double that of ISP or DWMP (1.52%, 1.76%; p<0.000). At week 12, apparent GI‐p was highest in ISP‐fed mice (12.8 ng/mL), being significantly increased over DMC (8.96 ng/mL; p<0.02). Purified diet feeding resulted in increased apparent GI‐p, regardless of protein source (12.1 ng/mL vs. 9.56 ng/mL; p<0.004). Whole‐gut transit time averaged 2.5 hours longer for mice fed purified diets (6.6 hrs vs. 4.1 hrs, p<0.0001). Division of the value of 4 h FITC concentration used as the GI‐p index, by gut transit time, abolished differences due to diet purification; raising the possibility, that gut permeability per se did not differ among dietary groups. Consistent with this, cytokines (TNF‐a, IL‐1b, IL‐6, IL‐10) measured in whole blood and intestine explants were generally low and non‐indicative of either tissue or systemic inflammation. Density distributions of NBD‐stained lipoproteins showed that diets containing milk protein increased lipoproteins of d < 1.019 g/mL (TRL) (3.44% v. 5.4%) and HDL (d = 1.063 – 1.18 g/mL) (64.2% v. 60.6%) and decreased LDL (d=1.019–1.063 g/mL) (30.5% v. 36.0%, p<0.04). Compared to all other diets, mice fed ISP had a 30% increase in small dense LDL (d=1.05 – 1.063 g/mL) (p<0.000) and a ~ 4.0% decrease in HDL‐2b (d = 1.063–1.091 g/mL) (p<0.000). Increasing FITC‐d concentrations in plasma were significantly associated with decreases in %TRL (p<0.0127). In diets providing macronutrient distributions following the USDA's Dietary Guidelines for Americans, degree of purification seemingly has a greater effect on GI‐P, motility, and nitrogen utilization than protein source, while milk protein‐containing diets resulted in improved lipoprotein profiles regardless of purification. Abstract:Phytosterols, the plant analogues of cholesterol, widely occur in the human diet. In this study, we investigated and compared the effects of stigmasterol and beta-sitosterol (both with purities >= 95%) on dextran sulfate sodium (DSS)-induced colitis in C57BL/6J male mice fed a high fat Western-style diet. Mice treated with DSS developed severe mucosal colitis, with a marked distortion and crypt loss of colonic surface epithelium. Both beta-sitosterol and stigmasterol significantly inhibited colon shortening, lowered fecal hemoglobin content, and reduced the severity of colitis in the middle and distal colon (p < 0.05). These phytosterols also significantly suppressed the activation of nuclear factor-kappa B. They also significantly decreased colony stimulating factor-1 and the nuclear translocation of inflammatory master regulator nuclear factor-kappa B. Stigmasterol significantly lowered the colonic inflammation score and the expression of cyclooxygenase-2 and colony stimulating factor-1, while beta-sitosterol was less or not effective. These results suggest that dietary intake of stigmasterol and beta-sitosterol ameliorates colitis. Such activities of stigmasterol and beta-sitosterol in humans remain to be investigated.
Beef, Casein, and Soy Proteins Differentially Affect Lipid Metabolism, Triglycerides Accumulation and Gut Microbiota of High-Fat Diet-Fed C57BL/6J Mice

Abstract:Consumption of dietary protein at recommended levels is considered a potential strategy to promote satiety and weight management, but how protein from different dietary sources effect the obesity development, lipid metabolism, and gut microbiota is not known. This study focused on the effects of beef, casein, and soy protein diet on lipid metabolism, triglycerides accumulation, and microbial diversity in colon of C57BL/6J mice, which were given either low-fat diets (LFD, 12% Kcal) or high-fat diets (HFD, 60% Kcal) for 12 weeks. Body and liver weight increased significantly in mice fed a beef protein HFD (HFB), whereas reduced cumulative energy intake was seen in a soy protein HFD (HFS) group. HFB-fed mice showed signs of impaired glucose metabolism and insulin resistance along with a significant elevation in the concentration of triglycerides, LDL-cholesterol, total cholesterol, IL1β, TNF-α, IL-6, and leptin in serum. HFB also enhanced lipid accumulation in liver with increased activity of genes important for lipogenesis and hepatic cholesterol metabolism. A 16S rRNA gene sequencing indicated that HFD, regardless of proteins, significantly enhanced the ratio of Firmicutes to Bacteroidetes in colonic microbiota. However, HFB not only reduced the abundance of Akkermansia, compared with LFD independent of proteins, but also decreased the abundance of butyrate-producing bacteria such as Anaerotruncus, Butyricicoccus, and Lactobacillus (P < 0.05) compared with HFS and HFC. In conclusion, consumption of HFB does not only affect the gut microbiota composition but also increases the problems related to metabolic syndromes like dyslipidemia, hypercholesterolemia, and triglycerides accumulation in liver, which lead to systemic inflammation and its associated comorbidities, for example, impaired glucose metabolism and insulin resistance.

Protein Quality and the Protein to Carbohydrate Ratio within a High Fat Diet Influences Energy Balance and the Gut Microbiota In C57BL/6J Mice

Abstract:Macronutrient quality and composition are important determinants of energy balance and the gut microbiota. Here, we investigated how changes to protein quality (casein versus whey protein isolate; WPI) and the protein to carbohydrate (P/C) ratio within a high fat diet (HFD) impacts on these parameters. Mice were fed a low fat diet (10% kJ) or a high fat diet (HFD; 45% kJ) for 21 weeks with either casein (20% kJ, HFD) or WPI at 20%, 30% or 40% kJ. In comparison to casein, WPI at a similar energy content normalised energy intake, increased lean mass and caused a trend towards a reduction in fat mass (P = 0.08), but the protein challenge did not alter oxygen consumption or locomotor activity. WPI reduced HFD-induced plasma leptin and liver triacylglycerol, and partially attenuated the reduction in adipose FASN mRNA in HFD-fed mice. High throughput sequence-based analysis of faecal microbial populations revealed microbiota in the HFD-20% WPI group clustering closely with HFD controls, although WPI specifically increased Lactobacillaceae/Lactobacillus and decreased Clostridiaceae/Clostridium in HFD-fed mice. There was no effect of increasing the P/C ratio on energy intake, but the highest ratio reduced HFD-induced weight gain, fat mass and plasma triacylglycerol, non-esterified fatty acids, glucose and leptin levels, while it increased lean mass and oxygen consumption. Similar effects were observed on adipose mRNA expression, where the highest ratio reduced HFD-associated expression of UCP-2, TNFα and CD68 and increased the diet-associated expression of β3-AR, LPL, IR, IRS-1 and GLUT4. The P/C ratio also impacted on gut microbiota, with populations in the 30/40% WPI groups clustering together and away from the 20% WPI group. Taken together, our data show that increasing the P/C ratio has a dramatic effect on energy balance and the composition of gut microbiota, which is distinct from that caused by changes to protein quality.

Abstract:Different protein sources can impact gut microbiota composition and abundance, and also participate in health regulation. In this study, mice were gavaged with yeast protein (YP), soybean protein isolate (SPI), and whey protein isolate (WPI) for 28 days. Body weights showed similar patterns across different protein administration groups. The ileum in YP-supplemented mice exhibited good morphology, and tight-junction (TJ) proteins were slightly upregulated. Immunoglobulin (Ig)A, IgM, and IgG levels in the ileum of different protein groups were significantly increased (p < 0.05). Interleukin (IL)-10 levels were significantly increased, whereas IL-6 levels were significantly reduced in the YP group when compared with the control (C) (p < 0.05). Glutathione peroxidase (GSH-Px) levels in the ileum were significantly increased in the YP group (p < 0.05). These results indicate that YP potentially improved intestinal immunity and inflammatory profiles. The relative abundances of Parabacteroides, Prevotella, and Pseudobutyrivibrio in the YP group were more enriched when compared with the C and SPI groups, and Parabacteroides was significantly upregulated when compared with the WPI group (p < 0.05). Overall, the results indicate that YP upregulates the beneficial bacteria and improves ileal immunity and anti-inflammatory capabilities.

Purified diets containing high levels of soluble fiber and grain-based diets promote similar gastrointestinal morphometry yet distinct microbial communities

Abstract:Grain-based diets (GBDs) are widely used in rodent studies but their utility is limited due to batch-to-batch variability resulting from inconsistent ingredients. Purified diets (PDs) are composed of only known and refined ingredients and offer a solution to the constraints of GBDs. However, unlike GBDs, PDs commonly used as control diets typically contain little to no soluble fiber. We therefore sought to identify a combination of fibers in PDs that best recapitulates the gastrointestinal morphometry and intestinal microbial composition of mice fed GBDs. Adult male mice (n=30) were randomly assigned to one of six diets—two GBDs and four PDs with varying insoluble and soluble fiber composition—for 28 days. 16S rRNA gene sequencing was used to compare microbial profiles across different gastrointestinal (GI) niches and diets. Gut microbiotas and cecal weights were distinct between mice fed the two GBDs, indicating that GBDs are unreliable controls in diet-based studies. Unexpectedly, intestinal microbial richness decreased as the amount of soluble fiber in the PDs increased and the addition of multiple soluble fibers did not rescue this effect. Mice fed PDs with high soluble fiber content (≥ 75% of dietary fiber was soluble fiber) best recapitulated GI morphometry of mice fed GBDs, but intestinal microbial communities were distinct between PD- and GBD-fed mice. Although supplementing PDs with soluble fiber improved GI morphometry, further research to determine the optimal mixture of soluble and insoluble fibers is required to more closely mirror the intestinal microbial diversity observed in mice fed GBDs.

The aims of this study were to investigate the effect of oral colitis-extracted proteins (CEP) on dextran sulfate sodium (DSS)-induced colitis in BALB/c mice and to explore the relative role of the intestinal mucosal gamma delta T cells.The effect of five low oral doses of CEP on colitis was evaluated by clinical manifestation and histological lesions. Serum cytokines were measured by enzyme-linked immunosorbent assay. The percentages of the intestinal mucosal gamma delta T cells were evaluated by flow cytometry.CEP-fed colitis mice showed less severe symptoms and histological injury than bovine serum albumin (BSA)-fed control mice. Tolerized mice developed an increase in TGF-beta 1 and no change in IFN-gamma serum levels. Increases in TCR gamma delta(+) T cells and CD8 alpha+TCR gamma delta(+) T cells in small intestinal mucosal lymphocytes and no quantitative change in large intestinal mucosal lymphocytes were demonstrated in colitis mice compared to untreated mice. The proportions of TCR gamma delta(+) T cells and CD8 alpha+TCR gamma delta(+) T cells in large intestinal mucosal lymphocytes from CEP-fed colitis mice were significantly higher compared to BSA-fed controls. The disease activity index negatively correlated with the percentages of large intestinal mucosal gamma delta T cells. Furthermore, mucosal repair in repair-period mice was also accompanied by increases in TCR gamma delta(+) T cells and CD8 alpha+TCR gamma delta(+) T cells in large intestinal mucosal lymphocytes.Improvement of DSS-induced colitis that resulted from oral administration of colitis-extracted proteins is associated with an increase in gamma delta T cells in large intestinal mucosa.

Abstract:We investigated how protein quantity (10%–30%) and quality (casein and whey) interact with dietary fat (20%–55%) to affect metabolic health in adult mice. Although dietary fat was the main driver of body weight gain and individual tissue weight, high (30%) casein intake accentuated and high whey intake reduced the negative metabolic aspects of high fat. Jejunum and liver transcriptomics revealed increased intestinal permeability, low-grade inflammation, altered lipid metabolism, and liver dysfunction in casein-fed but not whey-fed animals. These differential effects were accompanied by altered gut size and microbial functions related to amino acid degradation and lipid metabolism. Fecal microbiota transfer confirmed that the casein microbiota increases and the whey microbiota impedes weight gain. These data show that the effects of dietary fat on weight gain and tissue partitioning are further influenced by the quantity and quality of the associated protein, primarily via effects on the microbiota.

Abstract:This work was to assess possible impacts of novel insoluble fiber 8% bacterial cellulose (BC), soluble fiber 8% konjac glucomannan (KGM), and their mixture (4% BC/4% KGM) on fatty acid metabolism and intestinal microbiota of C57BL/6J mice fed with a high-fat diet (HFD). HFD-fed mice receiving the dietary fibers (DFs) for 16 weeks exhibited an improvement in lipid-associated cytokines and a decrease in inflammation factors, which was associated with the improved hepatic and serum fatty acid composition. The DFs, notably the mixed BC/KGM, elevated the HFD-caused decrease in the contents of acetic acid (from 23.9 ± 0.85 to 32.2 ± 0.84 mM/g; p < 0.05), propionic acid (from 6.53 ± 0.28 to 12.8 ± 0.58 mM/g; p < 0.05), and butyric acid (from 7.73 ± 0.43 to 13.5 ± 0.47 mM/g; p < 0.05). Furthermore, the mixed BC/KGM significantly decreased the abundance of Firmicutes (from 90.4 to 67.6%) and Mucispirillum (from 4.77 to 1.58%) and dramatically increased the abundance of Bacteroidetes (from 7.83 to 25.0%) and Akkermansia (from 0.69 to 2.80%) in the gut of HFD-fed mice at the genus level. Moreover, correlation analysis revealed that the multiplicity of gut microbiota was useful in sustaining colonic integrity through producing short-chain fatty acids to some extent. This finding suggests that a mixture of insoluble BC and soluble KGM has positive effects on modulation of the intestinal microecosystem in mice.

Abstract:Bovine whey protein has been demonstrated to exert a positive effect on energy balance, lipid metabolism, and nutrient absorption. Additionally, it affects gut microbiota configuration. Thus, whey protein is considered as good dietary candidate to prevent or ameliorate metabolic diseases, such as obesity. However, the relationship that links energy balance, metabolism, and intestinal microbial population mediated by whey protein intake remains poorly understood. In this study, we investigated the beneficial effects attributed to whey protein in the context of high‐fat diet (HFD) in mice at two different ages, with short or longer durations of whey protein supplementation. Here, a 5‐week dietary intervention with HFD in combination with either whey protein isolate (WPI) or the control nonwhey milk protein casein (CAS) was performed using 5‐week or 10‐week‐old C57BL/6J mice. Notably, the younger mice had no prior history of ingestion of WPI, while older mice did. 5‐week‐old HFD‐WPI‐fed mice showed a decrease in weight gain and changes in the expression of genes within the epidydimal white adipose tissue including those encoding leptin, inflammatory marker CD68, fasting‐induced adipose factor FIAF and enzymes involved in fatty acids catabolism, relative to HFD‐CAS‐fed mice. Differences in β‐diversity and higher proportions of Lactobacillus murinus , and related functions, were evident within the gut microbiota of HFD‐WPI mice. However, none of these changes were observed in mice that started the HFD dietary intervention at 10‐weeks‐old, with an extended period of WPI supplementation. These results suggest that the effect of whey protein on mouse body weight, adipose tissue, and intestinal parameters depends on diet duration and stage of life during which the diet is provided. In some instances, WPI influences gut microbiota composition and functional potential, which might orchestrate observed metabolic and physiological modifications.

Abstract:Abstract Objectives Short chain fatty acids (SCFAs), which are gut microbial fermentation byproducts with suggested positive health effects, have emerged as a therapeutic modality against metabolic diseases like obesity and type 2 diabetes. Alluringly, in vivo SCFA levels are easily modifiable by consumption of fermentable fibers (FF). Most rodent studies on dietary FF supplementation report terminal increased cecal/fecal SCFA levels but the time course of this increase remains elusive. Also, there is limited information on the effect of this sustained SCFA increase on physiology. Thus, we investigated dietary FF-dependent temporal increases in plasma SCFA levels and its metabolic effects on a western diet (WD) mouse model. Methods C57BL/6J male mice (age 10 weeks) were fed test diets for 8 weeks. In Phase I, to establish time-course of plasma SCFA increase, mice were fed the following isocaloric diets: control (low fat + 0% FF); WD; control + 20% FF, where FF was fructooligosaccharides (FOS), inulin (In), guar gum (GG) or pectin (Pec). In Phase II mice were fed a control diet, or a WD with or without 20% FOS, Pec or GG. End points were weekly plasma SCFAs (by MS/MS), body weight, random glucose and insulin, and at the end of experimental period body fat composition and metabolic tests. Results Phase I. Compared to control, WD lowered while FF induced significant increases in total plasma SCFAs (FOS, Pec, GG > In) in a time dependent manner that plateaued beyond 2 weeks. All FF increased propionate and acetate but not butyrate. Phase II. WD caused metabolic dysfunctions (increased body weight and fat mass; glucose intolerance; insulin resistance; P < 0.0001 by two-way ANOVA) that were alleviated in mice fed FF enriched WD (Pec = GG > FOS). Compared to WD, food intake was similar except high in the WD-Pec group, while WD-Pec and GG showed higher energy expenditure. All 3 plasma SCFAs were significantly higher in all WD-FF groups. FF supplementation of the control diet showed no significant difference compared to control. Conclusions We conclude that: 1) FF feeding induced SCFA production reaches saturation within 2 weeks, suggesting selection of specific gut bacterial features; and 2) all FFs were protective from weight gain and its metabolic consequences. Abstract:PurposeThe impact of dietary nutrients on body growth performance and the composition of gut microbes and metabolites is well-established. In this study, we aimed to determine whether dietary protein can regulate the physiological indexes and changes the intestinal tissue morphology in rats, and if dietary protein was a crucial regulatory factor for the composition, function, and metabolic pathways of the gut microbiota.MethodA total of thirty male Sprague Dawley (SD) rats (inbred strain, weighted 110 ± 10 g) were randomly assigned to receive diets containing animal-based protein (whey protein, WP), plant-based protein (soybean protein, SP), or a blended protein (soybean-whey proteins, S-WP) for a duration of 8 weeks. To investigate the effects of various protein supplement sources on gut microbiota and metabolites, we performed a high throughput 16S rDNA sequencing association study and fecal metabolomics profiling on the SD rats. Additionally, we performed analyses of growth indexes, serum biochemical indexes, and intestinal morphology.ResultsThe rats in S-WP and WP group exhibited a significantly higher body weight and digestibility of dietary protein compared to the SP group (P < 0.05). The serum total protein content of rats in the WP and S-WP groups was significantly higher (P < 0.05) than that in SP group, and the SP group exhibited significantly lower (P < 0.05) serum blood glucose levels compared to the other two groups. The morphological data showed the rats in the S-WP group exhibited significantly longer villus height and shallower crypt depth (P < 0.05) than the SP group. The gut microbial diversity of the SP and S-WP groups exhibited a higher level than that of the WP group, and the microbiomes of the WP and S-WP groups are more similar compared to those of the SP group. The Arachidonic acid metabolism pathway is the most significant KEGG pathway when comparing the WP group and the SP group, as well as when comparing the SP group and the S-WP group.ConclusionThe type of dietary proteins exerted a significant impact on the physiological indices of SD rats. Intake of S-WP diet can enhance energy provision, improve the body’s digestion and absorption of nutrients, as well as promote intestinal tissue morphology. In addition, dietary protein plays a crucial role in modulating fecal metabolites by regulating the composition of the gut microbiota. Metabolomics analysis revealed that the changes in the levels of arachidonic acid metabolites and secondary bile acid metabolite induced by Clostridium_sensu_stricto_1 and [Eubacterium]_coprostanoligenes_group maybe the primarily causes of intestinal morphological differences.

Abstract:Whey protein isolate (WPI) is considered a dietary solution to obesity. However, the exact mechanism of WPI action is still poorly understood but is probably connected to its beneficial effect on energy balance, adiposity, and metabolism. More recently its ability to modulate the gut microbiota has received increasing attention. Here, we used a microbiota depletion, by antibiotic cocktail (ABX) administration, to investigate if the gut microbiota mediates the physiological and metabolic changes observed during high-fat diet (HFD)-WPI consumption. C57BL/6J mice received a HFD containing WPI (HFD-WPI) or the control non-whey milk protein casein (HFD-CAS) for 5 or 10 weeks. HFD-fed mice supplemented with WPI showed reduced body weight gain, adiposity, Ob gene expression level in the epidydimal adipose tissue (eWAT) and plasma leptin relative to HFD-CAS-fed mice, after 5- or 10-weeks intervention both with or without ABX treatment. Following 10-weeks intervention, ABX and WPI had an additive effect in lowering adiposity and leptin availability. HFD-WPI-fed mice showed a decrease in the expression of genes encoding pro-inflammatory markers (MCP-1, TNFα and CD68) within the ileum and eWAT, compared to HFD-CAS-fed mice, without showing alterations following microbiota depletion. Additionally, WPI supplementation decreased HFD-induced intestinal permeability disruption in the distal ileum; an effect that was reversed by chronic ABX treatment. In summary, WPI reverses the effects of HFD on metabolic and physiological functions through mainly microbiota-independent mechanisms. Moreover, we demonstrate a protective effect of WPI on HFD-induced inflammation and ileal permeability disruption, with the latter being reversed by gut microbiota depletion.
Dietary Milk or Isolated Legume Proteins Modulate Intestinal Microbiota Composition in Rats

Abstract:Shifts toward increased proteolytic fermentation, such as, for example, in athlete and high-protein weight loss diets, may alter the relative abundance of microbial species in the gut and generate bioactive, potentially deleterious metabolic products. In the current investigation, intestinal (caecal) microbiota composition was studied in rats fed diets differing only in their constituent proteins: milk (casein (CAS), lactalbumin (LA)) or legume (Cicer arietinum, Lupinus angustifolius) protein isolates (chickpea protein isolate (CPI), lupin protein isolate (LI)). ANOSIM and Discriminant Analysis showed significant (p < 0.05) differences at both family and genus levels in both microbiota composition and functionality as a consequence of feeding the different proteins. Differences were also significant (p < 0.05) for predicted functionality parameters as determined by PICRUSt analysis. LA induced a generally healthier microbiota composition than CAS, and higher amounts of Methanobrevibacter spp. and Methanogenic_PWY were found in the LI group. LEfSe analysis of bacterial composition and functional activities revealed a number of groups/functions able to explain the different effects found with milk and legume protein isolates. In conclusion, the mostly beneficial modulation of intestinal microbiota generally found with legume-based diets is likely to be due, at least in part, to their constituent proteins.

Abstract:Background: The current nutritional composition of the "American diet" (AD; also known as Western diet) has been linked to the increasing incidence of chronic diseases, including inflammatory bowel disease (IBD), namely Crohn disease (CD). Objectives: This study investigated which of the 3 major macronutrients (protein, fat, carbohydrates) in the AD has the greatest impact on preventing chronic inflammation in experimental IBD mouse models. Methods: We compared 5 rodent diets designed to mirror the 2011-2012 "What We Eat in America" NHANES. Each diet had 1 macronutrient dietary source replaced. The formulated diets were AD, AD-soy-pea (animal protein replaced by soy + pea protein), AD-CHO ("refined carbohydrate" by polysaccharides), AD-fat [redistribution of the ω-6:ω-3 (n-6:n-3) PUFA ratio; ∼10:1 to 1:1], and AD-mix (all 3 "healthier" macronutrients combined). In 3 separate experiments, 8-wk-old germ-free SAMP1/YitFC mice (SAMP) colonized with human gut microbiota ("hGF-SAMP") from CD or healthy donors were fed an AD, an AD-"modified," or laboratory rodent diet for 24 wk. Two subsequent dextran sodium sulfate-colitis experiments in hGF-SAMP (12-wk-old) and specific-pathogen-free (SPF) C57BL/6 (20-wk-old) mice, and a 6-wk feeding trial in 24-wk-old SPF SAMP were performed. Intestinal inflammation, gut metagenomics, and MS profiles were assessed. Results: The AD-soy-pea diet resulted in lower histology scores [mean ± SD (56.1% ± 20.7% reduction)] in all feeding trials and IBD mouse models than did other diets (P < 0.05). Compared with the AD, the AD-soy-pea correlated with increased abundance in Lactobacillaceae and Leuconostraceae (1.5-4.7 log2 and 3.0-5.1 log2 difference, respectively), glutamine (6.5 ± 0.8 compared with 3.9 ± 0.3 ng/μg stool, P = 0.0005) and butyric acid (4:0; 3.3 ± 0.5 compared with 2.54 ± 0.4 ng/μg stool, P = 0.006) concentrations, and decreased linoleic acid (18:2n-6; 5.4 ± 0.4 compared with 8.6 ± 0.3 ng/μL plasma, P = 0.01). Conclusions: Replacement of animal protein in an AD by plant-based sources reduced the severity of experimental IBD in all mouse models studied, suggesting that similar, feasible adjustments to the daily human diet could help control/prevent IBD in humans.
