Abstract:Field‐effect transistor‐based biosensors (FET biosensors) have gained significant attention in the context of the global SARS‐CoV‐2 pandemic owing to their label free and highly sensitive detection capabilities. In this comprehensive review, recent advances in the use of electrochemical‐transistor‐based biosensors for the label‐free detection of SARS‐CoV‐2 are analyzed. A general introduction to the electrochemical‐transistor‐based biosensor system is initiated, highlighting its critical technical requirements. Various structural designs and working principles of transistor‐based transducers are described, and the essential aspects of interface engineering for improved sensing performance are summarized. The applications of transistor‐based biosensors for the detection of SARS‐CoV‐2 are discussed in detail. Finally, perspectives for the future development of transistor‐based sensing systems are provided. This review aims to provide valuable insights and guidance for the design and optimization of biochemical sensor systems with the potential to impact health monitoring, disease diagnosis, and biosafety in the field of in vitro diagnostic products.

Abstract:Over the last few decades, field-effect transistor (FET)-based biosensors have demonstrated great potential across various industries, including medical, food, agriculture, environmental, and military sectors. These biosensors leverage the electrical properties of transistors to detect a wide range of biomolecules, such as proteins, DNA, and antibodies. This article presents a comprehensive review of advancements in the architectures of FET-based biosensors aiming to enhance device performance in terms of sensitivity, detection time, and selectivity. The review encompasses an overview of emerging FET-based biosensors and useful guidelines to reach the best device dimensions, favorable design, and realization of FET-based biosensors. Consequently, it furnishes researchers with a detailed perspective on design considerations and applications for future generations of FET-based biosensors. Finally, this article proposes intriguing avenues for further research on the topology of FET-based biosensors.

Abstract:The future of medical diagnostics calls for portable biosensors at the point of care, aiming to improve healthcare by reducing costs, improving access, and increasing quality – what is called the 'triple aim'. Developing point-of-care sensors that provide high sensitivity, detect multiple analytes, and provide real time measurements can expand access to medical diagnostics for all. Field-effect transistor (FET)-based biosensors have several advantages, including ultrahigh sensitivity, label-free and amplification-free detection, reduced cost and complexity, portability, and large-scale multiplexing. They can also be integrated into wearable or implantable devices and provide continuous, real-time monitoring of analytes in vivo, enabling early detection of biomarkers for disease diagnosis and management. This review analyzes advances in the sensitivity, parallelization, and reusability of FET biosensors, benchmarks the limit of detection of the state of the art, and discusses challenges and opportunities towards ultrasensitive, parallelized, and reusable FET biosensors for future healthcare applications.

Abstract:Two-dimensional (2d) materials have been used extensively in various fields due to their unique physical and chemical properties. Among their diverse applications, field-effect transistor biosensors (bio-FETs) promise a brilliant prospect in the fabrication of biodevices for diagnostics, especially point-of-care (PoC) based biomedical testing. The introduction of 2d nanomaterials as a sensing platform not only promotes their bioassay process but also provides label-free, low-cost, real-time, highly sensitive, and selective detection of different biomolecules. Several advantages make 2d-based bio-FETs valuable for biosensing applications, including their large effective surface area, environment-dependent conductivity, easy functionalization, and biocompatibility. This review focuses comprehensively on 2d-based bio-FETs and more on both dry and wet states. In this regard, these bio-FETs are classified into four categories including Apta, DNA (Geno), Immuno, and enzymatic biosensors. The configuration, detection mechanism, and electrical signal processing of each category are discussed in response to different biotargets. Finally, their promising roles as PoC testing devices for modern medicine are discussed in biomedical (real) samples.

Abstract:The vital utilization of biosensors in different domains has led to the design of much more precise and powerful biosensors, since they have the potential to attain information in a fast and simple manner compared to conventional assays. The present review describes the basic concepts, operation, and construction of biosensors and presented an ideology that choice of categorization, selection of immobilization method and advantages are crucial factors for an efficient and commercial biosensor. Amongst various biosensors, the field effect transistor (FET)-based biosensors have shown much more potential and immense advantages such as high detection ability and sensitivity for both neutral and charged biomolecules and, hence, have been explored comprehensively in the present review. This paper discusses the current challenges in device design by mainly focusing on the quantitative and qualitative performance parameters such as sensing surface properties, signal-to-noise ratio and various other factors, since consideration of these factors will eventually address the crucial concerns related to device design and practical limitations. The critical measures to translate the commercialization of biosensors in the market at a high pace have also been discussed. Hence, the discussion on device challenges illustrates that there is a scope of improvement in the areas such as short-channel effects, specificity and nanocavity filling factor for revolutionary advances in FET-based biosensors. Optimal selection of design rules and biosensing material has the potential to feature the next generation of biosensors. The present paper reports that following integrated multidisciplinary approaches and switching to nanotechnology in designing of FET-based biosensors can offer a lot of improvements in the practical key factors (such as low cost and reliability) and opportunities for the biosensors in the marketplace.

Abstract:An overview of interface engineering strategies to overcome the limitation of the Debye screening effect for the achievement of high‐performance FET‐based electronic devices is proposed, and their applications in biological environment analysis are systematically summarized. Promising advances in molecular medicine have promoted the urgent requirement for reliable and sensitive diagnostic tools. Electronic biosensing devices based on field‐effect transistors (FETs) exhibit a wide range of benefits, including rapid and label‐free detection, high sensitivity, easy operation, and capability of integration, possessing significant potential for application in disease screening and health monitoring. In this perspective, the tremendous efforts and achievements in the development of high‐performance FET biosensors in the past decade are summarized, with emphasis on the interface engineering of FET‐based electrical platforms for biomolecule identification. First, an overview of engineering strategies for interface modulation and recognition element design is discussed in detail. For a further step, the applications of FET‐based electrical devices for in vitro detection and real‐time monitoring in biological systems are comprehensively reviewed. Finally, the key opportunities and challenges of FET‐based electronic devices in biosensing are discussed. It is anticipated that a comprehensive understanding of interface engineering strategies in FET biosensors will inspire additional techniques for developing highly sensitive, specific, and stable FET biosensors as well as emerging designs for next‐generation biosensing electronics.

Abstract:The rise in antibiotic-resistant pathogens, highly infectious viruses, and chronic diseases has prompted the search for rapid and versatile medical tests that can be performed by the patient. Field-effect transistor (FET)-based electronic biosensing platforms are particularly attractive due to their sensitivity, fast turn-around time, potential for parallel detection of multiple pathogens, and compatibility with semiconductor manufacturing. However, an unmet critical need is a scalable, site-selective multiplexed biofunctionalization method with nanoscale precision for immobilizing different types of pathogen-specific bioreceptors on individual FETs, preventing parallel detection of multiple targets. Here, we propose a paradigm shift in FET biofunctionalization using thermal scanning probe lithography (tSPL) with a thermochemically sensitive polymer. This polymer can be spin-coated on fully-fabricated FET chips, making this approach applicable to any FET sensor material and technology. Crucially, we demonstrate the spatially selective multiplexed functionalization capability of this method by immobilizing different types of bioreceptors at prescribed locations on a chip with sub-20 nm resolution, paving the way for massively parallel FET detection of multiple pathogens. Antibody- and aptamer-modified graphene FET sensors are then realized, achieving ultra-sensitive detection of a minimum measured concentrations of 3 aM of SARS-CoV-2 spike proteins and 10 human SARS-CoV-2 infectious live virus particles per ml, and selectivity against human influenza A (H1N1) live virus.

Abstract:Field effect transistors (FETs)-based detection probes are powerful platforms for quantification in biological media due to their sensitivity, ease of miniaturization, and ability to function in biological media. Especially, FET-based platforms have been utilized as promising probes for label-free detections with the potential for use in real-time monitoring. The integration of new materials in the FET-based probe enhances the analytical performance of the developed probes by increasing the active surface area, rejecting interfering agents, and providing the possibility for surface modification. Furthermore, the use of new materials eliminates the need for traditional labeling techniques, providing rapid and cost-effective detection of biological analytes. This review discusses the application of materials in the development of FET-based label-free systems for point-of-care (POC) analysis of different biomedical analytes from 2018 to 2024. The mechanism of action of the reported probes is discussed, as well as their pros and cons were also investigated. Also, the possible challenges and potential for the fabrication of commercial devices or methods for use in clinics were discussed.

Abstract:This paper discusses the methods to enhancing the detection sensitivity of biosensors in three aspects: overcoming the influence of Debye shielding effects, promoting the binding efficiency of analytes and transistor surface, and improving the structures of device. Furthermore, the most commonly used definitions and representation parameters of sensitivity, including relative/absolute changes of output response and Detection limit (LoD), are reviewed. Biosensors based on field effect transistor (FET), also known as Bio‐FETs, offer numerous advantages, including high sensitivity, rapid response speed, low operating voltage, compact size, high reliability, and easily integrative. These features endow Bio‐FETs with tremendous performance in rapid analysis for samples with ultra‐low concentration at large‐scale, broadening their multiple potential applications. In recent years, the research based on Bio‐FETs has attracted more and more attention. A lot of effort is put into the challenge to improve their sensitivity, which can be influenced by various factors. This review focuses on the current approaches from multiple perspectives, including overcoming the influence of Debye shielding effects, promoting the binding efficiency of analytes and FETs surface, and improving the structures of FETs device. Correspondingly, novel strategies to increase the Debye length, facilitate high frequency electric field modulation, enhance the specific interaction efficiency, and enlarge the specific surface area (SBET) are also summarized. Additionally, there is a lack of a reliable standard method to characterize the sensitivity of FETs currently. Regarding this issue, the most commonly used definitions and representation parameters of sensitivity are also evaluated. Overall, this review outlines the potential future research directions to offer insights for enhancing the sensitivity of Bio‐FETs.

Abstract:The rapid advancement of wearable biosensors has revolutionized healthcare monitoring by screening in a non-invasive and continuous manner. Among various sensing techniques, field-effect transistor (FET)-based wearable biosensors attract increasing attention due to their advantages such as label-free detection, fast response, easy operation, and capability of integration. This review explores the innovative developments and applications of FET-based wearable biosensors for healthcare monitoring. Beginning with an introduction to the significance of wearable biosensors, the paper gives an overview of structural and operational principles of FETs, providing insights into their diverse classifications. Next, the paper discusses the fabrication methods, semiconductor surface modification techniques and gate surface functionalization strategies. This background lays the foundation for exploring specific FET-based biosensor designs, including enzyme, antibody and nanobody, aptamer, as well as ion-sensitive membrane sensors. Subsequently, the paper investigates the incorporation of FET-based biosensors in monitoring biomarkers present in physiological fluids such as sweat, tears, saliva, and skin interstitial fluid (ISF). Finally, we address challenges, technical issues, and opportunities related to FET-based biosensor applications. This comprehensive review underscores the transformative potential of FET-based wearable biosensors in healthcare monitoring. By offering a multidimensional perspective on device design, fabrication, functionalization and applications, this paper aims to serve as a valuable resource for researchers in the field of biosensing technology and personalized healthcare.

Abstract:During recent years, field-effect transistor biosensors (Bio-FET) for biomedical applications have experienced a robust development with evolutions in FET characteristics as well as modification of bio-receptor structures. This review initially provides contemplation on this progress by briefly summarizing remarkable studies on two aforementioned aspects. The former includes fabricating unprecedented nanostructures and employing novel materials for FET transducers whereas the latter primarily synthesizes compact molecules as bio-probes (antibody fragments and aptamers). Afterwards, a future perspective on research of FET-biosensors is also predicted depending on current situations as well as its great demand in clinical trials of disease diagnosis. From these points of view, FET-biosensors with infinite advantages are expected to continuously advance as one of the most promising tools for biomedical applications.

Abstract:Abstract Cancer is an incurable disease, and the treatment process is extremely painful. Early detection may ease the treatment process and prevent cancer from spreading beyond the primary disease area. However, conventional screening tests have long detection times and lack the required sensitivity for early detection. Consequently, traditional cancer biosensors, including amplification refractory mutation system, digital polymerase chain reaction, next generation sequencing, western blot, electrochemical, and mechanical biosensors, have been studied in recent years. Specifically, field effect transistor (FET) biosensors, are attractive pocketable devices with short detection time capabilities. Because FET biosensors have outstanding electrical and mechanical properties, FET biosensors have been studied for their efficacy in the early detection of cancer. Traditional detection methods of cancer biomarkers include the use of FET biosensors for the detection of cancer biomarkers, especially gene, antigen, and protein characteristics. This review presents the latest strategies in FET applications in cancer biosensing and compares their advantages and disadvantages regarding sensing principle, configuration, and performance. Especially, FET biosensors for the detection of cancer biomarkers, which include antibodies, nucleic acids, proteins are highlighted. Mechanical and electrical properties of FET devices and their effect on performance is discussed. This review provides a guiding role in the design and development of FET-based biosensors.

Abstract:The precise monitoring of environmental contaminants and agricultural plant stress factors, respectively responsible for damages to our ecosystems and crop losses, has nowadays become a topic of uttermost importance. This is also highlighted by the recent introduction of the so-called "Sustainable Development Goals" of the United Nations, which aim at reducing pollutants while implementing more sustainable food production practices, leading to a reduced impact on all ecosystems. In this context, the standard methods currently used in these fields represent a sub-optimal solution, being expensive, laboratory-based techniques, and typically requiring trained personnel with high expertise. Recent advances in both biotechnology and material science have led to the emergence of new sensing (and biosensing) technologies, enabling low-cost, precise, and real-time detection. An especially interesting category of biosensors is represented by field-effect transistor-based biosensors (bio-FETs), which enable the possibility of performing in situ, continuous, selective, and sensitive measurements of a wide palette of different parameters of interest. Furthermore, bio-FETs offer the possibility of being fabricated using innovative and sustainable materials, employing various device configurations, each customized for a specific application. In the specific field of environmental and agricultural monitoring, the exploitation of these devices is particularly attractive as it paves the way to early detection and intervention strategies useful to limit, or even completely avoid negative outcomes (such as diseases to animals or ecosystems losses). This review focuses exactly on bio-FETs for environmental and agricultural monitoring, highlighting the recent and most relevant studies. First, bio-FET technology is introduced, followed by a detailed description of the the most commonly employed configurations, the available device fabrication techniques, as well as the specific materials and recognition elements. Then, examples of studies employing bio-FETs for environmental and agricultural monitoring are presented, highlighting in detail advantages and disadvantages of available examples. Finally, in the discussion, the major challenges to be overcome (e.g., short device lifetime, small sensitivity and selectivity in complex media) are critically presented. Despite the current limitations and challenges, this review clearly shows that bio-FETs are extremely promising for new and disruptive innovations in these areas and others.

Abstract:Biosensors based on field-effect transistors (FETs) are suitable for use in miniaturized and cost-effective healthcare devices. Various semiconductive materials can be applied as FET channels for biosensing, including one- and two-dimensional materials. The signal transduction interface between the biosample and the channel of FETs plays a key role in translating electrochemical reactions into output signals, thereby capturing target ions or biomolecules. In this Review, distinctive signal transduction interfaces for FET biosensors are introduced, categorized as chemically synthesized, physically structured, and biologically induced interfaces. The Review highlights that these signal transduction interfaces are key in controlling biosensing parameters, such as specificity, selectivity, binding constant, limit of detection, signal-to-noise ratio, and biocompatibility.

Abstract:The emergence of rapidly expanding infectious diseases such as coronavirus (COVID-19) demands effective biosensors that can promptly detect and recognize the pathogens. Field-effect transistors based on semiconducting two-dimensional (2D) materials (2D-FETs) have been identified as potential candidates for rapid and label-free sensing applications. This is because any perturbation of such atomically thin 2D channels can significantly impact their electronic transport properties. Here, we report the use of FET based on semiconducting transition metal dichalcogenide (TMDC) WSe2 as a promising biosensor for the rapid and sensitive detection of SARS-CoV-2 in vitro. The sensor is created by functionalizing the WSe2 monolayers with a monoclonal antibody against the SARS-CoV-2 spike protein and exhibits a detection limit of down to 25 fg/μL in 0.01X phosphate-buffered saline (PBS). Comprehensive theoretical and experimental studies, including density functional theory, atomic force microscopy, Raman and photoluminescence spectroscopies, and electronic transport properties, were performed to characterize and explain the device performance. Glass adherence, random migration, response to MIF and killing of Salmonella typhimurium and Saccharomyces cerevesiae by guinea pig macrophages were decreased with temperatures below 37 degrees C. Killing of Sacch. cerevesiae by human macrophages was also reduced at temperatures less than 37 degrees C. Acid phosphatase and beta-N-acetylglucosaminidase (NAG) activity assayed at 37 degrees C did not change when the cells were preincubated for 1 and 5 hours at various temperatures. Impaired macrophage function with subnormal temperatures may contribute to enhanced susceptibility to infection of patients with chronic diseases such as renal failure and cirrhosis.

Abstract:This work reports flexible electrolyte‐gated field‐effect transistor (EGFET) based biosensors for rapid severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) detection with ultra‐high sensitivity, where the Indium Zinc Oxide (IZO) doping with polyethylenimine (PEI) serves as the semiconductor and functionalized with a specific antibody to access the presence of SARS‐CoV‐2 S1 proteins in virus. The pandemic of coronavirus disease 2019 (COVID‐19) reflects the great significance of rapid and accurate detection of pathogens by new sensing technologies. Antibody based biosensors with high sensitivity comparable to golden standard polymerase chain reaction (PCR) and miniaturized device features allow the detection of pathogens in portable and flexible formats. Herein, flexible metal oxide electrolyte‐gated field‐effect transistors (EGFETs) are reported to serve as the biosensors for rapid and ultrasensitive severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) detection. The semiconducting layer of the EGFETs associates with hybrid material of PEI doped metal oxides that not only improves the transistor performance, but also regulates microstructure forming higher surface‐to‐volume ratio, which brings more antibodies immobilization, resulting in higher sensitive, and faster response for detecting SARS‐CoV‐2. Comprehensive studies of materials and interfacing engineering of the EGFETs not only build the strong foundation for the EGFET sensors to show excellent sensitivity with a limit of detection from 0.14 fg ml−1 for SARS‐CoV‐2 S1 proteins, and 0.09 copies μl−1 for SARS‐CoV‐2 viruses, but also offer good mechanical properties to enable thin, soft flexible sensing platforms. This work provides a new strategy from materials to devices as innovative schemes for virus/pathogens detection.

Abstract:This review summarizes recent advances in graphene field effect transistor (GFET)‐based biosensors, focusing on i) fundamental concepts of FETs and the specific electrical properties of GFETs relevant to biosensing; ii) recent schemes to improve the performance of GFET biosensors from several aspects, such as the principle of operation and stability, sensitivity, and specificity; and iii) multi‐detection strategies for GFET biosensors. Recently, field‐effect transistors (FETs) have emerged as a novel type of multiparameter, high‐performance, highly integrated platform for biochemical detection, leveraging their classical three‐terminal structure, working principles, and fabrication methods. Notably, graphene materials, known for their exceptional electrical and optical properties as well as biocompatibility, serve as a fundamental component of these devices, further enhancing their advantages in biological detection. This review places special emphasis on recent advancements in graphene field‐effect transistor (GFET)‐based biosensors and focuses on four main areas: i) the basic concepts of FETs and the specific electrical properties of GFETs; ii) various state‐of‐the‐art approaches to enhance the performance of GFET‐based biosensors in terms of operating principles and the "3S"—stability, sensitivity, and specificity; iii) multiplexed detection strategies for GFET‐based biosensors; and iv) the current challenges and future perspectives in the field of GFET‐based biosensors. It is hoped that this article can profoundly elucidate the development of GFET biosensors and inspire a broader audience.
