Abstract:A sensitive HPLC method with simple extraction was developed for simultaneous determination of huperzine A (HupA) and huperzine B (HupB) in Huperzia serrata, H. crispata, H. miyoshiana, and Lycopodiastrum casuarinoides. In order to avoid conventional multiple-step and time-consuming sample preparation methods, direct reflux extraction with alkaline chloroform was adopted. The quantitative determination was conducted by reversed-phase HPLC with a photodiode array detector set at 308 nm. Separation was performed on a Luna C18 column (250 x 4.6 mm id, 5 pm) with methanol-0.2% aqueous acetic acid (18 + 82, v/v) mobile phase. The method was validated for accuracy, reproducibility, precision, and limits of detection and quantification. Quantification of the two active compounds in the samples was performed by this newly developed method, and the content of HupA and HupB varied substantially among four different species. The satisfactory results indicated that the developed method can readily be utilized for quality control of the species of Huperziaceae and Lycopodiaceae containing the two compounds.
Separation and Determination of Coumarins in the Root Bark of Three Citrus Plants by Micellar Electrokinetic Capillary Chromatography.

Abstract:A micellar electrokinetic capillary chromatography (MEKC) method for the determination of xanthyletin and xanthoxyletin in the roots bark of three Citrus plants was developed for the first time. The electrophoresis buffer was 10 mmol/L sodium borate containing 50 mmol/L sodium dodecyl sulfate, 20% (v/v) acetonitrile and 10 % (v/v) methanol (pH 9.8). The correlation coefficients of the calibration curves for xanthyletin and xanthoxyletin are 0.9993 and 0.9999, respectively, over the concentration ranges (16.0-800.0 mg/L; 8.0-400.0 mg/L).

Abstract:A simple and sensitive micellar electrokinetic capillary chromatography (MEKC) method was developed for the separation and determination of six flavonoids in Epimedium brevicornum Maxim. Field-enhanced sample injection with reverse migrating micelles (FESI-RMM) was used for on-line concentration of the flavonoids. An electrolyte containing 20 mM H3PO4, 100 mM SDS, 20% acetonitrile and 2% 2-propanol (pH 2.0) was chosen as the electrophoretic buffer. By optimizing the stacking conditions, about 40-360-fold improvement in the detection sensitivity was obtained for the flavonoids.

Abstract:The simultaneous determination of 20-hydroxyecdysone (1), 25-deoxy-11,20-dihydroxyecdysone (2), kaempferol 4,7-dimethyl ether (3) and biochanin A (4) in the fruit, leaf and root of Serratula strangulata Iljin has been investigated by micellar electrokinetic capillary chromatography for the first time. With an electrolyte containing 15 mmol/L borate, 60 mmol/L SDS, 20% (v/v) methanol, at pH 9.08 and 20 kV applied voltage, the four analytes were completely separated within 22.7 min. The effects of concentration of borate, electrolyte pH, the concentrations of SDS and organic modifier and the applied voltage on electrophoretic behavior and separation were studied. Regression equations revealed linear relationships (correlation coefficients: 0.9986 - 0.9997) between the peak areas of the analytes and the corresponding concentrations. The levels of analytes in the different parts of S. strangulata were easily determined with recoveries ranging from 95.3% to 104.0%.

Abstract:OBJECTIVE To establish a method for the preparation of high-purity huperzine A from the extract of Huperzia serrata.METHODS By using the polymer reversed phase medium and C1s reversed phase medium and two steps of medium pressure preparation in series,huperzine A monomer was isolated and prepared from the Huperzia serrata extract.RESULTS The purity of 99.7％ huperzine A monomer was obtained,and the overall recovery rate was 87.5％.CONCLUSION This method utilizes the characteristics of polymer medium and C18 medium to provide an efficient,green and economical way to obtain pharmaceutical grade huperzine A through technological linkage.

Abstract:Objective To establish a method for determination of several alkaloids in Chelidonium majus L.and perfect the quality standard of this kind of traditional Chinese medicine.Methods The RP-HPLC method was used with Inertsil ODS-3 column(4.6 mm×250 mm),and Acetonitrile-1% triethylamine(25∶75,adjusted to pH 3 with phosphoric acid) as mobile phase.The flow rate is 1ml/min,column temperature 35℃ and 274nm as detection wavelength.Results There were good linear relationships between the masses and the peak areas of Chelerythrine,Sanguinarine,Coptisine in the ranges of 0.0134~0.2520μg(r=0.999 7)、0.0760~0.6840μg(r=0.999 9) and 0.1416~2.5488μg(r=0.999 9),respectively.Conclusion The method is accurate,sensitivite,reliable and can be used repeatedly which is suitable for the quality evaluation of Chelidonium majus L..
Micellar electrokinetic chromatography for separation of a mixture of coptis alkaloids, scute flavonoids, and rhubarb anthraquinones and bianthrones

Abstract:Coptidis Rhizoma, Scutellariae Radix, and Rhei Rhizoma are three common Chinese herbs. There are many herbal formulas which contain either two or all three of the herbs mentioned above. Their bioactive components have already been identified, respectively. However, there is no report about separation of the 13 bioactive constituents of the three herbs at the same time. In order to assess these constituents of related Chinese herbal preparations, a micellar electrokinetic chromatography method was developed. While buffer pH and surfactant concentration affected the resolution of separation, acetonitrile percentage was found to significantly influence the resolution, peak shape, and elution window. Optimum separation of 13 compounds was achieved at pH 7.3 using a buffer mixture of 70% (v/v) 3 mM di-sodium tetraborate, 10 mM sodium dihydrogen phosphate, and 50 mM sodium deoxycholate with 30% (v/v) acetonitrile. When applying the developed method to analyze a model preparation, San-huang-xie-xin-tang, which contains all three herbs, 8 of the 13 bioactive constituents, could be determined. The present study proposed a method to assess San-huang-xie-xin-tang within short analysis time and also provided a possible starting point to evaluate related herbal preparations containing Coptidis Rhizoma, Scutellariae Radix, and Rhei Rhizoma.

Abstract:A simple and sensitive strategy using cyclodextrin-modified micellar electrokinetic chromatography with diode array detector was developed and applied for the simultaneous separation and determination of nine components in Sanyetangzhiqing (SYTZQ), a hypoglycemic and hypolipidemic agent. Several important parameters affecting separation performance were evaluated and optimized using single variable methods. Under the optimal conditions, baseline separation of the nine components, including four flavonoids (hyperoside, isoquercitrin, quercetin-3-O-glucuronoside, and astragalin), four phenolic acids (chlorogenic acid, rosmarinic acid, salvianolic acid B, and lithospermic acid), and a monoterpenoids (paeoniflorin), were achieved in less than 16 min. The correlation coefficients of the calibration curves were over 0.9996 for all the analytes. Intraday and interday precisions ranged from 0.4% to 4.8% and 1.7% to 5.0%, respectively. Recoveries of analytes varied from 95.3% to 105%. Validation results as well as the application to analyse SYTZQ samples demonstrated the applicability of the proposed method and thus provided an effective tool for the quality control of SYTZQ. Moreover, with the advantages of short time consuming, low energy consumption, high efficiency, and low cost, this method has laid a foundation for the determination and quality evaluation of multicomponents in Chinese herbal compounds.
[Separation and Determination of Ephedrine Alkaloids and 2,3,5,6-Tetramethyl Pyrazine in Ephedra Herba by HPLC].

Abstract:A sensitive and reliable high performance liquid chromatographic method(HPLC) has been developed for the first time for the simultaneous determination of the active ingredients of ephedrine alkaloids and 2,3,5,6-tetramethyl pyrazine (TMP) in Ephedra herba crude drug and two Chinese traditional medicines (Xiao-er qingfeiwan and Lu-si kewan). The HPLC assay was performed on a reversed phase C18 column (Nova-Pak C18, 3.9 mm i.d. x 150 mm) by using methanol-0.02 mol/L KH2PO4-acetic acid-triethyl amine (4:96:0.2:0.01, V/V) as mobile phase for the ephedrine alkaloids analysis and methanol-H2O-acetic acid (35:65:0.5, V/V) as mobile phase for TMP analysis. Regression equations revealed the linear relationships (correlation coefficients: 0.991-0.998) between the peak area of each constituent (E, PE, NE, NPE, TMP) and its concentration. The detection limits for E, PE, NE, NPE and TMP were 0.4 mg/L, 0.1 mg/L, 0.03 mg/L, 0.02 mg/L and 0.03 mg/L, respectively, and the recoveries ranged between 92%-103%. The contents of E, PE, NE, NPE, TMP in Ephedra herba, traditional medicine Xiao-er qingfeiwan and Lu-si kewan were determined respectively. The relative standard deviations (RSD) of the contents ranged between 1.1%-3%.

Abstract:A reverse-phase high performance liquid chromatographic method was developed for the determination of huperzine A in the extracts of Diphasiastrum complanatum(L.)Holub.The chromatographic analysis was carried out using a Diamonsil C18 column at 25 ℃.The mobile phase was acetonitrile-potassium phosphate monobasic(15/85,V/V),at a flow rate of 1ml/min and UV detection at 308 nm.The experimental results showed a good linear relationship between the peak area and mass of the huperzine A within the range of 0.224~4.48 μg(R=0.9999).The relative standard deviation(RSD) was 0.91%(n=5).The average recovery was 98.8%,The method is simple,rapid and accurate and provide a good way for the seeking of huperzine A.

Abstract:Reversed phase-hydrophilic interaction chromatography-tailored multiple reaction monitoring (RPLC-HILIC-tailored MRM) was developed for simultaneous determination of 27 components with wide polarity and wide range of content in Cistanches herba. RPLC was directly coupled with HILIC to achieve comprehensive retention of numerous compounds regardless of the polarity. Acquity UPLC HSS T3 column (100 mmx2.1 mm, 1.8 mu m) and Xbridge Amide column (150 mmx4.6 mm, 3.5 mu m) were employed for reversed phase separation and HILIC mode, respectively. To overcome the obstacle of incompatible mobile phases, dilution pumps were involved between two columns via a mixer. Pump A and pump B were responsible for delivering 0. 1 % formic acid and acetonitrile , respectively , into the RPLC column with a flow rate of 0. 15 mL/min by gradient elution. Pump C and pump D, served as dilution pumps, introduced the same mobile phase as pump A and pump B into the mixer at a flow rate of 1. 0 mL/min, which contributed to retention of polar compounds on HILIC column. For mass detection, tailored MRM with optimal collision energy was carried out by online energy-resolved MS to suppress mass response of abundant constituents, and to advance mass response of trace compounds. A total of 27 components in thirty-six batches of Cistanches Herba, such as amino acids , organic acids, phenylethanoid glycosides , lignans, and iridoids, were determined by RPLC-HILIC-tailored MRM. Satisfactory correlation coefficients were observed for all calibration equations with greater than 0. 9929 over their corresponding concentration ranges. The limits of detection (LODs) were 0. 0032-160 mu g/g and the limits of quantification (LOQs) were 0. 032-320 mu g/g. The average recoveries at three spiked levels were in the range of 74. 7% -125. 1% with relative standard deviations (RSDs) of 1. 6% -13. 7% . The developed method showed wide linear range, good accuracy and wide applicability , and had the potential for large-scale quantitative analysis of traditional Chinese medicine.

Abstract:OBJECTIVES Blunt SBI is infrequent and its diagnosis may be difficult, especially in the face of confounding variables. The purpose of this study was to evaluate methods for making the diagnosis of blunt SBI. METHODS Patients with blunt small bowel injury (SBI) were identified from the registries of 95 trauma centers for a 2-year period (1998-1999). Patients with SBI (cases) were matched by age and Injury Severity Score with a blunt trauma patient receiving an abdominal workup who did not have SBI (controls). RESULTS Logistic regression models were unable to differentiate SBI with perforation from SBI without perforation. Thirteen percent of patients with documented perforating SBI had normal abdominal computed tomographic scans preoperatively. CONCLUSION Alone or in combination, current diagnostic approaches lack sensitivity in the diagnosis of perforated SBI. Improvements in diagnostic methods and approaches are needed to ensure the prompt diagnosis of this uncommon but potentially devastating injury.
Simultaneous Determination of Five Alkaloids in the Mixture of Rhizoma Coptidis and Fructus Evodiae by MEKC

Abstract:Objective:To establish a method for the determination of five primary alkaloids (Berberine,Palmatine, Jatrorrhizine,Evodiamine and Rutaecarpine)in the Mixture of Rhizoma Coptidis and Fructus Evodiae by micellar electrophokinectic chromatography.Method:The separation was performed on a fused-silica capillary of 75 μm× 74 cm(65 cm effective length).20 mmol·L~(-1)phosphate buffer(including 50 mmol·L~(-1)SDS,pH 6.9)-acetoni- trile(8:5)was selected for the running buffer.The voltage applied was 25 kV.The sample was injected by air pres- sure (2 kPa,3 s).The detection wavelength was set at 225 nm.Results:The regression equations for Berberine,Pal- matine,Jatrorrhizine,Evodiamine and Rutaecarpine showed good linearity in the ranges of 0.027-1.345,0.018- 0.710,0.018-0.712,0.016-0.653 and 0.016-0.661 mg·mL~(-1) respectively.The recoveries (n=6) were 99.2%(RSD 3.2%),103.1%(RSD 2.1%),100.9%(RSD 1.5%),99.4%(RSD 2.9%),101.9%(RSD 3.3%),respectively.Five alkaloids in Rhizoma Coptidis,Frectus Evodiae and the Mixture of Rhizoma Coptidis and Fructus Evodiae were determinated.Conclusion:The method is simple and accurate.It can be applied to optimize the extraction process and quality control of the Mixture of Rhizoma Coptidis and Fructus Evodiae.

Abstract:Capillary electrophoresis with electrochemical detection has been employed for the determination of p -hydroxyacetophenone, chlorogenic acid, and caffeic acid in Herba Artemisiae Scopariae (the dried sprout of Artemisia scoparia Waldst. et Kit.). The effects of several important factors, such as the concentration and the acidity of the running buffer, separation voltage, injection time, and detection potential, were investigated to acquire the optimum conditions. The detection electrode was a 300-μm-diameter carbon disc electrode at a working potential of +0.90 V (relative to the saturated calomel electrode). The three analytes can be well separated within 11 min in a 40-cm-long fused-silica capillary at a separation voltage of 15 kV in 50 mM borate buffer (pH 9.2). The relation between peak current and analyte concentration was linear over about 3 orders of magnitude, with detection limits (signal-to-noise ratio of 3) of 0.31, 0.39, and 0.50 μM for p -hydroxyacetophenone, chlorogenic acid, and caffeic acid, respectively. The proposed method has been successfully applied to monitor the three bioactive constituents in real plant samples and to differentiate between different herbal drugs with satisfactory assay results.
Determination of Rutin and Quercetin in Plants by Capillary Electrophoresis with Electrochemical Detection.

Abstract:A method based on capillary electrophoresis with electrochemical detection (CE-ED) was developed for the determination of daidzein, rutin and quercetin. The effects of some important factors such as the acidity and concentration of running buffer, separation voltage, injection time, and detection potential were investigated to acquire the optimum conditions. The working electrode was a 300μm diameter carbon disc electrode positioned opposite the outlet of capillary. The three analytes could be well separated within 10min in a 40cm length capillary at a separation voltage of 12kV in a 100mmol/l borate buffer (BB, pH 9.0). The response was linear over three orders of magnitude with detection limits (S/N=3) ranged from 0.190×10−6 to 0.434×10−6mol/l for all compounds. This proposed method demonstrated good long-term stability and reproducibility with relative standard deviations (R.S.D.) of less than 5% for both migration time and peak current (n=7). It has been successfully applied for the determination of rutin and quercetin in Chinese traditional drug, Flos Sophorae buds and in the leaves of Ligustrum lucidum Ait. and Cinnamomum camphora (L.) Presl.
Rapid Determination of 3 Components with Different Polarities in Medicinal Mushrooms by Multistep Matrix Solid-Phase Dispersion and High-Performance Liquid Chromatography Analysis

Abstract:In this study, a multistep matrix solid-phase dispersion (MSPD) combining with a high-performance liquid chromatography method was developed for assaying 3 components of different polarities (mannitol, adenosine, and ergosterol) from mushroom samples. MSPD extraction was carried out using 1 g octadecyl-bonded silica as the sorbent material, 9% methanol, 20% methanol, and 100% methanol as eluting solvents for the elution of mannitol, adenosine, and ergosterol, respectively. Mannitol was separated on an NH2P-50 4E column and detected using an evaporative light scattering detector. Adenosine and ergosterol were separated on a Poroshell 120 SB-C18 column and measured at 260 nm and 283 nm, respectively. The developed method showed good linearity (R ≥ 0.9986) within the test range. The relative SD (RSD) of precisions were less than 1.4%, and the recoveries were 95.6%-97.0% (RSD ≤3.0%). Compared with the reported methods, the developed procedure could rapidly prepare components with different polarities (mannitol, adenosine, and ergosterol) from medical mushroom samples with less organic solvent and sample. The method is rapid and eco-friendly, which is helpful to improve the quality evaluation of medicinal mushrooms.
Separation and determination of ephedrine alkaloids and tetramethylpyrazine in Ephedra sinica Stapf by gas chromatography-mass spectrometry

Abstract:A simple, sensitive, and reliable method using gas chromatography (GC)-mass spectrometry (MS) is developed for the simultaneous determination of ephedrine alkaloids and 2,3,5,6-tetramethylpyrazine (TMP) in Ephedra sinica Stapf. The sample is extracted with ethyl ether and submitted to GC-MS for identification and quantitation without derivatization. The column used for GC is an HP-5 (30.0 m x 250 microm x 0.25 microm, 5% phenyl methyl siloxane), and the carrier gas is helium. The detection limits for ephedrine, pseudoephedrine, and TMP are 0.4 ng 0.7 ng, and 0.02 ng (signal-to-noise ratio of 3), respectively. The reproducibility of the total procedure is proved to be acceptable (RSD < 2%), and the recoveries are above 93%.
Simultaneous determination of eight alkaloids and oleandrin in herbal cosmetics by dispersive solid-phase extraction coupled with ultra high performance liquid chromatography and tandem mass spectrometry

Abstract:We utilized ultra-high performance liquid chromatography with tandem mass spectrometry and dispersive solid-phase extraction to develop a new method for the detection of nine analytes (scopolamine, cephaeline, strychnine, hyoscyamine, brucine, hydrastine, ajmalicine, colchicine, and oleandrin) in herbal cosmetics. Acetonitrile/water and 2-propylaminoethylamine were used to disperse and purify during the dispersive solid-phase extraction step. The analytes were separated by a Waters UPLC HSS T3 column and detected through electrospray ionization source in the positive mode with multi-reaction monitoring conditions. Under the optimal conditions, the calibration curves were linear in the range of 0.2-100.0 μg/L with the correlation coefficients higher than 0.995. The method limit of quantitation (S/N = 10) were 5.0 μg/kg for oleandrin and 1.0 μg/kg for the other eight alkaloids. The mean recoveries at three spiked concentration levels of 1.0-10.0 μg/kg were in the range of 86.9-116.5% with the intra-day relative standard deviations (n = 6) ranging from 2.4 to 8.8%, and inter-day relative standard deviations ranging from 2.7 to 5.7%. This method is accurate, simple and rapid, and has been applied to the quality supervision of herbal cosmetics in Guangzhou.

Abstract:Capillary zone electrophoretic (CZE) separation of pharmacologically active alkaloids, matrine, sophocarpine, sophoridine, oxymatrine, and oxysophocarpine, was systematically optimized, and their contents in different parts of Sophora species were quantitatively determined within 16 min under the following optimized conditions: 0.04 mol/L citrate buffer system at pH 4.5, applied voltage 30 kV, capillary temperature 25°C, and diode array detection at 201 nm. The recoveries are from 90 to 108%, with repeatability ranging from 0.7 to 4.2% (relative standard deviation) and the detection limits ranging from 0.032 to 0.070 mg/mL (signal to noise ratio=3). The proposed CZE method is suitable for routine analysis and quality control of herbal medicines that contain these alkaloids. © 2001 John Wiley & Sons, Inc. J Micro Sep 13: 221–226, 2001

Abstract:A rapid and efficient micellar electrokinetic capillary chromatography (MEKC) method was developed to analyze edgeworoside C (1), kaempferol-3-O-beta-D-glucoside (2) and rutin (3) in the alabastrum of Edgeworthia chrysantha Lindl. for the first time. The factors that affect the separation were studied, such as the concentrations of the buffer, SDS, and organic modifier, the apparent pH, the applied voltage and temperature. The analytes were well separated within 15 min with an electrolyte containing 25 mM Na(2)B(4)O(7), 30 mM NaH(2)PO(4), 60mM SDS and 15% acetonitrile (pH(*) 9.1) at 25 kV and 15 degrees C. The correlation coefficients between the peak areas of analytes and the corresponding concentrations were 0.9976-0.9981 under the optimum conditions. The relative standard deviations (R.S.D.) of the migration time and peak area were in the range 0.6-1.7 and 1.9-5.3%, respectively. The contents of analytes in E. chrysantha Lindl. collected from the different places were successfully determined with the recoveries ranging from 95.9 to 104.3%. And, the results demonstrated that there was significant difference between the two real samples.
