Abstract:ABSTRACT In soil, fungal colonization of plant roots has been traditionally studied by indirect methods such as microbial isolation that do not enable direct observation of infection sites or of interactions between fungal pathogens and their antagonists. Confocal laser scanning microscopy was used to visualize the colonization of tomato roots in heat-treated soil and to observe the interactions between a nonpathogenic strain, Fo47, and a pathogenic strain, Fol8, inoculated onto tomato roots in soil. When inoculated separately, both fungi colonized the entire root surface, with the exception of the apical zone. When both strains were introduced together, they both colonized the root surface and were observed at the same locations. When Fo47 was introduced at a higher concentration than Fol8, it colonized much of the root surface, but hyphae of Fol8 could still be observed at the same location on the root. There was no exclusion of the pathogenic strain by the presence of the nonpathogenic strain. These results are not consistent with the hypothesis that specific infection sites exist on the root for Fusarium oxysporum and instead support the hypothesis that competition occurs for nutrients rather than for infection sites.

Growth-Promoting and Protective Effect of Trichoderma atrobrunneum and T. simmonsii on Tomato against Soil-Borne Fungal Pathogens

Abstract:Trichoderma fungi are promising candidates for biocontrol agents and plant growth promoters. Trichoderma atrobrunneum and T. simmonsii were evaluated for the control of soil-borne phytopathogenic fungi, in the present study. Dual culture tests with Rhizoctonia solani and Fusarium oxysporum f. sp. lycopersici were used to conduct in vitro evaluation. In the presence of Trichoderma, phytopathogen’s growth rate was inhibited up to 59.70% for R. solani and 42.57% for F. oxysporum. Greenhouse trials with potted tomato plants demonstrated that Trichoderma caused a significant increase of stem height and fresh stem weight in pathogen-inoculated plants, compared with the negative control (plants artificially inoculated with the phytopathogen only). Except for T. simmonsii, plant growth was not significantly enhanced by a Trichoderma presence in the positive control (healthy plants). The overall performance of the two Trichoderma species studied was equivalent to that of the T. harzianum T22 commercial strain. All the tested species were found to be effective in suppressing colony growth and disease development of the soil borne pathogens in dual cultures and potted plants, indicating that they could be used as biocontrol agents. Our findings are discussed in the context of enhancing endophytic microorganisms’ application in crop production systems.
Biocontrol Activity of Nonpathogenic Strains of Fusarium oxysporum: Colonization on the Root Surface to Overcome Nutritional Competition

Abstract:Fusarium oxysporum is a soil-borne fungal pathogen that causes vascular wilts in a wide variety of crops. Certain nonpathogenic strains of F. oxysporum are known to protect crops against F. oxysporum pathogens. We assessed the biocontrol activities of nonpathogenic mutants of F. oxysporum ff. spp. melonis and lycopersici generated by disruption of the FOW2 gene, which encodes a Zn(II)2Cys6-type transcriptional regulator essential for their pathogenicity. Pre-inoculation of melon or tomato roots with strain Δ FOW2 conidia markedly reduced disease incidence caused by the parental wild-type strain in a concentration-dependent manner of conidial suspensions of Δ FOW2 strains. The biocontrol effect caused by the Δ FOW2 pre-inoculation lasted for at least 7 days. Pre-inoculation of melon roots with the wild-type or Δ FOW2 strain of F. oxysporum f. sp. lycopersici and nonpathogenic F. oxysporum strain also led to biocontrol activity against F. oxysporum f. sp. melonis , indicating that the biocontrol activity of Δ FOW2 strains is due to its nonpathogenic nature, not to the FOW2 disfunction. Conidial germination and hyphal elongation of only the wild-type strain were inhibited on melon root surface pre-inoculated with conidia of strains nonpathogenic to melon plants. Expression of defense-related genes was not significantly induced in roots and aboveground parts of melon seedlings preinoculated with Δ FOW2 conidia. Carbon source competition assay showed that nonpathogenic strains competed with the wild-type strain for a carbon source in soil. Strain Δ FOW2 also competed with the oomycete pathogen Pythium aphanidermatum for carbon source and protected melon plants from P. aphanidermatum. Our results suggest that the biocontrol activity of the nonpathogenic F. oxysporum strains used in this study mainly depends on their extensive colonization of the root surface and outcompeting pathogens for nutrients.

Abstract:Endophytic fungal-based biopesticides are sustainable and ecologically-friendly biocontrol agents of several pests and diseases. However, their potential in managing tomato fusarium wilt disease (FWD) remains unexploited. This study therefore evaluated effectiveness of nine fungal isolates against tomato fusarium wilt pathogen, Fusarium oxysporum f. sp. lycopersici (FOL) in vitro using dual culture and co-culture assays. The efficacy of three potent endophytes that inhibited the pathogen in vitro was assessed against FWD incidence, severity, and ability to enhance growth and yield of tomatoes in planta. The ability of endophytically-colonized tomato (Solanum lycopersicum L.) plants to systemically defend themselves upon exposure to FOL were also assessed through defence genes expression using qPCR. In vitro assays showed that endophytes inhibited and suppressed FOL mycelial growth better than entomopathogenic fungi (EPF). Endophytes Trichoderma asperellum M2RT4, Hypocrea lixii F3ST1, Trichoderma harzianum KF2R41, and Trichoderma atroviride ICIPE 710 had the highest (68.84-99.61%) suppression and FOL radial growth inhibition rates compared to EPF which exhibited lowest (27.05-40.63%) inhibition rates. Endophytes T. asperellum M2RT4, H. lixii F3ST1 and T. harzianum KF2R41 colonized all tomato plant parts. During the in planta experiment, endophytically-colonized and FOL-infected tomato plants showed significant reduction of FWD incidence and severity compared to non-inoculated plants. In addition, these endophytes contributed to improved growth promotion parameters and yield. Moreover, there was significantly higher expression of tomato defence genes in T. asperellum M2RT4 colonized than in un-inoculated tomato plants. These findings demonstrated that H. lixii F3ST1 and T. asperellum M2RT4 are effective biocontrol agents against FWD and could sustainably mitigate tomato yield losses associated with fusarium wilt.

Abstract:A mutant of the root pathogen Fusarium oxysporum f. sp. lycopersici, deficient in class V chitin synthase, has been shown previously to be nonvirulent. In this study, we tested the hypothesis that the cause of its avirulence could be the elicitation of the induced plant defence response, leading to the restriction of fungal infection. Co-inoculation of tomato plants with the wild-type strain and the DeltachsV mutant resulted in a significant reduction in symptom development, supporting a protective mechanism exerted by the mutant. The ability of the mutant to penetrate and colonize plant tissues was determined by scanning and transmission electron microscopy, as well as fluorescence microscopy using green fluorescent protein- or cherry fluorescent protein-labelled fungal strains. The extent of wild-type strain colonization in co-inoculated plants decreased steadily throughout the infection process, as shown by the quantification of fungal biomass using real-time polymerase chain reaction. The hypothesis that defence responses are activated by the DeltachsV mutant was confirmed by the analysis of plant pathogenesis-related genes using real-time reverse transcriptase-polymerase chain reaction. Tomato plants inoculated with the DeltachsV mutant showed a three fold increase in endochitinase activity in comparison with wild-type inoculated plants. Taken together, these results suggest that the perturbation of fungal cell wall biosynthesis results in elicitation of the plant defence response during the infection process.

Protection to Tomato Wilt Disease Conferred by the Nonpathogen Fusarium oxysporum Fo47 is More Effective Than that Conferred by Avirulent Strains

Abstract:Although the vascular pathogen Fusarium oxysporum is notorious for being the causal agent of Fusarium wilt disease, the vast majority of F. oxysporum strains are harmless soil and root colonizers. The latter F. oxysporum's are often endophytes colonizing roots intracellularly without negatively affecting plant fitness. Actually, most of them, like Fo47, are beneficial providing biological control to various root pathogens. Interestingly, also pathogenic F. oxysporum inoculated on a resistant host (i.e., avirulent F. oxysporum f. sp. lycopersici) can reduce susceptibility to virulent F. oxysporum strains via a mechanism called "cross protection." It has been hypothesized that cross protection is based on activation of a resistance protein of the host upon recognition of a cognate avirulence (Avr) protein of the pathogen. Currently, it is unknown whether the biocontrol activity of F. oxysporum endophytes utilizes similar mechanisms as cross protection conferred by avirulent pathogens, and whether both provide a quantitative similar level of protection. Here, we show that in tomato biocontrol activity of the Fo47 endophyte to the pathogen F. oxysporum f. sp. lycopersici is more effective than cross protection induced by avirulent F. oxysporum f. sp. lycopersici strains activating either I, I-2, or both resistance proteins upon recognition of Avr1 or the Avr2/Six5 pair, respectively. These findings imply that cross protection and biological control utilize different mechanisms to reduce susceptibility of the host to subsequent infections.

Abstract:Tomato (Lycopersicon esculentum Mill.) is the most consumed vegetable in the world after potato. In Cameroon, the plant is cultivated in almost all agroecological areas, however, yields remain low due to attacks by various pathogens and insects. Among the pathogens, Fusarium oxysporum f.sp. lycopersici (FOL) is a phytopathogenic fungus responsible for Fusarium wilt, a disease responsible for enormous economic losses. To contribute to the control of this microbial pathogen, the stimulatory effect of the tomato defence system of extracts of some plants in the tomato/FOL interaction was evaluated. Tomato plants were treated with the aqueous extracts (AE) of Callistemon citrinus (C. citrinus), Cymbopogon citratus (C. citratus), and Oxalis barrelieri (O. barrelieri ). After 4 days of spraying with the extracts, the plants were inoculated with a virulent strain of Fusarium oxysporum f.sp. lycopersici (FOL) under controlled conditions. Tomato roots were used to determine the contents of hydrogen peroxide (H2O2), phenols and malondialdehyde (MDA). The activities of the main antioxidant enzymes were also evaluated in tomato roots: catalase (CAT), phenylalanine ammonia lyase (PAL), peroxidase (POX) and superoxide dismutase (SOD). The results showed that treatment of tomato plants with plant extracts and their infection with FOL induced an increase in the contents of H2O2, phenols and MDA in tomato roots; an increase in PAL, POX, SOD activities and a reduction in CAT activity. Our results suggest that the increase and reduction of enzymatic activities, and the increase in the synthesis of some metabolites could mitigate the oxidative damage that takes place during the expansion of the pathogen. Aqueous extracts of C. citrinus, C. citratus and O. barrelieri at 10% (W/V) could be used as natural products to stimulate the tomato defence system against FOL. These results could contribute to the development of natural products to induce tomato resistance against FOL, thus improving productivity, quantity and quality of production.

Abstract:The antifungal effects of Citrullus colocynthis extract (Hexane, chloroform, methanol, and water) were tested in vitro on Fusarium oxysporum f. sp. lycopersici (Sacc.) W. C. Snyder & H. N. Hans (FOL), the causal agent of Fusarium wilt. Of these, methanol and water extract at 10% showed the highest inhibition of mycelial growth of FOL by 12.32 and 23.61 mm respectively. The antifungal compounds were identified through Fourier transform infrared (FT-IR) spectroscopy and gas chromatography-mass spectroscopy (GC-MS). The methanol extract was compatible with the biocontrol agent Trichoderma viride. The antagonistic fungi were mass-cultured under laboratory conditions using sorghum seeds. Both T. viride and C. colocynthis methanol extract was also tested alone and together against FOL under both in vitro and in vivo conditions. The combination of T. viride and C. colocynthis showed the highest percentage of antifungal activity (82.92%) against FOL under in vitro conditions. This study revealed that induced systemic resistance (ISR) in enhancing the disease resistance in tomato plants against Fusarium wilt disease. The combined treatment of T. viride and C. colocynthis significantly reduced the disease incidence and index by 21.92 and 27.02% in greenhouse conditions, respectively. Further, the induction of defense enzymes, such as peroxidase (PO), polyphenol oxidase (PPO), β-1,3-glucanase, and chitinase were studied. The accumulation of defense enzyme was greater in plants treated with a combination of T. viride and C. colocynthis compared to the control. Reduction of wilt disease in tomato plants due to the involvement of defense-related enzymes is presumed through this experiment.

Abstract:Studying plant early immunity, such as the unique immune mechanisms against pathogens, is an important field of research. Tomato wilt resulting from the infection by Fusarium oxysporum f. sp. lycopersici ( Fol ) is an important soil-borne vascular disease. In this study, we challenged tomato plants with Fol for a time-course RNA sequencing (RNA-seq) analysis. The result indicated that phenylpropanoid and flavonoid pathway genes were significantly enriched during the early invasion stage. Further study revealed that the flavonoids galangin and quercetin could effectively inhibit Fol growth and enhance wilt resistance in tomato. Moreover, the genes involved in plant-pathogen interactions, the MAPK signaling pathway, and plant hormone signal transduction were significantly enriched. These genes were also involved in plant pattern-triggered immunity (PTI) and effector-triggered immunity (ETI) signaling pathways. Strikingly, the transcription levels of pathogen-related protein 1 ( SlPR1 ) were dramatically increased at 2 days post Fol inoculation, implying that SlPR1 is important in early immunity in tomato. SlPR1 does not have direct antifungal activity. Instead, its C-terminal peptide CAPE1 could activate root defense responses, such as the reactive oxygen species (ROS) burst, salicylic acid (SA)/jasmonic acid (JA) production, and defense-related gene expression, which collectively increased tomato resistance to Fol infection. In addition, CAPE1 could induce systemic acquired resistance (SAR). Application of CAPE1 onto tomato leaves induced local resistance to the pathogen Botrytis cinerea and systemic resistance to Fol infection. These results advanced our understanding for the early immunity against Fol in tomato and provide potential strategy for tomato disease control.

Native soils with their microbiotas elicit a state of alert in tomato plants

Abstract:Several studies have investigated soil microbial biodiversity, but understanding of the mechanisms underlying plant responses to soil microbiota remains in its infancy. Here, we focused on tomato (Solanum lycopersicum), testing the hypothesis that plants grown on native soils display different responses to soil microbiotas. Using transcriptomics, proteomics, and biochemistry, we describe the responses of two tomato genotypes (susceptible or resistant to Fusarium oxysporum f. sp. lycopersici) grown on an artificial growth substrate and two native soils (conducive and suppressive to Fusarium). Native soils affected tomato responses by modulating pathways involved in responses to oxidative stress, phenol biosynthesis, lignin deposition, and innate immunity, particularly in the suppressive soil. In tomato plants grown on steam-disinfected soils, total phenols and lignin decreased significantly. The inoculation of a mycorrhizal fungus partly rescued this response locally and systemically. Plants inoculated with the fungal pathogen showed reduced disease symptoms in the resistant genotype in both soils, but the susceptible genotype was partially protected from the pathogen only when grown on the suppressive soil. The 'state of alert' detected in tomatoes reveals novel mechanisms operating in plants in native soils and the soil microbiota appears to be one of the drivers of these plant responses.

Abstract:Bithionol is a primary photosensitizer. Cross-reactions occurred with tetrachlorosalicylanilide (TCSA), tribromosalicylanilide (TBS), and hexachlorophene but not with trichlorocarbanilide (TCC). Photopatch tests produced relatively characteristic responses for each substance. Reactions to TBS and TCSA in patients with contact photodermatitis from bithionol represent true photo-cross-sensitivity. Reactions to hexachlorophene in these patients with contact photodermatitis from bithionol did not require light. These reactions represent the unusual phenomenon of crosssensitivity between a photosensitizer and a simple contact allergen.
Developing resistance against soil-borne Fusarium pathogen causing tomato wilt through in vitro cell line selection

Abstract:Numerous complex chemicals are being routinely used to prevent pathogens in various commercially important agri-horticultural crops that have some biosafety issues associated with human health and environment. Therefore, the present study was aimed to develop resistance in tomato against soil-borne fungal pathogen Fusarium oxysporum f. sp. lycopersici , through in vitro cell line selection approach. BLASTn analysis of sequenced 16S rRNA region of the fungus isolated from the wilt infected tomato plant was confirmed by ITS gene technology. Highest callus induction (79.63 ± 0.70%) along with shoot regeneration (78.68 ± 0.64%) was recorded on MS medium fortified containing 4.54 μM TDZ + 2.90 μM IAA and 2.27 μM TDZ + 1.0 μM 2,4-D using leaf explants, respectively. To carry out in vitro selection, developed calli using leaf explants were further subjected on selective medium followed by discontinuous selection cycles supplemented with 30% culture filtrate (CF) of targeted pathogen, where 21.32 ± 0.99 per cent average callus survival and 45.98 ± 0.55% average shoot induction (devoid of CF) were recorded. 12 moderately resistant lines were obtained from the total 48 cell lines during in vivo bioassay under glass house conditions. Two somaclones were confirmed at molecular level among the total 12 moderately developed resistant cell lines. The multilocation field trials are being done to check stable inheritance with generation wise advancements. It was inferred from the present study that these findings could be a cutting edge to develop resistance against various stresses in commercially important horticultural crops like tomato without posing any threat to environment.

Abstract:Fusarium wilt is considered one of the most destructive diseases for tomato plants. The novelty of this work was to investigate the antifungal and plant growth-promoting capabilities of some plant growth-promoting fungi (PGPF). Plant growth-promoting fungi (PGPF) improved the plant health and control plant infections. In this study, two fungal strains as PGPF were isolated and identified as Aspergillus fumigatus and Rhizopus oryzae using molecular method. The extracts of A. fumigatus and R. oryzae exhibited promising antifungal activity against F. oxysporum in vitro Moreover, antagonistic effect of A. fumigatus and R. oryzae against F. oxysporum causing tomato wilt disease was evaluated in vivo. Disease severity and growth markers were recorded and in vitro antagonistic activity assay of the isolated A. fumigatus and R. oryzae against Fusarium oxysporum was measured. Physiological markers of defense in plant as response to stimulate systemic resistance (SR) were recorded. Our results indicated that A. fumigatus and R. oryzae decreased the percentage of disease severity by 12.5 and 37.5%, respectively. In addition, they exhibited relatively high protection percentage of 86.35 and 59.06% respectively. Fusarium wilt was declined the growth parameters, photosynthetic pigments, total soluble carbohydrate, and total soluble protein, whereas content of free proline, total phenols, and the activity of antioxidant enzymes activity increased under infection. Moreover, application of A. fumigatus and R. oryzae on infected plants successfully recovered the loss of morphological traits, photosynthetic pigment total carbohydrates, and total soluble proteins in comparison to infected control plants. PGPF strains in both non-infected and infected plants showed several responses in number and density of peroxidase (POD) and polyphenol oxidase (PPO) isozymes.

Abstract:Aims: To isolate endophytic Streptomyces strains from tomato and examine their antimicrobial activity. Methods: Endophytic Streptomyces strains were isolated using surface-sterilization methods and identified by morphological characteristics. Antimicrobial activities were measured by the agar plate sensitivity method. Antifungal activity in vivo was measured by seedling mortality in infested soils. Results: Twenty-one per cent of endophytic streptomycete isolates produced antibacterial metabolites and 41% produced antifungal metabolites in S medium. Sixty-five per cent of the most frequently isolated strains inhibited the growth of Rhizoctonia solani by the antibiosis assay but only 32% produced metabolites active against R. solani in S medium. Growth promotion and enhanced disease resistance of seedlings inoculated with Streptomyces sp. strain S30 were observed in tomato but not in cucumber seedlings. Conclusions: Endophytic Streptomyces spp. strains were successfully isolated using stringent methods and strain S30 promoted growth and enhanced resistance to R. solani in tomato seedlings. Significance and impact of the study: Endophytic streptomycetes showing antifungal activity in vitro and in vivo may indicate the potential for their use as biocontrol agents particularly of R. solani disease of tomato seedlings.
Metabolomics and histopathological analysis of two tomato cultivars after co-infection with soil-borne pathogens (Southern root-knot nematode and Fusarium wilt fungus)

Abstract:Southern root-knot nematode (Meloidogyne incognita) and Fusarium wilt fungus (Fusarium oxysporum) are one of the most predominant pathogens responsible for substantial agricultural yield reduction of tomato. The current study planned to assess the effects of M. incognita (Mi) and F. oxysporum (Fo) and their co-infection on two tomato cultivars, Zhongza 09 (ZZ09) and Gailing Maofen 802 (GLM802). The present study examined the effects of co-infection on leaf morphology, chlorophyll content, leaf area, and histopathology. The present study used metabolomics to evaluate plant-pathogen interactions. The outcomes of the current study revealed that chlorophyll content and leaf area decreased more in GLM802 during co-infection. In co-infection (Fo + Mi), the chlorophyll content reduction in ZZ09 was 11%, while in GLM802 the reduction reached up to 31% as compared to control. Moreover, the reduction in leaf are in ZZ09 was 31%, however, in the GLM802 reduction was observed 54% as compared to control plants. Similarly, GLM802 stems exhibited larger brown patches on their vascular bundles than ZZ09 stems. The rate of browning of GLM802 stems was 247% more than ZZ09, during co-infection. Moreover, GLM802 roots exhibited a higher abundance of hyphae and larger galls than ZZ09 roots. In metabolic studies, glutathione, succinic acid, and 2-isopropylmalic acid decreased, whereas spermine and fumaric acid increased in GLM802 co-infected stems. It indicates that GLM802 is weakly resistant; therefore, F. oxysporum and other pathogens readily damage tissue. In the co-infected stem of ZZ09, L-asparagine and shikimic acid increased, but pipecolic acid, L-saccharine, and 2-isopropylmalic acid declined. L-asparagine was crucial in preserving the stability of nitrogen metabolism, chlorophyll synthesis, and leaf growth in ZZ09. Shikimic acid's substantial accumulation could explain the limited extent of browning observed in the vascular bundles of ZZ09. Thus, the present study provides insight into M. incognita and F. oxysporum co-infection in two tomato cultivars, which may aid breeding efforts to generate commercially viable resistant cultivars. However, further research on the relationship between M. incognita and F. oxysporum in different host plants is required in the future.
Characterization of Pathogenic and Nonpathogenic Fusarium oxysporum Isolates Associated with Commercial Tomato Crops in the Andean Region of Colombia

Abstract:In Colombia, tomato production under protected conditions represents an important economic contribution to the agricultural sector. Fusarium wilt diseases, caused by pathogenic formae speciales of the soil-borne fungus Fusarium oxysporum Schltdl., cause significant yield losses in tomatoes throughout the world. Investigation of the F. oxysporum–tomato pathosystem in Colombia is required to develop appropriate alternative disease management. In this study, 120 fungal isolates were obtained from four different departments in the Central Andean Region in Colombia from tomato crops with symptoms of wilt disease. A molecular characterization of the fungal isolates was performed using the SIX1, SIX3, and SIX4 effector genes of Fusarium oxysporum f. sp. lycopersici W.C. Snyder & H.N. Hansen (Fol). Additionally, we developed a new specific marker to distinguish between Fusarium oxysporum f. sp. radicis-lycopersici Jarvis & Shoemaker (Forl) and Fol isolates. Furthermore, a phylogenetic analysis using the Translation Elongation Factor 1-alpha (EF1a) gene was performed with the collected isolates. Two isolates (named Fol59 and Fol-UDC10) were identified as Fol race 2, four isolates were identified as Forl, six isolates were identified as F. solani, and most of the isolates were grouped within the F. oxysporum species complex. The phylogenetic tree of EF1a showed that most of the isolates could potentially correspond to nonpathogenic strains of F. oxysporum. Additional pathogenicity assays carried out with Fol59 and Fol-UDC10 confirmed that both isolates were highly virulent strains. This study represents a contribution to the understanding of the local interaction between tomatoes and F. oxysporum in Colombia.
