Abstract:A novel ultrasensitive and simple amplified immunosensing strategy is designed based on a surface-enhanced fluorescence (SEF) nanohybrid made from covalently conjugated thionine–gold nanoparticles (GNP–Th), as a novel amplified fluorescence label, and magnetic nanoparticles (MNPs), as a biological carrier, used for hepatitis B virus surface antigen (HBsAg) detection. This immunosensing strategy operates on the basis of the capture and then release of the amplified fluorescence label. Capturing of the antiHBs-antibody (Ab)-modified GNP–thionine hybrid (GNP–Th-Ab) is carried out through the formation of a two-dimensional (sandwich) probe between this amplified label and antiHBs-antibody-modified magnetic nanoparticles (MNP-Ab), in the presence of a target antigen and using an external magnetic force. Afterward, releasing of the captured fluorescence label is performed using a protease enzyme (pepsin) by a digestion mechanism of grafted antibodies on the GNP–thionine hybrid. As a result of antibody digestion, the amplified fluorescent hybrids (labels) are released into the solution. To understand the mechanism of enhanced fluorescence, the nature of the interaction between thionine and gold nanoparticles is studied using the B3LYP density functional method. In such a methodology, several new mechanisms and structures are used simultaneously, including a SEF-based metal nanoparticle–organic dye hybrid, dual signal amplification in a two-dimensional probe between the GNP–thionine hybrid and MNPs, and a novel releasing method using protease enzymes. These factors improve the sensitivity and speed, along with the simplicity of the procedure. Under optimal conditions, the fluorescence signal increases with the increment of HBs antigen concentration in the linear dynamic range of 4.6 × 10–9 to 0.012 ng/mL with a detection limit (LOD) of 4.6 × 10–9 ng/mL. The proposed immunosensor has great potential in developing ultrasensitive and rapid diagnostic platforms.

Abstract:The rampant hepatitis B virus (HBV) seriously endangers human health, and hepatitis B surface antigen (HBsAg) is its early diagnostic marker. Therefore, it is crucial to construct a fast and highly sensitive HBsAg detection method. Based on high-efficiency magnetic separation technology and fluorescent composite material labelling technology, an accurate, fast and sensitive fluorescent immunosensing system for HBsAg detection was developed. Immunomagnetic beads constructed from carboxyl-functionalized Fe3O4 nanoparticles (Fe3O4-COOH) with excellent magnetic response performance were used as efficient capture carriers for HBsAg. Immunofluorescence composite microspheres constructed based on ultra-stable polystyrene-coated CsPbBr3 perovskite nanocrystals (CPB@PSAA) with high hydrophilic properties, were excellent fluorescent markers for HBsAg. Using this sensitive sandwich fluorescence sensing system a good linear relationship within the range of 0.2-15 ng/mL was established between HBsAg concentration and fluorescence intensity with a limit of detection (LOD) of 0.05 ng/mL. The system obtained satisfactory results when tested on real human serum samples. The magnetic-assisted fluorescence immune-sandwich sensor system has broad application prospects in biomedicine such as rapid and early diagnosis and effective prevention of infectious diseases.

Abstract:Herein, a novel sandwich-type electrochemical immunosensor was fabricated based on Fe3O4 nanoflowers (Fe3O4 NFs) and heterogeneous chain reaction (HCR) signal amplification strategy for the sensitive detection of hepatitis B surface antigen (HBsAg). The aldehyde-functionalized Fe3O4 NFs are used as a supporting matrix to immobilize the hepatitis B surface antibody1 (HBsAb1). The biotin-modified single-strand DNA (biotin-S0) was connected onto the biotin-HBsAb2 via linkage of streptavidin (SA), followed by addition of methylene blue (MB) modified single strand DNA1 (MB-S1) and DNA2 (MB-S2) for HCR signal amplification. The designed immunosensor exhibited a detection linear range of 0.5 pg mL-1-0.25 ng mL-1 and a low detection limit of 0.16 pg mL-1, with excellent stability, selectivity and reproducibility. Furthermore, HBsAg is detected in the serum samples with a stable and fast response, indicating that the proposed immunosensor has a promising potential application in clinical analysis.

Abstract:Hepatitis B Surface Antigen (HBsAg) is usually recognized as an earliest diagnostic markers for Hepatitis B Virus (HBV) infections. The precise and sensitive determination of HBsAg is crucial for the diagnostic of HBV infection. In this paper, a colorimetric immunosensor has been successfully designed and fabricated based on conjugating magnetic separation and enzymatic catalysis, for point-of-care detection of HBsAg. The HBsAg was simultaneously captured by the biotinylated HBsAg primary antibody (biotin-Ab1) labeled immunomagnetic nanoparticles and detection antibody (Ab2). In conjugation with Ab2, horseradish peroxidase (HRP) labeled rabbit anti-goat secondary antibody was linked with chromogenic agents to induce the colorimetric signal. As a result, a good linear relation was achieved between the concentration of HBsAg and OD450 nm (absorbance values at 450 nm) in the 0.1–20 ng mL−1 ranges, demonstrating a great specificity to HBsAg with a limit of detection of 0.012 ng mL−1. Furthermore, considering its succeed in real human serum sample test, this immunosensor could make contribute to potential applications in rapid diagnosis, effective prevention of epidemic and infectious diseases.

Abstract:An immunochromatographic assay (ICA) based on red silica nanoparticles (red-SiNPs) was developed for quantitative detection of hepatitis B surface antigen (HBsAg) in serum. Red SiNPs have unique optical properties and remarkable stability compared to red latex microspheres. Red-SiNPs functionalized with anti-HBsAg mAb1 were employed as detection immuno-probes. The sandwich-type immunoreaction structures were formed on ICA strips, and the accumulation of red SiNPs-mAb1 on the test line produced characteristic red color bands, enabling visual detection of HBsAg without instrumentation. Quantitative detection was performed by researching the relationship between the HBsAg concentration and the corresponding peak area of the gray value of the test line with a camera and image analysis software Image J. Under optimal conditions, the visual limit of detection (vLOD) of the strip for the qualitative detection of HBsAg is 1 ng mL(-1), which is lower than that of the routinely used red latex microsphere-based ICA. In the quantitative detection, the peak area of the gray value of the test line and the HBsAg concentration show a good linearity when the HBsAg concentration is in the range of 5-500 ng mL(-1). The calibration equation was y = 370.6118 + 9.6673x (R2 = 0.9962) and the limit of detection (LOD) of the method was 0.97 ng mL(-1) (S/N = 3). Our proposed method provides promising and versatile opportunities for the rapid and quantitative screening of other proteins at relatively sensitive levels by combining a colored label with ICA.

Abstract:A novel thionine unites with platinum nanowire inlaid globular SBA-15 (Pt NWs@g-SBA-15/Thi) not only utilizes as an efficient electrical signal probe but also constitutes an amplifying system with double-deck gold film (D-Au film) have been applied to the fabrication of sandwich-type immunosensor for detecting hepatitis B surface antigen (HBs Ag). The D-Au film can accelerate the electron transfer on the electrode interface due to the tunneling effect between the two Au films and can improve the load capacity of primary antibodies (Ab1) because of the good biocompatibility. The Pt NWs@g-SBA-15/Thi with uniform globular morphology not only can effectively reduce the spatial limitation for loading the secondary antibodies (Ab2) but also can provide outstanding pore accessibility of guest species from outside and offer catalytically active sites in a large scale. Besides, the presence of Thi can well enhance the electrical conductivity of Pt NWs@g-SBA-15/Thi. With the good cooperation between D-Au film and Pt NWs@g-SBA-15/Thi, a linear relationship between current signals and the concentrations of HBs Ag was obtained in the wide range from 10 fg/mL to 100ng/mL and the detection limit of HBs Ag was 3.3 fg/mL (signal-to-noise ratio of 3). Furthermore, the designed immunosensor with excellent selectivity, reproducibility and stability shows excellent performance in detection of human serum samples and provides a promising capacity for detecting a wide range of other tumor markers in clinical application.

Abstract:Despite the availability of effective hepatitis B vaccinations, the hepatitis B virus remains a serious global health concern. It is expected that early detection could aid in initiating therapy before the infection progresses to liver damage. A silicon nanowire rectangular optical waveguide has been demonstrated theoretically to detect the surface antigen of hepatitis B "HBsAg" based on label-free surface sensing using finite-element method-based COMSOL Multiphysics. Different procedural segments of the biomarker detection have been mimicked on the surface of a waveguide as adlayers to investigate the device theoretically. Initially, the parameters of the waveguide have been optimized to provide a large interaction of light and bio-analyte, i.e., to provide high sensitivity. The analyses are first performed at the waveguide level based on the light-analyte interaction. Furthermore, performances of the sensor have been obtained by incorporating this waveguide structure in the sensing arm of the Mach-Zehnder interferometer. The device structure shows ultra-high surface sensitivities such as phase surface sensitivity of 7.03×2πrad/nm and MZI surface sensitivity of 3421.89 µW/nm with an excellent detection limit of 2.92×10-3pg/mm2 for HBsAg detection. The proposed device can measure the HBsAg concentration as low as 0.00973 ng/mL, which is significantly low to detect the infection in an early stage.

Abstract:In this work, a sandwich-type electrochemical immunosensor was fabricated to the effective detection of hepatitis B surface antigen (HBsAg). The designed electrochemical immunosensor was based on Au core and Pd shell nanodendrites loaded on amino functionalized molybdenum dioxide nanosheets ( NDs/NH2–MoO2 NSs) as the secondary antibody (Ab2) label and silver nanoparticles were loaded by electrodeposited (D-Ag NPs) on the surface of electrode as the platform. Because of the synergistic effect and abundant catalytic activity sites provided by surface dendrite structure, NDs were more effective than single gold and palladium nanoparticles in catalytic reduction of hydrogen peroxide (H2O2). MoO2 had the good catalytic capacity for reduction of H2O2 and favourable electrical conductivity. Hence, the obtained NDs/NH2–MoO2 NSs were more effective than NDs and NH2–MoO2 NSs in catalytic reduction of hydrogen peroxide attribute to a synergistic effect. Also, Ag NPs with admirable electrical conductivity and biocompatibility were used as sensing platforms and primary antibodies (Ab1) carriers, which can accelerate the electron transfer and improve the sensitivity of the immunosensor. Here, the proposed electrochemical immunosensor offered a wide linear interval from 10 fg mL-1 to 100 ng mL-1 and the lower limit of detection of 3.3 fg mL-1 (S/N=3) for detection of HBsAg under optimal experimental conditions. Furthermore, the accuracy of the actual serum sample analysis was satisfactory, which showed that the electrochemical immunosensor possessed a good application prospect in clinical detection.

Abstract:Herein, highly sensitive electrochemical immunosensor based on noble metal and metal oxide nanoparticles functionalized bimetal conductive metal-organic frameworks (c-MOFs) was proposed toward HBsAg detection. The nanorod-like architecture of Ni3(HITP)2c-MOFs was synthesised via a simple and facile route, followed by dopant coble ions formation cobaltosic oxide (Co3O4) nanoparticles decorated bimetal NiCo(HITP) c-MOFs nanocomposites. Benefiting from large specific surface area, high electrical conductivity and abundant active sites of desirable nanostructures, the resultant Co3O4NPS/NiCo(HITP) bimetal c-MOFs nanostructure exhibits fast electron transfer capability and effective specific binding between antigen and antibody. Meanwhile, different amounts of Au nanoparticles were further functionalized on Co3O4NPS/NiCo(HITP) nanocomposites to further improve the electrochemical properties. Under optimum condition, the as -proposed Au@Co3O4NPS/NiCo (HITP) electrochemical immunosensor exhibits superior sensing performances toward HBsAg including wide linear response range from 1 pg mL-1 to 100 ng mL-1 and low detection limit of 15 fg mL-1 (S/N = 3). Meanwhile, the as -fabricated electrochemical immunosensors also displays outstanding selectivity, repeatability and stability. Besides, quantitative detection of HBsAg also achieves satisfactory results in real sample detection. Thereby, our work not only provides a promising strategy to construct highly sensitive electrochemical immunosensor using hybrid c-MOFs nanomaterials but also presents a potential application in the early diagnosis of hepatitis B -related diseases.

Abstract:A multiple signal amplification strategy was designed for an ultrasensitive competitive immunoassay for Hg2+. This strategy was achieved using graphene conjugated with a large number of CdSe quantum dots to enhance the basal signal and enormous horseradish peroxidase (HRP) labeled with gold nanoparticles (AuNPs) to consume the coreactant H2O2 generated in situ. The immunosensor was constructed by immobilization of coating antigen on poly(diallyldimethylammonium chloride) graphene CdSe composites (PDDA-GN-CdSe), and a strong electrochemiluminescence (ECL) signal was obtained. When the immunosensor was immersed in antibody AuNPs HRP composites, the ECL signal greatly decreased, which was ascribed to the bound enzyme on the electrode surface. The self-produced coreactant H2O2 was consumed by o-phenylenediamine in the presence of enzyme, effectively decreasing the ECL intensity from the quantum dots. The Hg2+ in solution and the corresponding coating antigen competed for the limited antibody, and thus, the ECL intensity was linearly dependent on the logarithm of the mercury(II) concentration from 0.2 to 1000 ng mL(-1) with a detection limit of 0.06 ng mL(-1). The immunoassay exhibited good stability and accuracy and acceptable reproducibility, indicating that it provides a promising approach for the detection of trace mercury and other small molecular compounds in environmental samples.
Noble metal-molybdenum disulfide nanohybrids as dual fluorometric and colorimetric sensor for hepatitis B virus DNA detection

Abstract:Hepatitis B virus (HBV) infection is one of the global healthcare burdens, and its early diagnosis is crucial for the prevention of HBV-induced chronic hepatitis, liver fibrosis, cirrhosis, and hepatocellular carcinoma. Although different detection approaches have been reported, most of these methods either rely on sophisticated machines or procedures, which limit their use particularly in the high endemic, developing countries. In this work, we report a dual-sensing nanoplatform built on noble metal-molybdenum disulfide (MoS2) nanohybrids, and this platform can detect the HBV DNA target through either fluorometric or colorimetric readouts. The design with the silver nanocluster (AgNC)–MoS2 nanohybrid enables multiplex fluorescent detection, while the HBV DNA-regulated growth of platinum nanoparticles (PtNPs) on the MoS2 nanosheets offers signal-on colorimetric detection. Both AgNC–MoS2 and PtNP–MoS2 nanohybrids show high sensitivity with pico-molar detection limit and single nucleotide specificity, even with the spiked human serum. Collectively, the proposed nanohybrids possess their potential in the use of early HBV diagnosis, particularly suitable for the high endemic areas with limited medical and instrumental supports.

Abstract:Hepatitis B virus (HBV) infection is considered a major global health problem, causing various health issues, including cirrhosis, hepatitis, and liver cancer. The risk of developing such complications increases when the viral load is above 105 copies/mL. Early identification of HBV infection is imperative to preventing the spread of infection to other parts of the body. Although various sensing methods have been developed to identify HBV, researchers are still working toward developing cheap, easy, and sensitive detection methods. We developed a highly sensitive and rapid HBV detection method using nanomaterials on enzyme-linked immunosorbent assay (ELISA). Aptamer-antibody was utilized as the detection probe and immobilized on a zeolite-modified ELISA plate to detect the HBV biomarker hepatitis B surface antigen (HBsAg). To enhance the detection of HBsAg, aptamer and antibody were attached to gold nanoparticle through electrostatic interaction and immobilized on the zeolite-modified ELISA plate through amine linker. This probe-modified ELISA plate detected low levels of HBsAg, with a detection limit of 0.1 ng/mL. Furthermore, serum spiked experiments showed increment of absorbance with increasing HBsAg concentration, but control trials with other biomolecules showed no increment of absorbance, showing the specific and selective detection of HBsAg. This nanomaterial-modified ELISA plate can detect low levels of HBsAg and help in the diagnosis of HBV infection in its early stages.
Dual-label vertical flow-based electrochemical immunosensor for rapid and simultaneous detection of hepatitis B surface and e virus antigens

Abstract:The detection of hepatitis B virus (HBV)-associated biomarkers, especially hepatitis B surface (HBsAg) and hepatitis B e (HBeAg) antigens, is of critical significance for the early diagnosis and monitoring of disease progression. However, the detection of these biomarkers has seldom been reported. Herein, we first present a dual-label electrochemical vertical flow-based immunosensor (DL-eVFIS) for the simultaneous detection of HBsAg and HBeAg. DL-eVFIS was engineered to integrate multiple time-sequenced detection procedures into a single device, thus considerably shortening the overall assaying time to 6 min. To enable simultaneous determination, thionine (Th) and ferrocenecarboxylic acid (Fc) were employed as signal tags, as they demonstrate nonoverlapping specific reduction peaks. The appearance of target antigens resulted in a substantial increase in the electrochemical signal response of the redox tags. While requiring only a miniscule sample volume (5 μL), superior performance was achieved in terms of their limits of detection (LODs) at 1.66 pg/mL for HBsAg and 5.32 pg/mL for HBeAg. DL-eVFIS further demonstrates excellent specificity and selectivity in clinical sample analysis compared with the conventional method. This study contributes to next-generation point-of-care (POC) testing devices for HBV diagnosis and a wide range of biosensing applications.

Abstract:Au/Ag core-shell nanoparticles labeled by monoclonal antibody and surface-enhanced Raman scattering (SERS) probes were used for immunoassay analysis. Mouse polyclonal antibodies against hepatitis B surface antigen (mouse Anti-HBsAg PAb) were covalently immobilized on the silicon chips modified by a self-assembled monolayer of (3-amino-propyl) trimethoxysilane via glutaraldehyde (GA) activation. The Au/Ag core-shell nanoparticles were labeled by the mouse monoclonal antibody against hepatitis B surface antigen (mouse Anti-HBsAg MAb) and the probe molecule, p-mercaptobenzoic acid (MBA). Immunoreactions between the labeled Au/Ag core-shell nanoparticles and the corresponding antigens captured by the Anti-HBsAg PAb on the silicon chips were investigated by SERS.

Abstract:The combination of supramolecular chemistry and nanotechnology has potentially applied in the construction of biosensors, and thus improves the analytical performance and robustness of electron devices. Herein, a new sandwich-type DNA sensor was constructed for ultrasensitive determination of hepatitis B virus (HBV) DNA, a recognized marker for chronic hepatitis B. The water-soluble pillar[5]arene stabilized Pd NPs combined with reduced graphene oxide nanosheet (WP5-Pd/RGO) was synthesized and employed as supporting material for the modification of electrode surface. The probe DNA was immobilized onto the electrode surface through a new strategy based on the host-guest interaction between WP5 and methylene blue labeled DNA (MB-DNA). Moreover, MOF-derived cobalt sulfide nanobox was prepared to anchor the hydroxylatopillar[5]arene stabilized Au NPs (HP5-Au/CoS), which had superior electrocatalytic performance towards H2O2 reduction to achieve signal amplification. Under the optimized conditions, the proposed sensor displayed a linear relationship between amperometric currents and the logarithm of tDNA solution from 1 × 10-15 mol/L to 1 × 10-9 mol/L, and a low detection limit of 0.32 fmol/L. What's more, the DNA sensor had remarkable behaviors of stability, reproducibility, specificity, and accuracy, which provided a potential and promising prospect for clinical diagnosis and analysis.
New amperometric and potentiometric immunosensors based on gold nanoparticles/tris(2,2'-bipyridyl)cobalt(III) multilayer films for hepatitis B surface antigen determinations

Abstract:Two generic, fast, sensitive and novel electrochemical immunosensors have been developed. Initially, a layer of plasma-polymerized Nafion film (PPF) was deposited on the platinum electrode surface, then positively charged tris(2,2'-bipyridyl)cobalt(III) (Co(bpy)(3)(3+)) and negatively charged gold nanoparticles were assembled on the PPF-modified Pt electrode by layer-by-layer technique. Finally, hepatitis B surface antibody (HBsAb) was electrostatically adsorbed on the gold nanoparticles surface. Electrochemical behavior of the {Au/Co(bpy)(3)(3+)}(n) multilayer film-modified electrodes was studied. Cyclic voltammetry, electrochemical impedance spectroscopy (EIS) were adopted to monitor the regular growth of the multilayer films. The performance and factors influencing the performance of the resulting immunosensors were studied in detail. The multilayer film-modified immunosensor was used for hepatitis B surface antigen (HBsAg) determination via the amperometric and potentiometric immunosensor systems, and both systems provided the same linear ranges from 0.05 to 4.5 microg/mL with different detection limits for the amperometric system 0.005 microg/mL and for the potentiometric system 0.015 microg/mL. The immunosensors were used to analyse HBsAg in human serum samples. Analytical results of clinical samples show that the developed immunoassay is comparable with the enzyme-linked immunosorbent assays (ELISAs) method, implying a promising alternative approach for detecting HBsAg in the clinical diagnosis. In addition, the multilayer films also showed better stability for 1 month at least.

Abstract:As one of the most prominent global public health problems, which seriously threats public health security and human health, hepatitis B virus (HBV) infection is the main cause of liver cirrhosis and cancer. Herein, an innovative and sensitive biosensor for HBV-DNA detection was developed based on the surface etching of the helical gold nanorods (HGNRs) at single-particle level under a dark-field microscope (DFM). In this work, the unique HGNRs with high surface activity were functionalized as the optical probe, which were etched to the smooth gold nanorods and even to gold nanospheres, resulting in a distinct color and light scattering change of single particle with a more sensitive result of HBV-DNA detection than the common gold nanorods. Furthermore, the hybridization chain reaction amplification and DFM technique enabled the proposed strategy with a high sensitivity, offering the limit of detection (LOD) as low as 30.15 fM (3σ/k) and realizing sensitive detection of HBV-DNA from 0.05 to 10 pM. Moreover, the proposed strategy supplied the good specificity, which was capable of distinguishing mismatch sequence and other viral nucleic acids. In addition, this strategy was universal and provided a new idea for early diagnosis of other viral nucleic acids, such as hepatitis C virus (HCV) and human immunodeficiency virus (HIV).

Abstract:Immune-mediated diseases strongly associating with human leukocyte antigen (HLA) alleles are likely linked to specific antigens. These antigens are presented to T cells in the form of peptides bound to HLA molecules on antigen presenting cells, e.g. dendritic cells, macrophages or B cells. The identification of HLA-DR-bound peptides presents a valuable tool to investigate the human immunopeptidome. The lung is likely a key player in the activation of potentially auto-aggressive T cells prior to entering target tissues and inducing autoimmune disease. This makes the lung of exceptional interest and presents an ideal paradigm to study the human immunopeptidome and to identify antigenic peptides. Our previous investigation of HLA-DR peptide presentation in the lung required high numbers of cells (800 × 106 bronchoalveolar lavage (BAL) cells). Because BAL from healthy nonsmokers typically contains 10–15 × 106 cells, there is a need for a highly sensitive approach to study immunopeptides in the lungs of individual patients and controls. In this work, we analyzed the HLA-DR immunopeptidome in the lung by an optimized methodology to identify HLA-DR-bound peptides from low cell numbers. We used an Epstein-Barr Virus (EBV) immortalized B cell line and bronchoalveolar lavage (BAL) cells obtained from patients with sarcoidosis, an inflammatory T cell driven disease mainly occurring in the lung. Specifically, membrane complexes were isolated prior to immunoprecipitation, eluted peptides were identified by nanoLC-MS/MS and processed using the in-house developed ClusterMHCII software. With the optimized procedure we were able to identify peptides from 10 × 106 cells, which on average correspond to 10.9 peptides/million cells in EBV-B cells and 9.4 peptides/million cells in BAL cells. This work presents an optimized approach designed to identify HLA-DR-bound peptides from low numbers of cells, enabling the investigation of the BAL immunopeptidome from individual patients and healthy controls in order to identify disease-associated peptides.

Abstract:The detection of hepatitis B surface antigen (HBsAg) with an immunoassay using magnetite nanoparticles (NPs) that bind hepatitis B surface antibody (anti-HBs) was investigated. Magnetite NPs with an average size of 32 nm were synthesized and functionalized using tetraethyl orthosilicate, 3-aminopropyltriethoxysilane, glutaraldehyde, and streptavidin. The functionalized NPs were conjugated with biotinylated anti-HBs to capture HBsAg. HBsAgs were detected with a sensitivity, specificity and agreement of 98%, 97%, and 97.5%, respectively, when compared to the rapid test. In addition, the novel method could also enrich HBsAgs, using different tube sizes and can facilitate amplification for the detection of microorganisms.
