Abstract:Background/Aims: Fatty acid synthase (FAS) and human epidermal growth factor receptor 2 (HER2) are overexpressed in a series of cancers. However, few studies have investigated the potential role of FAS and HER2 in esophageal carcinoma. The aim of this study was to evaluate the expression of FAS and HER2 and the possible link between FAS/HER2 expression and the pathological prognostic varibles. Methodology: The frequency of FAS and HER2 expression was determined immunohistochemically. The overall survival rate was analysed by the Kaplan-Meier method and the log-rank test using SPSS 17.0 software. Results: The majority of the cases were esophageal squamous cell carcinomas (n=142). FAS and HER2 overexpression in the studied cases are 73.2% and 14.1%, respectively. There was a significant difference in FAS expression regarding tumor differentiation and FAS overexpression showed its prognostic value for patients with different tumor differentiation. Meanwhile, HER2 overexpression did not significantly relate to the clinicopathological characteristics of the tumors, with the only exception of the surgical margins. Conclusions: FAS and HER2 overexpression are common in esophageal carcinomas. FAS overexpression showed its prognostic value for esophageal carcinoma patients with different tumor differentiation, which lead us to consider FAS-inhibitors as potential candidates for target-based adjuvant therapies.

Abstract:Purpose: Lipid metabolism reprogramming is a major pathway in tumor evolution. This study investigated fatty acid synthase (FASN) mRNA expression in anthropometric adipose tissue and elucidated the prognostic value and potential mechanism of clear cell renal cell carcinoma (ccRCC). Materials and Methods: Transcription profiles were obtained from 533 ccRCC samples in The Cancer Genome Atlas (TCGA) cohorts. Real-time quantitative PCR (RT-qPCR) and immunohistochemistry were performed to detect FASN expression in 380 paired ccRCC and normal tissues from the Fudan University Shanghai Cancer Center (FUSCC). Visceral adipose tissue (VAT) and subcutaneous adipose tissue were at the level of the umbilicus as measured by magnetic resonance imaging (MRI). Non-targeted metabolomics and in vitro experiments were used to reveal the biological functions of FASN. Results: Increased FASN expression was significantly relevant to advanced T, N, and American Joint Committee on Cancer (AJCC) stages ( p < 0.01) and significantly correlated to poor progression-free survival (PFS) and overall survival (OS) of 913 ccRCC patients in FUSCC and TCGA cohorts. Pearson's correlation coefficient indicated that FASN amplification was positively correlated to VAT% ( r = 0.772, p < 0.001), which significantly correlated to poor PFS (HR = 2.066, p = 0.028) and OS (HR = 2.773, p = 0.023) in the FUSCC cohort. Transient inhibition or overexpression of FASN significantly regulated A498 and 786O cell proliferation and migration by regulating epithelial–mesenchymal transition. Inhibition of FASN led to a higher apoptotic rate and decreased lipid droplet formation compared with normal control in ccRCC cells. Non-targeted metabolomics showed that decreased de novo lipogenesis might be required to sustain an elevation of glycolytic activity in 786O cells by regulating galactinol, dl -lactate, N-acetylaspartylglutamate, and sucrose, thereby participating in carcinogenesis and progression of ccRCC. Conclusion: This study demonstrated that FASN expression is positively related to aggressive cell proliferation, migration, apoptosis, and lipid droplet formation and regulates metabolic disorders of the ccRCC microenvironment. Additionally, elevated FASN mRNA expression is significantly correlated to the abdominal obesity distribution, especially VAT%, which is a significant predictor of a poor prognosis for ccRCC patients.

Abstract:We studied a Japanese patient who developed parkinsonian symptoms over 3 months before the diagnosis of acquired immunodeficiency syndrome. Brain MRI showed multiple lesions with mass effect and ring enhancement in the basal ganglia and subcortical white matter suggesting Toxoplasma infection. Anti-Toxoplasma therapy and highly active antiretroviral therapy for 6 months allowed improvement of parkinsonism, brain MRI findings, and immune system.
Identification of MicroRNA Expression Profiles Related to the Aggressiveness of Salivary Gland Adenoid Cystic Carcinomas

Abstract:Adenoid cystic carcinoma (ACC) has been reported as the second most common carcinoma of the salivary glands. Few studies have associated miRNA expression with ACC aggressiveness. In this study, we evaluated the miRNA profile of formalin-fixed, paraffin-embedded (FFPE) samples of salivary gland ACC patients using the NanoString platform. We studied the miRNA expression levels associated with the solid growth pattern, the more aggressive histologic feature of ACCs, compared with the tubular and cribriform growth patterns. Moreover, the perineural invasion status, a common clinicopathological feature of the disease that is frequently associated with the clinical progression of ACC, was investigated. The miRNAs showing significant differences between the study groups were selected for target prediction and functional enrichment, which included associations with the disease according to dedicated databases. We observed decreased expression of miR-181d, miR-23b, miR-455, miR-154-5p, and miR-409 in the solid growth pattern compared with tubular and cribriform growth patterns. In contrast, miR-29c, miR-140, miR-195, miR-24, miR-143, and miR-21 were overexpressed in patients with perineural invasion. Several target genes of the miRNAs identified have been associated with molecular processes involved in cell proliferation, apoptosis, and tumor progression. Together, these findings allowed the characterization of miRNAs potentially associated with aggressiveness in salivary gland adenoid cystic carcinoma. Our results highlight important new miRNA expression profiles involved in ACC carcinogenesis that could be associated with the aggressive behavior of this tumor type.
Expression and Clinical Significance of MAPK,MMP-2 and MMP-9 in Salivary Adenoid Cystic Carcinoma

Abstract:PURPOSE: To investigate the expression of MAPK(p-ERK1/2、p-P38 and p-JNK),matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in human salivary adenoid cystic carcinoma(SACC) tissues,and to explore their relation with clinicopathological features.METHODS: SP immunochemistry staining method was employed to detect protein expression of MAPK,MMP-2 and MMP-9 in the paraffin-embedded specimens from 27 cases with human SACC and 15 cases with normal human salivary epithelial tissues.All data were analyzed by SPSS17.0 software package.RESULTS: Positive expression rates of p-ERK1/2,p-P38,p-JNK,MMP-2 and MMP-9 were 81.48%,88.89%,70.37%,81.49% and 77.78% in SACC tissues and 20%,26.67%,26.67%,26.67% and 26.67% in normal salivary tissues,respectively.The expression of p-ERK1/2,p-P38,p-JNK,MMP-2 and MMP-9 in SACC tissues increased compared with normal salivary tissues respectively(P0.01,P0.01,P0.05,P0.01 and P0.01).Positive expression rates of p-ERK1/2,p-P38,MMP-2 and MMP-9 at the third and forth clinical stage(88.89%,100%,94.44% and 94.44%) were higher than that at the first and second clinical stage(44.44%,66.67%,55.56% and 44.44%) in SACC.The differences were statistically significant(P0.01,P0.01,P0.01 and P0.05,respectively);Positive expression rate of p-JNK was not significantly correlated with clinical stages(P0.05);Positive expression rates of p-ERK1/2,p-P38,p-JNK,MMP-2 and MMP-9 were not significantly correlated with patient's gender,age,primary site,pathological type,neural invasion,local recurrence as well as distant metastasis(P0.05).p-ERK1/2,p-P38 and p-JNK had a positive relationship with MMP-2 and MMP-9 in protein expression in SACC,respectively(r=0.786,P0.001;r=0.796,P0.001;r =0.824,P0.001 and r=0.768,P0.001;r=0.581,P0.01;r=0.604;P0.01,respectively).CONCLUSION: Increased expression of p-ERK1/2,p-P38,p-JNK,MMP-2 and MMP-9 may be associated with clinical progression of SACC,and they may participate in the process of SACC initiation,progression,invasion and metastasis by activating the MAPK/MMPs pathway.
Expression of Cancer Cell-Derived IgG and Extra Domain A-containing Fibronectin in Salivary Adenoid Cystic Carcinoma.

Abstract:Objective: Cancer-IgG is a newly-discovered molecule, mainly derived from epithelial carcinoma cells and is significantly correlated with differentiation, metastasis, local invasion, and poor prognosis of many cancers. In our previous study we detected IgG expression in oral epithelial carcinoma, including salivary adenoid cystic carcinoma (SACC), using an IgG-specific commercial antibody. Here, we explored the correlation between cancer-IgG and clinicopathological features of SACC.Design: A total of 68 human SACC tissue specimens and 2 siRNAs were used to analyze the correlation between cancer-IgG and extra domain A (EDA(+))-containing fibronectin using the cancer-IgG-specific monoclonal antibody, RP215.Results: We found an unexpected correlation between cancer-IgG and EDA(+) fibronectin, both of which showed aberrant expression in SACC tissue samples. Both were highly expressed in SACC with nerve invasion. In our previous study, EDA(+) fibronectin overexpression in SACC cells decreased N-cadherin expression. In the present study, we used SACC-83 cells, wherein EDA(+) fibronectin is overexpressed and cancer-IgG is knocked down. EDA(+) fibronectin expression was reduced with cancer-IgG knockdown, while cancer-IgG expression did not affect EDA(+) fibronectin overexpression. Furthermore, knockdown of non-B cell-derived IgG in SACC cells decreased cellular motility (P < 0.05) as well as increased E-cadherin and alpha-smooth muscle actin levels.Conclusion: The results suggest that cancer IgG potentially regulates EDA(+) fibronectin, expression, thereby suggesting possible new therapeutic approaches for SACC.
Expression of Neural Cell Adhesion Molecule in 49 Cases with Salivary Adenoid Cystic Carcinoma and Its Significance

Abstract:[Purpose]To investigate expression of neural cell adhesion molecule (NCAM) in salivary adenoid cystic carcinoma(SACC) and its clinical significance. [Methods] The expression of NCAM in 49 cases with SACC was detected by immunohistochemical technique. [Results] The positive rate of NCAM was 57.14%(28/49). NCAM expression was not related to gender, tumor site and staging. NCAM expressions in SACC tubular subtype and in solid subtype were significantly different (χ2=4.07,P=0.04). NCAM expression with neural invasion was significantly higher than that without neural invasion (P0.05). [Conclusion] The NCAM closely correlates to neural invasion in SACC. NCAM plays an important role in neural invasion of SACC.
TGFβ signaling pathway in salivary gland tumors

Abstract:Objective Pleomorphic adenoma (PA), mucoepidermoid carcinoma (MEC), and adenoid cystic carcinoma (ACC) are the most prevalent salivary gland tumors. Their pathogenesis has been recently associated with complex molecular cascades, including the TGFβ signaling pathway. The aim of this study was to evaluate the expression of genes associated with the TGFβ signaling pathway ( TGFB1 , ITGB6 , SMAD2 , SMAD4 , FBN1 , LTBP1 , and c-MYC ) to map possible downstream alterations in the TGFβ cascade. Design Thirteen PA, 17 MEC, 13 ACC, and 10 non-neoplastic salivary gland samples were analyzed by real-time RT-PCR. Results Cases of PA presented increased TGFB1 , LTPB1 , c-MYC , and FBN1 expressions, whereas SMAD2 expression was decreased when compared to non-neoplastic tissue. MEC patients displayed increased expressions of TGFB1 , ITGB6 , FBN1 , and c-MYC and decreased expressions of SMAD2 and SMAD4 . ACC cases exhibited elevated expressions of the investigated genes except TGFB1 . The present results suggest that decreased expression of SMAD2 and SMAD4 does not impede the transcriptional regulation of c-MYC , especially in PA and MEC. Increased expressions of ITGB6 , TGFB1 , LTBP1 , and FBN1 appear to be related to the regulation of the TGFβ signaling pathway in these tumors. Additionally, we observed a higher expression of SMAD4 in ACC and a raised expression of ITGB6 and lowered expression of SMAD2 in MEC. Conclusions Our study demonstrated the differential expression of TGFβ cascade members in salivary gland tumors such as SMAD2/SMAD4 and c-MYC as well as the participation of ITGB6 , TGFB1 , LTBP1 , and FBN1 , contributing to the understanding of the mechanisms involved in tumor progression.

Abstract:Abstract Background: Growing evidence has indicated obesity one of the important etiological indicators of clear cell renal cell carcinoma (ccRCC). This study aims to investigated FASN mRNA expression in anthropometric adipose tissue, and elucidate prognostic value and potential functions in ccRCC patients. Methods: Transcriptional expression profiles was obtained from 380 paired ccRCC and adjacent normal tissues from Fudan University Shanghai Cancer Center (FUSCC) and 533 ccRCC samples from the Cancer Genome Atlas (TCGA) cohort. The visceral adipose tissue (VAT) and the subcutaneous adipose tissue (SAT) at the level of the umbilicus were measured using magnetic resonance imaging. The Kaplan-Meier method and log-rank test, with cooperation from Cox regression analysis, were used in survival analysis. Subsequently, we transiently transfected FASN plasmid into A498 and 786O cells, and investigated the role of FASN in ccRCC cell proliferation, apoptosis and migration in vitro. Related hub gens, functional annotations and significant signal pathways were predicted using integrated bioinformatics. Results: FASN mRNA expression was significantly higher in tumor than normal tissues in 913 ccRCC patients from FUSCC and TCGA cohorts. In addition, increased FASN mRNA expression was significantly relevant to advanced T stage ( p <0.001), N stage ( p =0.019), and AJCC stage ( p =0.002). Pearson’s correlation coefficient indicated that FASN amplification positively correlated with VAT% ( r =0.772, p <0.001). VAT% was significantly correlated with poor PFS and OS, with hazard ratios of 2.066 (1.113-7.261, p =0.028) and 2.773 (1.168-8.974, p =0.023) in FUSCC. Meanwhile, ccRCC patients with elevated FASN expression significantly responded for poor PFS and OS, with hazard ratios of 1.529 (1.135-2.061, p =0.005) and 1.450 (1.030-2.041, p =0.033). Next, after transfection efficiency was verified in A498 and 786O cells, we found transient inhibition and overexpression of FASN significantly regulates cells proliferation and migration abilities, and inhibition of FASN displayed higher apoptotic rate in ccRCC cells. Conclusion: In conclusion, this study demonstrated that FASN mRNA expression is positively related with aggressive proliferation, migration progression and predicts prognosis of ccRCC. In addition, we first reveal that elevated FASN mRNA expression is significantly correlated with abdominal obesity distribution, especially VAT%, which also is a significant predictor of poor prognosis in patients with ccRCC.
E‐CADERIN, N‐CADERIN, SLUG, SNAIL, and TWIST contribute to epithelial‐mesenchymal transition in salivary gland tumors

Abstract:Background Transcription factors are important in the epithelial‐mesenchymal transition process and are possibly related to the development of a more invasive tumor phenotype. Thus, the objective of this study was to analyze the expression and identify the localization of cellular markers related to the epithelial‐mesenchymal transition process in salivary gland tumors. Study Design The expression and localization of E‐CADERIN, N‐CADERIN, SLUG, SNAIL, and TWIST were evaluated, using immunohistochemistry, in 48 salivary gland tumors, being 17 pleomorphic adenomas (PA), 14 adenoid cystic carcinomas (ACC), and 17 mucoepidermoid carcinomas (MEC). these proteins were compared to clinical and histopathologic parameters. normal gland tissues were included for immunohistochemical comparisons. Results ACC and MEC cases showed higher expression of SNAIL compared to PA. MEC showed high expression of SLUG and TWIST. Low expression of N‐CADHERIN, SNAIL, and TWIST in ACC was frequent in T3 and T4. High expression of TWIST in MEC was more frequent at age ≥ 40 years A positive correlation was only observed between N‐cadherin/SNAIL in ACC, between SNAIL/TWIST in MEC, and between SLUG/TWIST in PA. Conclusion This study provided insight into EMT‐related proteins (E‐cadherin, N‐cadherin, SNAIL, SLUG, and TWIST) and their contribution to the maintenance of morphogenesis and the development of the salivary gland tumors and showed a positive correlation among N‐CADHERIN/SNAIL in ACC and SNAIL/TWIST in MEC.

Comprehensive Analysis of FASN in Tumor Immune Infiltration and Prognostic Value for Immunotherapy and Promoter DNA Methylation

Abstract:Fatty acid synthase (FASN) promotes tumor progression in multiple cancers. In this study, we comprehensively examined the expression, prognostic significance, and promoter methylation of FASN, and its correlation with immune cell infiltration in pan-cancer. Our results demonstrated that elevated FASN expression was significantly associated with an unfavorable prognosis in many cancer types. Furthermore, FASN promoter DNA methylation can be used as a tumor prognosis marker. Importantly, high levels of FASN were significantly negatively correlated with tumor immune infiltration in 35 different cancers. Additionally, FASN was significantly associated with tumor mutational burden (TMB) and microsatellite instability (MSI) in multiple malignancies, suggesting that it may be essential for tumor immunity. We also investigated the effects of FASN expression on immunotherapy efficacy and prognosis. In up to 15 tumors, it was significantly negatively correlated with immunotherapy-related genes, such as PD-1, PD-L1, and CTLA-4. Moreover, we found that tumors with high FASN expression may be more sensitive to immunotherapy and have a good prognosis with PD-L1 treatment. Finally, we confirmed the tumor-suppressive effect of mir-195-5p through FASN. Altogether, our results suggested that FASN may serve as a novel prognostic indicator and immunotherapeutic target in various malignancies.
Immunohistochemical expression of OCT4 and CD44 in major and minor salivary gland neoplasms

Abstract:The aim of this study was to identify tumor parenchyma cells exhibiting immunohistochemical profile of stem cells by evaluating the immunoreactivity of OCT4 and CD44 in a number of cases of salivary gland neoplasms. The sample consisted of 20 pleomorphic adenomas, 20 mucoepidermoid carcinomas, and 20 adenoid cystic carcinomas located in major and minor salivary glands. The expression of OCT4 and CD44 was evaluated by the percentage of positive cells and the intensity of expression. All studied cases showed positive expression of OCT4 and CD44 and higher values than the control groups. For OCT4, luminal and non-luminal cells were immunostained in the case of pleomorphic adenomas and adenoid cystic carcinomas. Moreover, the immunoreactivity of CD44 was particularly evident in the non-luminal cells of these lesions. In mucoepidermoid carcinomas, there was immunoreactivity for both markers in squamous and intermediate cells and absence of staining in mucous cells. For both markers, a significantly higher immunostaining was verified in neoplasms located in the major salivary glands compared with lesions in minor salivary glands (p<0.001). In the total sample and in minor salivary glands, malignant neoplasms exhibited higher immunoreactivity for OCT4 than pleomorphic adenoma. A significant moderate positive correlation (r = 0.444 and p ≤ 0.001) was found between OCT4 and CD44 immunoexpression in the total sample. The high expression of OCT4 and CD44 may indicate that these proteins play an important role in identifying tumor stem cells.

Abstract:Objective: To perform a comparative study focusing on the morphologic and immunohistochemical features of pleomorphic adenomas (PAs) and carcinoma ex pleomorphic adenomas (CXPAs) of the minor salivary glands. Study design: Forty PAs and 10 CXPAs were retrieved from the archives of 3 pathology laboratories, and the diagnoses were reviewed and confirmed. Immunohistochemical reactions were done for p53, Ki67, mammaglobin, and the metabolic markers of glucose transporter 1 (GLUT-1) and adipophilin. Results: The most common subtypes of CXPA were ductal carcinoma (n = 4), clear cell carcinoma (n = 3), and myoepithelial carcinoma (n = 2). The morphologic features suggestive of malignancy were the presence of mitosis, cellular pleomorphism, and extracapsular component. Compared to PA, CXPA demonstrated a higher number of Ki67- and p53-positive cells, in addition to higher expression of GLUT-1 and adipophilin. Mammaglobin expression was observed in the malignant areas, mainly in the ductal CXPA, being negative in the benign reminiscent PA. Conclusion: The diagnosis of CXPA based only on the morphologic aspects can be difficult; thus, an association of the morphologic and immunohistochemical features, particularly high number of Ki67- and p53-positive cells, is essential for a proper diagnosis. Other metabolic markers such as GLUT-1 and adipophilin may be also helpful, although their exact utility remains to be determined.
A Concordant Expression Pattern of Fatty Acid Synthase and Membranous Human Epidermal Growth Factor Receptor 2 Exists in Gastric Cancer and is Associated with a Poor Prognosis in Gastric Adenocarcinoma Patients

Abstract:Fatty acid synthase (FAS) and human epidermal growth factor receptor 2 (HER2) are overexpressed in gastric cancer (GC), and certain interactions have been found between FAS and HER2. A total of 94 patients were enrolled in the present study, each of whom underwent a D2 radical surgery in Zhongshan Hospital affiliated with Fudan University (Shanghai, China) between 2000 and 2005. The expression of FAS and HER2 was assessed by immunohistochemistry analysis of tissue microarrays generated from GC and non-tumor tissues. All data were analyzed by GraphPad Prism 5.0 to investigate the association between FAS and HER2 and to detect the potential association with prognosis. FAS (P<0.0001) and membranous HER2 (mHER2; P=0.0021) were overexpressed in the GC tissues, and a bidirectional and strong correlation was demonstrated between FAS and mHER2 in the tumor tissues. The expression of cytoplasmic HERZ (cHER2) was significantly lower in the GC tissues compared with the non-tumor tissues (P=0.0005), and cHER2 was expressed at a higher level in tumors that had better differentiation compared with poorly-differentiated tissues (P=0.0503). Patients with a concordant expression pattern of FAS and mHER2 showed a significantly poorer prognosis than the non-concordant group (P=0.0096; hazards ratio, 3.2801; 95% confidence interval, 1.5781-6.8176). GC tissues significantly overexpress FAS and mHER2 and the expression of these two markers is associated. Patients with a concordant expression of FAS and mHER2 are more likely to suffer a poor prognosis.

Abstract:Objectives To conduct a comprehensive proteomic analysis of normal salivary gland tissue, pleomorphic adenoma (PA), and carcinoma ex‐pleomorphic adenoma (CXPA), and validate the proteomic findings using immunohistochemistry. Methods Six normal salivary gland tissues, seven PA and seven CXPA samples underwent laser microdissection followed by liquid chromatography coupled to mass spectrometry. Protein identification and quantification were performed using MaxQuant software. Statistical analysis and functional enrichment were conducted using the Perseus platform and STRING tool, respectively. Immunohistochemistry was used for validation. Results Comparative proteomic analysis revealed 2680 proteins across the three tissue types, with 799 significantly altered between groups. Translocation protein SEC63 homolog, Annexin A6 and Biglycan were up‐regulated in CXPA compared to PA. Decorin was markedly up‐regulated in both PA and CXPA compared to normal salivary gland (log2 fold changes of 7.58 and 7.38, respectively). Validation confirmed elevated levels of Biglycan and Decorin in the extracellular matrix of CXPA compared to PA. Conclusions Proteomic analysis identified differential protein expression patterns associated with malignant transformation of PA into CXPA. Findings indicate a crucial role for extracellular matrix proteins, specifically Biglycan and Decorin, in the tumorigenic progression of PA and CXPA.

Abstract:PURPOSE:To investigate the expression of ErbB3 binding protein ebp1, E-cadherin, ICAM-1 and matrix metalloproteinase-9(MMP-9) in salivary adenoid cystic carcinoma(SACC), and to explore their relationship with clinical pathological features.METHODS:Two-step immunohistochemical staining method was used to detect the expression of ebp1 E-cadherin, ICAM-1 and MMP-9 in 33 cases with human SACC and 33 with para-cancerous normal tissues. All data were analyzed with SPSS17.0 software package.RESULTS:Positive expression rate of ebp1 in SACC was 84.85%, lower than in normal salivary tissues(96.97%). Ebp1 expression was significantly correlated to pathological pattern and clinical stage(P<0.05), but not correlated to gender and age. Positive expression rate of ebp1 at I-II stage was higher than that of SACC at III-IV stage; in regard to pathological typing, higher expression was found in adenoid tubular type than in solid type; the positive expression rate in patients with tumor metastasis was lower than in patients without metastasis (P<0.05). Expression of ebp1 had a positive relationship with E-cadherin (r=0.851,P<0.01), while a negative relationship was found with MMP-9 (r=-0.364,P<0.05).CONCLUSIONS:Expression of ebp1 may be associated with progression of SACC. Ebp1 has important role in the generation and evolution of adenoid cystic carcinoma, and can be used as a useful indicator for clinical assessment of tumor biological behavior and prognosis in patients with adenoid cystic carcinoma.

Abstract:Background:Aberrant crypt foci(ACF) is the earliest morphological precursor of epithelial neoplasia in colorectum, which may progress through adenoma to carcinoma.Fatty acid synthase(FAS) is highly expressed in most human carcinomas.Aims:To investigate the expression of FAS in rectal ACF in patients with colon cancer,and to study the significance of FAS overexpression in ACF in the pathogenesis of colorectal cancer.Methods:Thirty patients with sporadic colon cancer confirmed pathologically were enrolled in this study.The rectal mucosa was stained with 0.2% methylene blue and ACF was identified by magnifying colonoscopy.Large ACF(>30 crypts/focus) was selected and biopsied.Expression of FAS in rectal ACF and paired colon cancer tissue and normal colonic mucosa was determined by immunohistochemistry.Results:A total of 116 large ACF were selected and biopsied in 30 colon cancer patients,25 with dysplasia and 91 without dysplasia.Colon cancer tissue had a significantly higher positivity rate for FAS than ACF and normal mucosa(76.7%vs.32.8%and 10.0%,P<0.05);significant difference was also seen between ACF and normal mucosa(P<0.05).Expression rate of FAS was not different between ACF with and without dysplasia(36.0%vs.31.9%, P>0.05).Conclusions:FAS overexpression occurs early and plays an important role in the adenoma-carcinoma sequence of colorectal cancer.Inhibiting the progression of ACF by FAS inhibitors may provide an approach for chemoprevention of colorectal cancer.

Abstract:BackgroundSalivary duct carcinoma (SDC) is a highly aggressive disease which often metastasizes to distant sites, and there is no established standard therapy for this systemic disease. Given that SDC is biologically similar to breast and prostate cancer, anti-androgenic receptor (AR) and anti-human epidermal growth factor receptor 2 (HER2) therapies have the potential to exert effects, not only on patients with breast and prostate cancer but also on those with SDC.MethodsThe expression levels of HER2, epidermal growth factor receptor (EGFR), Ki-67, and AR were assessed in 32 patients with SDC, and their correlations with overall survival (OS) and disease-free survival (DFS) were analyzed retrospectively. SDC was classified into five subtypes using a method similar to that used for breast cancer.ResultsAnti-AR, HER2, and EGFR were positive in 23 (71.9 %), 14 (43.8 %), and 26 (81.3 %) cases, respectively. One or more of these 3 factors were positive in 30 (93.8 %) cases. The Ki-67 labeling index was greater than 15 % in all cases. While molecular status did not correlate with OS, EGFR and AR positivity were significantly associated with DFS in univariate analysis. Multivariate analysis revealed that EGFR was the only independent predictor of DFS.ConclusionsThe statuses of some molecules are useful to predict DFS in patients with SDC. Ki-67 overexpression suggests that cytotoxic agents are effective for SDC. Since the majority of SDCs express AR, HER2, and/or EGFR, assessing and targeting these molecules are promising strategies to improve the prognosis of unresectable, metastatic or recurrent SDC, and a classification system according to the molecular expression status may be useful to select appropriate therapy.

Abstract:Objective To investigate the expression of TGF-β1, MAPK (p-ERK1/2, p-p38 and p-JNK) in human salivary adenoid cystic carcinoma (SACC) tissues, and to explore their association and relation with clinical-pathological features. Methods SP immunochemistry staining method was applied to detect protein expression of TGF-β1, MAPK in the paraffin-embedded specimens from 27 cases of human SACC and 15 cases of normal human salivary epithelial tissues. All data were analyzed by SPSS 17.0 software package. Results The positive expression of TGF-β1, MAPK in SACC tissues increased compared with that in normal salivary tissues (P < 0.01, P < 0.01, P < 0.01 and P < 0.05 respectively). Positive expression rates of TGF-β1, p-ERK1/2 and p-p38 at the third and forth phases was higher than that at the first and second phases in SACC (P < 0.01, P < 0.05 and P < 0.05 respectively), except p-JNK(P > 0.05); their expression was not obviously correlated with patient′s other clinical-pathological features (P > 0.05); TGF-β1 had a positive relationship with MAPK in protein expression in SACC (r = 0.819, P < 0.001, r = 0.728, P < 0.001 and r = 0.640, P < 0.05 respectively). Conclusions Increased expression of TGF-β1, MAPK may be associated with clinical progression of SACC, and they may participate in the process of SACC initiation, progression, invasion and metastasis by TGF-β1 activated MAPK.
