Abstract:SummaryThis study investigated the aroma volatiles, sensory and chemical attributes of strawberry achenes and receptacle. Samples were subjected to chemical analysis in terms of total soluble solids (TSS), total acidity (TA), total phenolics, anthocyanin content, antioxidant capacity and volatile compounds. Descriptive tests indicated that fresh whole fruit had the best overall quality. Chemical evaluations revealed that the receptacle emitted more aroma volatiles but exhibited lower TSS, phenolics and antioxidant capacity than the achenes. Moreover, achenes plus receptacle contained higher levels of phenolics and antioxidant capacity and showed moderate aroma volatile profiles than frozen whole fruit. The processing of juice showed the higher loss of phenolics, antioxidant capacity and aroma volatiles. Achene induced a significant increase in phenolics and antioxidant capacity. Aroma volatiles were reduced by freezing. These results suggested that the fresh whole fruit puree exhibited better overall quality, which had better sensory attributes, more aroma volatiles and higher antioxidant capacity.

Abstract:Profound metabolic and proteomic changes involved in the primary and the secondary metabolism are required for the ripeness of fleshy fruit such as strawberries (Fragaria x ananassa). Here we present the quantitative proteomic profiling in parallel with metabolic and transcriptional profiling at five developmental stages of strawberry fruit ripening, and correlations between changes in representative metabolites and the abundance of related proteins were analyzed. Hierarchical clustering analysis of the quantitative proteomic profiling identified 143 proteins in strawberry fruit across five developmental stages. Meanwhile, both protein abundance and gene expression spanned a wide range of roles, such as the primary and the secondary metabolism, defense system, and response to stress stimuli. The decreased abundance of proteins contributed to the carbohydrate metabolism and the up-regulated expression of secondary biosynthetic proteins was found to be positively correlated with the accumulation of primary and secondary metabolites during strawberry development. Moreover, with the same annotations and high homology, the gene function of key genes involved in primary and secondary metabolism (FaTPI, FaPAL, FaMDH and FaME) was confirmed in Nicotiana via the transient expression assay, which provides further evidence for the role of those genes in metabolism of strawberry fruit. The results of the present study may serve as an important resource for the functional analysis of the proteome and offer new perspectives on regulation of fruit quality.

Abstract:The strawberry is one of the world's most popular fruits, providing humans with vitamins, fibers, and antioxidants. Cultivated strawberry (Fragaria × ananassa) is an allo-octoploid and highly heterozygous, making it a challenge for breeding, QTL mapping, and gene discovery. Some wild strawberry relatives, such as Fragaria vesca, have diploid genomes and are becoming laboratory models for the cultivated strawberry. Recent advances in genome sequencing and CRISPR-mediated genome-editing have greatly improved the understanding of various aspects of strawberry growth and development in both cultivated and wild strawberries. This review focuses on fruit quality traits that are most relevant to the consumers, including fruit aroma, sweetness, color, firmness, and shape. Recently available phased-haplotype genomes, SNP arrays, extensive fruit transcriptomes, and other big data have made it possible to locate key genomic regions or pinpoint specific genes that underlie volatile synthesis, anthocyanin accumulation for fruit color, and sweetness intensity or perception. These new advances will greatly facilitate marker-assisted breeding, the introgression of missing genes into modern varieties, and precise genome editing of selected genes and pathways. Strawberries are poised to benefit from these recent advances, providing consumers with fruit that is tastier, longer-lasting, healthier, and more beautiful.

Effect of Exogenous Sucrose on Anthocyanin Synthesis in Postharvest Strawberry Fruit.

Abstract:Sucrose acts as a vital signal that modulates fruit ripening. In current study, 50 mM sucrose was applied in strawberry fruit to investigate the regulation of sucrose in anthocyanin synthesis after harvest. The results showed that sucrose treatment increased the contents of glucose, fructose and sucrose, which were 19.76%, 15.83% and 16.50% higher, respectively, compared with control at the end of storage. The increase of glucose and fructose contents resulted from the activation of acid invertase by sucrose treatment. In addition, sucrose treatment specifically increased four pelargonidin derivatives, pelargonidin 3-glucoside, pelargonidin 3-rutinoside, pelargonidin 3-malonylglucoside and pelargonidin 3-methylmalonyglucoside, during the storage. Further, transcriptional profiles and enzyme activities analysis revealed that the accumulation of pelargonidin derivatives was related to the activation of the pentose phosphate pathway, shikimate pathway, phenylpropanoid pathway, and flavonoid pathway. These results provided new insights into the regulation of sucrose on the accumulation of individual anthocyanins.
Inheritance of esters and other volatile compounds responsible for the fruity aroma in strawberry

Abstract:Cultivated strawberry, Fragaria × ananassa , has a complex aroma due to the presence of more than 350 volatile organic compounds (VOCs). However, a mixture of only 19 compounds, called Key Volatile Compounds (KVC), can impart the main strawberry aroma. The octoploid nature of the cultivated strawberry species (2 n = 8 x = 56) adds complexity to the heritance of the accumulation of the volatiles responsible for aroma. An F1 population cross between two breeding parental lines, FC50 and FD54, was phenotyped for aroma by SPME GCMS during six harvests. A total of 58 compounds were identified: 33 esters, nine terpenes, seven aldehydes, four lactones, two furans, one acid, one alkane and one alcohol, of which 16 were KVCs. A total of 179 QTLs were found, and 85 of these were detected in at least three harvests, of which 50 QTLs were considered major (LOD > 4.0) and detected in five or six analyzed harvests. Several clusters of ester QTLs associated with fruity aroma were discovered, such as QTLs for esters that share hexanoate group that were mapped in LG4A ( Hexanoate_4A ), those that share acetate and octyl groups in LG6A ( Acetate_6A and Octyl_6A ) or those with the same methyl group in LG7B ( Methyl_7B ). Different terpene QTLs associated with floral aroma appear grouped in a cluster in LG3C ( Terpene_3C ). Some of these clusters of QTLs were validated in a second F2 population, a cross of "Camarosa" and "Dover," that was also phenotyped for three years. Selected SNPs from floral and fruity aroma QTLs were tested in a third population, which will most likely be useful for marker-assisted breeding (MAB).

Abstract:Strawberries are an economically important crop partly because they are very popular with consumers. Strawberry growers and breeders would benefit from knowing which cultivars and fruit traits consumers would prefer. The objectives of this study were to (1) determine consumer preference for the fruit of once-fruiting and repeat-fruiting strawberry cultivars available to grow in the Mid-Atlantic to help growers select the most suitable strawberry cultivars based on their marketability, critical sensory quality attributes, and physicochemical measurements; and (2) identify interrelationships between sensory attributes and physicochemical measurements to determine which approaches might result in breeding strawberries consumers will find most appealing. Strawberries from 10 cultivars were harvested and evaluated by a trained sensory panel and instrumentally. 'Flavorfest' fruits were rated highest of all 10 cultivars, and 'Albion' fruits were rated highest of the repeat-fruiting cultivars. Overall quality was correlated with sweetness and strawberry flavor and aroma, but not sourness. Separation of cultivar means for sweetness was better using an instrument to measure, rather than the panel evaluation. Overall appearance was correlated with visual freshness and glossiness. Light orange-red strawberries (higher L* and b*) appeared fresher and more appealing to the panels than darker more purple-red strawberries. Evaluations of strawberry fruits for berry size, juiciness, and texture were less straightforward.

Abstract:A quantitative proteomic investigation of strawberry fruit ripening employing stable isotope labeling by peptide dimethylation was conducted on ‘Mira’ and ‘Honeoye’ strawberry fruit. Postharvest physiological quality indices, including volatile production, total phenolics, total anthocyanins, antioxidant capacity, soluble solids and titratable acidity, were also characterized in white, pink and red fruit. More than 892 and 848 proteins were identified and quantified in the ‘Mira’ and ‘Honeoye’ fruit, respectively, using at least two peptides for each protein identification. Using the normalized ratio of protein abundance changes, proteins that changed two-fold or more were identified as proteins that are up- or down-regulated during fruit ripening. Among the quantified proteins, 111 proteins were common to both cultivars and represented five significant clusters based on quantitative changes. Among the up-regulated proteins were proteins involved in metabolic pathways including flavonoid/anthocyanin biosynthesis, volatile biosynthesis, antioxidant metabolism, stress responses and allergen formation. Proteins that decreased during fruit ripening were found to be responsible for methionine metabolism, antioxidant-redox, energy metabolism and protein synthesis. Our results show that strawberry ripening is a highly complex system involving multi-physiological processes made possible through changes in protein expression. This study provides new insights on the regulation of proteins during strawberry fruit ripening that lay the foundation for further targeted studies.
Quality Traits, Volatile Organic Compounds, and Expression of Key Flavor Genes in Strawberry Genotypes over Harvest Period

Abstract:Six strawberry genotypes were examined for fruit yield and size, important chemical traits (sugars, phenolics, anthocyanins, ascorbic acid, volatiles) and antioxidant properties (ferric reducing power). In addition, we determined the expression of genes and transcription factors (SAAT, FaNES1, FaFAD1, FaEGS2, FaEOBII and FaMYB10) controlling the main flavor and aroma traits, and finally evaluated the effect of the genotype and harvest time on the examined chemical and genetic factors, as well as their intercorrelations. The commercial varieties ‘Fortuna’, ‘Victory’, ‘Calderon’, ‘Rociera’, and two advanced selections Ber22/6 and Ber23/3 were cultivated under the same conditions at Berryplasma World Ltd. plantations (Varda, Ilia, Region of Western Greece). Strawberries were harvested at three different time points over the main harvest period in Greece, i.e., early March (T1), late March (T2) and late April (T3). ‘Fortuna’ exhibited the highest early and total yield, while ‘Calderon’, the highest average berry weight. General Linear Model repeated measures ANOVA demonstrated that the interaction of the genotype and harvest time was significant (p < 0.001) on all tested quality attributes and gene expression levels, showing that each genotype behaves differently throughout the harvest period. Exceptions were observed for: (a) the volatile anhydrides, fatty acids, aromatics and phenylpropanoids (all were greatly affected by the harvest time), and (b) lactones, furaneol and FaEGS2 that were affected only by the genotype. We observed significant intercorrelations among those factors, e.g., the positive correlation of FaFAD1 expression with decalactone and nerolidol, of SAAT with furaneol, trans-cinnamic acid and phenylpropanoids, and of FaEGS2 with decalactone and FaFAD1. Moreover, a strong positive correlation between SAAT and FaMYB10 and a moderate negative one between SAAT and glucose were also detected. Those correlations can be further investigated to reveal potential markers for strawberry breeding. Overall, our study contributes to a better understanding of strawberry physiology, which would facilitate breeding efforts for the development of new strawberry varieties with superior qualitative traits.

Variations in Quality Characteristics of Two Strawberry Cultivars during Fruit Ripening

Abstract:In the present study,the variations in fruit quality characteristics related to the flavor and physiological quality in two strawberry cultivars,‘Mira’ and ‘Honeoye’,were investigated during three ripening stages.Results showed that total soluble solids (TSS),volatile aromatic components and anthocyanins accumulation significantly increased,while total acid (TA),total phenolics and total flavonoid contents as well as antioxidant capacity declined during strawberry ripening.The correlations between the quality traits were analyzed by principal component analysis.It was indicated that there were high correlations (r =0.859 1-0.994 7) between the changes in total phenolics,flavonoids,total soluble solid or titratable acid and antioxidant capacity (FRAP) as well as oxygen radical absorbance capacity (ORAC) during strawberry ripening,while the correlation coefficients between total anthocyanin and ferric reducing antioxidant power (FRAP) as well as ORAC were only 0.315 0 and 0.385 3,respectively.Furthermore,a positive correlation between the maturity of strawberry fruits and the accumulation of aromatic volatile components and anthocyanins was also observed.

Abstract:Aroma is an important indicator of fruit flavor, but mechanisms of aroma formation in strawberries (Fragaria spp.) during natural ripening are still not clear. In this study, aroma compounds in strawberry cultivars were analyzed using gas chromatography-mass spectrometry (GC-MS). Richly creamy strawberry cultivars in particular expressed high levels of vanillin acetate and coumarin (up-regulated by 12.6- and 9.8-fold, respectively), while the aroma-free cultivars were dominated by differential changes in terpenes and alcohols. Further research using liquid chromatography-mass spectrometry (LC-MS) and RNA-Seq indicated that the activation of the phenylpropanoid biosynthesis and alpha-linolenic acid metabolic pathways constituted the key to formation of aroma compounds in creamy strawberry cultivars. The results of this study not only provide a well-defined database to detect aroma compounds in different strawberry cultivars but also explore the underlying mechanisms of creamy aroma formation in strawberries.

Abstract:Introduction: The strawberry industry in South Korea has witnessed a significant 65% growth over the past decade, surpassing other fruits and vegetables in production value. While sweetness and acidity are well-recognized flavor determinants, the role of volatile organic compounds (VOCs) in defining the desirable flavor profiles of strawberries is also crucial. However, existing research has predominantly concentrated on a limited range of commercial cultivars, neglecting the broader spectrum of strawberry varieties. Methods: This study embarked on developing a comprehensive VOC database for a diverse array of strawberry cultivars sourced both domestically and internationally. A total of 61 different strawberry cultivars from Korea (45), the USA (7), Japan (8), and France (1) were analyzed for their VOC content using Tenax TA Thermo Desorption tubes and Gas Chromatography-Mass Spectrometry (GC-MS). In addition to VOC profiling, heritability was assessed using one-way ANOVA to compare means among multiple groups, providing insights into the genetic basis of flavor differences. Results and discussion: The analysis identified 122 compounds categorized into esters, alcohols, terpenes, and lactones, with esters constituting the majority (46.5%) of total VOCs in Korean cultivars. 'Arihyang', 'Sunnyberry', and 'Kingsberry' exhibited the highest diversity of VOCs detected (97 types), whereas 'Seolhong' showed the highest overall concentration (57.5mg·kg -1 FW). Compared to the USA cultivars, which were abundant in γ-decalactone (a peach-like fruity aroma), most domestic cultivars lacked this compound. Notably, 'Misohyang' displayed a high γ-decalactone content, highlighting its potential as breeding germplasm to improve flavor in Korean strawberries. The findings underscore the importance of a comprehensive VOC analysis across different strawberry cultivars to understand flavor composition. The significant variation in VOC content among the cultivars examined opens avenues for targeted breeding strategies. By leveraging the distinct VOC profiles, particularly the presence of γ-decalactone, breeders can develop new strawberry varieties with enhanced flavor profiles, catering to consumer preferences for both domestic and international markets.

Integrated metabolomic and transcriptomic analysis reveals factors underlying differences in fruit quality between Fragaria nilgerrensis and Fragaria pentaphylla

Abstract:Background: Strawberries have become one of the most popular fruits because of their unique flavor and high nutritional value. Fruit quality and price are the most important criteria that determine consumer acceptability. Fragaria nilgerrensis and Fragaria pentaphylla are two wild Asian diploid strawberry species that differ in fruit color, taste, and aroma. To understand the molecular mechanisms involved in the formation of high-quality strawberry fruit, we integrated transcriptomics and metabolomics research methods to compare the metabolic and biosynthetic mechanisms of the two Fragaria species. Results: F. nilgerrensis fruit has higher amino acid and lipid contents and a higher sugar-to-acid ratio than F. pentaphylla fruit does, underlying their superior nutritional value, aroma, firmness, and taste. Compared with F. nilgerrensis fruit, F. pentaphylla fruit contained more flavonoids, indicating its enhanced color and health benefits. In addition, candidate structural genes that regulate the biosynthesis of flavonoids, amino acids, and glycerophospholipids in the two strawberry fruit were screened. Conclusions: The differences in aroma, firmness, and taste between F. nilgerrensis fruit and F. pentaphylla fruit are probably due to differences in their amino acid and lipid contents, as well as the difference in their sugar-to-acid ratios. Eight key structural genes that may play important roles in the biosynthesis of amino acids, lipids, and flavonoids were identified. © 2021 Society of Chemical Industry.
Decoding strawberry volatile cultivar diversity through comparative transcriptome analysis

Abstract:This study presents a comparative transcriptomic analysis of three commercial strawberry cultivars: ‘Rociera’, ‘Calderon’, and ‘Victory’, aimed at uncovering the molecular basis of their distinct flavor and aroma profiles. Through RNA sequencing, we analyzed the transcriptomic landscape of these varieties, uncovering a notable array of differentially expressed genes (DEGs) between them. Specifically, ‘Rociera’ showed a higher count of DEGs compared to ‘Victory’, suggesting significant differences in gene expression related to flavor and aroma between those cultivars. Our further investigations revealed pivotal metabolic pathways— such as those involving furanones, amino acids, fatty acids, and terpenoids—participating in generating volatile organic compounds in strawberries. These pathways exhibited variety-specific expression patterns, underlining the genetic determinants of each variety’s unique sensory characteristics. Gene Ontology (GO) enrichment analysis underscored important pathways like phototropism, sugar-mediated signaling, and terpenoid biosynthesis, highlighting the genetic intricacy that influences strawberry flavor and aroma. Additionally, our study identified 31,677 single nucleotide polymorphisms (SNPs) with a significant impact on genes associated with volatile compound biosynthesis. Notably, SNPs linked to key enzymes such as lipoxygenase 6, omega-6 fatty acid desaturase (FaFAD1), and phenylalanine ammonia-lyase 1 (PAL) were discovered, shedding light on the genetic variations that underlie flavor differences. Therefore, this research advances our comprehension of the genetic elements that impact strawberry fruit flavor and aroma, offering invaluable insights for molecular breeding endeavors focused on enhancing strawberry flavor.

Exploration of a European-centered strawberry diversity panel provides markers and candidate genes for the control of fruit quality traits

Abstract:Fruit quality traits are major breeding targets in cultivated strawberry ( ). Taking into account the requirements of both growers and consumers when selecting high quality cultivars is a real challenge. Here, we used an original diversity panel enriched with unique European accessions and the 50K FanaSNP array to highlight the evolution of strawberry diversity over the past 160 years, investigate the molecular basis of 12 major fruit quality traits by GWAS, and provide genetic markers for breeding. Results show that considerable improvements of key breeding targets including fruit weight, firmness, composition and appearance occurred simultaneously in European and American populations. Despite the high genetic diversity of our panel, we observed a drop in nucleotide diversity in certain chromosome regions, revealing the impact of selection. GWAS identified 71 associations with 11 quality traits and, while validating known associations (firmness, sugar), highlighted the predominance of new QTL, demonstrating the value of using untapped genetic resources. Three of the six selective sweeps detected are related to glossiness or skin resistance, two little-studied traits important for fruit attractiveness and, potentially, postharvest shelf-life. Moreover, major QTL for firmness, glossiness, skin resistance and susceptibility to bruising are found within a low diversity region of chromosome 3D. Stringent search for candidate genes underlying QTL uncovered strong candidates for fruit color, firmness, sugar and acid composition, glossiness and skin resistance. Overall, our study provides a potential avenue for extending shelf-life without compromising flavor and color as well as the genetic markers needed to achieve this goal.

Abstract:Background: Strawberries are bright in color, sweet and sour in taste, and rich in nutrients and flavonoid compounds such as anthocyanins and proanthocyanidins. The synthesis and accumulation of anthocyanins are the decisive factors that make strawberries appear bright red. From the perspective of plant breeding, a change in flesh color is an important goal. Methods: In this study, two strawberry plants with different flesh colors were selected, and transcriptome and metabolome analyses were performed during the color change period (S1) and ripening period (S2). Results: RNA-seq revealed a total of 13,341 differentially expressed genes (DEGs) between and within materials, which were clustered into 5 clusters. A total of 695 metabolites were detected via metabolome analysis, and 243 differentially regulated metabolites (DRMs) were identified. The anthocyanin biosynthesis, starch and sucrose metabolism and glycolysis/gluconeogenesis pathways were determined to be important regulatory pathways for changes in strawberry flesh color through a joint analysis of RNA-seq data and the metabolome. The leucoanthocyanidin reductase (LAR) and chalcone synthase (CHS) gene is a key gene related to anthocyanins, cinnamic acid, and phenylalanine. In addition, through joint RNA-seq and metabolome analyses combined with weighted gene co-expression network analysis (WGCNA), we identified 9 candidate genes related to strawberry flesh color. Conclusions: Our research findings have laid the groundwork for a more comprehensive understanding of the molecular mechanisms governing the color transformation in strawberry flesh. Additionally, we have identified novel genetic resources that can be instrumental in advancing research related to strawberry color change.

Identification of key aromas of grapefruit juice and study of their contributions to the enhancement of sweetness perception

Abstract:Odor-induced sweetness enhancement has become a method that does not change consumer acceptability and can effectively reduce the sugar content in food products. In this study, the volatile compounds in "Red Blush" grapefruit juice were investigated by headspace solid-phase microextraction, solvent-assisted flavor evaporation, and gas chromatography–mass spectrometry (GC–MS) combined with gas chromatography/olfactometry-associated taste (GC/O-AT). A total of 59 volatiles were identified by GC–MS, and 32 volatiles were identified as key aroma compounds with an odor activity value ≥ 1 or aroma intensity ≥ 3. GC-O/AT analysis screened 10 odorants associated with sweetness. Furthermore, we examined whether odor could enhance the sweetness in sucrose solutions or sucrose–citric acid mixed solutions through sensory experiments. The results showed that eight odorants could significantly achieve the sweetness enhancement effect at 50 g/L sucrose solution. Ethyl propanoate, ethyl butyrate, ethyl 2-methyl butyrate, ethyl hexanoate, and terpinolene could also enhance the sweet taste in the sucrose–citric acid mixed solution. This study provides more theoretical support for aroma–taste interactions in the citrus-flavored system and compensates for sugar reduction in the food industry.

Abstract:Aroma significantly contributes to flavor, which directly affects the commercial quality of strawberries. The strawberry aroma is complex as many kinds of volatile compounds are found in strawberries. In this review, we describe the current knowledge of the constituents and of the biosynthesis of strawberry volatile compounds, and the effect of postharvest treatments on aroma profiles. The characteristic strawberry volatile compounds consist of furanones, such as 2,5-dimethyl-4-hydroxy-3(2H)-furanone and 4-methoxy-2,5-dimethyl-3(2H)-furanone; esters, including ethyl butanoate, ethyl hexanoate, methyl butanoate, and methyl hexanoate; sulfur compounds such as methanethiol, and terpenoids including linalool and nerolidol. As for postharvest treatment, the present review discusses the overview of aroma volatiles in response to temperature, atmosphere, and exogenous hormones, as well as other treatments including ozone, edible coating, and ultraviolet radiation. The future prospects for strawberry volatile biosynthesis and metabolism are also presented. © 2018 Society of Chemical Industry.

Evaluation of Bioactive Compounds in Strawberry Fruits by a Targeted Metabolomic Approach

Abstract:Strawberry (Fragariaxananassa Duch.) is referred to as a functional fruit because of its high nutritional value. Here, we used a targeted metabolomics approach to investigate the quantitative variation in strawberry bioactive compound concentration across cultivars and breeding lines and during fruit ripening. We also compared the efficiency of 95% ethyl alcohol (EtOH), 50% EtOH, and water in extracting these compounds. High-performance liquid chromatography (HPLC) and multivariate analysis revealed that 50% EtOH extracted bioactive compounds more efficiently than 95% EtOH and water. Screening of targeted bioactive compounds extracted with 50% EtOH in six cultivars and 28 breeding lines revealed high abundance of anthocyanins, flavonols, flavanols, and cinnamic acid, which constituted up to 6.4 mg.g(-1) of fresh weight of fruits. The highest concentration of phenolic compounds was observed in breeding lines 'SB08', 'SB11', 'Arang', 'SB09', and 'SB10'. Furthermore, significant changes in the concentration of these compounds occurred during fruit ripening in four strawberry cultivars. Overall, our results highlight the chemotypic relationship between cultivars, breeding lines, and fruit ripening associated-changes in strawberries, and these findings provide a chemometric method for choosing optimal cultivars/breeding lines.
