Abstract:In this study, a calibration model based on Near-infrared spectroscopy (NIR) technique and chemometrics method was developed for rapid and non-destructive detecting the polysaccharide contents of lentinula edodes samples collected from different regions. The polysaccharide contents of these samples were firstly determined by standard phenol-sulphruic acid method. Then, NIR spectra of these samples were collected by using Fourier transform infrared spectrometry. Based on these experimental data, a random forest method was further used to distinguish the regions of these samples, with a classification accuracy of 96.6%. After that, a rapid, accurate, and quantitative model was established for predicting the polysaccharide contents of these samples. In the model establishing process, some signal pre-treatment methods were optimized, and the validation results with highest determination coefficient (R2) and low root mean square errors of prediction (RMSEP) were, 0.925 and 0.720, respectively. These results showed that combined NIR technique with chemometrics was an effective and green method for lentinula edodes quality control.
A Study on the Use of Near-Infrared Spectroscopy for the Rapid Quantification of Major Compounds in Tanreqing Injection

Abstract:In this paper we describe the strategy used in the development and validation of a near infrared spectroscopy method for the rapid determination of baicalin, chlorogenic acid, ursodeoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), and the total solid contents (TSCs) in the Tanreqing injection. To increase the representativeness of calibration sample set, a concentrating-diluting method was adopted to artificially prepare samples. Partial least square regression (PLSR) was used to establish calibration models, with which the five quality indicators can be determined with satisfied accuracy and repeatability. In addition, the slope/bias (S/B) method was used for the models transfer between two different types of NIR instruments from the same manufacturer, which is contributing to enlarge the application range of the established models. With the presented method, a great deal of time, effort and money can be saved when large amounts of Tanreqing injection samples need to be analyzed in a relatively short period of time, which is of great significance to the traditional Chinese medicine (TCM) industries.

Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.

Abstract:A rapid and simultaneous quantification of the main active compounds of the in-process extract solutions is beneficial to the process monitoring as well as ensuring quality consistency of the end products during the production of traditional Chinese medicine. Here we present a near infrared (NIR) spectroscopy-based method for rapid analysis of extract solutions in the production of compound E Jiao oral liquid. The partial least-squares regression (PLSR) models for four quality indicators (viz. total flavonoids, total saponins, total saccharides and soluble solid contents) were established and validated. The results showed that all of the four models exhibited satisfactory fitting and predictive capacity. The root mean squares error of prediction (RMSEP) was 0.0384 mg mL−1, 0.0154 mg mL−1, 3.80 mg mL−1 and 0.199% for total flavonoids, total saponins, total saccharides and soluble solid contents, respectively. This work here demonstrated that NIR spectroscopy coupled with PLSR calibration can offer a reliable and non-destructive alternative in the routine monitoring of the extraction process in the production of compound E Jiao oral liquid. The presented approach is expected to be equally applicable to the mixed decoction of other herbal medicines.

Abstract:A non-invasive analysis method for bottled Tanreqing injection was developed based on near infrared (NIR) spectroscopy. The NIR trans-mission spectra were collected using a home-made accessory. The quality indicators of Tanreqing injection, which include baicalin, ursodesoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), chlorogenic acid (CA), forsythin, total amino acids (TAA) and soluble solid content (SSC), were determined with standard methods. Several outlier samples were eliminated using the Dixon test, leverage and studentised residual test and analogous samples were amalgamated. Partial least squares regression (PLSR) was used for establishing the calibration models with the pre-treated spectra and the reference values. The root mean square errors of prediction ( RMSEP) values of external validation samples were 0.098 g L−1, 0.059 g L−1, 0.140 g L−1, 0.038g L−1, 0.007 g L−1, 0.053 g L−1 and 0.538 gL−1 for baicalin, CA, UDCA, CDCA, forsythin, TAA and SSC, respectively, which indicated that the established models possessed satisfactory predictive abilities. The presented method can reduce the workload of product inspection to a large extent. This will allow the sampling proportion to be increased and enable improved quality control during Tanreqing injection production.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Abstract:Objective:To develop a method for the rapid determination of tannins during the alcoholic-precipitation process of Danshen injection using near infrared spectroscopy.Method:The phosphomolybdium tungstic acid-casein method was used as reference method.Different spectra pretreatment methods were compared.The calibration model based on partial least square algorithm was developed to correlate the spectra with the corresponding values determined by the reference method.Then the model was validated by the samples of validation set.Result:The result showed that the best model was obtained after the NIR spectra were pretreated with the first derivative and Savitzky-Golay smoothing,and the wavelength range 5430-5839 cm-1 was used for calibration.The correlation coefficient of the developed NIR model was 0.964 and the RMSEP was 1.43 g·L-1.Conclusion:Owing to its fast and nondestructive properties,the presented method can be developed as a process analysis method for the alcohol-precipitation process of Chinese medicine.
An on-Line Near-Infrared Spectroscopy Approach for Content Determination in the Percolation Process of Compound Kushen Injection

Abstract:OBJECTIVE To develop an on-line analysis method based on near infrared spectroscopy(NIRS) for monitoring the percolation process of compound Kushen injection traditional Chinese medicines.METHODS Samples and correlative spectra were collected with online equipment in percolation process.The contents of total alkaloids and total solids measured with standard methods were used as reference values.Multivariate calibration models based on partial least squares(PLS) algorithm were developed to correlate the spectra and the reference values.RESULTS The correlation coefficients of the calibration models for total alkaloids and total solids were 0.997 9 and 0.996 0 with root mean square errors(RMSECs) of 0.072 1 g·L-1 and 0.081 1%,respectively.External validation samples collected on-line were used to evaluate the quantitative models.The correlation coefficients between the predicted value and the reference value were 0.991 5 and 0.999 0 with root mean square errors of prediction(RMSEPs) of 0.195 0 g·L-1 and 0.068 1%,respectively.CONCLUSION The presented method is rapid,accurate and reliable,which can be used for monitoring the percolation process of compound Kushen injection.
Development of Simple Identification Models for Four Main Catechins and Caffeine in Fresh Green Tea Leaf Based on Visible and Near-Infrared Spectroscopy.

Abstract:Catechin polyphenols and caffeine play an important role in tea quality. This study analyzed the visible and near-infrared (Vis-NIR) spectral variation and concentration differences of catechin and caffeine of fresh tea leaves (Camellia sinensis L.) in three varieties and six leaf positions, and a universal spectral model for quickly and accurately measurement of the catechin and caffeine content in the various varieties and leaf positions was established. It was found that all the catechins and caffeine were significantly influenced by variety and leaf position. While, the Vis/NIR spectrum as an indication of internal biochemical substances was successfully used to distinguish different varieties and leaf positions with the discrimination accuracy rates of 98.15% and 100%, respectively. The quantitative relationship between the chemical components and the data obtained by Vis-NIR spectroscopy was established based on multivariate regression analysis such as partial least squares (PLS) and multiple linear regression (MLR). Furthermore, competitive adaptive reweighted sampling (CARS) and successive projections algorithm (SPA) were used to select the characteristic wavelengths for the development of simple models. Results showed that the quantitative determination models obtained good performance with the determination coefficients (R2) of 0.949, 0.893, 0.968, 0.931 and 0.917 for epigallocatechin gallate (EGCG), epicatechin gallate (ECG), epigallocatechin (EGC), epicatechin (EC) and caffeine (CAF), respectively. Such high detection accuracy shows that the spectral detection model has a strong applicability for both varieties and leaf positions. The overall results of the study revealed the potential use of Vis-NIR spectroscopy as a rapid, simple and non-destructive method for the determination of four main catechins and caffeine in fresh tea leaves, and it will play a great role in the real-time detection of tea physiological information in tea garden.
Establishment of Predictive Model for Quantitative Analysis of Major Components in Jasmine Tea by Near Infrared Spectroscopy(NIRS)

Abstract:Taking 112 representative jasmine tea as research material, the quantitative calibration models for determining the contents of 13 chemical compositions such as total tea polyphenols (TP), free amino acids (AA), caffeine (CAFF), water extract, GA, GC, EGC, C, EC, EGCG, GCG, ECG, CG were established by NIRS combined with chemometrics. Q-value taking all important statistical values into account can evaluate the quality of models. Results showed that almost all the Q-value of models was 0.8~0.9, except for CG with a Q-value of 0.7702. Each of the 13 predictive models was applied for external inspection. Coefficients of correlation (R) between predicted value by NIR and the actual value were exceed 0.9, except CAFF and GCG was 0.7~0.8 and GC、EGCG、ECG、CG was 0.8~0.9. The established models present high stability and predictive accuracy. This paper provided a simple and convenient method for measuring the contents of major quality components in jasmine tea.
Quality Evaluation of Ephedrae Herba by Near Infrared Spectroscopy

Abstract:A novel method was developed for the quality control of Ephedrae herba by near-infrared (NIR) spectroscopy. First, qualitative models established by discriminant analysis and support vector machine were used for the preliminary screening of unqualified samples of E. herba. Then quantitative models of ephedrine and the total alkali (ephedrine and pseudoephedrine) were established by partial least squares regression and particle swarm optimization based least square support vector machine. The contents of test samples were predicted by the established NIR quantitative models. As a result, the accuracies of unqualified identification were 98.9% by discriminant analysis and 100% by support vector machine. The performance of the particle swarm optimization based least square support vector machine models were better than the partial least squares regression models. The correlation coefficients were both more than 0.98 and relative standard errors of calibrations were less than 9% in the calibration sets of particle swarm optimization based least square support vector machine models. As for the test sets, the correlation coefficients were both more than 0.93 and the relative standard errors of prediction were less than 13%, indicating satisfactory predicted results. All of these results demonstrated that NIR spectroscopy may be a powerful tool for the quality control of E. herba.
[On-Line Monitoring of Extraction Process of Danhong Injection Based on Near-Infrared Spectroscopy].

Abstract:OBJECTIVE:To establish a rapid quantitative analysis method for the quality control of Danhong injection extraction using near-infrared (NIR) spectroscopy.METHOD:Online collecting the NIR spectra during the mixed extraction process of Salvia miltiorrhiza and Carthamus tinctorius, partial least squares regression (PLSR) models were developed for the quality indicators rosmarinic acid (RA), salvia acid B (SaB), lithospermic acid (LA), hydroxysafflor yellow A (HSYA) and solid content (SSC), with HPLC and weight-loss method as reference methods.RESULT:The correlation coefficients of the cross validation for RA, SaB, LA, HSYA and SSC were 0.909 3, 0.915 2, 0.901 9, 0.747 7 and 0.931 4, respectively. And the root mean square errors of cross validation (RMSECV) were 0.012 1, 0.251, 0.017 7, 0.038 1 g x L(-1) and 0.359%, respectively.CONCLUSION:In this study, NIR spectroscopy was successfully applied to achieve the real-time determination of the contents of RA, SaB, LA and SSC, while the performance of the HSYA calibration model needed to be improved.
On-line monitoring the contents of active components in the solution for Fufangdanshen dripping pills by near-infrared spectroscopy

Abstract:Objective:To establish a rapid analysis method of determining the contents of Danshensu and protocatechuic aldehyde in the dripping solution of Fufangdanshen dripping pills,and on-line monitoring the homogeneity of the blending process.Method:Near-infrared(NIR) transflectance spectroscopy was first used to collect NIR spectra of the dripping solution.With HPLC analysis values as reference,the partial least square regression(PLSR) calibration models were built after wavelength selection and multiplicative scatter correction(MSC) + 1st derivative pretreatment,which were applied to on-line determine the contents of ingredients in the dripping solution of Fufangdanshen dripping pills,and on-line monitor the homogeneity and end-point of the blending process.Result:The PLSR calibration models of NIR spectral have high correlation coefficients(R) and low root mean square error of prediction(RMSEP).Danshensu:r=0.9929,RMSEP=0.368;Protocatechuic aldehyde:r=0.9937,RMSEP=0.159.The proposed method was successfully used in on-line monitoring the blending process of dripping solution.Conclusion:NIR transflectance spectroscopy is accurate,robust and can be used routinely to determine the contents of ingredients and the uniformity of distribution in the solution of dripping pills.
Fast Analytical Method on Content and Transfer Rate of Marker Component in Ethanol Precipitation of Cinobufacini by Near Infrared Spectroscopy

Abstract:The method of near infrared spectroscopy was established to analyze the content and transfer rate of the marker component, indole alkaloids, of cinobufacini in the process of ethanol precipitation. The samples of cinobufacini were collected, and the concentration of indole alkaloids was determined by the reference method, UV-vis spectroscopy. After the rejection of abnormal samples, the pretreatment of spectra and the choice of the corresponding wave band, the multivariate calibration models were established using PLS algorithm between spectra and the UV values. The samples in ethanol precipitation, which did not participate in modeling, were used to examine the performance of the models. The correlation coefficient of the calibration model was 0.9613. Root mean square error of calibration (RMSEC) and root mean square error of prediction (RMSEP) were 0.172 and 0.169, respectively. Furthermore, residual predictive deviation (RPD) of the model was 4.13, which meant quantity analysis was feasible. It is concluded that near infrared spectroscopy can be considered as a fast, effective and non-destructive analysis method applied in the process of ethanol precipitation for traditional Chinese medicine.
Rapid and Quantitative Determination of Multiple Quality Indicators(Indole Alkaloids, Etc.) in Concentration Process of Bufo Bufo Gargarizans Cantor Extract Using Fourier Transform Near Infrared Spectroscopy and Chemometrics

Abstract:OBJECTIVE To investigate the application of near infrared spectroscopy in rapid and quantitative determination of multiple quality indicators in concentration process of Bufo bufo gargarizans Cantor extract.METHODS NIR transmission spectra of concentrated solutions were collected in rectangular quartz cuvette with different optical pathlengths(2 and 5 mm).Calibration models of density,water content and indole alkaloids were developed by partial least squares regression(PLSR).The established models were applied to predict the unknown samples to test the performance of the models.RESULTS The correlation coefficients of the density,water content and indole alka.The residual predictive deviations(RPD) were all above 3,and the relative standard errors of predictions(RSEP) were less than 10%,indicating the satisfactory model performance and predictive ability.Moreover,better calibration results were obtained with an optical path length of 2 mm for density and water content,and 5 mm for indole alkaloids.The results revealed that the optical path length has influence on the NIR analysis results,and the appropriate path length for the NIR analysis should be determined by comparison analysis.CONCLUSION NIR spectroscopy technique provides a novel efficient and environmental friendly approach for the fast simultaneous determination of three key quality indicators(density,water content and indole alkaloids) in the concentration process of Bufo bufo gargarizans Cantor extract.
Rapid Quantitative Determination of Multi-Indicators in Lonicerae Japonicae Flos by Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a set of quantitative calibration models based on near-infrared (NIR) spectroscopy and chemometrics methods for rapid determination of chlorogenic acid and luteoloside content in Lonicerae Japonicae Flos at the same time, which is useful for the quality control process. METHODS The PLSR algorithm was finally used to build NIR quantitative models between the spectra and the content of chlorogenic acid and luteoloside determined by the reference methods. The validation set was used to identify the predictive performance of the established models. RESULTS For PLSR models, the correlation coefficients of calibration and cross-validation were both >0.90, and the root mean square errors of cross validation (RMSECV) of two indicators were 0.469 and 0.012 4, respectively. External validation proved that the root mean square errors of prediction (RMSEP) were 0.478 and 0.010 5, which were close to RMSEC values, respectively. The ratio of RMSECV to RMSEP was close to 1, and the relative standard errors (RSEP) for chlorogenic acid and luteoloside content were 10.19% and 14.76%. This established method was initiatively proved to be accurate and stable. CONCLUSION NIR can be used for the rapid determination of chlorogenic acid and luteoloside content in Lonicerae Japonicae Flos at the same time, which is beneficial and crucial for the quality control of this raw material in TCM pharmaceutical factory.
Nondestructive Qualitative and Quantitative Analysis of Yaobitong Capsule Using Near-Infrared Spectroscopy in Tandem with Chemometrics.

Abstract:The purpose of the study is to present a nondestructive qualitative and quantitative approach of hard-shell capsule using near-infrared (NIR) spectroscopy combined with chemometrics. The Yaobitong capsule (YBTC) was used for demonstration of the proposed approach and the NIR spectra were collected using a handheld fiber probe (FP) without the damage of capsule shell. By comparing the differences and similarities of the NIR spectra of capsule shells, contents and intact capsules, a preliminary conclusion can be drawn that the NIR spectra contained the information of the contents. Characteristic variables were selected by competitive adaptive weighted resampling (CARS) method, and least squares support vector machine (LSSVM) method based on particle swarm optimization (PSO) algorithm was applied to the construction of quantitative models. The relative standard error of prediction (RSEP) values of five saponins including notoginsenoside R1, ginsenoside Rg1, Re, Rb1, and Rd were 3.240%, 5.468%, 5.303%, 5.043%, and 3.745%, respectively. In addition, for qualitative model, three different types of adulterated capsules were designed. The model established by data driven version of soft independent modeling of class analogy (DD-SIMCA) demonstrated a satisfactory result that all adulterated capsules were identified accurately after an appropriate number of principal components (PCs) were chosen. The results indicated that although the NIR spectra collection was affected by capsule shell, sufficient content information can be obtained for quantitative and qualitative analysis after combining with chemometrics. It further proved that acquired NIR spectra do contain the effective component information of the capsule. This study provided a reference for the rapid nondestructive quality analysis of traditional Chinese medicine (TCM) capsule without damaging capsule shell.
Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. The results indicate that NIRs in combination with chemometrics has great potential in improving the efficiency in production. (c) 2020 Elsevier B.V. All rights reserved.

Abstract:OBJECTIVES To investigate the feasibility of using near-infrared spectroscopy for rapid determination of main organic acids in Ginkgo biloba leaf extract (EGBL). METHODS Main organic acids in EGBL were assayed using the HPLC method. Critical factors of the chromatographic separation were optimized by a novel analytical quality by design approach. Partial least squares-discriminant analysis (PLS-DA) was performed to screen the marker components, and principal component analysis (PCA) was utilized to distinguish the different samples. Then, spectral quantification potential was investigated using PLS and support vector machine (SVM) approaches. For modelling, different spectral preprocessing and wavelength selection methods were systematically compared. KEY FINDINGS It was found that quinic acid, protocatechuic acid and 6-hydroxykynurenic acid were identified as possible index components. PLS-DA based on contents and PCA based on near-infrared spectra can both effectively distinguish the different EGBL samples. The calibration models with wonderful prediction performance can be both developed by the PLS and SVM algorithms. CONCLUSIONS NIR spectroscopy combined with chemometrics can realize the rapid and non-destructive qualitative and quantitative analysis of EGBL. The proposed method may be applied to quality control of EGBL and other natural products in commercial use.
Near-infrared Spectroscopy On-Line and Real-Time Monitoring of Extraction Process of Xuebijing Injection

Abstract:Objective:To establish a rapid quantitative analysis model for quality control of Xuebijing Injection extraction with application of near-infrared(NIR) spectroscopy,and quickly determine the end point of extraction process,so as to realize on-line monitoring of the extraction process of Xuebijing Injection.Methods: Taking mixed extraction process of Ligusticum chuanxiong Hort.,Salvia miltiorrhiza Bge.and Angelica sinensis(Oliv.) Diels for an example,partial least squares regression(PLSR) models were developed for the parameters of interest: total solid content and ferulic acid.The established PLS calibration models were used for on-line and real-time monitoring of the extraction process,and the moving block of standard deviation(MBSD) was used to identify the extraction end point.Furthermore,a model updating method was adopted to improve the stability and prediction accuracy of the calibration models.Results: The correlation coefficients of total solid content and ferulic acid models have reached 0.986.When the established models were applied to on-line monitoring,the correlation coefficients of prediction(r) and the residual predictive deviations(RPD) were above 0.967 and 3,respectively,and the relative standard errors of predictions(RSEP) were less than 10%.The result of MBSD was generally coincided with the variation of actual-measured ferulic acid concentration.In addition,the predictive accuracy was significantly improved after model updating with new samples.Conclusions: In this study,NIR spectroscopy was successfully applied to predicting total solid content and ferulic acid concentration in real-time and rapid determination of the extraction end point.Moreover,to solve the problem of calibration invalidation,the method of model updating was utilized to extend the use of established models and obtain better prediction results.
