Abstract:Microfluidics is used to manipulate fluid flow in micro-channels to fabricate drug delivery vesicles in a uniform tunable size. Thanks to their designs, microfluidic technology provides an alternative and versatile platform over traditional formulation methods of nanoparticles. Understanding the factors that affect the formulation of nanoparticles can guide the proper selection of microfluidic design and the operating parameters aiming at producing nanoparticles with reproducible properties. This review introduces the microfluidic systems' continuous flow (single-phase) and segmented flow (multiphase) and their different mixing parameters and mechanisms. Furthermore, microfluidic approaches for efficient production of nanoparticles as surface modification, anti-fouling, and post-microfluidic treatment are summarized. The review sheds light on the used microfluidic systems and operation parameters applied to prepare and fine-tune nanoparticles like lipid, poly(lactic-co-glycolic acid) (PLGA)-based nanoparticles as well as cross-linked nanoparticles. The approaches for scale-up production using microfluidics for clinical or industrial use are also highlighted. Furthermore, the use of microfluidics in preparing novel micro/nanofluidic drug delivery systems is presented. In conclusion, the characteristic vital features of microfluidics offer the ability to develop precise and efficient drug delivery nanoparticles.

Biogeosciences Estimating mixed layer nitrate in the North Atlantic Ocean

Abstract:Here we present an equation for the estimation of nitrate in surface waters of the North Atlantic Ocean (40 ◦ N to 52 N, 10 W to 60 W). The equation was derived by multiple linear regression (MLR) from nitrate, sea surface temperature (SST) observational data and model mixed layer depth (MLD) data. The observational data were taken from merchant vessels that have crossed the North Atlantic on a regular basis in 2002/2003 and from 2005 to the present. It is important to find a robust and realistic estimate of MLD because the deepening of the mixed layer is crucial for nitrate supply to the surface. We compared model data from two models (FOAM and Mercator) with MLD derived from float data (using various criteria). The Mercator model gives a MLD estimate that is close to the MLD derived from floats. MLR was established using SST, MLD from Mercator, time and latitude as predictors. Additionally a neural network was trained with the same dataset and the results were validated against both model data as a “ground truth” and an independent observational dataset. This validation produced RMS errors of the same order for MLR and the neural network approach. We conclude that it is possible to estimate nitrate concentrations with an uncertainty of ±1.4 μmol L−1 in the North Atlantic.

Abstract:Droplet microfluidics offers exquisite control over the flows of multiple fluids in microscale, enabling fabrication of advanced microparticles with precisely tunable structures and compositions in a high throughput manner. The combination of these remarkable features with proper materials and fabrication methods has enabled high efficiency, direct encapsulation of actives in microparticles whose features and functionalities can be well controlled. These microparticles have great potential in a wide range of bio-related applications including drug delivery, cell-laden matrices, biosensors and even as artificial cells. In this review, we briefly summarize the materials, fabrication methods, and microparticle structures produced with droplet microfluidics. We also provide a comprehensive overview of their recent uses in biomedical applications. Finally, we discuss the existing challenges and perspectives to promote the future development of these engineered microparticles.

Abstract:Microfluidic technology has been highly useful in nanovolume sample preparation, separation, synthesis, purification, detection and assay, which are advantageous in drug development. This review highlights the recent developments and trends in microfluidic applications in two areas of drug development. First, we focus on how microfluidics has been developed as a facile tool for the fabrication of drug carriers including microparticles and nanoparticles. Second, we discuss how microfluidic chips could be used as an independent platform or integrated with other technologies in drug toxicity screening. Challenges and future perspectives of microfluidic applications in drug development have also been provided considering the present technological limitations.

Abstract:This review explores significant advancements in polymer science and fabrication processes that have enhanced the performance and broadened the application scope of microfluidic devices. Microfluidics, essential in biotechnology, medicine, and chemical engineering, relies on precise fluid manipulation in micrometer-sized channels. Recent innovations in polymer materials, such as flexible, biocompatible, and structurally robust polymers, have been pivotal in developing advanced microfluidic systems. Techniques like replica molding, microcontact printing, solvent-assisted molding, injection molding, and 3D printing are examined, highlighting their advantages and recent developments. Additionally, the review discusses the diverse applications of polymer-based microfluidic devices in biomedical diagnostics, drug delivery, organ-on-chip models, environmental monitoring, and industrial processes. This paper also addresses future challenges, including enhancing chemical resistance, achieving multifunctionality, ensuring biocompatibility, and scaling up production. By overcoming these challenges, the potential for widespread adoption and impactful use of polymer-based microfluidic technologies can be realized.

Abstract:Conventional drug administration usually faces the problems of degradation and rapid excretion when crossing many biological barriers, leading to only a small amount of drugs arriving at pathological sites. Therapeutic drugs delivered by drug delivery systems to the target sites in a controlled manner greatly enhance drug efficacy, bioavailability, and pharmacokinetics with minimal side effects. Due to the distinct advantages of microfluidic techniques, microfluidic setups provide a powerful tool for controlled synthesis of drug delivery systems, precisely controlled drug release, and real-time observation of drug delivery to the desired location at the desired rate. In this review, we present an overview of recent advances in the preparation of nano drug delivery systems and carrier-free drug delivery microfluidic systems, as well as the construction of in vitro models on-a-chip for drug efficiency evaluation of drug delivery systems. We firstly introduce the synthesis of nano drug delivery systems, including liposomes, polymers, and inorganic compounds, followed by detailed descriptions of the carrier-free drug delivery system, including micro-reservoir and microneedle drug delivery systems. Finally, we discuss in vitro models developed on microfluidic devices for the evaluation of drug delivery systems, such as the blood–brain barrier model, vascular model, small intestine model, and so on. The opportunities and challenges of the applications of microfluidic platforms in drug delivery systems, as well as their clinical applications, are also discussed.

Abstract:The microfluidic method primainly utilizes two incompatible liquids as continuous phase and dispersed phase respectively. It controls the formation of droplets by managing the microchannel structure and the flow rate ratio of the two phases. Droplet-based microfluidics is a rapidly expanding interdisciplinary research field encompassing physics, biochemistry, and Microsystems engineering. Droplet microfluidics offer a diverse and practical toolset that enables chemical and biological experiments to be conducted at high speeds and with greater efficiency compared to traditional instruments. The applications of droplet-based microfluidics are vast, including areas such as drug delivery, owing to its compatibility with numerous chemical and biological reagents and its ability to carry out various operations. This technology has been extensively researched due to its promising features. In this review, we delve into the materials used in droplet generation-based microfluidic devices, manufacturing techniques, methods for droplet generation in channels, and, finally, we summarize the applications of droplet generation-based microfluidics in drug delivery vectors, encompassing nanoparticles, microspheres, microcapsules, and hydrogel particles. We also discuss the challenges and future prospects of this technology across a wide array of applications.

Abstract:Additive manufacturing plays a vital role in the food, mechanical, pharmaceutical, and medical fields. Within these fields, medical additive manufacturing has led to especially obvious improvements in medical instruments, prostheses, implants, and so forth, based on the advantages of cost-effectiveness, customizability, and quick manufacturing. With the features of precise structural control, high throughput, and good component manipulation, microfluidic techniques present distinctive benefits in medical additive manufacturing and have been applied in the areas of drug discovery, tissue engineering, and organs on chips. Thus, a comprehensive review of microfluidic techniques for medical additive manufacturing is useful for scientists with various backgrounds. Herein, we review recent progress in the development of microfluidic techniques for medical additive manufacturing. We evaluate the distinctive benefits associated with microfluidic technologies for medical additive manufacturing with respect to the fabrication of droplet/fiber templates with different structures. Extensive applications of microfluidic techniques for medical additive manufacturing are emphasized, such as cell guidance, three-dimensional (3D) cell culture, tissue assembly, and cell-based therapy. Finally, we present challenges in and future perspectives on the development of microfluidics for medical additive manufacturing.

Abstract:Microfluidic devices have been widely used since 1990s for diverse manipulations of particles (a general term of beads, cells, vesicles, drops, etc.) in a variety of applications. Compared to the active manipulation via an externally imposed force field, the passive manipulation of particles exploits the flow-induced intrinsic lift and/or drag to control particle motion with several advantages. Along this direction, inertial microfluidics has received tremendous interest in the past decade due to its capability to handle a large volume of samples at a high throughput. This inertial lift-based approach in Newtonian fluids, however, becomes ineffective and even fails for small particles and/or at low flow rates. Recent studies have demonstrated the potential of elastic lift in non-Newtonian fluids for manipulating particles with a much smaller size and over a much wider range of flow rates. The aim of this article is to provide an overview of the various passive manipulations, including focusing, separation, washing and stretching, of particles that have thus far been demonstrated in non-Newtonian microfluidics.

Abstract:Ultrasound (US)-responsive microparticles show broad potential in controlled drug delivery systems. Compare with the traditional micron-scale material fabrication methods, capillary microfluidic technology features superior advantages in large-scale production, batch-to-batch similarity, high encapsulation efficiency, low cost, and so on. The excellent maneuverability and customizability of the capillary microfluidic devices allow the production of microparticles with various functionalities and fine-tuned chemical compartments. Moreover, the flexible regulation of the particle size and core-shell ratio can be easily realized by modulating the capillary orifices and flow rates of microfluidic channels. In this review, we introduce the fabrication of US-responsive microparticles with specific core-shell structures via capillary microfluidic methods, from single emulsion to triple emulsions. Then, we address some particular examples, where the drug delivery and US-triggered cargo release capacity of these microfluidic microparticles are demonstrated. Finally, we conclude the advanced achievements of the US-responsive microfluidic microparticles, summarize the obstacles to the development of this interdisciplinary field, and prospect their future applications.

Abstract:The development of nanomedicine or medical nanotechnology, has brought important new ways to the development of medicines and biotechnology products. As a result of groundbreaking discoveries in the use of nanoscale materials significant commercialization initiatives have been launched and are at the forefront of the rapidly expanding field of nanotechnology by using smart particles. Microfluidic technologies use nano-and micro-scale manufacturing technologies to develop controlled and reproducible liquid microenvironments. Lead compounds with controlled physicochemical properties can be obtained using microfluidics, characterized by high productivity, and evaluated by biomimetic methods. Microfluidics, for example, can not only produce nanoparticles in a well-controlled, reproducible, and high-throughput manner, but it can also continuously create three-dimensional environments to mimic physiological and/or pathological processes. Materials with smart properties can be manipulated to respond in a controllable and reversible way, modifying some of their properties as a result of external stimuli such as mechanical stress or a certain temperature. All in all, microfluidic technology offers a potential platform for the rapid synthesis of various novel drug delivery systems. Therefore, these smart particles are equally necessary as the drug in drug delivery.

Abstract:Microfluidics is an emerging technology that can be employed as a powerful tool for designing lipid nano-microsized structures for biological applications. Those lipid structures can be used as carrying vehicles for a wide range of drugs and genetic materials. Microfluidic technology also allows the design of sustainable processes with less financial demand, while it can be scaled up using parallelization to increase production. From this perspective, this article reviews the recent advances in the synthesis of lipid-based nanostructures through microfluidics (liposomes, lipoplexes, lipid nanoparticles, core-shell nanoparticles, and biomimetic nanovesicles). Besides that, this review describes the recent microfluidic approaches to produce lipid micro-sized structures as giant unilamellar vesicles. New strategies are also described for the controlled release of the lipid payloads using microgels and droplet-based microfluidics. To address the importance of microfluidics for lipid-nanoparticle screening, an overview of how microfluidic systems can be used to mimic the cellular environment is also presented. Future trends and perspectives in designing novel nano and micro scales are also discussed herein.

Abstract:Overview of latest advances in droplet microfluidic formation of highly monodispersed microparticles, involving a plethora of types including spherical, nonspherical, and Janus type, as well as core–shell, hole–shell, and controllable multicompartment particles, is provided. The critical analysis of the current status and existing challenges is also conducted, and the future development in the emerging fields by microfluidic emulsification is envisioned. The remarkable control function over the functional material formation process enabled by droplet microfluidic emulsification approaches can lead to the efficient and one‐step encapsulation of active substances in microparticles, with the microparticle characteristics well regulated. In comparison to the conventional fabrication methods, droplet microfluidic technology can not only construct microparticles with various shapes, but also provide excellent templates, which enrich and expand the application fields of microparticles. For instance, intersection with disciplines in pharmacy, life sciences, and others, modifying the structure of microspheres and appending functional materials can be completed in the preparation of microparticles. The as‐prepared polymer particles have great potential in a wide range of applications for chemical analysis, heavy metal adsorption, and detection. This review systematically introduces the devices and basic principles of particle preparation using droplet microfluidic technology and discusses the research of functional microparticle formation with high monodispersity, involving a plethora of types including spherical, nonspherical, and Janus type, as well as core–shell, hole–shell, and controllable multicompartment particles. Moreover, this review paper also exhibits a critical analysis of the current status and existing challenges, and outlook of the future development in the emerging fields has been discussed.

Abstract:In the past two decades, microfluidics-based particle production is widely applied for multiple biological usages. Compared to conventional bulk methods, microfluidic-assisted particle production shows significant advantages, such as narrower particle size distribution, higher reproducibility, improved encapsulation efficiency, and enhanced scaling-up potency. Herein, an overview of the recent progress of the microfluidics technology for nano-, microparticles or droplet fabrication, and their biological applications is provided. For both nano-, microparticles/droplets, the previously established mechanisms behind particle production via microfluidics and some typical examples during the past five years are discussed. The emerging interdisciplinary technologies based on microfluidics that have produced microparticles or droplets for cellular analysis and artificial cells fabrication are summarized. The potential drawbacks and future perspectives are also briefly discussed.
Microfluidic approaches for producing lipid-based nanoparticles for drug delivery applications

Abstract:The importance of drug delivery for disease treatment is supported by a vast literature and increasing ongoing clinical studies. Several categories of nano-based drug delivery systems have been considered in recent years, among which lipid-based nanomedicines, both artificial and cell-derived, remain the most approved. The best artificial systems in terms of biocompatibility and low toxicity are liposomes, as they are composed of phospholipids and cholesterol, the main components of cell membranes. Extracellular vesicles-biological nanoparticles released from cells-while resembling liposomes in size, shape, and structure, have a more complex composition with up to hundreds of different types of lipids, proteins, and carbohydrates in their membranes, as well as an internal cargo. Although nanoparticle technologies have revolutionized drug delivery by enabling passive and active targeting, increased stability, improved solubilization capacity, and reduced dose and adverse effects, the clinical translation remains challenging due to manufacturing limitations such as laborious and time-consuming procedures and high batch-to-batch variability. A sea change occurred when microfluidic strategies were employed, offering advantages in terms of precise particle handling, simplified workflows, higher sensitivity and specificity, and good reproducibility and stability over bulk methods. This review examines scientific advances in the microfluidics-mediated production of lipid-based nanoparticles for therapeutic applications. We will discuss the preparation of liposomes using both hydrodynamic focusing of microfluidic flow and mixing by herringbone and staggered baffle micromixers. Then, an overview on microfluidic approaches for producing extracellular vesicles and extracellular vesicles-mimetics for therapeutic applications will describe microfluidic extrusion, surface engineering, sonication, electroporation, nanoporation, and mixing. Finally, we will outline the challenges, opportunities, and future directions of microfluidic investigation of lipid-based nanoparticles in the clinic.

Abstract:Microfluidic systems provide a powerful platform for preparing, functionalizing, and manipulating nano-/micro-materials. In this review, we summarize the characteristics of state-of-the-art microfluidic techniques and their application to processing novel nano-/micro-materials with tunable size, shape, morphology, composition, and properties. A variety of synthesis strategies based on novel microfluidic devices are presented and compared, including their integration with real-time monitoring, external fields, and automatic feedback control. Novel functional nano-/micro-materials with complex structures have been controllably prepared and post-processed to obtain high-quality products. Strategies for controlling and tuning physical and chemical properties are also examined in terms of the design and function of microfluidic platforms. We provide an outlook on future developments in the processing of nano-/micro-materials by microfluidic technologies. Abstract:Three-dimensional (3D) printing, also called additive manufacturing (AM) or rapid prototyping (RP), is a layer by layer manufacturing method and now has been widely used in many areas such as organ printing, aerospace and industrial design. Now 3D printed microfluidics attract more and more interests for its rapid printing in the lab. In this review, we focused on the advances of 3D printed microfluidic chips especially the use in the chemistry and biology (vascularization and organs on chips). Based on the brief review of different 3D printing methods, we discussed how to choose the suitable 3D printing methods to print the desired microfluidics. We predict that microfluidics will be evolved from 2D chips to 3D cubes, printed hydrogel-based microfluidics will be reported and widely used, sensors & actuators could be integrated in the microfluidics during printing, and rapid assembling chips with printed microfluidic modules will be popular in the near future.

Abstract:Nanoparticles (NPs) have attracted tremendous interest in drug delivery in the past decades. Microfluidics offers a promising strategy for making NPs for drug delivery due to its capability in precisely controlling NP properties. The recent success of mRNA vaccines using microfluidics represents a big milestone for microfluidic NPs for pharmaceutical applications, and its rapid scaling up demonstrates the feasibility of using microfluidics for industrial-scale manufacturing. This article provides a critical review of recent progress in microfluidic NPs for drug delivery. First, the synthesis of organic NPs using microfluidics focusing on typical microfluidic methods and their applications in making popular and clinically relevant NPs, such as liposomes, lipid NPs, and polymer NPs, as well as their synthesis mechanisms are summarized.
