Abstract:Agaricus bisporus (Lange) Sing. Chaidam, a special brown mushroom with thick body indigenous in Chaidam basin, was used for fungal chitosan extraction. FTIR, XRD and DSC spectra showed that fungal chitosan was similar to commercial chitosan from aquatic sources. Fungal chitosan and commercial chitosan were used to coat on betulinic acid-loaded liposomes modified with biosurfactants mannosylerythritol lipid A (MEL-A), respectively. After chitosan coating, the mean size, zeta potential and encapsulation efficiency of both liposomes increased. The liposomes coated with fungal chitosan were discovered to have smaller size and higher zeta potential. Furthermore, the wall material MEL-A and coating material chitosan endue liposomes with increased antioxidant capacity. Fungal chitosan coated liposomes also have stronger antioxidant effects than commercial chitosan. The findings implied that the fungal chitosan coated liposomes modified with MEL-A can be considered as a promising delivery system with enhanced antioxidant effects for bioactive compounds.
Deciphering the Role of Burkholderia Cenocepacia Membrane Proteins in Antimicrobial Properties of Chitosan

Abstract:Chitosan, a versatile derivative of chitin, is widely used as an antimicrobial agent either alone or mixed with other natural polymers. Burkholderia cenocepacia is a multidrug-resistant bacteria and difficult to eradicate. Our previous studies shown that chitosan had strong antibacterial activity against B. cenocepacia. In the current study, we have investigated the molecular aspects for the susceptibility of B. cenocepacia in response to chitosan antibacterial activity. We have conducted RNA expression analysis of drug efflux system by RT-PCR, membrane protein profiling by SDS–PAGE, and by LC-MS/MS analysis following the validation of selected membrane proteins by real-time PCR analysis. By RT-PCR analysis, it was found that orf3, orf9, and orf13 were expressed at detectable levels, which were similar to control, while rest of the orf did not express. Moreover, shotgun proteomics analysis revealed 21 proteins in chitosan-treated cells and 16 proteins in control. Among them 4 proteins were detected as shared proteins under control and chitosan-treated cells and 17 proteins as uniquely identified proteins under chitosan-treated cells. Among the catalog of uniquely identified proteins, there were proteins involved in electron transport chain and ATP synthase, metabolism of carbohydrates and adaptation to atypical conditions proteins which indicate that utilization and pattern of chitosan is diverse which might be responsible for its antibacterial effects on bacteria. Moreover, our results showed that RND drug efflux system, which display the ability to transport a variety of structurally unrelated drugs from a cell and consequently are capable of conferring resistance to a diverse range of chemotherapeutic agents, was not determined to play its role in response to chitosan. It might be lipopolysaccharides interaction with chitosan resulted in the destabilization of membrane protein to membrane lyses to cell death. Membrane proteome analysis were also validated by RT-qPCR analysis, which corroborated our results that of membrane proteins.
Novel antifungal mechanism of oligochitosan by triggering apoptosis through a metacaspase-dependent mitochondrial pathway in Ceratocystis fimbriata

Abstract:The antifungal effects of oligochitosan (OCS) against Ceratocystis fimbriata that causes black rot disease in sweet potato and its apoptosis mechanism were evaluated. OCS restrained the mycelial growth and spores germination of C. fimbriata, and decreased the ergosterol content of cell membrane. Transmission electron microscopy observation and flow cytometry analysis revealed that OCS induced morphology changes with smaller size and increased granularity of C. fimbriata, which was the typical feature of apoptosis. To clarify the apoptosis mechanism induced by OCS, a series of apoptosis-related parameters were analyzed. Results showed that OCS induced reactive oxygen species accumulation, Ca2+ homeostasis dysregulation, mitochondrial dysfunction and metacaspase activation, coupled with hallmarks of apoptosis including phosphatidylserine externalization, DNA fragmentation, and nuclear condensation. In summary, OCS triggered apoptosis through a metacaspase-dependent mitochondrial pathway in C. fimbriata. These findings have important implications for the application of OCS to control pathogens in food and agriculture.

Abstract:Chitosan has wide-spectrum antimicrobial activity but knowledge of its antifungal mechanism is still incomplete. In this study, transcriptome of Penicillium expansum upon chitosan treatment was analyzed by RNA-Seq. KEGG enrichment analysis revealed that endocytosis as well as other physiological pathways was regulated by chitosan treatment. Clathrin adaptor protein mu-subunit (PeCAM) gene, which encodes a protein associated with clathrin-dependent endocytosis, was up-regulated after chitosan treatment. Deletion of PeCAM resulted in changes of conidial, hyphal and colonial morphology. Confocal microscopy images of the distribution of fluorescein isothiocyanate-labeled chitosan confirmed cellular internalization of chitosan. However, deletion of PeCAM almost completely blocked uptake of chitosan into fungal cells and ΔPeCAM mutant exhibited less sensitivity to chitosan compared with wild type, suggesting that chitosan uptake is mediated by clathrin-dependent endocytosis and internalized chitosan also plays an important role in its antifungal activity. Collectively, our results provide a new insight into the antifungal mechanism of chitosan.

Abstract:Heteroleptic metal complexes containing CuII, CoII, and ZnII, incorporating curcumin and a Schiff base ligand (L), were synthesized and characterized, and their antifungal activity was evaluated. Their antifungal activities were investigated individually and in combination with fluconazole. Utilizing various analytical techniques such as UV-Vis, FT-IR, NMR, ESI-MS, TGA-DTG, elemental analyses, conductance, and magnetic susceptibility measurements, complex C1 ([Cu(Cur)LCl(H2O)]) was assigned a distorted octahedral geometry, while complexes C2 ([Co(Cur)LCl(H2O)]) and C3 ([Zn(Cur)LCl(H2O)]) were assigned octahedral geometries. Among these complexes, C2 exhibited the highest inhibitory activity against both FLC-susceptible and resistant strains of Candida albicans. Furthermore, C2 demonstrated candidicidal activity and synergistic interactions with fluconazole, effectively inhibiting the growth and survival of both FLC-resistant and FLC-sensitive C. albicans strains. The complex displayed a dose-dependent inhibition of drug efflux pumps in FLC-resistant C. albicans strains, indicating its potential to disrupt the cell membrane of these strains. The significant role of membrane efflux transporters in the development of antifungal drug resistance within Candida species has been extensively documented and our findings indicate that complex C2 specifically targets this crucial factor, thereby playing a pivotal role in mitigating drug resistance in C. albicans.

Abstract:As a destructive fungus-like plant pathogen, the oomycete Phytophthoracapsici is unable to synthesize its own ergosterol as the potential target of fungicide cinnamaldehyde (CA). In this study, CA exerted efficient inhibitory effects on both mycelial growth (EC50=0.75 mM) and zoospore germination (MIC=0.4 mM) of P. capsici. CA-induced immediate Ca(2+) efflux from zoospores could be confirmed by the rapid decrease in intracellular Ca(2+) content determined by using Fluo-3 AM and the increase in extracellular Ca(2+) concentration determined by using ICP-AES (inductively coupled plasma atomic emission spectrometry). Blocking Ca(2+) influx with ruthenium red and verapamil led to a higher level of CA-induced Ca(2+) efflux, suggesting the simultaneous occurrence of Ca(2+) influx along with the Ca(2+) efflux under CA exposure. Further results showed that EGTA-induced decrease in intracellular Ca(2+) gave rise to the impaired vitality of P. capsici while the addition of exogenous Ca(2+) could suppress the growth inhibitory effect of CA. These results suggested that Ca(2+) efflux played an important role in CA-induced growth inhibition of P. capsici. The application of 3-phenyl-1-propanal, a CA analog without α,β- unsaturated bond, resulted in a marked Ca(2+) influx in zoospores but did not show any growth inhibitory effects. In addition, exogenous cysteine, an antagonist against the Michael addition (the nucleophilic addition of a carbanion or another nucleophile) between CA and its targets, could attenuate CA-induced growth inhibition of P. capsici by suppressing Ca(2+) efflux. Our results suggest that CA inhibits the growth of P. capsici by stimulating a transient Ca(2+) efflux via Michael addition, which provides important new insights into the antimicrobial action of CA.

Abstract:The application of Chitosan (CS) in drug delivery systems, plant growth promotion, antibacterial potentiality and plant defense is significantly limited by its inability to dissolve in neutral solutions. In this work, CS with different molecular weights (M w ) has been oxidized, yielding five kinds of oxidized chitosan (OCS 1–5) with solubilities in neutral solutions. The results obtained from Fourier Transform Infrared Spectroscopy clearly showed the successful oxidation of the hydroxyl group to form aldehyde and carboxyl groups. And the CS derivatives showed the wrinkled and lamellar structures on the surface of OCS. The results of antifungal activity against Fusarium graminearum showed that the OCS dissolved in 2 % (V/V) acetic acid exhibited better performance of almost complete inhibition of mycelial growth compared with CS at the concentration of 500 μg/mL. Among the five OCS, OCS-4 exhibited the best antifungal effect and had the lowest EC 50 value of 581.68 μg/mL in samples. OCS-4 displayed superior promoting effect on seed germination with a germination potential of 62.2 % at a concentration of 3 g/L and a germination rate of 74.5 %. Additionally, the other four OCS also showed excellent antifungal activity with dose-dependent manners. These results indicated that the OCS had excellent antifungal potential in agricultural production.

Abstract:Chitosan-based nanocomposites (CS NCs) are gaining considerable attention as multifaceted antifungal agents. This study investigated the antifungal activity of NCs against two phytopathogenic strains: Fusarium solani (F. solani) and Alternaria solani (A. solani) . Moreover, it sheds light on their underlying mechanisms of action. The NCs, CS-ZnO, CS-CuO, and CS-SiO 2 , were characterized using advanced methods. Dynamic and electrophoretic light scattering techniques revealed their size range (60–170 nm) and cationic nature, as indicated by the positive zeta potential values (from +16 to +22 mV). Transmission electron microscopy revealed the morphology of the NCs as agglomerates formed between the chitosan and oxide components. X-ray diffraction patterns confirmed crystalline structures with specific peaks indicating their constituents. Antifungal assessments using the agar diffusion technique demonstrated significant inhibitory effects of the NCs on both fungal strains (1.5 to 4-fold), surpassing the performance of the positive control, nystatin. Notably, the NCs exhibited superior antifungal potency, with CS-ZnO NCs being the most effective. A. solani was the most sensitive strain to the studied agents. Furthermore, the tested NCs induced oxidative stress in fungal cells, which elevated stress biomarker levels, such as superoxide dismutase (SOD) activity and protein carbonyl content (PCC), 2.5 and 6-fold for the most active CS-CuO in F. solani respectively Additionally, they triggered membrane lipid peroxidation up to 3-fold higher compared to control, a process that potentially compromises membrane integrity. Laurdan fluorescence spectroscopy highlighted alterations in the molecular organization of fungal cell membranes induced by the NCs. CS-CuO NCs induced a membrane rigidifying effect, while CS-SiO 2 and CS-ZnO could rigidify membranes in A. solani and fluidize them in F. solani . In summary, this study provides an in-depth understanding of the interactions of CS-based NCs with two fungal strains, showing their antifungal activity and offering insights into their mechanisms of action. These findings emphasize the potential of these NCs as effective and versatile antifungal agents.

Abstract:As the increasing number of the individuals suffering from AIDs, chemotherapy, and radiotherapy, pathogenic fungi, which may rapidly grow and invade the host tissues in these immune-compromised patients, is becoming great threat to human health. In this study, we constructed a novel fungal pathogen-responsive assembly of cuprous oxide (Cu2O) nanoparticles (NPs) for specific targeting and inhibiting growth and biofilm formation of the representative fungal pathogen, Candida albicans (C. albicans). This assembly was formed by coating the initial Cu2O NPs with both phosphatidylethanolamine (PE) and bovine serum albumin (BSA), followed by hydrophobic/electrostatic interaction-driven formation of the Cu2O-PE-BSA microaggregates. The formed microaggregates could be induced for disassembly by the fungal pathogen C. albicans, leading to close binding of the NPs to the cell wall of the pathogen. Both confocal microscopy and viability assays showed that the assembly strongly inhibited growth and biofilm formation of the pathogen, but had extreme low toxicity to mammalian cells. In vivo mouse wound model further revealed that the assembly had high capacity of healing the fungus-infected wounds and reduced the fungal burden of the wound tissues. This study sheds a novel light on facile development of pathogen-responsive nano-assemblies for efficient and safe antifungal therapy.

The Sodium/Calcium Exchanger PcNCX1-Mediated Ca2+ Efflux is Involved in Cinnamaldehyde-Induced Cell-Wall Defects of Phytophthora Capsici

Abstract:Phytophthora capsici is one of the devastating pathogens, causing foliar blight, root rot, and fruit rot in peppers. Cinnamaldehyde (CA) is a natural compound coming from Cinnamomum cassia. The medicinal properties of CA have been widely identified. Limited knowledge is known about the application of CA in agriculture. In this study, CA significantly inhibited P. capsici, which further suppressed Phytophthora blights in both pepper seedlings and pepper fruits. Treatment with CA resulted in collapsed and fragmented hyphae, accompanying the increase in MDA (malondialdehyde) content and the decrease in intercellular glycerol content in hyphae. CA also inhibited the growth of wild type yeast. The yeast mutant ΔYvc1 with a deletion of Yvc1 (a Ca2+ transporter) showed decreased sensitivity to CA. The transformation of PcNCX1, a sodium/calcium exchanger from P. capsici, into ΔYvc1 restored its sensitivity to CA. The transformant carrying PcNCX1 also showed restored Ca2+ efflux upon CA treatment. RNA-seq analysis showed that CA treatments resulted in the down-regulation of a set of genes encoding for calcium-related proteins. Collectively, our study demonstrates that the antifungal activity of CA against P. capsici may be associated with PcNCX1-mediated Ca2+ efflux. Our results provide crucial insights into the antimicrobial action of CA.
Chitosan-mediated copper nanohybrid attenuates the virulence of a necrotrophic fungal pathogen Macrophomina phaseolina

Abstract:This study reported the synthesis and characterization of chitosan-copper nanoparticles (Ch-CuNPs) using a 1% copper sulfate solution in 0.2% w/v chitosan. The Ch-CuNPs, displaying a stable brick-red hue, showed an absorption peak at 572 nm, indicative of monodisperse nanoparticle formation and surface plasmon resonance. X-ray diffraction confirmed the face-centered cubic structure with peaks at 36.78°, 43.38°, 50.56°, and 74.26°, and an average particle size of 87–89 nm. FTIR analysis showed interactions between chitosan and copper, particularly around 3370 –3226 cm−1, 1633 cm−1, and 680 cm−1. In vitro assays revealed that Ch-CuNPs inhibited Macrophomina phaseolina growth by 18–71% at 0.03–0.09% concentrations, achieving complete inhibition at 0.12–0.15%, with PCA analysis confirming that growth peaked at lower concentrations and sharply declined at higher levels. Ch-CuNPs also altered fungal morphology and enzyme activity, with notable degradation at higher concentrations. The Cu uptake by the fungus peaked at 29.9% with 0.03% Ch-CuNPs and decreased at higher concentrations. FTIR analysis showed shifts and disappearance of peaks in fungal biomass treated with Ch-CuNPs, indicating molecular interactions and potential structural changes. This study underscores the potential of Ch-CuNPs as an effective antifungal agent and elucidates their interaction mechanisms.

Encapsulating insoluble antifungal drugs into oleic acid-modified silica mesocomposites with enhanced fungicidal activity

Abstract:Recently, with increasing medical practices, including organ transplantation and tumor chemotherapy, fungal infections, particularly the occurrence of drug-resistant fungal strains, remain a severe problem for the public health, which cause worse complications in the immunocompromised patients. The search for efficacious yet safe antifungal agents is in high demand in precision medicine. However, fungicides are often poorly water soluble for oral absorption, which is difficult for pharmaceutical efficacy evaluation. In this study, lipophilic oleic acid (OA)-grafted mesoporous silica (SBA-15) was facilely modified by cetyltrimethylammonium bromide (CTAB), which acts as an efficient antifungal drug matrix of itraconazole (ITZ). Characterized by physicochemical methods, the rod-like SBA-15-OA-CTAB/ITZ composite with retained mesostructural regularity shows that the loading amount of ITZ in the mesopore is ∼18%, contributing to the enhanced antifungal activity against Aspergillus fumigatus (A. fumigatus) and Candida albicans (C. albicans). The antimicrobial mechanism study suggests that the reactive oxygen species (ROS) were formed when fungal cells were incubated with the formulated ITZ, while there was no ROS formation in the presence of pure ITZ, which may result from the quaternary ammonium moieties of CTAB in the nanocomposites. Due to the potential toxicity of CTAB on mammalian cells, the as-synthesized mesoporous SBA-15-OA-CTAB/ITZ provides an alternative molecular design for the formulation improvement of a lipophilic antifungal drug applicable for external uses such as topical therapy.

Abstract:This paper aims to identify the interaction of chitosan (CTS) with bacterial membrane and delineate the inhibition mechanism of CTS towards Gram-negative bacteria. The inhibition activity of CTS against Escherichia coli (E. coli) was evaluated by measuring O.D610nm. CTS have increased the permeabilities of the outer membrane (OM) and inner membrane (IM) of E. coli, and ultimately disrupted the cell membrane with the release of cellular contents. Morphologies of bacteria treated with CTS were observed by Transmission electron microscopy (TEM), which indicated that it was the primary inhibition action for CTS to damage the bacterial cell membranes. The interaction between CTS and a model phospholipids membrane was studied, and the precipitates formed with CTS and phosphatidylcholine (PC) suggested the damage of bacterial cell membranes was likely to be caused by the electrostatic interaction between NH3+ groups of CTS and carbonyl and phosphoryl groups of the phospholipid components of the cell membrane.

Abstract:Fungal infections have been a concern for decades, yet effective and approved antifungal agents are limited. We recently developed a potential method to enhance the antifungal activity of a small chitin-binding domain (LysM) from Pteris ryukyuensis chitinase A (PrChiA) by the site-specific introduction of a palmitoyl (C16) group catalyzed by microbial transglutaminase (MTG). Herein, we attempted the conjugation of a series of lipid-peptide substrates with LysM genetically fused with a C-terminal MTG-reactive Q-tag (LysM-Q) to yield LysM-lipid conjugates (LysM-lipids) with different lengths (LysM-C12, -C14, and -C16) and different numbers of alkyl chains [LysM-(C12) , - (C14) , and -(C16) ]. The enzymatic conjugation proceeded smoothly for all LysM-lipids, except for LysM-(C16) because of the low aqueous dispersibility of the hydrophobic (C16) lipid-peptide substrate. The combination of amphotericin B (AmB) with LysM-C14 or LysM-C16 exhibited the highest antifungal performance against Trichoderma viride whereas alterations in the number of alkyl chains were not effective in enhancing the antifungal activity of the LysM-lipids. Fluorescent microscopic analysis showed that the fungal cell wall was stained with C14- and C16-modified LysM-muGFP fusion proteins when combined with AmB, suggesting a synergistic action of AmB and LysM-lipids with a suitable lipid length. All LysM-lipids showed minimum cytotoxicity toward mammalian cells, suggesting that LysM-lipids could be a safe additive in the development of new antifungal formulations.

Abstract:The current work aims to take full advantage of the structural superiority and hepatoprotective effect of nutriment trans-p-coumaric acid (TPCA) to decrease the solubility and dissolution rate of antimycotic agent 5-fluorocytosine (FLCY) by co-crystallization strategie, thereby giving a novel understanding into conquering dose-limiting hepatotoxicity induced by quick and almost complete absorption of FLCY. Toward that end, FLCY-TPCA, as the first cocrystal of FLCY with phenolic acid nutriment of hydroxycinnamic acid types, is orientationally self-assembled and fully characterized. The single crystal structure identifies that TPCA molecules in the crystal lattice capture the polar sites in situ of FLCY via robust hydrogen bond interplays, thus creating a vital planar construction (VPC) possessing hydrophobe feature, which results in the simultaneous down-regulation for the cocrystal in solubility and dissolution rate versus parent FLCY. The outcomes of the experiment are forcefully supported by related theory researches. Molecular electrostatic potential (MEP) calculation manifests that the VPC polarity in the cocrystal is lower than that of free FLCY, which conforms with the reduction in polarity due to intensifying hydrogen-bonds energy based on topology analysis with atoms in molecules (AIM) theory, hence conferring a conspicuous improvement in hydrophobicity on the cocrystal. Furthermore, the molecular planarity analysis on account of Multiwfn's wave function calculation module confirms the convergence in planarity at a two-dimensional infinite level, which undoubtedly contributes to accomplishing a tight three-dimensional supramolecular superposition landscape, corresponding with the coinstantaneous reinforcement of π-π stacking interaction analyzed by the reduced density gradient (RDG), and all these further support the fact that the hydrophobicity of FLCY-TPCA cocrystal is significantly enhanced. As a result, the current contribution exemplifies the feasible cocrystallization clues by integrated experimental with systematic theoretical approaches to overcoming intractable toxicity issues of antifungal agents.

Abstract:Planktonic bacteria tend to form sessile community architectures to shield resident bacteria from various environmental stresses. The formed biofilm leads to the failure of conventional antimicrobial therapy. Extracellular macromolecules, including extracellular DNA (eDNA), proteins, lipids, and polysaccharides, crosslink into gel-like structures through electrostatic forces in the mature biofilm matrix. The stereo-structural integrity and chemical inertia of the extracellular polymeric matrix result in comprehensive antimicrobial resistance to antibacterial polysaccharides. Herein, an ionic gelation method was employed to functionalize cationic chitosan nanoparticles (CSNPs) with chloroquine and deoxyribonuclease. The modification involved shifting eDNA chirality through a DNA-intercalating agent, chloroquine, and hydrolyzing an eDNA scaffold with deoxyribonuclease. The antibiofilm activity was assessed against a standard Staphylococcus aureus strain and clinical subtype isolates. Functional modifications targeting eDNA improved the chitosan anti-biofilm efficiency (residual biomass decreased from 74.2 to 90.3 % to 16.7-24.6 %) by disrupting the biofilm matrix. The functional CSNPs worked as a sensitizer prodrug, contributing to a bactericidal process of chitosan itself (cell wall damage increased from 11.38-18.16 % to 55.2-61.4 %) by dispersing the biofilm-enclosed bacteria. In vivo, the bacterial burden of infected mouse joints was reduced by 4.1 lg CFU/mL. Our results indicate the potential of this chitosan-based anti-infection strategy in biofilm-related infections.

Abstract:Bowen's disease of the skin may have differing clinical presentations depending on the skin surface on which it presents, but has the same histologic features of squamous cell carcinoma in situ wherever it occurs. The etiologic factors include ultraviolet light and human papillomavirus infection. The choice of therapy requires a consideration for retention of form, function and cosmosis while offering a high cure rate. The immunodualtory agent imiquimod has been shown to be an effective treatment on a variety of skin surfaces.
Synthesis, antimicrobial activity, antioxidant activity and molecular docking of novel chitosan derivatives containing glycine Schiff bases as potential succinate dehydrogenase inhibitors

Abstract:Succinate dehydrogenase (SDH) is an important inner mitochondrial membrane-bound enzyme involved in redox reactions during the tricarboxylic acid cycle. Therefore, a series of novel chitosan derivatives were designed and synthesized as potential microbicides targeting SDH and precisely characterized by FTIR, 1H NMR and SEM. Their antifungal and antibacterial activities were evaluated against Botrytis cinerea, Fusarium graminearum, Staphylococcus aureus and Escherichia coli. The bioassays revealed that these chitosan derivatives exerted significant antifungal effects, with four of the compounds achieving 100 % inhibition of Fusarium graminearum merely at a concentration of 0.5 mg/mL. Additionally, CSGDCH showed 79.34 % inhibition of Botrytis cinerea at a concentration of 0.1 mg/mL. In vitro antibacterial tests revealed that CSGDCH and CSGDBH have excellent Staphylococcus aureus and Escherichia coli inhibition with MICs of 0.0156 mg/mL and 0.03125 mg/mL, respectively. Molecular docking studies have been carried out to explore the binding energy and binding mode of chitosan and chitosan derivatives with SDH. The analyses indicated that chitosan derivatives targeted the active site of the SDH protein more precisely, disrupting its normal function and ultimately repressing the growth of microbial cells. Furthermore, the chitosan derivatives were also evaluated biologically for antioxidation, and all of these compounds had a greater degree of reducing power, superoxide radical, hydroxyl radical and DPPH-radical scavenging activity than chitosan. This research has the potential for the development of agricultural antimicrobial agents.
Cinnamaldehyde inhibits the growth of Phytophthora capsici through disturbing metabolic homoeostasis

Abstract:Background: Phytophthora capsici Leonian (P. capsici) can cause wilting and roots rotting on pepper and other cash crops. The new fungicide cinnamaldehyde (CA) has high activity against this pathogen. However, its potential mechanism is still unknown. Methods: In order to gain insights into the mechanism, isobaric tags for relative and absolute quantification (iTRAQ)-based quantitative proteomics was used to analyze P. capsici treated with CA. The iTRAQ results were evaluated by parallel reaction monitoring (PRM) analysis and quantitative real-time PCR (qRT-PCR) analysis. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis was used to speculate the biochemical pathways that the agent may act on. Results: The results showed that 1502 differentially expressed proteins were identified, annotated and classified into 209 different terms (like metabolic process, cellular process, single-organism process) based on Gene Ontology (GO) functional enrichment analysis and nine different pathways (glyoxylate and dicarboxylate metabolism, fatty acid metabolism and so on) based on the Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. This study suggested that CA disordered fatty acid metabolism, polysaccharide metabolism and leucine metabolism. Based on PRM analysis, five proteins including CAMK/CAMK1 protein kinase, glucan 1,3-beta-glucosidase, 1,3-beta-glucanosyltransferase, methylcrotonoyl-CoA carboxylase subunit alpha and isovaleryl-CoA dehydrogenase were down-regulated in P. capsici treated with CA. Furthermore, the qRT-PCR analysis showed that the gene expression level of the interested proteins was consistent with the protein expression level, except for CAMK/CAMK1 protein kinase, acetyl-CoA carboxylase and fatty acid synthase subunit alpha. Conclusions: CA destroyed the metabolic homoeostasisof P. capsici, which led to cell death. This is the first proteomic analysis of P. capsici treated with CA, which may provide an important information for exploring the mechanism of the fungicide CA against P. capsici.
