Abstract:Mercury presents a potential risk to soil organisms, yet our understanding of mercury bioaccumulation in soil dwelling organisms is limited. The influence of soil geochemistry and digestive processes on both methylmercury (MeHg) and total mercury (THg) bioavailability to earthworms (Pheretima guillemi) was evaluated in this study. Earthworms were exposed to six mercury-contaminated soils with geochemically contrasting properties for 36 days, and digestive fluid was concurrently collected to solubilize soil-associated mercury. Bioaccumulation factors were 7.5-31.0 and 0.2-0.6 for MeHg and THg, respectively, and MeHg accounted for 17-58% of THg in earthworm. THg and MeHg measured in soils and earthworms were negatively associated with soil total organic carbon (TOC). Earthworm THg and MeHg also increased with increasing soil pH. The proportion of MeHg and THg released into the digestive fluid (digestive solubilizable mercury, DSM) was 8.3-18.1% and 0.4-1.3%, respectively. The greater solubilization of MeHg by digestive fluid than CaCl2, together with a biokinetic model-based estimate of dietary MeHg uptake, indicated the importance of soil ingestion for MeHg bioaccumulation in earthworms.

Abstract:This study was conducted to find an appropriate approach for the assessment of bioavailability of DDTs in soil to both earthworm and vegetables. Four chemical approaches—Soxhlet extraction with n-hexane, n-butanol agitation extraction, water agitation extraction, and matrix solid-phase microextraction (matrix-SPME)—were used to assess the relationships between the extractability of 1,1-dichloro-2,2-bis(p-chlorophenyl) ethylene (p,p′-DDE), 1,1,1-trichloro-2-(p-chlorophenyl)-2-(o-chlorophenyl) ethane (o,p′-DDT), 1,1-dichloro-2,2-bis(p-chlorophenyl) ethane (p,p′-DDD), and 1,1,1-trichloro-2,2-bis(p-chlorophenyl) ethane (p,p′-DDT) in soil and their amounts uptaken by the earthworm (Eisenia foetida), Chinese cabbage (Brassica campestris L. spp.), and cole (Brassica napus L.). These results indicated that the extractability and bioavailability of DDTs in soil decreased with time of aging. Correlation analysis showed that n-butanol extraction or 12-h matrix-SPME could be used to assess the bioavailability of DDTs to the earthworm, and Soxhlet extraction, n-butanol extraction, or 12-h matrix-SPME could be used to predict the bioavailability of DDTs to both Chinese cabbage and cole. As a solventless, time-efficient, and negligible-depletion technique, it could be concluded that matrix-SPME is a better approach to predict the bioavailability of DDTs to both the earthworm and vegetables, compared with Soxhlet extraction, n-butanol extraction, and water extraction.

Abstract:This study aimed to optimize the methods for sampling and analyzing methylmercury (MeHg) concentrated within diffusive gradients in thin films (DGT) and its application to different water bodies. We explored the elution solution for MeHg, comprised of 1.13 mM thiourea and 0.1M HCl, optimizing its volume to 50 mL. In addition, we found that it is necessary to analyze the entire extraction solution after adjusting its pH, to ensure completion of the ethylation reaction. The DGT samplers were deployed in two distinct aquatic environments (i.e., Okjeong Lake and Nakdong River) for up to 6 weeks, and this study demonstrated to predict the time-weighted average concentration with a diffusion coefficient of 7.65 × 10 −6 cm 2 s −1 for MeHg in the diffusive gel. To assess the diffusive boundary layer (DBL) effects, the DGT samplers with different agarose diffusive gel thickness were deployed. The mass of MeHg accumulated in the DGT resin at a given time decreased with increasing diffusive gel thickness, because of creating longer diffusion pathways within thicker gels. The labile MeHg concentration estimated by the DGT in Okjeong Lake and Nakdong River are found in the range of 61–111 and 55–105 pg L −1 , respectively, which were found to be similar to the grab sampling data. Additionally, this study evaluated depth-dependent MeHg in Okjeong Lake. The vertical profile results showed that the concentration of MeHg at the depth of 2.3 and 15.7 m are about 1.5 and 4.6 times of the DGT installed at 0.3 m of the surface layer, respectively, suggesting potential mercury methylation in deep waters. These findings have practical implications for predicting bioavailability, assessing risks, and formulating strategies for water body management and contamination remediation.

Abstract:Mercury-contaminated sediment and water contain various Hg species, with a small fraction available for microbial conversion to the bioaccumulative neurotoxin monomethylmercury (MeHg). Quantification of this available Hg pool is needed to prioritize sites for risk management. This study compared the efficacy of diffusive gradient in thin-film (DGT) passive samplers to a thiol-based selective extraction method with glutathione (GSH) and conventional filtration (<0.2 μm) as indicators of Hg bioavailability. Anaerobic sediment slurry microcosms were amended with isotopically labeled inorganic Hg "endmembers" (dissolved Hg2+, Hg-humic acid, Hg-sorbed to FeS, HgS nanoparticles) with a known range of bioavailability and methylation potentials. Net MeHg production (expressed as percent of total Hg as MeHg) over 1 week correlated with mass accumulation of Hg endmembers on the DGTs and only sometimes correlated with the 0.2 μm filter passing Hg fraction and the GSH-extractable Hg fraction. These results suggest for the first time that inorganic Hg uptake in DGTs may indicate bioavailability for methylating microbes. Moreover, the methylating microbial community assessed by hgcA gene abundance was not always consistent with methylation rates between the experiments, indicating that knowledge of the methylating community should target the transcript or protein level. Altogether, these results suggest that DGTs could be used to quantify the bioavailable Hg fraction as part of a method to assess net MeHg production potential in the environment.

Abstract:Historically as part of its national security mission, the U.S. Department of Energy's Y-12 National Security Facility in Oak Ridge, TN acquired a significant fraction of the world's supply of elemental mercury. During the 1950s and 1960s, a large amount of elemental mercury escaped confinement and is still present in the watershed surrounding the Y-12 facility. Earthworms are key components in natural food chains, providing a food source for many small mammals and important food sources for small birds. The objectives of this study were to investigate the current status of mercury distribution and speciation and determination of mercury bioavailability to native earthworms in floodplain soils of East Fork Poplar Creek (EFPC) after decades of U.S. Department of Energy's remediation. The present study clearly shows that the total mercury in a tested floodplain field of EFPC was significantly below the US Department of Energy target 400mg Hg/kg. The major mercury form in the current floodplain soils of EFPC is mainly the non-cinnabar mercury bound form in soil silicates (4M HNO3-extractable residual fraction). The results show strong linear relationships between mercury concentrations in native earthworms (both mature and immature groups) and the non-cinnabar mercury form. Native earthworms may be used as a potential mercury ecological bio-indicator (bio-marker) for demonstrating mercury bioavailability and ecotoxicity in the ecosystem.
Appraising ecotoxicological risk of mercury species and their mixtures in sediments to aquatic biota using diffusive gradients in thin films (DGT)

Abstract:Mercury (Hg) is a global, persistent and inevitable pollutant, the toxicity of which is mostly reflected in its species including inorganic Hg (InHg) and methyl mercury (MeHg). Using diffusive gradients in thin films (DGT) is deemed as a reliable technique to determine the bioavailability of pollutants. This study is the first attempt to assess the integrated toxicity of mercury species mixtures in sediments to the aquatic biota based on the DGT technique. In the course, the Daya Bay under serious anthropogenic influences was selected as the study case. The results showed that the DGT concentrations of InHg and MeHg were detected as 0.30-1.93 μg/L and 0.28-1.94 μg/L respectively in the surface sediments collected from the Daya Bay. In terms of the toxicity of single mercury species, the risk quotient (RQ) values of InHg and MeHg significantly exceeded 1, indicating that the adverse effects of InHg and MeHg should not be ignored. In terms of the integrated toxicity of mercury species mixtures, the probabilistic biological risk assessment results demonstrate that Daya Bay features low (3.32%) probability of toxic effects in its surface sediments to the aquatic biota.

Abstract:Measuring the isotopic composition of Hg in natural waters is challenging due to the ultratrace level of aqueous Hg (ng L-1). At least 5 ng of Hg mass is required for Hg isotopic analysis. Given the low Hg concentration in natural waters, a large volume of water (>10 L) is typically needed. The conventional grab sampling method is time-consuming, laborious, and prone to contamination during transportation and preconcentration steps. In this study, a DGT (diffusive gradients in thin films) method based on aminopropyl and mercaptopropyl bi-functionalized SBA-15 nanoparticles was developed and extended to determine the concentration and isotopic composition of aqueous Hg for the first time. The results of laboratory analysis showed that Hg adsorption by DGT induces ∼ -0.2‰ mass-dependent fractionation (MDF) and little mass-independent fractionation (MIF). The magnitude of MDF exhibits a dependence on the diffusion-layer thickness of DGT. Since Hg-MDF can occur in a broad range of environmental processes, monitoring the δ202Hg of aqueous Hg using the DGT method should be performed with caution. Field results show consistent MIF signatures (Δ199Hg) between the DGT and conventional grab sampling method. The developed DGT method serves as a passive sampling method that effectively characterizes the MIF of Hg in waters to understand the biogeochemical cycle of Hg at contaminated sites.

Abstract:The environmental and human health risks of arsenic (As) mainly depend on its bioavailability in the soil. However, precise determination of bioavailable As in situ is still challenging due to the complexity of soil types and hydrogeological conditions in polluted sites. Diffusive gradients in thin films (DGT) is one of the reliable analysis technology in the in situ analysis of bioavailable chemicals in waters, sediments and farmland soils. However, few studies have investigated the application of DGT in evaluating the bioavailability of heavy metals in brownfield soils. In this study, DGT was extended applied to evaluate the availability of As in 16 contaminated soil samples from seven brownfield sites. The correlation between the bioavailable As measured by DGT and the bioaccessible As determined by the Unified BRAGE method (UBM) assay was also analyzed. In addition, the human health risks were evaluated, and the DGT-based prediction model for bioavailable As was also developed (R 2=0.852 , P < 0.001). The results indicated that considering bioavailability in risk assessments can improve the accuracy of calculations for site-specific health risks and provide guidance for effective remediation. The study will demonstrate the effectiveness of DGT in characterizing bioavailable heavy metals in contaminated site soils and provide decision-making support for the soil remediation.

Eisenia fetida impact on cadmium availability and distribution in specific components of the earthworm drilosphere

Abstract:Although the potential of vermiremediation for restoring metal-contaminated soils is promising, the effects of earthworms on the availability of soil metals are still debatable. Most previous studies considered the soil as a "whole black box." Mobilization or immobilization of metals are affected by earthworm activities within drilosphere hotspots under different soil conditions, which has not been specifically studied. Therefore, an improved 2D terrarium was designed to study the impact of earthworm activities on cadmium (Cd) fate in the drilosphere hotspots (burrow wall soils, burrow casts, and surface casts) of different artificially spiked Cd treatments (CK: 0 mg kg-1; LM: 1 mg kg-1; and HM: 5 mg kg-1) with different organic amendments (2% and 10%). The results revealed that Cd increased earthworm activities with the highest cast production in HM and the highest burrow length in LM. Earthworms exhibited a stronger tendency to reduce total Cd concentration by 4.48-13.58% in casts of LM soils, while 3.37-5.22% in burrow walls under HM treatments. Overall, earthworms could increase the availability of Cd in casts under all conditions (55.46-121.01%). The organic amendments decreased the total Cd concentration and increased the availability of Cd in the disturbed soil. A higher amount of organic amendment significantly decreased total Cd concentration of the drilosphere by 1.16-5.83% in LM and HM treatments, while increasing DTPA-Cd concentrations in all components by 23.13-55.20 %, 14.63-35.11%, and 3.30-11.41% in CK, LM, and HM treatments, respectively, except for earthworm non-disturbed soil and no-earthworm soil in HM treatments. Redundancy analysis (RDA) revealed that the moisture, pH, and total carbon contents in soil are the main factors affecting Cd bioavailability. In this study, we decoded the "black box" of soil by making it relatively simple to better understand the effects and mechanisms of earthworm activities on soil metal availability and consequently provided comprehensive insights for using earthworms in soil vermiremediation.

Abstract:Mercury uptake dynamics in the earthworm Pheretima guillemi, including the dissolved uptake rate constant (ku) from pore-water and assimilation efficiencies (AEs) from mercury-contaminated soil, was quantified in this study. Dissolved uptake rate constants were 0.087 and 0.553 L g(-1) d(-1) for inorganic mercury (IHg) and methylmercury (MeHg), respectively. Assimilation efficiency of IHg in field-contaminated soil was 7.2%, lower than 15.4% of spiked soil. In contrast, MeHg exhibited comparable AEs for both field-contaminated and spiked soil (82.4-87.2%). Within the framework of biodynamic model, we further modelled the exposure pathways (dissolved exposure vs soil ingestion) to source the accumulated mercury in Pheretima guillemi. The model showed that the relative importance of soil ingestion to mercury bioaccumulation depended largely on mercury partitioning coefficients (K(d)), and was also influenced by soil ingestion rate of earthworms. In the examined field-contaminated soil, almost (>99%) accumulated IHg and MeHg was predicted to derive from soil ingestion. Therefore, soil ingestion should be carefully considered when assessing mercury exposure risk to earthworms.

Abstract:Mercury (Hg) is a toxic, non-essential element for living organisms, frequently present in high concentrations in soils from industrial areas. The total, dissolved, and labile Hg concentrations in garden soils and their accumulation in edible vegetables (onion, garlic, lettuce, and parsley) grown on contaminated soils in localities situated a former mining area were evaluated. The labile Hg fraction was estimated by diffusive gradient in thin films (DGT). The soil-to-vegetable transfer factors, as well as the health risk by exposure to Hg, were calculated based on the labile Hg concentration in soil. The total Hg concentration in soil varied widely (0.11–3.77 mg kg −1 ), Hg in soil solution ranged between 2.14 and 20.2 μg L −1 and labile Hg between 1.13 and 18.6 μg L −1 . About 36–96% (84% on average) of the Hg concentration in soil solution was found in labile form. Multivariate analysis revealed significant correlations between the labile Hg concentration in soil and Hg accumulated in vegetables. The hazard indices showed that, although the study area is affected by legacy pollution, exposure to soil and consumption of locally grown vegetables do not pose health risks.

Abstract:To assess the mixing characteristics of the right ventricle and pulmonary artery, radioiodinated (131)I serum albumin and indocyanine green dye were injected simultaneously in 16 subjects. One indicator was injected into the atrium and the other into the ventricle, or both were injected at different sites in the ventricle. Washout curves were obtained by rapid catheter sampling alternately just above or just below the pulmonic valve. The washout of radioisotope was also recorded with a precordial scintillation detection probe. Indicator washout from the ventricular inflow tract was rapid, while washout from the region of the ventricular apex was quite slow. Protosystolic dips in indicator concentration, noted in curves drawn below the pulmonic valve, suggest that the ventricle emptied sequentially. Flow values computed from curves sampled below the valve, when compared with reference values, suggest that a significant volume of atrial blood passed through the ventricle without mixing, or mixing to only a small extent, with the residual volume of the chamber. Peak concentration of indicator was higher below the pulmonic valve than above. This finding, plus the close agreement between flow values computed from curves sampled above the valve and the reference values, indicates that further mixing occurred above the valve. Ventricular volumes computed from washout downslopes are systematically falsely high. This overestimate appeared to be larger in normal subjects than in patients with low stroke volumes. Progressive mixing of blood leaving the atrium occurs during its passage through the ventricle, pulmonic valve, and pulmonary artery and permits accurate estimation of flow.

Abstract:This work sets out to investigate biological responses of Eisenia fetida to Cd, based on the bioavailable rather than total concentration of Cd, in soils. E. fetida was cultured for 14d in three selected Chinese soils amended with 0.1-40mgkg-1 Cd. Potentially bioavailable concentrations of Cd were measured in soil solution, in extractions using CaCl2 and HAc solutions, and using the technique of diffusive gradients in thin-films (DGT). Antioxidant system responses in E. fetida to Cd were measured as biological endpoints. Biological responses were more highly correlated with Cd concentrations evaluated using bioavailable methods than with total concentrations. Cd concentration measured using DGT and CaCl2 extraction provided the narrowest ranges of lowest observed effect concentration (LOEC) values based on biological responses, indicating the potential use of these measurements in management of soil quality and setting soil remediation standards. The LOEC values obtained from 15 field soils contaminated by Cd were similar to those from the three Cd-amended soils and suggested that DGT in particular can be a good tool to predict stress responses of E. fetida to Cd in soils. The study shows the potential of combining biological response and DGT measurements in risk assessment.

Abstract:The genotoxicity of single and combined soil pollution by mercury (Hg) and bromoxynil (BX, 3, 5-dibromo-4-hydroxybenzonitrile) on earthworm (Eisenia fetida) were investigated by the single cell microgel electrophoresis (SCGE) assay. Tail length (TL), Olive tail moment (OTM) and Tail DNA% (TD) were used to evaluate DNA damage of the coelomocytes on earthworms after 14-d exposure. The results showed that a dose-response relationship was established between the concentration of single pollutant and DNA damage. There is a negative effect of pollutant on earthworms. The results of DNA damage co-exposure to the mixture was observed to be different compared to the exposure to a single pollutant. DNA damage increased with the increasing concentration of BX co-exposure to Hg (p<0.05, p<0.01). Furthermore, the results revealed that BX was the contribution of main factor of DNA damage in the combined experiments. It can be proved that Eisenia fetida is a suitable bio-indicator to measure the acute toxicity of mixed pollutants in soil and the OTM may be considered as a valuable and sensitive parameter to diagnose adverse effects caused by Hg or BX in soil environment.

Abstract:Although strong in vivo-in vitro correlations (IVIVCs) between relative bioavailability (RBA) and bioaccessibility of soil Pb were well reported, knowledge on the fractions of bioaccessible Pb in simulated gastrointestinal (GI) fluids that are available for absorption into the systemic circulation is limited. Here, Pb-RBA in 14 Pb-contaminated soils were assessed using an in vivo mouse bioassay and compared to Pb bioaccessibility by the gastrointestinal phase of the UBM (Unified Bioaccessibility research group of Europe (BARGE) Method) in vitro assay with and without 0.45 μm filtration of GI fluid. Results showed good IVIVC between Pb-RBA and Pb bioaccessibility without filtration ( r 2 = 0.62), while Pb bioaccessibility with filtration provided a poor correlation with Pb-RBA ( r 2 = 0.16). This suggested that besides dissolved Pb ions, Pb-complexes formed in the UBM gastrointestinal fluid might also contribute to bioavailable Pb. To ascertain this, DGT (diffusive gradients in thin-films) devices which can measure both Pb2+ ions and labile inorganic and organic Pb-complexes were introduced to the UBM fluids to measure Pb DGT-bioaccessibility, which showed strong correlation to Pb-RBA ( r 2 = 0.71). With increasing diffusive gel thickness which could enhance release of Pb ions from Pb-complexes, Pb DGT-bioaccessibility increased by 3.4-5.7 times, while inclusion of dialysis membrane within DGT devices significantly decreased Pb DGT-bioaccessibility by inhibiting diffusion of Pb complexes to binding gel. These results confirmed the contribution of Pb-complexes to Pb bioavailability, providing new insights to Pb bioavailability.
Hormetic Effects of Mercury on Survival of Eisenia Fetida (Oligochaeta)

Abstract:The article describes a laboratory experiment to determine the hormetic effects of inorganic mercury (Hg) on the earthworm Eisenia fetida (E. fetida) in OECD soils. In an experiment lasting 17d, survival rates of exposure to mercury chloride were investigated. 7d and 14d LC50 of mortality were 155.97 and 143.67mg/kg, respectively. There are a significant dose-response relationship between survival rates and Hg concentration (0, 50, 100,150, 200 and 250 mg/kg). Survival of E. fetida is a stable and sensitive biomarker to diagnose adverse effects of Hg in soil environment. Sub-chronic treatment (0.78, 1.56, 3.125, 6.25, 12.5, 25, 50, 100 mg/kg) caused a significant increase at low doses yet an inhibitory at high levels in survival of earthworms over 7 d. The OECD soil exposure of the mature E. fetida showed a hormetic stimulation of 114-132% of survival rates. It indicated that inorganic Hg is capable of causing hormetic-like biphasic effects on earthworms and high dose of pollutants may conceal the hormesis of exposure to Hg.

Abstract:The purpose of this review was to survey the recent applications of the diffusive gradients in thin films (DGT) technique in the assessment of mobility and bioavailability of nutrients and potentially toxic elements (PTEs) in agricultural soil. Many studies compared the capabilities of the DGT technique with those of classical soil chemical extractants used in single or sequential procedures to predict nutrients and PTE bioavailability to crops. In most of the published works, the DGT technique was reported to be superior to the conventional chemical extraction and fractionation methods in obtaining significant correlations with the metals and metalloids accumulated in crops. In the domain of nutrient bioavailability assessment, DGT-based studies focused mainly on phosphorous and selenium labile fraction measurement, but potassium, manganese, and nitrogen were also studied using the DGT tool. Different DGT configurations are reported, using binding and diffusive layers specific for certain analytes (Hg, P, and Se) or gels with wider applicability, such as Chelex-based binding gels for metal cations and ferrihydrite-based hydrogels for oxyanions. Overall, the literature demonstrates that the DGT technique is relevant for the evaluation of metal and nutrient bioavailability to crops, due to its capacity to mimic the plant root uptake process, which justifies future improvement efforts.

Abstract:Identifying the bioavailability and release-desorption mechanism of heavy metals (HMs) in soil is critical to understand the release risk of HMs. Simultaneously, the mechanistic investigation of affecting the bioavailability of HMs in soil is necessary, such as the grain-size distribution and soil mineralogy. Herein, the bioavailability of HMs (Cu, Cd, Ni, Pb, and Zn) in different area soils near a typical copper-smelter was evaluated by the sequential extraction technique (BCR), diffusive gradients in thin-films (DGT), and DGT-induced fluxes in sediments (DIFS) model. Results showed that the HMs proportion of the residual fraction in all soils was the highest. The average bioavailability concentration (CDGT) of Cu and Cd in industrial soil was the highest, with 45.12 μg· L−1 and 9.06 μg· L−1. The result of DIFS model revealed that the decreased order of the mean value of desorption rate constant (K-1) was Cd > Zn > Ni > Cu > Pb, 5.91 × 10−5, 4.96 × 10−5, 2.89 × 10−5, 9.64 × 10−6, and 8.69 × 10−6, respectively. According to the spatial distribution of release potential (R-value), the release potential of labile-Cu in agricultural soil was the highest, which was mainly attributed to fertilizer application in farmland. Simultaneously, the reduced hydroxyl was also related to the agricultural activities, resulting in the weakened adsorption capacity of HMs by soil. Redundancy analysis (RDA) results showed that the bioavailability of Cd, Ni, and Zn was mainly driven by soil pH, while the bioavailability of Cu and Pb was primarily driven by dissolved organic carbon (DOC). Meanwhile, carbonate minerals had a positive correlation with the bioavailability of Cd, Ni, and Zn, which could promote the release of HMs in mining soil as chemical weathering progresses. In conclusion, this study provides a structured method which can be used as a standard approach for similar scenarios to determine the geochemical fractionation, bioavailability, and release kinetics of heavy metals in soils.
