Identification of proteomic signatures of exposure to marine pollutants in mussels (Mytilus edulis)

Abstract:Bivalves and especially mussels are very good indicators of marine and estuarine pollution, and so they have been widely used in biomonitoring programs all around the world. However, traditional single parameter biomarkers face the problem of high sensitivity to biotic and abiotic factors. In our study, digestive gland peroxisome-enriched fractions of Mytilus edulis (L., 1758) were analyzed by DIGE and MS. We identified several proteomic signatures associated with the exposure to several marine pollutants (diallyl phthalate, PBDE-47, and bisphenol-A). Animals collected from North Atlantic Sea were exposed to the contaminants independently under controlled laboratory conditions. One hundred and eleven spots showed a significant increase or decrease in protein abundance in the two-dimensional electrophoresis maps from the groups exposed to pollutants. We obtained a unique protein expression signature of exposure to each of those chemical compounds. Moreover a set of proteins composed a proteomic signature in common to the three independent exposures. It is remarkable that the principal component analysis of these spots showed a discernible separation between groups, and so did the hierarchical clustering into four classes. The 14 proteins identified by MS participate in alpha- and beta-oxidation pathways, xenobiotic and amino acid metabolism, cell signaling, oxyradical metabolism, peroxisomal assembly, respiration, and the cytoskeleton. Our results suggest that proteomic signatures could become a valuable tool to monitor the presence of pollutants in field experiments where a mixture of pollutants is often present. Further studies on the identified proteins could provide crucial information to understand possible mechanisms of toxicity of single xenobiotics or mixtures of them in marine ecosystems.
Fast analysis of relevant contaminants mixture in commercial shellfish

Abstract:One of the major challenges currently faced is to develop systematic ways of addressing chemical mixtures in environmental assessment. With this purpose, a simple, rapid, and sensitive method for the detection and quantification of a mixture of relevant contaminants in molluscs has been developed. The method is based on QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) and Ultra-High Performance Liquid Chromatography-High Resolution Mass Spectrometry (UHPLC-HRMS). It includes a mixture of 23 compounds formed by pesticides, endocrine disruptors and pharmaceuticals (metolachlor, simazine, desethylatrazine, atrazine, thiabendazole, diazinon, malathion, bentazone, MCPA, propanil, acetamiprid, imidacloprid, caffeine, bisphenol A, triclosan, triclocarban, methylparaben, ethylparaben, propylparaben, 1H-benzotriazole, sulfamethoxazole, venlafaxine and carbamazepine). The method was developed and validated in 4 different types of shellfish of high commercial interest such as mussel (Mytilus galloprovincialis), oyster (Crassostrea gigas), cockle (Cerastoderma edule) and razor shell (Solen marginatus). The mean percentage of recoveries obtained for all the compounds in each mollusc type (intra-specie) ranged from 96% to 107% showing the good performance of the method developed. The relative standard deviation was under 10% for the intra-day and 17% inter-day analyses. Method detection limits and method quantification limits were below 10 ng/g dry weight for all the species and compounds targeted. Finally, the method was applied to aquaculture samples, oysters and cockles, from Ebro Delta (Spain), after some episodes of mortality occurred in 2017. A high level of bisphenol A was detected in C. edule which may explain the mortality suffered by this organism. C. gigas presented low levels of metolachlor, bentazone, acetamiprid, and methylparaben.
Fast Determination of Multiple Antibiotics in Mussels Based on Extraction and UPLC-MS/MS

Abstract:A fast and simple method for simultaneous determination of fifteen antibiotics (sulfadiazine, ciprofloxacin, penicillin, spechromycin and roxithromycin) in mussels using ultrasound extraction, solid phase extraction purification and ultra performance liquid chromatography-tandem mass spectrometric was established. The mussel samples were extracted by ultrasonic extraction, followed by solid-phase extraction (SPE) as the cleanup procedure. Ultra performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-MS/MS) was used for identification and quantification of antibiotics. The performance of two kinds of SPE cartridges, Oasis HLB and Oasis PRiME HLB, was compared, with the latter showing better results. Recoveries ranged from 64% to 121% at spiking levels of 50 ng/g, with RSD being 0.5%-19%, and from 67% to 117% at spiking levels of 100 ng/g, with RSD being 1%-9%. Limits of detection (LODs) ranged from 0.004 to 0.5 ng/g (dry weight) and limits of quantification (LOQs) ranged from 0.013 to 1.67 ng/g (dry weight). The recoveries were reasonable and the detection limit was low. Finally, the method was successfully applied to the determination of the target antibiotics in mussel samples (Hyriopsis cumingii) collected from Poyang Lake. 9 out of 15 selected antibiotics were detected in the collected mussel tissues. The concentration and detection frequency of trimethoprim in mussels were the highest. The concentration of trimethoprim was 78.8 ng/g, followed by spectinmycin (41.2 ng/g) and ciprofloxacin (39.8 ng/g). © 2018 Peking University.
Multiresidue analytical method for the determination of 41 multiclass organic pollutants in mussel and fish tissues and biofluids by liquid chromatography coupled to tandem mass spectrometry

Abstract:In this work, the full optimisation and validation procedure to analyse a wide set of emerging organic contaminants in biotissues (mussel and fish muscle, liver, gills and brain) and biofluids (fish plasma and bile) is described. The target families include artificial sweeteners, industrial products, hormones, pharmaceutical and personal care products, pesticides and phytoestrogens. Different clean-up strategies (hydrophilic–lipophilic-balanced (HLB) solid-phase extraction, Florisil solid-phase extraction and liquid–liquid extraction followed by HLB solid-phase extraction and microextraction based on polyethersulfone polymer) were evaluated for the clean-up of focused ultrasonic solid–liquid extraction (FUSLE) extracts before the analysis by liquid chromatography–triple quadrupole tandem mass spectrometry (LC-QqQ-MS/MS). The methods afforded satisfactory apparent recovery values (71–126%) using isotopically labelled analytes and matrix-matched calibration approach, regardless of the matrix. Method detection limits in the range of 4–48 ng/g and 0.3–111 ng/L were obtained for biotissues and biofluids, respectively. The developed method was applied to determine the uptake and tissue distribution in juvenile gilt-head bream (Sparus aurata) during 7 days in seawater, and unexpectedly, perfluoro-1-butanesulfonate tended to accumulate in liver and, to a lesser extent, in muscle and gills. Furthermore, real mussel samples collected in the Basque coast were also analysed and the presence of the highly consumed valsartan (7 ng/g) and telmisartan (6.8 ng/g) compounds in bivalves is reported for the first time here.Graphical abstractᅟ
Simultaneous determination of PCBs, OCPs and PAHs in mussel by ultrasound-assisted cloudy extraction and gas chromatography–tandem mass spectrometry

Abstract:A method for the simultaneous determination in mussel of 8 polychlorinated biphenyls (PCBs), 23 organochlorine pesticides (OCPs), and 35 polycyclic aromatic hydrocarbons (PAHs), including alkyl-PAHs, was optimised using ultrasound-assisted cloudy extraction and gas chromatography–tandem mass spectrometry (GC-MS/MS). Optimal selection of the extraction solvent and the dispersing solvent contributed to a high extraction yield and a clean extract. The ranges of the lower limits of detection of PCBs, OCPs and PAHs were 0.012–0.058, 0.01–0.29 and 0.01–0.5 μg kg−1, respectively. The feasibility of the proposed method was validated by analysing standard reference materials of mussel with satisfactory results. The precision achieved by this method was in the range of 0.677–2.69% (PCBs), 1.14–6.60% (OCPs) and 0.694–7.46% (PAHs), and its accuracy was in the range of 101–104% (PCBs), 99.6–106% (OCPs) and 101–110% (PAHs). The advantages of the method include the simultaneous measurement of 66 analytes and the simplicity, low cost and high sensitivity of the procedure. When the proposed method was used to analyse the target compounds in 11 mussel samples, the analytical results displayed a concentration range of 0.41–0.45 µg kg−1 for PCBs, 0.26–6.49 µg kg−1 for OCPs, and 3.48–30.69 µg kg−1 for PAHs.
Simultaneous determination of triazine herbicides and their metabolites in shellfish by HPLC-MS/MS combined with Q/E-Orbitrap HRMS

Abstract:Triazine herbicides are used extensively in agriculture and aquaculture worldwide because of their broad effectiveness in weed control. However, after they are discharged into the sea, they seriously contaminate aquatic ecosystems and threaten aquatic organisms, especially shellfish. Currently, there are no established methods for the detection and confirmation of triazine herbicides and their metabolites in biological matrixes. Hence, the food safety of aquatic products cannot be accurately evaluated, which creates a technical barrier against international aquatic product trade. In this study, for the first time, a method was developed for the analysis and confirmation of seven triazine herbicides and 13 metabolites in shellfish, based on alkaline acetonitrile extraction and neutral Al2O3 cartridge purification coupled with internal standard calibration. Specifically, quantitative and qualitative analysis was conducted using high-performance liquid chromatography-triple quadrupole mass spectrometry (HPLC-MS/MS), and accurate identification was carried out by quadrupole orbitrap high-resolution mass spectrometry (Q/E Orbitrap HRMS). The results showed that target analytes demonstrated good linearity within the corresponding range (R2 > 0.995). The limit of detection and limit of quantitation of the proposed method were 0.1 and 0.3 μg/kg, respectively. The average recoveries of analytes were between 70.0% and 120% when spiked at three levels with blank oyster (Crassostrea gigas) as the matrix, and the relative standard deviations (RSDs) were all less than 12% (n=6). The proposed method was successfully applied for the detection of triazine herbicide residues in oyster samples during actual breeding, and the presence of DIP, HP, DEHA, and other metabolites in positive samples was confirmed by Q/E Orbitrap HRMS. This method exhibits high accuracy, high sensitivity, and good reproducibility. It has promising application prospects in the field of hazard analysis and the positive identification of aquatic products.

Abstract:Since the last decades, light has been shed on the pollution of aquatic ecosystems. Considering apolar compounds in water, analysis of the dissolved phase is not sufficient due to their possible bioaccumulation. Additional analysis of sediments, biota, or sentinel species is necessary. Among sentinel species, Gammarus fossarum is a small shrimp of 30 mg that lives naturally in the river. Its ability to bioaccumulate makes it a good bioindicator of river pollution. Nevertheless, micropollutants are difficult to extract from gammarids due to their small size and their high level of lipids. Extracted interferences can lead to analytical difficulties. Targeted micropollutants in this work were organotins, personal and pharmaceutical care products (PPCPs), pesticides, flame retardants, and perfluorinated compounds. A sample preparation based on QuEChERS followed by liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) was developed: different salts (acetate and citrate) and purification (addition of hexane, dispersive SPE) were tested and validated. Targeted molecules used to be analyzed by LC-MS/MS, except organotins which are principally analyzed in gas chromatography. One of the main challenges was to quantify them also in LC-MS/MS to implement an original multi-residue method. The analysis of 40 compounds was finally validated according to ICH guidelines, with LOQs ranging from 0.04 ng.g-1 to 313.5 ng.g-1. Regarding Perfluorooctanesulfonic acid (PFOS) and the sum of hexabromocyclododecane, LOQs reached the environmental quality standards in biota which are respectively of 9.1 and 167 ng.g-1. Finally, the method was applied to 15 real samples. Many compounds were quantified: perfluorinated compounds, drugs such as ketoprofen, and even cosmetics products such as octocrylene. This is the first study to quantify as many emerging compounds, especially organotins, in a sentinel species such as G. fossarum.

Abstract:Per- and polyfluoroalkyl substances (PFAS) are known to bioaccumulate in aquatic organisms, such as shellfish, and have been linked to adverse human health outcomes. Increasing attention has been focused on method development for the detection of PFAS in various media; however, these methods are typically tedious, require high solvent volumes, and are time consuming. The present method used a Quick, Easy, Cheap, Effective, Rugged, and Safe (QuEChERS) extraction approach and analysis by ultra-performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS) to detect 28 PFAS in Eastern oyster ( Crassostrea virginica ) tissue. This method was validated using limit of detection, limit of quantitation, and precision and accuracy studies. Limits of detection ranged from 0.2 to 5.8 ng g −1 , limits of quantitation ranged from 0.66 to 19.2 ng g −1 , and recoveries spanned 52.1–105.9% at the 100 ng mL −1 analyte level. Analysis of 12 unknown oyster composite samples revealed the detection of PFHxA, PFPeS, PFOA, PFHpA and PFOS and at least two compounds were detected in each oyster sample. This validated method proved to be an efficient and environmentally friendly sample preparation method for PFAS analysis in complex tissue media, such as oysters.

Abstract:The extant methods for detecting multi-residues in aquatic products can avoid the disadvantages of complex pretreatment, long time consumption, single detection type, and low sensitivity. Therefore, developing a method that enables a multi-residue detection of veterinary drugs is very important. QuEChERS is a simple, rapid, and efficient method for detecting multi-drug residues. In this work, QuEChERS pretreatment combined with ultra-performance liquid chromatography–tandem mass spectrometry was proposed to identify sulfamide, tetracycline, quinolone, and malachite green residues in oyster, scallop, squid, and cuttlefish. The sample was extracted by using acetonitrile solution containing 0.1% acetic acid, while the extract was purified by using the QuEChERS method. The target analytes were separated on an ACQUITY UPLC BEH chromatographic column by using acetonitrile solution as a mobile phase for the gradient elution. Quantification was performed by using the internal standard method. The recovery of the 21 tested compounds was within the range of 65.26 to 113.5% and the relative standard deviation was less than 6.78%, thereby proving the good repeatability of the method. The intra- and inter-day precisions were less than 6.26% and 6.02%, respectively, while the limits of detection and quantification were less than 0.8 μg/kg and 2.5 μg/kg, respectively. The proposed method was then applied to detect residues in 150 samples of oysters, scallops, squid, and cuttlefish obtained from eight regions, including Shenzhen, Dongguan, Zhaoqing, Shantou, Maoming, Jiangmen, Shanwei, and Huizhou in Guangdong Province. No residues were detected in any of these samples. In sum, the proposed detection method has good reproducibility, high sensitivity, and reliability.

Abstract:The amount of macrolide (MAL) residues in aquatic products, including oleandomycin (OLD), erythromycin (ERM), clarithromycin (CLA), azithromycin (AZI), kitasamycin (KIT), josamycin (JOS), spiramycin (SPI), tilmicosin (TIL), tylosin (TYL), and roxithromycin (ROX), was determined using solid-phase extraction and ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS). The residues were extracted with 1% ammonia acetonitrile solution and purified by neutral alumina adsorption. Chromatographic separation was completed on an ACQUITY UPLC BEH C18 column with acetonitrile–0.1% formic acid aqueous solution as the mobile phase, and mass spectrometry detection was performed by multiple reaction monitoring scanning with the positive mode in an electrospray ion source (ESI+). Five isotopically labeled compounds were used as internal standards for quality control purposes. The findings indicated that across the mass concentration span of 1.0–100 μg/L, there was a strong linear correlation (R2 > 0.99) between the concentration and instrumental response for the 10 MALs. The limit of detection of UPLC-MS/MS was 0.25–0.50 μg/kg, and the limit of quantitation was 0.5–1.0 μg/kg. The added recovery of blank matrix samples at standard gradient levels (1.0, 5.0, and 50.0 μg/kg) was 83.1–116.6%, and the intra-day precision and inter-day precisions were 3.7 and 13.8%, respectively. The method is simple and fast, with high accuracy and good repeatability, in line with the requirements for accurate qualitative and quantitative analysis of the residues for 10 MALs in aquatic products.

Development and Validation of a Hybrid Screening and Quantitative Method for the Analysis of Eight Classes of Therapeutants in Aquaculture Products by Liquid Chromatography-Tandem Mass Spectrometry

Abstract:A method using reverse-phase ultra-high-performance liquid chromatography coupled with tandem mass spectrometry is described for eight classes of therapeutants that are used in marine aquaculture products. Validation studies to evaluate recovery, precision, method detection limits, and measurement uncertainty were performed at three levels, using three representative matrices [salmon (fatty fish), tilapia (lean fish), and shrimp (crustaceans)] to assess the method performance for use as a screening or determinative (quantitative and confirmatory) method. A total of 16 sulfonamides (plus 2 potentiators), 2 tetracyclines, 11 (fluoro)quinolones, 7 nitroimidazoles, 3 amphenicols, 5 steroids, and 3 stilbenes met the quantitative criteria for method validation. An additional 5 triphenylmethane dyes, 2 sulfonamides, 2 tetracyclines, and 1 amphenicol met the required performance for use as a screening method. Limits of detection (LODs) for the compounds that met the quantitative criteria ranged from 0.1 to 5 μg/kg, while LODs for compounds from the screening group ranged from 0.1 to 30 μg/kg. This method provides a comprehensive approach to the determination of different classes of compounds in aquaculture products.
Superior and Novel Detection of Small, Polar Metabolites in Saline Samples Using Pre-Extraction Benzoyl Chloride Derivatization and Ultra-High Performance Liquid Chromatography Tandem Mass Spectrometry (UHPLC MS/MS).

Abstract:Dissolved metabolites support microbial systems via nutritional and energetic utilization and chemical signaling. However, the full scope and magnitude of these processes in marine systems is unknown largely because the mass spectrometric identification and quantification of these compounds from saline matrices has been hampered by poor extraction of small, polar compounds using common solid phase extraction resins (e.g. PPL resin, Bond Elut, Agilent). Here we utilized pre-extraction derivatization to improve the extraction, quantification, and chromatographic resolution of targeted dissolved metabolites in seawater and saline culture medium detected with ultra-high performance liquid chromatography electrospray ionization tandem mass spectrometry (UHPLC-ESI-MSMS) with Orbitrap detection. Metabolites were derivatized with benzoyl chloride by their primary and secondary amine and alcohol functionalities and quantified using stable isotope-labeled internal standards (SIL-IS) produced from 13C-labeled benzoyl chloride. We optimized derivatization, extraction, and sample preparation for field and culture samples, and we evaluated matrix-derived biases. This method quantifies 73 metabolites in seawater, of which 50 have an extraction efficiency <2% without derivatization. This method is sensitive (detection limits = pM to nM) and substantially improves sample throughput by shortening processing time ~10-20x
Validation of a confirmatory method for lipophilic marine toxins in shellfish using UHPLC-HR-Orbitrap MS

Abstract:Lipophilic marine toxins are produced by harmful microalgae and can accumulate in edible filter feeders such as shellfish, leading to an introduction of toxins into the human food chain, causing different poisoning effects. During the last years, analytical methods, based on liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS), have been consolidated by interlaboratory validations. However, the main drawback of LC-MS/MS methods remains the limited number of compounds that can be analyzed in a single run. Due to the targeted nature of these methods, only known toxins, previously considered during method optimization, will be detected. Therefore in this study, a method based on ultra-high-performance liquid chromatography coupled to high-resolution Orbitrap mass spectrometry (UHPLC-HR-Orbitrap MS) was developed. Its quantitative performance was evaluated for confirmatory analysis of regulated lipophilic marine toxins in shellfish flesh according to Commission Decision 2002/657/EC. Okadaic acid (OA), dinophysistoxin-1 (DTX-1), pectenotoxin-2 (PTX-2), azaspiracid-1 (AZA-1), yessotoxin (YTX), and 13-desmethyl spirolide C (SPX-1) were quantified using matrix-matched calibration curves (MMS). For all compounds, the reproducibility ranged from 2.9 to 4.9 %, repeatability from 2.9 to 4.9 %, and recoveries from 82.9 to 113 % at the three different spiked levels. In addition, confirmatory identification of the compounds was effectively performed by the presence of a second diagnostic ion ((13)C). In conclusion, UHPLC-HR-Orbitrap MS permitted more accurate and faster detection of the target toxins than previously described LC-MS/MS methods. Furthermore, HRMS allows to retrospectively screen for many analogues and metabolites using its full-scan capabilities but also untargeted screening through the use of metabolomics software.

Abstract:An accurate and efficient method was developed for the determination of azaspiracid shellfish toxins (azaspiracids-1, -2, and -3), neurotoxic shellfish toxins (brevetoxins-2 and -3), diarrhetic shellfish toxins (okadaic acid and dinophysistoxins-1 and -2), and the amnesic shellfish toxin (domoic acid) in mussels ( Mytilus galloprovincialis ). Lipophilic marine biotoxins (azaspiracids, brevetoxins, and okadaic acid group) were extracted with 0.5% acetic acid in methanol under heating at 60°C to improve the extraction efficiency of okadaic acid group toxins and then cleaned up with a C18 solid-phase extraction cartridge. Domoic acid was extracted with 50% aqueous methanol and then cleaned up with a graphitized carbon solid-phase extraction cartridge. Lipophilic marine biotoxins and domoic acid were quantified by reversed-phase liquid chromatography coupled to electrospray ionization tandem mass spectrometry. The developed method had insignificant matrix effects for the nine analytes and good recoveries in the range of 79.0% to 97.6% at three spiking levels for all analytes except brevetoxin-2 (43.8–49.8%). The developed method was further validated by analyzing mussel tissue certified reference materials, and good agreement was observed between certified and determined values.

Abstract:The presence of pharmaceuticals and personal care products (PPCPs) in aquatic systems has raised concern about their potential adverse effects on aquatic organisms. Considering the fact that the physiological/biological effects of PPCPs are triggered when their concentrations in the organism exceeds the respective threshold values, it is important to understand the bioconcentration and toxicokinetics of PPCPs in aquatic organisms. In the present study, we developed a convenient analytical method for the determination of 65 pharmaceuticals and 7 personal care products (log Kow = 0.14-6.04) in plasma and whole-body tissues of fish. The analytical method consists of ultrasound-assisted extraction in methanol/acetonitrile (1:1, v/v,) acidified with acetic acid-ammonium acetate buffer (pH 4), cleanup on a HybridSPE®-Phospholipid cartridge (zirconia-coated silica cartridge), and quantification with liquid chromatography-tandem mass spectrometry (LC-MS/MS). Acceptable accuracy (internal standard-corrected recovery: 70%-120%) and intra- and inter-day precision (coefficient of variation: <15%) were obtained for both plasma and whole-body tissue samples. In addition, low method detection limits were achieved for both plasma (0.0077 to 0.93 ng mL-1) and whole-body tissue (0.022 to 4.3 ng g - 1 wet weight), although the developed method is simple and fast - a batch of 24 samples can be prepared within 6 h, excluding the time for measurement with LC-MS/MS. The developed method was successfully applied to the analysis of PPCPs in plasma and whole-body tissue samples of fish collected in a treated wastewater-dominated stream, for a comprehensive evaluation of their bioconcentration properties. The analytical method developed in the present study is sufficiently accurate, sensitive, and rapid, and thus highly useful for the comprehensive evaluation of PPCP residues in fish and would aid in future exposome and risk assessment.

Abstract:BACKGROUND Lipopolysaccharide (LPS) stimulates osteoclast differentiation through toll-like receptors (TLRs) 2 and 4, and hydrogen sulfide (H(2)S) induces osteoclast differentiation. If H(2)S activates TLRs, H(2)S may enhance the effects of LPS on osteoclast differentiation. The purpose of the present study is to examine the combined effects of sodium hydrogen sulfide (NaHS, an H(2)S donor drug) and LPS on osteoclast differentiation and TLR expression in rat periodontal tissue. METHODS Twenty-eight male Wistar rats (8 weeks old) were divided into four groups (n = 7 per group): a control (no treatment) group and three experimental groups (NaHS group, LPS group, and a combination [NaHS + LPS] group). At 1 day after topical application of NaHS and/or Porphyromonas gingivalis LPS into the gingival sulcus of first molars, the number of tartrate-resistant acid phosphate (TRAP)-positive osteoclasts in the periodontal tissue was counted. Expression of TLR2 and TLR4 mRNAs and proteins in the gingival was also assessed. RESULTS The number of TRAP-positive osteoclasts was significantly higher in the combination group than in any other group (P <0.01). The combination group had 11.0-fold higher TLR4 mRNA levels than the control group. TLR4 protein levels were also higher in the combination group than in the NaHS or LPS group. However, the TLR2 mRNA and protein levels were not significantly different in the combination group and the LPS group. CONCLUSION In rat periodontal tissue, NaHS and LPS had an additive effect on osteoclast differentiation through activation of the TLR4 pathway but not the TLR2 pathway.
Automated on-line solid-phase extraction coupled to liquid chromatography–tandem mass spectrometry for determination of lipophilic marine toxins in shellfish

Abstract:Automated on-line solid-phase extraction (SPE) coupled to liquid chromatography-tandem mass spectrometry (LC-MS/MS) has been developed for fast determination of lipophilic marine toxins in shellfish samples. The direct coupling of an on-line SPE column to LC-MS/MS was accomplished using column switching techniques. Suitable chromatographic separation was performed on a reversed-phase C18 column under alkaline conditions (pH 11). The selected reversed-phase C18 SPE column allowed rapid and efficient on-line desalting of hydrolysed shellfish samples, avoiding signal suppression during mass spectrometry detection. Furthermore, the on-line SPE procedure allowed reducing matrix effects observed in raw and hydrolysed shellfish extracts. The proposed method was evaluated in terms of linearity, precision, accuracy and limits of detection (LODs). Quantitative recovery (97-102%) and satisfactory inter-day precision (RSD<8%) were achieved for all target compounds. LODs in the sub-μgkg-1 level (0.37-0.68μgkg-1) were obtained for all toxins except for okadaic acid, which showed a value of 2.75μgkg-1. Several mussel samples from North-western Spain were finally analysed in order to demonstrate the applicability of the method. Okadaic acid was the predominant toxin in all samples, although other lipophilic toxins were also detected.

Abstract:This work describes a reliable, cheap, easy and fast method for analysis of nine bisphenols in mussel samples by gas chromatography-mass spectrometry after trimethylsilylation. The modified method consisted of miniaturized matrix solid phase dispersion (micro-MSPD) in a glass Pasteur pipette using Captiva EMR (enhanced matrix removal)-lipid as the sorbent. Good linearity was obtained in the work range (1–500 μg L −1 ) with a correlation coefficient ( R 2 ) ≥ 0.998. The method accuracy and precision were determined at two concentration levels. The results show recoveries ranging from 55 to 111%. The precision varied from 1.95 to 11.4% (RSD). The whole quantification limits were between 0.056 and 3.42 μg per kg dry weight. The analytical procedure was applied for the analyses of five mussel samples collected from Galician Rias. The major compound was BPA, and wild mussels from Rías de Ferrol, Vigo and A Coruña had the highest levels. The proposed method is suitable for the analysis of BPA and its analogues in mussel samples.
