Abstract:Coevolution between plants and insects is thought to be responsible for generating biodiversity. Extensive research has focused largely on antagonistic herbivorous relationships, but mutualistic pollination systems also likely contribute to diversification. Here we describe an example of chemically-mediated mutualistic species interactions affecting trait evolution and lineage diversification. We show that volatile compounds produced by closely related species of Zamia cycads are more strikingly different from each other than are other phenotypic characters, and that two distantly related pollinating weevil species have specialized responses only to volatiles from their specific host Zamia species. Plant transcriptomes show that approximately a fifth of genes related to volatile production are evolving under positive selection, but we find no differences in the relative proportion of genes under positive selection in different categories. The importance of phenotypic divergence coupled with chemical communication for the maintenance of this obligate mutualism highlights chemical signaling as a key mechanism of coevolution between cycads and their weevil pollinators.

Tissue-Specific Emission of (E)-Α-bergamotene Helps Resolve the Dilemma when Pollinators Are Also Herbivores

Abstract:More than 87% of flowering plant species are animal-pollinated [1] and produce floral scents and other signals to attract pollinators. These floral cues may however also attract antagonistic visitors, including herbivores [2]. The dilemma is exacerbated when adult insects pollinate the same plant that their larvae consume. It remains largely unclear how plants maximize their fitness under these circumstances. Here we show that in the night-flowering wild tobacco Nicotiana attenuata, the emission of a sesquiterpene, (E)-α-bergamotene, in flowers increases adult Manduca sexta moth-mediated pollination success, while the same compound in leaves is known to mediate indirect defense against M. sexta larvae [3, 4]. Forward and reverse genetic analyses demonstrated that both herbivory-induced and floral (E)-α-bergamotene are regulated by the expression of a monoterpene-synthase-derived sesquiterpene synthase (NaTPS38). The expression pattern of NaTPS38 also accounts for variation in (E)-α-bergamotene emission among natural accessions. These results highlight that differential expression of a single gene that results in tissue-specific emission of one compound contributes to resolving the dilemma for plants when their pollinators are also herbivores. Furthermore, this study provides genetic evidence that pollinators and herbivores interactively shape the evolution of floral signals and plant defense.
Volatile profiling of cacti: a preliminary assessment of the taxonomic and evolutionary significance of volatile compounds in Cylindropuntia, Grusonia, Consolea, Opuntia, Quiabentia, and Tacinga

Abstract:Volatile analyses of cacti have previously been performed on the flowers, fruits, and consumed stems. During our own investigations, we and others have observed that the cut stems of certain species of the Graveolens clade of Cylindropuntia emitted odors similar to those of rancid butter or cyanoacrylate. Some species of Consolea, Opuntia, Quiabentia, and Tacinga were found to produce similar odors. Fresh samples of Cylindropuntia and these other genera were collected, and the volatile compound profiles were analyzed by solid phase micro-extraction gas chromatography mass spectrometry. Linear discriminate analysis found the compounds to be characteristic of the odiferous cacti as the aldehydes hexanal, 2-hexenal, and nonanal; the alcohol phenethyl alcohol; the terpene β-phellandrene; the ketone β-ionone; and the diol 5-pentyl-1,3-benzenediol. Compounds characteristic of the non-odiferous cacti are the ketones 6-methyl-2-heptanone, 2-octanone, and 1,3-dihydro-5-methyl-2H-benzimidazol-2-one; the alkanes undecane, tridecane, pentadecane, and heptadecane; the aromatics p-cymene and 1,2,3,5-tetramethyl benzene; the esters octyl formate, methyl benzoate, and methyl salicylate; the aldehyde 2-octenal; the alcohol cyclooctyl alcohol; the imine methoxy-phenyl-oxime; the terpene 1-methyl-2-(2-propenyl)-benzene; and nine unknown compounds. Putative hybrid cacti were found to have a unique volatile profile in comparison to the parents. Additionally, differing infraspecific chromosome races, diploids (n = 11) and tetraploids (n = 22), were found to have differing volatile profiles with some compounds increasing with an increase in chromosome number while other compounds decreased with an increase in chromosome number.

Dynamic regulation of volatile terpenoid production and emission from Chrysanthemum morifolium capitula

Abstract:Flower-associated communities consist of both mutualistic and antagonistic organisms. We have limited knowledge on how flowers regulate volatiles to balance their defense against antagonists and the attraction of beneficial organisms necessary for reproductive success. Asteraceae is the largest family among flowering plants. Its representatives are characterized by unique inflorescence called capitulum, which has been reduced to a reproduction unit resembling a single flower. Here, we chose Chrysanthemum morifolium, a model species of Asteraceae, to investigate how the capitulum balances the accumulation and emission of floral terpenoid volatiles that are implicated in defense and pollinator attraction, respectively. Our results showed that the capitula of C. morifolium produce and emit complex mixtures of monoterpenoids and sesquiterpenoids. The highest concentrations of terpenoids were detected in the bud stage of the capitula. In contrast, the capitulum reached the highest emission level prior to full blooming. The disc florets were the dominant organs of terpenoid accumulation and emission in the full-openness stage. To understand the molecular basis of volatile terpenoid biosynthesis in C. morifolium, experiments were designed to study terpene synthase (TPS) genes, which are pivotal for terpene biosynthesis. Eight CmCJTPS genes were identified in the transcriptomes of C. morifolium, and the proteins encoded by five genes were found to be biochemically functional. CmCJTPS5 and CmCJTPS8 were the multi-product enzymes catalyzing the monoterpenoid and sesquiterpenoid formation, which closely matched the major terpenoids produced in the flower heads. The five functional terpene synthase genes exhibited similar temporal expression patterns but diverse spatial expression levels, suggesting tissue-specific functions. Altogether, our results illustrate the dynamic patterns of accumulation and emission of floral volatile terpenoids implicated in defense and attracting pollinators in C. morifolium, for which both the regulation of TPS gene expression and the regulation of release may play critical roles.

Complex Sexual Deception in an Orchid Is Achieved by Co-opting Two Independent Biosynthetic Pathways for Pollinator Attraction

Abstract:Sexually deceptive orchids lure their specific male pollinators using volatile semiochemicals that mimic female sex pheromones. To date, the semiochemicals known to be involved consist of blends of chemically and biosynthetically related compounds. In contrast, we report that (S)-β-citronellol and 2-hydroxy-6-methylacetophenone, two biosynthetically distinct compounds, are the active semiochemicals in Caladenia plicata, which is pollinated by male Zeleboria sp. thynnine wasps. They are also sex pheromone components of the female Zeleboria. A 1:4 blend elicits a high rate of attempted copulation (∼70%) in bioassays, equivalent to rates observed at orchid flowers. Whereas β-citronellol is well known, 2-hydroxy-6-methylacetophenone appears to be previously unknown as a floral volatile. Production of the two compounds is restricted to glandular sepal tips; thus, differential expression analysis of contrasting floral tissue transcriptomes was employed to illuminate the biosynthesis. As expected, production of (S)-β-citronellol commences with the terpene synthase GES1 catalyzing the irreversible conversion of geranyl diphosphate (GPP) to geraniol. Contrary to prediction, biosynthesis subsequently proceeds in three steps, commencing with the oxidation of geraniol to geranial by alcohol dehydrogenase ADH3, followed by the enantioselective reduction of a double bond in geranial by geranial reductase GER1 to give (S)-β-citronellal. Finally, ADH3-catalyzed reduction of (S)-β-citronellal results in (S)-β-citronellol. In line with previous work on insects showing that 2-hydroxy-6-methylacetophenone is derived from a polyketide pathway, we report a differentially expressed polyketide synthase (PKS) gene candidate. Thus, in this unique example of sexual deception, pollination is achieved by co-opting and regulating two independent biosynthetic pathways of floral volatile compounds. VIDEO ABSTRACT.
Divergence in floral scent and morphology, but not thermogenic traits, associated with pollinator shift in two brood-site-mimicking Typhonium (Araceae) species

Abstract:Background: Flowers which imitate insect oviposition sites probably represent the most widespread form of floral mimicry, exhibit the most diverse floral signals and are visited by two of the most speciose and advanced taxa of insect - beetles and flies. Detailed comparative studies on brood-site mimics pollinated exclusively by each of these insect orders are lacking, limiting our understanding of floral trait adaptation to different pollinator groups in these deceptive systems. Methods: Two closely related and apparent brood-site mimics, Typhonium angustilobum and T. wilbertii (Araceae) observed to trap these distinct beetle and fly pollinator groups were used to investigate potential divergence in floral signals and traits most likely to occur under pollinator-mediated selection. Trapped pollinators were identified and their relative abundances enumerated, and thermogenic, visual and chemical signals and morphological traits were examined using thermocouples and quantitative reverse transcription-PCR, reflectance, gas chromatography-mass spectrometry, floral measurements and microscopy. Key results: Typhonium angustilobum and T. wilbertii were functionally specialized to trap saprophagous Coleoptera and Diptera, respectively. Both species shared similar colour and thermogenic traits, and contained two highly homologous AOX genes (AOX1a and AOX1b) most expressed in the thermogenic tissue and stage (unlike pUCP). Scent during the pistillate stage differed markedly - T. angustilobum emitted a complex blend of sesquiterpenes, and T. wilbertii, a dung mimic, emitted high relative amounts of skatole, p-cresol and irregular terpenes. The species differed significantly in floral morphology related to trapping mechanisms. Conclusions: Functional specialization and pollinator divergence were not associated with differences in anthesis rhythm and floral thermogenic or visual signals between species, but with significant differences in floral scent and morphological features, suggesting that these floral traits are critical for the attraction and filtering of beetle or fly pollinators in these two brood-site mimics.

Abstract:As a fundamental step towards a clean, renewable source of H2, a novel physical chemical process within molten NaOH in which an external single, small bad gap photosentizer, such as Si, can drive the energetics of water cleavage is demonstrated, and is accomplished by tuning (decreasing) the water splitting electrochemical potential, EH2O, rather than tuning the photosensitizer band gap; this diminished potential is due to (i) a thermodynamic temperature induced decrease of EH2O with increasing temperature, and (ii) a partial recombination of the cleavage products.

Abstract:Abstract Deceptive pollination often involves volatile organic compound emissions that mislead insects into performing nonrewarding pollination. Among deceptively pollinated plants, Arum maculatum is particularly well-known for its potent dung-like volatile organic compound emissions and specialized floral chamber, which traps pollinators—mainly Psychoda phalaenoides and Psychoda grisescens—overnight. However, little is known about the genes underlying the production of many Arum maculatum volatile organic compounds, and their influence on variation in pollinator attraction rates. Therefore, we performed de novo transcriptome sequencing of Arum maculatum appendix and male floret tissue collected during anthesis and postanthesis, from 10 natural populations across Europe. These RNA-seq data were paired with gas chromatography–mass spectrometry analyses of floral scent composition and pollinator data collected from the same inflorescences. Differential expression analyses revealed candidate transcripts in appendix tissue linked to malodourous volatile organic compounds including indole, p-cresol, and 2-heptanone. In addition, we found that terpene synthase expression in male floret tissue during anthesis significantly covaried with sex- and species-specific attraction of Psychoda phalaenoides and Psychoda grisescens. Taken together, our results provide the first insights into molecular mechanisms underlying pollinator attraction patterns in Arum maculatum and highlight floral chamber sesquiterpene (e.g. bicyclogermacrene) synthases as interesting candidate genes for further study.

Complex floral morphology and scent chemistry mediate hawkmoth pollination of an African orchid in the diverse Habenaria clade

Abstract:Abstract One explanation for the evolution of complex floral morphology in orchids is that it can function as a component of ‘lock-and-key’ mechanisms that ensure pollen transfer by specific flower visitors. We investigated the roles of floral morphology and scent in the pollination of Bonatea antennifera, an African orchid that is phylogenetically embedded in the hyper-diverse Habenaria clade. Using camera traps and direct observations, we established that B. antennifera is pollinated by an assemblage of short-tongued hawkmoths with proboscides ~4 cm in length, corresponding to the spur length of the orchid. Pollinaria are attached between the palps of these hawkmoths via an intricate mechanism involving a wedge-shaped rostellum chamber and a tooth in the floral spur that deflect the head and proboscis of the hawkmoth to either side of the spur entrance. This functional subdivision of the flower results in removal of a single pollinarium per visit. Scent production is synchronized with the peak activity of hawkmoths at dusk, and electrophysiology experiments showed that hawkmoth antennae respond to several of the dominant volatile terpenoid compounds [e.g. R-(−)-linalool, caryophyllene epoxide, and (E)-nerolidol] emitted by the flowers. These results illustrate the functional significance of complex floral morphology and scent for pollen transfer in orchids.

Metabolic Profiling of Terpene Diversity and the Response of Prenylsynthase-Terpene Synthase Genes During Biotic and Abiotic Stresses in Dendrobium Catenatum

Abstract:Dendrobium catenatum is a widely cultivated Chinese orchid herb rich in abundant secondary metabolites, such as terpenes. However, terpene distribution and characterization of terpene biosynthesis-related genes remain unknown in D. catenatum. In this study, metabolic profiling was performed to analyze terpene distribution in the root, stem, leaf, and flower of D. catenatum. A total of 74 terpene compounds were identified and classified. Clustering analysis revealed that terpene compounds exhibited a tissue-specific accumulation, including monoterpenes in the flowers, sesquiterpenes in the stems, and triterpenes in the roots. Transcriptome analysis revealed that the ‘terpenoid backbone biosynthesis’ pathway was only significantly enriched in root vs. flower. The expression of terpene biosynthesis-related genes was spatiotemporal in the flowers. Prenylsynthase-terpene synthases (PS-TPSs) are the largest and core enzymes for generating terpene diversity. By systematic sequence analysis of six species, 318 PS-TPSs were classified into 10 groups and 51 DcaPS-TPSs were found in eight of them. Eighteen DcaPS-TPSs were regulated by circadian rhythm under drought stress. Most of the DcaPS-TPSs were influenced by cold stress and fungi infection. The cis-element of the majority of the DcaPS-TPS promoters was related to abiotic stress and plant development. Methyl jasmonate levels were significantly associated with DcaTPSs expression and terpene biosynthesis. These results provide insight into further functional investigation of DcaPS-TPSs and the regulation of terpene biosynthesis in Dendrobium.
Extreme divergence in floral scent among woodland star species (Lithophragma spp.) pollinated by floral parasites

Abstract:Backgrounds and aims: A current challenge in coevolutionary biology is to understand how suites of traits vary as coevolving lineages diverge. Floral scent is often a complex, variable trait that attracts a suite of generalized pollinators, but may be highly specific in plants specialized on attracting coevolved pollinating floral parasites. In this study, floral scent variation was investigated in four species of woodland stars (Lithophragma spp.) that share the same major pollinator (the moth Greya politella, a floral parasite). Three specific hypotheses were tested: (1) sharing the same specific major pollinator favours conservation of floral scent among close relatives; (2) selection favours 'private channels' of rare compounds particularly aimed at the specialist pollinator; or (3) selection from rare, less-specialized co-pollinators mitigates the conservation of floral scent and occurrence of private channels. Methods: Dynamic headspace sampling and solid-phase microextraction were applied to greenhouse-grown plants from a common garden as well as to field samples from natural populations in a series of experiments aiming to disentangle the genetic and environmental basis of floral scent variation. Key results: Striking floral scent divergence was discovered among species. Only one of 69 compounds was shared among all four species. Scent variation was largely genetically based, because it was consistent across field and greenhouse treatments, and was not affected by visits from the pollinating floral parasite. Conclusions: The strong divergence in floral scents among Lithophragma species contrasts with the pattern of conserved floral scent composition found in other plant genera involved in mutualisms with pollinating floral parasites. Unlike some of these other obligate pollination mutualisms, Lithophragma plants in some populations are occasionally visited by generalist pollinators from other insect taxa. This additional complexity may contribute to the diversification in floral scent found among the Lithophragma species pollinated by Greya moths.

Abstract:Chemical defenses, repellents, and attractants are important shapers of species interactions. Chemical attractants could contribute to the divergence of coevolving plant-insect interactions, if pollinators are especially responsive to signals from the local plant species. We experimentally investigated patterns of daily floral scent production in three Lithophragma species (Saxifragaceae) that are geographically isolated and tested how scent divergence affects attraction of their major pollinator—the floral parasitic moth Greya politella (Prodoxidae). These moths oviposit through the corolla while simultaneously pollinating the flower with pollen adhering to the abdomen. The complex and species-specific floral scent profiles were emitted in higher amounts during the day, when these day-flying moths are active. There was minimal divergence found in petal color, which is another potential floral attractant. Female moths responded most strongly to scent from their local host species in olfactometer bioassays, and were more likely to oviposit in, and thereby pollinate, their local host species in no-choice trials. The results suggest that floral scent is an important attractant in this interaction. Local specialization in the pollinator response to a highly specific plant chemistry, thus, has the potential to contribute importantly to patterns of interaction specificity among coevolving plants and highly specialized pollinators.

Complex reproductive secretions occur in all extant gymnosperm lineages: a proteomic survey of gymnosperm pollination drops

Abstract:Complex protein-containing reproductive secretions are a conserved trait amongst all extant gymnosperms; the pollination drops of most groups include carbohydrate-modifying enzymes and defence proteins. Pollination drops are aqueous secretions that receive pollen and transport it to the ovule interior in gymnosperms (Coniferales, Cycadales, Ginkgoales, Gnetales). Proteins are well established as components of pollination drops in conifers (Coniferales) and Ephedra spp. (Gnetales), but it is unknown whether proteins are also present in the pollination drops of cycads (Cycadales), Ginkgo (Ginkgoales), Gnetum (Gnetales), or in the pollination drops produced by sterile ovules occurring on pollen plants in the Gnetales. We used liquid chromatography-tandem mass spectrometry followed by database-derived protein identification to conduct proteomic surveys of pollination drops collected from: Ceratozamia hildae, Zamia furfuracea and Cycas rumphii (Cycadales); Ginkgo biloba (Ginkgoales); Gnetum gnemon and Welwitschia mirabilis, including pollination drops from both microsporangiate and ovulate plants (Gnetales). We identified proteins in all samples: C. hildae (61), Z. furfuracea (40), C. rumphii (9), G. biloba (57), G. gnemon ovulate (17) and sterile ovules from microsporangiate plants (25) and W. mirabilis fertile ovules (1) and sterile ovules from microsporangiate plants (138). Proteins involved in defence and carbohydrate modification occurred in the drops of most groups, indicating conserved functions for proteins in pollination drops. Our study demonstrates that all extant gymnosperm groups produce complex reproductive secretions containing proteins, an ancient trait that likely contributed to the evolutionary success of seed plants.
Evolution and diversity of floral scent chemistry in the euglossine bee-pollinated orchid genus Gongora

Abstract:• Background and aims: Animal-pollinated angiosperms have evolved a variety of signalling mechanisms to attract pollinators. Floral scent is a key component of pollinator attraction, and its chemistry modulates both pollinator behaviour and the formation of plant-pollinator networks. The neotropical orchid genus Gongora exhibits specialized pollinator associations with male orchid bees (Euglossini). Male bees visit orchid flowers to collect volatile chemical compounds that they store in hind-leg pouches to use subsequently during courtship display. Hence, Gongora floral scent compounds simultaneously serve as signalling molecules and pollinator rewards. Furthermore, because floral scent acts as the predominant reproductive isolating barrier among lineages, it has been hypothesized that chemical traits are highly species specific. A comparative analysis of intra- and inter-specific variation of floral scent chemistry was conducted to investigate the evolutionary patterns across the genus. • Methods: Gas chromatography-mass spectrometry (GC-MS) was used to analyse the floral scent of 78 individuals belonging to 28 different species of Gongora from two of the three major lineages sampled across the neotropical region. Multidimensional scaling and indicator value analyses were implemented to investigate the patterns of chemical diversity within and among taxonomic groups at various geographic scales. Additionally, pollinator observations were conducted on a sympatric community of Gongora orchids exhibiting distinct floral scent phenotypes. • Key results: A total of 83 floral volatiles, mainly terpenes and aromatic compounds, were detected. Many of the identified compounds are common across diverse angiosperm families (e.g. cineole, eugenol, β-ocimene, β-pinene and terpinen-4-ol), while others are relatively rare outside euglossine bee-pollinated orchid lineages. Additionally, 29 volatiles were identified that are known to attract and elicit collection behaviour in male bees. Floral scent traits were less variable within species than between species, and the analysis revealed exceptional levels of cryptic diversity. Gongora species were divided into 15 fragrance groups based on shared compounds. Fragrance groups indicate that floral scent variation is not predicted by taxonomic rank or biogeographic region. • Conclusions: Gongora orchids emit a diverse array of scent molecules that are largely species specific, and closely related taxa exhibit qualitatively and quantitatively divergent chemical profiles. It is shown that within a community, Gongora scent chemotypes are correlated with near non-overlapping bee pollinator assemblies. The results lend support to the hypothesis that floral scent traits regulate the architecture of bee pollinator associations. Thus, Gongora provides unique opportunities to examine the interplay between floral traits and pollinator specialization in plant-pollinator mutualisms.
Pollination of Chamaepentas nobilis (Rubiaceae) by long‐proboscid hawkmoths in south‐central Africa: Trait mismatching and floral scent chemistry

Abstract:Flowers usually evolve a depth that either matches or slightly exceeds the tongue length of their pollinators. In the case of an African shrub pollinated by hawkmoths, pollen is transferred on their extremely long tongues, which exceed the depth of the flowers. This allows the plant to be co‐pollinated by hawkmoths that vary on tongue length. The evolution of extremely long (>10 cm) floral tubes in angiosperms is closely linked with adaptation for pollination by long‐proboscid hawkmoths. In most cases, pollen is placed on the head or body of these moths, selecting for floral tubes that match or slightly exceed their proboscis length as this ensures contact with reproductive parts of the flower. However, in the case of Chamaepentas nobilis (Rubiaceae), anthers are inserted inside the c. 12‐cm floral tubes and coat the proboscis of visiting hawkmoths with sticky pollen, meaning that insects with proboscides longer than the floral tube can be effective pollinators, despite trait mismatching. Direct observations and camera trapping on granite outcrops in central Zambia showed that C. nobilis was visited both by Agrius convolvuli (proboscis length c. 13 cm) and Xanthopan morganii (proboscis length c. 17 cm), which are the only moth species in Africa that can access the small amounts of dilute nectar at the base of the floral tube. Pollen on the proboscides of captured hawkmoths was confirmed to originate from C. nobilis. Key floral advertising traits of C. nobilis include early evening anthesis, the highly reflective white limbus, and the evening production of scent dominated by oxygenated aromatic and terpenoid compounds known to elicit antennal responses of A. convolvuli. This study shows that that A. convolvuli and X. morganii share floral resources and jointly pollinate C. nobilis, despite their proboscides differing in length and being mismatched with the corolla tube length.

Abstract:<h3 class="a-plus-plus">Abstract</h3> Nocturnal flowering plants often release strong scents to attract their pollinators. Among night active flower visitors are cyclocephaline scarab beetles, which have been demonstrated to respond to uncommon volatile organic compounds released in high amounts by their host plants. In Araceae, the molecular structure of several such compounds is yet to be unveiled. We investigated headspace floral scent samples of Philodendron squamiferum, Thaumatophyllum mello-baretoanum, and Xanthosoma hylaeae by a variety of approaches, leading to the identification of novel compounds. Dehydrojasmone, (Z)-4-methylene-5-(pent-2-en-1-yl)cyclopent-2-en-1-one (1), (Z)-3-methylene-2-(pent-2-en-1-yl)cyclopentyl acetate (isojasmyl acetate, 3), and (E)-4,8-dimethylnona-1,3,7-trien-5-yl acetate (4) had not been previously reported, while full analytical data of the recently described (Z)-3-methylene-2-(pent-2-en-1-yl)cyclopentan-1-ol (isojasmol, 2) are presented here. All these compounds are derived from more common precursors, (Z)-jasmone and (E)-4,8-dimethyl-1,3,7-nonatriene, likely through biosynthetic “post-processing”.

The key role of 4-methyl-5-vinylthiazole in the attraction of scarab beetle pollinators: a unique olfactory floral signal shared by Annonaceae and Araceae

Abstract:Cyclocephaline scarabs are specialised scent-driven pollinators, implicated with the reproductive success of several Neotropical plant taxa. Night-blooming flowers pollinated by these beetles are thermogenic and release intense fragrances synchronized to pollinator activity. However, data on floral scent composition within such mutualistic interactions are scarce, and the identity of behaviorally active compounds involved is largely unknown. We performed GC-MS analyses of floral scents of four species of Annona (magnoliids, Annonaceae) and Caladium bicolor (monocots, Araceae), and demonstrated the chemical basis for the attraction of their effective pollinators. 4-Methyl-5-vinylthiazole, a nitrogen and sulphur-containing heterocyclic compound previously unreported in flowers, was found as a prominent constituent in all studied species. Field biotests confirmed that it is highly attractive to both male and female beetles of three species of the genus Cyclocephala, pollinators of the studied plant taxa. The origin of 4-methyl-5-vinylthiazole in plants might be associated with the metabolism of thiamine (vitamin B1), and we hypothesize that the presence of this compound in unrelated lineages of angiosperms is either linked to selective expression of a plesiomorphic biosynthetic pathway or to parallel evolution.
The dilemma of being a fragrant flower: the major floral volatile attracts pollinators and florivores in the euglossine-pollinated orchid Dichaea pendula

Abstract:Volatile organic compounds (VOCs) mediate both mutualistic and antagonistic plant–animal interactions; thus, the attraction of mutualists and antagonists by floral VOCs constitutes an important trade-off in the evolutionary ecology of angiosperms. Here, we evaluate the role of VOCs in mediating communication between the plant and its mutualist and antagonist floral visitors. To assess the evolutionary consequences of VOC-mediated signalling to distinct floral visitors, we studied the reproductive ecology of Dichaea pendula, assessing the effects of florivores on fruit set, the pollination efficiency of pollinators and florivores, the floral scent composition and the attractiveness of the major VOC to pollinators and florivores. The orchid depends entirely on orchid-bees for sexual reproduction, and the major florivores, the weevils, feed on corollas causing self-pollination, triggering abortion of 26.4 % of the flowers. Floral scent was composed of approximately 99 % 2-methoxy-4-vinylphenol, an unusual floral VOC attractive to pollinators and florivores. The low fruit set from natural pollination (5.6 %) compared to hand cross-pollination (45.5 %) and low level of pollinator visitation [0.02 visits (flower hour)−1] represent the limitations to pollination. Our research found that 2-methoxy-4-vinylphenol mediates both mutualistic and antagonistic interactions, which could result in contrary evolutionary pressures on novo-emission. The scarcity of pollinators, not florivory, was the major constraint to fruit set. Our results suggest that, rather than anti-florivory adaptations, adaptations to enhance pollinator attraction and cross-pollination might be the primary drivers of the evolution of VOC emission in euglossine-pollinated flowers.

Pollinator Preferences for Floral Volatiles Emitted by Dimorphic Anthers of a Buzz-Pollinated Herb

Abstract:Floral scents attract pollinators to plant rewards; in nectarless flowers, pollen grains are the only reward. Thus, pollen not only fertilizes ovules, but also feeds pollinators. This dilemma is resolved by specialization of anthers (i.e., heteranthery): feeding anthers that feed pollinators and pollinating anthers for fertilization. We hypothesized that the chemical composition of floral volatiles differs between the two types of anther and influences pollination preference for feeding anthers. We used Solanum rostratum as a study model because its heterantherous flowers produce a floral scent that suggests a close association with their pollinators. The main aim of this study was to determine the chemical composition of the two types of anther and to investigate how they influence foraging behaviour of pollinators. To characterize this composition, we used solid phase microextraction and hexane extraction followed by gas chromatography-mass spectrometry. We registered 12 volatile compounds in S. rostratum floral extracts, mainly aromatic and sesquiterpene compounds. The proportion of these compounds differed between feeding and pollinating anthers. Some of these compounds were probably emitted by osmophores located in both anther types. Also, we used electroantennography to investigate Melipona solani antennal response to floral volatiles. The M. solani antennae are receptive to the highest floral extract dose tested. Finally, we conducted two behavioural bioassays to test bee attraction for each type of floral extract: a) multiple-choice in a feeding arena using M. solani and b) Y-olfactometer bioassay using Bombus impatiens. Both bee species preferred feeding anthers in bioassays. In conclusion, heteranthery involves chemical differentiation (i.e., proportion of volatiles compounds) in anther specialization that influences bee preference for feeding anthers over pollinating anthers.
Multi-omics analysis the differences of VOCs terpenoid synthesis pathway in maintaining obligate mutualism between Ficus hirta Vahl and its pollinators

Abstract:Inroduction Volatile organic compounds (VOCs) emitted by the receptive syconia of Ficus species is a key trait to attract their obligate pollinating fig wasps. Ficus hirta Vahl is a dioecious shrub, which is pollinated by a highly specialized symbiotic pollinator in southern China. Terpenoids are the main components of VOCs in F. hirta and play ecological roles in pollinator attraction, allelopathy, and plant defense. However, it remains unclear that what molecular mechanism difference in terpenoid synthesis pathways between pre-receptive stage (A-phase) and receptive stage (B-phase) of F. hirta syconia. Methods Transcriptome, proteome and Gas Chromatography-Mass Spectrometer (GC-MS) were applied here to analyze these difference. Results and discussion: Compared to A-phase syconia, the genes ( ACAT2 , HMGR3 , GGPS2 , HDR , GPS2 , TPS2 , TPS4 , TPS10-4 , TPS14 ) related to the terpenoid synthesis pathway had higher expression level in receptive syconia (B-phase) according to transcriptome sequencing. Seven differentially expressed transcription factors were screened, namely bHLH7 , MYB1R1 , PRE6 , AIL1 , RF2b , ANT , VRN1 . Specifically, bHLH7 was only specifically expressed in B-phase. 235 differentially expressed proteins (DEPs) were mainly located in the cytoplasm and chloroplasts. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that the DEPs were mainly enriched in the metabolic process. A total of 9 terpenoid synthesis proteins were identified in the proteome. Among them, 4 proteins in methylerythritol phosphate (MEP) pathway were all down-regulated. Results suggested the synthesis of terpenoids precursors in B-phase bracts were mainly accomplished through the mevalonic acid (MVA) pathway in cytoplasm. Correlation analysis between the transcriptome and proteome, we detected a total of 1082 transcripts/proteins, three of which are related to stress. From the VOCs analysis, the average percent of monoterpenoids emitted by A-phase and B-phase syconia were 8.29% and 37.08%, while those of sesquiterpenes were 88.43% and 55.02% respectively. Monoterpenes (camphene, myrcene, camphor, menthol) were only detected in VOCs of B-phase syconia. To attract pollinators, B-phase syconia of F. hirta need more monoterpenoids and less sesquiterpenes. We speculate that transcription factor bHLH7 may regulate the terpenoid synthesis pathway between A- and B-phase syconia. Our research provided the first global analysis of mechanism differences of terpenoid synthesis pathways between A and B phases in F. hirta syconia.
