Abstract:Chemodynamic therapy (CDT) as an emerging modality in cancer treatment, its implementation remains a daunting challenge by the lack of smart Fenton catalyst under acidic tumor microenvironments. Herein, we have successfully constructed a Fe3O4 @MIL-10 0 heterojunction by growing Fe-based metal-organic framework (MIL-100) onto the surface of Fe3O4 nanoparticles. The as-made heterojunction after encapsu-lating glucose oxidase (termed FMG) is demonstrated as a pH-responsive intelligent Fenton nanosystem with the synergistic effect of starvation therapy (ST). Density functional theory (DFT) calculations reveal that such heterojunction could greatly reduce the energy barrier of the Fenton reaction, which better ex-plains the mechanism of Fenton performance improvement. Moreover, the encapsulated glucose oxidase has successfully activated the ST process, in which its generated H2O2 and gluconic acid further improve the CDT efficiency. More O 2 from the enhanced CDT in turn promotes the enzymatic reaction of glucose oxidase. The Fenton/cascade enzymatic reaction operates in a self-feedback manner as proposed. In vitro and in vivo experiments demonstrate that such intelligent Fenton nanoreactors provide a powerful anti-cancer mechanism for effective tumor ablation with enough safety. This work provides insights into the developments of MOF-based heterojunctions as powerful anticancer treatment nanoreactors. Abstract:The Fenton reaction-based chemodynamic therapy is a form of cancer therapy, and its efficacy can be significantly improved by promoting catalytic reactions involving iron ions. A system with high catalytic capacity and low biological toxicity that effectively inhibits tumor progression is required for optimal treatment. In this study, iron-loaded carbonaceous nanoparticles (CNPs@Fe) with Fenton catalytic activity were fabricated and applied for the chemodynamic therapy of cancer. The carbonaceous nanoparticles derived from glucose via a caramelization reaction demonstrated high biocompatibility. Besides, aromatic structures in the carbonaceous nanoparticles helped accelerate electron transfer to enhance the catalytic decomposition of H 2 O 2 , resulting in the formation of highly reactive hydroxyl radicals ( ̇OH). At pH 6.0 (representing weak acidity in the tumor microenvironment), the Fenton catalytic activity of CNPs@Fe in the decomposition of H 2 O 2 was 15.3 times higher than that of Fe 2+ and 28.3 times higher than that of Fe 3 O 4 via a chromogenic reaction. The reasons for the enhancement were revealed by analyzing the chemical composition of carbonaceous nanoparticles using high-resolution mass spectra. The developed Fenton agent also demonstrated significant therapeutic effectiveness and minimal side effects in in vitro and in vivo anticancer studies. This work proposes a novel approach to promote the generation of reactive oxygen species (ROS) for the chemodynamic therapy of cancer.

One‐Pot Approach to Fe 2+ /Fe 3+ ‐Based MOFs with Enhanced Catalytic Activity for Fenton Reaction

Abstract:Smart theragnostic nanoplatforms exhibit great promise in clinical tumor treatment. The Fe-based Fenton reaction in tumor sites may generate reactive oxygen species to kill cancer cells with negligible side effects on normal tissues. However, its efficiency and duration are limited by the low intracellular concentration of H2O2, weak acidicity of tumor tissue, and low catalytic activity of conventional Fenton reagents. Herein, a facile strategy is proposed to efficiently overcome these obstacles. An efficient enzymatic/Fenton-starvation nanoreactor PMs loaded with glucose oxidase and perfluoropentane (PGPMs) is constructed through synthesizing methoxy-PEG-carboxymethy-modified iron (Fe2+/Fe3+)-based metal–organic frameworks (PMs), followed by loading glucose oxidase (GOx) and perfluoropentane (PFP). PGPMs accumulating in the tumor tissue exhibit tumor microenvironment-responsive biodegradable behavior and unusual catalytic activity for Fenton reaction advantageous over Fe3+-based MOFs. Meanwhile, encapsulation of GOx into PGPMs further significantly increases the catalytic activity for Fenton reaction and also induces starvation therapy. PGPMs also exhibit considerable capabilities of ultrasound and tumor microenvironment-responsive T2 MR imaging applicable for contrast-enhanced diagnosis. Both in vitro and in vivo studies demonstrate the great diagnostic and therapeutic potentials of this nanoreactor in tumor.

Abstract:The pursuit of mesoporous Fe-based nanoagents addresses the field of developing alternative Fe-bearing nanoagents for synergistic cancer therapy with the expectation that the use of an essential element may avoid the issues raised by the exogenous administration of other metal element-based nanoagents. Herein, we highlight the interface-engineered mesoporous FeB (mFeB) where the core mFeB is interfacially oxidized into an FeOOH nanosheet loaded with the chemotherapeutic drug doxorubicin (DOX) and further encapsuled within the double-sulfide-bonded SiO2 outer layer, denoted as mFeB@DOX-ss-SiO2, which can realize programmed drug release for synergistic cancer theranostics. When only in a tumor microenvironment, the nanoagent can be activated to release DOX from the mFeB and FeOOH nanosheets as well as expose the easily oxidized mFeB to spontaneously transform to FeOOH nanosheets with Fenton activity to facilitate chemodynamic therapy (CDT). In addition, the high photothermal conversion efficiency of mFeB@DOX-ss-SiO2 would promote CDT. Also, owing to the inherent nature of ferromagnetism and red fluorescence of DOX, mFeB@DOX-ss-SiO2 can realize T2-weighted magnetic resonance imaging and fluorescence imaging. In vivo mouse model experiments demonstrate that mFeB@DOX-ss-SiO2 with good biocompatibility realizing CDT/photothermal therapy/chemotherapy achieved complete tumor suppression. This study opens up a new way to explore theranostic nanoagents.

Abstract:Metal‐organic framework (MOF)‐based drug delivery nanomaterials for cancer therapy have attracted increasing attention in recent years. Here, we propose an enhanced chemodynamic anti‐tumor therapy strategy by promoting the Fenton reaction by using core‐shell zeolitic imidazolate framework‐8 (ZIF‐8)@Fe3O4 as a therapeutic platform. Carboxymethyl cellulose (CMC) was used as a stabilizer of Fe3O4, which then decorated on the surface of ZIF‐8 via the electrostatic interaction and served as an efficient Fenton reaction trigger. Meanwhile, the pH‐responsive ZIF‐8 scaffold acted as a container to encapsulate the chemotherapeutic drug doxorubicin (DOX). The obtained DOX‐ZIF‐8@Fe3O4/CMC nanoparticles concomitantly accelerated DOX release and generated more hydroxyl radicals by targeting the lysosomes in cancer cells. In vitro and in vivo studies verify that the DZFC nanoparticles trigger GPX4‐dependent ferroptosis via the activation of the c‐Jun N‐terminal kinases (JNK) signaling pathway, following to achieve the chemo/ferroptosis synergistic anti‐tumor efficacy. No marked toxic effects were detected during DZFC treatment in a tumor‐bearing mouse model. This composite nanoparticle remarkably suppressed the tumor growth with minimized systemic toxicity, opening new horizons for the next generation of theragnostic nanomedicines. Abstract:Fenton reaction, a typical inorganic reaction, is broadly utilized in the field of wastewater treatment. Recently In case of its ability to inhibit the growth of cancer cells, it has been frequently reported in cancer treatment. Using the unique tumor microenvironment in cancer cells, many iron-based nanoparticles have been developed to release iron ions in cancer cells to induce Fenton reaction. In this mini review, we outline several different types of iron-based nanoparticles and several main means to enhance Fenton reaction in cancer cells. Finally, we discussed the advantages and disadvantages of iron-based nanoparticles for cancer therapy, prospected the future development of iron-based nanoparticles. It is believed that iron-based nanoparticles can make certain contribution to the cause of human cancer in the future.
Fenton-like nanoparticles capable of H2O2 self-supply and glutathione consumption for chemodynamic and chemotherapy of cancer

Abstract:Chemodynamic therapy (CDT) utilizing the Fenton reaction to convert hydrogen peroxide (H2O2) into cytotoxic hydroxyl radicals (˙OH) has recently drawn extensive interest in tumor treatment. However, the therapeutic efficiency of CDT often suffers from high concentrations of glutathione (GSH), insufficient endogenous H2O2 and inefficient Fenton activity. Herein, a GSH-depleting and H2O2 self-providing nanosystem that can efficiently load copper ions and doxorubicin (DOX) (MSN-Cu2+-DOX) to induce enhanced CDT and chemotherapy is proposed. The results show that MSN-Cu2+-DOX could release Cu2+ and DOX under acidic conditions. Particularly, both the released Cu2+ and Cu2+ in MSN-Cu2+-DOX are available for ˙OH production via a Fenton-like reaction for CDT. Meanwhile, Cu2+ undergoes a reduction to Cu+ by depleting overexpressed GSH, thereby enhancing CDT. Moreover, the released DOX could not only be used for chemotherapy, but also promote the generation of endogenous H2O2 to improve the efficiency of a Cu-based Fenton-like reaction. Resultantly, this nanosystem featuring Fenton-like activity, GSH consumption, H2O2 self-sufficiency and chemotherapy exhibits a great antitumor effect with a tumor inhibition ratio of 93.05%. Overall, this study provides a promising strategy to enhance CDT for effective tumor therapy.

Abstract:Fenton reaction-mediated oncotherapy is an emerging strategy which uses iron ions to catalytically convert endogenous hydrogen peroxide into hydroxyl radicals, the most reactive oxygen species found in biology, for efficient cancer therapy. However, Fenton reaction efficiency in tumor tissue is typically limited due to restrictive conditions. One strategy to overcome this obstacle is to increase the temperature specifically at the tumor site. Herein, a tumor-targeting iron sponge (TTIS) nanocomposite based on graphdiyne oxide, which has a high affinity for iron is described. TTIS can accumulate in tumor tissue by decoration with a tumor-targeting polymer to enable tumor photoacoustic and magnetic resonance imaging. With its excellent photothermal conversion efficiency (37.5%), TTIS is an efficient photothermal therapy (PTT) agent. Moreover, the heat produced in the process of PTT can accelerate the release of iron ions from TTIS and simultaneously enhance the efficiency of the Fenton reaction, thus achieving a combined PTT and Fenton reaction-mediated cancer therapy. This work introduces a graphdiyne oxide-based iron sponge that exerts an enhanced antitumor effect through PTT and Fenton chemistry.

Abstract:It is the challenging goal in cancer biomedicine to search novel cancer-therapeutic modality with concurrent high therapeutic efficiency on combating cancer and low side effects to normal cells/tissues. The recently developed nanocatalytic cancer therapy based on catalytic Fenton reaction represents one of the promising paradigms for potential clinical translation, which has got fast progress very recently. This progress report discusses the rational design and fabrication of Fenton reaction-based nanocatalysts for triggering the in-situ Fenton chemical reaction within tumor microenvironment to generate highly toxic hydroxyl radicals (•OH), which is highly efficient for killing the cancer cells and suppressing the tumor growth. Several strategies for optimizing the nanocatalytic cancer-therapeutic efficiency of Fenton reaction have been highlighted, including screening high-performance Fenton nanocatalysts, increasing peroxide-hydrogen amounts as the reactants, changing the Fenton-reaction conditions (e.g., temperature, acidity and photo-triggering), and Fenton reaction-based synergistic cancer therapy such as some sequential nanocatalytic reactions with improved therapeutic outcome. The facing challenges and future developments of Fenton reaction-based nanocatalytic cancer therapy are also discussed for further promoting the clinical translation of this emerging cancer-therapeutic modality to benefit the cancer patients.

Abstract:Recently, nanoparticle‐triggered in situ catalytic Fenton/Fenton‐like reaction is widely explored for tumor‐specific chemodynamic therapy (CDT). However, despite the great potential of CDT in tumor treatment, insensitive response to the relatively high pH of the tumor sites and the insufficient intratumoral H2O2 level leads to limited efficiency of most Fenton/Fenton‐like reactions, which greatly imped its clinical conversion. This paper reports the fabrication of Fenton‐type bimetallic peroxides for ultrasensitive chemodynamic therapy with high pH‐activated, synergistic effect/H2O2 self‐supply‐mediated cascade Fenton chemistry for the first time. The observations reveal that these bimetallic peroxides exhibit an ultrasensitive acid‐activated decomposition‐mediated Fenton‐like reaction at the relatively high pH of 6.5–7.0, accompanied with highly increased •OH generation efficiency (especially, 40–60‐fold increase at pH 7.0) by the metal‐mediated synergistic effect‐enhanced Fenton chemistry as well as in situ self‐generated H2O2 supplement. Moreover, the bimetallic peroxides exhibit high tumor accumulation which along with a high‐efficiency tumor catalytic‐therapeutic with negligible side effects in vivo. Developing these novel bimetallic peroxides, together with the already demonstrated capacity of the key metals (Fe, Mn, Cu, etc.) for magnetic resonance imaging or photodynamic/immune‐enhanced therapy, will propel interest in development of smart high‐efficiency nanoplatform for cancer theranostics.

Tumor-targeted supramolecular catalytic nanoreactor for synergistic chemo/chemodynamic therapy via oxidative stress amplification and cascaded Fenton reaction

Abstract:Utilizing intratumoral iron mediated Fenton reaction to induce cancer cells destruction, chemodynamic therapy (CDT) is an emerging tumor-specific therapeutic strategy. However, the CDT efficacy is severely restrained by insufficient endogenous H2O2 and catalytic ferrous ions in tumor site. Herein, based on innovative small molecule prodrug design concept, an acid-labile ferrocene-modified cinnamaldehyde prodrug (Fc-CA) was first synthesized. Skillfully combining ROS-generator CA with Fenton catalyst Fc in its molecular structure, Fc-CA could synchronously elevate H2O2 level and catalytic iron amount for enhanced CDT efficacy. Then, hydrophobic Fc-CA was efficiently encapsulated into hydrophilic cyclodextrin-modified hyaluronic acid conjugate (HA-CD) via host-guest interactions, to spontaneously form pH/redox dual-responsive supramolecular catalytic nanoreactor (HA-CD/Fc-CA NPs). The nanoreactor with high drug loading and stability, excellent catalytic activity and tumor specificity could preferentially accumulate in tumor tissues via EPR effect and CD44 receptor-mediated internalization. Through the cleavage of acid-labile hydrazone bond under acidic lysosome environments, the nanoreactor would be dissociated and release pre-protected CA for reinforced oxidation therapy and amplified H2O2 level, which would in turn react with Fc through cascaded Fenton reaction, to generate highly cytotoxic •OH for enhanced cancer killing in a positive feedback process. Both in vitro and in vivo results verified excellent cascade-amplifying therapeutic performance and favorable biocompatibility of this nanoreactor. This study provides an innovative strategy for exploiting versatile organic therapeutic nanoplatforms, simultaneously activating tumor-specific oxidative stress amplification and cascaded enhanced Fenton reaction for superior cancer therapy with excellent biosafety.
Synthesis of Iron Nanometallic Glasses and Their Application in Cancer Therapy by a Localized Fenton Reaction

Abstract:Metallic glasses and cancer theranostics are emerging fields that do not seem to be related to each other. Herein, we report the facile synthesis of amorphous iron nanoparticles (AFeNPs) and their superior physicochemical properties compared to their crystalline counterpart, iron nanocrystals (FeNCs). The AFeNPs can be used for cancer theranostics by inducing a Fenton reaction in the tumor by taking advantage of the mild acidity and the overproduced H2 O2 in a tumor microenvironment: Ionization of the AFeNPs enables on-demand ferrous ion release in the tumor, and subsequent H2 O2 disproportionation leads to efficient (.)OH generation. The endogenous stimuli-responsive (.)OH generation in the presence AFeNPs enables a highly specific cancer therapy without the need for external energy input.

Abstract:Chemodynamic therapy based on Fe2+-catalyzed Fenton reaction holds great promise in cancer treatment. However, low-produced hydroxyl radicals in tumor cells constitute its severe challenges because of the fact that Fe2+ with high catalytic activity could be easily oxidized into Fe3+ with low catalytic activity, greatly lowering Fenton reaction efficacy. Here, we codeliver CuS with the iron-containing prodrug into tumor cells. In tumor cells, the overproduced esterase could cleave the phenolic ester bond in the prodrug to release Fe2+, activating Fenton reaction to produce the hydroxyl radical. Meanwhile, CuS could act as a nanocatalyst for continuously catalyzing the regeneration of high-active Fe2+ from low-active Fe3+ to produce enough hydroxyl radicals to efficiently kill tumor cells as well as a photothermal therapy agent for generating hyperthermia for thermal ablation of tumor cells upon NIR irradiation. The results have exhibited that the approach of photothermal therapy nanomaterials boosting transformation of Fe3+ into Fe2+ in tumor cells can highly improve Fenton reaction for efficient chemodynamic therapy. This strategy was demonstrated to have an excellent antitumor activity both in vitro and in vivo, which provides an innovative perspective to Fenton reaction-based chemodynamic therapy.

Abstract:Chemodynamic therapy (CDT) shows great potential in tumor treatment, in which tumor cells are killed by hydroxyl radicals (·OH) generated by Fenton-catalysis-mediated hydrogen peroxide (H2O2) disproportionation. Fabricating a high-performance tumor microenvironment (TME)-responsive Fenton nanocatalyst and realizing sustainable ·OH production are essential for effective tumor elimination but still challenging. Herein, we reported a sustainable amorphous Fenton nanosystem (SAFN) for visualization-guided synergistic tumor therapy, highlighting the superiority of cascade catalysis and the sustainability of the Fenton nanosystem. The SAFN was fabricated by rational integration of a polydopamine (PDA) core, an amorphous ferric phosphate (FePi) payload, glucose oxidase (GOx) decoration, and a bull serum albumin coating. In an acidic TME, Fe(III) ions were released on demand from FePi and reduced to high-activity Fe(II) by PDA. The reducing capacity of PDA was stronger under acidic conditions, due to C=O exposure. Fe(II) and H2O2 are sustainably supplied by the PDA-reducing and GOx catalytic chemistry, achieving long-lasting ·OH production for enhanced CDT (ECDT). Localized hyperthermia can be generated by PDA under near-infrared laser irradiation for low-temperature photothermal therapy (LT-PTT) in a GOx-induced energy-shortage TME. In addition, a remarkable antitumor effect is achieved by this sustainable nanosystem through the synergistic effect of the starvation/LT-PTT/ECDT strategy.

Abstract:In this study, iron oxide core-shell mesoporous silica nanoparticles (Fe3O4@MSN) were prepared via the hydrolysis of tetraethyl orthosilicate on the surfaces of Fe3O4 nanoparticles, and these were further conjugated with folate (PEG-FA) and mitochondrial targeting triphenylphosphonium (TPP) to form Fe3O4@MSN-TPP/PEG-FA. A reactive oxygen species (ROS) promoting synergistic combined chemotherapy platform was designed through Fe3O4@MSN-TPP/PEG-FA encapsulating doxorubicin (DOX) and 3-amino-1,2,4-triazole (AT) for cancer therapy. DOX could stimulate the activation of nicotinamide adenine dinucleotide phosphate oxidases (NOXs), which change oxygen into superoxide radicals, which could be further triggered to produce hydrogen peroxide (H2O2) using the superoxide dismutase (SOD) enzyme. AT, as a catalase inhibitor, was employed to inhibit catalase activity to protect the production of H2O2. Thereafter, H2O2 was catalyzed with the help of Fe2+/Fe3+ to form highly toxic free hydroxyl radicals through Fenton reactions, which could induce cell death via synergistic DOX therapy. From in vitro assays, the prepared DOX/AT-loaded Fe3O4@MSN-TPP/PEG-FA showed remarkable inhibition efficiency (3.23% cell viability and 88.1% cell apoptosis) towards MGC-803 cells. This work has created a novel approach to gradually promote the production of ROS and combine this with chemotherapy to enhance anticancer efficacy.

Abstract:The recently developed reactive oxygen species (ROS)-manipulated nanocatalytic medicine in cancer treatment based on the Fenton reaction, defined as chemodynamic therapy (CDT), has been extensively studied and got fast progress. However, the heterogeneity and complexity of tumors impair the oxidation efficiency of Fenton reaction to some degree. For this reason, various modified strategies, such as Fenton-like reaction, photo-enhanced Fenton reaction, and Fenton catalytic-enhanced synergistic therapy, are being explored to improve the efficiency of CDT and traditional therapeutic methods. In this review, the recent progress in the design and application of Fenton nanocatalysts that employ Fenton or modified Fenton reaction for CDT is highlighted. Then, the latest signs of improvement in Fenton catalysis-enhanced cancer synergistic therapy are systematically presented. Finally, the purpose of this review is also to provide a better understanding of Fenton's reaction in cancer treatment and to anticipate the obstacles and challenges in the future development of CDT-based nanocatalytic medicine and combined therapy.

Abstract:Still today, cancer remains a threat to human health. Possible common treatments to cure this disease include chemotherapy (CT), radiotherapy (RT), photothermal therapy (PTT), and surgical resection, which give unreasonable results because of their limited efficiency and also lead to side-effects. Hence, different strategies are now being exploited to not only enhance the efficiency of these traditional therapeutic methods or treat the tumor cells but also curtail the side effects. A latest method with authentic proof of chemodynamic therapy (CDT) utilizing the Fenton reaction is now gaining importance. This approach, which is developed based on the high level of hydrogen peroxide (H2O2) in a tumor microenvironment (TME), can be used to catalyze the Fenton reaction to generate cancer cell-killing reactive oxygen species (ROS). The selection of materials is extremely important and nanomaterials offer the most likely method to facilitate CDT. Among various materials, metal-organic frameworks (MOFs) which have been extensively applied in medical areas are regarded as a promising material and possess potential for the next generation of nanotechnology. This review focuses on summarizing the use of MOFs in CDT and their synergetic therapeutics as well as the challenges, obstacles, and development.

Abstract:Full-process radiosensitization, that is, pre-increasing radiation sensitivity of cancer cells, magnifying •OH formation during ionizing irradiation, and intervention on the resultant DNA repair for final cells death, could enhance the overall radiotherapeutic effects, but has not yet been achieved. Herein, Hf-nMOFs with Fe3+ ions uniformly dispersed (Hf-BPY-Fe) were constructed to integratedly improve radiotherapeutic effects via a multifaceted mechanism. The in vitro experiments demonstrated that persistent reactive oxygen species stress from Hf-BPY-Fe-activated in situ Fenton reaction reassorted cell cycle distribution, consequently contributing to increased tumoral radiosensitivity to photon radiation. Upon irradiation during the course of radiation therapy, Hf4+ in Hf-BPY-Fe gave substantial amounts of high-energy electrons, which partially converted H2O to •OH and, meanwhile, relaxed to a low-energy state in nMOF pores, leading to an electron-rich environment. These aggregated electrons facilitated the reduction from Fe3+ to Fe2+ and further promoted the production of •OH in the Fenton process to attack DNA. The Hf-BPY-Fe postponed the DNA damage response process by interfering with certain proteins involved in the DNA repair signaling pathway. The in vivo experiments showed improved radiotherapeutic effects from integrated contributions from Fe3+-based Fenton reaction and Hf4+-induced X-ray energy conversion in tumors. This work provides a nMOFs-based full-process radiosensitizing approach for better radiotherapeutic efficacy.
