Abstract:This paper presents the performance of electronic holography for imaging applications of biological and medical interest involving turbid media. It demonstrates that digital holography is useful for biological studies in the areas of estuary sediment erosion and imaging unstained living cells. In particular, we demonstrate the recording geometries of classical 'in-line' back illumination using a collimated beam for measuring micro-panicles eroded in a cylindrical observation chamber; we also demonstrate a phase contrast set-up using a magnifying lens for observing biomedical micro-objects of unstained living cells in physiological saline. Practical results are presented for the studies of sediment erosion and transport, which provide information about how polymeric substances produced by the organisms in sediment and erosion shear velocity affect the sediment particle size when erosive stresses are applied to sediment beds.

Abstract:Imaging systems that exploit arrays of photodetectors in curvilinear layouts are attractive due to their ability to match the strongly nonplanar image surfaces (i.e., Petzval surfaces) that form with simple lenses, thereby creating new design options. Recent work has yielded significant progress in the realization of such "eyeball" cameras, including examples of fully functional silicon devices capable of collecting realistic images. Although these systems provide advantages compared to those with conventional, planar designs, their fixed detector curvature renders them incompatible with changes in the Petzval surface that accompany variable zoom achieved with simple lenses. This paper describes a class of digital imaging device that overcomes this limitation, through the use of photodetector arrays on thin elastomeric membranes, capable of reversible deformation into hemispherical shapes with radii of curvature that can be adjusted dynamically, via hydraulics. Combining this type of detector with a similarly tunable, fluidic plano-convex lens yields a hemispherical camera with variable zoom and excellent imaging characteristics. Systematic experimental and theoretical studies of the mechanics and optics reveal all underlying principles of operation. This type of technology could be useful for night-vision surveillance, endoscopic imaging, and other areas that require compact cameras with simple zoom optics and wide-angle fields of view.

Abstract:Hypobromous acid (HOBr) is profoundly implicated in many physiological and pathological conditions. In vivo, the production of HOBr is a double-edged sword for organisms. Appropriate amount of HOBr have effective antibacterial activity, however, abnormal production of HOBr can easily cause tissue damages. Unfortunately, the detailed cellular functions of HOBr are still unclear, which are mainly because highly specific and sensitive sensing of HOBr in living organism remains to be challenged. Based on that, a new naphthalimide-derived 1-(2aminoethyl)piperidine-containing highly selective fluorescent probe NAEP-HOBr for bioimaging endogenous HOBr was designed. Probe NAEP-HOBr possesses satisfactory sensitivity reflected in low detection limit (200 nM), pleasurable selectivity over others biological molecules, and rapid response for HOBr (similar to 1 min). Moreover, probe NAEP-HOBr is competent for tracking endogenous HOBr in living cells and zebrafish, which further provides an effective and powerful tool for tracking the fluctuations of endogenous HOBr in living organisms.

Abstract:We have developed a coherent Raman imaging platform using broadband coherent anti-Stokes Raman scattering (BCARS) that provides an unprecedented combination of speed, sensitivity, and spectral breadth. The system utilizes a unique configuration of laser sources that probes the Raman spectrum over 3,000 cm$^{-1}$ and generates an especially strong response in the typically weak Raman "fingerprint" region through heterodyne amplification of the anti-Stokes photons with a large nonresonant background (NRB) while maintaining high spectral resolution of $<$ 13 cm$^{-1}$. For histology and pathology, this system shows promise in highlighting major tissue components in a non-destructive, label-free manner. We demonstrate high-speed chemical imaging in two- and three-dimensional views of healthy murine liver and pancreas tissues and interfaces between xenograft brain tumors and the surrounding healthy brain matter.

Abstract:Hypobromous acid (HOBr), one of the significant reactive oxygen species (ROS) that acts as an important role in human immune system, however the increasing level of HOBr in human body can cause the disorder of eosinophils (EPO), leading to oxidative stress in organelles, and further causing a series of diseases. In this study, a ratiometric fluorescent probe DMBP based on Nile red skeleton was developed to detect HOBr specifically by the electrophilic substitution with HOBr. DMBP emits near-infrared (NIR) fluorescence at 653 nm, after reacting with HOBr, the emission wavelength of DMBP shifted blue and a new peak appeared at 520 nm, realizing a ratiometric examination of HOBr with a limit of detection of 89.00 nM. Based on its sensitive and specific response to HOBr, DMBP was applied in the visual imaging of HOBr in HepG2 cells and zebrafish. Foremost, probe DMBP has excellent lysosome targeting ability and NIR emission reduced the background interference of biological tissues, providing a potential analytical tool to further investigate the role of HOBr in lysosome.

Abstract:A new fluorescent probe SWJT-23 with lysosomal targeting ability for detection of hypobromous acid (HBrO) was synthesised based on the naphthalimide skeleton. This probe exhibited a fast response (within 3s), a low detection limit (1.24 nM), excellent selectivity and a high fluorescence quantum yield ( Φ = 0.490). Moreover, SWJT-23 not only realized the sensitive detection of HBrO in cells and water samples, but also was fabricated as a paper-based sensor. In consequence, SWJT-23 is expected to be an efficient and powerful tool for monitoring HBrO in organisms and the environment in realistic scenarios.

Abstract:As an essential reactive oxygen species (ROS), hypobromous acid (HOBr) plays a vital role in the immune process. Nevertheless, excessive HOBr can cause a series of diseases including cancers. Therefore, the development of the in situ detection methods for intracellular HOBr in biosystems is urgent and important to further uncover the detailed physiological and pathological effects of HOBr. In this work, a simple water-soluble near-infrared (NIR) fluorescence dye was reasonably designed and synthesized, which was satisfactory chosen as a highly selective and sensitive fluorescent probe for the rapid detection of HOBr based on the electrophilic bromination reaction mechanism. This probe can perform the specific detection of HOBr, which facilitates the accurate imaging of HOBr in biosystems. Finally, this probe was successfully applied to trace mitochondrial HOBr fluctuations with high resolution in living cells and zebrafish.

Abstract:Shortwave infrared (SWIR) imaging provides enhanced tissue penetration and reduced autofluorescence in clinical and pre-clinical applications. However, existing applications often lack the ability to probe chemical composition and molecular specificity without the need for contrast agents. Here, we present a SWIR imaging approach that visualizes spontaneous Raman scattering with remarkable chemical contrast deep within tissue across large fields of view. Our results demonstrate that Raman scattering overcomes autofluorescence as the predominant source of endogenous tissue background at illumination wavelengths as short as 892 nm. We highlight the versatility of SWIR Raman imaging through in vivo monitoring of whole-body tissue composition dynamics and non-invasive detection of fatty liver disease in mice, and identification of calcification and lipids in unfixed human atherosclerotic plaques. Moreover, our approach facilitates the visualization of nerves embedded in fatty tissue, a major advancement for surgical applications. With a simple wide-field setup orthogonal to fluorescence, SWIR Raman imaging holds promise for rapid adoption by clinicians and biologists. This technique opens new possibilities for contrast agent-free visualization of pathophysiology in whole animals and intraoperative imaging in humans.

Abstract:Large-scale synthesis of DNA and RNA is a crucial technology for modern biological research ranging from genomics to nucleic acid therapeutics and for technological research ranging from nanofabrication of materials to molecular-level writing of digital data. Maskless Array Synthesis (MAS) is a versatile and efficient approach for creating the required complex microarrays and libraries of DNA and other nucleic acids for these applications and, more generally, for the synthesis of sequence-defined engineered and biological oligomers. MAS uses digital photomasks displayed by a digital micromirror device (DMD) illuminated by an appropriate light source and imaged into a photochemical reaction chamber with an optical relay system. Previously, Offner relay systems were used for imaging, but modern DMD formats with more and smaller micromirrors favor a different solution. We present a desktop MAS optical system with the larger numerical aperture and larger field of view required by 1080p and other large-format DMDs. The resulting catadioptric relay is well suited to modern DMDs in this application, and is corrected for first order axial and lateral color, enabling the use of high-power LEDs as inexpensive and long-lasting light sources spanning the ultraviolet-to-violet to perform the required photochemistry. Additional characteristics of the system, including high contrast and low scatter, make it ideal for reducing the error rates in photochemical synthesis of biomolecules.

Rational construction of deep-red fluorescent probe for rapid detection of HClO and its application in bioimaging and paper-based sensing

Abstract:Hypochlorous acid (HClO), the core bactericidal substance of the human immune system, plays a vital role in many physiological and pathological processes in the human body. In this work, we designed and synthesized a novel deep-red fluorescent probe TCF-ClO for the determination of hypochlorous acid through theoretical analysis. The results showed that probe TCF-ClO exhibited excellent characteristics of long-wavelength emission (635 nm), fast response (< 1 min), and low detection limit (24 nM). In addition, it had been successfully used for imaging of HClO in living HeLa cells. More importantly, the TCF-ClO composited paper-based sensing material was successfully constructed. The RGB/gray value was obtained from a mobile phone and computer, which could achieve the quantitative detection of HClO, with a linear detection range of 0-50 μM and a detection limit of 1.09 μM (RGB mode)/3.38 μM (gray mode). The function of the paper-based sensor extended from qualitative to quantitative detection of HClO, and it is expected to become a portable device widely used in the environmental area.

Portable Electronic Endoscopic Imaging System

Abstract:The paper presents a low-power, inexpensive and portable endoscopic imaging system. A 1.3 million pixels CMOS sensor is considered as an image capture. The sensor and the lens system are designed to minify the cannula diameter of the endoscope and therefore minimize the incision size for insertion. LVDS is used for image data transmission between the sensor and CPU to realize a long distance, high speed and low noise system. An ARM 920T based microcontroller is employed as the control core for the image transmission module, display module and other modules. The camera interface and LCD controller are integrated in the microcontroller and both have a dedicated DMA supports to transmit image data though AHB to or from frame buffer located in system memory without CPU intervention. The image is displayed on an 8 inch LCD screen with 800 x 600 resolution and 16 bits of color depth. With the maximum capture and display rate of 15 fps, this system can provide a clear image enough for laparoscopy or industrial application. And with integrated camera, light source and video display function, it can also be used as a portable, miniature and inexpensive endoscope.

Abstract:Oral drug delivery is a preferred method for drug administration because it is economical and convenient to the patient [1]. Uptake from the gut to the blood is mediated by the intestinal barrier, which exhibits some selectivity, i.e., mechanisms in the intestinal barrier define which compounds are absorbed and which are not. The limited understanding of these mechanisms impedes the design and development of new oral drugs. On the horizon: Emerging organ-on-a-chip models of the intestine may provide new insights into structure-function relationships in biological barriers like the gut-blood interface of the intestine. These small ‘artificial organs’ can host biological cells, which mimic the natural behavior of intestinal cells, thus providing a highly controlled platform for fundamental bioresearch and, ultimately, drug-screening [2]. Our group develops and implements optical imaging technologies specifically geared for imaging organ-on-a-chip systems. At the SPIE conference “Optical Methods for Inspection, Characterization, and Imaging of Biomaterials V” we will present the application of two-photon microscopy for imaging of relatively thick 3D printed organ-on-a-chip systems and discuss imaging-challenges related to sample-induced aberrations and scattering. We will also present our ongoing work to implement an adaptive optics-enabled lattice light sheet microscope [3], which (we anticipate) will enable high-resolution imaging inside complex organ-on-a-chip systems by compensating the sample-induced aberrations. A laboratory-based X-ray phase contrast microscopy system for targeting in unstained soft-tissue samples

Abstract:We propose an imaging system and methodology for mapping soft-tissue samples in three dimensions, with micron-scale and isotropic spatial resolution, with low-concentrations as well as in the absence of heavy metal staining. We used hard X-ray phase-contrast imaging for the X-ray ability to non-destructively probe the internal structure of opaque specimens and for enhanced contrast obtained by exploiting phase effects, even in cases with reduced or absent staining agents. To demonstrate its applicability to soft-tissue specimens, we built a compact system that is easily deployable in a laboratory setting. The imaging system is based on a conventional rotating anode X-ray tube and a state-of-the-art custom-made radiation detector. The system's performance is quantitatively assessed on a calibration standard. Its potential for soft-tissue microscopy is demonstrated on two biological specimens and benchmarked against gold-standard synchrotron data. We believe that the approach proposed here can be valuable as a bridging imaging modality for intravital correlative light electron microscopy and be applied across disciplines where the three-dimensional morphology of pristine-condition soft tissues is a key element of the investigation.

Abstract:Visualization of isotope distribution within the living body has created important medical diagnostic procedures during the last decade. The static gamma-ray cameras reviewed here are often replacing the earlier mechanical scanners because of their ability to reduce picture exposure times: in extreme cases from minutes to tenths of seconds. Motion picture photography of changing isotope distribution can thus be used to follow many physiological processes which were previously a mystery, and these dynamic studies offer completely new diagnostic methods. This paper reviews the development of static gamma-ray cameras, discusses the problems of their design, and attempts to evaluate the limitations of each type. Methods of judging camera quality are discussed, and there is also brief mention of analytical methods for picture assessment.

Abstract:We describe the synthesis, properties, and application of two reversible fluorescent probes, mCy-TemOH and Cy-TemOH, for HOBr sensing and imaging in live cells. The two probes contain a hydroxylamine functional group for the monitoring of HOBr oxidation/ascorbic acid reduction events.
