Abstract:Algae could offer a potential source of fine chemicals, pharmaceuticals and biofuels. In this study, a green synthesis of dispersed cuboidal gold nanoparticles (AuNPs) was achieved using red algae, Gelidium amansii reacted with HAuCl4. It was found to be 4-7 nm sized cubical nanoparticles with aspect ratio of 1.4 were synthesized using 0.5 mM of HAuCl4 by HRSEM analysis. The crystalline planes (111), (200), (220), (311) and elemental signal of gold was observed by XRD and EDS respectively. The major constitutes, galactose and 3,6-anhydrogalactose in the alga played a critical role in the synthesis of crystalline AuNPs with cubical dimension. Further, the antibacterial potential of synthesized AuNPs was tested against human pathogens, Escherichia coli and Staphylococcus aureus. The synthesized AuNPs found biocompatible up to 100 ppm and high concentration showed an inhibition against cancer cell. This novel report could be helped to exploration of bioresources to material synthesis for the application of biosensor and biomedical application.
Biosynthesis of Gold Nanoparticles by the Extreme Bacterium Deinococcus Radiodurans and an Evaluation of Their Antibacterial Properties

Abstract:Deinococcus radiodurans is an extreme bacterium known for its high resistance to stresses including radiation and oxidants. The ability of D. radiodurans to reduce Au(III) and biosynthesize gold nanoparticles (AuNPs) was investigated in aqueous solution by ultraviolet and visible (UV/Vis) absorption spectroscopy, electron microscopy, X-ray diffraction (XRD), dynamic light scattering (DLS), Fourier transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS). D. radiodurans efficiently synthesized AuNPs from 1 mM Au(III) solution in 8 h. The AuNPs were of spherical, triangular and irregular shapes with an average size of 43.75 nm and a polydispersity index of 0.23 as measured by DLS. AuNPs were distributed in the cell envelope, across the cytosol and in the extracellular space. XRD analysis confirmed the crystallite nature of the AuNPs from the cell supernatant. Data from the FTIR and XPS showed that upon binding to proteins or compounds through interactions with carboxyl, amine, phospho and hydroxyl groups, Au(III) may be reduced to Au(I), and further reduced to Au(0) with the capping groups to stabilize the AuNPs. Biosynthesis of AuNPs was optimized with respect to the initial concentration of gold salt, bacterial growth period, solution pH and temperature. The purified AuNPs exhibited significant antibacterial activity against both Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) bacteria by damaging their cytoplasmic membrane. Therefore, the extreme bacterium D. radiodurans can be used as a novel bacterial candidate for efficient biosynthesis of AuNPs, which exhibited potential in biomedical application as an antibacterial agent.
Bioprospecting a Native Silver-Resistant Bacillus Safensis Strain for Green Synthesis and Subsequent Antibacterial and Anticancer Activities of Silver Nanoparticles.

Abstract:Green nanomaterials have gained much attention due to their potential use as therapeutic agents. The present study investigated the production of silver nanoparticles (AgNPs) from a silver-resistant Bacillus safensis TEN12 strain, which was isolated from metal contaminated soil and taxonomically identified through 16S rRNA gene sequencing. The formation of AgNPs in bacterial culture was confirmed by using UV-vis spectroscopy with an absorption peak at 426.18 nm. Fourier transform infrared (FTIR) spectroscopy confirmed the involvement of capping proteins and alcohols for stabilization of AgNPs. Moreover, X-ray diffraction analysis (XRD), scanning and transmission electron microscopy (SEM and TEM) confirmed the crystalline nature and spherical shape of AgNPs with particle size ranging from 22.77 to 45.98 nm. The energy dispersive X-ray spectroscopy (EDX) revealed that 93.54% silver content is present in the nano-powder. AgNPs showed maximum antibacterial activity (20.35 mm and 19.69 mm inhibition zones) at 20 mu g mL(-1) concentration against Staphylococcus aureus and Escherichia coli, respectively and significantly reduced the pathogen density in broth culture. Furthermore, AgNPs demonstrated significant anticancer effects in the human liver cancer cell line (HepG2) in MTT assay, whereas, no cytotoxic effects were demonstrated by AgNPs on normal cell line (HEK293). The present study suggests that the biogenic AgNPs may substitute chemically synthesized drugs with wider applications as antibacterial and anticancer agents. (C) 2020 The Authors. Biosynthesis, characterization, and antibacterial activity of gold nanoparticles

Abstract:Background: In recent decades focus of research has been toward an alternative antibacterial agent because of growing bacterial resistance and side effects of antibacterial agents. In the current study, the antibacterial activity of gold nanoparticles has been evaluated on selected human pathogens. Methods: In this study, we used panchagavya (PG) to synthesize gold nanoparticles, and the resulting nanoparticles (PG-AuNPs) were characterized by several spectroscopic techniques. In addition, antibacterial activity of PG-AuNPs against Escherichia coli, Bacillus subtilis, and Klebsiella pneumoniae were studied by well diffusion method. Results: The synthesis of AuNPs was affirmed by a colour change, which was further validated by UV-vis spectra with a maximum absorption peak at 527 nm. Bandgap energy was calculated as 2.13 eV by Tauc method from the UV result. The presence of amino acids and proteins in PG was responsible for the conversion of gold ions to AuNPs, according to FTIR analysis. (111), (200), (220), and (311) crystallographic planes were observed by XRD; further crystalline nature was validated by SAED analysis. The size and zeta value were found to be 53.29 nm and -9.8 mV respectively. Spherical shaped nanoparticles and elemental structure of PG-AuNPs were confirmed by HRTEM and EDS analysis. The antibacterial activity of PG-AuNPs showed the maximum and minimum zone of inhibition against K. pneumoniae (17.12 ± 0.14 mm) and B. subtilis (11.42 ± 0.58 mm). Conclusion: Antibacterial activity of PG-AuNPs was found to be strong against gram negative bacteria and moderate against gram positive bacteria. Based on the result, it was concluded that PG-AuNPs could be used to combat antibiotic drug resistance. Besides, in vitro and in vivo toxicity studies of PG-AuNPs should be conducted.
Biosynthesis and characterisations of silver nanoparticles with filamentous cyanobacterium Lyngbya sp. with in vitro antibacterial properties against MDR pathogenic bacteria

Abstract:This study describes phycocompounds of the non-N2-fixing filamentous cyanobacterium Lyngbya sp., which has potential bio-reducing and stabilizing heavy metal-accumulating properties for synthesizing silver nanoparticles (AgNPs), whose formation was confirmed by the colour change of the Lyngbya sp.-AgNP solution from pale green to deep brown. The reduction of 'Lyngbya sp.-AgNPs', called Lsp-AgNPs, was proved by UV-visible photo-spectrometry analysis with an obtained peak value at 426 nm. Lsp-AgNPs were characterised by analytical techniques, XRD, FESEM, DLS and FTIR. The XRD analysis with 5-70 theta was obtained at 2ϴ angles ranging from 38.79º with intensity, indicating the crystal structure of Lsp-AgNPs. The FESEM analysis indicated the area size at 20-50 µm; in the DLS analysis, the peak at 400 d nm indicated the size and distribution of Lsp-AgNPs. In FTIR analysis, the peaks were obtained at wavenumbers 3338, 1639, and 542 cm-1, which indicated the presence of N-H, -OH and C=O functional groups in Lsp-AgNPs. Those had in vitro antibacterial activities against Gram-negative Escherichia coli (MTCC 443) and Pseudomonas aeruginosa (MTCC 1688) and Gram-positive Staphylococcus aureus (MTCC 7443) bacterial strains with zone of inhibitions (ZOI) of 16, 12 and 14 mm, respectively, with comparing the antibiotic gentamycin as a positive control, as was monitored with agar-well diffusion method. Furthermore, the MIC value was 50 mg/ml, and MBC values of 65 mg/ml of Lsp-AgNPs were effective against those bacteria. Thus, Lsp-AgNPs had potential antibacterial activities against MDR pathogenic S. aureus, E. coli and P. aeruginosa. In conclusion, MDR pathogenic bacteria could be controlled as prodrugs in the future.
Green Synthesis of Gold Nanoparticles by Aqueous Extract of Zingiber officinale: Characterization and Insight into Antimicrobial, Antioxidant, and In Vitro Cytotoxic Activities

Abstract:The main challenge for researchers in the biomedical sectors concerns the development of new active compounds through cost-effectiveness, rapid, simple, and ecofriendly methods to overcome antibiotic resistance to pathogenic microbes. Herein, gold nanoparticles (AuNPs) were fabricated by a green approach through the reduction of chloroauric acid (HAuCl4) by harnessing the metabolites present in the aqueous extract of Zingiber officinale rhizome. UV-Vis spectroscopy, FT-IR, XRD, TEM, DLS, and zeta potential were used to characterize phytosynthesized AuNPs. In addition, the antimicrobial effect was investigated against Gram-positive bacteria (Bacillus subtilis and Staphylococcus aureus), Gram-negative bacteria (Pseudomonas aeruginosa and Escherichia coli), unicellular fungi (Candida albicans), and multicellular fungi (Aspergillus brasiliensis). Antioxidant and in vitro cytotoxic activities were assessed. TEM and XRD showed the successful formation of spherical and crystalline structures, with sizes in the range of 5–53 nm (average size: 15.11 ± 8.5 nm). The zeta potential value (i.e., –28.8 mV) explains the high stability of synthesized AuNPs. AuNPs exhibit promising activity against prokaryotic and eukaryotic microorganisms with variable inhibition zones and low MIC values in the range of 25–6.25 μg mL−1. Phytosynthesized AuNPs exhibit DPPH scavenging activity with percentages of 87.6 ± 0.5% at a maximum concentration (1000 μg mL−1), which can be compared with ascorbic acid (97.3 ± 0.2%). Moreover, the AuNPs displayed a target-oriented effect for cancer cell lines HepG2 and MCF7 at low IC50 concentrations of 131.9 ± 9.34 and 288.23 ± 31.39 μg mL−1 compared to the normal oral epithelial OEC cell line (487.612 ± 3.53 μg mL−1). Overall, the phytosynthesized AuNPs show wide activities that enable their use in various biomedical and biotechnological applications.

Abstract:The increase in bacterial infections and the emergence of antibiotic resistance has led to the development of bio-derived and environmentally friendly nanomaterials to counteract bacterial activity, and silver nanoparticles AgNPs are a promising way to achieve this. The present work aimed to synthesize AgNPs from the blue-green algae Athrospira Platensis and evaluate their antibacterial efficacy against Enterococcus faecalis, Staphylococcus aureus, Proteus mirabilis, and Pseudomonas aeruginosa isolated from human urinary tract infections. The results were shown and confirmed by peak UV spectra at 450. Through SEM and DLS analyses, the shapes of the AgNPs were determined to be spherical and with sizes ranging from 15 to 100 nm. The zeta potential values of AgNPs were found to be −20 ± 3.2 mV indicating the absence of AgNPs aggregates. Synthetic AgNPs produced by A. platensis showed a strong antibacterial effect against bacteria isolated from human urinary tract infections. The diameters of the inhibitory zone for S. aureus E. faecalis P. mirabilis and P. aeruginosa 14 mm, 16 mm, 15 mm, and 12 mm, respectively. AgNPs showed the highest activity against E. faecalis strain, with the diameter of the inhibitory zone reaching 16 ± 0.2 mm at a concentration of 800 μg/mL. These results will be a useful resource for studying the effects of man-made nanoparticles on the environmental safety and represent a promising step toward realizing AgNP-based antimicrobial drug production.

Abstract:The current study is designed to synthesize gold nanoparticles using Ajuga bracteosa extract, which is a highly known medicinal herb found in the northern Himalayas. The synthesized gold nanoparticles were initially characterized by UV-Vis spectrophotometer, SEM, FTIR, pXRD, and, GC-MS. Antibacterial efficacy of A . bracteosa extract, AuNps, and AuNps-free supernatant activity was checked against highly pathogenic clinical isolates of Escherichia coli , Staphylococcus aureus , and Pseudomonas aeruginosa via agar well diffusion method, assuming that supernatant might have active compounds. The Nps-free supernatant showed the maximum antibacterial activity against E . coli (20.8±0.3 mm), Staphylococcus aureus (16.5±0.5), and Pseudomonas aeruginosa (13±0.6). While green synthesized AuNps showed effective antibacterial activity ( Escherichia coli (16.4±0.3mm), Staphylococcus aureus (15.05±0.5mm), and Pseudomonas aeruginosa (11.07±0.6mm)) which was high compared to A . bracteosa extract. Anticancer activity was assessed by MTT assay on U87 and HEK293 cell lines. Aj-AuNps have an antigrowth effect on both the cell lines however Aj-AuNps-free supernatant which was also evaluated along with the Aj-AuNps, showed high toxicity toward HEK293 cell line compared to U87. Further, the GC-MS analysis of supernatant showed the presence of resultant toxic compounds after the reduction of gold salt, which include Trichloromethane, Propanoic acid, 2-methyl-, methyl ester, Methyl isovalerate, Pentanoic acid, 2-hydroxy-4-methyl-, Benzene-propanoic acid, and alpha-hydroxy. Based on the observation small molecular weight ligands of Ajuga bracteosa were analyzed in-silico for their binding efficacy towards selected membrane proteins of our target pathogens. RMSD is also calculated for the best docked protein ligand pose. The results revealed that among all listed ligands, Ergosterol and Decacetylajugrin IV have high virtuous binding affinities towards the membrane proteins of targeted pathogens. The current findings revealed that the Aj-AuNps are good antibacterial as well as anticancerous agents while the Nps-free supernatant is also exceedingly effective against resistant pathogens and cancer cell lines.

Abstract:Background: In recent years, there has been a notable increase in the level of attention devoted to exploring capabilities of nanoparticles, specifically gold nanoparticles AuNPs, within context of modern times. AuNPs possess distinct biophysical properties, as a novel avenue as an antibacterial agent targeting Streptococcus Mutans and Candida Albicans. The aim of this study to create a nano-platform that has the potential to be environmentally sustainable, in addition to exhibiting exceptional antimicrobial properties against Streptococcus Mutans as well as Candida Albicans. Methods: this study involved utilization of Pelargonium Graveolens leaves extract as a cost effective and environmentally sustainable approach for the green synthesis of AuNPs. Subsequently, physicochemical characteristics were assessed employing a variety of analytical methods, including as transmission electron microscopy, X-ray diffraction, Field Emission Scanning Electron Microscope, Zeta potential, Ultraviolet visible absorption spectroscopy, and Energy dispersive X-ray spectroscopy and Fourier transform infrared spectroscopy. The antimicrobial efficacy against Streptococcus Mutans and Candida Albicans was evaluated. Nanoparticles of various shapes, including hexagonal, spherical, semi-spherical, and triangular, were synthesized. These nanoparticles exhibited a mean particle size of 294nm and demonstrated low degree of aggregation. These nanoparticles exhibited long-term stability and were capable of facilely combining with diverse bioactive compounds. Results: The study demonstrated that AuNPs which is synthesized by green methods display potent antimicrobial properties. Conclusion: Utilization of Pelargonium Graveolens AuNPs may exhibit a promising potential as an antibacterial agent against Streptococcus Mutans and Candida Albicans. Nanoparticles (NPs) have the potential to serve as a novel approach for addressing pathogen infections as well as for biomedical, dental and pharmaceutical purposes in the future.

Abstract:The World Health Organization (WHO) reports that the emergence of multidrug-resistant and the slow advent of novel and more potent antitumor and antimicrobial chemotherapeutics continue to be of the highest concern for human health. Additionally, the stability, low solubility, and negative effects of existing drugs make them ineffective. Studies into alternative tactics to tackle such tenacious diseases was sparked by anticancer and antibacterial. Silver (Ag) and gold (Au) nanoparticles (NPs) were created from Trichoderma saturnisporum , the much more productive fungal strain. Functional fungal extracellular enzymes and proteins carried out the activities of synthesis and capping of the generated nano-metals. Characterization was done on the obtained Ag-NPs and Au-NPs through UV–vis, FTIR, XRD, TEM, and SEM. Additionally, versus methicillin-sensitive Staphylococcus aureus (MSSA) and methicillin-resistant Staphylococcus aureus (MRSA), Pseudomonas aeruginosa , and Klebsiella pneumoniae , the antibacterial activities of Ag-NPs and Au-NPs were assessed. In particular, the Ag-NPs were more effective against pathogenic bacteria than Au-NPs. Furthermore, antibiofilm study that shown Au-NPs had activity more than Ag-NPs. Interestingly, applying the DPPH procedure, these noble metallic NPs had antioxidant activity, in which the IC 50 for Ag-NPs and Au-NPs was 73.5 μg/mL and 190.0 μg/mL, respectively. According to the cytotoxicity evaluation results, the alteration in the cells was shown as loss of their typical shape, partial or complete loss of monolayer, granulation, shrinking, or cell rounding with IC 50 for normal Vero cell were 693.68 μg/mL and 661.24 μg/mL, for Ag-NPs and Au-NPs, respectively. While IC 50 for cancer cell (Mcf7) was 370.56 μg/mL and 394.79 μg/mL for Ag-NPs and Au-NPs, respectively. Ag-NPs and Au-NPs produced via green synthesis have the potential to be employed in the medical industry as beneficial nanocompounds.

Abstract:In recent times, gold nanoparticles (AuNPs) have attracted research attention owing to their versatility and physicochemical properties. The present study has been envisaged to investigate the photocatalytic potential of AuNPs synthesized using the cell-free supernatant of a potential probiotic strain Lactobacillus acidophilus against multi-drug-resistant (MDR) strains of enteroaggregative Escherichia coli (EAEC) and methicillin-resistant Staphylococcus aureus (MRSA). The AuNPs were synthesized in 60 min at ambient temperature using cell-free supernatant of L. acidophilus MTCC 10307. These probiosynthesized AuNPs were assessed by UV-diffuse reflection spectroscopy to obtain a characteristic peak at 565 nm. The X-ray diffraction analysis revealed the crystallinity of AuNPs, which was further confirmed using transmission electron microscopic imaging to reveal uniform-sized AuNPs (8.55 ± 1.11 nm) with spherical morphology. In an attempt to investigate the photocatalytic dye degradation potential, the probiosynthesized AuNPs completely degraded methylene blue within 60 min. Moreover, the AuNPs exhibited potential antimicrobial activity against MDR strains of EAEC and MRSA, as evidenced by the micro broth dilution technique. Furthermore, the probiosynthesized AuNPs revealed excellent photocatalytic disinfection properties (within 120 min) against the tested MDR-bacterial strains. In conclusion, the probiosynthesized AuNPs could serve as an effective photocatalyst which could further be employed for various environmental bioremediation processes, including wastewater treatment, as well as the removal of antibiotic and industrial pollutants.

Abstract:A tremendous increase in the green synthesis of metallic nanoparticles has been noticed in the last decades, which is due to their unique properties at the nano dimension. The present research work deals with synthesis mediated by the actinomycete Streptomyces tendae of silver nanoparticles (AgNPs), isolated from Little and Greater Rann of Kutch, India. The confirmation of the formation of AgNPs by the actinomycetes was carried out by using a UV-Vis spectrophotometer where an absorbance peak was obtained at 420 nm. The X-ray diffraction pattern demonstrated five characteristic diffraction peaks indexed at the lattice plane (111), (200), (231), (222), and (220). Fourier transform infrared showed typical bands at 531 to 1635, 2111, and 3328 cm−1. Scanning electron microscopy shows that the spherical-shaped AgNPs particles have diameters in the range of 40 to 90 nm. The particle size analysis displayed the mean particle size of AgNPs in aqueous medium, which was about 55 nm (±27 nm), bearing a negative charge on their surfaces. The potential of the S. tendae-mediated synthesized AgNPs was evaluated for their antimicrobial, anti-methicillin-resistant Staphylococcus aureus (MRSA), anti-biofilm, and anti-oxidant activity. The maximum inhibitory effect was observed against Pseudomonas aeruginosa at (8 μg/mL), followed by Escherichia coli and Aspergillus niger at (32 μg/mL), and against Candida albicans (64 μg/mL), whereas Bacillus subtilis (128 μg/mL) and Staphylococcus aureus (256 μg/mL) were much less sensitive to AgNPs. The biosynthesized AgNPs displayed activity against MRSA, and the free radical scavenging activity was observed with an increase in the dosage of AgNPs from 25 to 200 μg/mL. AgNPs in combination with ampicillin displayed inhibition of the development of biofilm in Pseudomonas aeruginosa and Streptococcus pneumoniae at 98% and 83%, respectively. AgNPs were also successfully coated on the surface of cotton to prepare antimicrobial surgical cotton, which demonstrated inhibitory action against Bacillus subtilis (15 mm) and Escherichia coli (12 mm). The present research integrates microbiology, nanotechnology, and biomedical science to formulate environmentally friendly antimicrobial materials using halotolerant actinomycetes, evolving green nanotechnology in the biomedical field. Moreover, this study broadens the understanding of halotolerant actinomycetes and their potential and opens possibilities for formulating new antimicrobial products and therapies.

Abstract:In this study, we report a green synthesis of pharmaceutically active gold nanoparticles from marine red alga Acanthophora spicifera by the reduction of chloroauric acid. The formation of A. spicifera-mediated gold nanoparticles (As-AuNPs) was characterized by several analytical techniques. The crystalline and face-centered cubic (fcc) structure were confirmed by X-ray diffraction (XRD) analysis. Electron microscopy results confirmed that As-AuNPs were spherical and the average size of particles was < 20 nm. As-AuNPs hold a significant level of antioxidant activities than A. spicifera extract. As-AuNPs exhibited the highest antibacterial activity against Vibrio harveyi than Staphylococcus aureus at 100 µg/ml. Furthermore, As-AuNPs exhibited the utmost cytotoxicity against human colon adenocarcinoma (HT-29) cells and registered the half-maximal inhibitory concentration (IC50) at 21.86 µg/ml. These findings authenticated that the synthesized As-AuNPs possess a broad spectrum of biological activities, and it can be effectively applied in the field of aquaculture and biomedical application.

Abstract:Biofilm development by bacteria is a serious issue in many different sectors, including the food, pharmaceutical, water, petroleum, and others. Because biofilms are difficult to remove due to their capacity to survive in bad environments, attempts were undertaken throughout this work to biologically manufacture gold nanoparticles with possible antibiofilm capabilities. During this study, 19 morphologically distinct fungal cultures were isolated from soil samples and used to synthesize gold nanoparticles. Among 19 fungi, 7 were producing gold nanoparticles and these particles were subjected to antimicrobial activity. Among 7 gold nanoparticles, nanoparticles synthesized from S2-10 isolate of fungi exhibited high antimicrobial activity toward biofilm-forming drug-resistant bacteria including Klebsiella pneumoniae (14.6 ± 0.57 mm), Aeromonas hydrophila (13.3 ± 0.57 mm) and Escherichia coli (10.0 ± 1.0 mm), and was used for further studies. The chosen nanoparticle also shown strong anti-biofilm action against test microorganisms on a variety of material substrates, including plastic, metal, and glass. These nanoparticles also exhibited inhibition of microcolony/initial biofilm, mature and preformed biofilm formed by Klebsiella pneumoniae (53.42 ± 4.45%, 62.14 ± 7.45%, and 24.12 ± 9.87%, respectively), Aeromonas hydrophila (43.89 ± 0.95%, 59.79 ± 1.19%, and 54.85 ± 6.50%, respectively) and Escherichia coli (45.28 ± 6.88%, 45.17 ± 3.94%, and 32.39 ± 4.16%, respectively). The physicochemical characterization of potential nanoparticles was conducted using UV-visible Spectrophotometer, X-ray diffractionSpectroscopy (XRD), Energy Dispersive X-ray (EDX) spectroscopy, Fourier Transform Infrared Spectroscopy (FTIR), and Scanning Electron Microscopy (SEM). These physicochemical analyses confirmed the formation of gold nanoparticles which were found to be spherical in shape with the size between 30 and 40 nm. Furthermore, the potential fungal isolate S2-10 was characterized as Aspergillus niger isolate CJ14 with the help of morphological and molecular techniques and deposited at National Center for Biotechnology Information (NCBI) (Accession number MN519438.1). Hence, it can be concluded that the gold nanoparticles produced using A. niger isolate CJ14 contain significant antibiofilm properties and can be used for developing antibiofilm products for future applications.

We now demonstrate that this structure is readily accessible by a solvent-free mechanochemical milling approach, highlighting the importance of mechanochemical reaction environments in evaluating chemical reactivity.
Bio-Synthesis of Aspergillus terreus Mediated Gold Nanoparticle: Antimicrobial, Antioxidant, Antifungal and In Vitro Cytotoxicity Studies

Abstract:Gold nanoparticles (GNP) were bio-fabricated utilizing the methanolic extract of the endophytic isolate Aspergillus terreus. The biosynthesised gold nanoparticles (GNP023) were characterised using UV-visible spectroscopy (UV-Vis); transmission electron microscopy (TEM), Fourier-transform nfrared spectroscopy (FTIR) and X-ray diffraction (XRD) studies. The bio-fabricated GNP023 displayed a sharp SPR peak at 536 nm, were spherically shaped, and had an average size between 10-16 nm. The EDX profile confirmed the presence of gold (Au), and XRD analysis confirmed the crystalline nature of GNP023. The antimicrobial activity of GNP023 was investigated against several food-borne and phytopathogens, using in vitro antibacterial and antifungal assays. The maximum zone of inhibition was observed for S. aureus and V. cholera at 400 μg /mL, whereas inhibition in radial mycelial growth was observed against Fusarium oxysporum and Rhizoctonia solani at 52.5% and 65.46%, respectively, when challenged with GNP023 (200 μg/mL). Moreover, the gold nanoparticles displayed significant antioxidant activity against the ABTS radical, with an IC50 of 38.61 µg/mL, and were non-toxic when tested against human kidney embryonic 293 (HEK293) cells. Thus, the current work supports the application of myco-synthesised gold nanoparticles as a versatile antimicrobial candidate against food-borne pathogens.

Abstract:Antimicrobial resistance (AMR) has become an alarming threat to the successful treatment of rapidly growing bacterial infections due to the abuse and misuse of antibiotics. Traditional antibiotics bear many limitations, including restricted bioavailability, inadequate penetration and the emergence of antimicrobial-resistant microorganisms. Recent advances in nanotechnology for the introduction of nanoparticles with fascinating physicochemical characteristics have been predicted as an innovative means of defence against antimicrobial-resistant diseases. The use of nanoparticles provides several benefits, including improved tissue targeting, better solubility, improved stability, enhanced epithelial permeability and causes minimal side effects. However, except for gold nanoparticles (AuNPs), the biological safety of the majority of metal nanoparticles remains a serious problem. AuNPs appear to be promising for drug delivery and medicinal applications because of their minimal toxicity, biocompatibility, functional flexibility, chemical stability and versatile biological activities, such as their antiviral, antifungal, anti-inflammatory and antimicrobial properties. Hence, we are focusing on the gold nanoparticles possessing antimicrobial activity in this article. This review will cover recent strategies in the preparation of gold nanoparticles, with special emphasis placed on antibiotics-coated AuNPs with enhanced antimicrobial properties and how they fight against disease-causing bacteria and eradicate biofilms, along with their activities and physicochemical properties.

Therapeutic effect of biosynthetic gold nanoparticles on multidrug-resistant Escherichia coli and Salmonella species isolated from ruminants

Abstract:Background and aim: Multidrug-resistant (MDR) pathogenic microorganisms have become a global problem in ruminants as a result of the intensive use of antibiotics, causing the development of resistance among gut microbiota. The antibiotic-resistant microorganisms can be transferred from diseased animals to humans. This study aimed to determine the prevalence of MDR Escherichia coli and Salmonella spp. isolated from cattle, buffaloes, sheep, and goats suffering from respiratory signs, diarrhea, and mastitis and to screen the antibiotic sensitivity of selected isolated bacteria. It also detected antibiotic-resistance genes by polymerase chain reaction (PCR), produced green gold nanoparticles (AuNPs) using plant extracts (Artemisia herba-alba and Morus alba), and evaluated the antimicrobial activities of these biosynthesized nanoparticles on selected pathogens (E. coli and Salmonella spp.). Materials and methods: MDR E. coli and Salmonella spp. were investigated using fecal samples (n=408), nasal swabs (n=358), and milk samples (n=227) of cattle, buffaloes, sheep, and goats with or without clinical signs, including respiratory manifestations, pneumonia, diarrhea, and mastitis, from different governorates in Egypt. E. coli and Salmonella spp. were isolated and identified on selective media, which were confirmed by biochemical reactions and PCR. Antimicrobial susceptibility testing against 10 commonly used antibiotics was performed using the Kirby-Bauer disk diffusion method. Antibiotic resistance genes blaTEM, blaSHV, blaOXA , and bla CTX-M were detected by PCR. The antibacterial effect of the biosynthesized AuNPs was evaluated by MIC and well diffusion assay. The biosynthesized AuNPs were also characterized by ultraviolet-visible spectrophotometry and transmission electron microscopy (TEM). Results: Among all fecal samples, the prevalence of E. coli was 18.4% (183/993) and that of Salmonella spp. was 16.7% (66/408), as determined by cultural and molecular tests. All isolates of E. coli and Salmonella spp. were 100% resistant to ampicillin (AM) and amoxicillin and highly resistant to cefoxitin and AM-sulbactam. The total rate of resistance genes in E. coli was 61.2% (112/183), while that in Salmonella was 63.6% (42/66) for pathogens isolated from ruminants with respiratory manifestations, pneumonia, diarrhea, and mastitis. Among the resistance genes, blaTEM had the highest prevalence rate in E. coli (25.9%, 21/81) while blaSHV had the lowest (9.8%, 8/81) in fecal swabs. AuNPs were successfully synthesized using aqueous leaf extract of A. herba-alba and M. alba as bioreducing agents. TEM analysis showed particle size of 10-42 nm for A. herba-alba and M. alba AuNPs. The biosynthesized AuNPs showed antibacterial activity against MDR E. coli and Salmonella spp. Conclusion: Rapid and accurate diagnostic methods are the cornerstone for effective treatment to reduce the risk of antimicrobial-resistant pathogenic microorganisms. This is particularly important for overcoming the increasing rate of MDR in ruminants with respiratory manifestations, pneumonia, diarrhea, and mastitis. This can be complemented by the development of AuNPs synthesized in an environmentally friendly manner AuNPs using natural plant extracts for the treatment of antibiotic-resistant microorganisms.

Abstract:Given the urgent need for novel methods to control the spread of multidrug-resistant microorganisms, this study presents a green synthesis approach to produce silver nanoparticles (AgNPs) using the bark extract from Anadenanthera colubrina (Vell.) Brenan var. colubrina. The methodology included obtaining the extract and characterizing the AgNPs, which revealed antimicrobial activity against MDR bacteria. A. colubrina species is valued in indigenous and traditional medicine for its medicinal properties. Herein, it was employed to synthesize AgNPs with effective antibacterial activity (MIC = 19.53–78.12 μM) against clinical isolates from the ESKAPEE group, known for causing high hospitalization costs and mortality rates. Despite its complexity, AgNP synthesis is an affordable method with minimal environmental impacts and risks. Plant-synthesized AgNPs possess unique characteristics that affect their biological activity and cytotoxicity. In this work, A. colubrina bark extract resulted in the synthesis of nanoparticles measuring 75.62 nm in diameter, with a polydispersity index of 0.17 and an average zeta potential of −29 mV, as well as low toxicity for human erythrocytes, with a CC50 value in the range of 961 μM. This synthesis underscores its innovative potential owing to its low toxicity, suggesting applicability across several areas and paving the way for future research.

Abstract:In this study, Cannabis sativa roots extract has been employed for the biosynthesis of silver nanoparticles (AgNPs). The appearance of reddish-brown colour followed by absorption peak of AgNPs at 408 nm through UV-vis spectrophotometry suggested biosynthesis of AgNPs. The size of the particles ranged from 90-113 nm, confirmed using DLS and TEM along with zeta potential of -25.3 mV. The FTIR provided information regarding the phytochemical capping. The study was further elaborated for determining AgNPs antibacterial, antioxidant, and cellular toxicity using MIC, DPPH, MTT, and haemolytic assays, respectively. The AgNPs were significantly effective against Staphylococcus aureus (Gram-positive), as compared to that of Pseudomonas aeruginosa, Klebsiella pneumoniae, and Escherichia coli (Gram-negative). AgNPs also exhibited remarkable antioxidant potential wherein 58.01 ± 0.09% free radical scavenging was observed at a concentration of 100 µg/ml. AgNPs revealed lower cytotoxicity where cell viability was observed to be 52.38 ± 0.6% at a very high concentration of 500 µg/ml in HEK 293 cells. Further, very low toxicity was seen in RBCs i.e. 6.47 ± 0.04% at a high concentration of 200 µg/ml. Thus, the current study beholds anticipation that Cannabis sativa ethanolic root extract-mediated AgNPs may play a vital role in therapeutic.
