Abstract:Dysregulation of metabolism is one of the cancer hallmarks. However, little is known whether long coding RNAs (lncRNAs) play a role in the regulation of metabolism in pancreatic ductal adenocarcinoma (PDAC). In the present study, we focused on lncRNA LINC00901 because a high level of LINC00901 is associated with the poor outcome of PDAC patients. Functional studies showed that LINC00901 promotes PDAC cell proliferation and invasion, whereas knockout (KO) of LINC00901 impairs these abilities of PDAC cells. Furthermore, the xenograft animal model suggested that LINC00901 promotes tumorigenesis. To determine the underlying mechanism of LINC00901-mediated tumorigenesis, we performed RNA-seq analysis in LINC00901 overexpression or KO cells. Gene Set Enrichment Analysis (GSEA) analysis showed that gene sets involved in glucose biological processes such as ‘‘MYC targets”, “Glycolysis” and “Gluconeogenesis” are significantly enriched in the LINC00901 overexpression group, whereas these genes are down-regulated in the LINC00901 KO group. The top-scoring genes recurring in the three gene sets include key glycolysis-related genes, lactate dehydrogenase A (LDHA), hexokinase 2 (HK2), and enolase 1 (ENO1), all of which were validated by qRT-PCR and western blot. Moreover, we found that LINC00901 upregulates c-Myc by stabilizing c-Myc RNA. Together, these results suggest that LINC00901 is an oncogenic lncRNA capable of regulating glucose metabolism, leading to PDAC development. Abstract:Abstract Background Mammary carcinomas have been associated with a high-fat diet, and the rate of breast cancer in overweight post-menopausal women is up to 50% higher than in their normal-weight counterparts. Epidemiological studies suggest that prolactin (PRL) plays a role in the progression of breast cancer. The current study examined breast cancer as a metabolic disease in the context of altered fatty acid catabolism by examining the effect of PRL on carnitine palmitoyl transferase 1 (CPT1), an enzyme that shuttles long-chain fatty acids into the mitochondrial matrix for β-oxidation. The effect of PRL on the adenosine 5'-monophosphate-activated protein kinase (AMPK) energy sensing pathway was also investigated. Methods MCF-7 and MDA-MB-231 breast cancer cells and 184B5 normal breast epithelial cells treated with 100 ng/ml of PRL for 24 hr were used as in vitro models. Real-time PCR was employed to quantify changes in mRNA levels and Western blotting was carried out to evaluate changes at the protein level. A non-radioactive CPT1 enzyme activity assay was established and siRNA transfections were performed to transiently knock down specific targets in the AMPK pathway. Results PRL stimulation increased the expression of CPT1A (liver isoform) at the mRNA and protein levels in both breast cancer cell lines, but not in 184B5 cells. In response to PRL, a 20% increase in CPT1 enzyme activity was observed in MDA-MB-231 cells. PRL treatment resulted in increased phosphorylation of the α catalytic subunit of AMPK at Thr172, as well as phosphorylation of acetyl-CoA carboxylase (ACC) at Ser79. A siRNA against liver kinase B1 (LKB1) reversed these effects in breast cancer cells. PRL partially restored CPT1 activity in breast cancer cells in which CPT1A, LKB1, or AMPKα-1 were knocked down. Conclusions PRL enhances fatty acid β-oxidation by stimulating CPT1 expression and/or activity in MCF-7 and MDA-MB-231 breast cancer cells. These PRL-mediated effects are partially dependent on the LKB1-AMPK pathway, although the regulation of CPT1 is also likely to be influenced by other mechanisms. Ultimately, increased CPT1 enzyme activity may contribute to fueling the high energy demands of cancer cells. Targeting metabolic pathways that are governed by PRL, which has already been implicated in the progression of breast cancer, may be of therapeutic benefit.

Abstract:Background: KRAS mutations are chief genetic event in pancreatic cancer. Recently approved inhibitors, sotorasib and adagrasib specifically target KRASG12C, a minor mutation in pancreatic cancer. Further, patients developed acquired resistance by generating secondary KRAS mutations, posing an added obstacle for allele specific inhibitors, warranting alternative strategies to tackle KRAS-dependent cancers. KRAS forms proteolipid signaling platforms on plasma membrane (PM) termed nanoclusters. We have shown mutant KRAS nanoclusters selectively enrich phosphatidylserine (PS) lipid, being particularly receptive to unsaturated PS, suggesting lipid acyl chain metabolism is crucial for KRAS function. In cells, lysophosphatidylcholine acyltransferases 3 (LPCAT3) preferentially catalyzes synthesis of unsaturated PS. We tested the hypothesis that inhibition of LPCAT3 depletes unsaturated PS in the PM, perturbs the PS-dependent PM interactions and oncogenic functions of KRAS. Methods: LPCAT3 knockdown (KD) mediated by siRNA in doxycycline (Dox) inducible KRASG12D murine cells (iKRAS) and CRISPR-Cas9 in human pancreatic tumor line (MiaPaCa-2). LPCAT3 inhibition achieved using small molecule inhibitors (SMIs): HTS3 and HTS4. Shotgun lipidomics characterized re-modeling of PS lipidomes. PM distribution and nanoclustering of KRAS was evaluated by super-resolution electron microscopy (EM). Anti-tumor effect of LPCAT3 inhibition was examined in vitro by proliferation, colony formation and soft agar assays and cellular signaling by western blot. Results: LPCAT3KD decreased polyunsaturated PS levels in MiaPaCa-2 and mislocalized mutant KRASG12V on the PM with a 4-fold reduction in KRAS PM distribution (p<0.0013) and disrupted KRAS nanoclustering, without affecting that of HRASG12V in transformed MiaPaCa-2. LPCAT3KD reduced colony formation ability in MiaPaCa-2. LPCAT3 siRNA reduced KRAS-dependent MAPK signaling, and significantly reduced colony formation only upon Dox-induced expression of KRASG12D, while having no effect in uninduced iKRAS. An integrated knowledgebase tool canSAR.ai generated a druggability score of 77% suggesting LPCAT3 is highly druggable by chemistry-based assessment. Indeed, recently discovered LPCAT3 inhibitors, significantly reduced MAPK signaling, proliferation and colony formation in mutant KRAS-dependent tumor cells while having no significant effect in WT KRAS cells corroborating mutant KRAS specificity of LPCAT3. Conclusion: We demonstrate targeting unsaturated PS lipid metabolism by inhibiting LPCAT3 disrupts KRAS signaling platform and perturbs its oncogenic functions in pancreatic cancer cells. Since various KRAS mutants require unsaturated PS to propagate signaling, our approach targets diverse KRAS mutants, uniquely addressing drug resistance issue. Abstract:Abstract Background Pancreatic ductal adenocarcinoma (PDAC) is a profoundly aggressive and fatal cancer. One of the key factors defining its aggressiveness and resilience against chemotherapy is the existence of cancer stem cells (CSCs). The important task of discovering upstream regulators of stemness that are amenable for targeting in PDAC is essential for the advancement of more potent therapeutic approaches. In this study, we sought to elucidate the function of the nuclear receptor subfamily 5, group A, member 2 (NR5A2) in the context of pancreatic CSCs. Methods We modeled human PDAC using primary PDAC cells and CSC-enriched sphere cultures. NR5A2 was genetically silenced or inhibited with Cpd3. Assays included RNA-seq, sphere/colony formation, cell viability/toxicity, real-time PCR, western blot, immunofluorescence, ChIP, CUT&Tag, XF Analysis, lactate production, and in vivo tumorigenicity assays. PDAC models from 18 patients were treated with Cpd3-loaded nanocarriers. Results Our findings demonstrate that NR5A2 plays a dual role in PDAC. In differentiated cancer cells, NR5A2 promotes cell proliferation by inhibiting CDKN1A. On the other hand, in the CSC population, NR5A2 enhances stemness by upregulating SOX2 through direct binding to its promotor/enhancer region. Additionally, NR5A2 suppresses MYC, leading to the activation of the mitochondrial biogenesis factor PPARGC1A and a shift in metabolism towards oxidative phosphorylation, which is a crucial feature of stemness in PDAC. Importantly, our study shows that the specific NR5A2 inhibitor, Cpd3, sensitizes a significant fraction of PDAC models derived from 18 patients to standard chemotherapy. This treatment approach results in durable remissions and long-term survival. Furthermore, we demonstrate that the expression levels of NR5A2/SOX2 can predict the response to treatment. Conclusions The findings of our study highlight the cell context-dependent effects of NR5A2 in PDAC. We have identified a novel pharmacological strategy to modulate SOX2 and MYC levels, which disrupts stemness and prevents relapse in this deadly disease. These insights provide valuable information for the development of targeted therapies for PDAC, offering new hope for improved patient outcomes. Pyroptosis-related Lncrna Pairs to Estimate the Molecular Features and Prognostic Outcomes of Pancreatic Ductal Adenocarcinoma.

Abstract:Pyroptosis is a form of programmed cell death associated with inflammatory alterations. However, the intrinsic mechanisms and underlying correlation of pyroptosis-related lncRNAs (PRLs) in pancreatic ductal adenocarcinoma (PDAC) remain unclear. The objective of the current research was to identify pyroptosis-related lncRNAs and a prognostic model to predict the prognosis of patients. We extracted pyroptosis-related lncRNAs to construct a risk model and validated them at Fudan University Shanghai Cancer Center. Crosstalk between lncRNA SNHG10 and GSDMD was found to regulate pyroptosis levels. A new algorithm was used to establish a 0 or 1 PRL pair matrix and prognostic model. Six pyroptosis-related lncRNA pairs were identified and utilized to construct a risk model. The low-risk groups exhibited better prognoses than the high-risk groups. The area under the curve (AUC) indicated extremely high accuracy, reaching 0.810 at 1 year, 0.850 at 2 years, and 0.850 at 3 years in the training set. Patients with different risk scores exhibited distinct metabolic, inflammatory, and immune microenvironments as well as tumor mutation landscapes. Additionally, 9 commonly used chemotherapeutic drugs exhibited different sensitivities between the high- and low-risk groups. To conclude, we propose that pyroptosis exhibits a close correlation with PDAC. Our risk model based on PRL pairs may be beneficial for the accurate estimation of prognostic outcomes, the immune microenvironment, and drug sensitivity, bringing therapeutic hope for patients with PDAC.

Abstract:New therapeutic targets are needed for pancreatic ductal adenocarcinoma (PDAC), a leading cause of cancer-related death worldwide. Phospholipase D (PLD) is a lipid-signaling enzyme that appears to play a critical role in cell proliferation, survival/apoptosis, and downstream oncogenic transformation. Elevated PLD1 or PLD2 (the two classical mammalian isoforms) activity has been shown to promote tumor progression in various cancer types, however, the role and mechanisms of PLD in pancreatic cancer are not yet understood. Thus, the objective is to determine the role of PLD1 regulation in pancreatic carcinogenesis through genetic and pharmacological inhibition. To evaluate the role of PLD1 in tumor growth, we subcutaneously transplanted human MIA PaCa-2 pancreatic cancer cells expressing endogenous PLD1 levels (Ctr KD cells), or MIA PaCa-2 cells in which PLD1 was knocked down (Pld1 KD cells), into immunodeficient nude mice. After 18 days post implantation, tumors that arose from Pld1-KD cells presented a significantly smaller tumor size (p<0.05) and had reduced levels of Bcl-xL and Bcl-2, compared to controls (Ctr KD). Then, to assess the role of PLD1 in the tumor microenvironment, we implanted KPC pancreatic cancer cells into C57BL/6 wild-type (WT) or C57BL/6 PLD1 knockout (Pld1−/−) mice with and without treatment with gemcitabine. Fourteen days after tumor implantation, the growth of KPC tumors was attenuated in Pld1−/− mice by 49% (p<0.05). Treatment with gemcitabine reduced tumor growth by 51.6% (p<0.05) in WT mice and by 79.5% (p<0.03) in Pld1−/− mice. Moreover, we evaluated the effects of PLD1 genetic ablation by crossing LSL-KrasG12D/+; Ptf1Cre/+ (KC) mice into PLD1 knockout (Pld1−/−) mice to generate KC; Pld1−/− mice. At 8 months of age, KC; Pld1−/− mice had significantly smaller pancreases (p<0.05) compared to KC mice. Interestingly, PLD1 ablation increased phosphorylation of ERK and c-Jun in KC; Pld1−/− mice compared to KC mice. Finally, to assess whether pharmacological inhibition of PLD can inhibit pancreatic carcinogenesis, KC mice received daily intraperitoneal injections of a small molecule inhibitor of PLD1 and PLD2 (FIPI) at a dosage of 3 mg/kg of body weight for a duration of 5 weeks. Treatment with FIPI reduced acinar-to-ductal metaplasia (p<0.05) and reduced cell proliferation (p<0.01), when compared to vehicle-treated KC controls. Additionally, FIPI treatment was safe demonstrating no significant differences in body weight between the groups and no significant alteration in the serum levels of liver enzymes, including aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase. In conclusion, these findings reveal PLD1 plays a critical role in pancreatic carcinogenesis and might represent a novel therapeutic target. Abstract:PRL1 and PRL3, members of the protein tyrosine phosphatase family, have been associated with cancer metastasis and poor prognosis. Despite extensive research on their protein phosphatase activity, their potential role as lipid phosphatases remains elusive. Methods: We conducted comprehensive investigations to elucidate the lipid phosphatase activity of PRL1 and PRL3 using a combination of cellular assays, biochemical analyses, and protein interactome profiling. Functional studies were performed to delineate the impact of PRL1/3 on macropinocytosis and its implications in cancer biology. Results: Our study has identified PRL1 and PRL3 as lipid phosphatases that interact with phosphoinositide (PIP) lipids, converting PI(3,4)P2 and PI(3,5)P2 into PI(3)P on the cellular membranes. These enzymatic activities of PRLs promote the formation of membrane ruffles, membrane blebbing and subsequent macropinocytosis, facilitating nutrient extraction, cell migration, and invasion, thereby contributing to tumor development. These enzymatic activities of PRLs promote the formation of membrane ruffles, membrane blebbing and subsequent macropinocytosis. Additionally, we found a correlation between PRL1/3 expression and glioma development, suggesting their involvement in glioma progression. Conclusions: Combining with the knowledge that PRLs have been identified to be involved in mTOR, EGFR and autophagy, here we concluded the physiological role of PRL1/3 in orchestrating the nutrient sensing, absorbing and recycling via regulating macropinocytosis through its lipid phosphatase activity. This mechanism could be exploited by tumor cells facing a nutrient-depleted microenvironment, highlighting the potential therapeutic significance of targeting PRL1/3-mediated macropinocytosis in cancer treatment.

Abstract:Abstract Pancreatic ductal adenocarcinoma (PDAC) is predicted to be the second leading cause of cancer death in the US by 2025, with rising incidence. Mutant KRAS in >90% of PDAC cases promotes activation of the MAPK pathway, yet targeting the MAPK pathway clinically has failed to improve patient outcomes. SHP2 canonically promotes KRAS activation and is essential for PDAC development in mouse models. Because targeting MAPK is insufficient, yet SHP2 loss blocks PDAC development, we hypothesized SHP2 has both MAPK dependent and independent functions. Initial studies revealed that SHP2 inhibition with SHP099 in murine KPC [K8484: KRASG12D/+, p53R172H/+, Pdx1Cre/+] cells required concentrations exceeding 50 μM to elicit a minimal ERK inhibition. This led us to predict that there may be a tumor suppressive mechanism of SHP2 inhibition that is KRAS/MAPK independent. We utilized available RNAseq data (NCBI GEO: PRJNA558508) of PDAC MiaPaCa2 cells treated with the MEK inhibitor, Trametinib, or SHP099 to compare the direct effects of inhibition on transcriptional outcomes. We identified inositol phosphate signaling as a candidate target, whose downstream consequence is Ca2+ flux. We measured IL-1β induced Ca2+ flux in a panel of human PDAC cells exposed to MEK or SHP2 inhibition. Trametinib treated cells showed no effect on Ca2+ flux, however, SHP099 led to a drastic depletion of free Ca2+ and response to stimulus. We then validated the Ca2+ response to IL-1β stimulus in Crispr-Cas9 SHP2-knockout K8484 cells. While we did not observe a difference in basal free calcium, we still found a reduction in response to IL-1β stimulus. Because Ca2+ is an important co-factor for mitochondrial metabolism, we then interrogated metabolic effects. We tested mitochondrial function using a Seahorse Mito-Stress test and found that while MEK inhibition failed to affect mitochondrial metabolism, SHP2 inhibition ablated it. A Glycolysis-Stress Test revealed that SHP2 inhibition led to glucose consumption primarily through anaerobic respiration. TMT-labeled phospho-proteomic analysis comparing SHP2KO and MEK inhibited K8484s revealed SHP2 dependent deficiencies in mitochondrial proteins, among others, involved in mitochondrial fission and lipid metabolism. Western diets have been shown to promote tumor progression in GEM models of PDAC, so we used KRASG12D/+, SHP2fl/fl or wt./wt, Ptf1aCre/+ (SKC and KC) mice to demonstrate that while diet-induced obesity affects KC tumor burden, the lack of SHP2 in SKCs prevents the diet associated discrepancy in tumor progression. Collectively, these data led us to conclude that while SHP2 has been studied as a contributor to KRAS signaling in PDAC, the outcomes of its role specifically on mitochondrial metabolism have yet to be appreciated or thoroughly investigated, opening the path for a new approach to its contribution to tumor progression. Abstract:Background: Reprogrammed cellular metabolism has been refocused on in the past decade, as many cancer cells rewire various metabolic pathways to facilitate their survival, unlimited cell growth, and division. RNA Polymerase II-Associated Factor 1 (PAF1)/Pancreatic Differentiation 2 (PD2) is a core subunit of the human PAF1 Complex (PAF1C) that regulates the RNA polymerase II function during transcriptional elongation in normal cells; however, its overexpression has been implicated in promoting pancreatic tumorigenesis and metastasis through epigenetic regulation, chemoresistance, radioresistance and maintenance of cancer stem cells. While most of these studies have provided evidence of the multifunctional nature of PAF1/PD2 in PDAC progression, no study has investigated the role of PAF1/PD2 during the metabolic rewiring of cancer. Methods: In this study, we sought to examine the role of PAF1/PD2 in the metabolic rewiring of cancer cells in pancreatic ductal adenocarcinoma (PDAC) and decipher how PAF1/PD2 mediates this metabolic reprogramming in PDAC. Pancreatic cancer cell lines were transfected with shRNAs to knock down PAF1/PD2. Metabolic genes regulated by PAF1/PD2 were identified by qPCR and western blot upon PAF1/PD2 depletion. Metabolic assays were performed to investigate the role of PAF1/PD2. Immunoprecipitations identified proteins that interact with PAF1/PD2. Confocal microscopy confirmed the co-localization of PAF1/PD2 with its protein partners. We performed chromatin immunoprecipitation (ChIP) - Polymerase chain reaction to confirm the binding of the PAF1 sub-complex to its target gene. Results: Our results showed that pancreatic cancer cells depleted PAF1/PD2 downregulate genes involved in aerobic glycolysis compared to control cells. Also, the lactate release assay indicated that more lactate was produced in control cells than in cells with PAF1/PD2 knockdown. Interestingly, we identified that HIF1α interacts with PAF1, specifically in pancreatic cancer cells. PAF1 and HIF1α co-localization was observed in pancreatic cancer cell lines. We also observed that the PAF1/PD2- HIF1α complex bound to the promoter region of LDHA to regulate the expression of LDHA in pancreatic cancer cells, reprogramming the metabolism to utilize the aerobic glycolysis pathway preferentially. Conclusions: In conclusion, our study shows that PAF1/PD2 rewires the metabolism of PDAC by interacting with HIF1 α to regulate the expression of LDHA, which is the rate-limiting step of aerobic glycolysis. Abstract:Insulin resistance is one of the major contributing factors in the development of metabolic diseases. The mechanisms responsible for insulin resistance, however, remain poorly understood. Although numerous functions of the prolactin receptor (PRLR) have been identified, a direct effect on insulin sensitivity has not been previously described. The aim of our current study is to investigate this possibility and elucidate underlying mechanisms. Here we show that insulin sensitivity is improved or impaired in mice injected with adenovirus that overexpress or knock down PRLR expression, respectively. Similar observations were obtained in in vitro studies. In addition, we discovered that the signal transducer and activator of transcription-5 pathway are required for regulating insulin sensitivity by PRLR. Moreover, we observed that PRLR expression is decreased or increased under insulin-resistant (db/db mice) or insulin-sensitive (leucine deprivation) conditions, respectively, and found that altering PRLR expression significantly reverses insulin sensitivity under both conditions. Finally, we found that PRLR expression levels are increased under leucine deprivation via a general control non-derepressible 2/mammalian target of rapamycin/ribosomal protein S6 kinase-1-dependent pathway. These results demonstrate a novel function for hepatic PRLR in the regulation of insulin sensitivity and provide important insights concerning the nutritional regulation of PRLR expression.

Abstract:Pancreatic ductal adenocarcinoma (PDAC) has a dismal 5-year survival of 12%. RNF43 is frequently mutated in pancreatic cystic neoplasms, including intraductal papillary mucinous neoplasms (IPMNs), which are precursor lesions of PDAC. RNF43 encodes for an E3 ubiquitin ligase and was initially identified as a negative Wnt regulator. To elucidate the role of RNF43-mutated neoplasms in an organ specific context, we have generated a genetically engineered mouse model which has pancreas specific loss of RNF43 (Hosein, Gastroenterology 2022). In this study we examine some of the metabolic dependencies created by Rnf43 loss on the background of mutant Kras, and how the Kras;Rnf43 model differs from the commonly used Kras;p53 model of PDAC. We have generated cell lines harboring mutant Kras and bi-allelic loss of Rnf43 from the resulting lesions in these mice (henceforth referred to as Kras;Rnf43 lines).Isogenic Kras;Rnf43 cell lines with re-expression of full-length Rnf43 (Kras;Rnf43-RNF43OE) were generated to confirm if observed phenotypes were Rnf43 dependent. Single-cell RNA sequencing (scRNA-seq) was performed for comparison between murine PDAC arising in Kras;Rnf43 versus Kras;p53 mice. We further compared metabolic parameters in Kras;Rnf43 and Kras;Rnf43-RNF43OE cells using the Seahorse platform. Mitochondrial ultrastructure by transmission electron microscopy (TEM) was analyzed in Kras;Rnf43 compared to Kras;p53 cells. To visualize endoplasmic reticulum (ER)-mitochondria contact sites (ERMCS), a split green fluorescent protein-based contact site sensor (SPLICS) was transfected and analyzed. Compared to Kras;p53 PDAC, Kras;Rnf43 tumors demonstrated upregulated mitochondrial transcripts and gene signatures of ER stress. Further, the ER stress response protein IRE1 was markedly elevated in Kras;Rnf43 cells versus Kras;p53 cells. The Kras;Rnf43 lines had enhanced mitochondrial oxidative phosphorylation (OXPHOS) activity and demonstrated evidence of enhanced mitochondrial quality control. ERMCS is a dynamic contact region between ER and mitochondria, which regulates biological processes including calcium homeostasis, ER stress and mitochondrial dynamics. The Kras;Rnf43 cells had significantly increased ERMCS compared to Kras;p53 cells. Finally, using IACS-010759, a small molecule inhibitor of OXPHOS, we demonstrate that Kras;Rnf43 cells had both increased in vitro sensitivity as well as in vivo susceptibility to tumor growth inhibition in orthotopic models, which translated into increased overall survival of mice. In conclusion, we provide evidence showing that bi-allelic loss of RNF43 is associated with increased ER stress and enhanced IRE1 dependent stress response. ER stress is mitigated via increased mitochondrial OXPHOS and ERMCS. Importantly, our results suggest that this creates a metabolic synthetic essentiality in the context of RNF43 loss, providing an avenue to suppress the progression of RNF43-mutated IPMNs, and potentially other solid cancers by targeting mitochondrial OXPHOS. Abstract:BackgroundThe abnormal expression of leucine-rich-alpha-2-glycoprotein 1 (LRG-1) is reported to be associated with multiple malignancies, but its role in the progression of pancreatic ductal adenocarcinoma (PDAC) remains to be determined.MethodsThe expression of LRG-1 was assessed in PDAC tissues by RT-PCR, Western blot and immunohistochemistry. LRG-1-silenced or overexpressed cell lines were constructed using shRNA or LRG-1-overexpressing plasmids. EdU incorporation assay, Transwell invasion and wound-healing assays were performed to evaluate the proliferation, invasion and migration of PDAC cells. In addition, protein expression of the mitogen-activated protein kinase (MAPK) pathway was detected using Western blot. Finally, Co-immunoprecipitation assay was conducted in search of the potential interaction between LRG-1 and epidermal growth factor receptor (EGFR).ResultsThe expression of LRG-1 in PDAC tissue was significantly higher than that in adjacent normal tissue, and high LRG-1 expression predicted poor survival and a late tumor stage. In addition, LRG-1 markedly promoted the viability, proliferation, migration and invasion of PDAC cells in vitro and facilitated tumor growth in vivo. More importantly, we revealed that these bioactivities of LRG-1 might result from its selective interaction with EGFR, which might further activate the p38/MAPK signaling pathways.ConclusionLRG-1 may prove to be a promising biomarker for predicting prognosis of PDAC patients. Inhibition of LRG-1 or its downstream pathway could be a potential therapeutic target for the treatment of PDAC.

Abstract:Mutational activation of the KRAS proto-oncogene is the initiating mutational event in pancreatic ductal adenocarcinoma (PDAC) and ~90% of patients harbor KRAS mutations. PDAC is marred by a 12% five-year overall survival rate, and developing effective therapeutic strategies remains a priority. Accumulating evidence suggests that all KRAS mutations are not created equal. While the KRASG12R mutation is rare in lung and colorectal cancers (<1%), it is the third most common KRAS mutation in PDAC, accounting for approximately 20% of all cases. However, KRASG12R cannot interact with the lipid kinase PI3Ka, a well-characterized RAS effector necessary for KRAS-driven tumorigenesis. Previous studies in mice models of cancer have demonstrated that ablation of the KRAS:PI3Ka interaction limits tumorigenesis. Despite the inability of KRASG12R to activate PI3Ka directly, AKT signaling is robustly activated in KRASG12R-mutant PDAC. The mechanisms that allow the KRASG12R mutant to overcome the inability to activate PI3K and promote PDAC are unclear. We recently developed a Ptf1a-CREERT2;KrasLSL-G12R genetically engineered mouse model to study KRASG12R in an in vivo context. This model does not develop pancreas lesions or tumors. We have uncovered two unique characteristics of human PDAC that we hypothesize allows for the KRASG12R mutation to produce tumors only in human pancreatic tissue. First, we have found that all four PI3K isoforms are overexpressed in human PDAC, and the PI3Kd and PI3Kg isoforms are specifically upregulated in KRASG12R-mutant PDAC. Second, PTEN is oxidized in PDAC. PTEN oxidation results in an intramolecular disulfide bond, which inhibits the phosphatase activity of PTEN and leads to hyperactivated PI3K signaling. Critically, mouse pancreas tissue only expresses the PI3Ka/b isoforms and PTEN is in the reduced state, demonstrating two significant differences between mouse models and human disease. Additionally, we have determined that PTEN becomes fully oxidized in nutrient-restricted medium (low glucose/glutamine), a common strategy employed to mimic the pancreatic tumor microenvironment in cell culture. Using PTEN oxidation-resistant variants, we demonstrate that KRASG12R-mutant PDAC cell lines are reliant on oxidized (inactivated) PTEN for proliferation. To confirm that PTEN inactivation can aid KRASG12R-mediated tumorigenesis in mouse models, we generated at KrasG12R/+;Ptenfl/fl genetically engineered mouse model, and the results of this model will be presented herein. Thus, our data demonstrate that increased PI3K isoform expression, coupled with PTEN oxidation, creates a unique environment that allows KRASG12R to initiate and promote pancreatic tumorigenesis. As PTEN oxidation overcomes the need for KRAS to activate PI3K signaling in PDAC, these results indicate that directly targeting KRAS alone will be insufficient at reducing tumor growth in humans and clinically successful therapeutic strategies will have to develop alternative methods to reduce PI3K signaling in addition to directly targeting KRAS activity. Abstract:Introduction: Acinar cells undergo de-differentiation after injury to a progenitor-like cell type with ductal characteristics termed acinar-to-ductal metaplasia (ADM). In the absence of oncogenic mutation, the ADM lesions resolve and reform the acinar compartment (we terms this as adaptive ADM). However, in the presence of oncogenic Kras mutations (we terms this as oncogenic ADM), acinar cells undergo neoplastic transformation after ADM and evolve to pancreatic intraepithelial neoplasia (PanIN), a well-known precursor of pancreatic ductal adenocarcinoma (PDAC). Herein, we explore the role of the Paired-Related Homeobox1 transcriptional factor (Prrx1) in adaptive ADM and the relationship between Prrx1 and mutant Kras in oncogenic ADM. Methods: We generated tamoxifen-inducible Ptf1aCreERT;Rosa26YFP/YFP (abbreviated as Prrx1cWT) mice and bred in the Prrx1fl/fl allele to make (abbreviated Prrx1cKO ) mice and subjected them to caerulin (CCK analog) induced acute pancreatitis, a well-estabished model. We created a time course for both mouse genotypes subjected to acute pancreatitis for histological analysis. Quantification of ADM was performed through automated cell-counting of immunofluorescence (IF) staining. Then, we performed 3D ex vivo acinar cultures of Prrx1cWT and Prrx1cKO mice. We used an exogenous adenoviral vector to induce Cre expression and knock out Prrx1, using an empty vector as a control. Dissociated acinar cell cultures in collagen were utilized for the evaluation of TGFβ signaling by western blotting (WB). Additionally, we generated novel Pdx1-Cre;LSLKrasG12D/+;Prrx1fl/fl;Rosa26YFP/YFP (KCY Prrx1 KO) mice, in which mutant Kras is efficiently expressed and Prrx1 is deleted in a pancreas-specific manner. KCY Prrx1 WT and KO mice were sacrificed at 3 months and 5 months for histological analysis. Results: IF staining revealed that Prrx1cKO mice had fewer ADM lesions compared to Prrx1cWT mice at Day 3 post-caerulein. Ex vivo cultures demonstrated a substantial increase of cystic structures, which are representative of ADM in Prrx1cWT cultures compared to Prrx1cKO cultures. Additionally, using a novel nucleofection protocol, we demonstrated that overexpression of the Prrx1b isoform (associated with a progenitor cell state) leads to a notable increase in ADM. WB of acinar protein lysates from either Prrx1cWT or Prrx1cKO mice showed significant differences in TGFβ levels, and a >2-fold higher expression of phosphorylated SMAD2 in Prrx1cWT mice. IF staining also showed that KCY Prrx1 KO mice had fewer ADM lesions compared to Prrx1 WT mice at at the 5 months time point. Conclusions: Prrx1 can influence adaptive ADM formation. Moreover, Prrx1 can influence ADM formation in the presence of oncogenic KRAS. Our data suggest that Prrx1 facilitates PDAC progression through ADM formation. Abstract:Abstract More than 90% of human pancreatic ductal adenocarcinoma (PDAC) cases are driven by activating mutations in KRAS. RMC-7977 is a potent inhibitor of GTP-bound RAS proteins (RAS(ON), including both wild type and mutant variants of KRAS, NRAS, and HRAS. The related investigational agent, RMC-6236, is a first-in-class, potent, orally bioavailable, RASMULTI(ON) inhibitor currently in Phase 1/1b clinical trials (NCT05379985). We performed preclinical studies in a range of models of PDAC, including the highly chemoresistant K-rasLSL.G12D/+, p53LSL.R172H/+, Pdx1Cretg/+ (KPC) genetically engineered mouse. RMC-7977 exhibited broad anti-tumor activity at tolerable doses, provoking radiographic responses in KPC pancreatic tumors and extending overall survival by 3-fold, the largest response observed yet in this model. RMC-7977 dosing produced a metronomic effect on RAS signaling in tumor and normal tissues. This yielded tumor-selective effects on apoptosis and proliferation, consistent with RAS oncogene addition in PDAC. PDAC tumors are characterized by an expansive stroma that harbors multiple subtypes of cancer associated fibroblasts (CAFs) that manufacture a fibrotic, inflammatory extracellular matrix. CAF proliferation is driven by signals emanating from the malignant epithelial cells, such as Sonic Hedgehog (SHH), a secreted ligand that drives downstream Hedgehog pathway signaling in myofibroblastic CAFs (myCAFs). To understand the impact of RAS-GTP inhibition on paracrine signaling to CAFs in PDAC, To understand the dynamic responses of malignant epithelial cells following GTP-RAS inhibition, we performed single cell RNA sequencing (scRNAseq) on KPC pancreatic tumors treated with RMC-7977 or vehicle at multiple timepoints. Using the ARACNe and VIPER algorithms, we performed single cell regulatory network analysis on the expression profiles of >210,000 cells to calculate the activities of thousands of transcriptional regulatory protein in each cell. We found that the RAS-GTP inhibition blocked SHH expression in malignant epithelial cells, resulting in loss of downstream Hh pathway activity in CAFs within 24 hours of treatment. By one week of treatment, myCAFs were significantly depleted whereas inflammatory CAFs (iCAFs) were significantly accumulated. Strikingly, CAF heterogeneity was restored in KPC pancreatic tumors that acquired resistance to RMC-7977, despite continued suppression of Hedgehog pathway signaling. Consistent with prior studies of Smoothened inhibitors, the depletion of myCAFs from RAS-GTP inhibition was associated with the rapid opening and hypersprouting of intra-tumoral blood vessels. Ongoing studies using the mutant-selective KRASG12D inhibitor RM-044 should discriminate between direct effects of wild type RAS inhibition in CAFs versus the paracrine consequences of mutant RAS inhibition in malignant epithelial cells. Abstract:KRAS is the most frequently mutated oncogene in pancreatic ductal adenocarcinoma (PDAC), with KRASG12D driving initiation and progression of ~40% of tumors. Mutant KRAS proteins exist predominantly in the GTP-bound (RAS(ON)) state, leading to excessive downstream signaling via interaction with effectors such as RAF kinases. RMC-9805 is an orally bioavailable, mutant-selective covalent inhibitor of the active state of KRASG12D. RM-044, used here, is the corresponding representative preclinical tool compound. RM-044, KRASG12D and cyclophilin A form a stable, high affinity tri-complex resulting in the disruption of interactions with downstream KRAS effectors and suppression of KRASG12D(ON) signaling. RM-044 decreased RAS pathway activity and inhibited proliferation in multiple human KRASG12D PDAC cell lines in vitro. While these data are encouraging, we sought to study models that more faithfully represent the heterogeneity and complex tumor microenvironment found in the tumors of patients with KRASG12D PDAC. To address this, we generated and characterized pancreatic organoids from the lox-STOP-lox-KrasG12D genetically engineered mouse model (GEMM) and introduced expression of the KrasG12D mutation and p53 loss (KP) through a vector containing a p53 sgRNA and Cre recombinase. GEMM-derived KP pancreatic organoid lines treated with RM-044 showed concentration-dependent proliferation inhibition in vitro. KP organoids were implanted orthotopically into mouse pancreas leading to the development of PDAC tumors in immunocompetent mice. Histopathological examination of KP organoid derived tumors revealed molecular and phenotypic features translatable to the tumors of patients with PDAC. In this orthotopic model, oral administration of RM-044 inhibited MAPK pathway activation and led to deep and durable tumor growth suppression at a well-tolerated dose. In summary, mutant-selective, covalent inhibition of KRASG12D(ON) inhibits cell proliferation and drives tumor growth suppression in both human and murine preclinical models of KRASG12D PDAC. The generation of preclinical models that allow for in-depth translational analyses of KRASG12D(ON) inhibition in vitro and in vivo can support identification of optimal therapeutic regimens. Collectively, these preclinical results indicate that direct targeting of KRASG12D(ON) in PDAC has the potential to translate into clinical benefit for patients with pancreatic cancer. Abstract:Abstract Pancreatic intraepithelial neoplasia (PanIN) is a precursor of pancreatic ductal adenocarcinoma (PDAC), which commonly occurs in the general populations with aging. Although most PanIN lesions (PanINs) harbor oncogenic KRAS mutations that initiate pancreatic tumorigenesis; PanINs rarely progress to PDAC. Critical factors that promote this progression, especially targetable ones, remain poorly defined. We show that peroxisome proliferator-activated receptor-delta (PPARδ), a lipid nuclear receptor, is upregulated in PanINs in humans and mice. Furthermore, PPARδ ligand activation by a high-fat diet or GW501516 (a highly selective, synthetic PPARδ ligand) in mutant KRAS G12D ( KRAS mu ) pancreatic epithelial cells strongly accelerates PanIN progression to PDAC. This PPARδ activation induces KRAS mu pancreatic epithelial cells to secrete CCL2, which recruits immunosuppressive macrophages and myeloid-derived suppressor cells into pancreas via the CCL2/CCR2 axis to orchestrate an immunosuppressive tumor microenvironment and subsequently drive PanIN progression to PDAC. Our data identify PPARδ signaling as a potential molecular target to prevent PDAC development in subjects harboring PanINs.

Abstract:Abstract The majority of pancreatic ductal adenocarcinoma (PDAC) patients present with late-stage, metastatic disease that is often resistant to therapeutics, resulting in the lowest survival rate of all major cancers. Metabolic rewiring in response to oncogenic signals plays a critical role in PDAC cell survival, tumor growth, and metastasis. In contrast to normal epithelial cells, these metabolic alterations make PDAC tumors dependent on glutamine for survival, highlighting a unique metabolic vulnerability that can be therapeutically exploited. However, during times of nutrient stress, PDAC cells can circumvent this vulnerability by engulfing extracellular fluids to replenish amino acids, including glutamine, in a process called, macropinocytosis. Macropinocytosis occurs downstream of oncogenic KRAS, a small GTPase that is almost universally mutated in PDAC patients. The inhibition of macropinocytosis in vivo reduces PDAC tumor growth, underscoring the importance of this pathway to cancer cell survival. However, the signaling mechanisms that regulate this process and the contribution of macropinocytic signaling to aggressive cancer phenotypes remains poorly understood. Protein phosphatase 2A (PP2A) is a heterotrimeric complex that regulates a wide variety of cell signaling pathways, including KRAS, and is commonly deregulated in human PDAC tumors. Recently, PP2A has been implicated as an important regulator of macropinocytosis, but the mechanism by which this occurs is unknown. We demonstrate that the genetic loss of the specific PP2A subunit, B56a, accelerates the formation of KRAS-driven pancreatic lesions in an in vivo mouse model, implicating this subunit as a critical negative regulator of KRAS phenotypes during PDAC progression. Here, we show that acute PP2A-B56a activation prevents the fusion of macropinosomes with lysosomes, robbing PDAC cells of critical metabolic nutrients and driving cell death. Consistent with these findings, PP2A activation also reduces glutamine levels and suppresses oxidative phosphorylation. Finally, we determined that PP2A activating compounds can function synergistically with metabolic inhibitors, supporting a new therapeutic strategy in this aggressive and deadly cancer. Together, our results implicate PP2A as a critical suppressor of PDAC metabolic plasticity and highlight the use of PP2A activating compounds to prevent PDAC nutrient scavenging. Abstract:Purpose[JP1] : Molecular phenotyping of PDAC [PJ2] has revealed an aggressive squamous/basal/quasi-mesenchymal[JP3] subtype, clinically presenting as highly metastatic and harboring genetic amplifications of KRAS. Our previous work indicated that a squamous identity gene, Parathyroid Hormone Like Hormone (PTHLH; encoding for the PTHrP protein), is frequently co-amplified with KRAS and skews PDAC towards a squamous identity. Further, PTHrP is secreted by tumor cells and promotes tumor growth and metastasis through autocrine signaling. However, pathological analysis of PTHrP-null tumors indicated that the tumor microenvironment (TME) was re-organized upon loss of PTHrP. Thus, we hypothesized that PTHrP is a tumor-derived factor that shapes the TME through its systemic secretion and mobilization of immunosuppressive myeloid cells. Methods: In vivo experiments used KrasG12D; p53R172H; Pdx1-Cre; YFP (KPCY) and KPCY-PthlhloxP/WT (KPCY-Pthlhhet) for spontaneous tumor formation in immune competent mice. Orthotopic implantation experiments used Pthlh overexpressing KPCY tumor cells (herein KPCY-PthlhOE) or empty vector control cells (KPCY-PthlhEV). Tumor immunophenotyping was done using flow cytometry. Cytokine expression was performed using qPCR and ELISA. Results: In vivo investigation of tumors from KPCY-Pthlhhet mice revealed less MDSC and greater CD8+ T cell infiltration than KPCY controls, demonstrating the necessity of PTHrP for immunosuppression[PJ4]. Orthotopic implantation of KPCY-PthlhOE cell lines reduced overall TME immune cell recruitment, while simultaneously increasing the proportion of polymorphonuclear myeloid-derived suppressor cells (PMN-MDSC) infiltration. A concomitant decrease in the frequency of T cells in the KPCY-PthlhOE tumors was also observed, showing the sufficiency of PTHrP to organize an immunosuppressive TME[PJ5]. Additionally, a reduction of PD-1+ and CD44high CD8+ T cells revealed they were less activated. Interestingly, recombinant human PTHrP was sufficient to induce MDSC mobilization in vivo.[JP6] To interrogate the mechanism of MDSC trafficking into the TME, cytokine production by was analyzed in vitro. KPCY-PthlhOE exhibited a pronounced increase and decrease of Cxcl1 and Cxcl10 expression, respectively, relative to the KPCY-EV control. Conclusions: The work herein describes a new mechanism for the mobilization and infiltration of immune suppressive MDSC in the tumor microenvironment, initiated by PTHrP. The PTHrP-dependent immunosuppressive TME excludes and inhibits effector immune cells, such as CD8+ T cells. This phenomenon has been described in squamous-like PDAC but may also apply to other squamous adenocarcinomas. [PJ7] Further elucidation of the mechanism by which PTHrP is the genesis of the immune suppressive TME may spur the development of novel treatments for these more clinically severe cancer subtypes.
