Abstract:Due to the disconnection of surviving neural elements after spinal cord injury (SCI), such patients had to suffer irreversible loss of motor or sensory function, and thereafter enormous economic and emotional burdens were brought to society and family. Despite many strategies being dealing with SCI, there is still no effective regenerative therapy. To date, significant progress has been made in studies of SCI repair strategies, including gene regulation of neural regeneration, cell or cell-derived exosomes and growth factors transplantation, repair of biomaterials, and neural signal stimulation. The pathophysiology of SCI is complex and multifaceted, and its mechanisms and processes are incompletely understood. Thus, combinatorial therapies have been demonstrated to be more effective, and lead to better neural circuits reconstruction and functional recovery. Combinations of biomaterials, stem cells, growth factors, drugs, and exosomes have been widely developed. However, simply achieving axon regeneration will not spontaneously lead to meaningful functional recovery. Therefore, the formation and remodeling of functional neural circuits also depend on rehabilitation exercises, such as exercise training, electrical stimulation (ES) and Brain–Computer Interfaces (BCIs). In this review, we summarize the recent progress in biological and engineering strategies for reconstructing neural circuits and promoting functional recovery after SCI, and emphasize current challenges and future directions.

Abstract:Spinal cord injury (SCI) is different from peripheral nerve injury; it results in devastating and permanent damage to the spine, leading to severe motor, sensory and autonomic dysfunction. SCI produces a complex microenvironment that can result in hemorrhage, inflammation and scar formation. Not only does it significantly limit regeneration, but it also challenges a multitude of transplantation strategies. In order to promote regeneration, researchers have recently begun to focus their attention on strategies that manipulate the complicated microenvironment produced by SCI. And some have achieved great therapeutic effects. Hence, reconstructing an appropriate microenvironment after transplantation could be a potential therapeutic solution for SCI. In this review, first, we aim to summarize the influential compositions of the microenvironment and their different effects on regeneration. Second, we highlight recent research that used various transplantation strategies to modulate different microenvironments produced by SCI in order to improve regeneration. Finally, we discuss future transplantation strategies regarding SCI.

Abstract:Spinal cord injury (SCI) is a debilitating condition with limited treatment options, necessitating the exploration of innovative regenerative approaches. Nanosystems have emerged as promising tools for neural tissue engineering and drug delivery in SCI. This review provides a concise overview of the current progress and future directions in nanosystems-enabled regenerative therapies for SCI. It highlights the underlying mechanisms of SCI, including pathophysiology, inflammatory response, and axonal degeneration. Various nanosystems, such as nanoparticles and nano-scaffolds, are discussed for their potential in promoting neural tissue regeneration, neuroprotection, and controlled drug delivery. Additionally, the challenges of clinical translation, safety considerations, and regulatory aspects are addressed, along with the assessment of biocompatibility and adverse effects. This review highlights the immense potential of nanosystems in SCI treatment and emphasizes the need for further research to overcome remaining hurdles and facilitate their clinical implementation.

Abstract:Spinal cord injury (SCI) is a tremendous disaster in a person’s life. It interrupts the brain–body neuronal circuits, resulting in functional deficits. Pathogenesis of SCI is a progressive and comprehensive event. In clinical trials, attempts to promote nerve regeneration and functional recovery after SCI have met with failures. Recently, with the development of transcriptome sequencing and biomaterials, researchers have struggled to explore novel efficient therapeutic treatments for SCI. Here, we summarize the recent progress that has been made in SCI repair based on the lesion microenvironment, neural circuits, and biomaterial scaffolds. We also propose several important directions for future research, including targeted-microRNA therapy, blood vessel interventions, and multiple treatment combinations. In short, we hope this review will enlighten researchers in the field and pave the way for SCI therapy.
Biomechanics of the Aortic Valve in the Continuous Flow VAD-Assisted Heart

Abstract:The biomechanics of the aortic valve are altered in patients with ventricular assist devices (VADs). During high VAD flow and low cardiac function, transvalvular pressure is high, and the aortic valve remains closed throughout the cardiac cycle. This condition has been linked to the development of aortic valve fusion and incompetence during VAD use. Thus, physicians try to maintain pulsatile flow to assure periodic valve opening. The aim of this study was to determine the extent of aortic valve opening and alterations in valve leaflet strain before and during VAD support using a specially designed mock loop. The results showed that diastole is prolonged during VAD use. In addition, there is a reduction in valve opening area, producing a VAD-related functional stenosis. The average leaflet strain increased during VAD support, primarily due to an increase in the minimum strain, during systole, rather than the maximum strain during diastole. The findings support our hypothesis that altered biomechanics in the VAD-assisted heart results in increased strain in the aortic valve leaflets, which can stimulate soft tissue remodeling. The implication for clinical use is that valve opening during parallel VAD flow is reduced compared with normal flow conditions. Consequently, current clinical practice for VAD patients may not be achieving sufficient valve opening to prevent changes such as fusion and incompetence.

Abstract:Axon degeneration and neurons after spinal cord injury (SCI) can lead to the serious sensory and motor dysfunctions. The effective treatment of SCI still remains a grand clinical challenge for current medicine. Development of biological scaffolds with drugs are of great significance to provide a supportive environment for re-establishment of cellular connections and growth of new tissues after SCI. In this study, we presented the synthesis of strontium-doped hydroxyapatite (Sr-HAP) with high biocompatibility and osteoconductivity, which was a promising biomaterial for repair of hard/soft tissues. The Sr-HAP nanoparticles was obtained by a classical precipitation process followed by the loading on macroporous chitosan (CS) scaffold. The CS/Sr-HAP nanocomposite was characterized with SEM, XRD, and FT-IR techniques, which was used to carry drugs for guiding nerve regeneration. We have performed the in vivo experiment by transplanting the scaffolds carried with chymopapain into an acute rat with SCI model. The robust neurogenesis of neural stem cells (NSCs) was observed, which then transformed into the functional neurons and the injured gap can be reconnected. This study suggests that the CS/Sr-HAP nanocomposite scaffold has great potential for promoting the spinal cord repair. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Spinal cord injuries (SCIs) are devastating. In SCIs, a powerful traumatic force impacting the spinal cord results in the permanent loss of nerve function below the injury level, leaving the patient paralyzed and wheelchair-bound for the remainder of his/her life. Unfortunately, clinical treatment that depends on surgical decompression appears to be unable to handle damaged nerves, and high-dose methylprednisolone-based therapy is also associated with problems, such as infection, gastrointestinal bleeding, femoral head necrosis, obesity, and hyperglycemia. Nanomaterials have opened new avenues for SCI treatment. Among them, performance-based nanomaterials derived from a variety of materials facilitate improvements in the microenvironment of traumatic injury and, in some cases, promote neuron regeneration. Nanoparticulate drug delivery systems enable the optimization of drug effects and drug bioavailability, thus contributing to the development of novel treatments. The improved efficiency and accuracy of gene delivery will also benefit the exploration of SCI mechanisms and the understanding of key genes and signaling pathways. Herein, we reviewed different types of nanomaterials applied to the treatment of SCI and summarized their functions and advantages to provide new perspectives for future clinical therapies.

Abstract:Spinal cord injuries lead to significant loss of motor, sensory, and autonomic functions, presenting major challenges in neural regeneration. Achieving effective therapeutic concentrations at injury sites has been a slow process, partly due to the difficulty of delivering drugs effectively. Nanoparticles, with their targeted delivery capabilities, biocompatibility, and enhanced bioavailability over conventional drugs, are garnering attention for spinal cord injury treatment. This review explores the current mechanisms and shortcomings of existing treatments, highlighting the benefits and progress of nanoparticle-based approaches. We detail nanoparticle delivery methods for spinal cord injury, including local and intravenous injections, oral delivery, and biomaterial-assisted implantation, alongside strategies such as drug loading and surface modification. The discussion extends to how nanoparticles aid in reducing oxidative stress, dampening inflammation, fostering neural regeneration, and promoting angiogenesis. We summarize the use of various types of nanoparticles for treating spinal cord injuries, including metallic, polymeric, protein-based, inorganic non-metallic, and lipid nanoparticles. We also discuss the challenges faced, such as biosafety, effectiveness in humans, precise dosage control, standardization of production and characterization, immune responses, and targeted delivery in vivo. Additionally, we explore future directions, such as improving biosafety, standardizing manufacturing and characterization processes, and advancing human trials. Nanoparticles have shown considerable progress in targeted delivery and enhancing treatment efficacy for spinal cord injuries, presenting significant potential for clinical use and drug development.

Abstract:Spinal Cord Injury (SCI) causes irreversible functional loss of the affected population. The incidence of SCI keeps increasing, resulting in huge burden on the society. The pathogenesis of SCI involves neuron death and exotic reaction, which could impede neuron regeneration. In clinic, the limited regenerative capacity of endogenous cells after SCI is a major problem. Recent studies have demonstrated that a variety of stem cells such as induced Pluripotent Stem Cells (iPSCs), Embryonic Stem Cells (ESCs), Mesenchymal Stem Cells (MSCs) and Neural Progenitor Cells (NPCs) /Neural Stem Cells (NSCs) have therapeutic potential for SCI. However, the efficacy and safety of these stem cellbased therapy for SCI remain controversial. In this review, we introduce the pathogenesis of SCI, summarize the current status of the application of these stem cells in SCI repair, and discuss possible mechanisms responsible for functional recovery of SCI after stem cell transplantation. Finally, we highlight several areas for further exploitation of stem cells as a promising regenerative therapy of SCI.

Abstract:Spinal cord injury (SCI) is a devastating event, and there are still no effective therapies currently available. Neural stem cells (NSCs) have gained increasing attention as promising regenerative therapy of SCI. NSCs based therapies of various neural diseases in animal models and clinical trials have been widely investigated. In this review we aim to summarize the development and recent progress in the application of NSCs in cell transplantation therapy for SCI. After brief introduction on sequential genetic steps regulating spinal cord development in vivo, we describe current experimental approaches for neural induction of NSCs in vitro. In particular, we focus on NSCs induced from pluripotent stem cells (PSCs). Finally, we highlight recent progress on the NSCs, which show great promise in the application to regeneration therapy for SCI.

Abstract:Purpose of this review: Manipulating or re-engineering the damaged human spinal cord to achieve neuro-recovery is one of the foremost challenges of modern science. Addressing the restricted permission of neural cells and topographically organised neural tissue for self-renewal and spontaneous regeneration, respectively, is not straightforward, as exemplified by rare instances of translational success. This review assembles an understanding of advances in nanomedicine for spinal cord injury (SCI) and related clinical indications of relevance to attempts to design, engineer, and target nanotechnologies to multiple molecular networks. Recent findings: Recent research provides a new understanding of the health benefits and regulatory landscape of nanomedicines based on a background of advances in mRNA-based nanocarrier vaccines and quantum dot-based optical imaging. In relation to spinal cord pathology, the extant literature details promising advances in nanoneuropharmacology and regenerative medicine that inform the present understanding of the nanoparticle (NP) biocompatibility–neurotoxicity relationship. In this review, the conceptual bases of nanotechnology and nanomaterial chemistry covering organic and inorganic particles of sizes generally less than 100 nm in diameter will be addressed. Regarding the centrally active nanotechnologies selected for this review, attention is paid to NP physico-chemistry, functionalisation, delivery, biocompatibility, biodistribution, toxicology, and key molecular targets and biological effects intrinsic to and beyond the spinal cord parenchyma. Summary: The advance of nanotechnologies for the treatment of refractory spinal cord pathologies requires an in-depth understanding of neurobiological and topographical principles and a consideration of additional complexities involving the research's translational and regulatory landscapes.

Prestack Elimination of Complex Multiples: a Gulf of Mexico Subsalt Example

Abstract:Summary Since the surface-related multiple elimination technique popularized by the DELPHI consortium is independent of subsurface structure, it can theoretically be used in any complex environment. In practice, however, the limited offset range of field data introduces time-, dip- and offsetdependent amplitude artifacts into the multiple model. This, plus the fact that sources and receivers are located below the water surface, make it impossible to extract a relevant wavelet, which is a critical step in the method. Consequently, this technique is not adaptive enough to provide adequate multiple attenuation. A new method has been developed that eliminates the need for wavelet extraction in the DELPHI technique. It utilizes the kinematically accurate multiple model and applies a pattern-recognition algorithm to remove the actual multiples. This adaptive technique offers optimum multiple attenuation without damaging the primaries. However, its effectiveness is still somewhat limited by the amplitude inaccuracies of the multiple model. In areas where first-order multiples can be isolated, it is possible to extract a posteriori the seismic wavelet using a simple matching filter between estimated and modeled multiples. Studies of these extracted wavelets have shown that they are indeed offset-dependent, confirming the limitations of the single wavelet approach. Thus, the pattern-recognition technique always gives better results than the costly non-linear wavelet inversion associated with the DELPHI technique.
Improving Functional Recovery after Severe Spinal Cord Injury by a Noninvasive Dual Functional Approach of Neuroprotection and Neuromodulation

Abstract:Abstract Despite tremendous unmet medical needs, there is no effective pharmacological treatment to promote functional recovery after spinal cord injury (SCI). Although multiple pathological events have been implicated in SCI, the development of a noninvasive pharmacological approach to simultaneously target the different mechanisms involved in SCI remains a formidable challenge. In this study, we report the development of a noninvasive nanodrug delivery system that consists of ROS-responsive amphiphilic copolymers and an encapsulated neurotransmitter-conjugated KCC2 agonist. We show that upon intravenous administration, the nanodrugs were able to enter the injured spinal cord due to blood spinal cord barrier disruption and ROS-responsive disassembly. Remarkably, once in the injured spinal cord, these nanodrugs exhibited dual functions: scavenging ROS accumulated in the lesion to protect spared connections and increasing neuronal excitability in the injured spinal cord through targeted delivery of the KCC2 agonist to inhibitory neurons. Thus, the noninvasive treatment led to significant functional recovery in the rats with contusive SCI. Together, these findings provide a much-needed translational pharmacological approach for treating severe SCI.

Abstract:Spinal cord is the major communicating system between the brain and the body, injury to the spinal cord will completely cut or damage the information and signal exchanges between brain and different parts of the body. Spinal cord injury (SCI) regulates the physical, cellular, and molecular function of the body. SCI could cause impairment in blood flow, breathing, temperature, body pressure and sensory appendages. Thus, serious steps must be taken to prevent and heal SCI in human body by severe damages and shocks. Nanoparticles are emerging in the field of biomedicine and gained attention as drug carriers, imaging, mediator for supportive chemical and accelerating the immunes cells. In recent time, nanoparticles are gaining interest towards improving or modifying the immune system for the treatment of SCI. The drug delivery efficacy of nanoparticles makes them easy to incorporate and load with molecules used for the treatment of SCI, also it enhances the recovery time by targeting localization, altering the signalling pathways and cellular uptake. Researchers are using nanoparticles with different synthesis methods, morphology, incorporating materials, stability and site of target. But exploring the ability of nanoparticles towards improving the healing mechanism, regeneration of SCI and accelerating the treatment time is the major necessity. Thus considering the lesser exposure of nanoparticles in the area of spinal cord injury and regeneration of cells, the present review give a brief knowledge about the different types of nanoparticles and its role in SCI therapy.

Abstract:Hydrogel and nanoparticles (NPs) are the most effective, widely studied therapeutic material strategies for spinal cord injury (SCI). Hydrogels can bridge the tissue gap and balance the tissue microenvironment, while NPs can effectively target payload drugs. This article comprehensively reviews the use of hydrogels and NPs in the repair and regeneration of SCI, to provide multimodal treatment strategies reference. Spinal cord injury (SCI) is a severe neurodegenerative disease caused by mechanical and biological factors, manifesting as a loss of motor and sensory functions. Inhibition of injury expansion and even reversal of injury in the acute damage stage of SCI are important strategies for treating this disease. Hydrogels and nanoparticle (NP)‐based drugs are the most effective, widely studied, and clinically valuable therapeutic strategies in the field of repair and regeneration. Hydrogels are 3D flow structures that fill the pathological gaps in SCI and provide a microenvironment similar to that of the spinal cord extracellular matrix for nerve cell regeneration. NP‐based drugs can easily penetrate the blood‐spinal cord barrier, target SCI lesions, and are noninvasive. Hydrogels and NPs as drug carriers can be loaded with various drugs and biological therapeutic factors for slow release in SCI lesions. They help drugs function more efficiently by exerting anti‐inflammatory, antioxidant, and nerve regeneration effects to promote the recovery of neurological function. In this review, the use of hydrogels and NPs as drug carriers and the role of both in the repair of SCI are discussed to provide a multimodal strategic reference for nerve repair and regeneration after SCI.

Abstract:Abstract Spinal cord injury (SCI) disrupts the structural and functional connectivity between the higher center and the spinal cord, resulting in severe motor, sensory, and autonomic dysfunction with a variety of complications. The pathophysiology of SCI is complicated and multifaceted, and thus individual treatments acting on a specific aspect or process are inadequate to elicit neuronal regeneration and functional recovery after SCI. Combinatory strategies targeting multiple aspects of SCI pathology have achieved greater beneficial effects than individual therapy alone. Although many problems and challenges remain, the encouraging outcomes that have been achieved in preclinical models offer a promising foothold for the development of novel clinical strategies to treat SCI. In this review, we characterize the mechanisms underlying axon regeneration of adult neurons and summarize recent advances in facilitating functional recovery following SCI at both the acute and chronic stages. In addition, we analyze the current status, remaining problems, and realistic challenges towards clinical translation. Finally, we consider the future of SCI treatment and provide insights into how to narrow the translational gap that currently exists between preclinical studies and clinical practice. Going forward, clinical trials should emphasize multidisciplinary conversation and cooperation to identify optimal combinatorial approaches to maximize therapeutic benefit in humans with SCI.

Abstract:The effective regeneration and functional restoration of damaged spinal cord tissue have been a long-standing concern in regenerative medicine. Treatment of spinal cord injury (SCI) is challenging due to the obstruction of the blood-spinal cord barrier (BSCB), the lack of targeting of drugs, and the complex pathophysiology of injury sites. Lipid nanovesicles, including cell-derived nanovesicles and synthetic lipid nanovesicles, are highly biocompatible and can penetrate BSCB, and are therefore effective delivery systems for targeted treatment of SCI. We summarize the progress of lipid nanovesicles for the targeted treatment of SCI, discuss their advantages and challenges, and provide a perspective on the application of lipid nanovesicles for SCI treatment. Although most of the lipid nanovesicle-based therapy of SCI is still in preclinical studies, this low immunogenicity, low toxicity, and highly engineerable nanovesicles will hold great promise for future spinal cord injury treatments.

Abstract:Spinal cord injury (SCI) is damage to the spinal cord due to trauma or health conditions, resulting in lesions in the spinal cord. Currently, available treatment includes surgical intervention to decompress or stabilize a dislocated loose spine, steroid drugs to reduce inflammation, and subsequent rehabilitation. As there is a rising number of SCI globally, radical treatments to recover spinal cord functions have become highly anticipated. The development of new treatments is indeed progressing. Various therapeutic drug candidates are being developed in clinical trials, including neuroprotective/neurotrophic factors, antibodies for repulsive guidance molecules, and cell transplantation. Among them, with advances in stem cell biology, cell transplantation therapy is currently a promising therapeutic development for SCI. In particular, there have been various reports regarding the realization of regenerative medicine using human induced pluripotent stem cells (iPSCs). This review will introduce the advantages of cell-based therapy based on iPSC-derived neural stem/progenitor cells (iPSC-NS/PCs) and some of their mechanisms of action for functional improvement, which have recently been elucidated. Potential challenges and methodologies to realize the clinical application of iPSC-NS/PCs not only for the subacute phase but also for the chronic phase of SCI will be presented. Finally, we also introduce recent research with a view to the clinical application of spinal cord regenerative therapy and discuss future prospects.
