Abstract:The Multi-Ion Toxicity (MIT) Model uses electrochemical theory to predict the transepithelial potential (TEP) across the gills as an index of major ion toxicity in freshwater animals. The goal is to determine environmental criteria that will be protective of aquatic organisms exposed to salt pollution. In recent studies, TEP disturbances above baseline (ΔTEP) during short-term exposures to major ions have been proven as indicative of their toxicity to fish, in accord with the MIT model. However, the acute 1-h exposures used in these previous studies might not be realistic relative to the 24 h or 96 h test periods used for toxicity assessment. To address this temporal inconsistency, the current study investigated both the TEP responses to serial concentrations of 10 major salts (NaCl, Na2SO4, NaHCO3, KCl, K2SO4, KHCO3, CaCl2, CaSO4, MgCl2, MgSO4) and plasma ion levels in juvenile rainbow trout after they had been pre-exposed to 50% of the 96h-LC50 levels of these same salts for 4 days. The pre-exposures caused no mortalities. In general, plasma ions (Na+, K+, Ca2+, Mg2+, Cl-) were well-regulated; however, pre-exposure to sulfate salts resulted in the greatest number of alterations in plasma ion levels. TEP responses remained largely similar to those of naïve trout (without salt pre-exposure). All salts caused hyperbolic concentration-dependent increases in TEP that were well-described by the Michaelis-Menten equation. In the pre-exposed trout, the variation of ∆TEP at the 96h-LC50 concentrations was only 2.2-fold, compared to nearly 28-fold variation among the molar concentrations of the various salts at the 96h-LC50s, identical to the conclusion for naïve trout. Overall, the results remove the temporal inconsistency of previous tests and remain supportive of the MIT model. In addition, the recorded alterations in certain plasma ions, baseline TEP, and Michaelis-Menten constants improve our knowledge on specific physiological responses after extended major ion exposure.

Abstract:The acute toxicity of organic pollutants to fish, Daphnia magna, Tetrahymena pyriformis, and Vibrio fischeri was investigated. The results indicated that the Toxicity Ratio (TR) threshold of log TR =1, which has been based on the distribution of toxicity data to fish, can also be used to discriminate reactive or specifically acting compounds from baseline narcotics for Daphnia magna and Vibrio fischeri. A log TR=0.84 is proposed for Tetrahymena pyriformis following investigation of the relationships between the species sensitivity and the absolute averaged residuals (AAR) between the predicted baseline toxicity and the experimental toxicity. Less inert compounds exhibit relatively higher toxicity to the lower species (Tetrahymena pyriformis and Vibrio fischeri) than the higher species (fish and Daphnia magna). A greater number of less inert compounds with log TR greater than the thresholds was observed for Tetrahymena pyriformis and Vibrio fischeri. This may be attributed to the hydrophilic compounds which may pass more easily through cell membranes than the skin or exoskeleton of organisms and have higher bioconcentration factors in the lower species, leading to higher toxicity. Most of classes of chemical associated with excess toxicity to one species also exhibited excess toxicity to other species, however, a few classes with excess toxicity to one species exhibiting narcotic toxicity to other species and thus may have different MOAs between species. Some ionizable compounds have log TR much lower than one because of the over-estimated log KOW. The factors that influence the toxicity ratio calculated from baseline level are discussed in this paper.
Subtypes and One-Year Survival of First-Ever Stroke in Chinese Patients: The Nanjing Stroke Registry

Abstract:Background: Morbidity and mortality of stroke have been investigated extensively in Western populations, while data concerning case fatality and cause of death after stroke are very limited in mainland China. This study aimed to analyze the 1-year survival and predictors of case fatality in Chinese patients with first-ever stroke. Methods: Subjects are patients registered in the Nanjing Stroke Registry Program. Information concerning cardiovascular risk factors and stroke characteristics were collected, and patients were followed after registration. Ischemic strokes were classified according to TOAST criteria as large-artery atherosclerosis (LAA), cardiac embolism stroke (CES), small-vessel stroke (SVS), or other determined and undetermined causes (UND). One-year case fatality was analyzed by the Kaplan-Meier method, and predictors of case fatality were evaluated by the Cox proportional hazards model. Results: A total of 752 patients with first-ever stroke were included, of which 142 (18.9%) were identified as intracerebral hemorrhage (ICH), 120 (16.0%) as LAA, 123 (16.4%) as SVS, 160 (21.3%) as CES and 216 (28.7%) as UND. The overall survival rate was 86.4% at the end of the 1-year follow-up. Patients with SVS have the highest survival rate (92.7%), followed by UND (89.4%), CES (88.1%) and LAA (84.2%). Patients with ICH have the lowest survival rate (76.8%). Survival rates of patients with different subtypes of stroke presented a significant difference (χ2 = 19.3, p < 0.001). For patients deceased during the first year after the index stroke, 33.3% of deaths were caused by the first stroke, 18.6% by recurrent stroke, 16.7% by cardiovascular comorbidities, 14.7% by nonvascular conditions and 16.7% died of undetermined causes. Advanced age, hypertension, hyperlipidemia, diabetes mellitus (DM), atrial fibrillation (AF), history of transient ischemic attack and cigarette smoking were associated with an increased risk of death 1 year after stroke. Conclusions: The case fatality rate and predictors for mortality of Chinese patients with first-ever stroke are similar to those reported for other populations. The significant influence of cardiovascular disease on the first-year survival rate emphasizes the importance of acute stroke management and control of hypertension, DM, AF and other predictors for decreasing case fatality and improving prognosis.
Revealing the toxicity of monovalent and trivalent thallium to medaka fish in controlled exposure conditions

Abstract:Thallium (Tl) is a rare earth element increasingly being used in high-technology manufacturing. It is also an emerging pollutant with high exposure and toxicity risks to aquatic ecosystems. Tl exists in the environment in a monovalent [thallous, Tl(I)] or trivalent [thallic, Tl(III)] state. Currently, the stability of the two Tl species in natural water is uncertain and the toxicity in algae and daphnia are inconsistent due to lack of robust characterization of Tl species and matrix effects, while studies with fish are sparse. In this study, larvae of medaka fish (Oryzias latipes) were dosed with environmentally relevant concentrations of Tl(I) or Tl(III) spiked into synthetic and natural river water for 7 days to observe the toxic effects of two Tl species on fish. The transformation of Tl(I) and Tl(III) in water was analyzed by high performance liquid chromatography coupled with inductively coupled plasma and mass spectrometry. Analytical and toxicity results showed that Tl(I) is more stable presenting higher mortality and bioconcentration in medaka than Tl(III) in different water matrices. Tl(I)-induced LC50 and body burden in treated fish were highly correlated with its competitive ion, potassium (K), especially in waters containing medium K levels. This study provides reliable evidence regarding the stability and toxicity of Tl(I) and Tl(III) as well as the interaction of aqueous K versus Tl(I) in fish. Such information is useful for justifying water-quality guidelines and ecological risks of Tl pollution in natural water ecosystems.

Abstract:Triphenyltin (TPT) has attracted considerable attention owing to its vitality, bioaccumulation, and lurking damage. TPT widely exists in complex salinity areas such as estuaries and coastal regions. However, there are few studies on the toxicological behavior of TPT under different salinity. In the study, juvenile Nile tilapia (Oreochromis niloticus) were utilized as model animals to investigate the effects of environmental relevant TPT exposure on the osmoregulation and energy metabolism in gill under different salinity. The results showed that salinity and TPT single or combined exposure affected the morphology of the gill tissue. After TPT exposure, Na+-K+-ATPase (NKA) activity significantly decreased at 0 ppt, while NKA and Ca2+-Mg2+-ATPase (CMA) activities significantly increased at 15 ppt. In addition, significantly higher succinate dehydrogenase (SDH) and lactate dehydrogenase (LDH) activities were found in the control fish compared to the TPT-exposed ones at 15 ppt. Quantitative real-time PCR results showed that TPT exposure affected the expression of osmoregulation and energy metabolism-related genes under different salinity. Overall, TPT exposure interfered with osmoregulation and energy metabolism under different salinity. The study will provide reference data for assessing the toxicity of organotin compounds in complex-salinity areas.

Role of the luminal composition on intestinal metal toxicity, bioavailability and bioreactivity: An in vitro approach based on the cell line RTgutGC

Abstract:The bioavailability of metal complexes is poorly understood. To evaluate bioavailability and toxicity of neutral and charged complexes as well as free metal ions, Visual Minteq, a chemical equilibrium model, was used to design media containing different metal species. Two non-essential (silver and cadmium) and two essential (copper and zinc) metals were selected. The rainbow trout (Oncorhynchus mykiss) gut cell line (RTgutGC) was used to investigate bioavailability, bioreactivity and toxicity of the different metal species. Toxicity was measured using a multiple endpoint cytotoxicity assay, bioavailability by measuring intracellular metal concentration, and bioreactivity by quantification of mRNA level of the metal responsive genes, metallothionein (MT), glutathione reductase (GR) and zinc transporter 1 (ZnT1). Speciation calculations showed that silver and cadmium preferentially bind chloride, copper phosphate and bicarbonate, and zinc remained primarily as a free ion. Cysteine avidly complexed with all metals reducing toxicity, bioavailability and bioreactivity. Silver and copper toxicity was not affected by inorganic metal speciation, whereas cadmium and zinc toxicity was decreased by chloride complexation. Moreover, reduction of calcium concentration in the medium increased toxicity and bioavailability of cadmium and zinc. Bioavailability of silver and zinc was reduced by low chloride while cadmium bioavailability was increased by low chloride and in presence of bicarbonate. Copper bioavailability was not affected by the medium composition. Cadmium and silver were more bioreactive, independently from the medium composition, in comparison to copper and zinc (i.e., higher induction of MT and GR). Cadmium was the only metal able to induce MT in presence of cysteine. ZnT1 was induced by cadmium in low-chloride, by zinc in low-chloride low-calcium and by cadmium and copper in the bicarbonate media. Overall, this study demonstrates that metal complexation alone is not sufficient to explain metal toxicity, and that anion exchange mechanisms play a role in metal uptake and bioreactivity.

Abstract:The increasing salinization of freshwater streams from anthropogenic land uses and activities is a growing global environmental problem. Increases in individual ions (such as sodium or chloride) and combined measures such as total dissolved solids (TDS) threaten drinking water supplies, agricultural and economic interests, and the ecological health of freshwater streams. Because the toxicity of high ionic strength waters depends on the specific ion composition, few water quality standards exist to protect freshwater streams from salinization. In this study, we used a novel approach to develop site‐specific and ecologically‐relevant TDS thresholds for the protection of aquatic life. The first step of the approach was to characterize the ion composition of the waterbody or region of interest and prepare artificial samples to match that composition. Using a combination of standardized toxicity test species and more ecologically relevant field‐collected species, toxicity tests were then conducted on these artificial samples prepared at a range of TDS concentrations. The advantage of this approach is that water quality criteria can be developed for easy to measure generalized parameters such as TDS while ensuring that the criteria are protective of instream aquatic life and account for the complex interactions of the various ions contributing to salinization. We test this approach in Sand Branch, Loudoun County, Virginia, USA, where salinization from hard rock mining and urban runoff has impaired aquatic life. Acute and chronic TDS thresholds of 938 and 463 mg/L, respectively, were developed in this stream and used for Total Maximum Daily Load development in the watershed. The approach provides a potential model for establishing protective thresholds for other waterbodies impacted by salinization.This article is protected by copyright. Proteome profile of spinneret from the silkworm, Bombyx mori

Abstract:The silkworm spinneret is an important tissue for silk fibrillogenesis and spinning. All biochemical processes during silk fibrillogenesis are correlated with silk properties. Understanding the role of spinneret in silk fibrillogenesis may help to reveal the mechanism of silk fibrillogenesis as well as improve silk quality for commercial purposes. Thus, we profiled the proteome of silkworm spinneret. A total of 1572 proteins and 232 differential abundance proteins were identified. Silk fibrillogenesis‐related proteins, such as cuticle proteins, ion‐transporting proteins, muscular proteins, and energy metabolic proteins, were abundant in spinneret. Metabolic pathway and GO enrichment analyses revealed that the identified proteins were involved in energy metabolism, chitin binding, and cuticle construction. Active energy metabolism may provide abundant energy for the muscle contraction as well as ion and water exchange. The chitin binding and cuticle construction process may provide sufficient shear forces for silk formation. Our data suggest that silkworm spinneret provides a suitable physiological and biochemical environment for silk fibrillogenesis. Global responses to tris(1-chloro-2-propyl) phosphate and tris(2-butoxyethyl) phosphate in Escherichia coli: Evidences from biomarkers, and metabolic disturbance using GC-MS and LC-MS metabolomics analyses

Abstract:Tris(1-chloro-2-propyl) phosphate (TCPP) and tris(2-butoxyethyl) phosphate (TBEP) as pollutants of emerging concern have aroused the rising attention due to their potential risks on aquatic ecosystem and public health. Nevertheless, there is a lack of toxicological mechanisms exploration of TCPP and TBEP at molecular levels. Herein, the toxicity effects and molecular mechanism of them were fully researched and summarized on Escherichia coli (E.coli). Acute exposure to them significantly activated antioxidant defense system and caused lipid peroxidation, as proved by the changes of antioxidant enzymes and MDA. The ROS overload resulted in the drop of membrane potential as well as the downregulated synthesis of ATPase, endorsing that E. coli cytotoxicity was ascribed to oxidative stress damage induced by TCPP and TBEP. The combination of GC-MS and LC-MS based metabolomics validated that TCPP and TBEP induced metabolic reprogramming in E.coli. More specifically, the responsive metabolites in carbohydrate metabolism, lipids metabolism, nucleotide metabolism, amino acid metabolism, and organic acids metabolism were significantly disturbed by TCPP and TBEP, confirming the negative effects on metabolic functions and key bioprocesses. Additionally, several biomarkers including PE(16:1(5Z)/15:0), PA(17:1(9Z)/18:2(9Z,12Z)), PE(19:1(9Z)/0:0), and LysoPE(0:0/18:1(11Z)) were remarkably upregulated, verifying that the protection of cellular membrane was conducted by regulating the expression of lipids-associated metabolites. Collectively, this work sheds new light on the potential molecular toxicity mechanism of TCPP and TBEP on aquatic organisms, and these findings using GC-MS and LC-MS metabolomics generate a fresh insight into assessing the effects of OPFRs on target and non-target aquatic organisms.

Abstract:Whole effluent toxicity (WET) tests are increasingly used to monitor compliance of consented discharges, but few studies have related toxicity measured using WET tests to receiving water impacts. Here we adopt a four-stage procedure to investigate the toxicity and biological impact of a point source discharge and to identify the major toxicants. In stage 1, standard WET tests were employed to determine the toxicity of the effluent. This was then followed by an assessment of receiving water toxicity using in situ deployment of indigenous (Gammarus pulex) and standard (Daphnia magna) test species. The third stage involved the use of biological survey techniques to assess the impact of the discharge on the structure and functioning of the benthic macroinvertebrate community. In stage 4, toxicity identification evaluations (TIE) were used to identify toxic components in the effluent. Receiving-water toxicity and ecological impact detected downstream of the discharge were consistent with the results of WET tests performed on the effluent. Downstream of the discharge, there was a reduction in D. magna survival, in G. pulex survival and feeding rate, in detritus processing, and in biotic indices based on macroinvertebrate community structure. The TIE studies suggested that chlorine was the principal toxicant in the effluent.
The Mitigating Effect of High Salinity on Chronic Nitrate Toxicity on Hexagrammos Otakii in Recirculating Aquaculture Systems

Abstract:Nitrate nitrogen, the end product of nitrogen conversion within the biofilter of recirculating aquaculture systems (RAS), has recently been recognized as a potentially toxic pollutant to fish. However, the influence of salinity on nitrate toxicity in juvenile Hexagrammos otakii (H. H. otakii) ) remains an area of uncertainty. In our study, a 5-week chronic expose experiment was conducted on juvenile H. otakii, , with a salinity of 30 PSU and nitrate-N of 12 mg/ Las control treatment (CT). Additionally, three treatment groups were established with salinities of 12, 21, and 30 PSU, all maintained at a nitrate-N concentration of 150 mg/L (T1, T2, and T3 respectively). The findings revealed pronounced decreases in growth and survival across all treatments, with the effects becoming more pronounced at lower salinity levels. Specifically, T1 exhibited elevated plasma nitrite concentrations and significantly reduced white and red blood cell counts, as well as hemoglobin levels. Histopathological evaluations of gill and liver tissues highlighted notable damage, particularly in the T1 group, impacting oxygen uptake and various metabolic functions. Gene ontology (GO) terms showed significant enrichment of categories about ion channel activity, gas transport, wnt signaling pathway and so on in the gill tissue among groups. In liver tissue, significant changes were observed in ion regulation, oxidoreductase activity, and immune response between the T1 and CT groups, though T2 and T3 showed minimal variances from CT. Furthermore, KEGG analysis indicated that PPAR signaling pathway in both gill and liver tissue was significantly inhibited in T1 group, which was a hallmark of tissue damage and disease development. This research demonstrates the protective role of increased salinity against nitrate toxicity in H. otakii culture. These findings have profound implications for understanding the interplay between salinity and nitrate toxicity, with potential applications in improving aquaculture health and management in RAS.

Neurotoxicity of tetramethylammonium ion on larval and juvenile zebrafish: Effects on neurobehaviors and multiple biomarkers

Abstract:Tetramethylammonium hydroxide (TMAH) is an important compound that utilized and released by the rapidly expanding semiconductor industry, which could hardly be removed by the conventional wastewater treatment techniques. As a cholinergic agonist, the tetramethylammonium ion (TMA+) has been reported to induce toxicity to muscular and respiratory systems of mammals and human, however the toxicity on aquatic biota remains poorly known. We investigated the neurotoxic effects of TMA+ exposure on zebrafish, based on neurobehavior tests and a series of biomarkers. Significant inhibitions on the swimming distance of zebrafish larvae were observed when the exposure level exceeded 50 mg/L, and significant alterations on swimming path angles (straight and deflective movements) occurred even at 10 mg/L. The tested neurobehavioral endpoints of zebrafish larvae were significantly positively correlated with reactive oxygen species (ROS) and malondialdehyde (MDA), significantly negatively related with the activities of antioxidant enzymes, but not significantly correlated with the level of acetylcholinesterase (AChE). Such relationship indicates that the observed neurotoxic effects on swimming behavior of zebrafish larvae is mainly driven by oxidative stress, rather than the alterations of neurotransmitter. At the highest exposure concentration (200 mg/L), TMA+ evoked more severe toxicity on zebrafish juveniles, showing significantly stronger elevation on the MDA activity, and greater inhibitions on the activities of antioxidant enzymes and AChE, suggesting juveniles were more susceptible to TMA+ exposure than larval zebrafish.

Abstract:In this research, the continuous physiological changes of zebrafish (Danio rerio) in 0.1 μg/L thallium (Tl) in 15 days were investigated. The results showed that Tl(I) stress had a significant positive linear correlation with zebrafish ammonia nitrogen excretion (ANE) (p < 0.001), and the mean value of ANE in Tl(I) treatment (435 ± 227 mg/kg/h) was approximately 2 times higher than in the control group (239 ± 168 mg/kg/h), which suggested that ANE was suitable for Tl(I) stress assessment. A substantial difference based on oxygen consumption rate (OCR) between the control group (587 ± 112 mg/kg/h) and Tl(I) treatment (260 ± 88 mg/kg/h) with a high significance p < 0.001 could be observed, and the results indicated that Tl(I) played a negative role in OCR of zebrafish. The characteristics of both ANE and OCR changes under slight Tl(I) stress could be reflected by the ammonia quotient (AQ). It was noteworthy that AQ increased rapidly in first 6 h from 0.66 to 4.50, which was 3 times higher than 1.2, indicating rapid increase in both anaerobic energy utilization and protein metabolism in 0.1 μg/L Tl(I) exposure. It is concluded that the physiological changes of zebrafish based on metabolism can be regarded as a sensitive biological indicator of Tl(I) pollution, which could work as a substitute of potassium that disrupts the normal biological metabolism in the process of transport.

Toxicity of representative mixture of five rare earth elements in juvenile rainbow trout (Oncorhynchus mykiss) juveniles

Abstract:Abstract Rare earth elements (REEs) are contaminants of increasing interest due to intense mining activities for commercial purposes and ultimately released in the environment. We exposed juvenile rainbow trout ( Oncorhynchus mykiss ) to a representative mixture of the five most abundant REEs for 96 h at concentrations similar found in lakes contaminated by mining activities at 0.1, 1, 10, and 100X whereas the 1x mixture contained cerium (Ce, 280 μg/L), lanthanum (La, 140 μg/L), neodymium (Nd, 120 μg/L), praseodymium (Pr, 28 μg/L), and samarium (Sm, 23 μg/L). We investigated the expression of 14 genes involved in oxidative stress, DNA repair, tissue growth/proliferation, protein chaperoning, xenobiotic biotransformation, and ammonia metabolism in the liver. In addition, DNA damage, oxidative stress (lipid peroxidation or LPO), inflammation (cyclooxygenase or COX activity), detoxification mechanisms (glutathione-S-transferase activity or GST), and labile zinc were determined in gills. The data revealed that genes involved in oxidative stress-catalase ( cat ), heat shock proteins 70 ( hsp70 ), and glutamate dehydrogenase ( glud ) were upregulated while glutathione S-transferase ( gst ) and metallothionein ( mt ) gene expressions were downregulated. The mixture was genotoxic and increased labile Zn in gills of exposed trout. These changes occurred at concentrations 600 times lower than the LC 50 for this mixture indicating effects below the 1X concentration. Based on principal component analysis and concentration-dependent reponses, the following sublethal effects were considered the most important/significant: DNA strand breaks (genotoxicity), labile Zn, cat , gst , hsp70 , sparc , mt , and glud. These effects of fish juveniles are likely to occur in environments under the influence of mining activities.

Abstract:Toxaphene is an organochlorine insecticide, widely used in cereals, vegetables, fruits, tea and other pests. However, in killing pests, increasing agricultural production, while toxaphene aquatic creatures threat. In order to enrich aquatic organisms toxicological data, evaluate the risk to aquatic organisms toxaphene healthy growth, we use the traditional toxicological approach to study the toxic effects of toxaphene goldfish (Carassius auratus)'s. Study 5 toxaphene concentration 7d goldfish exposed catalase (CAT), superoxide dismutase (SOD), acetylcholinesterase (TChE), glutathione peroxidase (GSH-PX), malondialdehyde (MDA) effects, the results showed that: toxaphene in 0.1,0.2,0.3,0.4, 0.5 μg / L in five concentrations, these five enzymes promoting effect, with more toxaphene concentration high, goldfish liver CAT activity is stronger, the stronger the brain and liver SOD activity, the activity of the brain TChE weaker, the stronger the brain activity of GSH-PX, and the contrast with the blank, P <0.01, illustrate the five enzymes do toxaphene toxicity testing of biomarkers.
Antioxidant defense system, immune response and erythron profile modulation in gold fish, Carassius auratus, after acute manganese treatment

Abstract:The manganese contamination has become a global problem, recently, because it is perceived as a real threat to the human health and the environment. It is well-known that overexposure to Mn2+ may have negative physiological effects on fish and other organisms inhabiting heavy metal polluted waters. To the best of our knowledge, studies relating with manganese effects on fish antioxidant enzyme response in the blood, immunocompetence and erythron profile alteration, are scarce. In this study, the acute sub-lethal effects of manganese on blood antioxidant response, immune status and erythron profile were determined by exposing the freshwater model organism, Carassius auratus, to two doses of this metal (3.88 ± 0.193 mg/L and 7.52 ± 0.234 mg/L Mn2+) for 96 h. Significant increases in blood antioxidant enzyme activity like superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST), were observed in fish exposed to manganese. Furthermore, plasmatic glucose and cortisol levels increased, while total protein decreased significantly. White blood cell differential count revealed a significant increase in monocyte and neutrophil number and a significant decrease of lymphocyte's number in fish exposed to manganese compared with those of control group. That can be considered as a clear evidence of altered immune system. Measured of erythron profile revealed a significant increasing of cellular and nuclear alteration of red blood cells, with karryorhectic, dividing and micronucleated erythrocytes in exposed fish, indicating the cytotoxic and genotoxic effects Mn2+ ions. Our data shown also that manganese could trigger antioxidant response, modulate immune response and induce erythron profile modification leading to eryptosis, compromising the blood oxygen carrying capacity, and overall health status in fish. This may suggest those parameters consider as useful biomarkers for monitoring effects of sub-lethal metal exposure on fish.

Abstract:The response of supraspinal neurons to acute or delayed treatment with GDNF following a spinal cord injury was examined. A cervical level 3 hemisection lesion cavity was created by tissue aspiration in adult, female rats. In one experiment gel foam saturated with GDNF was placed into the lesion cavity immediately after injury to determine if the extent of axonal retraction was affected by neurotrophic factor treatment. One week prior to sacrifice animals received a microinjection of biotinylated dextran amine (BDA) into the red nucleus and reticular formation to label descending spinal pathways by anterograde transport mechanisms. Animals were sacrificed 1 or 4 weeks after injury and treatment with GDNF. The terminal end of injured BDA-labeled rubrospinal and reticulospinal tract axons was identified and the distance from the lesion was measured. In comparison to PBS-treated animals, GDNF-treatment resulted in a significant decrease in the extent of axonal retraction of both rubrospinal and reticulospinal tract axons at 1 week after spinal cord injury for both tracts. At 4 weeks after injury the mean distance from the lesion was less than 240 microm following GDNF-treatment for both tracts, compared to over 480 microm following PBS-treatment. In the second experiment injured supraspinal neurons were labeled by retrograde transport of True Blue that had been placed into the lesion cavity. One month later scar tissue was removed from the cavity by aspiration to enlarge the cavity by approximately 500 microm in a rostral direction. GDNF-saturated gel foam was placed into the cavity for 60 min prior to apposition of an autologous peripheral nerve (PN) graft to the rostral cavity wall. One month later Nuclear Yellow was applied to the distal end of the PN graft and animals were sacrificed after 2 days. The number of supraspinal neurons containing both True Blue and Nuclear Yellow was counted as a measure of axonal regeneration by chronically injured neurons. There was a seven-fold increase in the number of regenerating neurons after GDNF-treatment, with the majority (65%) of dual-labeled neurons located within the reticular formation. These results indicate that GDNF has neuroprotective effects when provided acutely after injury and promotes axonal regeneration when provided in a chronic injury situation.
A tiered toxicity testing strategy for assessing early life stage toxicity in estuarine fish (Mugilogobius chulae): A case study on tris (1-chloro-2-propyl) phosphate ester

Abstract:The estuarine ecological environment faces significant threats from contaminants of emerging concern (CECs); yet, the risk posed by CECs to resident organisms remains poorly understood. Here, we employed tiered toxicity testing to investigate the adverse effects and potential mechanisms of tris (1-chloro-2-propyl) phosphate (TCPP) on the early life stages of an estuarine fish, Mugilogobius chulae. TCPP affected the development of M. chulae embryos, including survival, morphology, hatching, and behavior. A concentration-dependent transcriptomic analysis showed that TCPP disrupted 12 neurodevelopment-related KEGG pathways in M. chulae embryos, with five of the 30 % top-ranked pathways related to neurotransmitter signaling. Besides the cholinergic synapse signaling pathway, the glutamatergic signaling pathway (including NMDAR and AMPAR subtypes) may also mediate TCPP-induced neurodevelopmental toxicity. The NMDAR subtype GRIN2B was downregulated at high concentrations. Molecular dynamics simulations revealed a strong interaction between TCPP and GRIN2B, with TCPP binding to the residues Ile153 and Ile188. The results suggest that NMDARs play a crucial role in TCPP-induced neurodevelopmental toxicity toward M. chulae. AOP network analysis predicted that TCPP may impact cognitive functions and memory. Our study provides a novel testing strategy for identifying the mechanisms of toxicity of CECs, a crucial component of ecological risk assessment.
