Abstract:The high performance and increased tumor-targeting accumulation of magnetic nanocrystals (MNCs) are the most important considerations in cancer targeted magnetic hyperthermia (TMH). To achieve these goals, our study was firstly done using well-established fluorescence/magnetic Mn-Zn ferrite MNCs (core size: 14 nm) as multi-modal imaging contrast agents and highly-efficient "heat generators", which were coated with a biocompatible PEG-phospholipid (DSPE-PEG2000) and further modified by a cyclic tripeptide of arginine-glycine-aspartic acid (RGD). By using a mouse model bearing breast carcinoma (4T1), we then systematically compared PEGylated MNCs (MNCs@PEG)- and RGD-PEGylated MNCs (MNCs@RGD)-mediated tumor targeting abilities by intravenous administration. The MNCs@PEG-based passive targeting could successfully accumulate at the tumor due to the enhanced permeability and retention (EPR) effects, but the non-targeted localization might make the MNCs@PEG "leaking" from larger pores of tumor fenestrated vascular networks. Our designed MNCs@RGD, simultaneously functionalized with PEG and RGD ligands, might promote a synergistic effect including efficient tumor vasculature active targeting and EPR-mediated passive targeting, improving total MNC concentration and retention time in tumor tissues. By combining fluorescence/magnetic resonance (MR)/thermal multi-modal imaging-guided diagnostics and continuous TMH treatment under an alternating current magnetic field (ACMF, 2.58 kA m(-1), 390 kHz), the tumor surface could be heated to approximately 43-44 °C based on the MNC-mediated repeated injections. Sufficient temperature elevation induced the apoptosis of tumor cells, and inhibited the tumor angiogenesis. Compared with MNCs@PEG, the active MNCs@RGD-based tumor targeting MR image was significantly more efficient due to both the higher and long-lasting tumor accumulation, but its antitumor efficacy was not obviously improved in the TMH treatments. To achieve a singularly promising tumor TMH therapy, a greatly increased MNC content in tumor was needed. This insight indicated that not only the tumor vasculature targeting, but also the active tumor cells targeting of MNCs should receive considerable attention in future clinical TMH therapy application.

Abstract:Magnetic hyperthermia therapy (MHT) and chemodynamic therapy (CDT) are non-invasive in situ treatments without depth limitations and with minimum adverse effects on surrounding healthy tissue. We herein report a mitochondria-targeting magnetothermogenic nanozyme (Ir@MnFe2O4 NPs) for highly efficient cancer therapy. An iridium(III) complex (Ir) acts as a mitochondria-targeting agent on the surface of MnFe2O4 NPs. On exposure to an alternating magnetic field (AMF), the Ir@MnFe2O4 NPs induce a localized increase in temperature causing mitochondrial damage (MHT effect). Meanwhile glutathione (GSH) reduces Fe(III) to Fe(II) on the NPs surface, which in turn catalyzes the conversion of H2O2 to cytotoxic •OH (CDT effect). The depletion of GSH (a •OH scavenger) increases CDT efficacy, while the localized increase in temperature increases the rate of conversion of both Fe(III) to Fe(II) and H2O2 to •OH further enhancing the CDT effect. In addition, the disruption of cellular redox homeostasis due to CDT, leads to greater sensitivity of the cell towards MHT. This nanoplatform integrates these excellent therapeutic properties, with two-photon microscopy (TPM) (demonstrated in vitro) and magnetic resonance imaging (MRI) (demonstrated in vivo) to enable the precise and effective treatment of cancer.
High-performance PEGylated Mn-Zn ferrite nanocrystals as a passive-targeted agent for magnetically induced cancer theranostics.

Abstract:An effective magnetic nanocrystals (MNCs)-mediated theranostics strategy as a combination of simultaneous diagnostics and heating treatment of tumors by using magnetic resonance imaging (MRI) and alternating current magnetic field (ACMF) is successfully developed. In this strategy, we had firstly synthesized a well-established Mn–Zn ferrite MNCs coated with PEG-phospholipids (1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol copolymers, DSPE-PEG2000). The monodisperse PEGylated MNCs with core–shell structure (15 nm) exhibited excellent performance, such as high magnetism of 98 emu g−1 Fe, relaxivity coefficient (r2) of 338 mm−1 s−1, and specific absorption rate (SAR) value of 324 W g−1 Fe. It was proved that the obtained MNCs with an average diameter of 48.6 nm can drastically minimize the recognition and phagocytosis of macrophages, simultaneously improve their biocompatibility in vitro. These advantages endowed them with efficient passive targeting ability in vivo for prominent tumor MRI and magnetically induced heating when exposed to ACMF, based on enhanced permeability and retention (EPR) effects. To ensure sufficient accumulation of MNCs within tumors for targeted hyperthermia, we described the use of MNCs with a well-tolerated intravenous single dose of 18 mg Fe/kg mouse body weight, achieving repeatedly injection and hyperthermia within a subcutaneous breast cell carcinoma mouse model. With an ACMF of 12 A at 390 kHz, the tumor surface sites could be heated to approximately 43 °C in 30 min based on MNCs-mediated intravenous injections. The long-lasting hyperthermia could effectively induce the apoptosis of tumor cells, inhibit the angiogenesis of tumor vessels, and finally suppress the tumor growth within a certain period of time.

Abstract:Magnetic nanoparticle (MNP)-mediated magnetic hyperthermia (MHT) under an alternating magnetic field (AMF) causes tumor regression via reactive oxygen species (ROS) generation. However, less therapeutic efficacy has been reported due to the generation of low levels of ROS in a hypoxic tumor microenvironment. Therefore, improved treatments are required to generate relatively high levels of ROS to promote irreversible oxidative damage to the tumor cells. Herein, we report a magnetothermodynamic (MTD) therapy, as a robust and versatile approach for cancer treatment, by combining the magnetothermodynamic-related ROS and heat-related immunological effect in order to overcome the aforementioned obstacle. The synergistic therapy was achieved by the development of vitamin k3 (Vk3)-loaded copper zinc ferrite nanoparticles (Vk3@Si@CuZnIONPs) as an efficient MTD agent. The in vitro results unveiled that enhanced ROS production under the influence of AMF is a predominant aspect in yielding an assertive anticancer response. The in vivo antitumor response was assessed in an ectopic tumor model of A549 lung adenocarcinoma by MTD. The tumor inhibition rate of 69% was achieved within 20 days of MTD treatment, exhibiting complete tumor eradication within 30 days. The validation of antitumor response was marked by severe apoptosis (TUNEL, Caspase-3) in the Vk3@Si@CuZnIONPs + AMF-treated group. The higher expression level of heat shock proteins and proinflammatory cytokines (IL-6, TNF-α, IL-1α, IL-1β) was speculated to play a role in the activation of immune response for faster tumor regression in the MTD-treated group. Therefore, by implementing a dual ROS and heat-mediated immunogenic effect, the antitumor efficiency of future cancer magnetotherapies will be greatly enhanced.

Abstract:Cancer cells have higher heat sensitivity compared to normal cells; therefore, hyperthermia is a promising approach for cancer therapy because of its ability to selectively kill cancer cells by heating them. However, the specific and rapid heating of tumor tissues remains challenging. This study investigated the potential of magnetic nanoparticles (MNPs) modified with tumor-homing peptides (THPs), specifically PL1 and PL3, for tumor-specific magnetic hyperthermia therapy. The synthesis of THP-modified MNPs involved the attachment of PL1 and PL3 peptides to the surface of the MNPs, which facilitated enhanced tumor cell binding and internalization. Cell specificity studies revealed an increased uptake of PL1- and PL3-MNPs by tumor cells compared to unmodified MNPs, indicating their potential for targeted delivery. In vitro hyperthermia experiments demonstrated the efficacy of PL3-MNPs in inducing tumor cell death when exposed to an alternating magnetic field (AMF). Even without exposure to an AMF, an additional ferroptotic pathway was suggested to be mediated by the nanoparticles. Thus, this study suggests that THP-modified MNPs, particularly PL3-MNPs, hold promise as a targeted approach for tumor-specific magnetic hyperthermia therapy.

Abstract:In this work, we report a ultrahigh specific absorption rate (SAR) performance in Zn-substituted magnetite superparamagnetic (SPM) nanoparticles (NPs) for potential application in magnetic hyperthermia (MHT)-based cancer treatment. Although MHT shows promise in cancer therapy, challenges such as low heating performance, agglomeration of magnetic nanoparticles (MNPs) in blood veins, cytotoxicity, and hemocompatibility have hindered clinical uses. To overcome these challenges, we adopted a reverse-micelles based coprecipitation synthesis approach to prevent MNPs agglomeration and optimized the substitution of Zn ions in magnetite to enhance heating performance. Calorimetric measurements showed SAR values of 118 W/g and 181 W/g for magnetite NPs using the initial slope method (ISM) and Box Lucas method (BLM), respectively. Through our compositional optimizations, we achieved a significant increase in SAR value by consciously substituting Zn 2+ ions in the magnetite lattice. Specifically, we obtained SAR values of 325 W/g and 579 W/g (>300% increase) for Zn 0.3 Fe 2.7 O 4 MNPs using ISM and BLM, respectively. This enhancement can be attributed to improved saturation magnetization (174–257 kA/m) and magneto-crystalline anisotropy (12–24 kJ/m 3 ). The increase in saturation magnetization in the magnetite MNPs can be explained by the higher magnetic moment resulting from increased Zn concentration up to Zn x Fe 3-x O 4 (x = 0.3), strengthening the J AB interaction. However, further increases in Zn concentration lead to a decrease in saturation magnetization due to non-collinearity, as described by the Yafet-Kittle model. Our optimized MNPs exhibit improved heating performance, enabling the use of lower MNPs concentrations for cancer treatment, reducing potential toxicity effects. Biocompatibility investigations demonstrated a low hemolysis rate ( 92% cell viability) in MTT assays for all compositions, confirming their potential suitability for clinical applications. This study offers a promising approach to enhance SAR performance, addressing MHT-based cancer treatment challenges, and improving clinical suitability.

Abstract:IntroductionTumor cells can effectively be killed by heat, e.g. by using magnetic hyperthermia. The main challenge in the field, however, is the generation of therapeutic temperatures selectively in the whole tumor region. We aimed to improve magnetic hyperthermia of breast cancer by using innovative nanoparticles which display a high heating potential and are functionalized with a cell internalization and a chemotherapeutic agent to increase cell death.MethodsThe superparamagnetic iron oxide nanoparticles (MF66) were electrostatically functionalized with either Nucant multivalent pseudopeptide (N6L; MF66-N6L), doxorubicin (DOX; MF66-DOX) or both (MF66-N6LDOX). Their cytotoxic potential was assessed in a breast adenocarcinoma cell line MDA-MB-231. Therapeutic efficacy was analyzed on subcutaneous MDA-MB-231 tumor bearing female athymic nude mice.ResultsAll nanoparticle variants showed an excellent heating potential around 500 W/g Fe in the alternating magnetic field (AMF, conditions: H = 15.4 kA/m, f = 435 kHz). We could show a gradual inter- and intracellular release of the ligands, and nanoparticle uptake in cells was increased by the N6L functionalization. MF66-DOX and MF66-N6LDOX in combination with hyperthermia were more cytotoxic to breast cancer cells than the respective free ligands. We observed a substantial tumor growth inhibition (to 40% of the initial tumor volume, complete tumor regression in many cases) after intratumoral injection of the nanoparticles in vivo. The proliferative activity of the remaining tumor tissue was distinctly reduced.ConclusionThe therapeutic effects of breast cancer magnetic hyperthermia could be strongly enhanced by the combination of MF66 functionalized with N6L and DOX and magnetic hyperthermia. Our approach combines two ways of tumor cell killing (magnetic hyperthermia and chemotherapy) and represents a straightforward strategy for translation into the clinical practice when injecting nanoparticles intratumorally.

Abstract:Tumor-associated macrophages (TAMs) play an important role in tumor development and progression. In particular, M2 TAMs can promote tumor growth by facilitating tumor progression and malignant behaviors. Selectively targeted elimination of M2 TAMs to inhibit tumor progression is of great significance for cancer treatment. Iron oxide nanoparticles based magnetic hyperthermia therapy (MHT) is a classical approach to destroy tumor tissue with deep penetration depth. In this study, we developed a typical M2 macrophage-targeted peptide (M2pep) functionalized superparamagnetic iron oxide nanoparticle (SPIO) for magnetic resonance imaging (MRI)-guided MHT in an orthotopic breast cancer mouse model. The obtained multifunctional SPIO-M2pep with a hydrodynamic diameter of 20 nm showed efficient targeting capability, high transverse relaxivity (149 mM(-1) s(-1)) and satisfactory magnetic hyperthermia performance in vitro. In vivo studies demonstrated that the SPIO-M2pep based MRI can monitor the distribution of nanoparticles in tumor and indicate the suitable timing for MHT. The M2 macrophage-targeted MHT significantly reduced the tumor volume and the population of pro-tumoral M2 TAMs in tumor. In addition, the SPIO-M2pep based MHT can remodel the tumor immune microenvironment (TIME). The multifunctional SPIO-M2pep with M2 macrophage-targeting ability, high magnetic hyperthermia efficiency, MR imaging capability and effective role in remodeling the TIME hold great potential to improve clinical cancer therapy outcomes. Redox-Responsive Functional Iron Oxide Nanocrystals for Magnetic Resonance Imaging-Guided Tumor Hyperthermia Therapy and Heat-Mediated Immune Activation

Abstract:Magnetic nanoparticle-mediated mild hyperthermia is an emerging approach for tumor treatment and antitumor immune activation. However, the precise delivery of magnetic hyperthermia nanoagents inside tumor for mild magnetic hyperthermia therapy (MHT) via intravenous administration remains challenging. In this study, we developed a tumor microenvironment (TME)-responsive nanosystem based on cleavable methoxypolyethene glycol (mPEG) and transactivator of transcription (TAT) cell-penetrating peptide dual-decorated Fe3O4 nanocrystals (@Fe3O4) and utilized it to facilitate effective magnetic resonance (MR) imaging, tumor MHT, and mild heat-mediated immune stimulation. The as-constructed @Fe3O4 exhibited supreme sensitivity to the reducing environment in the TME for detaching the mPEG shell and strongly inhibited the proliferation of tumor cells with exposure to an alternating magnetic field. In vivo experiments in CT26 tumor-bearing mice showed that this nanodelivery system received an enhanced T2-weighted MR imaging-guided tumor MHT by achieving an 85.5% tumor inhibition rate and induced a magnetic hyperthermia–immune synergistic therapy. Notably, the structure–variable delivery system showed a lower intravenous dosage than previously reported agents at a temperature of 43–44 °C induced by MHT. Abstract:A therapeutic breakthrough in cancer treatment has recently been made by using magnetic nanoparticles (MNPs) for heating in hyperthermia therapy. Unapproachable tumors are being effectively destroyed by focused heat produced by MNPs. The major challenges regarding practical application of this therapy include the control and improvement of induction heating ability of MNPs and hyperthermia temperature range between 42–47 °C, for secure treatment at targeted area. Here, in this study, we have reported the synthesis and characterization of core–shell structured silica-coated cobalt ferrite nanoparticles (SiO2 coated CoFe2O4 MNPs), which are potential candidates for use as heat source in magnetic hyperthermia therapy. The SiO2 coated CoFe2O4 MNPs were synthesized using the reverse micelle method, with the SiO2 coating performed simultaneously during MNP synthesis. Various analytical tools were utilized for the characterization. The structural measurements were probed by X-ray diffraction (XRD). Monodisperse MNPs with nearly spherical core–shell structure was revealed by Transmission electron microscopy (TEM) results. The average diameter of MNPs obtained from TEM analysis was 15 nm. The surface coating of cobalt ferrite MNPs with silica was verified by Fourier transform infrared spectrometry (FTIR). The saturation magnetization values obtained using vibrating sample magnetometer (VSM) measurements were 45.74 emu/g, revealing the superparamagnetic nature of the nanoparticles. The heating efficiency of different concentrations of synthesized nanoparticles was evaluated in an aqueous solution under an alternating magnetic field of strength 5.5 kA/m at a frequency of 260 kHz. A saturation temperature of 42 °C was attained at an optimum concentration of 1.7 mg/mL, while a magnetic field strength of 3.9 kA/m achieved 42 °C at a fixed concentration of 3.5 mg/ml. The obtained specific absorption rate (SAR) values for all the samples were between 56.23 to 100.1 W/g. The correlation between SAR values, MNP concentration, and applied magnetic field strength was also examined. The high saturation magnetization, efficient heating rates, and high SAR values make our synthesized silica-coated cobalt ferrite nanoparticles promising candidates for magnetic hyperthermia treatments, potentially improving therapeutic outcomes for cancer patients.

Abstract:Cells of the obligate halophile Halobacterium cutirubrum were grown to the stationary phase in the presence of 32P-orthophosphate, 35S-sulfate, or 14C-labelled precursors (acetate, malonate, mevalonate, glycerol). 32P was incorporated into the phosphatide components in proportion to their relative concentrations, thus: phosphatidyl glycerophosphate (diether analogue) > unidentified minor phosphatide > phosphatidyl glycerol (diether analogue). 35S-Sulfate was incorporated mostly into the glycolipid sulfate ester component, but small amounts of 35S also appeared in a more polar sulfolipid and in a minor unidentified sulfolipid.14C-Mevalonate was the most efficient precursor for the phytanyl chains, followed in decreasing order by acetate and glycerol. Abstract:Magnetic hyperthermia using ferrite nanoparticles has emerged as a eutherapeutic approach for cancer treatment. In this study, we had constructed folate conjugated poly(ethyleneimine) (PEI) modified magnetic nanoparticles (FA-PEI-MNPs) that generate heat in an alternating magnetic field (AMF). Transmission electron microscope (TEM) showed that FA-PEI-MNPs were nearly spherical in shape. The mean diameter of these nanoparticles determined by DLS was 30.7 +/- 7.7 nm. The cytotoxicity assay showed that FA-PEI-MNPs exhibited no significant cytotoxicity against L929 cells. FA-PEI-MNPs have good hyperthermia effect on KB cells in vitro by magnetic field induced hyperthermia, especially at 45 degrees C and 47 degrees C. In in vivo hyperthermia treatment using FA-PEI-MNPs in a subcutaneous tumor xenograft model of KB cells tumor, the tumor temperature increased to 48 degrees C whereas the body temperature remained at around 37.5 degrees C. Thermotherapy with FA-PEI-MNPs led up to prolongation of survival over magnetic field controls, 66.7% (4/6) of subcutaneous tumors regressed completely. Histological examinations after treatment revealed large necrotic areas close to particle deposits. These results suggest that hyperthermia using FA-PEI-MNPs is an effective and specific therapy for tumor. This therapy may provide an alternative way to treat recurrent cancer refractory to other modalities.
Genetically engineered magnetic nanocages for cancer magneto-catalytic theranostics

Abstract:The clinical applications of magnetic hyperthermia therapy (MHT) have been largely hindered by the poor magnetic-to-thermal conversion efficiency of MHT agents. Herein, we develop a facile and efficient strategy for engineering encapsulin-produced magnetic iron oxide nanocomposites (eMIONs) via a green biomineralization procedure. We demonstrate that eMIONs have excellent magnetic saturation and remnant magnetization properties, featuring superior magnetic-to-thermal conversion efficiency with an ultrahigh specific absorption rate of 2390 W/g to overcome the critical issues of MHT. We also show that eMIONs act as a nanozyme and have enhanced catalase-like activity in the presence of an alternative magnetic field, leading to tumor angiogenesis inhibition with a corresponding sharp decrease in the expression of HIF-1α. The inherent excellent magnetic-heat capability, coupled with catalysis-triggered tumor suppression, allows eMIONs to provide an MRI-guided magneto-catalytic combination therapy, which may open up a new avenue for bench-to-bed translational research of MHT.

Abstract:At present, iron oxide magnetic nanoparticles (MNPs) are commonly developed for cancer treatment. It combines with the external magnetic field for magnetically controlled targeting (accumulating at the tumor site), magnetic hyperthermia (promoting the production of ROS and activating the maturation of immune‐related cells), and magneto‐mechanical effect of particles (disrupting cellular structure, promoting cell apoptosis or necrosis). Iron oxide nanoparticles (MNPs) demonstrate notable benefits in magnetic induction, attributed to their distinctive physical and chemical attributes. Emerging cancer treatment utilizing magnetic fields have also gathered increasing attention in the biomedical field. However, the defects of difficult dispersion and poor biocompatibility of MNPs seriously hinder their application. In order to overcome its inherent defects and maximize the therapeutic potential of MNPs, various functionalized MNPs have been developed, and numerous combined treatment methods based on MNPs have been widely studied. In this review, we compare and analyze the common nanoparticles based on MNPs with different sizes, shapes, and functional modifications. Additionally, we introduced the therapeutic mechanisms of the strategies, such as magnetically controlled targeting, magnetic hyperthermia, and magneto‐mechanical effect, which based on the unique magnetic induction capabilities of MNPs. Finally, main challenges of MNPs as smart nanomaterials were also discussed. This review seeks to offer a thorough overview of MNPs in biomedicine and a new sight for their application in tumor treatment.

Abstract:Immunotherapy is based on the systemic immunity activation against malignacies, which is becoming one of the most promising modalities in solid tumor therapy. Unfortunately, the outcomes of the present immunotherapy remain unsatisfactory or even disappointing in most types of solid tumors, largely because of the immunosuppressive tumor microenvironment and hardly permeable barriers of solid tumors against immune cell infiltrations. Herein, a novel strategy of concurrently stimulating innate and adaptive immunity while enhancing immune cell infiltration in solid tumor, is proposed using a rationally designed magnetic nanocatlytic medicine (ZnCoFe 2 O 4 @ZnMnFe 2 O 4 -PBA, ZCMFP) for mild magnetic hyperthermia therapy (MHT) mediated immune response activations. Briefly, ZCMFP generates reactive oxygen species (ROS) catalyzed by released Fe 2+/3+ ions from the nanomedicine in response to acidic tumor microenvironment (TME), provoking the innate immunity by triggering M1 polarization of tumor-associated macrophages (TAMs). Meanwhile, the co-released Mn 2+ promotes the maturation of dendritic cells (DCs) via STING pathway for antigen presentation, which awakes T cells and thus activates the adaptive immunity. The mild MHT also regulates heat shock protein (HSP) and reduces vascular endothelial growth factor (VEGF) intratumorally, resulting in the elevated expressions of adhesion molecules (ICAM-1/VCAM-1) by endothelial cells, which facilitates the recruitment and infiltration of effector immune cells. This strategy features robust and durable immune responses leading to the marked inhibitions on the growths both orthotopic and metastasis tumors, providing a possible solution for solid tumor immunotherapy in clinics.

Abstract:Hypothesis Multicore flower-like iron oxide nanoparticles (IONPs) are among the best candidates for magnetic hyperthermia applications against cancers. However, they are rarely investigated in physiological environments and their efficacy against cancer cells has been even less studied. The combination of magnetic hyperthermia, using multicore IONPs, with selected bioactive molecules should lead to an enhanced activity against cancer cells. Experiments Multicore IONPs were synthesized by a seeded-growth thermal decomposition approach. Then, the cytotoxicity, cell uptake, and efficacy of the magnetic hyperthermia approach were studied with six cancer cell lines: PANC1 (pancreatic carcinoma), Mel202 (uveal melanoma), MCF7 (breast adenocarcinoma), MB231 (triple-negative breast cancer line), A549 (lung cancer), and HCT116 (colon cancer). Finally, IONPs were modified with a chemotherapeutic drug (SN38) and tumor suppressor microRNAs (miR-34a, miR-182, let-7b, and miR-137), to study their activity against cancer cells with and without combination with magnetic hyperthermia. Findings Two types of multicore IONPs with very good heating abilities under magnetic stimulation have been prepared. Their concentration-dependent cytotoxicity and internalization have been established, showing a strong dependence on the cell line and the nanoparticle type. Magnetic hyperthermia causes significant cell death that is dramatically enhanced in combination with the bioactive molecules.

Abstract:Photodynamic therapy is widely used in clinics and for anti-bacterial applications. The major challenge is the limited depth of tissue penetration of light and poor targetability. In this study, magnetite nanoparticles were used as a highly sensitive T-weighted MR imaging contrast agents to target the tumor and mimic horseradish peroxidase (HRP), which could catalyze the decomposition of hydrogen peroxide to generate reactive oxygen species (ROS) to inhibit the tumor in vivo. In these experiments, MNPs were demonstrated to possess the enzyme-mimicking activity in different pH values, and the activity was dependent on the size of the MNPs: the smaller the size, the higher the activity. We demonstrated that MNPs showed highly efficient anti-bacterial (E. coli) activity in presence of HO. The E. coli inhibition ratio reached nearly 100% at optimal concentration. The anti-tumor activity was evaluated through HeLa cell viability under treatment with MNPs and HO. Consequently, the cell viability was significantly decreased and more than 80% of HeLa cells were dead after treatment with MNPs and HO under different pH values. MR imaging was used to demonstrate the tumor targetability of 13 nm MNPs in vitro and in vivo. Consequently, the relaxivity of the 13 nm MNPs was determined to be r = 104 s mM. The MR signal was much more negative and the intensity was significantly diminished with the increase of the concentration of 13 nm MNPs in vitro. The tumor signal was clearly visualized and a 3-fold decrease of the MR signal intensity of the tumor site of the mice was observed after 24 h-post treatment with the 13 nm MNPs. Finally, the tumor inhibition in vivo was investigated using 6 nm MNPs in BALB/c nude female mice bearing subcutaneously implanted HeLa cells on the right flank. The results show statistically significant efficacy in delaying tumor growth from day 6, and an approximately 99% tumor inhibition ratio was shown by the combination of MNPs and HO after treatment for 17 days. By leveraging the passive targeting and MR imaging properties, we expect that the enzyme-mimicking MNPs could be used for cancer theranostics and may open up a new avenue for the treatment of epidermal infections.
